Lipase-catalyzed optical resolution of trifluoro(aryl)ethanols   总被引:2,自引:0,他引:2  
Optical resolutions of racemic 2,2,2-trifluoro-1-(aryl)ethanols — (1-naphthyl), (2-naphthyl), (4-methylnaphthyl), (phenyl), (1-pyrenyl) — were achieved by lipase-catalyzed enantioselective acetylations with vinyl acetate as an acetyl donor in octane, and (S)-acetates and (R)-alcohols were obtained. Among the lipases tested, lipase from Pseudomonas aeruginosa (lipase LIP, Toyobo) showed good enantioselectivity for above ethanols. However, no acetylation occurred with sterically hindered alcohols — (9-phenanthryl), (9-anthryl), (2-methylnaphthyl), (2, 4, 6-trimethylphenyl) — by various lipases. The resolutions of the three alcohols were carried out by the enantioselective alcoholysis or hydrolysis of their chloroacetates by lipase LIP.  相似文献

Transesterification of 4-allyl-1-hydroxymethylazetidin-2-ones using vinyl acetate in the presence of the lipase from Pseudomonas cepacia led to the corresponding (R)-acetates and the remaining (S)-alcohols in high yields and excellent ee's (E: 32–71). Chemo-enzymatic approaches for the synthesis of the family of aromatase inhibitory drug via lipase-catalyzed kinetic resolution of (±)-4-cyano-4-phenyl-1-hexanol (2) as appropriate precursors were described. Enzymatic transesterification of primary alcohol (±)-2 using Pseudomonas cepacia (Amano PS, PCL) provided the enantiopure alcohol (R)-(−)-2 with 99% ee at conversion of 86%, while that of (±)-2 using Pseudomonas fluorescens (Amano AK, LAK) provided the (S)-(+)-2 with 96% ee at conversion of 86%. Chemical transformation of substrate (R)-(−)-2 gave (R)-(+)-aminoglutethimide (1) in enantioselectively high yield.  相似文献

From the dichloromethane extract of the flowers of Helichrysum gymnocomum (Asteraceae) two known flavonoids, 4 and 5, and a known acylphloroglucinol, 3B, were isolated. In addition to 1 and 2, the 4′,6′-dibenzyloxy-2′-hydroxy derivative of 2′,4′,6′-trihydroxychalcone and 5,7-dibenzyloxy derivative of pinocembrin, respectively, are reported in Nature for the first time. A compound 3A, related to 3B has the structure 2-methyl-1-[2,4,6-trihydroxy-3-(2-hydroxy-3-methyl-3-butenyl)phenyl]-1-propanone. Compounds 1, 2, 3A, 3B, 4 and 5 have MIC values below 64 μg/ml against a selection of pathogens, with 3B having the highest sensitivity (6.3–45 μg/ml) for eight of the ten pathogens tested, including Staphylococcus aureus (6.3 μg/ml) and methicillin and gentamycin resistant strain of S. aureus (7.8 μg/ml). With the exception of 2, the other compounds had notable activity (45–63 μg/ml) towards Pseudomonas aeruginosa.  相似文献

Pseudomonas fluorescens DSM 7155 after growth on phenylacetonitrile as sole nitrogen source contained an inducible nitrilase which consists of two different functional subunits (40 and 38 kDa). The nitrilase catalysed the exclusive hydrolysis of arylacetonitrile substrates into the equivalent carboxylic acids plus ammonia as major products. The corresponding amides were formed at low levels (<5%) during nitrile hydrolysis but were not substrates for the purified enzyme. The native enzyme, which had a pH optimum of 9 and a temperature optimum of 55°C, was activated (140–160%) by the thiol protectant 2-mercaptoethanol (50–100 mM). The purified nitrilase catalysed the hydrolysis of the two enantiomers of racemic 2-(methoxy)-mandelonitrile to the corresponding acid at significantly different rates: at 50% overall conversion the predominant product was the (R)-acid (enantiomeric excess=92%) whereas at 85% overall conversion the ee% of the (R)-acid had decreased to 27%.  相似文献

