摘要:【目的】评估不同类型煤炭生物降解转化为甲烷的潜力,研究原位煤层的微生物群落结构特征。【方法】分别在原位模拟、补加烃降解产甲烷菌系和补加碳源下厌氧培养煤样,利用气相色谱监测甲烷产生趋势,及高通量测序技术研究原位煤层的细菌和古菌群落。【结果】10个样品中有3个高成熟度煤样可以被厌氧降解转化为甲烷。通过生物强化和添加外源底物可以促进HF煤样的产甲烷潜力。其中SL 煤样中的古菌类群主要是氢营养型产甲烷菌Methanoculleus和乙酸营养型产甲烷菌Methanosaeta为主,细菌类群主要 属于Firmicutes(54.4%)、Proteobacteria(30.9%)、未培养微生物(10.8%)、Caldiserica(1.5%)及Thermotogae(1.3%)。【结论】不同成熟度煤样降解产气潜力不同,在部分原位煤层中可能存在参与烃降解与甲烷产生的功能菌。  相似文献

不同pH缓冲液对由乙酸产甲烷菌群结构的影响   总被引:1,自引:0,他引:1  
【目的】研究不同p H缓冲液对乙酸产甲烷过程及对细菌和古菌群落结构的影响。【方法】分别添加磷酸盐(PB)、4-羟乙基哌嗪乙磺酸(HEPES)、哌嗪-1,4-二乙磺酸(PIPES)和Na HCO3/CO2缓冲液到乙酸产甲烷菌系中,定期监测甲烷产生趋势,到稳定期后收集菌体,进行16S rRNA基因的末端限制性片段多态性分析(T-RFLP)。【结果】发现PB组的乙酸产甲烷菌系延滞期约为40d,显著高于其他组的20-24 d(P0.05);Na HCO3/CO2组乙酸转化为甲烷的比例为(88.3±0.5)%,显著高于其他组的77%-81%(P0.05);不同缓冲液组的最大甲烷比生长速率为0.46-0.57 d-1(P0.05);Na HCO3/CO2组的细菌群落变化最明显,主要是未培养细菌(unclassified bacteria)、螺旋菌科细菌(Spirochaetaceae)和未培养WWE1类群的丰度较其他组分别增加到(15.5±9.4)%、(7.3±4.6)%和(17.6±6.3)%,而互养菌科(Synergistaceae)的细菌丰度降低到(8.9±8.1)%。AC+PB组中的古菌类群发生了明显变化,以竹节状甲烷鬃毛菌(Methanosaeta harundinacea)相关的产甲烷古菌占主导(97±2%),而在HEPES、PIPES和Na HCO3/CO2组和不加缓冲液组中同时存在两类乙酸营养型产甲烷古菌M.harundinacea和联合鬃毛甲烷菌(Methanosaeta concilii),以及属于甲烷杆菌目(Methanobacteriales)的氢营养型产甲烷古菌。【结论】在乙酸产甲烷菌系中加入PB增加了甲烷产生的延滞期,加入Na HCO3/CO2增加了甲烷产量,但是添加p H缓冲液不会影响到菌系的最大甲烷比生长速率。加入PB和Na HCO3/CO2都会显著改变微生物的菌群结构。这些研究为设计适宜的产甲烷菌系生长条件提供了参考。  相似文献

【目的】通过分子生态学手段筛选适合互营烃降解菌Syntrophus sp.生长的非烃碳源。【方法】利用实验室驯化获得的正十六烷烃降解产甲烷菌系M82为接种物,添加不同碳源(正十二烷二元酸、正十四烷二元酸、正十六烷烃、十六烷酸钠、乳酸钠和丙酸钠)传代培养,通过PCR-DGGE和qPCR技术研究不同碳源条件下Syntrophaceae科细菌的丰度与变化趋势;利用T-RFLP方法分析古菌群落结构。【结果】菌系M82可以利用多种脂肪酸生长并产生甲烷,但是细菌群落结构发生了变化,只在添加正十二烷二元酸和正十四烷二元酸的培养液中检测到了代表Syntrophaceae细菌的条带,并且每毫升菌液中Syntrophaceae细菌的log丰度分别达到7.4和7.6,比添加其它几种非烃碳源的实验组丰度高2-3个单位。古菌群落结构主要由乙酸营养型产甲烷古菌(Methanosaeta)和氢营养型产甲烷古菌(Methanoculleus)组成。【结论】Syntrophus sp.细菌可以利用正十二烷二元酸和正十四烷二元酸这两种非烃碳源生长,这为我们定向分离互营烃降解菌和研究起始烃降解机制和代谢机理提供了依据。  相似文献

