The 1.709 or satellite IV repeated DNA family originally isolated from the domestic cow was analyzed using Southern blotting, pulsed field gel electrophoresis, fluorescence in situ hybridization, and DNA sequencing in species belonging to the genera Bos, Bison, Bubalus, Syncerus, Boselaphus, and Tragelaphus. Hybridization indicates that the family has been amplified in Bos, Bison, Bubalus, and Syncerus but not in Boselaphus or Tragelaphus. Pericentromeric, higher-order repeat substructure exists in all species, with multimeric arrays ranging in size from 10 to 1500 kb. Sequence analysis of a 492-bp PCR product revealed comparable levels (0.2–4.5%) of intra- and interspecific divergence when species of Bos and Bison were compared, supporting the idea that species of these two genera should be recognized under the genus Bos. Alternatively, all Syncerus sequences cluster as a monophyletic group on an evolutionary tree and differ from those of Bos/Bison by about 13%. Comparing these findings with the fossil record indicates that concerted evolution has occurred since Bos/Bison and Syncerus last shared a common ancestor (5.0 MYA) but before the radiation of the genus Bos (2.5 MYA): GenBank accession numbers AY517856-AY517904. Background

It has been established that mammalian egg zona pellucida (ZP) glycoproteins are responsible for species-restricted binding of sperm to unfertilized eggs, inducing the sperm acrosome reaction, and preventing polyspermy. In mammals, ZP apparently represents a barrier to heterospecific fertilization and thus probably contributes to reproductive isolation between species. The evolutionary relationships between some members of the tribe Bovini are complex and highly debatable, particularly, those involving Bos and Bison species for which interspecific hybridization is extensively documented. Because reproductive isolation is known to be a major precursor of species divergence, testing evolutionary patterns of ZP glycoproteins may shed some light into the speciation process of these species. To this end, we have examined intraspecific and interspecific genetic variation of two ZP genes (Zp2 and Zp3) for seven representative species (111 individuals) from the Bovini tribe, including five species from Bos and Bison, and two species each from genera Bubalus and Syncerus.  相似文献

Polymorphism in mitochondrial DNA (mtDNA) of yak (Bos grunniens)   总被引:3,自引:0,他引:3

Mitochondrial DNAs (mtDNA) from 21 yaks (Bos grunniens) were assayed for restriction fragment length polymorphisms by using 20 restriction endonucleases, six of which (AvaI, AvaII, BglII, EcoRI, HindIII, and HpaI) detected polymorphism. Four different mtDNA haplotypes were identified. Combining this with previous reports about the mtDNA RFLPs of B. indicus and B. taurus, there are obvious differences in mtDNA polymorphism between the yak and other Bos species. We estimated that the divergence times between the ancestor of B. grunniens and the ancestor of B. taurus or B. indicus were about 1.2–2.2 and 1.01–2.02 million years ago, respectively.  相似文献

Noncoding regions from the genes encoding aromatase cytochrome P450 and lactoferrin have been sequenced in ten bovine and one cervid species for an investigation of the evolutionary relationships within the tribe Bovini. The evolutionary rate of DNA-nucleotide alterations along the ancestral bovine lineage amounts to 0.38% per million years, as estimated from this combined 0.478-kb-single copy nuclear (scn) DNA sequence data set. Whereas rate homogenity is apparent within the Bovini, the relative rate test suggests that the boselaphine lineage (as represented by Boselaphus) has evolved at only about one third of the rate found within the Bovini. Consistent with other results, the scnDNA data provide evidence for (i) a monophyletic origin of the Bovini, (ii) a sister group position of the Boselaphini, and (iii) two different clades within the Bovini, the buffaloes (Bubalus and Syncerus) and the cattle (Bos/Bibos and Bison). Surprisingly, the results indicate very clearly that the enigmatic dwarf buffalo of Sulawesi Island (Anoa depressicornis) is most closely related to Boselaphus and that the divergence from the true Bovini occurred close to the base of bovine cladogenesis in the Middle Miocene (≈ 14—12 million years ago).  相似文献

Bos taurus-Bos indicus twin calves were produced by transferring Day 6–8 Bos indicus or Bos taurus embryos to previously inseminated Bos taurus or Bos indicus cows. Embryos were transferred to 112 recipients of which 64 (57.1%) were diagnosed as pregnant by rectal palpation at 7–8 weeks gestation. Twin foetuses were diagnosed in 38 (65.5%) of 58 cows in which the location and number of foetuses was recorded. Substantial foetal losses occurred after 7–8 weeks and only 44 cows calved giving 21 sets of twins and 23 singletons. Six of the single calves were from transferred embryos. The duration of gestation of the twin pregnancies (282 days) was similar to that of single Bos taurus pregnancies (282 days for Jerseys and 286 days for Friesians) and 14 days shorter than single Bos indicus pregnancies (296 days). Consequently Bos indicus members of mixed-species twins were premature at birth and required intensive care to ensure their survival.  相似文献

