RNA polymerase II from larvae of the brine shrimp, Artemia salina, was highly purified by two cycles of DEAE-cellulose chromatography followed by centrifugation through discontinuous sucrose gradients. Gradient fractions were subjected to elctrophoresis is polyacrylamide gels containing sodium dodecyl sulfate. The subunit structure of RNA polymerase II was determined by quantitative comparison of the polypeptides and enzyme activity present in each gradient fraction. The enzyme contains one copy of each of four subunits with estimated molecular weights of 170,000, 130,000, 36,000 and 24,000. The total molecular weight agrees well with the molecular weight estimated for the native enzyme by density gradient centrifugation.  相似文献

The main classes of RNA polymerase in cultured chick embryo skeletal muscle cells were identified and their levels determined during differentiation in vitro. Cell cultures contained a high proportion of muscle cells (>80%) and exhibited a high degree of fusion (>80%). The levels of RNA polymerases I and II increased by 1.8 and 1.5 times, respectively, from prefusion myoblasts (24 hr) to postfusion myotubes (120 hr). RNA polymerase III, a single peak on DEAE-Sephadex chromatography, was less than 7% of the total activity. The results suggest that the rates of synthesis of the main RNA types, which reportedly decline during myogenesis, are not determined by the relative levels of the main RNA polymerases and are more likely a reflection of chromatin template availability.  相似文献

Partial purification and characterization of DNA-dependent RNA-polymerases from nauplius larvae of the brine shrimp, Artemia salina, are described. Fractionation of solubilized RNA-polymerases on columns of DEAE-cellulose yielded partially purified preparations of RNA polymerases I and II. The properties of these enzymes were found to be similar to properties of corresponding enzymes from other animal sources. A significant change in the relative amounts of polymerases I and II occurs between 36 and 72 hr of development. Polymerase activity obtained from 36-hr nauplii consisted of approximately equal amounts of polymerases I and II, whereas polymerase II accounted for more than 80% of the activity recovered from 72-hr nauplii. Total polymerase activity was lower at 72 than at 36 hr. The significance of these changes in relation to the decrease in RNA synthesis in vivo that occurs after 36 hr is discussed.  相似文献

The DNA-dependent RNA polymerases of Schneider 2-L cells of Drosophila melanogaster are described. These cells contain five readily detectable forms of this enzyme, polymerases Ia, Ib, IIIa, II, and IIIb, which elute from DEAE-Sephadex at 0.08, 0.12, 0.15, 0.20, and 0.22 m ammonium sulfate, respectively. RNA polymerases IIIa and IIIb, which each constitute about 5–10% of the total RNA polymerase activity in Drosophila embryos, are found to constitute 30 and 10%, respectively, of the total polymerase activity in cultured cells. The two form III polymerases are further characterized by in vitro response to divalent cations and ionic strength, template utilization, and sensitivity to -amanitin. Verification of the class III designation of these two polymerases is provided by their sensitivity to only very high levels of -amanitin (50% inhibition at approximately 800 µg/ml), their 10-fold greater activity on poly[d(A–T)], and their elution from DEAE-cellulose at lower ionic strengths than from DEAE-Sephadex.This work was supported by the Natural Sciences and Engineering Research Council.  相似文献
