Capture of carp Cyprinus carpio from holding tanks by dipnet, or from semi-natural conditions by rod and line, elicits a physiological stress response characterized by elevation of plasma cortisol levels. The transfer of carp to keepnets subsequent to capture does not increase or reduce the magnitude or duration of this response and in both cases plasma cortisol levels have returned to pre-stress levels within 24 h of the initial disturbance. The postcapture plasma cortisol elevation is accompanied by disturbances in plasma glucose and lactate levels but these are less consistent in severity and duration than the cortisol response. These data suggest that the retention of fish in keepnets following capture, does not represent a source of stress additional to that imposed by capture and has no effect on the rate of recovery of the fish from the initial capture stress.  相似文献

Tropical labrids Hemigymnus melapterus sampled underwater had low plasma levels of cortisol, glucose, and lactate. Plasma cortisol levels were elevated by capture stress within 5–6 min, while glucose and lactate levels were not. Plasmalevels of cortisol and glucose increased after 2–4 h of handling and transport to the laboratory. Levels of cortisol and glucose fell with laboratory acclimation back to values similar to those found in wild fish. Parasitism by gnathiid isopods across an order of magnitude of isopod numbers had no effect on plasma levels of cortisol or glucose. Thus, H. melapterus has a stress response similar to that shown by temperate species, and relatively high parasite loads are not apparently stressful to fish in the wild. This may be related to the counterproductive effects of physiological stress responses on the immune system or behaviour-modulated processes that counter parasitic invasion.  相似文献

The effects of confinement and exercise on the stress response of the spiny damselfish Acanthochromis polyacanthus were investigated in a laboratory stock of fish. Cultured spiny damselfish had basal plasma cortisol values (<16 ng ml−1) similar to those found in wild fish, and basal plasma glucose and lactate levels that were similar to those found in other teleosts. Plasma cortisol concentrations increased in response to stress with a latency period of 5–10 min. Removal of the stressor resulted in partial recovery of cortisol levels by 24 h. Plasma glucose levels increased in response to stress in all experiments with significant increases occurring within 15 min of the imposition of stress. Elevations in plasma glucose concentrations were not initially reflected in changes in liver or muscle glycogen content, with significant reductions in liver glycogen concentrations only occurring in response to extended periods of stress. In contrast to many temperate species, plasma lactate concentrations did not consistently increase in response to stress, suggesting that the stress response in spiny damselfish is not strongly characterized by anaerobiosis.  相似文献

To simulate swimming in a trawl, age 3 year brown trout Salmo trutta (L.) were made to swim against a flow of 0·5 m s−1 for 60 min. To simulate cold shock, similar to placing them in a chilling tank, the fish were kept for 10 min in a tank containing ice and water. To simulate the combined stressors, the fish were first made to swim followed by a cold shock. The fish were in a comatose state 10 min after cold shock and combined stressors but conscious after swimming only. All the fish survived until the end of the studied recovery period (maximum 24 h). Cold shock after swimming (combined stressors v . swimming only) did not produce higher blood cortisol, lactate or glucose concentrations 10 min after the treatment. The effect of cold shock, however, was evident in the delayed start of recovery in cortisol and glucose concentrations. All the stress indicators used decreased to the levels for undisturbed fish within 24 h, except in the case of glucose after the combined stressors.  相似文献

A rapid increase in common carp Cyprinus carpio plasma cortisol levels was noted, in two experiments, after 30 mins of a 3 h net confinement, which was sustained while the fish were held in the nets. After release from the nets, cortisol levels returned to control values in 1 h. Plasma glucose and free fatty acid levels were elevated by the confinement. Glucose was increased after 30 min but returned to basal levels after 22 h of recovery while free fatty acids were not elevated until 3 h of confinement and remained high for the duration of the recovery period. After confinement for 3 h, plasma lactate levels were reduced and remained low for a further 1 h. No change in either plasma triglyceride or cholesterol levels were found during the study. Confinement had no effect on haematocrit levels but blood haemoglobin levels were reduced. In both experiments hypochloraemia occurred in response to confinement. However, values returned to pre-confinement levels 22 h after confinement. These results show that rearing isogenic carp strains, under identical conditions, results in a reproducable response to an acute stressor and that these carp respond in a similar manner to other teleost species.  相似文献