Body measurement traits, influenced by genes and environmental factors, play numerous important roles in the value assessment of productivity and economy. Growth differentiate factor 5 (GDF5), involved in the development and maintenance of bone and cartilage, is an important candidate gene for body measurement traits selection through marker-assisted selection (MAS). In this study, based on the PCR-RFLP technology, we discovered and evaluated the potential association of the single nucleotide polymorphism (SNP) (T586C in exon 1) of the bovine GDF5 gene with body measurement traits in 985 Bos taurus breed, 42 Bos indicus breed and 76 Bos indicus × Bos taurus individuals. As the SNP marker, there were the significant effects on the Body length (BL) in the Bos taurus (BT) and Bos indicus × Bos taurus (BMY) populations (P < 0.05). In BT population, animals with the genotype TT had lower mean values for BL and Hip width (HW) than these with the TC and CC genotype (P < 0.01). In BMY population, animals with the genotype TC had lower mean values for BL than these with the genotype CC (P < 0.05). These results suggest that the SNP of the GDF5 gene could be a very useful genetic marker for body measurement traits in the bovine reproduction and breeding.  相似文献

The relative importance of dry- and wet-bulb temperatures on cutaneous function inBos indicus andBos taurus females under humid tropical climatic conditions was evaluated. The parameters investigated were sweating rate and skin temperature, while the species utilised were zebu White Fulani (Bos indicus) and German Brown and German Black and White (Bos taurus). The sweating rate, irrespective of breed, differed with the site of sampling and was more influenced by dry-bulb (59%) than by wet-bulb temperature (41%). Skin temperature responded more to wet-bulb temperature in White Fulani and German Black and White cattle, but not in German Brown cattle. It is concluded that the response of the animals, with respect to sweating, was similar but that the efficiency of sweating, judged by the lowering of skin temperature, was higher inBos indicus than inBos taurus. This, in part, may explain the higher degree of comfort demonstrated byBos indicus under tropical conditions.  相似文献

Isoelectric focusing of red cell hemolysates revealed several isozymes that stain for NADH-methemoglobin reductase. Evidence for two different genetic loci controlling the banding patterns was obtained. One locus controlled a single band present in all animals tested. The second locus controlled ten different banding patterns that could be accounted for by four codominant alleles. Band B occurred in Bison bison. Bands A and C occurred in Bos indicus and band D occurred in both Bos indicus and Bos taurus. Bands A, C, and D were not observed in Bison bison and bands A, B, and C were not observed in Bos taurus.  相似文献

The Bovini tribe contains domestic and wild cattle‐like species, several of which are cross‐fertile. We present a completely resolved Y‐chromosomal phylogeny, which is better in agreement with autosomal phylogeny, morphological data, cross‐fertility and estimates of divergence times than mitochondrial data. The tree links Bos grunniens (yak) to Bison, so the commonly accepted Bos genus is not monophyletic. Therefore, we advocate the term Poephagus as designation of yak. This work illustrates the resolving power of Y‐chromosomal variation for cladistic studies of closely related species. © The Willi Hennig Society 2008.  相似文献

Progesterone concentrations were determined by radioimmunoassay in the plasma of heifers of German Brown Swiss, Holstein-Friesian and White Fulani cattle during two consecutive estrous cycles in the dry and wet seasons in a tropical climate. Seasonal differences in the levels were not significant but peak levels were significantly higher in the Bostaurus than in the Bosindicus heifers. The low levels at estrus and the gradual rise five days after estrus were similar among the heifers. The levels on the 1st and 2nd days preceding estrus were significantly higher in the Bostaurus than in the Bosindicus heifers, and suggest a possible tendency for the former to be more susceptible to infertility arising from high progesterone levels 1 and 2 days preceding estrus and insemination. The genital tract and ovaries at the mature luteal phase were generally smaller in the Bosindicus than in the Bostaurus heifers.  相似文献

