Diet of juvenile mutton Lutjanus analis, dog Lutjanus jocu and lane Lutjanus synagris snappers were studied in the tropical Brazilian estuarine system of the Piraquê‐açú and Piraquê‐mirim Rivers to determine how these species share the resources in this restricted space. The three species prey principally upon Peracarida (L. synagris: relative importance index IRIc = 29%), Natantia (L. analis and L. synagris: IRIc = 39 and 38%, respectively), Reptantia (L. analis and L. jocu: IRIc = 28 and 43%, respectively) and Teleostei (L. jocu: IRIc = 24%). The three species use estuaries as nursery habitats but food overlap was not biologically significant due to a combination of interspecific differences in size, spatial distribution, microhabitat preferences and seasonal patterns of abundance and prey choice. Large marine protected areas incorporating essential habitats for all life stages are suggested to be the best tool for the management of these economically important species.  相似文献

This is a paper about how men from Panapompom, an island in Milne Bay Province of Papua New Guinea (PNG), understand how they relate to white people and imagine the future. Until recently, men from Panapompom understood themselves to be engaged in a project of ‘development’, in which they would become more and more similar to white people. This was a desirable future. However, changes in the way Panapompom men work for money have resulted in a very different imagination of the future—one in which Panapompom people are not getting whiter, but blacker, and hence more and more excluded from the lives to which they aspire. Men now dive for bêche‐de‐mer, work which they regard as being particularly hard and dangerous. Diving has profound effects on the skin, blackening and hardening it, leading Panapompom men to liken themselves to the machines that create the wealth that white people use. These ‘mechanising’ effects that diving has on the black body lead men to see white people as the sole beneficiaries of the bêche‐de‐mer industry, and black people as mere tools or extensions. For bêche‐de‐mer divers, value and desired forms of life are lodged in Australia, Europe or America, while they find themselves excluded from this future by their growing blackness.  相似文献

Nitrogen‐fixing bacteria (rhizobia) form a nodule symbiosis with legumes, but also induce certain effects on non‐host plants. Here, we used a split‐root system of barley to examine whether inoculation with Rhizobium sp. strain NGR234 on one side of a split‐root system systemically affects arbuscular mycorrhizal (AM) root colonization on the other side. Mutant strains of NGR234 deficient in Nod factor production (strain NGRΔnodABC), perception of flavonoids (strain NGRΔnodD1) and secretion of type 3 effector proteins (strain NGRΩrhcN) were included in this study. Inoculation resulted in a systemic reduction of AM root colonization with all tested strains. However, the suppressive effect of strain NGRΩrhcN was less pronounced. Moreover, levels of salicylic acid, an endogenous molecule related to plant defense, were increased in roots challenged with rhizobia. These data indicate that barley roots perceived NGR234 and that a systemic regulatory mechanism of AM root colonization was activated. The suppressive effect appears to be Nod factor independent, but enhanced by type 3 effector proteins of NGR234.  相似文献

The paleoenvironmental setting in which the Ediacara Biota lived, died, and was preserved in the eponymous Ediacara Member of the Rawnsley Quartzite of South Australia is an issue of long‐standing interest and recent debate. Over the past few decades, interpretations have ranged from deep marine to shallow marine to terrestrial. One of the key features invoked by adherents of the terrestrial paleoenvironment hypothesis is the presence of iron oxide coatings, inferred to represent the upper horizons of paleosols, along fossiliferous sandstone beds of the Ediacara Member. We find that these surficial oxides are characterized by (234U/238U) values which are not in secular equilibrium, indicating extensive fluid‐rich alteration of these surfaces within the past approximately 2 million years. Specifically, the oxide coatings are characterized by (234U/238U) values >1, indicating interaction with high‐(234U/238U) fluids derived from alpha‐recoil discharge. These oxides are also characterized by light “stable” δ238/235U values, consistent with a groundwater U source. These U isotope data thus corroborate sedimentological observations that ferric oxides along fossiliferous surfaces of the Ediacara Member consist of surficial, non‐bedform‐parallel staining, and sharply irregular patches, strongly reflecting post‐depositional, late‐stage processes. Therefore, both sedimentological and geochemical evidence indicate that Ediacara iron oxides do not reflect synsedimentary ferruginization and that the presence of iron oxides cannot be used to either invoke a terrestrial paleoenvironmental setting for or reconstruct the taphonomic pathways responsible for preservation of the Ediacara Biota. These findings demonstrate that careful assessment of paleoenvironmental parameters is essential to the reconstruction of the habitat of the Ediacara Biota and the factors that led to the fossilization of these early complex ecosystems.  相似文献

