The effects ofcyanide (CN) on whole cell current measured with the perforated-patchmethod were studied in adrenal medullary cells. Application of CNproduced initially inward and then outward currents at 52 mV ormore negative. As the membrane potential was hyperpolarized, amplitudeand latency of the outward current (Io) by CNbecame small and long, respectively. A decrease in the externalNa+ concentration did not affectthe latency for CN-inducedIo but enhancedthe amplitude markedly. The CNIo reversedpolarity at 85 mV, close to the Nernst potential forK+, and was suppressed by theK+ channel blockers curare andapamin but not by glibenclamide, suggesting thatIo is due to theactivation of Ca2+-dependentK+ channels. Consistent with thisnotion, the Ca2+-mobilizingagents, muscarine and caffeine, also producedIo. Exposure toCN in a Ca2+-deficient medium for4 min abolished caffeine- or muscarine-induced Io withoutdevelopment ofIo, and additionof Ca2+ to the CN-containingsolution inducedIo. We concludethat exposure to CN producesCa2+-dependentK+ currents in an externalCa2+-dependent manner, probablyvia facilitation of Ca2+ influx.

Increased extracellular osmolarity ([Os]e) suppresses stimulated hormone secretion from anterior pituitary cells. Ca2+ influx may mediate this effect. We show that increase in [Os]e (by 18–125%) differentially suppresses L-type and T-type Ca2+ channel currents (IL and IT, respectively); IL was more sensitive than IT. Hyperosmotic suppression of IL depended on the magnitude of increase in [Os]e and was correlated with the percent decrease in pituitary cell volume, suggesting that pituitary cell shrinkage can modulate L-type currents. The hyperosmotic suppression of IL and IT persisted after incubation of pituitary cells either with the actin-disrupter cytochalasin D or with the actin stabilizer phalloidin, suggesting that the actin cytoskeleton is not involved in this modulation. The hyperosmotic suppression of Ca2+ influx was not correlated with changes in reversal potential, membrane capacitance, and access resistance. Together, these results suggest that the hyperosmotic suppression of Ca2+ influx involves Ca2+ channel proteins. We therefore recorded the activity of L-type Ca2+ channels from cell-attached patches while exposing the cell outside the patch pipette to hyperosmotic media. Increased [Os]e reduced the activity of Ca2+ channels but did not change single-channel conductance. This hyperosmotic suppression of Ca2+ currents may therefore contribute to the previously reported hyperosmotic suppression of hormone secretion. In mammalian cardiac myocytes, calcium released into the dyadic space rapidly inactivates calcium current (ICa). We used this Ca2+ release-dependent inactivation (RDI) of ICa as a local probe of sarcoplasmic reticulum Ca2+ release activation. In whole cell patch-clamped rat ventricular myocytes, Ca2+ entry induced by short prepulses from —50 mV to positive voltages caused suppression of peak ICa during a test pulse. The negative correlation between peak ICa suppression and ICa inactivation during the test pulse indicated that RDI evoked by the prepulse affected only calcium channels in those dyads in which calcium release was activated. Ca2+ ions injected during the prepulse and during the subsequent tail current suppressed peak ICa in the test pulse to a different extent. Quantitative analysis indicated that equal Ca2+ charge was 3.5 times less effective in inducing release when entering during the prepulse than when entering during the tail. Tail Ca2+ charge injected by the first voltage-dependent calcium channel (DHPR) openings was three times less effective than that injected by DHPR reopenings. These findings suggest that calcium release activation can be profoundly influenced by the recent history of L-type Ca2+ channel activity due to potentiation of ryanodine receptors (RyRs) by previous calcium influx. This conclusion was confirmed at the level of single RyRs in planar lipid bilayers: using flash photolysis of the calcium cage NP-EGTA to generate two sequential calcium stimuli, we showed that RyR activation in response to the second stimulus was four times higher than that in response to the first stimulus. excitation-contraction coupling  相似文献

