Arbuscular mycorrhizal (AM) symbiosis is known to stimulate plant drought tolerance. However, the mechanisms underlying the synergistic responses of the symbiotic partners to drought stress are largely unknown. A split-root experiment was designed to investigate the molecular interactions between a host plant and an AM fungus (AMF) under drought stress. In the two-compartment cultivation system, an entire or only a half root system of a maize plant was inoculated with an AMF, Rhizophagus intraradices, in the presence of localized or systemic drought treatment. Plant physiological parameters including growth, water status, and phosphorus concentration, and the expression of drought tolerance-related genes in both roots and R. intraradices were recorded. Although mycorrhizal inoculation in either one or both compartments systemically decreased abscisic acid (ABA) content in the whole root system subjected to systemic or local drought stress, we observed local and/or systemic AM effects on root physiological traits and the expression of functional genes in both roots and R. intraradices. Interestingly, the simultaneous increase in the expression of plant genes encoding D-myo-inositol-3-phosphate synthase (IPS) and 14-3-3-like protein GF14 (14-3GF), which were responsible for ABA signal transduction, was found to be involved in the activation of 14-3-3 protein and aquaporins (GintAQPF1 and GintAQPF2) in R. intraradices. These findings suggest that coexpression of IPS and 14-3GF is responsible for the crosstalk between maize and R. intraradices under drought stress, and potentially induces the synergistic actions of the symbiotic partners in enhancing plant drought tolerance.  相似文献

Arbuscular mycorrhizal (AM) symbiosis can improve date palm growth and alleviate drought-related impacts than non-mycorrhizal plants due to the ability of AMF for modifying plant metabolism and physiology.

Abstract

Date palm (Phoenix dactylifera L.) is an important agricultural and commercial crop in the North of Africa and Middle Eastern countries. During the last decade, date palm plantations were subjected to degradation due to an extensive exploitation and to drastic environmental conditions such as drought. Currently, there is a growing interest in the valorization of water due to environmental problems and economic aspects. The use of arbuscular mycorrhizal fungi (AMF) can offer a possibility to overcome these problems. The objective of this study was to study the influence of different Glomus species—Glomus intraradices, G. mosseae and Complex Aoufous (native AMF)—on the development of date palm grown under two water regimes (optimal irrigation, 75 % of field capacity or water deficit, 25 % of field capacity). Our results revealed that the beneficial effect of mycorrhizal symbiosis on plant growth depended on the fungal species and the water regime applied to the palm date seedling. While the native Complex Aoufous was the most effective in increasing the shoot height and biomass under well-watered conditions, G. intraradices was the most beneficial fungus for improving growth of plants that undergo restricted water supply. This positive effect of G. intraradices under drought conditions was not related to an enhancement of the antioxidant enzymatic activities in leaves; the association of palm date with G. intradices caused an increase in the elasticity of cell walls in leaves and allowed maintaining high water content in leaves without lowering leaf water potential under stressful conditions. The adequate selection of the AMF species is crucial for improving growth of palm date seedlings, and it must be in accordance with the water regime that will be applied to plants.  相似文献

The symbiosis of plants with arbuscular mycorrhizal fungi (AMF) may become parasitic if the cost:benefit ratio (carbon:phosphorus ratio) increases. In case of mycorrhizal parasitism, a plant may prevent growth depression through the reduction of root colonization as a form of control over the symbiosis. In this greenhouse study, we attempted to manipulate the cost:benefit ratio of the arbuscular mycorrhizal symbiosis by shading and/or phosphorus (P) fertilization in the differentially mycotrophic plant species Hieracium pilosella and Corynephorus canescens. By repeated sampling of soil cores, we assessed the temporal progress of plant investment towards mycorrhizal structures as a measure of plant control over the AMF. Unexpectedly, we found no obvious treatment effects on mycorrhizal growth dependency (MGD), most likely caused by constant N-limitation in AM plants being enhanced by P-fertilization and shade probably not exacerbating plant C-budget for AMF. This highlights the importance of N:P:C stoichiometry for the outcome of the symbiosis. Nevertheless, we found possible control mechanisms in shaded H. pilosella, with considerably higher resource investments into root than into hyphal growth, while root colonization was only marginally suppressed. This control only manifested after 4 weeks of growth under potentially detrimental conditions, emphasizing the importance of time in plant control over the arbuscular mycorrhizal symbiosis. In contrast, the less mycotrophic C. canescens did not exhibit obvious changes in mycorrhizal investments in reaction to shading and P-fertilization, possibly because the low mycotrophy and AMF colonization already imposes a functioning control mechanism in this species. Our study suggests that highly mycotrophic plants may have a stronger need to keep AMF in check than less mycotrophic plants, which may have implications for the role of mycotrophy in the outcome of symbiotic interactions in natural situations.  相似文献