Genome fingerprinting with a hypervariable minisatellite sequence of phage M13 DNA was used to study the genetic variation in individual species of the genera Bosand Bison(subfamily Bovinae) and in their interspecific and intergeneric hybrids. DNA fingerprints were obtained for domestic cow Bos taurus primigenius, vatussy Bos taurus macroceros, banteng Bos javanicus, gaur Bos gaurus, wisent Bison bonasus, bison Bison bison, and for the interspecific and intergeneric hybrids. Compared with the original species, most hybrids showed a greater variation in number and size of hybridization fragments. An association was revealed between the number of hybridization fragments and degree of consanguinity of interspecific hybrids resulting from unique crossing of domestic cow and banteng. Pairwise similarity coefficients were calculated to construct a dendrogram of genetic similarity, which reflected the relationships between the parental species and hybrids varying in degree of consanguinity. The applicability of the method for identifying interspecific and intergeneric hybrids and for studying the consequences of hybridization in the subfamily Bovinae is discussed.  相似文献

Bos taurus indicus cattle are less susceptible to infestation with Rhipicephalus (Boophilus) microplus than Bos taurus taurus cattle but the immunological basis of this difference is not understood. We compared the dynamics of leukocyte infiltrations (T cell subsets, B cells, major histocompatibility complex (MHC) class II-expressing cells, granulocytes) in the skin near the mouthparts of larvae of R. microplus in B. t. indicus and B. t. taurus cattle. Previously naïve cattle were infested with 50,000 larvae (B. t. indicus) or 10,000 larvae (B. t. taurus) weekly for 6 weeks. One week after the last infestation all of the animals were infested with 20,000 larvae of R. microplus. Skin punch biopsies were taken from all animals on the day before the primary infestation and from sites of larval attachment on the day after the first, second, fourth and final infestations. Infiltrations with CD3+, CD4+, CD8+ and γδ T cells followed the same pattern in both breeds, showing relatively little change during the first four weekly infestations, followed by substantial increases at 7 weeks post-primary infestation. There was a tendency for more of all cell types except granulocytes to be observed in the skin of B. t. indicus cattle but the differences between the two breeds were consistently significant only for γδ T cells. Granulocyte infiltrations increased more rapidly from the day after infestation and were higher in B. t. taurus cattle than in B. t. indicus. Granulocytes and MHC class II-expressing cells infiltrated the areas closest to the mouthparts of larvae. A large volume of granulocyte antigens was seen in the gut of attached, feeding larvae.  相似文献

A small number of west African Bos taurus cattle breeds, including the N'Dama, constitute a valuable genetic resource by virtue of their ability to remain productive under trypanosomiasis challenge. However, introgression of Bos indicus genes into the trypanotolerant breeds, particularly by introduction of zebu bulls, is a threat to this resource. This work describes the characterization and cloning of a bovine randomly amplified polymorphic DNA (RAPD) that is generated in polymerase chain reaction (PCR) with the 10 base primer ILO1065 from Bos indicus male templates, but not from B. taurus male templates or female templates of either type. Male-specific sequences with homology to the RAPD also occur in B. taurus breeds. This suggests that the polymorphism may be due to base substitution(s) in an ILO1065 priming site, or insertion/deletion events either affecting priming sites or occurring between sites on the cattle Y chromosome. We have shown that cattle, whether of B. indicus or B. taurus phenotype, which possess a typically B. indicus metaphase Y chromosome on the basis of QFQ banding, have a B. indicus ILO1065-generated genotype. The ILO1065-primed RAPD can be used in a simple dot blot assay as a probe of RAPD-PCR products, to provide a convenient, reliable and effective means of detecting introgression of zebu genes in B. taurus cattle populations.  相似文献

Reproductive function is mediated by season in the Indian breeds of cattle (Bosindicus). The reproductive endocrinology of Bosindicus cattle differs from that of Bostaurus breeds; the estrus is shorter and less intense and occurs late in relation to an estrogen stimulus. Moreover, the Bosindicus female has a smaller preovulatory surge of luteinizing hormone (LH), which occurs earlier relative to the onset of estrus, and she ovulates sooner after the onset of estrus. The corpus luteum is smaller and contains less progesterone, and the serum progesterone concentration is lower in Bosindicus females. Furthermore, they have fewer preovulatory LH surges than Bostaurus females and their luteal cells are less responsive to LH in vitro during the winter. Their fertility is lower during the late fall and winter months. For Bosindicus cattle, recovery of transferable embryos and survival of embryos in the recipient are at their maximum from July through October.  相似文献