Properties of ATP-dependent K(+) channels in adrenocortical cells   总被引:6,自引:0,他引:6  
Bovine adrenocortical zona fasciculata (AZF)cells express a novel ATP-dependent K+-permeable channel(IAC). Whole cell and single-channel recordings were used to characterize IAC channels withrespect to ionic selectivity, conductance, and modulation bynucleotides, inorganic phosphates, and angiotensin II (ANG II). Inoutside-out patch recordings, the activity of unitaryIAC channels is enhanced by ATP in the patchpipette. These channels were K+ selective with nomeasurable Na+ or Ca2+ conductance. Insymmetrical K+ solutions with physiological concentrationsof divalent cations (M2+), IACchannels were outwardly rectifying with outward and inward chordconductances of 94.5 and 27.0 pS, respectively. In the absence ofM2+, conductance was nearly ohmic. Hydrolysis-resistantnucleotides including AMP-PNP and NaUTP were more potent than MgATP asactivators of whole cell IAC currents. Inorganicpolytriphosphate (PPPi) dramatically enhancedIAC activity. In current-clamp recordings, nucleotides and PPPi produced resting potentials in AZFcells that correlated with their effectiveness in activatingIAC. ANG II (10 nM) inhibited whole cellIAC currents when patch pipettes contained 5 mMMgATP but was ineffective in the presence of 5 mM NaUTP and 1 mM MgATP.Inhibition by ANG II was not reduced by selective kinase antagonists.These results demonstrate that IAC is adistinctive K+-selective channel whose activity isincreased by nucleotide triphosphates and PPPi.Furthermore, they suggest a model for IAC gatingthat is controlled through a cycle of ATP binding and hydrolysis.

The hyperpolarization-activated, cyclic nucleotide-gated (HCN) channels, or cardiac (If)/neuronal (Ih) time- and voltage-dependent inward cation current channels, are conventionally considered as monovalent-selective channels. Recently we discovered that calcium ions can permeate through HCN4 and Ih channels in neurons. This raises the possibility of Ca2+ permeation in If, the Ih counterpart in cardiac myocytes, because of their structural homology. We performed simultaneous measurement of fura-2 Ca2+ signals and whole cell currents produced by HCN2 and HCN4 channels (the 2 cardiac isoforms present in ventricles) expressed in HEK293 cells and by If in rat ventricular myocytes. We observed Ca2+ influx when HCN/If channels were activated. Ca2+ influx was increased with stronger hyperpolarization or longer pulse duration. Cesium, an If channel blocker, inhibited If and Ca2+ influx at the same time. Quantitative analysis revealed that Ca2+ flux contributed to 0.5% of current produced by the HCN2 channel or If. The associated increase in Ca2+ influx was also observed in spontaneously hypertensive rat (SHR) myocytes in which If current density is higher than that of normotensive rat ventricle. In the absence of EGTA (a Ca2+ chelator), preactivation of If channels significantly reduced the action potential duration, and the effect was blocked by another selective If channel blocker, ZD-7288. In the presence of EGTA, however, preactivation of If channels had no effects on action potential duration. The effect of oxidants on voltage-dependent K+ currents was examined in mouse colonic smooth muscle cells. Exposure to either chloramine-T (Ch-T), an agent known to oxidize both cysteine and methionine residues, or the colon-specific oxidant monochloramine (NH2Cl) completely suppressed the transient outward K+ current (Ito) while simultaneously enhancing the sustained delayed rectifier K+ current (Idr). In contrast, the cysteine-specific oxidants hydrogen peroxide (H2O2) and 5,5'-dithiobis(2-nitrobenzoic acid) (DTNB) exhibited partial and slow suppression of Ito by inducing a shift in channel availability of -18 mV without affecting Idr. After enhancement by NH2Cl or Ch-T, Idr was sensitive to 10 mM tetraethylammonium but not to other K+ channel blockers, suggesting that it represented activation of the resting Idr and not a separate K+ conductance. Extracellular dithiothreitol (DTT) partially reversed the effect of H2O2 and DTNB on Ito but not the actions of NH2Cl and Ch-T on either Idr or Ito. Dialysis of myocytes with GSH (5 mM) or DTT (5 mM) prevented suppression