Sexually immature rainbow trout were acclimated to small-volume (1 m3) holding tanks and then exposed to short-term stress to examine the relationship between feeding, stress, plasma ghrelin levels and other plasma stress parameters. Plasma ghrelin levels showed an increase 24 h after a single feed, plasma lactate and glucose levels decreased over the same period and plasma cortisol levels were low and constant. One hour of confinement stress resulted in elevations of plasma cortisol, glucose and lactate and depression of plasma ghrelin levels. In a separate experiment, 2 h of confinement stress also depressed feeding immediately after stress, concomitant with increases in plasma cortisol, lactate and glucose; however, in this case there was no change in plasma ghrelin concentrations. A repeat of the 2-h confinement experiment using fish that had not been acclimated to small-volume holding tanks produced a more marked elevation in plasma cortisol and a stronger suppression of feeding post-stress but in this case also, there was no change in plasma ghrelin levels. The results of this study confirm that feeding in rainbow trout is suppressed by confinement stress although the effect is transitory in this domesticated stock. Similar to that in other fishes, plasma ghrelin levels appear to be modulated by feeding status and may be influenced by stress, suggesting an orexigenic role for ghrelin in rainbow trout.  相似文献

The response of six species of freshwater fishes, from the families Cyprinidae (common carp Cyprinus carpio, roach Rutilus rutilus and chub Leuciscus cephalus) and Salmonidae (rainbow trout Oncorhynchus mykiss, brown trout Salmo trutta and Arctic charr Salvelinus alpinus), to a standardized stressor was evaluated. A 6 h period of confinement resulted in changes to plasma cortisol, glucose, amino acid and lactate levels compared with unconfined controls. There were significant differences in the response profiles both within and between families. The cyprinid species exhibited higher and more sustained stress‐induced increases in plasma cortisol and glucose than the salmonid species. In cyprinids, plasma lactate and plasma amino acid concentration showed less disturbance following stress than in salmonids. The results of the study, together with an evaluation of previously published data for eight salmonid species and six cyprinid species, support the hypothesis that differences in core elements of the stress response exist between species of fishes, and that this variation may have a systematic basis.  相似文献

The present study investigated the interaction of feeding times with two dietary fat levels on physiological responses to handling stress in juvenile Beluga sturgeon. Fish were fed with two different diets (high energy; HE: 24% fat and low energy; LE: 12% fat) for 8 weeks at two feeding times; 09:00 and 16:00 (during the day) or 21:00 and 04:00 (during the night). At the end of the trial, blood samples were taken and the resting fish were held in a net out of water for 90 s as the handling stressor. Three hours after application of stress, post‐stress blood was taken. Cortisol, glucose, and lactate concentrations were considered as stress indicators. The mean values of cortisol and lactate levels did not reveal a significant difference between pre‐ and 3‐hr post‐stress samples, but the average concentration of glucose showed a significant difference. Cortisol, lactate and glucose concentrations were not influenced by the difference in the diets. Moreover, the cortisol and lactate concentrations were not affected by the different feeding times, while glucose levels were significantly affected by the feeding times with the lowest level in fish fed during the day. With respect to stress indicators, the results revealed that feeding times affected pre‐ and post‐stress secondary response to handling stress, but the rate and magnitude of metabolites (cortisol, glucose and lactate) were not affected by dietary fat levels. Therefore, it is necessary to examine the best feeding times with the interaction of feed ingredients in sturgeon farming.  相似文献

Cadmium is a non-essential metal with a wide distribution that has severe toxic effects on aquatic animals. Changes in hematology and muscle physiology were examined in silver carp (Hypophthalmichthys molitrix) exposed to environmentally relevant levels of cadmium (0.01 mg L?1) for 96 h. Cadmium exposure induced significant increases in the red blood cell count, and in the plasma concentrations of cortisol, glucose, and lactate. This suggests that the dose of cadmium was sufficient to cause stress, possibly associated with impaired gas exchange at the gills. There were no changes in hemoglobin concentration or plasma protein concentration. Significant decreases in muscle energy fuels (ATP and glycogen), and increases in muscle lactate persisted until the end of the exposure period, respectively. The changes in muscle lactate and protein in silver carp differed from those observed in response to exposure of fish to cadmium and heavy metals in other studies. The study highlights the importance of selecting unpolluted release sites with suitable water conditions for the survival of newly released individuals for stock enhancement of the Xiangjiang River.  相似文献

