Discrete red patches of water were observed in South San FranciscoBay (USA) on 30 April 1993, and examination of live samplesshowed that this red tide was caused by surface accumulationsof the pigmented ciliate Mesodinium rubrum. Vertical profilesshowed strong salinity and temperature stratification in theupper 5 m, peak chlorophyll fluorescence in the upper meter,and differences in the small-scale density structure and fluorescencedistribution among red patches. Events preceding this Mesodiniumred tide included: (i) heavy precipitation and run-off, allowingfor strong salinity stratification; (ii) a spring diatom bloomwhere the chlorophyll a concentration reached 50 mg m–3;(ii) depletions of dissolved inorganic N and Si in the photiczone; and (iv) several days of rapid warming and stabilizationof the upper surface layer. The planktonic calanoid copepod, Sinocalanus doerrii, a nativeof the rivers of mainland China was found in 1978 in California'sSacramento-San Joaquin Estuary during routine plankton sampling.Previous plankton surveys in 1963 and from 1972 to the presentindicate that the introduction occurred a relativery short timebefore specimens were first caught. The most probable mode ofintroduction is ballast water from Japanese freighters previouslydocking in China. Two differently colored membrane preparations were separatedfrom the prochlorophyte, Prochloron sp., by mechanical disintegrationof the cells followed by sucrose density gradient centrifugation.An orange-colored preparation, containing zeaxanthin as themajor constituent pigment, seemed to comprise the cytoplasmicmembrane. The other green-colored membrane preparation, containingß-carotene and chlorophyll a and b as major pigmentconstituents, was identified as the thylakoid membrane. Salps were gently captured in a perspex chamber by a SCUBA diver,who then injected a suspension of 14C-labeled phytoplanktonand 3H-labeled bacteria into the chamber. After 0.5 h incubationin situ, filtering rates were estimated from incorporation ofthe two isotopes, and expressed as a function of each salp'ssize. Both bacteria and phytoplankton were grazed by Thalia,the latter at higher rates. Weight-specific grazing rates increasedwith increasing size of salp. Estimated hourly rations rangedfrom 1% of bodily C for small Thalia feeding only on phytoplanktonto 8% of bodily C for large Thalia feeding on all particulateorganic C. Dark-grown cells of a mutant strain of Chlorella regularis containedchlorophyll a and protochlorophyll, phytyl ester of protochlorophyllide.Under illumination, protochlorophyll was quantitatively anddirectly converted into chlorophyll a. The photoconversion wasdependent on light intensity and temperature and proceeded ina cell-free preparation. We exposed natural zooplankton communities to in situ levelsof sunlight for 3 days at different depths in two north temperatelakes: one oligotrophic and one eutrophic. Natural fluxes ofUV-B radiation (280–320 nm) were manipulated with cutofffilters (Mylar®). There was substantial mortality in someof the zooplankton exposed to UV-B in the oligotrophic lake,but not in the eutrophic lake. Reproduction of Diaptomus wassuppressed by UV-B down to 6 m in the oligotrophic lake. Thesedata suggest that natural levels of UV-B radiation in the oligotrophiclake may prevent some species of zooplankton from continuouslyexploiting the warm surface waters during summer stratification.In the more eutrophic lake, UV-B is a less important constraintin the vertical distribution of zooplankton. The growth rates of Appendicularia sicula, Fritillaria borealissargassi, Fritillaria haplostoma, Oikopleura dioica and Oikopleuralongicauda were determined from microcosms incubated in situat 23C in Jamaican waters. Experiments were conducted fromoligotrophic offshore waters, through mesotrophic Lime Cay andeutrophic Kingston Harbour in both natural and nutrient-enhancedphytoplankton communities. Length-weight relationships werecalculated for two of these species: O.longicauda log W=2.47log TL –6.10 and F.haplostoma log W=2.44 log TL –7.37,where weight (W) is in micrograms and trunk length (TL) is inmicrometres. Instantaneous growth rates averaged 1.7–2.5day–1 for the five species and were observed as high as3.3 day–1 These instantaneous rates are equivalent todaily specific growth rates averaging 4.6–11.4 and rangingup to 28. In larger genera, growth rates were related positivelyto picoplankton and nanoplankton concentration, and negativelyto the biomass of larvaceans, but in the smallest species growthwas unrelated to these factors. A series of 4-day manipulations of zooplankton biomass and nutrientavailability was performed in enclosures in three lakes to determinespecies-specific algal responses to herbivory and nutrient enrichment.Algal performance in enclosures was compared to the relationshipsbetween weekly algal growth rates and the zooplankton in situ.When in situ growth rates were significant functions of zooplanktonbiomass, the responses were generally consistent with responsesin the enclosure experiments. The importance of both nutrientsand zooplankton in mediating algal growth was demonstrated bynumerous observations: strong algal community response to enrichment,unimodal or positive responses of certain algal taxa to zooplanktonbiomass, differences in degree of nutrient limitation amongthe algal response types, lack of nutrient limitation of non-grazedalgal taxa and a preponderance of taxa with no net responseto increasing zooplankton biomass. Variation in the zooplanktoncommunity may be the largest source of variability in nutrientsupply rate during summer in stratified lakes, and causes substationalvariability in the algae. The recruitment of Aphanizomenon flos-aquae out of the sedimentswas measured in the shallow, hypereutrophic Agency Lake, Oregon.There were two main episodes of recruitment, the latter of whichaccounted for 8.2% of the water column population increase duringits main growth phase. Phosphorus (P) content was higher innewly recruited cells than in water column cells. Histiobalantium