Greenhouse experiments were conducted on various crops (cucumber, tomato, eggplant, green bean) to ascertain the effects of Break-thru® (polyether-polymethylsiloxane-copolymer, a silicone surfactant) and an oil emulsion, on Beauveria bassiana (Balsamo) Vuillemin (Bb) applications for the control of the two spotted spider mite, Tetranychus urticae Koch. The objectives were to compare a) the efficacy of Bb control when applied in aqueous Break-thru® or an oil emulsion; b) the effects of various concentrations of Bb conidia, as affected by each surfactant; and c) the effects of Break-thru® on the activity of the fungus. Conidia were suspended either in an aqueous Break-thru® or an emulsifiable formulation at different conidial concentrations (1.05 × 106, 2.1 × 106 and 4.2 × 106 conidia ml?1) and sprayed onto leaves 2 weeks after artificial pest inoculation. Two sprays were performed, with an interval of one week from one spray to another, and T. urticae population counts (both motile and egg stages) were made on plant leaves 7 days after each spray. Bb conidia in Break-thru® were more efficacious than conidia in emulsifiable formulation. With the highest rate of conidia (4.2 × 106 conidia ml?1), mortality of adult mites ranged from 60 ± 4.2 (mean ± SE) to 85.7 ± 4.3% in the Break-thru® suspension and 39.4 ± 7 to 61.3 ± 6% in the oil emulsion. Leaf damage index was also substantially reduced from 70% in the unsprayed control to 40% by the application of Bb conidia at the highest rate with Break-thru®. Break-thru® can be combined with Bb in the integrated management of T. urticae and Isolate R444 is a promising candidate for the control of the pest.  相似文献

Two isolates of Beauveria bassiana (Bb1 and Bb5 strains) were grown in solid-state and submerged cultures (SSC and SbC, respectively) in order to obtain conidia and dry mycelium preparations. The samples obtained at laboratory scale were tested as alternatives for their further use as mycoinsecticides to control the sugarcane borer Diatraea saccharalis. The spore yields obtained in SSC indicated that the Bb5 strain was able to produce around three times more conidia per gram of initial dry matter (3.7×1010) than the Bb1 strain (1.3×1010) in a solid medium composed of wheat bran and bran husk. Under this culture conditions, the former showed not only satisfactory spore yields, but also a higher bioinsecticidal activity than the latter. Laboratory bioassays carried out with conidia of both strains against D. saccharalis larvae indicated that the Bb5 strain promotes an insect mortality of 82.5% whereas only 21.3% was observed with the Bb1 strain preparations. Concerning the samples obtained in SbC, in which a culture medium based on glucose and yeast extract was employed, it is interesting to point out that although both strains showed similar behaviors with yields of approximately 1.50×1010 conidia per gram of dry mycelium, both preparations failed in their bioinsecticidal activities. Despite satisfactory yields, bioinsecticidal activity of the Bb1 and Bb5 strains dropped drastically showing a larvae mortality below 2%.  相似文献

Beauveria bassiana has a high insecticidal potential to control the tarnished plant bug, Lygus lineolaris, a significant pest of strawberries. Screening experiments showed that L. lineolaris adults were susceptible to several B. bassiana isolates. Another screening test with Coleomegilla maculata, a natural enemy found in strawberries, was also performed in order to select the isolate having lower entomopathogenic impact on this insect. Based on data obtained from both insect species and on the ecozone origin of the B. bassiana isolates, INRS‐IP and INRS‐CFL isolates were selected for further experiments. The LC50 values of these two isolates against L. lineolaris adults were 7.8 × 105 and 5.3 × 105 conidia/ml, and average survival time (AST) values were 4.46 and 4.37 days at a concentration of 1 × 108 conidia/ml respectively. Results also indicated that L. lineolaris nymphs are susceptible to the selected isolates. During field experiments, using a randomized block design with four replicates, INRS‐IP and INRS‐CFL isolates were applied at two rates (1 × 1011 and 1 × 1013 conidia/ha) weekly during a period of 4 weeks. These multiple applications triggered a significant reduction of L. lineolaris nymphal populations in strawberries. Twenty‐four days after the first application, a significant difference was observed between the mean population densities of surviving nymphs in all B. bassiana‐treated plots (less than one insect per five plants) compared with those in control plots (four insects per five plants). During the field experiment, persistence of insecticidal activity and viability of B. bassiana conidia were also monitored. The results showed the presence of viable and infective conidia up to 6 days after each application on strawberry foliage. Moreover, the multiple applications of B. bassiana at the rate of 1 × 1013 conidia/ha triggered a significant reduction in strawberry fruit injuries induced by L. lineolaris feeding behaviour compared with the control plots.  相似文献