内蒙古自治区二连盆地、海拉尔盆地是我国重要的煤层气产区,其中生物成因煤层气是煤层气的重要来源,但复杂物质转化产甲烷相关微生物群落结构及功能尚不清楚。【目的】研究煤层水中的微生物代谢挥发性脂肪酸产甲烷的生理特征及群落特征。【方法】以内蒙古自治区二连盆地和海拉尔盆地的四口煤层气井水作为接种物,分别添加乙酸钠、丙酸钠和丁酸钠厌氧培养;定期监测挥发性脂肪酸降解过程中甲烷和底物的变化趋势,应用高通量测序技术,分析原始煤层气井水及稳定期产甲烷菌液的微生物群落结构。【结果】除海拉尔盆地H303煤层气井微生物不能代谢丙酸外,其他样品均具备代谢乙酸、丙酸和丁酸产生甲烷的能力,其生理生态参数存在显著差异,产甲烷延滞期依次是乙酸<丁酸<丙酸;最大比产甲烷速率和底物转化效率依次是丙酸<乙酸<丁酸。富集培养后,古菌群落结构与煤层气井水的来源显著相关,二连盆地优势古菌为氢营养型产甲烷古菌Methanocalculus (相对丰度13.5%–63.4%)和复合营养型产甲烷古菌Methanosarcina (7.9%–51.3%),海拉尔盆地的优势古菌为氢营养型产甲烷古菌Methanobact...  相似文献

【背景】芦苇湿地是甲烷主要的排放源之一,产甲烷古菌是唯一产生大量甲烷的生物,而盐碱湿地芦苇根际土优势甲烷途径鲜有研究。【目的】调查扎龙低温盐碱湿地芦苇根际土中的优势产甲烷途径。【方法】通过16S rRNA基因扩增子测序,分析扎龙湿地芦苇生长季根际土壤深度0–20 cm的产甲烷古菌和细菌组成。用已知的产甲烷底物三甲胺、甲醇、乙酸和H2/CO2,以及高盐环境植物和细菌的相似相容物质——甜菜碱(被细菌还原成三甲胺)在pH 8.0培养获得芦苇根际土的产甲烷富集物。测定各种富集物的产甲烷速率鉴定芦苇根际土的优势产甲烷途径;测定甜菜碱富集物中的16S rRNA基因多样性,并用RT-qPCR定量优势细菌和古菌的物种组成,从而推测协同代谢甜菜碱产甲烷的细菌和古菌类群。【结果】扎龙盐碱湿地芦苇根际土含有氢营养型的甲烷杆菌属(Methanobacterium,36.42%)、偏好低氢的Rice Cluster Ⅱ (11.55%)、乙酸营养型的甲烷鬃菌属(Methanosaeta,11.29%)、甲基营养型的甲烷八叠球菌属(Methanosarcina,6.53...  相似文献

【背景】电化学厌氧消化(electrochemical anaerobic digestion,EAD)系统的代谢途径由具备不同功能的微生物所主导,其代谢通量与功能微生物丰度、活性及群落结构相关。【目的】探究EAD产甲烷代谢通量与微生物的关系。【方法】采用代谢通量分析(metabolic flux analysis,MFA)方法,以pH为扰动因子得到微生物群落与产甲烷通量的响应关系。【结果】pH 7.5扰动时产甲烷通量最大为0.398 4±0.029 3,较对照组(pH 6.9)的0.297 4±0.012 7和扰动组(pH 6.3)的0.136 5±0.012 0分别提高了25%和65%。另外,平均有33.8%±3.1%的氢气(通量)用于还原二氧化碳产甲烷和乙酸,平均有21.0%±2.6%的乙酸(通量)转化为甲烷。此外,产甲烷通量与Mariniphaga、Methanosaeta和Desulfomicrobium的丰度呈正相关,与Sedimentibacter的丰度呈负相关且影响显著。【结论】在EAD产甲烷体系中产甲烷菌和产酸菌共存时,pH值略大于7.0的环境有利于甲烷的生成,改变E...  相似文献

