Background and aims

Nitrogen (N) availability affects water uptake from the roots, which decreases upon N deprivation and increases upon resupply. The aim of this study was to reveal possible mechanisms of regulation of water transport in roots through physiological and morphological adaptations to N availability.

Methods

The effects of continuous N deprivation and following resupply on root morphology, osmotic hydraulic conductivity, and expression of genes for aquaporins (water channels) were examined in rice (Oryza sativa L.) plants. The effect of local N availability was examined by using a split-root system.

Results

N deprivation decreased the expression of root-specific aquaporin genes, whereas N resupply increased their expression. Changes in aquaporin gene expression were correlated with changes in hydraulic conductivity. N deprivation increased dry matter allocation to the roots. In a split-root experiment, the expression of root-specific aquaporin genes was down-regulated in the N-deprived half, whereas it was up-regulated in the N-supplied half.

Conclusion

Our results suggest that expression of genes for root-specific aquaporins underlies the changes in conductivity during continuous N deprivation and resupply. Rice plants seem to adapt to N availability through coordinated adjustment of root proliferation and abundance of aquaporins.  相似文献

Background and aims

Whether root Zn uptake during grain filling or remobilization from pre-anthesis Zn stores contributes more to grain Zn in wheat is subject to an on-going debate. This study investigated the effects of N nutrition and post-anthesis Zn availability on the relative importance of these sources.

Methods

Durum wheat plants were grown in nutrient solution containing adequate Zn (0.5?μM) and three different N levels (0.5; 1.5; 4.5?mM). One third of the plants were harvested when they reached anthesis. One half of the remaining plants were grown to maturity with adequate Zn, whereas the Zn supply to the other half was discontinued at anthesis. Roots, straw and grains were harvested separately and analyzed for Zn and N.

Results

Depending on the N supply, Zn remobilization from pre-anthesis sources provided almost all of grain Zn when the Zn supply was withheld at anthesis; otherwise up to 100?% of grain Zn could be accounted for by Zn taken up post-anthesis. By promoting tillering and grain yield and extending the grain filling, higher N supply favored the contribution of Zn uptake to grain Zn accumulation.

Conclusion

Remobilization is critical for grain Zn accumulation when Zn availability is restricted during grain filling. However, where root uptake can continue, concurrent Zn uptake during grain development, favored by higher N supply, overshadows net remobilization.  相似文献

Background and aim

Root system activity is affected by abiotic stresses, which often creates spatial differences in root conditions. This is expected to influence plants ability to cope with suboptimal conditions.

Methods

Changes in root system activity were determined as 15N root uptake in top and bottom layers of potted tomato plants (Solanum lycopersicum L.), after waterlogging the bottom layer for 24 h or 5 d. The plants were grown in peat-based media; non-compacted or highly-compacted, resulting in differences in gas diffusion, air permeability and oxygen availability.

Results

The roots were affected by short-term waterlogging (24 h) by decreasing uptake in the bottom layer and increasing uptake in the pot top layer. Long-term waterlogging (5 d) decreased the 15N root uptake more in both layers. Root uptake recovered fast (within 6 h) after short-term waterlogging, whereas recovery of long-term waterlogged roots took more than 24 h, suggesting production of new root biomass. Despite affecting physical properties, medium compaction did not affect root uptake. Aboveground biomass was affected by waterlogging by increasing the dry matter percentage, decreasing nitrogen (N) percentage and increasing starch content.

Conclusions

The results confirmed that root uptake in different layers of small pots could be distinguished by the 15N technique, which was applicable under potentially denitrifying conditions. The results demonstrated that during short-term stress in part of the root system plants increased uptake from the non-affected parts of the root system, probably as compensation for suboptimal conditions.  相似文献

Aims

We determined whether nitrogen (N) deposition on phosphorus (P)-limited soil could increase Schima superba growth or alter root formation or P efficiency. The effects of N deposition on S. superba were also used to investigate the N/P requirements of plants of different provenances.

Methods

One-year-old S. superba seedlings from eight geographic areas were grown in P-limited soil and treated with N (0, 50, 100, and 200 -kg?N?ha?1?year?1; i.e., control, N50, N100, or N200, respectively). Seedling growth, root development, phosphorus acquisition efficiency (PAE), and phosphorus utilization efficiency (PUE) were measured.