The filamentous fungus Aspergillus fumigatus has become the most important airborne fungal pathogen causing life‐threatening infections in immunosuppressed patients. We established a 2‐D reference map for A. fumigatus. Using MALDI‐TOF‐MS/MS, we identified 381 spots representing 334 proteins. Proteins involved in cellular metabolism, protein synthesis, transport processes and cell cycle were most abundant. Furthermore, we established a protocol for the isolation of mitochondria of A. fumigatus and developed a mitochondrial proteome reference map. 147 proteins represented by 234 spots were identified.  相似文献

Recôncavo Baiano is an area favourable for the occurrence of citrus greasy spot (CGS) (Mycosphaerella citri), but there has been no study of this pathosystem in Brazil. This work aimed to characterise the temporal patterns of CGS‐induced defoliation in sweet orange cultivars ‘Bahia’ (Washington Navel) and ‘Pêra’. Temperature, rainfall and relative humidity were recorded, as well as weekly defoliation (fallen leaves/canopy m2 or m3). Considering the mean of fallen leaves per canopy m2, and mean canopy area, the total annual defoliation was estimated to be around 32 000 leaves per plant for ‘Bahia’ and 18 500 for ‘Pêra’ sweet orange. Spectral density analysis showed that defoliation has a 5‐week‐long main cycle for both cultivars. The proportion of symptomatic fallen leaves was never below 0.97. The monthly number of fallen leaves per canopy area was positively correlated with the mean CGS incidence on leaves. Defoliation was significant, resulting in a low leaf density throughout the year. Many defoliation cycles and the very high proportion of symptomatic fallen leaves assure a constant inoculum supply. Based on these results, CGS cannot be considered a minor disease, at least in Recôncavo Baiano.  相似文献

The detection of novel stimuli is a memory‐dependent process. The presented stimulus has to be compared with memory contents to judge its novelty. In addition, the novelty of stimuli activates attention‐related processes that facilitate memory formation. To determine the involvement of limbic and neocortical brain structures in novelty detection, we exposed mice to a novel gustatory stimulus (0.5% saccharin) added to their drinking fluid. We then compared the novelty‐induced expression of the two immediate‐early genes (IEGs) c‐fos and arg 3.1, with their expression in mice familiarized with the same stimulus or mice not exposed to that stimulus. Exposure to taste novelty increased expression of c‐fos and arg 3.1 mRNA in the cingulate cortex and deep layers of the parietal cortex. In addition, c‐fos mRNA expression was increased in the amygdala and arg 3.1 mRNA was increased in the dentate gyrus. Expression of c‐fos and arg 3.1 was elevated 30 min after the exposure to novelty. For arg 3.1, a second peak of expression was found 4.5 h after presentation of the novel stimulus. Our results indicate that the amygdala, the dentate gyrus, and the cingulate and parietal cortices may be involved in novelty detection and associated cognitive events, and suggest that c‐fos and arg 3.1 play distinct roles in these processes. © 1999 John Wiley & Sons, Inc. Abstract