The effects of various spray application parameters on the efficacy of a clay-based wettable powder formulation of Beauveria bassiana strain GHA conidia against western flower thrips, Frankliniella occidentalis, were evaluated in a series of greenhouse tests. With the aim of optimizing spray application methods to maximize biopesticide efficacy, a series of independent experiments was conducted that varied four spray parameters: application interval, rate, volume, and spray-program timing. Impatiens crops infested with western flower thrips were treated with (1) multiple sprays at the rate of 5×1013 conidia in 935 L aqueous carrier ha?1 applied at 3-, 5-, and 7-day intervals, (2) weekly sprays at rates of 5×1013, 1×1014, and 2.0×1014 conidia in 935 L carrier ha?1, (3) weekly sprays at a rate of 2.0×1014 conidia in volumes of 935, 1870, and 3740 L water ha?1, and (4) multiple sprays at the rate of 2.0×1014 conidia in 3740 L carrier ha?1 applied at 5-day intervals in spray programs initiated before versus after the onset of flowering. Pollen-bearing impatiens flowers were sampled twice weekly to estimate thrips population density, and adult female and second-instar thrips were collected 24 h post application for determination of acquired dose (conidia/insect). Numbers of conidia inoculated onto thrips increased with increasing spray frequency and volume. Dose was unexpectedly not directly correlated with application rate when volume was held constant, suggesting that thrips avoided concentrated spray residues. Statistically significant thrips population reductions relative to controls were achieved only when three to four sprays were applied at the highest label rate in the highest volume at < 7-day intervals. Applications against thrips infesting young, preflowering impatiens crops were not consistently more effective than applications in older crops. The most effective treatment programs reduced pest populations by 30–40% compared to untreated controls; this slowed, but did not stop, the growth of pest populations. Results indicate that use of fungi for thrips management will require integration with other control agents.  相似文献

The strawberry bud weevil, Anthonomus signatus (Say) (Coleoptera: Curculionidae) is a serious pest of strawberry crops in Canada. The results of previous laboratory screening and bioassay experiments showed that A. signatus adults were susceptible to several isolates of Beauveria bassiana (Balsmo) Vuillemin. Based on these results, the INRS-IP and INRS-CFL isolates were selected for field trails. Using a randomized block design with four replicates, three applications of the INRS-IP and INRS-CFL isolates were applied at a rate of 1×1013 conidia/ha at 7-day intervals. The multiple applications of B. bassiana triggered a significant reduction of A. signatus adult populations in strawberries. Twenty-four days after the first application, a significant difference was observed between the mean numbers of surviving adults in all B. bassiana-treated plots compared to control plots. During the field experiment, the persistence of insecticidal activity and the viability of B. bassiana conidia were also monitored. Results showed the presence of viable and infective conidia on strawberry foliage up to 6 days after each application. Moreover, the multiple applications of B. bassiana at a rate of 1×1013 conidia/ha resulted in a significant reduction of damaged strawberry fruits compared with control plots, where fruit loss was caused by A. signatus feeding and egg laying.  相似文献