Results

S. superba responded positively to N supplementation. Seedling growth and dry biomass were highest with N100 treatment and lowest with N200. Root biomass and acquisition of soil P were greatest with N100. Significant differences were observed among plants of different geographical provenances. PAE and PUE had a strong relationship with root growth in plants subjected to N100 treatment.

Conclusion

A threshold for N and P requirements related to different genetic conditions and soil nutrients may exist for S. superba. Root growth and PAE can be divided into three categories based on soil nitrate levels. Nutrients were found to control root morphology and to enhance aboveground differences.  相似文献

Background and aims

Litter decomposition is regulated by e.g. substrate quality and environmental factors, particularly water availability. The partitioning of nutrients released from litter between vegetation and soil microorganisms may, therefore, be affected by changing climate. This study aimed to elucidate the impact of litter type and drought on the fate of litter-derived N in beech seedlings and soil microbes.

Methods

We quantified 15N recovery rates in plant and soil N pools by adding 15N-labelled leaf and/or root litter under controlled conditions.

Results

Root litter was favoured over leaf litter for N acquisition by beech seedlings and soil microorganisms. Drought reduced 15N recovery from litter in seedlings thereby affecting root N nutrition. 15N accumulated in seedlings in different sinks depending on litter type.

Conclusions

Root turnover appears to influence (a) N availability in the soil for plants and soil microbes and (b) N acquisition and retention despite a presumably extremely dynamic turnover of microbial biomass. Compared to soil microorganisms, beech seedlings represent a very minor short-term N sink, despite a potentially high N residence time. Furthermore, soil microbes constitute a significant N pool that can be released in the long term and, thus, may become available for N nutrition of plants.  相似文献

Background and aims

Accurate predictions of nutrient acquisition by plant roots and mycorrhizas are critical in modelling plant responses to climate change.

Methods

We conducted a field experiment with the aim to investigate root nutrient uptake in a future climate and studied root production by ingrowth cores, mycorrhizal colonization, and fine root N and P uptake by root assay of Deschampsia flexuosa and Calluna vulgaris.

Results

Net root growth increased under elevated CO2, warming and drought, with additive effects among the factors. Arbuscular mycorrhizal colonization increased in response to elevated CO2, while ericoid mycorrhizal colonization was unchanged. The uptake of N and P was not increased proportionally with root growth after 5 years of treatment.

Conclusions

While aboveground biomass was unchanged, the root growth was increased under elevated CO2. The results suggest that plant production may be limited by N (but not P) when exposed to elevated CO2. The species-specific response to the treatments suggests different sensitivity to global change factors, which could result in changed plant competitive interactions and belowground nutrient pool sizes in response to future climate change.  相似文献

Arabidopsis Ca 2+ -ATPase ACA8 plays a role in sucrose signalling during early seedling development by integrating developmental signals with carbon source availability.

Abstract

Calcium (Ca2+) is an essential signal transduction element in eukaryotic organisms. Changes in the levels of intracellular Ca2+ affect multiple developmental processes in plants, including cell division, polar growth, and organogenesis. Here, we report that the plasma-membrane-localised Arabidopsis Ca2+-ATPase ACA8 plays a role in sucrose signalling during early seedling development. Disruption of the ACA8 gene elevated the expression of genes that encode transporters for Ca2+ efflux. The seedlings that carried a T-DNA insertion mutation in ACA8 experienced water stress during early development. This response was unrelated to inadequate osmoregulatory responses and was most likely caused by disruption of cell membrane integrity and severe ion leakage. In addition, aca8-1 seedlings displayed a significant decline in photosynthetic performance and arrested root growth after removal of sucrose from the growth medium. The two phenomena resulted from impaired photosynthesis, reduced cell proliferation in the root meristem and the sucrose control of cell-cycle events. All of the stress-response phenotypes were rescued when expression of ACA8 was restored in aca8-1 mutant. Taken together, our results indicate that ACA8-mediated Ca2+ signalling contributes to modulate early seedling development and coordinates root development with nutrient availability.  相似文献

Background and aims

Incorporating soybean (Glycine max) genotypes with a high nitrogen fixation potential into cropping systems can sustainably improve the livelihoods of smallholder farmers in Western Kenya. Nitrogen fixation is, however, often constrained by low phosphorus (P) availability. The selection of soybean genotypes for increased P efficiency could help to overcome this problem. This study investigated the contribution of different root traits to variation in P efficiency among soybean genotypes.