本研究构建了以丁酸为唯一碳源的厌氧消化反应器,利用16S rRNA基因测序技术分析氯四环素(Chlortetracycline,CTC)单独抑制及CTC与颗粒活性炭(Granular active carbon,GAC)协同作用下,互营丁酸氧化微生物群落结构的动态变化,探究环境胁迫下微生物之间的相互作用及其对CTC及CTC和GAC协同作用的响应。结果表明,原始反应器群落中,已知的互营丁酸氧化菌Syntrophomonas(11.6%)和乙酸营养型产甲烷古菌Methanosaeta(48.5%)分别在细菌和古菌群落中占主导优势。添加40mg/L和50 mg/L CTC条件下,甲烷产量分别降低40.4%和49.3%。Syntrophomonas对CTC表现出耐受性,但与其呈正相关联系的细菌(如unclassified Firmicutes和unclassified Comamonadaceae)以及乙酸氧化菌Tepidanaerobacter活性被CTC明显抑制,从而影响丁酸降解率,同时造成代谢产物积累,导致产甲烷量降低。单独添加GAC以及在40 mg/L和50 mg/L CTC抑制下添加GAC,甲烷产量分别降低2.9%、48.5%和64.7%。共现网络分析结果显示,添加GAC明显增强了Geobacter以及与其呈正相关联系的细菌(Azonexus等)的活性。而产甲烷古菌Methanosaeta和Methanoculleus与Azonexus等大部分细菌呈负相关,因此,添加GAC可能间接影响了产甲烷古菌的活性。  相似文献

青藏高原三个盐碱湖的产甲烷菌群和产甲烷代谢途径分析   总被引:1,自引:0,他引:1  
【目的】分析青藏高原不同类型盐碱湖中的优势产甲烷菌群和优势产甲烷代谢途径。【方法】以不同盐度和植被类型的公珠错、昆仲错和无植被的兹格塘错的沉积物为研究对象,通过高通量测序和q PCR定量古菌16S r RNA多样性分析优势古菌类群;模拟原位盐浓度及p H,比较不同产甲烷底物(甲醇、三甲胺、乙酸和H_2/CO_2)富集沉积物的产甲烷速率,分析其优势产甲烷菌代谢类型。通过添加产甲烷抑制剂(2-溴乙烷磺酸盐),检测沉积物中产甲烷底物积累,确定不同盐碱湖中主要的产甲烷途径。【结果】昆仲错的优势菌群包括甲基/乙酸型的甲烷八叠球菌科(Methanosarcinaceae,11%),乙酸型的甲烷鬃菌科(Methanosaetaceae,7.9%)和氢型甲烷菌甲烷杆菌目(Methanomicrobiales,7.4%);公珠错和兹格塘错的优势菌群为甲烷鬃菌科(Methanosaetaceae)分别占15%和15.3%,及甲烷杆菌属(Methanobacterium)和甲基型的甲烷叶菌属(Methanolobus)。公珠错和昆仲错分别以乙酸和甲醇产甲烷速率最高,而兹格塘错从不同底物产甲烷速率无差异。抑制甲烷产生后,公珠错主要积累乙酸,昆仲错主要积累甲醇;兹格塘错不仅甲烷排放低,也无产甲烷物质显著积累。【结论】昆仲错沉积物中的甲烷主要来自甲醇,公珠错中的甲烷主要来自乙酸,而兹格塘错产甲烷和底物积累不活跃。因而推测高原盐碱湖主要的产甲烷途径和菌群可能与周围植被类型的相关性更高,而与盐度的直接相关性较低。  相似文献