Neuropeptide Y (NPY) recognition by the human neuroblastoma cell lines SiMa, Kelly, SH‐SY5Y, CHP‐234, and MHH‐NB‐11 was analyzed in radioactive binding assays using tritiated NPY. For the cell lines CHP‐234 and MHH‐NB‐11 binding of [3H]propionyl‐NPY was observed with Kd‐values of 0.64 ± 0.07 nM and 0.53 ± 0.12 nM, respectively, determined by saturation analysis with non‐linear regression. The receptor subtype was determined by competition analysis using the subtype selective NPY analogues [Leu31, Pro34]‐NPY (NPY‐Y1, NPY‐Y5), [Ahx5‐24]‐NPY (NPY‐Y2), [Ala31, Aib32]‐NPY (NPY‐Y5), NPY [3‐36] (NPY‐Y2, NPY‐Y5), and NPY [13‐36] (NPY‐Y2). Both cell lines, CHP‐234 and MHH‐NB‐11, the latter one being characterized for NPY receptors for the first time, showed exclusive expression of NPY‐Y2 receptors. In both cell lines binding of NPY induced signal transduction, which was monitored as reduction of forskolin‐induced cAMP production in an ELISA.  相似文献

The effects of global change on biological systems and functioning are already measurable, but how ecological interactions are being altered is poorly understood. Ecosystem resilience is strengthened by ecological functionality, which depends on trophic interactions between key species and resilience generated through biogenic buffering. Climate‐driven alterations to coral reef metabolism, structural complexity and biodiversity are well documented, but the feedbacks between ocean change and trophic interactions of non‐coral invertebrates are understudied. Sea cucumbers, some of the largest benthic inhabitants of tropical lagoon systems, can influence diel changes in reef carbonate dynamics. Whether they have the potential to exacerbate or buffer ocean acidification over diel cycles depends on their relative production of total alkalinity (AT) through the dissolution of ingested calcium carbonate (CaCO3) sediments and release of dissolved inorganic carbon (CT) through respiration and trophic interactions. In this study, the potential for the sea cucumber, Stichopus herrmanni, a bêche‐de‐mer (fished) species listed as vulnerable to extinction, to buffer the impacts of ocean acidification on reef carbonate chemistry was investigated in lagoon sediment mesocosms across diel cycles. Stichopus herrmanni directly reduced the abundance of meiofauna and benthic primary producers through its deposit‐feeding activity under present‐day and near‐future pCO2. These changes in benthic community structure, as well as AT (sediment dissolution) and CT (respiration) production by S. herrmanni, played a significant role in modifying seawater carbonate dynamics night and day. This previously unappreciated role of tropical sea cucumbers, in support of ecosystem resilience in the face of global change, is an important consideration with respect to the bêche‐de‐mer trade to ensure sea cucumber populations are sustained in a future ocean.  相似文献

Series of agonists and antagonists were examined for their actions on a Bombyx moriα‐adrenergic‐like octopamine receptor (OAR) stably expressed in HEK‐293 cells. The rank order of potency of the agonists was clonidine>naphazoline>tolazoline in Ca2+ mobilization assays, and that of the antagonists was chlorpromazine>yohimbine. These findings suggest that the B. mori OAR is more closely related to the class‐1 OAR in the intact tissue than to the other classes. N′‐(4‐Chloro‐o‐tolyl)‐N‐methylformamidine (DMCDM) and 2‐(2,6‐diethylphenylimino)imidazolidine (NC‐5) elevated the intracellular calcium concentration ([Ca2+]i) with EC50s of 92.8 µM and 15.2 nM, respectively. DMCDM and NC‐5 led to increases in intracellular cAMP concentration ([cAMP]i) with EC50s of 234 nM and 125 nM, respectively. The difference in DMCDM potencies between the cAMP and Ca2+ assays might be due to “functional selectivity.” The Ca2+ and cAMP assay results for DMCDM suggest that the elevation of [cAMP]i, but not that of [Ca2+]i, might account for the insecticidal effect of formamidine insecticides. © 2009 Wiley Periodicals, Inc.  相似文献