Methods

Eight genotypes were grown in a Ferralsol amended with suboptimal (low P) and optimal (high P) amounts of soluble P. Root hair growth was visualized by growing plants in a novel agar system where P intensity was buffered by Al2O3 nanoparticles.

Results

In the pot trial, P uptake was unaffected among the genotypes at high P but differed about 2-fold at low P. The genotypes differed in P uptake efficiency but not in P utilization efficiency. Regression analysis and mechanistic modeling indicated that P uptake efficiencies were to a large extent related to root hair development (length and density) and, to a lower extent, to colonization by mycorrhizal fungi.

Conclusion

Breeding for improved root hair development is a promising way to increase P uptake efficiency in soybean.  相似文献

Backgrounds and aims

Plant nutrition strategies play a crucial role in community structure and ecosystem functioning. However, these strategies have been established only for nitrogen (N) acquisition, and it is not known whether similar strategies hold for other macronutrients such as sulphur (S). The aim of our study was to determine whether strategies for S acquisition of some grassland species were similar to those observed for N acquisition, and to analyse the relationships between these plant strategies and the soil microbial activity involved in soil organic S mineralisation.

Methods

We used three exploitative and three conservative grass species grown with and without S fertilisation. We measured a set of plant traits, namely root and shoot biomass, leaf area, root length, N and S content, leaf nutrient use efficiency, and sulphate uptake rates in plants, and one microbial trait linked to S mineralisation, namely soil arylsulphatase activity.

Results

The set of plant traits differentiated exploitative from conservative species. Close relationships were found between traits associated with strategies for N acquisition, namely total N content and Leaf N Use Efficiency (LNUE), and traits associated with strategies for S acquisition, namely total S content and Leaf S Use Efficiency (LSUE). Exploitative species exhibited similar or lower sulphate uptake capacities per unit of biomass than conservative species, but acquired more S through their larger root systems. Greater arylsulphatase activity was observed in the rhizosphere of the most exploitative species.

Conclusion

Overall, our results show that nutrient strategies defined in grassland species for N acquisition can be extended to S.  相似文献

Background and Aims

For plants growing in living walls, the growth potential is correlated to the roots ability to utilize resources in all parts of the growing medium and thereby to the spatial root distribution. The aim of the study was to test how spatial root distribution was affected by growing medium, planting position and competition from other plants.

Methods

Five species (Campanula poscharskyana cv. ‘Stella’, Fragaria vesca cv. ‘Småland’, Geranium sanguineum cv. ‘Max Frei’, Sesleria heufleriana and Veronica officinalis cv. ‘Allgrün’) were grown in three growing media (coir and two of rockwool) in transparent boxes under greenhouse conditions. Root frequency was registered and the activity of individual root systems was studied via 15N uptake and plant dry weight was measured.

Results

Plants in coir had stronger root growth in all parts of the medium than plants in rockwool. Upwards root growth was limited for plants in the middle or lower parts of the medium and 15N measurements confirmed that only plants in the bottom of the box had active roots in the bottom of the medium. The species differed in root architecture and spatial root distribution.

Conclusions

The choice of growing medium, plant species and planting position is important for a living wall as it affects the spatial root growth of the plants.  相似文献

Aims

The aim was to quantify the nitrogen (N) transferred via the extra-radical mycelium of the arbuscular mycorrhizal fungus Glomus intraradices from both a dead host and a dead non-host donor root to a receiver tomato plant. The effect of a physical disruption of the soil containing donor plant roots and fungal mycelium on the effectiveness of N transfer was also examined.

Methods

The root systems of the donor (wild type tomato plants or the mycorrhiza-defective rmc mutant tomato) and the receiver plants were separated by a 30 μm mesh, penetrable by hyphae but not by the roots. Both donor genotypes produced a similar quantity of biomass and had a similar nutrient status. Two weeks after the supply of 15?N to a split-root part of donor plants, the shoots were removed to kill the plants. The quantity of N transferred from the dead roots into the receiver plants was measured after a further 2 weeks.