Stenoma impressella is one of the most important defoliator pests in oil palm plantations in Colombia. To identify an alternative method for its control was characterized biologically and molecularly two strains of Cordyceps cateniannulata (CPIsp1201 and IPIsp1201) and three strains of Beauveria bassiana (CPBb0502; CPBb0411; CPBb0404) against S. impressella larvae. Virulence was evaluated under laboratory conditions. In an oil palm leaflet, individual larvae obtained from the insect colony were inoculated with 5 μl of a conidial suspension containing 1 × 107 conidia/ml. The five strains were pathogenic against S. impressella larvae. CPIsp1201 and IPIsp1201 strains resulted in the highest mortality and were subsequently evaluated in two bioassays using a dose of 1 × 1013 conidia/ha. In the first bioassay, performed under shaded conditions, leaves of oil palms were infested with 75 larvae from the breeding/treatment. The second bioassay was performed in the field using natural populations. No differences were found between strains in both bioassays and the different dosages (5 × 1012, 1 × 1013, and 1.5 × 1013 conidia/ha). Finally, the two strains were evaluated under oil palm plantation conditions at a dose of 1 × 1013 conidia/ha in 126 naturally infested palms. Larval mortality caused by the strains IPIsp1201 and CPIsp1201 (79.5% and 70.5%, respectively) was higher than the natural mortality registered in the control (37.3%). Cordyceps cateniannulata used at 1 × 1013 conidia/ha was effective at controlling S. impressella.  相似文献

To evaluate the pathogenicity of Metarhizium anisopliae (Metschinkoff) Sorokin (Deuteromycotina: Hyphomycetes) a bioassay was designed under laboratory conditions against Rhyzopertha dominica F. (Coleoptera: Bostrychidae) on stored wheat. The fungus was applied at the dose rates of 8 × 103, 8 × 105, 8 × 107 and 8 × 109 conidia/kg of wheat and the bioassay was conducted at 25°C with 60% relative humidity. The data regarding the mortality was recorded after 7 and 14 days exposure intervals. All the treatments gave the significant mortality of R. dominica and M. anisopliae of 8 × 109 conidia/kg was found to be the most effective after a 14-day exposure interval. There was greater production of progeny when the low rate of M. anisopliae was applied to wheat. Overall, our study showed that M. anisopliae is vigorous when applied at a high dose rate which revealed an effective control of R. dominica and also played a pivotal role in the integrated pest management program (IPM) of stored wheat insect pests.  相似文献

The effect of either untreated or treated adults of the spider mite, Tetranychus urticae Koch (Acari: Tetranychidae) by Beauveria bassiana (Bals.) Vuillemin (Ascomycota: Hypocreales) DEBI008 at 1×106 (conidia/ml) was investigated on developmental stages and life table parameters of Phytoseiulus persimilis Athias-Henriot (Acari: Phytoseiidae) under laboratory conditions. Four time intervals (0, 24, 48 and 72 h post-inoculation of spider mites) were considered for studying the predator characteristics as different treatments. Duration of each life stage, longevity, reproduction rate, intrinsic rate of natural increase (r m ), net reproductive rate (R 0), mean generation time (T) and finite rate of increase (λ) of the P. persimilis were calculated on both untreated and B. bassiana treated spider mite adults. Data analysis showed that longevity and fecundity of predatory mites fed on untreated and treated mites (time interval 0) were higher in comparison with other time intervals after inoculation. The entomopathogenic fungus adversely affected longevity and fecundity of the predatory mite. Fertility life table parameters of predatory mites fed on T. urticae treated by B. bassiana at different time intervals showed that T, R 0, λ and r m are strongly affected by the fungus presence and these parameters had significant differences among time treatments. The least r m value was observed in the time interval of 72 h post-inoculation. The fitness of T. urticae was affected by B. bassiana 24, 48 and 72 h post-inoculation of mite adults, and consequently it caused decreased longevity of P. persimilis and accordingly a decrease in the intrinsic rate of natural increase of the predator.  相似文献

