Third instar larvae of Heliothis virescens and Helicoverpa zea could be distinguished based on the hydrocarbons of their surface lipids. Hydrocarbons were the major components of the surface lipids and a distinctive capillary gas chromatographic profile could be obtained from a hexane extract of the surface lipids of a single larva. Analysis of hexane extracts of the surface lipids by capillary gas chromatography-mass spectrometry (CGC-MS) showed several obvious differences between the two species: (1) in their gas chromatographic profiles; (2) in the presence of a major alkene, hentriacontene, only in H. zea; (3) in H. virescens the CGC-MS peak with a KI of approximately 2860 was 2-methyloctacosane, but in H. zea it was a mixture of 4-methyloctacosane plus 9,13- and 8,12-dimethyloctacosanes; and (4) in the methyl branch positions of dimethyl-branched alkanes with carbon backbones of C31, C33, C35, C45, C47, C49 and C51. The methyl branch points of H. virescens dimethylalkanes were separated by seven or nine methylenes, while in H. zea the methyl branch points of the dimethylalkanes were separated by three and sometimes five methylenes.  相似文献

The effects on dietary self-selection of phagostimulation and metabolic feedback from nutrients were seperated by using two unusual carbohydrates alone or in combination: mannitol, which is nutritive but not phagostimulatory for H. zea larvae; and sorbose, which is weakly phagostimulatory but not nutritive. Last instar H. zea larvae recognized and responded positively to mannitol by metabolic feedback when it was included in a diet with casein. When offered a choice of two diets, one containing casein but lacking a utilizable carbohydrate and another lacking casein but including either mannitol or sorbose, they did not self-select for either mannitol or sorbose. However, in a similar experiment, in which the carbohydrate diet contained a mixture of mannitol and sorbose, there was a positive response, demonstrating that both metabolic feedback and phagostimulation are required to elicit self-selection.
Résumé Les chenilles d'H. zea peuvent choisir un mélange nutritif optimal parmi deux régimes séparés, répondant à des besoins alimentaires différents. Cette aptitude à l'autosélection pose la question des voies de sélection d'un régime optimal par l'insecte. L'autosélection est probablement réalisée par un feedback métabolique des aliments; la stimulation sensorielle par les aliments pourrait jouer elle aussi un rôle. Notre objectif est de séparer ces deux facteurs et d'en déterminer l'importance relative.Des L 5 venant de muer ont pu s'alimenter sur 2 régimes nutritionnellement complets, mais dont l'un contenait des carbohydrates (sucrose) mais non des protéines, tandis que l'autre contenait des protéines (caséine) mais non des carbohydrates. L'auto-sélection caséine/sucrose a été 81/19; elle correspond à la portion idéale pour le dernier stade des chenilles H. zea établie lors d'expériences antérieures (Waldbauer et al., 1984a). Pour comparer l'importance relative du feedback métabolique et de la stimulation sensorielle, 2 carbohydrates ont été substitués au sucrose, qui est à la fois nutritif et phagostimulant: le mannitol, nutritif mais ne stimulant pas de prise de nourriture, et le sorbose, faiblement stimulant mais non nutritif. Quand elles ont le choix, les chenilles ne sélectionnent ni mannitol, ni sorbose, mais consomment presque exclusivement le régime alternatif à la caséine. Quand sorbose et mannitol sont mélangés et proposés ensemble, la réponse est positive, montrant que le feedback métabolique et la stimulation sensorielle sont impliqués tous deux dans l'autosélection alimentaire.Des expériences en deux étapes d'autosélection ont montré que le feedback métabolique est de première importance. Dans la première, les chenilles avaient le choix entre caséine seule et caséine et mannitol. Les chenilles ont consommé significativement plus d'aliment avec mannitol que sans carbohydrate, bien que les goûts des deux régimes aient du être identiques, puisque le mannitol n'a pas de goût pour H. zea. La caséine a été vraisemblablement assez phagostimulante pour maintenir suffisamment les chenilles sur l'aliment avec mannitol et ainsi permettre au feedback métabolique du mannitol d'influer sur leur maintien sur cet aliment sans en rechercher un autre.Dans d'autres expériences, les chenilles ont eu le choix entre un régime alimentaire classique avec de la caséinne et d'autres n'ayant plus leur caséine mais avec des quantités décroissantes de sucrose. Elles n'ont pas réagi en consommant plus d'aliments avec du sucrose dilué, mais en consommant plus de caséine. Cependant, si du mannitol, pourtant non phagostimulant, est ajouté à des aliments avec du sucrose dilué pour remplacer le sucrose manquant, les chenilles ne compensent plus en consommant plus d'aliments avec de la caséine, mais de la même façon qu'elles le feraient avec un aliment contenant la concentration adéquate de sucrose, montrant qu'elles perçoivent ainsi le feedback métabolique du mannitol, qui est pourtant sans goût.