【背景】桂林会仙湿地位于喀斯特峰丛洼地和峰丛平原的过渡区,主要由岩溶地下河补给,水质呈现富钙偏碱特征,是一处典型的岩溶湿地环境,湿地沉积物以厌氧环境为主,独特的环境特征使之成为研究新型厌氧微生物的理想场所。氢型产甲烷菌和乙酸型产甲烷菌是环境中有机质厌氧降解的重要参与者,但目前会仙湿地产甲烷菌生理生态功能的研究十分有限。【目的】揭示乙酸盐营养型产甲烷条件和氢气营养型产甲烷条件下岩溶湿地环境古菌群落结构的变化规律和产甲烷菌的主要类群,探讨岩溶环境古菌的功能和相互关系。【方法】以会仙岩溶湿地沉积物为研究材料,分别以乙酸盐和氢气为唯一能源物进行富集培养,结合未添加任何能源底物的对照处理,动态监测产甲烷通量,分别构建了3个共包含146条古菌16S rRNA基因全长序列的文库,以及3个共包含138条甲基辅酶M还原酶基因mcr A基因序列的文库,通过构建系统发育树比较分析不同产甲烷底物培养条件下典型岩溶湿地古菌群落的变化规律。【结果】从乙酸型和氢型产甲烷条件培养物顶空气中都检测到了几乎等浓度的甲烷产生,甲烷八叠球菌是mcr A文库中主要的古菌类群,其中包含了Methanosarcina属古菌、Zoige Cluster I (ZC-I)和ANME-2d Cluster (AD)类群古菌,以及两个相对独立且不包含任何参考序列的分支KT-I和KT-II,可能代表产甲烷菌的新类群;经过两种产甲烷条件的富集培养后,ZC-I、AD和KT-II类群古菌的序列数占比有较为显著的增加(45%–155%);深古菌(Bathyarchaeota)是所有古菌16Sr RNA基因文库的优势类群,序列占比为88%–100%,其中MCG-11亚群最为丰富,占所有深古菌的84%,并且在乙酸盐产甲烷条件下增加了17%。【结论】会仙湿地沉积物中蕴涵着丰富的新型古菌序列,沉积物中主要的氢型产甲烷菌和乙酸型产甲烷菌都来自甲烷八叠球菌目,深古菌在岩溶环境和乙酸型产甲烷条件下可能都发挥着重要的作用。  相似文献

【目的】揭示芦岭煤田微生物群落组成,并分析其潜在的产甲烷类型及产甲烷途径。【方法】采集芦岭煤田的煤层气样品和产出水样品,分别分析样品的地球化学性质特征;利用Illumina HiSeq高通量测序技术分析产出水中的微生物群落结构;采用添加不同底物的厌氧培养实验进一步证实芦岭煤田生物成因气的产甲烷类型。【结果】该地区煤层气为生物成因和热成因的混合成因气;古菌16S rRNA基因分析表明在产出水中含有乙酸营养型、氢营养型和甲基营养型的产甲烷菌。丰度较高的细菌具有降解煤中芳香族和纤维素衍生化合物的潜力。厌氧富集培养结果表明,添加乙酸盐、甲酸盐、H2+CO2为底物的矿井水样均有明显的甲烷产生。【结论】芦岭煤田具有丰富的生物多样性,该地区同时存在三种产甲烷类型。本研究为利用微生物技术提高煤层气的采收率,实现煤层气的可持续开采提供科学依据。  相似文献

Methanogenic bacteria comprise a selected group of microorganisms that derive their energy for growth from the hydrogen-dependent reduction of CO2 to methane or the disproportionation of reduced one-carbon compounds and acetate to CO2 and methane. In the reduction and oxidation steps at the formyl, hydroxymethyl and methyl level the one-carbon unit remains bound to the reduced form of methanopterin, a pterin derivative typical of methanogenic bacteria. In addition, the reduced methanopterin, 5,6,7,8-tetrahydromethanopterin, is involved in a number of anabolic reactions. Methanopterin is structurally and functionally the counterpart of folic acid found in other organisms. In this review the occurrence and properties of methanopterin and its derivatives, as well as the biosynthesis and the role in the different catabolic and anabolic reactions are discussed against the background of folic acid biochemistry.  相似文献

Methanogenic archaebacteria have developed unique ways of dealing with osmotic stress. While several of them have transport systems capable of internalizing betaine, an osmolyte in many eubacteria, in general they have developed de novo synthesis of a novel series of beta-amino acids as compatible solutes. 13C-NMR spectroscopy has been the key tool in elucidating both the identity of these organic osmolytes and in investigating their dynamics.  相似文献

