Microsatellite DNA based analysis of the pattern of genetic diversity among three coastal and five freshwater populations of pikeperch Sander lucioperca in the northern part of the Baltic Sea drainage basin indicated marked genetic differentiation between the coastal and lake populations. The F(st) between these population groups was as high as 0.25 and R(st) =0.32. In general, the lake populations showed higher genetic diversity than the coastal ones. In terms of genetic distance, the three coastal populations (Vanhankaupunginlahti, V?stanfj?rd and Taivassalo) grouped tightly together. The freshwater samples formed a looser group, in which the northern Lake Kemij?rvi showed greater distance from the southern lakes than these did from each other. The two lake populations originally established through stockings (Lakes Painio and Averia) grouped near to their source population of Lake Lohjanj?rvi and their diversity level was nearly the same. Safeguarding the unique Baltic coastal populations of S. lucioperca against gene flow from increasing hatchery releases using freshwater S. lucioperca should be a high management priority. 相似文献

A morphometric truss network system and meristic counts were used to investigate the hypothesis of population fragmentation of pikeperch Sander lucioperca among four major fishing areas in the Caspian Sea drainage. Univariate analysis of variance showed significant differences between the means of the four groups for 20 out of 32 standardized morphometric measurements (27 truss, five traditional characters) and six out of nine meristic counts. The first two canonical discriminant functions accounted for 93 and 92.3% of the total variability in morphometric and meristic characteristics, respectively. A dendogram derived from cluster analysis of Mahalanobis distances and a discriminant analysis among the groups confirmed three distinct populations in these regions, while there was a relatively high degree of overlap between all locations when using meristic characters. The most relevant morphometric characteristics were the measured distances on body heights and the caudal peduncle area. This study indicated that the morphometric truss network system was more adequate than meristic characters for separation among the populations. 相似文献

To investigate the relative importance of homogenizing factors, such as gene flow, and diversifying factors, such as drift, genetic variation in pikeperch ( Sander lucioperca ) in two Fennoscandian regions (North and South) was analysed with microsatellites. Allelic richness and the degree of differentiation were significantly higher in the North ( F ST = 0·20) than in the South ( F ST = 0·064). In northern areas, assignments of genotypes were almost exclusively to the population of origin, but in southern areas, the proportion of correct assignments was significantly lower. Most samples exhibited significant heterozygote deficits, and the level of relatedness was higher than expected from randomness. These combined results suggest that there has been more gene flow between populations in southern areas than in northern areas, where the importance of genetic drift has been greater. Effective population sizes were small ( c. 100) and did not differ between areas. The effect of a common history appears minor, and thus processes such as genetic drift and gene flow have been more influential in shaping the patterns of genetic diversity in this species. 相似文献

Experiments were carried out on sperm cryopreservation of two European percid fish species, the pikeperch (Sander lucioperca) and the Volga pikeperch (S. volgensis). Two experiments were conducted on pikeperch sperm. In the first, the effects of three extenders (Glucose, KCl, Sucrose) and two cryoprotectants (dimethyl-sulfoxide: DMSO, methanol: MeOH) were tested on motility and fertilization. In the second, the effects of two dilution ratios (1 : 1, 1: 9) and two cryoprotectants (DMSO, MeOH) on hatching were investigated. In the experiment on Volga pikeperch the suitability of using cryopreservation for fertilization was investigated. In the first experiment on pikeperch the highest post-thaw motility (28 +/- 21%) and fertilization rate (43 +/- 12%) was found with DMSO as cryoprotectant in combination with Glucose extender. In the second, the highest hatch rate (41 +/- 22%) was observed with MeOH as cryoprotectant and 1 : 1 sperm dilution ratio, however no significant difference was found among the results. In the experiment on Volga pikeperch hatch rates with cryopreserved sperm (60 +/- 2%) did not significantly differ from the control (60 +/- 6%). Contamination of sperm with urine seems to be a key problem in the success of sperm cryopreservation of these species. 相似文献

A comparative proteomic approach was used to assess the protein expression profile in the liver of 34 days old pikeperch larvae fed from day 10 post hatching, with three isoproteic and isolipidic formulated diets varying by their phospholipid (PL) contents (% dry diet weight): 1.4% (PL1), 4.7% (PL5) and 9.5% (PL9). Using 2D-DIGE minimal labelling of liver extracts, we were able to show 56 protein spots with a differential intensity (p < 0.05) depending on the dietary PL content. Among these spots, 11 proteins were unambiguously identified using nanoLC-MS/MS tandem mass spectrometry. In the PL9 larvae, our results indicate that the glycolytic pathway could be down-regulated due to the under-expression of the fructose biphosphate aldolase B and the phosphoglucomutase 1. Meanwhile, propionyl coenzyme A carboxylase (a gluconeogenic enzyme) was under-expressed. In addition, another gluconeogenic and lipogenic enzyme, pyruvate carboxylase, was identified in 3 different spots as being under-expressed in fish fed with the intermediate PL level (PL5). A high PL content increased the expression of sarcosine dehydrogenase, an enzyme involved in methionine metabolism, along with vinculin, a structural protein. Moreover, several stress proteins (glutathione S-transferase M, glucose regulated protein 75 and peroxiredoxin-1) were modulated in response to the dietary PL level and fatty acid composition. In the larvae fed with the lowest dietary PL content (PL1), over-expression of both GSTM and GRP75 might indicate a cellular stress in this experimental treatment, while the under-expression of Prx1 might indicate a lower defence against oxidative stress. In conclusion, this nutriproteomic approach showed significant modifications of protein expression in the liver of pikeperch larvae fed different PL contents, highlighting the importance of these nutrients and their influence on metabolism processes and on stress response. 相似文献