This paper presents results of olfactometer experiments with the egg parasitoid Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) and its host the corn earworm moth, Heliothis zea (Boddie) (Lepidoptera: Noctuidae). The sex pheromone of the host significantly reduced the total number of border crossings between odour fields in the olfactometer. Also, female parasitoids made significantly more visits to the calling moth odour field than to the opposite control field in the olfactometer. Further, the wasps spent significantly more time in the olfactometer field containing the sex pheromone released by calling virgin moths, than in control fields. If non-calling virgin moths were used as odour source, the response was reversed and wasps were repelled by the odour of the moths, and the numbers of visits were evenly distributed over the four flow fields. These results are discussed in the context of foraging ecology of egg parasitoids.
Résumé Des expériences menées en olfactométrie avec le parasite oophage Trichogramma pretiosum Riley (Hymenoptera: Trichogrammatidae) et son hôte, Heliothis zea (Boddie) (Lepidoptera: Noctuidae) donnent les résultats suivants. La présence de la phéromone sexuelle de l'hôte réduit de façon significative le nombre de passages entre les champs odorisés. De mème, les parasites visitent plus fréquemment le champ qui dispense l'odeur de la femelle en appel que le champ témoin placé à l'opposé. Par ailleurs, le temps passé dans le champ qui contient la phéromone sexuelle émise par les femelles vierges en appel est significativement supérieur à la durée de visite des champs témoins. Si l'on utilise comme source d'odeur des femelles vierges qui ne sont pas en appel, la réponse est inversée, les parasites sont alors repoussés par l'odeur de ces papillons et le nombre de visites est distribué de façon aléatoire entre les quatre champs. Ces résultats sont discutés dans le contexte de l'écologie du comportement de recherche chez les parasites oophages.

Susceptibility of laboratory and field colonies of Helicoverpa zea (Boddie) and Heliothis virescens F. to Vip3A insecticidal protein was studied in diet incorporation and diet overlay assays from 2004 to 2008. Responses of field populations were compared to paired responses of University of Arkansas laboratory susceptible H. zea (LabZA) and H. virescens (LabVR) colonies. After 7 d of exposure, observations were made on number of dead larvae (M) and the number of larvae alive but remaining as first instars (L1). Regression estimates using M (LC50) and M plus L1 (MIC50) data were developed for laboratory and field populations. Susceptibility of laboratory and field populations exposed to Vip3A varied among different batches of protein used over the study period. Within the same batch of Vip3A protein, susceptibilities of laboratory colonies of both species (LabZA and LabVR) were similar. Field colonies were significantly more susceptible to Vip3A than the respective reference colonies of both species. Within field populations, susceptibility to Vip3A varied up to 75-fold in H. zea and 132-fold in H. virescens in LC50 estimates. Variabilities in MIC50s were up to 59- and 11-fold for H. zea and H. virescens, respectively.  相似文献