Results

Up to 10.6 % of donor-root 15N was recovered in the receiver plants when inoculated with the arbuscular mycorrhizal fungus (AMF). The quantity of 15N derived from the mycorrhizal wild type roots clearly exceeded that from the only weakly surface-colonised rmc roots. Hyphal length in the donor rmc root compartments was only about half that in the wild type compartments. The disruption of the soil led to a significantly increased AMF-mediated transfer of N to the receiver plants.

Conclusions

The transfer of N from dead roots can be enhanced by AMF, especially when the donor roots have been formerly colonised by AMF. The transfer can be further increased with higher hyphae length densities, and the present data also suggest that a direct link between receiver mycelium and internal fungal structures in dead roots may in addition facilitate N transfer. The mechanical disruption of soil containing dead roots may increase the subsequent availability of nutrients, thus promoting mycorrhizal N uptake. When associated with a living plant, the external mycelium of G. intraradices is readily able to re-establish itself in the soil following disruption and functions as a transfer vessel.  相似文献

Background & Aims

Searching for root traits underpinning efficient nutrient acquisition has received increased attention in modern breeding programs aimed at improved crop productivity. Root models provide an opportunity to investigate root-soil interactions through representing the relationships between rooting traits and the non-uniform supply of soil resources. This study used simulation modelling to predict and identify phenotypic plasticity, root growth responses and phosphorus (P) use efficiency of contrasting Lupinus angustifolius genotypes to localised soil P in a glasshouse.

Methods

Two L. angustifolius genotypes with contrasting root systems were grown in cylindrical columns containing uniform soil with three P treatments (nil and 20 mg P kg?1 either top-dressed or banded) in the glasshouse. Computer simulations were carried out with root architecture model ROOTMAP which was parameterized with root architectural data from an earlier published hydroponic phenotyping study.

Results

The experimental and simulated results showed that plants supplied with banded P had the largest root system and the greatest P-uptake efficiency. The P addition significantly stimulated root branching in the topsoil, whereas plants with nil P had relatively deeper roots. Genotype-dependent root growth plasticity in response to P supply was shown, with the greatest response to banded P.

Conclusions

Both experimental and simulation outcomes demonstrated that 1) root hairs and root proliferation increased plant P acquisition and were more beneficial in the localised P fertilisation scenario, 2) placing P deeper in the soil might be a more effective fertilisation method with greater P uptake than top dressing, and 3) the combination of P foraging strategies (including root architecture, root hairs and root growth plasticity) is important for efficient P acquisition from a localised source of fertiliser P.  相似文献

Background and aims

Biochar has been proposed as a tool to enhance phytostabilisation of contaminated soils but little data are available to illustrate the direct effect on roots in contaminated soils. This work aimed to investigate specific root traits and to assess the effect of biochar amendment on contaminant availability.

Methods

Amendment with two different types of biochar, pine woodchip and olive tree pruning, was assessed in a rhizobox experiment with maize planted in a soil contaminated with significant levels of copper and arsenic.

Results

Amendment was found to significantly improve root traits compared to the control soil, particularly root mass density and root length density. Copper uptake to plants and ammonium sulphate extractable copper was significantly less in the biochar amended soils. Arsenic uptake and extractability varied with type of biochar used but was not considered to be the limiting factor affecting root and shoot development.

Conclusions

Root establishment in contaminated soils can be enhanced by biochar amendment but choice of biochar is key to maximising soil improvement and controlling contaminant availability.  相似文献

Background and aims

Urea is the major nitrogen (N) form supplied as fertilizer in agriculture. However, urease, a nickel-dependent enzyme, allows plants to use external or internally generated urea as a nitrogen source. Since a urease inhibitor is frequently applied in conjunction with urea fertilizer, the N-metabolism of plants may be affected. The aim of this study was to determine physiological and molecular effects of nickel deficiency and a urease inhibitor on urea uptake and assimilation in oilseed rape.

Methods

Plants were grown on hydroponic solution with urea as the sole N source under three treatments: plants treated with nickel (+Ni) as a control, without nickel (?Ni) and with nickel and phenylphosphorodiamidate (+Ni+PPD). Urea transport and assimilation were investigated.