Aims

The study aims (1) to evaluate the effect of Mesorhizobium tianshanense on plant proline and polyamine levels of Lotus tenuis and its modulatory effect during plant response to short-term salt stress and (2) to compare these effects with those caused by mycorrhizal symbiosis.

Methods

Experiments consisted of a randomized factorial design of two factors: salinity (two levels, 0 and 150 mM NaCl) and symbiosis (three levels, uninoculated, Glomus intraradices, and M. tianshanense).

Results

Salinization led to increased proline levels regardless of plant organ and symbiotic status, excepting mycorrhizal L. tenuis roots. Salinity diminished the total polyamine level of control and rhizobial plants but not in mycorrhizal ones. Variations in the pattern response of the three individual polyamines (putrescine, spermidine, and spermine) differed in accordance with the symbiotic status of the plant, highlighting a divergence on proline and polyamine metabolisms between rhizobial and mycorrhizal symbiosis.

Conclusions

Spermidine and spermine contributed the most with the salt-induced root polyamine increment observed upon salinization in roots of nodulated plants, suggesting that these polyamines might mediate an adaptive role of the plant–M. tianshanense symbiosis in L. tenuis plants growing in a saline environment.  相似文献

Background and aims

The biological restoration of saline habitats could be achieved by using halophyte plant species together with adapted arbuscular mycorrhizal fungi (AMF). An interesting plant to be used in restoration of saline environments, Asteriscus maritimus, is highly mycotrophic. The aim of this study was to assess the effectiveness of native and allochthonous AMF to enhance the establishment and growth of the halophyte A. maritimus under saline conditions.

Methods

We studied the symbiotic effectiveness of four AMF strains (three native fungal isolates from a saline soil and one allochthonous, from collection) in A. maritimus subjected to increasing salinity stress. We measured plant physiological parameters by which AMF may ameliorate the detrimental effects of salinity stress.

Results

A. maritimus plants showed a high mycorrhizal dependency, even in absence of salt stress. Plants inoculated with native AMF had higher shoot dry weight, efficiency of photosystem II, stomatal conductance and accumulation of glutathione than those inoculated with the collection AMF at the highest level of salinity. Moreover, at this salt level, only 30 % of A. maritimus plants inoculated with the collection AMF survived, while with the three native AMF, the rate of survival was 100 %.

Conclusions

Results points out the importance of native AMF inoculation in the establishment, survival and growth of A. maritimus plants. Inoculation with these native AMF enhanced A. maritimus salt tolerance by increasing efficiency of photosystem II, stomatal conductance and glutathione content and by reducing oxidative damage. Thus, the use of adequate native AMF inocula could be a critical issue for success in recovering saline degraded areas.  相似文献

Background and Aims

Water solubility of zinc (Zn) fertilisers affects their plant availability. Further, simultaneous application of Zn and phosphorus (P) fertiliser can have antagonistic effects on plant Zn uptake. Arbuscular mycorrhizas (AM) can improve plant Zn and P uptake. We conducted a glasshouse experiment to test the effect of different Zn fertiliser materials, in conjunction with P fertiliser application, and colonisation by AM, on plant nutrition and biomass.

Methods

We grew a mycorrhiza-defective tomato genotype (rmc) and its mycorrhizal wild-type progenitor (76R) in soil with six different Zn fertilisers ranging in water solubility (Zn sulphate, Zn oxide, Zn oxide (nano), Zn phosphate, Zn carbonate, Zn phosphate carbonate), and supplemental P. We measured plant biomass, Zn and P contents, mycorrhizal colonisation and water use efficiency.