We measured changes in free and total plasma cortisol levels, plasma glucose, gill hsp70 levels, and growth in haddock (Melanogrammus aeglefinus) subjected to a long-term handling stress (15 s out of water, each day, for 4 weeks), and the effect of this long-term stress on the ability of haddock to respond to an acute stressor. The acute stressor was a single handling stress, and fish were sampled at 1, 6, and 12 h post-stress. During the long-term stress study, free and total plasma cortisol levels increased significantly (10-fold) in the stressed group after the second week. However, the percentage of free cortisol was already significantly elevated by the first week (control 17%, stressed 55%), and remained high during the second week (control 35% and stressed 65%). After 3 and 4 weeks of handling, both free and total cortisol declined in stressed fish to levels that were not significantly different from pre-stress values. Control fish grew significantly more than stressed fish (by 32% and 18%, respectively) over the 4 week study, and condition factor only increased in control fish. Although fish from the control group showed elevated total plasma cortisol levels (to 47 ng mL(-1)) 1 h after the acute stress, and the levels in stressed fish were comparable to those for the control fish, no significant increase in plasma cortisol was measured in the group subjected to the long-term stress. Free plasma cortisol levels did not increase significantly in either group following the acute stress. However, free plasma cortisol levels were significantly higher in long-term stress group, as compared with the control group, at 6 h post-stress. Plasma glucose and gill hsp70 levels were not altered by either the long-term stress or acute stressor. Our data indicate that cortisol (free and total), but not glucose or hsp70, appears to be adequate to assess short- and long-term stress in haddock.  相似文献

This study was carried out to investigate the effect of an acute stressor on the variation of some physiological and immunological parameters of Siberian sturgeon (Acipenser baerii) juveniles. Fish, reared in 3 tanks for 10 weeks, were used for this study. The acute stress of fish consisted of 2 min of air exposure stress. Plasma levels of cortisol, glucose, and lactate as well as lysozyme activity in plasma were measured before stress and 1 hr, 3 hr, 6 hr, 9 hr, 12 hr, and 24 hr after stress. The plasma cortisol significantly increased in the highest level 1 hr after stress, yet it gradually declined after 3 hr. The glucose significantly increased only 1 hr after stress. There was no significant difference between plasma lactate prestress and poststress. Moreover, lysozyme activity was enhanced by stress, thus reaching the highest level 9 hr after stress. The results of this study indicate that Siberian sturgeon not only have a rapid response to acute stress, but also a great capacity for recovery from stress, thus returning physiological parameters to prestress levels after 6 hr.  相似文献

Eurasian perch (Perca fluviatilis) is a promising aquaculture candidate, but the growth performance of this non-domesticated species may be negatively affected by its stress responsiveness to intensive culture conditions. To evaluate this potential problem, juvenile Eurasian perch were exposed to a standardized handling stressor twice a week for an 8-week period. A similar study was conducted on domesticated rainbow trout (Oncorhynchus mykiss) for comparison of intra- and inter-specific differences. The stressed fish of both species showed lower body growth than the non-stressed control fish, however, the final mean body mass was 35.4% lower in the stressed Eurasian perch than in the non-stressed controls, compared to 22.8% difference between the two groups in rainbow trout. The stress responsiveness was examined by comparing the post-stress cortisol and glucose levels in repeatedly stressed fish and fish exposed to the stressor only once. The cortisol stress response in both species strongly indicated a habituation to the repeated stressor. Thus, repeatedly stressed Eurasian perch reached maximum cortisol levels of 130 ng/mL after 0.5 h compared to 200 ng/mL in the fish stressed once, while considerably smaller differences in cortisol levels were shown between the repeatedly and single stressed rainbow trout. Rainbow trout also showed lower post-stress glucose levels in the repeatedly stressed fish compared to the single stressed fish. In contrast, the glucose levels in both groups of Eurasian perch increased abruptly after stress treatment and remained elevated at approximately 19 mM for 6 h; levels were three times as high as the peak levels 3 h post-stress in rainbow trout. Together, the habituation of the stress response shown in both species did not eliminate the growth difference found in the repeatedly stressed fish versus the control fish. Further, the lower growth performance of Eurasian perch compared to rainbow trout could partly be due to the increased energy consumption in the more stress responsive Eurasian perch.  相似文献