Collaborative research was conducted at the USDA-ARS Subtropical Agricultural Research Center in southern Texas to assess the microbial control potential of Beauveria bassiana and Paecilomyces fumosoroseus against Bemisia whiteflies. Laboratory assays demonstrated the capacity of both pathogens to infect Bemisia argentifolii nymphs on excised hibiscus leaves incubated at relative humidities as low as 25% at 23 ± 2°C (ca. 35% infection by B. bassiana and P. fumosoroseus resulted from applications of 0.6–1.4 × 103 conidia/mm2 of leaf surface). In small-scale field trials using portable air-assist sprayers, applications at a high rate of 5 × 1013 conidia in 180 liters water/ha produced conidial densities of ca. 1–2.5 × 103 conidia/mm2 on the lower surfaces of cucurbit leaves. Multiple applications of one isolate of P. fumosoroseus and four isolates of B. bassiana made at this rate at 4- to 5-day intervals provided >90% control of large (third- and fourth-instar) nymphs on cucumbers and cantaloupe melons. The same rate applied at 7-day intervals also provided >90% control in zucchini squash, and a one-fourth rate (1.25 × 1013 conidia/ha) applied at 4- to 5-day intervals reduced numbers of large nymphs by >85% in cantaloupe melons. In contrast to the high efficacy of the fungal applications against nymphs, effects against adult whiteflies were minimal. The results indicated that both B. bassiana and P. fumosoroseus have strong potential for microbial control of nymphal whiteflies infesting cucurbit crops.  相似文献

Barley, Hordeum vulgare, one of the important crops in the word, is used in malting, feed and food industries. The red flour beetle, Tribolium castaneum, was found wherever grains or other dried foods are stored. Disinfestations of barley using chemical methods to kill insects, in this research, for the first time we isolated the pathogenic KB512 of entomopathogenic fungi Beauveria bassiana from soil and insects, which produced aerial and submerged conidia and blastospores in laboratory conditions. We investigated the best conditions for the production and utilisation of spore suspension to spray the larvae of T. castaneum, which is one of the important pests in Kerman province (Iran). One hundred and eighty isolates that naturally infected by T. castaneum were reared during spring and summer seasons 2010–2011. The pathogenicity test was carried out with direct spray. To bioassay the isolates, three concentrations of the spore suspension were prepared as follows: 1?×?106 and 1?×?108 conidia/ml. The pests were sprayed by aerial conidial suspension, which was prepared by 0.01% Tween 80 in distilled water, and the controls were sprayed by 0.01% Tween 80 in distilled water. After spraying the pests, the plates were incubated at 25?±?1?°C and 80% of relative humidity. Then, the treated pests were monitored every day for the fungal growth and mortality.  相似文献

The efficacy of aqueous and emulsifiable formulations of the fungus Metarhizium anisopliae isolate ICIPE78 was evaluated on the population density of Tetranychus urticae infesting common bean plants under screenhouse and field conditions. Synthetic acaricide abamectin was included as a check. Bean plants were artificially infested with T. urticae and allowed to multiply. Three treatments were applied in the screenhouse and 1 treatment in field trials. Mite density was recorded 2 d before spraying and weekly postspraying. The number of pods per plant, number of seeds per pod, and the dry weight of seeds per plant were recorded only in the screenhouse trials. In both screenhouse and field trials, fungal formulations applied at the concentration of 108 conidia/mL and the acaricide reduced the population density of mites as compared to the controls. There were significant differences in T. urticae population densities between the treatments at the various post‐spraying sampling dates. In the screenhouse, the mite densities were near zero from 3‐week postspraying in the treated leaves. At 4‐week postspraying, there were no more leaves in the untreated control (T1) and in the control water + Silwet‐L77 (T2). Fungal formulations were as effective as abamectin in reducing mite densities in both screenhouse and field experiments. There were significant differences in the production parameters during the 2 screenhouse trials, with fungal and abamectin treatments generally having the highest yield. Results of this study underline the potential of the M. anisopliae isolate ICIPE78 as an alternative to acaricides for T. urticae management.  相似文献

An isolate of Bipolaris eleusines was investigated as a potential candidate for biocontrol of barnyardgrass and additional weeds in paddy rice fields and for safety to selected crop species under greenhouse conditions. Barnyardgrass appeared more susceptible at the three-leaf stage than older plants, and disease severity increased as the fungal inoculum increased from 1 × 105 to 1 × 107 conidia/ml when sprayed till run-off. The high application rate caused 73% mortality of barnyardgrass, relative to the non-treated control, but increasing application rate to >1 × 107 conidia/ml did not enhance efficacy (P >.05). This B. eleusines isolate showed no pathogenicity to rice (Oryza sativa spp. indica, O. sativa spp. japonica and an O. sativa hybrid), corn, wheat or any dicot crop species tested while causing slight infection on sorghum and barley. We conclude that B. eleusines, with high efficacy against barnyardgrass and demonstrated safety to rice, is a promising mycoherbicide candidate worthy of further evaluation and development for control of barnyardgrass in paddy rice fields.  相似文献