Results

Whereas water solubility of the Zn fertilisers was not correlated with plant biomass or Zn uptake, plant Zn and P contents differed among Zn fertiliser treatments. Plant Zn and P uptake was enhanced when supplied as Zn phosphate carbonate. Mycorrhizal plants took up more P than non-mycorrhizal plants; the reverse was true for Zn.

Conclusions

Zinc fertiliser composition and AM have a profound effect on plant Zn and P uptake.  相似文献

Aims

The most common metric of arbuscular mycorrhizal fungal (AMF) abundance is percent root length colonized (PRLC) by mycorrhizal structures. Frequently, plants with greater PRLC are assumed to receive more nutrients (such as phosphorus, P) from their mycorrhizal symbionts, leading to greater plant growth. Nevertheless, the functional significance of this metric remains controversial. In this review, I discuss whether manipulations of PRLC generally led to changes in plant biomass and P content, and whether AMF taxa and plant functional groups influence these relationships.

Methods

I conducted a meta-analysis of laboratory- and field-based trials in which mycorrhizal colonization was directly altered compared to unmanipulated controls. For each trial, I calculated (1) the difference in PRLC (ΔPRLC) between the treatments, and (2) the response ratio of plant biomass. In a subset of these studies, the response ratio of P content of host plants could also be calculated.

Results

The response ratio of plant biomass and P content rose significantly and exponentially as ΔPRLC increased. Nevertheless, ΔPRLC explained only a fraction of the variation in response ratios in each case. Moreover, AMF taxa varied in their effects on biomass per unit ΔPRLC. In addition, plant functional groups differed in effects on plant P content per unit ΔPRLC, with C4 grasses responding most strongly.

Conclusions

It appears that as the extent to which plant roots are colonized by AMF increases, plant growth and P content often increase, although substantial variability exists among trials. As others have found, a likely mechanism for this relationship is increased transfer of P (and perhaps other nutrients) through the more-prevalent mycorrhizal structures.  相似文献

The ascomycetous dark septate endophytic (DSE) fungi characterized by their melanized hyphae can confer abiotic stress tolerance in their associated plants in addition to improving plant growth and health. In this study inoculation of the DSE fungus Nectria haematococca Berk. & Broome significantly improved all the plant growth parameters like the plant height, stem girth, leaf characteristics and plant biomass of drought-stressed tomato. Root characters like the total root length, primary root diameter, 2nd order root number and diameter, root hair number and length were also significantly influenced by the fungal inoculation. Nevertheless, N. haematococca inoculation did not affect root colonization by native arbuscular mycorrhizal (AM) fungi and no significant correlation existed between the AM and DSE fungal variables examined. The proline accumulation in shoots of N. haematococca inoculated plants was significantly higher than uninoculated plants. The present study clearly indicates for the first time the ability of the DSE fungus, N. haematococca in inducing the drought stress tolerance and promoting the growth of the host plant under water stress.  相似文献

The objective of this study was to investigate the effects of arbuscular mycorrhizal fungus (AMF) inoculation on plant growth and drought tolerance in seedlings of a promising oilseed crop, Sacha Inchi (Plukenetia volubilis L.), under well-watered or drought conditions. AMF inoculation was applied in four treatments: without AMF inoculation, Glomus versiforme, Paraglomus occultum, or combination of both microorganism inoculations. The results showed that AMF colonization significantly enhanced the growth of Sacha Inchi seedlings regardless of soil water conditions, and the greatest development was reached in plants dually inoculated under well-watered conditions. G. versiforme was more efficient than P. occultum. Plants inoculated with both symbionts had significantly greater specific leaf area, leaf area ratio and root volume when compared with the uninoculated control, G. versiforme, and P. occultum treatments alone, indicating a synergistic effect in the two AMF inoculation. Photosynthetic rate and water-use efficiency were stimulated by AMF, but not stomatal conductance. Inoculation with AM fungus increased antioxidant enzymes activities including guaiacol peroxidase and catalase, thus lowering hydrogen peroxide accumulation and oxidative damage, especially under drought stress conditions. However, proline content showed little change during drought stress and AMF colonization conditions, which suggested that proline accumulation might not serve as the main compound for osmotic adjustment of the studied species. These results indicate that AMF inoculation stimulated growth and enhanced drought tolerance of Sacha Inchi seedlings, through alterations in morphological, physiological and biochemical traits. This microbial symbiosis might be an effective cultivation practice in improving the performance and development for Sacha Inchi plants.  相似文献

