Bioremediation is considered a promising eco-efficient alternative for industrial wastewater treatment. Particular attention is currently being given to biological degradation of synthetic dyes and more specifically to colour removal by fungi. This work looks at the extracellular enzymatic system of strain Euc-1. Its ability to decolourize 14 xenobiotic azo dyes was evaluated and compared with the well-known species Phanerochaete chrysosporium. Strain Euc-1 is a mesophilic white-rot basidiomycete, the main secreted ligninolytic enzyme being laccase (0.38 U ml–1). Although low manganese-dependent peroxidase activity (0.05 U ml–1) was also detected, neither lignin peroxidase nor aryl alcohol oxidase could be found in batch culture. Optimum pH values of 4.0 and 5.0 were obtained in the laccase-catalysed oxidation of guaiacol and syringaldazine, respectively. Laccase activity increased with the temperature rise up to 50–60 °C and remarkable thermal stability was observed at 50 °C with a half-life of 12 h and no deactivation within the first 2 h. Solid-plate decolourization studies showed that basidiomycete Euc-1 decolourized 11 azo dyes whereas P. chrysosporium only two. Moreover, it is shown that purified laccase from basidiomycete Euc-1 efficiently decolourizes the azo dye acid red 88.  相似文献

利用东方栓孔菌Trametes orientalis菌丝体制得的生物吸附剂,对刚果红、结晶紫、铬天青、亚甲基蓝和中性红5种染料进行了脱色试验,对脱色效果较好的染料进行了影响因子优化,并通过红外光谱分析、化学修饰等方法研究其吸附作用的机制。结果显示:东方栓孔菌菌丝体生物吸附剂对结晶紫有相对较好的脱色效果;在优化的影响因子中,0%–3.0%(m/v)浓度范围内,盐度对染料脱色有促进作用;表面活性剂促使脱色率增加,吐温60浓度为1.5%(v/v)时脱色率可达83.84%;温度为43℃、初始pH为3.0、振荡培养10d后吸附率最高达91.54%。红外光谱分析及化学修饰结果表明菌丝体对染料的吸附作用主要是由它们之间的静电作用力所致。  相似文献

Two strains of Phanerochaete chrysosporium and a local isolate of white-rot fungus, if pre-cultured in a high nitrogen medium with glucose, could decolorize two azo dyes (Amaranth and Orange G) and a heterocyclic dye (Azure B). When starch was used in the pre-cultivation medium, decoloration of Orange G occurred if the medium also contained 12mM NH4Cl, whether or not veratric acid was present. In medium containing 1.2mM NH4Cl and veratric acid, greater decoloration occurred with one strain of P. chrysosporium and the local isolate. 以平皿培养方式对采集自中国和芬兰的白腐真菌菌株降解6种不同结构的人工染料的能力进行了筛选研究。在40株菌株中,黑管孔菌Bjerkandera adusta Y5012,一色齿毛菌Cerrena unicolor Y5002,硬毛粗盖孔菌Funalia trogii Y4997,香栓孔菌Trametes suaveolens D8325和云芝栓孔菌Trametes versicolor Y4946对刚果红、橙黄G、茜素红、结晶紫、中性红和亚甲基蓝均显示出较强的脱色能力。对一色齿毛菌Cerrena unicolor Y5002的液体培养脱色条件进行了研究,其最适碳源和氮源分别为蔗糖和麦芽浸粉;在不同橙黄G浓度下均获得较高的脱色率,因此浓度为500mg/L的橙黄G未对该菌的脱色能力产生抑制作用,而浓度为400mg/L茜素红则对其脱色作用产生明显抑制。对菌丝生物量和染料脱色率的研究表明,在不同碳源和氮源条件下,两者之间具有明显的正相关性。  相似文献

Eleven strains of ectomycorrhizal fungi belonging to seven species have been cultured on a cadmium-contaminated growth medium in order to determine their in vitro cadmium tolerance. Four strains were collected from a zinc and cadmium-polluted soil. Radial growth rate was a sensitive parameter to detect Cd toxicity. A wide differential response to Cd was obtained between the individual species. A clear relation between Cd tolerance and site origin of the isolates did not exist, although such a relationship was found when strains are compared within one species. Cd-sensitive and Cd-tolerant strains of Suillus bovinus were studied in more detail. Two isolates were grown on media with combinations of two non-toxic zinc concentrations and three cadmium levels. Adding a higher Zn concentration to the medium resulted in a reduction of the toxic effect of Cd. This antagonistic effect also resulted in a lowered Cd concentration in the mycelium.  相似文献