In order to solve discrepancies between earlier assignments we have reinvestigated the stereoisomerism of the spheroidene molecule bound to reaction centers (RC) of Rhodobacter sphaeroides. A stable cis isomer could be extracted and purified from the reaction centres by working at very low ambient light. Resonance Raman spectroscopy showed that this cis isomer assumed the same configuration as that of the RC-bound molecule. Proton-NMR spectroscopy of the extracted isomer permitted to assign it the 15–15′ mono cis configuration. Comparisons between resonance Raman spectra of the native form and of the 15 cis extract showed that, in the reaction center, 15 cis spheroidene is in addition twisted into a non-planar conformation. Comparisons of extraction-induced changes in relative intensities of Raman bands of the 760–1060 cm−1 regions, which largely correspond to out-of-plane modes, further indicated that the out-of-plane twist of RC-bound spheroidene should predominantly affect C8–C12 and/or C8′–C12′ regions of the molecule rather than the central region. Comparisons between difference electronic absorption spectra of RC-bound spheroidene and of RC-bound methoxyneurosporene showed that the out-of-plane twisting of both these native forms results in a drastic weakening of their 1C ← 1A electronic transitions, compared with those of the planar, 15 cis forms. Finally, it is proposed, on the basis of their resonance Raman spectra, that spirilloxanthin bound to RCs of Rhodospirillum rubrum as well as dihydroneurosporene or dihydrolycopene bound to RCs of Rhodopseudomonas viridis shares 15 cis configurations and out-of-plane twisting with carotenoids bound to RCs of various strains of Rb. sphaeroides.  相似文献

The biosynthesis of poly(3-hydroxyalkanoates) (PHAs) by Pseudomonas cichorii YN2 cultured with C6–C12 1-alkenes was studied. PHAs containing repeating units with terminal epoxide groups were obtained when C7–C12 1-alkenes were fed separately as the only carbon source, but no epoxidized unit was detected when 1-hexene was fed. The content of epoxidized units in the polymers was in the range of 4–20 mol%, which was not dependent on the C atom length of the 1-alkene used as a substrate. The polymers produced undergo a glass transition at around −40 °C, and number average molecular weights were in the range of 1 50 000–2 00 000 as determined by GPC relative to polystyrene, with Mw/Mn ratios of 1.9–2.5. As an intermediate, the corresponding 1,2-epoxyalkane was found in the culture medium. According to this result, the epoxidation of the 1-alkene is the initial step in the synthetic pathway of the epoxy unit in the polymer.  相似文献

A new method for the oxidation of primary ad secondary allylic alcohols to the corresponding aldehydes/acids and ketones respectively is described utilizing Nocardia corallina B-276. Regioselective derivatisation of resveratrol (1) at positions 3, 5 or 4′ was achieved by a chemo-enzymatic procedure based on standard chemical reactions and esterification or alcoholysis in organic solvents catalysed by the commercially available Pseudomonas cepacia (PcL) and Candida antarctica (CaL) lipases.  相似文献

A 125-member library of synthetic linear undecapeptides was prepared based on a previously described peptide H-K1KLFKKILKF10L-NH2 (BP76) that inhibited in vitro growth of the plant pathogenic bacteria Erwinia amylovora, Xanthomonas axonopodis pv. vesicatoria, and Pseudomonas syringae pv. syringae at low micromolar concentrations. Peptides were designed using a combinatorial chemistry approach by incorporating amino acids possessing various degrees of hydrophobicity and hydrophilicity at positions 1 and 10 and by varying the N-terminus. Library screening for in vitro growth inhibition identified 27, 40 and 113 sequences with MIC values below 7.5 μM against E. amylovora, P. syringae and X. axonopodis, respectively. Cytotoxicity, bactericidal activity and stability towards protease degradation of the most active peptides were also determined. Seven peptides with a good balance between antibacterial and hemolytic activities were identified. Several analogues displayed a bactericidal effect and low susceptibility to protease degradation. The most promising peptides were tested in vivo by evaluating their preventive effect of inhibition of E. amylovora infection in detached apple and pear flowers. The peptide H-KKLFKKILKYL-NH2 (BP100) showed efficacies in flowers of 63–76% at 100 μM, being more potent than BP76 and only less effective than streptomycin, currently used for fire blight control.  相似文献