of Ito by H2O2 and DTNB but did not alter the effects of NH2Cl or Ch-T on either Idr or Ito. Ch-T and NH2Cl completely blocked Ito generated by murine Kv4.1, 4.2, and 4.3 in Xenopus oocytes, an effect not reversible by intracellular DTT. In contrast, intracellular DTT reversed the effect of H2O2 and DTNB on the cloned channels. Weinvestigated the contribution of sialic acid residues to theK+ currents involved in the repolarization of mouseventricular myocytes. Ventricular K+ currents had a rapidlyinactivating component followed by slowly decaying and sustainedcomponents. This current was produced by the summation of threedistinct currents: Ito, which contributed to thetransient component; Iss, which contributed tothe sustained component; and IK,slow, whichcontributed to both components. Incubation of ventricular myocytes withthe sialidase neuraminidase reduced the amplitude ofIto without alteringIK,slow and Iss. We foundthat the reduction in Ito amplitude resultedfrom a depolarizing shift in the voltage of activation and a reductionin the conductance of Ito. Expression of Kv4.3channels, a major contributor to Ito in theventricle, in a sialylation-deficient Chinese hamster ovary cell line(lec2) mimicked the effects of neuraminidase on the ventricularIto. Furthermore, we showed that sialylatedglycolipids have little effect on the voltage dependence ofIto. Finally, consistent with its actions onIto, neuraminidase produced an increase in theduration of the action potential of ventricular myocytes and thefrequency of early afterdepolarizations. We conclude that sialylationof the proteins forming Kv4 channels is important in determining thevoltage dependence and conductance of Ito and that incomplete glycosylation of these channels could lead to arrhythmias.

We have previously demonstrated that the sarcolemmalNa+-K+pump current(Ip) in cardiacmyocytes is stimulated by cell swelling induced by exposure tohyposmolar solutions. However, the underlying mechanism has not beenexamined. Because cell swelling activates stretch-sensitive ionchannels and intracellular messenger pathways, we examined their rolein mediating Ipstimulation during exposure of rabbit ventricular myocytes to ahyposmolar solution.Ip was measuredby the whole cell patch-clamp technique. Swelling-induced pumpstimulation altered the voltage dependence ofIp. Pumpstimulation persisted in the absence of extracellularNa+ and under conditions designedto minimize changes in intracellular Ca2+, excluding an indirectinfluence on Ipmediated via fluxes through stretch-activated channels. Pumpstimulation was protein kinase C independent. The tyrosine kinaseinhibitor tyrphostin A25, the phosphatidylinositol 3-kinase inhibitorLY-294002, and the protein phosphatase-1 and -2A inhibitor okadaic acidabolished Ipstimulation. Our findings suggest that swelling-induced pumpstimulation involves the activation of tyrosine kinase,phosphatidylinositol 3-kinase, and a serine/threonine proteinphosphatase. The effects ofintracellular nucleotide triphosphates on time-dependent changes inmuscarinic receptor cation currents (Icat) wereinvestigated using the whole cell patch-clamp technique in guinea pigileal muscle. In the absence of nucleotide phosphates in the patchpipette, Icat evoked every 10 min decayedprogressively. This decay was slowed dose dependently by inclusion ofmillimolar concentrations of ATP in the pipette. This required acomparable concentration of Mg2+, was mimicked by UTP andCTP, and was attenuated by simultaneous application of alkalinephosphatase or inhibitors of tyrosine kinase. In contrast, a suddenphotolytic release of millimolar ATP (probably in the free form) causeda marked suppression of Icat. Submillimolarconcentrations of GTP dose dependently increased the amplitude ofIcat as long as ATP and Mg2+ were inthe pipette, but, in their absence, GTP was ineffective at preventingIcat decay. The decay ofIcat was paralleled by altered voltage-dependentgating, i.e., a positive shift in the activation curve and reduction inthe maximal conductance. It is thus likely that ATP exerts tworeciprocal actions on Icat, throughMg2+-dependent and -independent mechanisms, and that theenhancing effect of GTP on Icat is essentiallydifferent from that of ATP.