An olfactometer was used to determine the effect of pheromones released by females of the bollworms Heliothis armigera (Hübner) and H. zea (Boddie) on females of the same species. Four combinations of virgin and mated females were tested for repellency of one to the other. Evidence is presented that females of the two bollworms were repelled by females of the same species. In addition, extracts of virgin female abdomens of each species repelled virgin females of the other species.
Résumé L'examen en olfactomètre a porté sur les réactions face à d'autres femelles de la même espèce, de femelles vierges ou ayant copulé d'Heliothis armigera Hübner et H. zea Boddie. Le lot comprenait 8 femelles, vierges ou ayant copulé en présence d'une femelle vierge ou ayant copulé. Les 4 combinaisons possibles de femelles vierges et de femelles ayant copulé ont été examinées avec 12 répétitions pour chaque espèce. Un extrait de l'extrémité de l'abdomen de femelles vierges d'une espèce a été présenté aux femelles de l'autre espèce pour examiner les possibilités de réactions interspécifiques aux phéromones.Pour chaque espèce, les réactions interspécifiques de répulsion entre femelles ont été hautement significatives par rapport aux témoins, à l'exception toutefois des réactions de femelle ayant copulé face à des femelles ayant elles aussi copulé. Les répulsions moyennes chez H. armigera et H. zea pour les 8 femelles de chaque expérience ont été: a) vierges en présence d'une vierge: 7,33 et 7,66; b) vierges en présence d'une femelles ayant copulé: 5,76 et 5,58; c) femelles ayant copulé en présence d'une vierge: 4,67 et 4,83. Les différences sont hautement significatives entre chaque paire de moyennes et entre chaque paire et le lot témoin; 3,17; 3,17; 3,42; 4,00 pour H. armigera; 3,17; 3,50; 2,83 et 3,75 pour H. zea.Les femelles vierges des deux espèces, H. armigera et H. zea ont présenté une réaction de répulsion en présence d'un extrait de l'abdomen de l'autre espèce; les répulsions moyennes étant respectivement 5,53 et 5,33 contre 3,83 et 3,58 pour le lot trémoin.On peut en conclure que ces répulsions doivent entraîner une tendance à la répartition uniforme.

Local dispersal ofHelicoverpa zea (Boddie) (Lepidoptera: Noctuidae) males as they emerged following the F1 and F2 generations in dent corn was examined in a 4 km diam. area in South Carolina, USA. Males were marked at artificial nectar feeding stations using RbCl, and recovered in traps baited with a synthetic pheromone. Effects of nocturnal boundary layer winds, crop pattern at the pheromone traps, and crop pattern between the marking area and pheromone traps were examined. Local movement of captured male moths was predominantly downwind in both flights in both years of the study. Crop pattern at the traps had no effect on capture of marked moths. During some flights, flowering crops between the marking area and traps had a negative effect on the number of marked moths captured.  相似文献

The parasitoidEucelatoria bryani Sabrosky regulates the larval behavior of its hostHelicoverpa zea (Boddie). Parasitized third, fourth and fifth instars burrow into the soil 0.7–3.4 days earlier than unparasitized larvae that normally enter the soil to pupate at the end of the fifth and final larval instar. Parasitized third instars molt once then burrow as fourth instars, one instar earlier than normal. WhenE. bryani pupariated on the soil surface in the field, none survived to the adult stage. However,E. bryani adults emerged from 49.2% of hosts that had burrowed into the soil. By accelerating the timing ofH. zea burrowing behavior and causing host larvae to enter the soil before death,E. bryani ensures its pupariation in an environment with improved protection against natural enemies and lethal temperatures.  相似文献

We have developed a monoclonal antibody that recognizes the larval and adult stages ofMicroplitis croceipes(Cresson) (Hymenoptera: Braconidae) andCotesia marginiventris(Cresson) (Hymenoptera: Braconidae), two principal endoparasitoids of the polyphagous pestsHelicoverpa zea(Boddie) (Lepidoptera: Noctuidae) andHeliothis virescens(F.) (Lepidoptera: Noctuidae). The antibody has been incorporated into an indirect enzyme-linked immunosorbent assay that can be used to distinguish parasitized from unparasitized pests. The antibody cross-reacts with several different hymenopteran parasitoids, but not with any of the noctuid pests we have assayed. An immunoassay based on a broadly reactive antibody such as this one enables comprehensive detection of hymenopteran parasitism with a single antibody reagent.  相似文献

We examined longevity, fecundity, and oviposition strategies ofEucelatoria bryani Sabrosky (Diptera: Tachinidae), a gregarious endoparasitoid ofHelicoverpa zea (Boddie) andHeliothis virescens (F.) (Lepidoptera: Noctuidae). Longevity of adult femaleE. bryani was not related to body size. In contrast to longevity, largerE. bryani females had greater potential fecundity than smaller females, as determined by the number of embryonated eggs present in the common oviduct. However, female parasitoid size did not affect primary clutch size (number of eggs deposited in a host). Because embryos in eggs located in the ovisac were larger than those located elsewhere in the common oviduct, maximum primary clutch size may be physiologically limited by the number of fully mature eggs a female has available at one time.E. bryani females adjusted primary clutch size in response to host size, for bothH. zea andH. virescens. This adjustment appears to be adaptive because females did not overexploit hosts by depositing more larvae than a host could support. Adult emergence was not related to host size. Although host weight positively influencedE. bryani progeny weight, increases in progeny size with host size were counterbalanced by increases in primary clutch size with host size.  相似文献