1. Plasma magnesium, cortisol, lactate and ascorbic acid were examined in common carp subjected to various dietary treatments and following handling stress. 2. Under conditions of satisfied dietary magnesium and ascorbate requirements, plasma cortisol concentration after stress increased less pronouncedly than in fish fed large doses of ascorbate and/or magnesium. 3. Plasma lactate increased significantly in all groups after stress, although the increase seemed to be more severe (detrimental) in fish on large doses of ascorbate, either as ascorbic acid (AA) or ascorbic monophosphate Mg salt (AP). 4. Large doses of dietary ascorbate, both AA and AP, resulted in a significant increase of total ascorbate concentration in kidney and hepatopancreas of carp in comparison to pre-experimental level. 5. Kidney total ascorbate concentration decreased by 10-23% in all groups but one in which fish fed diet supplemented with AA displayed a significant increase (30%) of tissue ascorbate. The opposite trend was found in hepatopancreas of AA group with 21.5% ascorbate depletion. 6. The present results suggest that plasma cortisol and kidney (steroidogenesis site) and hepatopancreas ascorbate concentration responses to stress may not be related. Our results also do not support the hypothesis of the primary role of the high concentration of ascorbate in the kidney inhibiting steroidogenesis.  相似文献

Endocrine, metabolic and osmoregulatory changes in Pacific halibut upon capture and at intervals following a 30‐min air exposure were measured. Concentrations of cortisol, sodium, and chloride in plasma and serum peaked two hours after the stressor. Thirty minutes after a 30‐min air exposure concentrations of glucose in plasma had increased significantly from levels obtained immediately after the stressor and remained elevated for up to 4 h. Plasma lactate was also elevated 30 min after the stress treatment and lactate concentrations increased significantly at each subsequent sampling interval, 2 and 4 h. There was a significant linear increase in plasma lactate for fish sampled from 1.5 to 6.5 h after capture. Incidence of delayed mortality was low; of 22 experimentally stressed animals only one died over a 10‐day monitoring period. These data illustrate the difficulties in using single time‐point plasma indices of stress to assess condition of animals after capture since the time‐course of physiological changes associated with the stress response vary with parameter measured and may take hours to be fully expressed.  相似文献

This study investigated the effect of high‐dose vitamin C supplementation on growth, tissue ascorbic acid concentrations and physiological response to transportation stress in juvenile silver pomfret, Pampus argenteus (initial average weight 6.2 ± 0.2 g). Three practical diets were formulated to contain 100 (control), 450 and 800 mg ascorbic acid/kg diet, respectively, supplied as l‐ascorbyl‐2‐polyphosphate. Each diet was fed to triplicate groups of fish in circular tanks (3 m ø, 1.5 m depth) (60 fish/tank) for 9 weeks. Growth did not change significantly with dietary vitamin C levels, although an improvement tendency with an increase in vitamin C supplementation was observed. Ascorbic acid concentrations in the liver and muscle of fish fed diets containing graded levels of vitamin C were positively correlated with dietary levels of this vitamin. Tissue ascorbic acid concentrations significantly increased with increasing vitamin C supplementation. After 9 weeks, the fish were subjected to transportation stress for 4 h to determine the influence of high vitamin C supplementation on the physiological response to this stressor. Serum cortisol, glucose and lactate levels significantly increased in stressed fish. Serum cortisol and glucose concentrations after stress were significantly higher in fish fed the control diet (7.91 μg L?1 and 0.80 mm , respectively) than in the other groups. However, there were no significant differences in serum cortisol and glucose levels after stress between the 450 and 800 mg kg?1 diets. No significant change could be found in serum lactate levels after stress among the different treatments. In conclusion, the dietary administration of high doses of vitamin C could reduce stress in silver pomfret and increase the survival of fish under stress conditions.  相似文献