Aims

Nutrients play a key role in arbuscular mycorrhizal (AM) symbiosis. We quantified the response of AM symbiosis of seedlings and adult plants of Plantago lanceolata to fertilization under field conditions in managed grasslands differing in nutrient availability and soil moisture.

Methods

The AM symbiosis was measured as the total extent of AM fungal colonization and frequency of arbuscules or vesicles, and as the relative proportions of morphotypes. We further examined the effects of the surrounding vegetation upon AM symbiosis.

Results

Fertilization decreased total AM colonization and relative arbuscular frequency of the whole mycorrhizal community and of Acaulospora and “fine endophyte” morphotypes in seedling roots, but it had no effect upon the mycorrhiza in adult plants. The decline in arbuscular frequency in seedling roots due to fertilization was greater at the sites with higher nutrient availability and lower N:P ratio. Seedlings surrounded by more forbs had a greater total AM colonization and higher vesicular frequency.

Conclusions

Increased nutrient availability in the initial stages of seedling development has a prominent effect upon AM symbiosis development, but these effects seem to diminish over the long term, as evidenced by the results obtained for adult plants and from the limited effects of parameters characterizing long-term nutrient availability.  相似文献

Background and aims

Year of release of a cultivar reflects the agricultural and breeding practices of its time; we hypothesize that there are differences in mycorrhizal responsiveness of new high yielding and old crop plants and landraces. We evaluated the importance of the year of release on mycorrhizal responsiveness, arbuscular mycorrhizal (AM) fungal root colonization and P efficiency. We also analyzed the effect of experimental treatments, P efficiency (P acquisition and P utilization efficiency) and AM fungal root colonization on a potential mycorrhizal responsiveness trend for year of release.

Methods

We conducted a meta-analysis on 39 publications working on 320 different crop plant genotypes.

Results

New cultivars were less intensely colonized but were more mycorrhiza-responsive (and possibly dependent) compared to ancestral genotypes. This trend was potentially influenced by the moderator variables density, pre-germination, plant, plant type and AMF species. AM root colonization was also important for the mycorrhizal responsiveness trend for year of release, but P efficiency was not.

Conclusions

With the data available we could find no evidence that new crop plant genotypes lost their ability to respond to mycorrhiza due to agricultural and breeding practices.  相似文献

Aims and Background

Many plants preferentially grow roots into P-enriched soil patches, but little is known about how the presence of arbuscular mycorrhizal fungi (AMF) affects this response.

Methods

Lotus japonicus (L.) was grown in a low-P soil with (a) no additional P, (b) homogeneous P (28 mg pot?1), (c) low heterogeneous P (9.3 mg pot?1), and (d) high heterogeneous P (28 mg pot?1). Each P treatment was combined with one of three mycorrhiza treatments: no mycorrhizae, Glomus intraradices, indigenous AMF. Real-time PCR was used to assess the abundance of G. intraradices and the indigeneous AMF G. mosseae and G. claroideum.

Results

Mycorrhization and P fertilization strongly increased plant growth. Homogeneous P supply enhanced growth in both mycorrhizal treatments, while heterogeneous P fertilization increased biomass production only in treatments with indigenous AMF inoculation. Preferential root allocation into P-enriched soil was significant only in absence of AMF. The abundance of AMF species was similar in P-enriched and unfertilized soil patches.

