Salinity is an increasing environmental problem in agricultural ecosystems and is not adequately represented in conventional life cycle assessment (LCA) impact categories. It is often not the total quantity of salts emitted or the proportion of salt accumulated in the soil profile that is the primary mechanism for deteriorating soil conditions for irrigated salinity, rather the ratio of major cations in the soil matrix and the potential for colloid dispersion and reduced permeability. A soil salinisation potential (SP) is proposed as an indicator for irrigated salinity and potential soil degradation from poor irrigation practices. The indicator uses the threshold electrolyte concentration concept that predicts the adjusted sodium adsorption ratio (SAR)/ Electrical conductivity (EC) ratio that soil will no longer flocculate, but potentially disperse. The SAR is converted to a threshold EC and compared to the measured EC in order to develop a site-specific irrigation equivalence factor (EF). This site/region/process specific EF is then used to weight the sodium load to soil and repeated for each stage throughout the entire life cycle to determine the overall Salinisation Potential (SP). The data required for calculating the SP is generally readily available either on site or from the water chemistry of the local watercourses. Preliminary calculations simply require the volume, pH, electrical conductivity (EC), alkalinity and the concentrations of Na, Ca, and Mg of the irrigation water. The site/process/region specific nature of the indicator ensures a quantitative measure to enable comparisons between different systems and is useful for identifying stages in the life cycle of a product (particularly food products), where the potential for soil salinisation and soil degradation is most severe.  相似文献

Neurokinin A (NKA), substance P (SP) and the two peptides combined (SP + NKA) were injected intracutaneously on the forearm and into the temporal muscle of healthy volunteers. Pain intensity, cutaneous wheal and flare responses and tenderness of the temporal muscle were quantitated. SP but not NKA induced cutaneous pain. This relates the algesic effect of SP to the specific N-terminal amino acid sequence of the peptide, not shared by NKA. NKA, however, potentiated the algesic effect of SP as SP + NKA induced a significantly prolonged cutaneous pain sensation. Both peptides induced wheals, but only SP induced flare. These results confirm previous studies relating wheal formation to the identical C-terminal amino acid sequence of the two peptides and flare reaction to the N-terminal part of SP. Injections into the temporal muscle did not cause pain or tenderness.  相似文献

Evoked striatal field potentials are seldom used to study corticostriatal communication in vivo because little is known about their origin and significance. Here we show that striatal field responses evoked by stimulating the prelimbic cortex in mice are reduced by more than 90% after infusing the AMPA receptor antagonist CNQX close to the recording electrode. Moreover, the amplitude of local field responses and dPSPs recorded in striatal medium spiny neurons increase in parallel with increasing stimulating current intensity. Finally, the evoked striatal fields show several of the basic known properties of corticostriatal transmission, including paired pulse facilitation and topographical organization. As a case study, we characterized the effect of local GABA(A) receptor blockade on striatal field and multiunitary action potential responses to prelimbic cortex stimulation. Striatal activity was recorded through a 24 channel silicon probe at about 600 μm from a microdialysis probe. Intrastriatal administration of the GABA(A) receptor antagonist bicuculline increased by 65±7% the duration of the evoked field responses. Moreover, the associated action potential responses were markedly enhanced during bicuculline infusion. Bicuculline enhancement took place at all the striatal sites that showed a response to cortical stimulation before drug infusion, but sites showing no field response before bicuculline remained unresponsive during GABA(A) receptor blockade. Thus, the data demonstrate that fast inhibitory connections exert a marked temporal regulation of input-output transformations within spatially delimited striatal networks responding to a cortical input. Overall, we propose that evoked striatal fields may be a useful tool to study corticostriatal synaptic connectivity in relation to behavior.  相似文献