The use of (R)-specific enoyl-coenzyme A (CoA) hydratase (PhaJ) provides a powerful tool for polyhydroxyalkanoate (PHA) synthesis from fatty acids or plant oils in recombinant bacteria. PhaJ provides monomer units for PHA synthesis from the fatty acid ß-oxidation cycle. Previously, two phaJ genes (phaJ1Pa and phaJ2Pa) were identified in Pseudomonas aeruginosa. This report identifies two new phaJ genes (phaJ3Pa and phaJ4Pa) in P. aeruginosa through a genomic database search. The abilities of the four PhaJPa proteins and the (R)-3-hydroxyacyl-acyl carrier protein [(R)-3HA-ACP] dehydrases, FabAPa and FabZPa, to supply monomers from enoyl-CoA substrates for PHA synthesis were determined. The presence of either PhaJ1Pa or PhaJ4Pa in recombinant Escherichia coli led to the high levels of PHA accumulation (as high as 36–41 wt.% in dry cells) consisting of mainly short- (C4–C6) and medium-chain-length (C6–C10) 3HA units, respectively. Furthermore, detailed characterizations of PhaJ1Pa and PhaJ4Pa were performed using purified samples. Kinetic analysis revealed that only PhaJ4Pa exhibits almost constant maximum reaction rates (Vmax) irrespective of the chain length of the substrates. The assay for stereospecific hydration revealed that, unlike PhaJ1Pa, PhaJ4Pa has relatively low (R)-specificity. These hydratases may be very useful as monomer-suppliers for the synthesis of designed PHAs in recombinant bacteria.  相似文献

New di-Schiff base type crown ethers crosslinked chitosan (CCTS-1) was synthesized by the reaction of 4,4′-diformyldibenzo-18-c-6 crown ether with crosslinked chitosan. New di-secondary amine type crown ethers crosslinked chitosan (CCTS-2) was prepared by the reaction between CCTS-1 and sodium borohydride. Their structures were confirmed by Fourier transform infrared spectral analysis, X-ray powder diffraction analysis and elemental analysis. The adsorption rates by CCTS-2 for Ag+ for 1 h were 96% at pH 6.0, and Ag+ initial concentration 0.5 mmol/l. The complexes of CCTS [Bull. Chem. Soc. Jpn 60 (1987) 444], CCTS-2 and silver ion against three bacteria were studied. The bacteriostasis zone diameters of the complex of CCTS-2 and Ag+ (CCTS-2–Ag+) containing 0.00355 mmol Ag+ against Staphylococcus Aureus, Escherichia coli and Pseudomonas aeruginona are 11, 10 and 7.5 mm, respectively, while those of the complex of CCTS and Ag+ (CCTS–Ag+) under similar conditions were 11, 10 and 6.0 mm, respectively. This research will be useful for designing crosslinked-chitosan-based adsorption for preconcentration of Ag+ for medical bacteriostasis.  相似文献

The lipopolysaccharide (LPS) of a galU mutant of Pseudomonas aeruginosa PA103, a serogroup O11 strain, was sequentially extracted with phenol–chloroform–petroleum ether (PCP) followed by hot phenol–water extraction of the bacterial pellet remaining after PCP extraction. LPS was found in both the PCP extract as well as in the water phase of the hot phenol–water extract. Analysis of the carbohydrate portion released by mild acid hydrolysis of both LPS preparations, both before and after removal of all phosphate groups by treatment with aqueous HF, was performed by glycosyl composition and linkage analyses as well as by NMR and mass spectrometric analyses. The results showed that the carbohydrate portion of these two LPS extracts contained the same structure: namely, -GalN(Ala)-(1→3)--(7-Cm)HepII-(1→3)--HepI-(1→5)--Kdo-(2→. The oligosaccharide preparation from PCP-extracted LPS consisted of a variety of structures containing up to six phosphate groups present as mono-, pyro-, and possibly triphosphate, primarily located on the HepI residue with some molecules having a monophosphate on HepII. The oligosaccharide preparation from the hot phenol–water-extracted LPS contained a similar variety of structures, but with an additional structure in which HepI contained a PPEA group at O-2. In addition, PAGE immunoblot analysis of the crude cellular extract with anti-A-antibodies revealed the presence of A-band material in both PA103 and the galU mutant. The A-band material was purified and characterized by glycosyl composition and linkage analyses, as well as by NMR spectroscopy, which confirmed that the A-band rhamnan polysaccharide was present but not as typical LPS since lipid-A or LPS core oligosaccharide components were not detected.  相似文献

Enantioselective acetylation (desymmetrization) of prochiral 2-(ferrocenylmethyl)propane-1,2-diol (1), 2-(2-ferrocenylethyl)propane-1,2-diol (2) and 2-(3-ferrocenylpropyl)propane-1,2-diol (3) into chiral monoacetates [(+)-4-(+)-6], with a series of microbial lipases in benzene at 27°C, revealed the lipase from Pseudomonas sp (PSL) as the most selective. Acetylation was fastest and most enantioselective for conversion 1→(+)-4 by PSL (97% e.e.). By comparison of the compounds (+)-4-(+)-6 with their benzene analogues of the known (R) absolute configuration, on the basis of their elution orders on Chiracel OD, and the same direction of their optical rotations, an R-configuration is proposed for (+)-monoacetates 4-6.  相似文献