sp. was found regularly in the pelagic zoneof Lake Constance, FRG, over five annual cycles. Maxima of upto 6400 cells l–1 were recorded in late summer, with similarnumbers in the 0–8 and 8–20 m depth intervals. Onan annual average, the population accounted for 10–17%of the total biomass of planktonic ciliates. In the laboratory,Histiobalantium grew well on a diet of the cryptophyte Rhodomonassp. Maximum growth rates obtained in batch cultures were 0.21and 0.33 day–11 at 9 and 18°C, respectively. In situexperiments using diffusion chambers yielded positive growthrates in autumn and winter. The highest values recorded at theambient temperatures 5, 14 and 17°C were 0.17, 0.32 and0.40 day–1, respectively. Euglena gracilis was grown under controlled levels of iron suchthat chlorophyll synthesis was halted, although the cells continuedto grow at a normal rate. Chloroplasts isolated by zonal centrifugationfrom iron-deficient cells, even at a very early stage of deficiency,contained less than half the normal levels of RNA and ribosomes.Total RNA was unaffected. A reverse-phase h.p.l.c. technique was used to estimate theconcentration of chlorophyll b in phytoplankton cultures, fecalpellets of Calanus pacificus, and suspended paniculate matterfrom the Central North Pacific, Oregon coastal waters, and DabobBay (a temperate fjord in Puget Sound, WA, USA). The purposewas to assess the distribution of this pigment in the euphoticzone and its effect on the fluorometnc estimation of phaeopigments.Analyses of natural waters confirm high chlorophyll b concentrations(median mass ratio of b:a > 0.3) at the depth of the chlorophylla maximum in tropical waters while values for temperate planktonare relatively low (median mass ratio of chl b:a = 0.05) andpatchy. Zooplankton fecal pellets showed a significant enrichmentin chlorophyll b, suggesting grazing as a mechanism to explainhigh concentrations of this pigment at the bottom of the euphoticzone. It is estimated that the presence of chlorophyll b couldcause an average overestimation of phaeopigment concentrationby the fluorometnc technique of 38% between 0 and 200 m in theCentral North Pacific. Fine-scale vertical (5 – 40 m) and horizontal (50 –500 m) patterns of temperature, chlorophyll and abundance ofzooplankton species were sampled with a pump filtration systemin the surface waters offshore of San Diego in May and October,1978. Intense and consistent patterns were most apparent invertical profiles. Herbivorous zooplankton were more consistentlyassociated with the estimated primary productivity maximum thanwith the deeper chlorophyll maximum layer, which representeda phytoplankton biomass maximum. Predators were positively correlatedwith abundant potential prey species. Variations in body lengthwith depth suggest that these fine-scale patterns were sufficientlystable to influence zooplankton growth. Etiolated oat protoplasts were treated with dibutyryl cAMP tostudy possible function of cAMP in the development by measuringthe protoplast swelling. The mean diameter of protoplasts inthe absence of any chemical treatment was 33.58±1.26(SE) µm, which increased to 36.96±0.86 µmin the presence of 100 µM dibutyryl cAMP. Prostacyclin,a potent activator of adenyl cyclase, also showed a significantswelling effect (diameter 38.01±0.98 µm). Cladocerans from three lakes in northwest Iowa were examinedfor attached diatoms over a period of 1 year to assess the seasonality,substrate preference and attachment site specificities of epizoicdiatoms. Over 90% of the cladocerans had attached diatoms immediatelyfollowing ice thaw, when the lakes were not yet thermally stratified,surface water temperatures were low(10–15°C)and watertransparency was at its annual high. Mean maximum density ofdiatoms on cladocerans averaged among lakes at this time was3000 cells animal–1. The dominant epizoic diatom was Synedracyclopum Brutschy, which was found most often on large, pelagicDaphnia pulicaria and Daphnia galeata mendotae. Heavily infestedanimals had diatoms concentrated on the second antennae andanterior ventral margin of the carapace. Animals with <5epizoic diatoms were more likely to have cells attached to thepostabdomen or tail spine. Effects of chloramphenicol, cycloheximide, puromycin and ethionineon the light-independent and subsequent light-dependent processesof chlorophyll formation in "glucose-bleached" cells of Chlorellaprotothecoides were studied. These substances, except puromycin,strongly suppressed different phases of chlorophyll formation.Ethionine most strongly suppressed the light-independent phaseand chloramphenicol an early, relatively short process in thelight-dependent phase of chlorophyll formation. Cycloheximideseverely suppressed all phases of chlorophyll formation. The present study characterized the rate of production of extracellularlyreleased organic carbon (ROC) by phytoplankton, its molecularweight distribution, subsequent utilization and transformationby bacterioplankton in situ. Primary production rate of phytoplanktonwas high during the study due to continuous blooms of smalldinoflagellates and ranged from 59.8 to 298.7µg CI–1h–1. The rate of organic carbon release varied from 1.3to 123.7 µg CI–1 h–1 and constituted from4.0 to 68.9% of the total carbon fixed in photosynthesis. TheROC was fractionated on molecular weight (MW) basis. A low MWfraction less than 500 daltons (18.5% of ROC), a fraction ofMW 10 000–30 000 daltons (30% of ROC), and high MW fractionof > 300 000 daltons (15.4% of ROC), were the most dominantin ROC. Bacterioplankton utilized a significant portion of ROC,ranging from 18 to 77%. Part of the utilized ROC incorporatedby the bacterioplankton (31 – 56%), and the remainderwas respired (mineralized). ROC not utilized by bacteria wascomposed of high MW compounds. Lemna paucicostata 6746, a short-day plant, flowers in agedHutner's medium even under continuous light, when the endogenousnitrogen level decreases to below 1.6 µmg fr wt. At thesenitrogen levels, daylength-independent flowering of the plantcan be induced even in fresh Hutner's medium.