Red‐bellied piranhas (Pygocentrus nattereri) are widely caught with different intensities throughout the region of Solimões‐Amazonas River by local fishermen. Thus, the management of this resource is performed in the absence of any information on its genetic stock. P. nattereri is a voracious predator and widely distributed in the Neotropical region, and it is found in other regions of American continent. However, information about genetic variability and structure of wild populations of red‐bellied piranha is unavailable. Here, we describe the levels of genetic diversity and genetic structure of red‐bellied piranha populations collected at different locations of Solimões‐Amazonas River system. We collected 234 red‐bellied piranhas and analyzed throughout eight microsatellite markers. We identified high genetic diversity within populations, although the populations of lakes ANA, ARA, and MAR have shown some decrease in their genetic variability, indicating overfishing at these communities. Was identified the existence of two biological populations when the analysis was taken altogether at the lakes of Solimões‐Amazonas River system, with significant genetic differentiation between them. The red‐bellied piranha populations presented limited gene flow between two groups of populations, which were explained by geographical distance between these lakes. However, high level of gene flow was observed between the lakes within of the biological populations. We have identified high divergence between the Catalão subpopulation and all other subpopulations. We suggest the creation of sustainable reserve for lakes near the city of Manaus to better manage and protect this species, whose populations suffer from both extractive and sport fishing.  相似文献

Striatin, an intraneuronal, calmodulin‐binding protein addressed to dendrites and spines, is expressed in the motor system, particularly the striatum and motoneurons. Striatin contains a high number of domains mediating protein–protein interactions, suggesting a role within a dendritic Ca2+‐signaling pathway. Here, we explored the hypothesis of a direct role of striatin in the motor control of behaving rats, by using an antisense strategy based on oligodeoxynucleotides (ODN). Rats were treated by intracerebroventricular infusion of a striatin antisense ODN (A‐ODN) or mismatch ODN (M‐ODN) delivered by osmotic pumps over 6 days. A significant decrease in the nocturnal locomotor activity of A‐ODN–treated rats was observed after 5 days of treatment. Hypomotricity was correlated with a 60% decrease in striatin content of the striata of A‐ODN–treated rats sacrificed on day 6. Striatin thus plays a role in the control of motor function. To approach the cellular mechanisms in which striatin is involved, striatin down‐regulation was studied in a comparatively simpler model: purified rat spinal motoneurons which retain their polarity in culture. Treatment of cells by the striatin A‐ODN resulted in the impairement of the growth of dendrites but not axon. The decrease in dendritic growth paralleled the loss of striatin. This model allows analysis of the molecular basis of striatin function in the dynamic changes occurring in growing dendrites, and offers clues to unravel its function within spines. © 1999 John Wiley & Sons, Inc. Nine diazotrophic bacteria were isolated from surface-sterilized roots and culms of wheat variety Malviya-234, which is grown with very low or no inputs of nitrogen fertilizer. Out of the nine bacteria, four showed indole acetic acid (IAA) production, and five were positive for P solubilization. One isolate, WM234C-3, showed appreciable level of nitrogenase activity, IAA production, and P solubilization ability, and was further characterized with a view to exploiting its plant growth promoting activity. Based on 16S rDNA sequence analysis, this isolate was identified as Achromobacter xylosoxidans. Diazotrophic nature of this particular isolate was confirmed by Western blot analysis of dinitrogenase reductase and amplification of nifH. Analysis of the nifH sequence showed close homology with typical diazotrophic bacteria. Endophytic nature and cross-infection ability of WM234C-3 were tested by molecular tagging with gusA fused to a constitutive promoter followed by inoculation onto rice seedlings in axenic conditions. At 21 days after inoculation, the roots showed blue staining, the most intense color being at the emergence of lateral roots and root tips. Microscopic observation confirmed colonization of gus-tagged WM234C-3 in the intercellular spaces of cortical as well as vascular zones of roots. Inoculation of gus-tagged WM234C-3 to rice plants resulted in significant increase in root/shoot length, fresh weight, and chlorophyll a content. Plant growth promoting features coupled with cross-infection ability suggest that this endophytic bacterium may be exploited as agricultural agent for various crops after a thorough and critical pathogenicity test.  相似文献