The effect of ambient salinity changes (0.9, 6 and 12 psu) on the levels of dissolved ammonia (DA), ninhydrin positive substances (NPS), trimethylamine (TMA) and trimethylamine oxide (TMAO) in the blood and tissue of medium-acclimated Sander lucioperca L. (also Stizostedion lucioperca) were investigated. In freshwater, blood and tissue total free amino acid levels (measured as NPS) were 3.62 mM and 60.61 mM, respectively. The NPS content increased significantly (P<0.05) in the tissue and blood on acclimation to 6 and 12 psu salinities. The mass-specific tissue TMAO concentration of pikeperch acclimated to normal freshwater was 0.413+/-0.084 micromol TMAO g(-1). Results reveal that TMAO levels are positively influenced by the external salinity medium where significant differences in mean levels occurred between the groups (P<0.05). The calculated p[NH(3)] and [NH(4)(+)] gradients reveal that the [NH(3)] gradient was consistently low (cf. the [NH(4)(+)] gradient). The gradient of p[NH(3)] decreased with the medium increased salinities. The results suggest that freshwater pikeperch may be able to resist salinity changes by manipulation of nitrogen metabolism. Free amino acids and TMAO are involved in mediating response to salinity exposure in freshwater pikeperch. 相似文献

In order to facilitate studies on the genetic structure of wild populations as well as to monitor genetic changes in cultured stocks, nine polymorphic microsatellite loci were isolated from pike-perch (Sander lucioperca). Single loci allele numbers varied between two (loci MSL-3 and MSL-7) and six (loci MSL-1 and MSL-2), and observed heterozygosities ranged from 0.36 (locus MSL-2) to 1.00 (locus MSL-9) in a test panel of 25 individuals. Only one pair of loci (MSL-5 and MSL-8) displayed significant linkage disequilibrium after sequential Bonferroni corrections. Hardy-Weinberg tests revealed significant excesses of heterozygotes at three loci (MSL-1, MSL-7, and MSL-9). 相似文献

The genetic polymorphism of the pike perch Sander lucioperca, sampled on the territory of the Republic of Kazakhstan (Lake Balkhash, Urals River, Syrdarya River and Shardara water reservoir, North Aral Sea) and in the Volga River, and the bersh Sandler volgensis (Lake Balkhash, Volga River) populations was examined at six microsatellite loci and the mitochondrial DNA cytochrome b gene. All examined loci were successfully amplified in both species and produced quite different allele profiles, which allows them to be used for accurate species identification of pike perch and bersh. At the same time, neither nuclear nor mitochondrial markers revealed statistically significant differentiation between the examined pike perch samples, excluding some isolation of the population from Volga River. 相似文献

The spawning sites of pikeperch were mapped using artificial spawning nests in the clay‐turbid Lake Tuusulanjärvi in southern Finland to protect the most important spawning areas. Altogether 272 artificial spawning nests were set before actual spawning on potential spawning grounds. The first observations of eggs were made on 26 May at a surface water temperature of 12°C. No new eggs were found after 10 June at water temperature of 16°C. The nests were removed on 18 June when the larvae had already hatched. The study demonstrated that artificial spawning nests could be used to evaluate both duration and spatial dimensions of spawning of pikeperch. 相似文献

The objective of this study was to control the reproductive cycle of pikeperch (Sander lucioperca) through determining the effects of different photoperiods and handling stress on the reproduction quality, timing and quality of spawning, fertilization, sex steroids, and cortisol concentrations. In this study, 72 pikeperch broodstocks with an average weight of 1367 ± 55.3 g were exposed to different photoperiods including constant light (24L:0D), constant darkness (0L:24D), and 12 h of light, 12 h of darkness (12L:12D) for 40 days. Half of the broodstocks of each photoperiod treatment were exposed to handling stress at a specific time of the day. Applying different photoperiods caused changes in the timing of broodstocks' spawning, so that fish under 24L:0D spawned earlier than those of other photoperiods, and stressed fish of the 0L:24D photoperiod had a delayed spawning compared to others. Also, the spawning of the broodstocks at different photoperiods which were exposed to handling stress was either delayed or did not occur at all. The highest and lowest spawnings were observed in the morning and at night, respectively. Fertilization percentage, number of eggs per gram, sex steroids including estradiol, progesterone, and testosterone, as well as cortisol and calcium concentrations did not show any significant difference in different photoperiods and handling stress. In stressed males of the 24L:0D photoperiod, there only was a significant decrease of testosterone concentration compared to the beginning of the experiment. Results indicated that the spawning performance of pikeperch broodstocks could be considerably stimulated using an effective photoperiod. Similarly, pikeperch broodstocks in culture systems are usually affected by handling stress, and this stress could lead to a poor reproductive performance and inhibition of spawning. 相似文献