Densities ofAmblyomma americanum (L.) onBos indicus, B. taurus andB. indicus x B. taurus cattle are compared over a 3-year period, and the growth rate (rate of increase or decrease) of parasitic tick populations on each cattle genotype is estimated.Average log10 densities of parasiticA. americanum larvae are significantly (P=0.05) lower onB. indicus cattle than onB. taurus andB. indicus x B. taurus cattle. Average log densities of nymphal and adult ticks onB. taurus cattle are significantly higher than onB. indicus cattle but neither cattle genotype differs in this regard fromB. indicus x B. taurus cattle.Estimated annual tick population growth rates (log10) for parasiticA. americanum are positive onB. taurus cattle (+0.84 larvae, +0.09 nymphs, +0.22 adults calf–1 year–1), but are negative onB. indicus (–0.18 nymphs, –0.14 adults calf–1 year–1) andB. indicus x B. taurus cattle (–0.45 larvae, –0.24 nymphs, –0.14 adults calf–1 year–1). Populations of parasitic larvae were not detected onB. indicus cattle.  相似文献

Summary It is suggested that the low calving rate of crosses between Bos taurus cows and Brahman bulls in the F2 generation, in later generations and in breeds derived from the cross, could be due to the difference between the Bos taurus Y and the Bos indicus Y if this difference is due to a translocation between the Y and an autosome. It is suggested that such a translocation could set up a balanced polymorphism and perpetuate low calving rates against selection.  相似文献

Indigenous cattle of India belong to the species, Bos indicus and they possess various adaptability and production traits. However, little is known about the genetic diversity and origin of these breeds. To investigate the status, we sequenced and analyzed the whole mitochondrial DNA (mtDNA) of seven Indian cattle breeds. In total, 49 single-nucleotide variants (SNVs) were identified among the seven breeds analyzed. We observed a common synonymous SNV in the COII gene (m.7583G?>?A) of all the breeds studied. The phylogenetic analysis and genetic distance estimation showed the close genetic relationship among the Indian cattle breeds, whereas distinct genetic differences were observed between Bos indicus and Bos taurus cattle. Our results indicate a common ancestor for European Zwergzebu breed and South Indian cattle. The estimated divergence time demonstrated that the Bos indicus and Bos taurus cattle lineages diverged 0.92 million years ago. Our study also demonstrates that ancestors of present zebu breeds originated in South and North India separately ~30,000 to 20,000 years ago. In conclusion, the identified genetic variants and results of the phylogenetic analysis may provide baseline information to develop appropriate strategies for management and conservation of Indian cattle breeds.  相似文献

Although the complete bovine mitochondrial DNA molecule has been previously sequencedand sequence comparisons of the mitochondrial displacement loop have been performed, detailed sequence information is limited on coding regions of mitochondrial DNA within and among breeds of Bos taurus and Bos indicus. This study analysed polymorphism of the mitochondrial DNA transfer RNA genes for trypto-phan, alanine, asparagine, cysteine, tyrosine and the origin of light strand replication among Ayrshire, Canadian, Belgium Blue, Brown Swiss, Hereford, Jersey, Limousine, Piedmon-taise, Red Angus, Simmental (Bos taurus) and a Nellore (Bos indicus). Nucleotide sequence analysis of a 420-bp fragment of mitochondrial DNA comprising the five transfer RNA genes showed 100% homology among single individuals of the Bos taurus breeds. The Nellore breed showed guanine to adenine substitutions in the DHU arm of asparagine tRNA and in the origin of light-strand replication. This equates to a 0.5% sequence difference between the Nellore andBos taurus breeds and may reflect an independent evolutionary origin of the species.  相似文献

The number of oocytes recovered from Bos taurus indicus females subjected to ovum pick-up averaged two to four times greater compared to Bos taurus taurus females. The objective of the present study was to test the hypothesis that this difference in oocyte yield was due to more preantral follicles in the ovaries of Bos indicus females. Ovaries (n = 64) from Nelore (Bos indicus) fetuses (n = 10), heifers (n = 12), and cows (n = 10), and Aberdeen Angus (Bos taurus) fetuses (n = 10), heifers (n = 12), and cows (n = 10) were cut longitudinally into halves, fixed, and processed for histological evaluation. The number of preantral follicles was estimated by counting them in each histological section, using the oocyte nucleus as a marker and employing a correction factor. The average number of preantral follicles in the ovaries of Bos indicus vs Bos taurus was (mean ± SD) 143,929 ± 64,028 vs 285,155 ± 325,195 for fetuses, 76,851 ± 78,605 vs 109,673 ± 86,078 for heifers, and 39,438 ± 31,017 vs 89,577 ± 86,315 for cows (P > 0.05). The number of preantral follicles varied greatly among individual animals within the same category, as well as between breeds. In conclusion, we inferred that the higher oocyte yield from Bos indicus females was not due to a greater ovarian reserve of preantral follicles. Therefore, mechanisms controlling follicle development after the preantral stage likely accounted for differences between Bos indicus and Bos taurus females in number of oocytes retrieved at ovum pick-up.  相似文献