Epithelial–mesenchymal transition (EMT), via activation of Wnt signaling, is prevailing in embryogenesis, but postnatally it only occurs in pathological processes, such as in tissue fibrosis and tumor metastasis. Our prior studies led us to speculate that EMT might be involved in the loss of limbal epithelial stem cells in explant cultures. To examine this hypothesis, we successfully grew murine corneal/limbal epithelial progenitors by prolonging the culture time and by seeding at a low density in a serum‐free medium. Single cell‐derived clonal growth was accompanied by a gradient of Wnt signaling activity, from the center to the periphery, marked by a centrifugal loss of E‐cadherin and β‐catenin from intercellular junctions, coupled with nuclear translocation of β‐catenin and LEF‐1. Large‐colony‐forming efficiency at central location of colony was higher than peripheral location. Importantly, there was also progressive centrifugal differentiation, with positive K14 keratin expression and the loss of p63 and PCNA nuclear staining, and irreversible EMT, evidenced by cytoplasmic expression of α‐SMA and nuclear localization of S100A4; and by nuclear translocation of Smad4. Furthermore, cytoplasmic expression of α‐SMA was promoted by high‐density cultures and their conditioned media, which contained cell density‐dependent levels of TGF‐β1, TGF‐β2, GM‐CSF, and IL‐1α. Exogenous TGF‐β1 induced α‐SMA positive cells in a low‐density culture, while TGF‐β1 neutralizing antibody partially inhibited α‐SMA expression in a high‐density culture. Collectively, these results indicate that irreversible EMT emerges in the periphery of clonal expansion where differentiation and senescence of murine corneal/limbal epithelial progenitors occurs as a result of Smad‐mediated TGF‐β‐signaling. 甘孜藏族自治州(以下简称甘孜州)生态状况良好而经济相对滞后,评估甘孜州生态系统服务功能及其价值,对甘孜州社会经济的可持续发展具有一定的促进作用。通过核算甘孜州的生态系统生产总值(GEP),包括对甘孜州的产品提供服务、调节服务和文化服务进行价值评估,以期用直观的经济数据揭示甘孜州生态系统对人们的直接贡献。核算结果显示,甘孜州2010年全州生态系统生产总值(GEP)为7545.59亿元,人均71.18万元,是当年该州国内生产总值(GDP)和人均GDP的约61倍。其中,调节服务的价值最大,为6842.28亿元,占GEP总值的90.68%;产品提供服务和文化服务的价值分别为632.64亿元和70.67亿元,占GEP的8.38%和0.94%。研究认为甘孜州生态系统服务功能价值巨大,保护甘孜州生态系统就是保护人们的福祉,该结果可以为甘孜州的生态系统管理、生态保护和生态补偿提供重要依据。  相似文献

The conversion of crystalline metal–organic frameworks (MOFs) into metal compounds/carbon hybrid nanocomposites via pyrolysis provides a promising solution to design electrocatalysts for electrochemical water splitting. However, pyrolyzing MOFs generally involves a complex high‐temperature treatment, which can destroy the coordinated surroundings within MOFs, and as a result not taking their full advantage of their electrolysis properties. Herein, a simple and room‐temperature boronization strategy is developed to convert nickel zeolite imidazolate framework (Ni‐ZIF) nanorods into ultrathin Ni‐ZIF/Ni? B nanosheets with abundant crystalline–amorphous phase boundaries. The combined experiment, and theoretical calculation results disclose that the ultrathin thickness allows fast electron transfer and ensures increased exposure of surface coordinatively unsaturated active sites while the crystalline–amorphous interface elaborately changes the potential‐determining step to energetically favorable intermediates. As a result, Ni‐ZIF/Ni? B nanosheets supported on nickel foam (NF) require overpotentials of 67 mV for the hydrogen evolution reaction and 234 mV for the oxygen evolution reaction to achieve a current density of 10 mA cm?2. Remarkably, Ni‐ZIF/Ni? B@NF as a bifunctional electrocatalyst for overall water splitting enables an alkaline electrolyzer with 10 mA cm?2 at an ultralow cell voltage of 1.54 V. The present work may open a new avenue to the design of MOF‐derived composites for electrocatalysis.  相似文献