Conclusion

Mycorrhization may completely override preferential root growth responses of plants to P- patchiness in soil. The advantage of this effect for the plants is to give roots more freedom to forage for other resources in demand for growth and to adapt to variable soil conditions.  相似文献

Aims

This study aimed to determine the effect of arbuscular mycorrhizal (AM) fungi and phosphorus (P) supply levels on β-carotene concentrations in sweet potato (Ipomoea batatas L.) tubers.

Methods

Two commercial AM fungal isolates of Glomus intraradices (IFP Glintra) and Glomus mosseae (IFP Glm) which differ in their life cycles were used. Sweet potato plants were grown in a horizontal split-root system that consisted of two root compartments. A root-free fungal compartment that allowed the quantification of mycelial development was inserted into each root compartment. The two root compartments were inoculated either with the same or with different AM isolates, or remained free of mycorrhizal propagules. Each fungal treatment was carried out in two P supply levels.

Results

In the low P supply level, mycorrhizal colonization significantly increased β-carotene concentrations in sweet potato tubers compared with the non-mycorrhizal plants. Glomus intraradices appeared to be more efficient in increasing β-carotene concentrations than G. mosseae. Dual inoculation of the root system with the two mycorrhizal fungi did not result in a higher increase in tuber β-carotene concentrations than inoculation with the single isolates. Improved P nutrition led to higher plant tuber biomass but was not associated with increased β-carotene concentrations.

Conclusions

The results indicate a remarkable potential of mycorrhizal fungi to improve β-carotene concentrations in sweet potato tubers in low P fertilized soils. These results also suggest that β-carotene metabolism in sweet potato tubers might be specifically activated by root mycorrhizal colonization.  相似文献

Background and Aim

Climate change models are limited by lack of baseline data, in particular carbon (C) allocation to – and dynamics within – soil microbial communities. We quantified seasonal C-assimilation and allocation by plants, and assessed how well this corresponds with intraradical arbuscular mycorrhizal fungal (AMF) storage and structural lipids (16:1ω5 NLFA and PLFA, respectively), as well as microscopic assessments of AMF root colonization.

Methods

Coastal Hypochoeris radicata plants were labeled with 13CO2 in February, July and October, and 13C-allocation to fine roots and NLFA 16:1ω5, as well as overall lipid contents and AM colonization were quantified.

Results

C-allocation to fine roots and AMF storage lipids differed seasonally and mirrored plant C-assimilation, whereas AMF structural lipids and AM colonization showed no seasonal variation, and root colonization exceeded 80 % throughout the year. Molecular analyzes of the large subunit rDNA gene indicated no seasonal AMF community shifts.

Conclusions

Plants allocated C to AMF even at temperatures close to freezing, and fungal structures persisted in roots during times of low C-allocation. The lack of seasonal differences in PLFA and AM colonization indicates that NLFA analyses should be used to estimate fungal C-status. The implication of our findings for AM function is discussed.  相似文献

Background and aims

Roots and mycorrhizas play an important role in not only plant nutrient acquisition, but also ecosystem nutrient cycling.

Methods

A field experiment was undertaken in which the role of arbuscular mycorrhizas (AM) in the growth and nutrient acquisition of tomato plants was investigated. A mycorrhiza defective mutant of tomato (Solanum lycopersicum L.) (named rmc) and its mycorrhizal wild type progenitor (named 76R) were used to control for the formation of AM. The role of roots and AM in soil N cycling was studied by injecting a 15N-labelled nitrate solution into surface soil at different distances from the 76R and rmc genotypes of tomato, or in plant free soil. The impacts of mycorrhizal and non-mycorrhizal root systems on soil greenhouse gas (CO2 and 14+15N2O and 15N2O) emissions, relative to root free soils, were also studied.

Results

The formation of AM significantly enhanced plant growth and nutrient acquisition, including interception of recently applied NO 3 ? . Whereas roots caused a small but significant decrease in 15N2O emissions from soils at 23?h after labeling, compared to the root-free treatment, arbuscular mycorrhizal fungi (AMF) had little effect on N2O emissions. In contrast soil CO2 emissions were higher in plots containing mycorrhizal root systems, where root biomass was also greater.