Several proteins belonging to the ATP-binding cassettesuperfamily can affect ion channel function. These include the cystic fibrosis transmembrane conductance regulator, the sulfonylurea receptor, and the multidrug resistance protein P-glycoprotein (MDR1).We measured whole cell swelling-activatedCl currents(ICl,swell) inparental cells and cells expressing wild-type MDR1 or aphosphorylation-defective mutant (Ser-661, Ser-667, and Ser-671replaced by Ala). Stimulation of protein kinase C (PKC) with a phorbolester reduced the rate of increase inICl,swell only incells that express MDR1. PKC stimulation had no effect on steady-stateICl,swell.Stimulation of protein kinase A (PKA) with 8-bromoadenosine3',5'-cyclic monophosphate reduced steady-state ICl,swell only inMDR1-expressing cells. PKA stimulation had no effect on the rate ofICl,swellactivation. The effects of stimulation of PKA and PKC onICl,swell wereadditive (i.e., decrease in the rate of activation and reduction insteady-stateICl,swell). The effects of PKA and PKC stimulation were absent in cells expressing thephosphorylation-defective mutant. In summary, it is likely thatphosphorylation of MDR1 by PKA and by PKC alters swelling-activated Cl channels by independentmechanisms and that Ser-661, Ser-667, and Ser-671 are involved in theresponses ofICl,swell tostimulation of PKA and PKC. It has been suggested that L-type Ca2+ channels play an important role in cell swelling-induced vasoconstriction. However, there is no direct evidence that Ca2+ channels in vascular smooth muscle are modulated by cell swelling. We tested the hypothesis that L-type Ca2+ channels in rabbit portal vein myocytes are modulated by hypotonic cell swelling via protein kinase activation. Ba2+ currents (IBa) through L-type Ca2+ channels were recorded in smooth muscle cells freshly isolated from rabbit portal vein with the conventional whole cell patch-clamp technique. Superfusion of cells with hypotonic solution reversibly enhanced Ca2+ channel activity but did not alter the voltage-dependent characteristics of Ca2+ channels. Bath application of selective inhibitors of protein kinase C (PKC), Ro-31–8425 or Go-6983, prevented IBa enhancement by hypotonic swelling, whereas the specific protein kinase A (PKA) inhibitor KT-5720 had no effect. Bath application of phorbol 12,13-dibutyrate (PDBu) significantly increased IBa under isotonic conditions and prevented current stimulation by hypotonic swelling. However, PDBu did not have any effect on IBa when cells were first exposed to hypotonic solution. Furthermore, downregulation of endogenous PKC by overnight treatment of cells with PDBu prevented current enhancement by hypotonic swelling. Bone marrow mesenchymal stem cells (MSCs) are used as a cell source for cardiomyoplasty; however, the cellular electrophysiological properties are not fully understood. The present study was to investigate the functional ionic channels in undifferentiated mouse bone marrow MSCs using whole cell patch-voltage clamp technique, RT-PCR, and Western immunoblotting analysis. We found that three types of ionic currents were present in mouse MSCs, including a Ca2+-activated K+ current (IKCa), an inwardly rectifying K+ current (IKir), and a chloride current (ICl). IKir was inhibited by Ba2+, and IKCa was activated by the Ca2+ ionophore A-23187 and inhibited by the intermediate-conductance IKCa channel blocker clotrimazole. ICl was activated by hyposmotic (0.8 T) conditions and inhibited by the chloride channel blockers DIDS and NPPB. The corresponding ion channel genes and proteins, KCa3.1 for IKCa, Kir2.1 for IKir, and Clcn3 for ICl, were confirmed by RT-PCR and Western immunoblotting analysis in mouse MSCs. Voltage-clamp studies offreshly isolated smooth muscle cells from rabbit portal veinrevealed the existence of a time-dependent cation current evoked bymembrane hyperpolarization (termed Ih). Both therate of activation and the amplitude of Ih wereenhanced by membrane hyperpolarization. Half-maximal activation ofIh was about 105 mV with conventional wholecell and 80 mV when the perforated patch technique was used. Incurrent clamp, injection of hyperpolarizing current produced a markeddepolarizing "sag" followed by rebound depolarization. Activationof Ih was augmented by an increase in theextracellular K+ concentration and