The synthesis of the 3-heptyl, and the eleven isomeric 3-methylheptyl-Δ8-tetrahydrocannabinols (3–7, R and S methyl epimers, and 8) has been carried out. The synthetic approach entailed the synthesis of substituted resorcinols, which were subjected to acid catalyzed condensation with trans-para-menthadienol to provide the Δ8-THC analogue. The 1′-, 2′- and 3′-methylheptyl analogues (3–5) are considerably more potent than Δ8-THC. The 4′-, 5′- and 6′-methylheptyl isomers (6–8) are approximately equal in potency to Δ8-THC.  相似文献

A newly isolated enantioselective esterase from Pseudomonas fluorescens KCTC 1767, which is currently considered as a biocatalyst for the production of a commercially valuable (S)-ketoprofen, has revealed a low structural and thermal stability. In order to enhance the stability, directed evolution was attempted on this enantioselective esterase by successive steps of an error prone and staggered extension PCR. After the second round of evolution, the best mutant 6–52 with enhanced thermal stability was selected and analyzed. DNA sequence analyses of 6–52 revealed that the three amino acid residues (L120P, I208V, and T249A) were changed and the mutation L120P was presumed as a structurally important residue due to its presence in all positive variants. The purified mutant 6–52, when incubated at 50 and 55 °C for 2 h, remained its activity over 30 and 10%, respectively, whereas there were no detectable activities in wild-type enzyme. The analysis of 6–52 in the presence of 15% ethanol showed 1.8-fold increase in the activity, compared to that of wild-type enzyme. The Km and Vmax values of 6–52 were estimated to be slightly increased, leading to 1.2-fold-higher the catalytic efficacy kcat/Km than that of wild-type enzyme. Additionally, the mutant 6–52 was more resistant to high substrate concentrations than that of wild-type enzyme.  相似文献

Four new peptides of the mastoparan family, characterized recently in the venom of three neotropical social wasps collected in the Dominican Republic, Polistes major major, Polistes dorsalis dorsalis and Mischocyttarus phthisicus were synthesized and tested for antimicrobial potency against Bacillus subtilis, Staphylococcus aureus, Escherichia coli (E.c.) and Pseudomonas aeruginosa, and for hemolytic and mast cells degranulation activities. As these peptides posses strong antimicrobial activity (minimal inhibitory concentration (MIC) values against Bacillus subtillis and E.c. in the range of 5–40 μM), we prepared 40 of their analogs to correlate biological activities, especially antimicrobial, with the net positive charge, hydrophobicity, amphipathicity, peptide length, amino acid substitutions at different positions of the peptide chain, N-terminal acylation and C-terminal deamidation. Circular dichroism spectra of the peptides measured in the presence of trifluoroethanol or SDS showed that the peptides might adopt -helical conformation in such anisotropic environments.  相似文献

Directional modifications of resibufogenin 1 by Mucor subtilissimus and Pseudomonas aeruginosa were carried out. The substrate was hydroxylated at C-12 by M. subtilissimus AS 3.2454, from which a major product 12-hydroxyresibufogenin 2 was obtained. Then product 2 was dehydrogenated by P. aeruginosa AS 1.860, which resulted in a new compound 12β-hydroxy-3-keto-resibufogenin 3.  相似文献

Racemic secondary alcohols were resolved via enantioselective acylation using succinic anhydride as acyl donor catalysed by lipase from Pseudomonas cepacia supported on celite (PS-C) in ionic liquid, 1-butyl-3-methylimidazolium hexafluorophosphate [bmim]PF6. The synthesis and antibacterial activity of novel 1β-methylcarbapenems 1a–f bearing oxyiminopyrrolidinylamide moiety at C-5 position of pyrrolidine are described. Most compounds exhibited comparable antibacterial activity to meropenem against a wide range of Gram-positive and Gram-negative organisms including Pseudomonas aeruginosa isolates. Of these carbapenems, 1a showed potent and broad spectrum of antibacterial activity and similar stability to DHP-I to meropenem. Against clinical isolates of 40 Gram-negative bacterial species including MDR and ESBL-producing strains, the selected carbapenem 1a possessed excellent in vitro activity except for MDR P. aeruginosa, and was comparable in potency to meropenem.  相似文献