摘要:【目的】本文在利用毕赤酵母成功分泌表达高活性三苯基甲烷类染料降解酶TpmD的基础上,研究不同有机溶剂和常见酶抑制剂对纯化后的重组脱色酶TpmD活性的影响。【方法】 将毕赤酵母分泌表达的重组酶TpmD经超滤浓缩及镍离子亲和层析柱纯化后得到纯化的重组TpmD酶。利用分光光度计法测定其对孔雀石绿脱色活性,并研究了各种有机溶剂和抑制剂对TpmD酶脱色活性的影响。通过脱色过程溶氧测定和全波长扫描分析反映了DTT与NADH作为TpmD辅因子时对孔雀石绿脱色反应的表观区别。【结果】甲醇对酶活性抑制作用较弱,重组酶在甲醇中能维持约90%的活性,乙醇和丙酮则使重组酶活性则迅速丧失。低浓度二甲基亚砜有利于重组酶活性的维持,30%的二甲基亚砜可抑制一半以上酶活性。L-半胱氨酸、叠氮化钠及低浓度的EDTA对重组酶活性影响较小,较高浓度的EDTA才显示较强抑制作用;表面活性剂SDS有强烈的抑制作用,极低的浓度也能完全抑制重组酶活性。抗氧化剂二硫苏糖醇(DTT)对重组脱色酶的作用十分独特,它可替代NADH辅助脱色反应并增加脱色速率,但在反应终点脱色总效率约为NADH作为辅酶时的92%。DTT辅助TpmD脱色孔雀石绿过程中的耗氧情况及脱色产物与NADH作为辅酶时明显不相同。【结论】发现不同有机溶剂和常见抑制剂对重组酶活性的影响存在较大差异,首次证实DTT能辅助TpmD脱色反应,这一脱色反应过程的作用机理与NADH为辅酶催化的反应原理完全不同。  相似文献

The effect of gallic acid, vanillic acid, and molasses spent wash (MSW) concentration on growth and decolourizing capability of four fungi (Geotrichum candidum, Coriolus versicolor, Phanerochaete chrysosporium and Mycelia sterilia) was studied. Fungal growth was inhibited to a varying extent in the presence of gallic and vanillic acid, except for G. candidum, which was unaffected by gallic acid. G. candidum and P. chrysosporium growth rates increased in the presence of increasing concentrations of MSW (up to 50% v/v), however, growth of M. sterilia and C. versicolor was inhibited at spent wash concentrations above 5% (v/v). Increasing the concentration of MSW from 6·25% (v/v) to 12·5% (v/v) increased the decolourizing ability of each fungus, except for M. sterilia. C. versicolor exhibited greatest colour removal with a reduction of 0·43 units at A475 (equivalent to 53% colour reduction) after 10 days growth in 12·5% (v/v) MSW.  相似文献

偶氮染料广泛应用于纺织、造纸和包装等行业,因其具有三致性、结构稳定且难降解,已成为染料废水处理的研究热点之一。本研究以白腐真菌作为脱色菌株,考察了不同白腐真菌对偶氮类染料酸性橙7(acid orange 7,AO7)的脱色降解,探讨了AO7染料的浓度、pH、温度以及脱色时间对染料脱色率的影响,同时应用紫外-可见光谱吸收法、红外光谱吸收法、高效液相色谱法和气相色谱-质谱法对AO7的降解产物进行分析,并对其产物进行植物毒性实验,以推断AO7可能的降解途径及其降解产物的毒性。结果表明:在pH 4.5、28℃条件下,刺芹侧耳(Pleurotus eryngii)和杂色云芝(Trametes versicolor)的混合菌丝脱色降解100 mg/L AO7,24 h脱色率可达93.46%。推测AO7可能的生物降解途径:AO7偶氮键断裂生成对氨基苯磺酸和1-氨基-2-萘酚;接着对氨基苯磺酸脱去磺酸基,生成对苯二酚;同时1-氨基-2-萘酚开环生成邻苯二甲酸和对羟基苯甲醛,之后进一步降解生成苯甲酸;最后对苯二酚和苯甲酸继续氧化成其他小分子中间体、H2O和CO_(2)。植物毒性实验表明,P.eryngii和T.versicolor混合菌丝对AO7脱毒效果较好。以上研究为探究白腐真菌在工业废水中降解偶氮类染料的应用奠定基础。  相似文献