【目的】研究微嗜酸寡养单胞菌(Stenotrophomonas acidaminiphila)CW117中磷酸吡哆胺氧化酶基因pnpox(phosphopyridoxamine oxidase,pnpox)在维生素B6(VB6)合成中的贡献及对黄曲霉毒素B1(aflatoxin B1,AFB1)的降解活性。【方法】采用基因插入突变方式,对菌株CW117中磷酸吡哆胺氧化酶基因pnpox进行突变,得到突变菌株。通过高效液相色谱法(high performance liquid chromatography,HPLC)检测突变株对AFB1的降解活性,以及突变株中吡哆醇和吡哆醛的合成情况,确定基因pnpox在寡养单胞菌体内VB6合成中的贡献和黄曲霉毒素降解代谢作用。【结果】成功构建了磷酸吡哆胺氧化酶基因突变子pnpox::pK19mobΩ2HMB,突变子吡哆醛的合成量较野生型菌株显著减少,吡哆醇合成量与野生型菌株无显著性差异;同时,突变子与野生型株CW117对AFB1的降解活性未发现显著性差异。【结论】菌株CW117中磷酸吡哆胺氧化酶在吡哆醛合成的过程中起着重要作用,该基因突变会导致VB6的严重缺乏,影响寡养单胞菌正常生长,但该基因对CW117降解黄曲霉毒素无显著性贡献。  相似文献

古丸菌纲(Archaeoglobi)是广古菌门下的纲级分类单元,包含古丸菌(Archaeoglobus)、地丸菌(Geoglobus)和铁丸菌(Ferroglobus)三个属,所属菌株均是严格嗜热厌氧菌,主要分布于海洋、陆地热液系统和油田环境中。Archaeoglobus属下的微生物是一类以硫酸盐、亚硫酸盐或硫代硫酸盐为电子受体代谢生成硫化氢(H2S)的化能自养或氢营养型微生物;而Geoglobus和Ferroglobus的成员则主要还原硝酸盐和铁离子。Archaeoglobi地理分布广泛,在元素生物地球化学循环过程中发挥着重要作用,是目前微生物生态学研究的一个热点。在进化方面,Archaeoglobi菌和产甲烷古菌具有较高的亲缘关系;同时,Archaeoglobi基因组中保留着部分产甲烷途径上的功能基因,最新研究表明部分未培养的Archaeoglobi基因组中含有完整的产甲烷通路。这些证据都表明Archaeoglobi菌的基因组特征可能是产甲烷古菌向硫酸盐还原菌进化的活化石。本文梳理了目前发现的11株Archaeoglobi菌株的生理生化特征和基因组分析结果,从化能自养、化能异养、硫化物呼吸、产乙酸、产甲烷等方面综述了已分离的Archaeoglobi菌的代谢特征,并基于宏基因组信息分析了未培养的Archaeoglobi菌基因组中的潜在代谢功能,为进一步分离培养此类未培养厌氧微生物提供理论指导。  相似文献

Newly-ecdysed last instar larvae ofH. zea grouped into 100-, 200-, 300-, or 400-mg categories were fed diet containing precocene II or given precocene II topically on the abdomen. The time for larvae to reach a maximal weight, time to pupation, growth rate, and the amount of precocene II excreted were calculated. Younger larvae of lower weights, which were fed or topically treated with precocene II required more time to reach their maximal weight, had a lower maximal weight, a lower growth rate, and required more time to pupate than control larvae. Older larvae represented by the largest weight category were less sensitive to precocene II, had a shorter delay in reaching maximal weight, and a shorter delay in the time to pupation than control larvae; larvae in the largest weight category that were fed precocene II also had smaller decreases in the growth rate. Growth rate declines for larvae given topical doses of precocene II, however, were largest for the oldest larvae. All larvae given a single topical dose excreted precocene II for several days and were most efficient at eliminating smaller doses; larger, older larvae excreted more precocene II than smaller, younger larvae. Age-dependent responses to precocene II indicate that growth and metabolic processes, as well as xenobiotic metabolism, change in last instar larvae.  相似文献