In total, 73 strains of methanogen isolates from our laboratory and 6 from culture collections were examined for the presence of plasmid DNA. Five strains were found to contain detectable plasmids. Multiple plasmids were found in two isolates, while three strains contained only one plasmid each. A physical map of the plasmid pT3 was constructed by use of six different restriction endonucleases. All sites were aligned with a single BgII site, and the position of the restriction sites was determined by double or sequential digestion of the plasmid DNA.  相似文献

Abstract: In the anaerobic treatment of sulfate-containing wastewater, sulfate reduction interferes with methanogenesis. Both mutualistic and competitive interactions between sulfate-reducing bacteria and methanogenic bacteria have been observed. Sulfate reducers will compete with methanogens for the common substrates hydrogen, formate and acetate. In general, sulfate reducers have better growth kinetic properties than methanogens, but additional factors which may be of importance in the competition are adherence properties, mixed substrate utilization, affinity for sulfate of sulfate reducers, relative numbers of bacteria, and reactor conditions such as pH, temperature and sulfide concentration. Sulfate reducers also compete with syntrophic methanogenic consortia involved in the degradation of substrates like propionate and butyrate. In the absence of sulfate these methanogenic consortia are very important, but in the presence of sulfate they are thought to be easily outcompeted by sulfate reducers. However, at relatively low sulfate concentrations, syntrophic degradation of propionate and butyrate coupled to HZ removal via sulfate reduction rather than via methanogenesis may become important. A remarkable feature of some sulfate reducers is their ability to grow fermentatively or to grow in syntrophic association with methanogens in the absence of sulfate.  相似文献

Abstract A blue fluorescent compound was extracted and purified from cells of Methanobacterium thermoautotrophicum . The compound was identified as 7-methylpterin on the basis of its (physico-) chemical properties and by comparison with 7-methylpterin prepared by organic synthesis. The compound is present in all methanogenic bacteria studied so far and it provides methanogenic bacteria the characteristic blue fluorescence observed upon fluorescence microscopy.  相似文献

NMR spectroscopy was used to determine the labeling patterns of the ribose moieties of ribonucleosides purified from Methanospirillum hungatei, Methanococcus voltae, Methanobrevibacter smithii, Methanosphaera stadtmanae, Methanosarcina barkeri and Methanobacterium bryantii labeled with 13C-precursors. In most methanogens tested ribose was labeled in a manner consistent with the operation of the oxidative branch of the pentose phosphate pathway. In contrast, transaldolase and transketolase reactions typical of a partial nonoxidative pentose phosphate pathway are hypothesized to explain the different labeling patterns and enrichments of carbon atoms observed in the ribose moiety of Methanococcus voltae. The source of erythrose 4-phosphate needed for the transaldolase reaction proposed in Methanococcus voltae, and for biosynthesis of aromatic amino acids in methanogenic bacteria in general, was assessed. Phenylalanine carbon atom C-7 was labeled by [1-13C]pyruvate in Methanospirillum hungatei, Methanococcus voltae, and Methanococcus jannaschii, the only methanogens which incorporated sufficient label from pyruvate for testing. Reductive carboxylation of a triose precursor (derived from pyruvate) to synthesize erythrose 4-phosphate is consistent with the labeling patterns observed in phenylalanine and ribose.Abbreviation TCA Tricarboxylic acid Issued as NRCC Publication No. 37382  相似文献

Acid-hydrolysis of the phospholipid fraction of Methanobacterium thermoautotrophicum, Methanobacterium formicicum and Methanospirillum hungatii demonstrated the presence of two neutral lipid products. Characterization of these lipids resulted in their identification as dialkyl glyceryl ether and diglycerol tetraethers. The ether-linked alkyl chains were identified as the 20- and 40-carbon branched chains for the diether and tetraether, respectively. M. thermoautotrophicum and M. formicicum were also characterized by the presence of acid-stable phospholipid components.  相似文献

Summary A methanogenic sludge showed denitrifying activity for acetate, glucose and effluents from methanogenic treatments as substrates; denitrifiers were present in a relatively high number. When glucose was used as substrate dissimilatory reduction of nitrate to ammonium occurred. Methane production from acetate was inhibited by denitrification and resumed after nitrite and nitrous oxide depletion.  相似文献