The behaviour of radio-tagged adult pikeperch (Stizostedion lucioperca (L.)) from two areas in the Danish River Gudenaa were recorded prior to, and during the spawning period. Eight of 13 tagged fish in the lower reaches of the river were located throughout the whole study. Five of these fish moved upstream to various sites in the river prior to spawning, which occurred from late April to June. The three remaining fish moved to the fjord. These movements were interpreted as a spawning migration, and it is suggested that the lower reaches of the River Gudenaa constitute an over-wintering area for pikeperch, which use different spawning areas. Ten pikeperch caught just downstream of an impassable hydropower plant were radio-tagged and translocated upstream of the dam to a reservoir. Within a week, half of the fish moved to a lake situated more than 30 km upstream the reservoir. This behaviour is hypothesised to be a homing response. The study reveals that the pikeperch is a highly mobile species with a complicated migration pattern, even in relatively small river systems. 相似文献

The European pond turtle, Emys orbicularis, inhabits a wide distribution area in the western Palaearctic. Polish populations of pond turtle represent the nominotypical
subspecies Emys orbicularis orbicularis. The mitochondrial DNA haplotype (cytb gene) variation among 131 turtles from 26 locations in five regions of Poland was investigated. Five haplotypes belonging
to three distinct lineages were identified. Two clades (I and II) were represented by two haplotypes each, while the other
clade (IV) was represented by one haplotype. Three haplotypes were reported for the first time in E. orbicularis. The eastern part of Poland is inhabited exclusively by turtles bearing haplotype Ia. The remaining four sequence variants
were recorded in western Poland where only the IIb haplotype is considered endemic. The distribution of the other haplotypes
in western Poland could thus reflect past introductions or accidental releases. The authors regarded the two locations (Drzeczkowo

Pikeperch and perch perform seasonal migrations between the Curonian Lagoon and the coastal waters of the Baltic Sea. The Curonian Lagoon is a freshwater basin, while salinity in the coastal waters varies between of 4.9–6.8 psu. In the Curonian Lagoon water temperature is generally higher than in the coastal waters. Field studies of growth and condition characteristics of pikeperch and perch were carried out in these water bodies with the aim to estimate growth differences of the two fish species under different salinity and temperature conditions. Additionally, an experimental study of the impact of salinity on the growth of perch young-of-the-year (YOY) was performed to test the hypothesis that a brackish environment positively influences percids. Field observations revealed that body length, condition factor, fatness coefficient and fat content in muscles were significantly higher in individuals inhabiting the cooler, brackish waters of the Baltic Sea than in individuals inhabiting the Curonian Lagoon. A positive effect of low salinity on growth was also established in the experimental study. Hence, the study results suggest that under certain temperature conditions, brackish waters beneficially affect the growth of pikeperch and perch. 相似文献

The present study examined the effects of feeding pike perch larvae Artemia, enriched with either docosahexanoic acid (DHA), arachidonic acid (ARA), oleic acid (OA), olive oil (OO) or a commercial enrichment DHA Selco (DS) on tissue lipid deposition, stress tolerance, growth and development, and metabolic rate. There was higher tissue retention of ARA than DHA at comparable inclusion levels. No differences were observed between diets on the percentage contribution of ARA or DHA to the fatty acid profile of tissues (head and trunk). Total fatty acid content (mgg(-1)) was significantly higher in the head, reflecting its high content of neural tissue. Observations on larval erratic behaviour and mortality following exposure to salinity stress suggested that high inclusions levels of DHA had an alleviating effect, while ARA did not. Particularly larval groups reared for 16 days on diets enriched with OO and OA had mortality rates approaching 100% within two hours. Interestingly, this tendency, although not as pronounced, was also apparent in juvenile fish after 120 days of rearing on a common diet. Standard metabolic rate in larvae on an OO enriched diet was significantly elevated, but otherwise no groups had significant changes to their respiratory physiology. In addition to increased stress challenge sensitivity, early feeding with OA had long term impact on pike perch neural development indicated by a smaller brain size in juvenile fish. In conclusion, lack of DHA in the diet of pikeperch larvae suggests that this long chain polyunsaturated fatty acid is involved in processes that increase stress tolerance and that lack of dietary DHA in early larval stage caused increased stress sensitivity and long-term impaired neural development, while it does not appear to affect metabolic rate at rest. 相似文献

The population of pikeperch in L. Peipsi was studied with respect to average number per haul, length distribution of fishes in experimental catches, condition factor (according to Fulton) and length-weight relationship in autumn 1993–1995.The relatively good growth rate and condition of pikeperch in the lake indicates that the waterbody is suitable for this fish. However, the length and age distribution of pikeperch in different years as well as the decreasing total number of fishes in catches demonstrate that the population of pikeperch is suppressed due to overfishing during the last years. 相似文献