was blocked rapidly byexternally applied Cs+ (1-5 mM). The bradycardic agentZD-7288 (10 µM), a selective inhibitor of Ih,produced a characteristically slow inhibition of the portal veinIh. The depolarizing sag recorded in current clamp was also abolished by application of 5 mM Cs+.Cs+ significantly decreased the frequency of spontaneouscontractions in both whole rat portal vein and rabbit portal veinsegments. Multiplex RT-PCR of rabbit portal vein myocytes using primers derived from existing genes for hyperpolarization-activated cation channels (HCN1-4) revealed the existence of cDNA clonescorresponding to HCN2, 3, and 4. The present study shows that portalvein myocytes contain genes shown to encode forhyperpolarization-activated channels and exhibit an endogenous currentwith characteristics similar to Ih in other celltypes. This conductance appears to determine, in part, the rhythmicityof this vessel.

In the brain,astrocytes represent a major target for endothelins (ETs), a family ofpeptides that can be released by several cell types and that havepotent and multiple effects on astrocytic functions. Four types ofK+ currents (IK) were detected invarious proportions by patch-clamp recordings of cultured striatalastrocytes, including the A-type IK, theinwardly rectifying IK IR, theCa2+-dependent IK(IK Ca), and the delayed-rectifiedIK (IK DR). Variationsin the shape of current-voltage relationships were related mainly todifferences in the proportion of these currents. ET-1 was found toregulate with opposite effects the two more frequently recorded outwardK+ currents in striatal astrocytes. Indeed, this peptideinduced an initial activation of IK Ca(composed of SK and BK channels) and a delayed long-lasting inhibitionof IK DR. In current-clamp recordings, theactivation of IK Ca correlated with a transient hyperpolarization, whereas the inhibition ofIK DR correlated with a sustaineddepolarization. These ET-1-induced sequential changes inmembrane potential in astrocytes may be important for the regulation ofvoltage gradients in astrocytic networks and the maintenance ofK+ homeostasis in the brain microenvironment.

Electrophysiological characterization of murine HL-5 atrial cardiomyocytes   总被引:1,自引:0,他引:1  
HL-5 cells are cultured murine atrial cardiomyocytes and have been used in studies to address important cellular and molecular questions. However, electrophysiological features of HL-5 cells have not been characterized. In this study, we examined such properties using whole cell patch-clamp techniques. Membrane capacitance of the HL-5 cells was from 8 to 62 pF. The resting membrane potential was –57.8 ± 1.4 mV (n = 51). Intracellular injection of depolarizing currents evoked action potentials (APs) with variable morphologies in 71% of the patched cells. Interestingly, the incidence of successful, current-induced APs positively correlated with the hyperpolarizing degrees of resting membrane potentials (r = 0.99, P < 0.001). Only a few of the patched cells (4 of 51, 7.8%) exhibited spontaneous APs. The muscarinic agonist carbachol activated the acetylcholine-activated K+ current and significantly shortened the duration of APs. Immunostaining confirmed the presence of the muscarinic receptor type 2 in HL-5 cells. The hyperpolarization-activated cation current (If) was detected in 39% of the patched cells. The voltage to activate 50% of If channels was –73.4 ± 1.2 mV (n = 12). Voltage-gated Na+, Ca2+, and K+ currents were observed in the HL-5 cells with variable incidences. Compared with the adult mouse cardiomyocytes, the HL-5 cells had prolonged APs and small outward K+ currents. Our data indicate that HL-5 cells display significant electrophysiological heterogeneity of morphological appearance of APs and expression of functional ion channels. The cellular mechanism for Cl and K+ secretion in the colonic epithelium requires K+ channels in the basolateral and apical membranes. Colonic mucosa from guinea pig and rat were fixed, sectioned, and then probed with antibodies to the K+ channel proteins KVLQT1 (Kcnq1) and minK-related peptide 2 (MiRP2, Kcne3). Immunofluorescence labeling for Kcnq1 was most prominent in the lateral membrane of crypt cells in rat colon. The guinea pig distal colon had distinct lateral membrane immunoreactivity for Kcnq1 in crypt and surface cells. In