【目的】吲哚-3-乙酸是调控植物生长发育和生理活动的重要激素,吲哚-3-乙酸N-乙酰转移酶YsnE在吲哚-3-乙酸合成中发挥重要作用,本研究拟解析解淀粉芽胞杆菌中YsnE参与吲哚-3-乙酸合成的代谢途径。【方法】通过基因ysnE缺失和强化表达,分析ysnE对吲哚-3-乙酸合成影响,结合吲哚-3-乙酸合成中间物(吲哚丙酮酸、吲哚乙酰胺、色胺和吲哚乙腈)添加和体外酶转化实验,解析ysnE参与吲哚-3-乙酸合成的代谢途径。【结果】明确了YsnE在解淀粉芽胞杆菌HZ-12吲哚-3-乙酸合成中发挥重要作用。发现ysnE缺失菌株中的吲哚丙酮酸、吲哚乙酰胺和吲哚乙腈利用显著降低,揭示了YsnE主要发挥吲哚丙酮酸脱羧酶YclB和吲哚乙酰胺水解酶/腈水解酶/腈水合酶YhcX的功能,并通过参与吲哚丙酮酸、吲哚乙酰胺和吲哚乙腈途径来影响吲哚-3-乙酸合成。【结论】初步揭示了YsnE通过影响吲哚丙酮酸、吲哚乙酰胺和吲哚乙腈途径参与吲哚-3-乙酸合成的代谢机理,为吲哚-3-乙酸合成途径解析和代谢工程育种构建吲哚-3-乙酸高产菌株奠定了基础。  相似文献

Cytochrome c oxidase (EC 1.9.3.1) is one of the components of the electron transport chain by which Nitrobacter, a facultative lithoautotrophic bacterium, recovers energy from nitrite oxidation. The genes encoding the two catalytic core subunits of the enzyme were isolated from a Nitrobacter winogradskyi gene library. Sequencing of one of the 14 cloned DNA segments revealed that the subunit genes are side by side in an operon-like cluster. Remarkably the cluster appears to be present in at least two copies per genome. It extends over a 5–6 kb length including, besides the catalytic core subunit genes, other cytochrome oxidase related genes, especially a heme O synthase gene. Noteworthy is the new kind of gene order identified within the cluster. Deduced sequences for the cytochrome oxidase subunits and for the heme O synthase look closest to their counterparts in other -subdivision Proteobacteria, particularly the Rhizobiaceae. This confirms the phylogenetic relationships established only upon 16S rRNA data. To explore a new approach of high expression of -amino acid oxidase (DAAO) in Pichia pastoris, a gene encoding DAAO from Trigonopsis variabilis (TvDAAO gene) deleted intron was prepared by PCR amplification and cloned into the intracellular expression vector pPIC3.5K. The expression plasmid pPIC3.5K-DAAO linearized by SalI was transformed into Pichia pastoris strain GS115 (hismut+). By means of MM and MD plates and PCR, the recombinant P. pastoris strains (his+mut+) were obtained. Activity assay and SDS-PAGE demonstrated that DAAO was intracellularly expressed in P. pastoris with the induction of methanol. The recombinant strain PD27 with the highest expression of DAAO was screened through activity assay and its high-density fermentation was carried out in a 1-l fermentor. Activity assay and SDS-PAGE demonstrated that DAAO was intracellularly expressed in P. pastoris with the induction of methanol. The recombinant cells with high expression of DAAO were screened and the high-density fermentation was carried out in a 1-l fermentor. Interestingly, the DAAO expression level reached up to 473 U/g dry cell weight in fermentation yield. Finally, 1-hexanol was used to break recombinant cells and the specific activity of DAAO was 1.46 U/mg protein in crude extraction.  相似文献