When various lignin-related para-phenolic benzoic acids, para-phenolic cinnamic acids, para-phenolic phenylpropionic acids, the corresponding unsubstituted and 4-O-methylated derivatives, and 4-hydroxyl substituted benzaldehydes were tested on the growth of eight white-rot fungi, methylation of the 4-hydroxy substituent resulted, in most cases, in increased inhibition of fungal growth. This effect was most notable with monomethoxylated compounds. When the aromatic ring contained additional methoxyl substituents, the toxicity of the 4-O-methylated derivative was less pronounced. Marked inhibition of fungal growth was also observed with aromatic compounds lacking a para-substituent. Higher concentrations of aromatic aldehydes were manifestly more toxic than the corresponding carboxylic acid.J.A. Nine white-rot fungal strains were screened for biodecolourization of brilliant green, cresol red, crystal violet, congo red and orange II. Dichomitus squalens, Phlebia fascicularia and P. floridensis decolourized all of the dyes on solid agar medium and possessed better decolourization ability than Phanerochaete chrysosporium when tested in nitrogen-limited broth medium. Growth of three strains of Tetracladium marchalianum was inhibited by Cd-, and, to a lesser extent, by Cu-and Zn-chloride. In the presence of 50 μM Cd(II), all strains increased total thiol and glutathione production to 6, 11, and 21 μmoles · mg−1 dry mass, respectively. Cd(II) also induced the synthesis of one to several compounds reacting with 5,5′-dithio-bis-(2-nitrobenzoic acid). In order to identify buffer-soluble thiolic compounds other than cysteine, γ-EC and γ-ECG (glutathione) were analyzed and confirmed by mass spectrometry. No water soluble sulfides were detectable in any of the culture filtrates, but Cd(II) exposure at a concentration of 50 μM raised sulfide levels in the mycelia of two of the strains between 3 and 7-fold, Cu(II) and Zn(II) had no effect. Energy Dispersive X-ray-analysis (EDX) and Electron Spectrometry-Images (ES-I) of one strain revealed increased levels of Cu and Zn in the cytoplasm and even higher levels in vacuolar precipitates. Zn and Cu are accumulated in the vacuoles as polyphosphates, identified by Electron Energy Loss-Spectrometry (EELS). Cd was found only in the vacuoles.  相似文献

Abstract. Uptake of phytotoxic amounts of metal by higher plants or algae can result in inhibition of several enzymes, and in increase in activity (= induction) of others. Two mechanisms of enzyme inhibition predominate: (1) binding of the metal to sulphydryl groups, involved in the catalytic actionor structural integrity of enzymes, and (2) deficiency of an essential metal in metalloproteins or metal-protein complexes, eventually combined with substitution of the toxic metal for the deficient element. Metal accumulation in the cellular compartment of the enzyme is a prerequisite for enzyme inhibition in vivo. The induction of some enzymes is considered to play a significant role in the stress metabolism, induced by metal phytotoxicity. Peroxidase induction is likely to be related to oxidative reactions at the biomembrane; several enzymes of the intermediary metabolism might be stimulated to compensate for metal-sensitive photosynthetic reactions. The induction of enzymes and metal-specific changes in isoperoxidase pattern can be used as diagnostic criteria to evaluate the phytotoxicity of soils, contaminated by several metals. Lines for future research on metal phytotoxicity are proposed, involving the study of inhibition and induction of enzymes at the different cell membranes (especially the plasmamembrane) in vivo.  相似文献

The white rot fungus, Fomes lividus, was isolated from the logs of Shorea robusta in the Western Ghats region of Tamil Nadu, India. The fungus was tested for decolorization of azo dyes such as orange G (50 M) congo red (50 M) amido black 10B (25 M) and also for colour removal from dye industry effluents. The results revealed that the fungus could remove only 30.8% of orange G in the synthetic solution, whereas congo red and amido black 10B were removed by 74.0 and 98.9% respectively. A dye industry effluent was treated by the fungus in batch and continuous mode. In batch mode treatment, a maximum decolorization of 84.4% was achieved on day 4, and in continuous mode a maximum decolorization of 37.5% was obtained on day 5. The colour removal by the basidiomycete fungus might be due to adsorption of the dyes to the mycelial surface and metabolic breakdown. These results suggested that the batch mode treatment of Fomes lividus is one of the most efficient ways for colour removal in dye industry effluents.  相似文献