Fourteen monosporal isolates of Hirsutella thompsonii grew vegetatively in various liquid media producing typical phialidic-like conidiophores. Hirsutella thompsonii var. synnematosa from Ivory Coast (HtIC) was the only pathotype which produced true conidia in submerged culture. HtIC began producing conidia after 3 days incubation reaching a peak ranging from 6.8 × 105 to 9.7 × 107 conidia/ml between 6 and 11 days. A concentration of 10 g/liter of corn steep liquor and 0.2% Tween 80 were essential for maximum conidiation. Submerged conidia had a smooth but somewhat rugose conidial walls, whereas aerially formed conidia were distinctly verrucose. Germination of submerged conidia ranged from 5.2 to 12.9% and were virulent causing 32.5% infection to adult citrus rust mites when sprayed on citrus foliage at 1.2 × 109 conidia/ml.  相似文献

Microsclerotia (MS) of Metarhizium brunneum strain F52 (Hypocreales: Clavicipitaceae) were processed as granules using three carriers: kaolin clay, microcrystalline cellulose (MCC), or diatomaceous earth (DE). In a series of experiments aimed at comparing viable conidial production, each MS carrier type was hydrated using a decreasing range of water activities on glycerol or polyethylene glycol (PEG200)-amended media. Conidial density and percent germination of conidia declined significantly as water activity was lowered. All three carrier types produced >2–6?×?109 viable conidia/gram at higher water activities (аw >0.987) but were still capable of producing 2.9?×?107?±?1.6?×?106 conidia/g (30% glycerol аw?=?0.883) and 1.9?×?107?±?6.5?×?106 conidia/g (30% PEG200 аw?=?0.924) at the lowest tested water activities. MS carrier types were sprayed onto 0.4?m length logs with or without hydromulch formulation. The median survival times of Asian longhorned beetles, Anoplophora glabripennis (Coleoptera: Cerambycidae), exposed by climbing double the length of the logs of hydromulch-treated bark, were 16.5–20.5?d while beetles exposed to logs without hydromulch formulation had median survival times of 22–25.5?d. Overall, experiments showed that there were few biologically significant differences between the MS carrier types.  相似文献

The cherry slugworm Caliroa cerasi is a significant destructive pest of sour cherries (Prunus cerasus) in Turkey. The potential of entomopathogenic fungi for controlling C. cerasi was evaluated. The effects of exposure methods and conidial concentrations (1 × 106, 1.5 × 106, 1 × 107 and 1.5 × 107 conidia/ml) on mature larvae of C. cerasi infected by Beauveria bassiana were investigated under laboratory conditions. Larvae sprayed directly with B. bassiana conidial suspensions and larvae exposed to B. bassiana-treated leaves resulted in 100% mortality within 2.90 and 2.77 days, respectively. The median lethal time (LT50) and days to mortality were highest in the 1.0 × 107 concentrations of conidia for both direct spray and leaf exposure. The present study suggests that B. bassiana has good potential for control of the cherry slugworm, C. cerasi.  相似文献

Dubas bug, Ommatissus lybicus is one of the major sucking pests of date palm. Data palm is the most important economic crop in Iran. In this research, we isolated the pathogenic strain KB 201, KB 206, KB 211, KB 215, KB 220 and KB 512, of entomopathogenic fungi Beauveria bassiana from soil and insects, which produced aerial and submerged conidia and blastospores in the laboratory conditions. We investigated the best conditions for the production and utilisation of the spore suspension to spray the nymph of O. lybicus, which is one of the important pests in Kerman province (Iran). A total of 180 isolates that naturally infected by O. lybicus were collected from date palm in Kerman province during spring and summer seasons 2010–2011. The pathogen city test was carried out with direct spray. To bioassay the isolates, three concentrations of the spore suspension were prepared as follows: 1?×?106, 1?×?107 and 1?×?108?conidia/ml. The pests were sprayed by aerial conidial suspension, which was prepared by 0.01% Tween 80 in distilled water, and the controls were sprayed by 0.01% Tween 80in distilled water. After spraying the pests, the plates were incubated at 25?±?1?°C and 80% of relative humidity. Then, the treated pests were monitored every day for the fungal growth and mortality.  相似文献