Cortisol release from fish head kidney during the acute phase of the stress response is controlled by the adrenocorticotropic hormone (ACTH) from the pituitary pars distalis (PD). Alpha-melanocyte-stimulating hormone (alpha-MSH) and beta-endorphin, from the pars intermedia (PI), have been implicated in cortisol release during the chronic phase. The present study addresses the regulation of cortisol release by ACTH and alpha-MSH in common carp (Cyprinus carpio) and includes characterization of their receptors, namely, the melanocortin-2 and melanocortin-5 receptors (MC2R and MC5R). We could not demonstrate corticotropic activity of alpha-MSH, beta-endorphin, and combinations of these. We do show a corticotrope in the PI, but its identity is as yet uncertain. Carp restrained for 1 and 7 days showed elevated plasma cortisol and alpha-MSH levels; cortisol is still elevated but lower at day 7 than day 1 of restraint. Interrenal response capacity is unaffected, as estimated by stimulation with a maximum dose ACTH in a superfusion setup. MC2R and MC5R appear phylogenetically well conserved. MC2R is predominantly expressed in head kidney; a low abundance was found in spleen and kidney. MC5R is expressed in brain, pituitary PD, kidney, and skin. Quantitative PCR analysis of MC2R and MC5R expression in the head kidney of restrained fish reveals MC2R mRNA downregulation after 7 days restraint, in line with lower plasma cortisol levels seen. We discuss regulation of corticosteroid production from a phylogenetic perspective. We propose that increased levels of alpha-MSH exert a positive feedback on hypothalamic corticotropin-releasing hormone release to sustain a mild stress axis activity.  相似文献

Crowding for 3 weeks significantly reduced the coefficient of condition of both brown trout and rainbow trout. However, acclimation of the hypothalamic-pituitary-interrenal (HPI) axis, as assessed by changes in plasma cortisol levels, occurred within 6 days for brown trout and within 10 days for rainbow trout. Blood lactate levels were significantly reduced in the crowded fish of both species throughout the experiment. Sexual maturation of the male fish significantly elevated the number of circulating red blood cells in both species, reduced the lactate levels in brown trout and elevated cortisol levels in the rainbow trout. Despite the relatively rapid interrenal acclimation, the numbers of thrombocytes and lymphocytes in the blood of both species were significantly reduced during the period of crowding and it is concluded that changes in the composition of circulating blood cells are more reliable indicators of chronic crowding stress than are plasma cortisol levels. These findings are discussed in relation to the role of the HPI axis in suppressing the defence systems of salmonid fish during periods of chronic stress.  相似文献

The effects of time of day and water temperature on the acute physiological stress response were investigated in young-of-the-year green sturgeon (Acipenser medirostris). The response to a 1-min air-emersion stressor was assessed during the day (08.00 h) and at night (20.00 h), as well as after acclimation to either 11 degrees C or 19 degrees C. Blood samples were collected prior to stress and at several times after exposure to the stressor, and plasma concentrations of cortisol, lactate, and glucose were determined. The magnitudes of cortisol (19.1 ng ml(-1) vs. 4.9 ng ml(-1)) and lactate (190.6 mg l(-1) vs. 166.7 mg l(-1)) were significantly higher in fish stressed at night when compared with the day. There were no significant differences in glucose levels between time periods. Although, acclimation temperature did not affect peak cortisol concentrations (56.7 and 50.3 ng ml(-1) at 11 degrees C and 19 degrees C, respectively), the duration of the response was significantly extended at 11 degrees C. Post-stressor lactate increases were similar between temperature groups, but at 11 degrees C post-stressor glucose levels were significantly increased through 6 h, suggesting stressor-induced glycogenolysis and gluconeogenesis or decreased glucose utilization. These data demonstrate that the physiological stress response in green sturgeon is modified by both time of day and temperature.  相似文献