Tetrabromobisphenol A (TBBPA) degradation was investigated using white rot fungi and their oxidative enzymes. Strains of the Trametes, Pleurotus, Bjerkandera and Dichomitus genera eliminated almost 1 mM TBBPA within 4 days. Laccase, whose role in TBBPA degradation was demonstrated in fungal cultures, was applied to TBBPA degradation alone and in combination with cellobiose dehydrogenase from Sclerotium rolfsii. Purified laccase from Trametes versicolor degraded approximately 2 mM TBBPA within 5 h, while the addition of cellobiose dehydrogenase increased the degradation rate to almost 2.5 mM within 3 h. Laccase was used to prepare TBBPA metabolites 2,6-dibromo-4-(2-hydroxypropane-2-yl) phenol (1), 2,6-dibromo-4-(2-methoxypropane-2-yl) phenol (2) and 1-(3,5-dibromo-4-hydroxyphen-1-yl)-2,2',6,6'-tetrabromo-4,4'-isopropylidene diphenol (3). As compounds 1 and 3 were identical to the TBBPA metabolites prepared by using rat and human liver fractions (Zalko et al., 2006), laccase can provide a simple means of preparing these metabolites for toxicity studies. Products 1 and 2 exhibited estrogenic effects, unlike TBBPA, but lower cell toxicity.  相似文献

The concentrations of essential amino acids in three, undigested invertebrate diets collected from the Clark Fork River (CFR) for cutthroat trout were similar to each other, but were c. 25–75% less than Artemia that were exposed to a mixture of arsenic, copper, cadmium, lead and zinc in the laboratory. The Artemia diet appeared less palatable and the texture, quantity and appearance of the intestinal contents differed between fish fed the Artemia and CFR diets. The Pb% in the fluid fraction of the intestinal contents was greater for the Artemia (29%) than for the CFR diets (10–17%), and the Cu% in the amino acid plus metal fraction of the intestinal contents was greater for the Artemia (78%) than for two of the three CFR diets (67% and 70%). Intestinal contents of fish fed invertebrate diets collected from various sites on the Coeur d'Alene River (CDA), Idaho, were similar in texture, quantity, and appearance. For fish fed the CDA diets, differences in the distribution of metals among fractions of the digestive fluids appeared to be related to concentrations of metals in the invertebrate diets. Pb% was lowest of all metals in the fluid portion of the intestinal contents. However, >80% of all metals in the hind gut were associated with the particulate fraction where they may still be available for uptake through pinocytosis.  相似文献

Six mitosporic fungi belonging to five species (Aspergillus flavus var. flavus, Aspergillus ochraceus, Cladosporium cladosporioides, Penicillium glabrum and Penicillium verrucosum) were selected from a screening on 258 fungal strains as the most promising for their ability to remove 2 model dyes in solid conditions. Hence they were tested in liquid conditions for their ability to decolourise 3 model dyes and 9 industrial dyes widely used in the textile industry. The influence of the culture medium, particularly its carbon:nitrogen ratio, on biomass development and decolourisation capacity was considered. All the strains were able to grow in the dyed media and displayed various degrees of decolourisation according to the dye and culture medium. The decolourisation was due to biosorption phenomena. Aspergillus ochraceus performed the highest decolourisation yield being able to remove all dyes over 90%. This strain was also found very effective, both in the living and inactivated form, against simulated effluents that mimicked the recalcitrance of real wastewaters being composed of ten different dyes at high concentration (1,000 ppm), in saline solution.  相似文献

The nitrogen (N) content of wood is usually suboptimal for fungal colonization. During decomposition of wood, an increasing fraction of the N becomes incorporated into fungal mycelium. Between 5 and 50% of the N in wood-degrading mycelium may be incorporated into chitin. Chitinolytic enzymes render this N available for re-utilization. Here, the activities of chitinolytic enzymes produced by wood-rotting fungi during degradation of spruce (Picea abies) wood were quantified in situ using fluorogenic 4-methylumbelliferyl substrates. A new method was developed that enables spatial quantification of enzyme activities on solid surfaces. All of the three tested fungi produced endochitinases, chitobiosidases and N-acetylhexosaminidases during colonization of wood. N-acetylhexosaminidase activity, and in some cases also chitobiosidase and endochitinase activities, were higher during secondary overgrowth of another fungus than during primary colonization of noncolonized wood. The results suggest that wood-degrading fungi degrade their own cell walls as well as the hyphae of earlier colonizers. Recycling of cell wall material within single mycelia and between fungal individuals during succession may lead to retention of N within woody debris.  相似文献

The fruit bodies of 97 species of wood-rotting fungi, mainly of Polyporaceae and related families, were examined for the distribution of triterpenes and sterols. Triterpene acids of lanostane group were detected exclusively from the fungi causing brown-rot of woods, while sterols were found to occur commonly in both brown-rot and white-rot fungi. The most abundant sterol was found to be ergosta-7,22-dien-3β-ol. The presence and absence of the triterpene acids is discussed from the point of view of fungal phylogeny.  相似文献