addition, Kcne3, an auxiliary subunit for Kcnq1, was detected in the lateral membrane of crypt and surface cells in guinea pig distal colon. Transepithelial short-circuit current (Isc) and transepithelial conductance (Gt) were measured for colonic mucosa during secretory activation by epinephrine (EPI), prostaglandin E2 (PGE2), and carbachol (CCh). HMR1556 (10 µM), an inhibitor of Kcnq1 channels (Gerlach U, Brendel J, Lang HJ, Paulus EF, Weidmann K, Brüggemann A, Busch A, Suessbrich H, Bleich M, and Greger R. J Med Chem 44: 3831–3837, 2001), partially (50%) inhibited Cl secretory Isc and Gt activated by PGE2 and CCh in rat colon with an IC50 of 55 nM, but in guinea pig distal colon Cl secretory Isc and Gt were unaltered. EPI-activated K+-secretory Isc and Gt also were essentially unaltered by HMR1556 in both rat and guinea pig colon. Although immunofluorescence labeling with a Kcnq1 antibody supported the basolateral membrane presence in colonic epithelium of the guinea pig as well as the rat, the Kcnq1 K+ channel is not an essential component for producing Cl secretion. Other K+ channels present in the basolateral membrane presumably must also contribute directly to the K+ conductance necessary for K+ exit during activation of Cl secretion in the colonic mucosa. This study examines whether fluid pressure (FP) modulates the L-type Ca2+ channel in cardiomyocytes and investigates the underlying cellular mechanism(s) involved. A flow of pressurized (16 dyn/cm2) fluid, identical to that bathing the myocytes, was applied onto single rat ventricular myocytes using a microperfusion method. The Ca2+ current (ICa) and cytosolic Ca2+ signals were measured using a whole cell patch-clamp and confocal imaging, respectively. It was found that the FP reversibly suppressed ICa (by 25%) without altering the current-voltage relationships, and it accelerated the inactivation of ICa. The level of ICa suppression by FP depended on the level and duration of pressure. The Ba2+ current through the Ca2+ channel was only slightly decreased by the FP (5%), suggesting an indirect inhibition of the Ca2+ channel during FP stimulation. The cytosolic Ca2+ transients and the basal Ca2+ in field-stimulated ventricular myocytes were significantly increased by the FP. The effects of the FP on the ICa and on the Ca2+ transient were resistant to the stretch-activated channel inhibitors, GsMTx-4 and streptomycin. Dialysis of myocytes with high concentrations of BAPTA, the Ca2+ buffer, eliminated the FP-induced acceleration of ICa inactivation and reduced the inhibitory effect of the FP on ICa by 80%. Ryanodine and thapsigargin, abolishing sarcoplasmic reticulum Ca2+ release, eliminated the accelerating effect of FP on the ICa inactivation, and they reduced the inhibitory effect of FP on the ICa. These results suggest that the fluid pressure indirectly suppresses the Ca2+ channel by enhancing the Ca2+-induced intracellular Ca2+ release in rat ventricular myocytes. To study and define the early time-dependent response (6 h) ofblocker-sensitive epithelial Na+channels (ENaCs) to stimulation ofNa+ transport by aldosterone, weused a new modified method of blocker-induced noise analysis todetermine the changes of single-channel current (iNa) channel open probability(Po), andchannel density(NT) undertransient conditions of transport as measured by macroscopic short-circuit currents(Isc). In threegroups of experiments in which spontaneous baseline rates of transportaveraged 1.06, 5.40, and 15.14 µA/cm2, stimulation of transportoccurred due to increase of blocker-sensitive channels.NT variedlinearly over a 70-fold range of transport (0.5-35µA/cm2). Relatively small andslow time-dependent but aldosterone-independent decreases ofPo occurredduring control (10-20% over 2 h) and aldosterone experimentalperiods (10-30% over 6 h). When thePo of control andaldosterone-treated tissues was examined over the 70-fold extendedrange of Na+ transport,Po was observedto vary inversely withIsc, falling from~0.5 to ~0.15 at the highest rates ofNa+ transport or ~25% per3-fold increase of transport. Because decreases ofPo from anysource cannot explain stimulation of transport by aldosterone, it isconcluded that the early time-dependent stimulation ofNa+ transport in A6 epithelia isdue exclusively to increase of apical membraneNT.