Using benzo(a)pyrene (BaP) as a probe for aryl hydrocarbon hydroxylase (AHH) activity, differences in mixed-function oxidase (MFO) activity were observed using microspectrofluorimetry in single living cells during long term treatment with 3-methylcholanthrene (3-MC) or carbaryl. Although these two compounds differ in chemical structure, similar effects were observed in 3T3 cell populations. The results suggest that the two compounds activate the same enzymatic system and that individual cells of a supposed homogeneous cell population are not equally sensitive to xenobiotics, i.e. subpopulations were observed which have differences in AHH activity.  相似文献

【目的】本研究旨在构建单增李斯特菌末端细胞色素aa3氧化酶亚基qoxB基因缺失株,并探索其在细菌生长及感染过程中发挥的生物学功能。【方法】利用同源重组方法构建获得缺失株ΔqoxB后,对野生株EGD-e和缺失株ΔqoxB的生长能力、细菌运动能力和细胞内黏附、侵袭、增殖及胞内迁移能力进行比较,同时利用荧光定量PCR方法检测ΔqoxB中鞭毛相关基因转录水平的变化。【结果】缺失qoxB基因后细菌在体外培养过程中生长能力没有差异,细菌的鞭毛运动能力显著降低,在30℃培养24 h和48 h后ΔqoxB运动圈直径分别较EGD-e下降35.86%和34.20%,且22个鞭毛相关基因转录水平显著降低。通过细胞感染试验发现缺失qoxB基因后细胞黏附、侵袭、增殖及胞内迁移能力均显著下降。【结论】本研究首次证实末端氧化酶亚基QoxB能降低单增李斯特菌的运动能力和对细胞的感染能力,此研究为进一步阐明末端细胞色素氧化酶影响单增李斯特菌的致病机制提供重要依据。  相似文献

Spodoptera exigua is one of the most renowned agricultural pest insects and relatively insensitive to Bacillus thuringiensis subsp. kurstaki strains which are widely used commercial products to control lepidopterans such as Heliothis armigera. In the current study, we have developed a new and efficient approach to screen and breed a B. thuringiensis subsp. kurstaki strain exhibiting high toxicity against S. exigua while retaining its high toxicity against H. armigera. UV and diethyl sulfate methods were used for mutagenesis, followed by an agar plug plate diffusion assay for preliminary screening of Zwittermicin A over-producing mutants, from which we obtained a mutant strain, designated here as B. thuringiensis subsp. kurstaki D1-23, with high toxicity against S. exigua. The toxicity of D1-23 against S. exigua and H. armigera was improved by 115.4 and 25.9%, respectively, compared to its parental commercial strain BMB005.  相似文献

采用正交试验设计法,研究了NAA、KT、2,4-D、蔗糖浓度和不同外植体类型等因素对银杏愈伤组织诱导的影响。结果表明:不同外植体类型对银杏愈伤组织诱导率影响最大,KT和NAA其次,2,4-D和蔗糖浓度最小。银杏愈伤组织诱导最佳培养基为MS+NAA 0.5 mg·L-1+KT 0.5 mg·L-1+蔗糖40 g·L-1,最佳外植体为茎段,其愈伤组织诱导率可达100%。  相似文献

The effects of notonectid-induced crests on growth and reproduction, and resource allocation to crest construction, moult losses and eggs of Daphnia carinata were measured. An attempt was made to elucidate the mechanisms of physiological costs of crest induction for this species. The crested morph of d. carinata reached a significantly larger size than the uncrested form. Reproductive output was similar in early instars, but the crested morph produced more eggs in latter broods. Instar duration was longer for the crested morph and age at first reproduction was delayed. Survival was also lower in this form. Crest construction required significant resources (equivalent to 60 eggs over a life time) but evidence is presented that these resources were obtained primarily by re-allocation of available material rather than collection of extra resources. The crested morph allocated significantly more resources to moulting than its uncrested counter-parts. The uncrested morph produced large eggs in early instars and progressively smaller ones in later instars. The crested morph produced only small eggs. The hypothesis is presented that the crest-induction strategy of D. carinata involves at least two separate sets of responses, each with its own costs and trade-offs. The first response is production of the crest. The cost of crest production is an increased cost of moulting. D. carinata off sets this cost by increasing instar duration and thus age at reproduction. The second response is increased size. D. carinata achieves this by reducing the fraction of available resources allocated to reproduction. The cost is lower reproductive output.  相似文献