Results

The results show that Ni-deficiency or PPD supply led to reduced growth and reduced 15N-uptake from urea. This effect was more pronounced in PPD-treated plants, which accumulated high amounts of urea and ammonium. Thus, Ni-deficiency or addition of PPD, limit the availability of N and decreased shoot and root amino acid content. The up-regulation of BnDUR3 in roots indicated that this gene is a component of the stress response to nitrogen-deficiency. A general decline of glutamine synthetase (GS) activity and activation of glutamate dehydrogenase (GDH) and increases in its expression level were observed in control plants. At the same time, in (?N) or (+Ni+PPD) treated plants, no increases in GS or GDH activities and expression level were found.

Conclusions

Overall results showed that plants require Ni as a nutrient (while most widely used nutrient solutions are devoid of Ni), whether they are grown with or without a urea supply, and that urease inhibitors may have deleterious effects at least in hydroponic grown oilseed rape.  相似文献

Background and Aims

Global change will likely express itself in southwestern United States arid lands through changes in amounts and timing of precipitation in response to elevated CO2 concentrations. In addition, increased nitrogen (N) deposition may occur due to increased urban development. This study addressed the effects of water and N availability on C allocation in arid land soil-plant systems.

Methods

Columns filled with Mojave Desert topsoil containing Larrea tridentata seedlings with two treatment levels each of N and soil moisture were labeled by exposure to 13C-enriched CO2.

Results

Increased soil moisture increased plant biomass, total 13C uptake, 13C levels in leaves, soil organic matter, and soil respiration, decreased relative C allocation to stems but increased allocation to soil organic matter. Increased soil N availability increased N uptake but decreased C allocation to soil respiration presumably due to decreased substrate supply for microbes. There was no detectable label in carbonate C, suggesting that this pool does not significantly contribute to ecosystem C fluxes.

Conclusions

Our study indicates that increased water availability causes increased C uptake with increased C allocation to soil organic matter in Larrea tridentata-dominated communities while increased N deposition will have a minimal impact on C sequestration.  相似文献

Aims

A simulation model to demonstrate that soil water potential can regulate transpiration, by influencing leaf water potential and/or inducing root production of chemical signals that are transported to the leaves.

Methods

Signalling impacts on the relationship between soil water potential and transpiration were simulated by coupling a 3D model for water flow in soil, into and through roots (Javaux et al. 2008) with a model for xylem transport of chemicals (produced as a function of local root water potential). Stomatal conductance was regulated by simulated leaf water potential (H) and/or foliar chemical signal concentrations (C; H?+?C). Split-root experiments were simulated by varying transpiration demands and irrigation placement.

Results

While regulation of stomatal conductance by chemical transport was unstable and oscillatory, simulated transpiration over time and root water uptake from the two soil compartments were similar for both H and H?+?C regulation. Increased stomatal sensitivity more strongly decreased transpiration, and decreased threshold root water potential (below which a chemical signal is produced) delayed transpiration reduction.

Conclusions

Although simulations with H?+?C regulation qualitatively reproduced transpiration of plants exposed to partial rootzone drying (PRD), long-term effects seemed negligible. Moreover, most transpiration responses to PRD could be explained by hydraulic signalling alone.  相似文献

Background

Water and nutritional restrictions are limiting factors for the growth of Eucalyptus trees in tropical climates. In the dry season, boron (B) uptake is severely affected.

Aims

The objectives of this study were to evaluate the phloem mobility of B and whether its deficiency can increase plant sensitivity to osmotic stress. It was also tested to what extent foliar application of B could mitigate the negative effects of drought under low B supply.

Methods

Seedlings of a drought tolerant Eucalyptus urophylla (Blake, S. T.) clone were grown in nutrient solution, subjected to low availability of B for 25 days, and then submitted to a progressive osmotic stress. After imposition of osmotic stress, B was applied to young or mature leaves.

Results

B applications, mainly to mature leaf, stimulated root growth and delayed dehydration under osmotic stress and led to an increased B translocation and carbon isotopic composition. The expression of B transporters and pectin metabolism genes were also increased in water-stressed plants supplied with B by foliar application.