Secretion of Cl and K+ in the colonic epithelium operates through a cellular mechanism requiring K+ channels in the basolateral and apical membranes. Transepithelial current [short-circuit current (Isc)] and conductance (Gt) were measured for isolated distal colonic mucosa during secretory activation by epinephrine (Epi) or PGE2 and synergistically by PGE2 and carbachol (PGE2 + CCh). TRAM-34 at 0.5 µM, an inhibitor of KCa3.1 (IK, Kcnn4) K+ channels (H. Wulff, M. J. Miller, W. Hänsel, S. Grissmer, M. D. Cahalan, and K. G. Chandy. Proc Natl Acad Sci USA 97: 8151–8156, 2000), did not alter secretory Isc or Gt in guinea pig or rat colon. The presence of KCa3.1 in the mucosa was confirmed by immunoblot and immunofluorescence detection. At 100 µM, TRAM-34 inhibited Isc and Gt activated by Epi (4%), PGE2 (30%) and PGE2 + CCh (60%). The IC50 of 4.0 µM implicated involvement of K+ channels other than KCa3.1. The secretory responses augmented by the K+ channel opener 1-EBIO were inhibited only at a high concentration of TRAM-34, suggesting further that KCa3.1 was not involved. Sensitivity of the synergistic response (PGE2 + CCh) to a high concentration TRAM-34 supported a requirement for multiple K+ conductive pathways in secretion. Clofilium (100 µM), a quaternary ammonium, inhibited Cl secretory Isc and Gt activated by PGE2 (20%) but not K+ secretion activated by Epi. We investigatedthe relationship between voltage-operatedCa2+ channel current and thecorresponding intracellular Ca2+concentration([Ca2+]i)change (Ca2+ transient) in guineapig gastric myocytes. Fluorescence microspectroscopy was combined withconventional whole cell patch-clamp technique, and fura 2 (80 µM) wasadded to CsCl-rich pipette solution. Step depolarization to 0 mVinduced inward Ca2+ current(ICa) andconcomitantly raised[Ca2+]i.Both responses were suppressed by nicardipine, an L-typeCa2+ channel blocker, and thevoltage dependence of Ca2+transient was similar to the current-voltage relation ofICa. When pulseduration was increased by up to 900 ms, peakCa2+ transient increased andreached a steady state when stimulation was for longer. The calculatedfast Ca2+ buffering capacity(B value), determined as the ratio ofthe time integral ofICa divided bythe amplitude of Ca2+ transient,was not significantly increased after depletion of Ca2+ stores by the cyclicapplication of caffeine (10 mM) in the presence of ryanodine (4 µM).The addition of cyclopiazonic acid (CPA, 10 µM), a sarco(endo)plasmicreticulum Ca2+-ATPase inhibitor,decreased B value by ~20% in areversible manner. When KCl pipette solution was used,Ca2+-activatedK+ current[IK(Ca)]was also recorded during step depolarization. CPA sensitivelysuppressed the initial peak and oscillations of IK(Ca) withirregular effects on Ca2+transients. The above results suggest that, in guinea pig gastric myocyte, Ca2+ transient is tightlycoupled to ICaduring depolarization, and global[Ca2+]iis not significantly affected byCa2+-inducedCa2+ release from sarcoplasmicreticulum during depolarization.