The pathogenicity of Beauveria bassiana to one of the major pests of citrus crops, Phyllocoptruta oleivora, was assessed by inoculating mites with different concentrations of conidia (1×106, 5×106, 1×107, 5×107 and 1×108). Treated mites were kept at controlled conditions (25 ± 0.5°C, 12 h photoperiod and 98% relative humidity) and mite survivorship was evaluated daily. Mortality was found to increase in time and was dependent on the conidia concentration, with values ranging from 24 to 91% for the lowest and highest conidia concentration, respectively. The calculated LC50 on the fifth day was 4.23×106 conidia/ml. Mean lethal time was 3.98, 9.79, 3.09 and 2.74 days for 5×106, 1×107, 5×107 and 1×108 conidia/ml, respectively. Conidia were found to adhere all over the mite body surface, especially at the anal region, where vegetative mycelium was found entering the mite body. We noticed the formation of small crystals inside the mite’s body that were produced during colonization of the body cavity by the fungus. This is the first report of B. bassiana pathogenicity for this species.  相似文献

Thermotolerance of entomopathogenic (insect-killing) fungi should be seriously considered before industrialization. This work describes the feasibility of millet grain as a substrate for production of thermotolerant Beauveria bassiana (Bb) GHA and ERL1170 and Metarhizium anisopliae (Ma) ERL1171 and ERL1540 conidia. First, conidial suspensions of the Bb isolates, produced on millet grain in polyethylene bags, were exposed to five temperatures (43–47°C) at 15-min intervals for up to 120 min (experiment I). Agar-based quarter-strength (¼) Sabouraud dextrose agar supplemented with yeast extract (SDAY) and whey permeate media served as controls. Millet-grain-based culture was superior in producing the most thermotolerant Bb conidia, followed by whey permeate agar and ¼SDAY-based cultures. Secondly, to compare the thermotolerance of conidia produced at the same conditions, the Bb isolates were then produced on agar-based millet powder medium, with ¼SDAY and whey permeate agar media as controls, and the two Ma isolates were added (experiment II). They were then exposed to the same temperatures as above. More thermotolerant Bb and Ma conidia were produced on millet powder agar than on whey permeate agar and ¼SDAY overall. These results suggest that millet grain can be used as a substrate to produce thermotolerant conidia in a mass production system.  相似文献

The ability of conidia of the human pathogenic fungus Aspergillus fumigatus to kill larvae of the insect Galleria mellonella was investigated. Conidia at different stages of the germination process displayed variations in their virulence as measured using the Galleria infection model. Non-germinating (‘resting’) conidia were avirulent except when an inoculation density of 1 × 107 conidia per insect was used. Conidia that had been induced to commence the germination process by pre-culturing in growth medium for 3 h were capable of killing larvae at densities of 1 × 106 and 1 × 107 per insect. An inoculation density of 1 × 105 conidia per insect remained avirulent. Conidia in the outgrowth phase of germination (characterised as the formation of a germ tube) were the most virulent and were capable of killing 100% of larvae after 5 or 24 h when 1 × 107 or 1 × 106 conidia, that had been allowed to germinate for 24 h, were used. Examination of the response of insect haemocytes to conidia at different stages of the germination process established that haemocytes could engulf non-germinating conidia and those in the early stages of the germination process but that conidia, which had reached the outgrowth stages of germination were not phagocytosed. The results presented here indicate that haemocytes of G. mellonella are capable of phagocytosing A. fumigatus conidia less than 3.0 μm in diameter but that conidia greater than this are too large to be engulfed. The virulence of A. fumigatus in G. mellonella larvae can be ascertained within 60–90 h if infection densities of 1 × 106 or 1 × 107 activated conidia (pre-incubated for 2–3 h) per insect are employed.  相似文献