Conclusions

Taken together, these data indicate that roots and AMF have an important role to play in plant nutrient acquisition and ecosystem N cycling.  相似文献

The influence of arbuscular mycorrhiza (AM) and drought stress on aquaporin (AQP) gene expression, water status, and photosynthesis was investigated in black locust (Robinia pseudoacacia L.). Seedlings were grown in potted soil inoculated without or with the AM fungus Rhizophagus irregularis, under well-watered and drought stress conditions. Six full-length AQP complementary DNAs (cDNAs) were isolated from Robinia pseudoacacia, named RpTIP1;1, RpTIP1;3, RpTIP2;1, RpPIP1;1, RpPIP1;3, and RpPIP2;1. A phylogenetic analysis of deduced amino acid sequences demonstrated that putative proteins coded by these RpAQP genes belong to the water channel protein family. Expression analysis revealed higher RpPIP expression in roots while RpTIP expression was higher in leaves, except for RpTIP1;3. AM symbiosis regulated host plant AQPs, and the expression of RpAQP genes in mycorrhizal plants depended on soil water condition and plant tissue. Positive effects were observed for plant physiological parameters in AM plants, which had higher dry mass and lower water saturation deficit and electrolyte leakage than non-AM plants. Rhizophagus irregularis inoculation also slightly increased leaf net photosynthetic rate and stomatal conductance under well-watered and drought stress conditions. These findings suggest that AM symbiosis can enhance the drought tolerance in Robinia pseudoacacia plants by regulating the expression of RpAQP genes, and by improving plant biomass, tissue water status, and leaf photosynthesis in host seedlings.  相似文献

Background and aims

Recent studies have shown that tree-based intercropping (TBI) systems support a more diverse soil microbial community compared to conventional agricultural systems. However, it is unclear whether differences in soil microbial diversity between these two agricultural systems have a functional effect on crop growth.

Methods

In this study, we used a series of greenhouse experiments to test whether crops respond differently to the total soil microbial community (Experiment 1) and to arbuscular mycorrhizal (AM) fungal communities alone (Experiment 2) from conventionally monocropped (CM) and TBI systems.

Results

The crops had a similar growth response to the total soil microbial communities from both cropping systems. However, when compared to sterilized controls, barley (Hordeum vulgare) and canola (Brassica napus) exhibited a negative growth response to the total soil microbial communities, while soybean (Glycine max) was unaffected. During the AM fungal establishment phase of the second experiment, ‘nurse’ plants had a strong positive growth response to AM fungal inoculation, and significantly higher biomass when inoculated with AM fungi from the CM system compared to the TBI system. Soybean was the only crop species to exhibit a significant positive growth response to AM fungal inoculation. Similar to the total soil microbial communities, AM fungi from the two cropping systems did not differ in their effect on crop growth.

Conclusion

Overall, AM fungi from both cropping systems had a positive effect on the growth of plants that formed a functional symbiosis. However, the results from these experiments suggest that negative effects of non-AM fungal microbes are stronger than the beneficial effects of AM fungi from these cropping systems.  相似文献

The objective of this study was to investigate the effects of arbuscular mycorrhizal fungus (AMF) inoculation on growth and drought tolerance of Poncirus trifoliata seedlings. The seedlings were inoculated with or without Glomus mosseae before exposure to a short-term (3 days) water depletion, and relevant physiological and biochemical parameters (plant height, chlorophyll content, relative water content, activity of antioxidant enzymes) and expression patterns of several stress-responsive genes were examined. Inoculation with G. mosseae led to growth promotion of the seedlings, as revealed by larger plant height and higher relative water and chlorophyll contents. When subjected to drought treatment, the AMF-inoculated (AM) plants showed better tolerance than the nonmycorrhizal (NAM) plants. Under drought, the AM plants exhibited higher level of proline and activity of two antioxidant enzymes, superoxide dismutase (SOD) and peroxidase (POD). In addition, mRNA abundance of four genes involved in reactive oxygen species homeostasis and oxidative stress battling was higher in the AM plants when compared with the NAM plants. These results indicate that AMF inoculation stimulated growth and enhanced drought tolerance of the seedlings, which may be due to activation of an arsenal of physiological, biochemical and molecular alterations.  相似文献