The phase behavior and ionic conductivity of tetraethylammonium bis(trifluoromethane‐sulfonyl)imide (Et4NTFSI) salt mixtures with LiTFSI have been examined. In addition, the phase behavior and crystal structure of neat LiTFSI is also reported. Two (1‐x) Et4NTFSI‐(x) LiTFSI (x = 0.50 and 0.67, where x is the mol fraction) mixed‐salt crystalline phases form. Large variations in ionic conductivity are observed; these are attributed to solid‐solid phase transitions of the neat Et4NTFSI salt creating disordered plastic crystalline phases and the formation of a low‐melting eutectic composition between the neat Et4NTFSI salt and the 1/1 Et4NTFSI/LiTFSI (x = 0.50) phase. Although Et4NTFSI and LiTFSI melt at 102 and 234 °C, respectively, the two salts form a eutectic system with a melting temperature of 32 °C. Based upon the findings reported, a new conductivity mechanism is proposed for plastic crystalline salt‐lithium salt electrolytes which is not ascribed to solid‐state diffusion/conduction.  相似文献

We report that the smb20752 gene of the alfalfa symbiont Sinorhizobium meliloti is a novel symbiotic gene required for full N2‐fixation. Deletion of smb20752 resulted in lower nitrogenase activity and smaller nodules without impacting overall nodule morphology. Orthologs of smb20752 were present in all alpha and beta rhizobia, including the ngr_b20860 gene of Sinorhizobium fredii NGR234. A ngr_b20860 mutant formed Fix? determinate nodules that developed normally to a late stage of the symbiosis on the host plants Macroptilium atropurpureum and Vigna unguiculata. However an early symbiotic defect was evident during symbiosis with Leucaena leucocephala, producing Fix? indeterminate nodules. The smb20752 and ngr_b20860 genes encode putative 3‐hydroxyisobutyryl‐CoA (HIB‐CoA) hydrolases. HIB‐CoA hydrolases are required for l ‐valine catabolism and appear to prevent the accumulation of toxic metabolic intermediates, particularly methacrylyl‐CoA. Evidence presented here and elsewhere (Curson et al., 2014 , PLoS ONE 9:e97660) demonstrated that Smb20752 and NGR_b20860 can also prevent metabolic toxicity, are required for l ‐valine metabolism, and play an undefined role in 3‐hydroxybutyrate catabolism. We present evidence that the symbiotic defect of the HIB‐CoA hydrolase mutants is independent of the inability to catabolize l ‐valine and suggest it relates to the toxicity resulting from metabolism of other compounds possibly related to 3‐hydroxybutyric acid.  相似文献

Objective: To date, no studies have examined dietary intake, physical activity, and body image in a large sample of Latin‐American and black women recruited using the same methodology. The aim of this study was to examine three potential correlates of obesity (dietary intake, body image, and physical activity) in a large sample of Latin‐American and black women across the weight spectrum. Research Methods and Procedures: Participants were black (n = 271) and Latin‐American (n = 234) adult women who completed a 24‐hour dietary recall and physical activity and body image questionnaires. Results: After controlling for BMI, education, marital status, and number of children, black women consumed more kilocalories, dietary fat (grams), and percent calories from fat than Latin‐American women, who consumed more carbohydrates (grams) and dietary fiber (total and soluble). Black women engaged in more sedentary behavior than Latin‐American women. Although Latin‐American women weighed less than black women, they perceived their current body image as heavier and reported greater body image dissatisfaction than black women. Black women also reported a higher ideal body image than Latin‐American women. Discussion: The combined effect of a diet higher in calories and fat, increased sedentary behavior, and more accepting body image could account for higher rates of obesity among black women. Future studies should further explore cultural attitudes and beliefs related to weight that could provide information for the development of culturally competent obesity interventions.  相似文献