Conclusions

B deficiency led to increased plant dehydration and decreased root growth under osmotic stress. The application of B to mature leaf of water-stressed plants proved effective in mitigating the negative effects of water deficit in root growth.  相似文献

Background and aims

Nutrient acquisition of forest stands is controlled by soil resource availability and belowground production, but tree species are rarely compared in this regard. Here, we examine ecological and management implications of nitrogen (N) dynamics during early forest stand development in productive commercial tree species with narrow (Populus deltoides Bartr. and Platanus occidentalis L.) and broad (Liquidambar styraciflua L. and Pinus taeda L.) site requirements while grown with a range of nutrient and water resources.

Methods

We constructed N budgets by measuring N concentration ([N]) and N content (N C ) of above- and belowground perennial and ephemeral tissues, determined N uptake (N UP ), and calculated N use efficiency (NUE).

Results

Forest stands regulated [N] within species-specific operating ranges without clear temporal or treatment patterns, thus demonstrating equilibrium between tissue [N] and biomass accumulation. Forest stand N C and N UP increased with stand development and paralleled treatment patterns of biomass accumulation, suggesting productivity is tightly linked to N UP . Inclusion of above- and belowground ephemeral tissue turnover in N UP calculations demonstrated that maximum N demand for narrow-sites adapted species exceeded 200?kg?N ha?1?year?1 while demand for broad-site adapted species was below this level. NUE was species dependent but not consistently influenced by N availability, suggesting relationships between NUE and resource availability were species dependent.

Conclusions

Based on early stand development, species with broad site adaptability are favored for woody cropping systems because they maintain high above- and belowground productivity with minimal fertilization requirements due to higher NUE than narrow site adapted species.  相似文献

Aims

In rice, seminal root elongation plays an important role in acquisition of nutrients such as N and P, but the extent to which different N forms and P concentrations affect root growth is poorly understood. This study aimed to examine N- and P-mediated seminal root elongation response and to identify putative QTLs associated with seminal root elongation.

Methods

Seminal root elongation was evaluated in 15 diverse wild and cultivated accessions of rice, along with 48 chromosome segment substitution lines (CSSLs) derived from a cross between the rice variety ‘Curinga’ and Oryza rufipogon (IRGC 105491). Root elongation in response to different forms of N (NH4 +, NO3 ? and NH4NO3) and concentrations of P was evaluated under hydroponic conditions, and associated putative QTL regions were identified.

Results

The CSSL parents had contrasting root responses to N and P. Root elongation in O. rufipogon was insensitive to N source and concentration, whereas Curinga was responsive. In contrast to N, seminal root elongation and P concentration was positively correlated. Three putative QTLs for seminal root elongation in response to N were detected on chromosome 1, and one QTL on chromosome 3 was associated with low P concentration.

Conclusions

Genetic variation in seminal root elongation and plasticity of nutrient response may be appropriate targets for marker-assisted selection to improve rice nutrient acquisition efficiency.  相似文献

Background and aims

Roots and mycorrhizas play an important role in not only plant nutrient acquisition, but also ecosystem nutrient cycling.

Methods

A field experiment was undertaken in which the role of arbuscular mycorrhizas (AM) in the growth and nutrient acquisition of tomato plants was investigated. A mycorrhiza defective mutant of tomato (Solanum lycopersicum L.) (named rmc) and its mycorrhizal wild type progenitor (named 76R) were used to control for the formation of AM. The role of roots and AM in soil N cycling was studied by injecting a 15N-labelled nitrate solution into surface soil at different distances from the 76R and rmc genotypes of tomato, or in plant free soil. The impacts of mycorrhizal and non-mycorrhizal root systems on soil greenhouse gas (CO2 and 14+15N2O and 15N2O) emissions, relative to root free soils, were also studied.

Results

The formation of AM significantly enhanced plant growth and nutrient acquisition, including interception of recently applied NO 3 ? . Whereas roots caused a small but significant decrease in 15N2O emissions from soils at 23?h after labeling, compared to the root-free treatment, arbuscular mycorrhizal fungi (AMF) had little effect on N2O emissions. In contrast soil CO2 emissions were higher in plots containing mycorrhizal root systems, where root biomass was also greater.

Conclusions