Ten qualitative traits were observed in two isolated human populations in Bosnia for prewar and postwar periods. Due to recent war in Bosnia and Herzegovina two million citizens was forced to migrate. Dramatic migration effects occurred in the investigated populations. Population characteristics found were geographic isolation, high levels of marital distance “zero”, patrilocality and propagation isolation. Relative recessive allele frequencies of ten qualitative traits were calculated. Average heterozigosity, genetic differentiation, total heterozigosity, within populations heterozigosity, genetic distance and relative measures based on results of genetic distance analysis were observed. The total number of observed individuals was 1875. We have observed 1209 individuals in prewar period (Bijela population 731 and Memici population 478) and 666 individuals in the postwar period (Bijela population 248 and Memici population 478). Results ofFst andGst analysis showed changed degree of genetic differentiation across observed loci. Significant changes in differentiation were recorded for crooked little finger and PTC tasting. Modest changes of gene differentiation were recorded for ear lobe type and thumb proximal extensibility. Genetic distance analisis between Bijela and Memici has lower value after then before the war, but genetic distance between Bijela-prewar and Bijela-postwar has higher value, as well as Memici-prewar and Memici-postwar genetic distance. Genetic distance between Bijela-prewar and Memiciprewar, as well as Bijela-postwar and Memici-postwar showed similarity. Constructed dendrogram based on results of genetic distance analyses indicated two clusters groups (1. Bijela-prewar, Memici-prewar; 2. Bijela-postwar, Memici-postwar). Changed genetic differentiation and results of genetic distance analyses indicated possibility of significant impact of forced migration in genetic structure of observed populations.  相似文献

To estimate how extensively the ensemble of denatured-state conformations is constrained by local side-chain–backbone interactions, propensities of each of the 20 amino acids to occur in mono- and dipeptides mapped to discrete regions of the Ramachandran map are computed from proteins of known structure. In addition, propensities are computed for the trans, gauche−, and gauche+ rotamers, with or without consideration of the values of phi and psi. These propensities are used in scoring functions for fragment threading, which estimates the energetic favorability of fragments of protein sequence to adopt the native conformation as opposed to hundreds of thousands of incorrect conformations. As finer subdivisions of the Ramachandran plot, neighboring residue phi/psi angles, and rotamers are incorporated, scoring functions become better at ranking the native conformation as the most favorable. With the best composite propensity function, the native structure can be distinguished from 300,000 incorrect structures for 71% of the 2130 arbitrary protein segments of length 40, 48% of 2247 segments of length 30, and 20% of 2368 segments of length 20. A majority of fragments of length 30–40 are estimated to be folded into the native conformation a substantial fraction of the time. These data suggest that the variations observed in amino acid frequencies in different phi/psi/chi1 environments in folded proteins reflect energetically important local side-chain–backbone interactions, interactions that may severely restrict the ensemble of conformations populated in the denatured state to a relatively small subset with nativelike structure.  相似文献

A subpopulation of transient ON/OFF ganglion cells in the turtle retina transmits changes in stimulus intensity as series of distinct spike events. The temporal structure of these event sequences depends systematically on the stimulus and thus carries information about the preceding intensity change. To study the spike events' intra-retinal origins, we performed extracellular ganglion cell recordings and simultaneous intracellular recordings from horizontal and amacrine cells. Based on these data, we developed a computational retina model, reproducing spike event patterns with realistic intensity dependence under various experimental conditions. The model's main features are negative feedback from sustained amacrine onto bipolar cells, and a two-step cascade of ganglion cell suppression via a slow and a fast transient amacrine cell. Pharmacologically blocking glycinergic transmission results in disappearance of the spike event sequence, an effect predicted by the model if a single connection, namely suppression of the fast by the slow transient amacrine cell, is weakened. We suggest that the slow transient amacrine cell is glycinergic, whereas the other types release GABA. Thus, the interplay of amacrine cell mediated inhibition is likely to induce distinct temporal structure in ganglion cell responses, forming the basis for a temporal code. Action Editor: Jonathan D. Gentianella aspera is a biennial plant of various nutrient-poor grasslands that has become rare in the landscapes outside the Alps of eastern Austria. Using AFLP fingerprinting we investigated: (1) effects of spatial structure on genetic structure in a large vineyard population that is confined to the embankments separating the grapevines; (2) temporal variation in genetic diversity and structure in this population; (3) relationships with other populations in a regional context. On the regional scale, moderate isolation by distance among populations was revealed by a Mantel test. Bayesian analysis of population structure indicated three spatially distinct gene pools and an additional one within the vineyard population. Within this population, spatial autocorrelation analysis revealed a positive correlation between genetic and spatial distance up to 50 m. Patterns found by PCoA were not in line with a priori defined subpopulations and indicated substantial gene flow across embankments. AMOVA revealed low differentiation among both the subpopulations that were found on the linear embankments and among two local groups of these subpopulations. We found, however, striking differences in the among-group variation between the 2 years, i.e., between two local groups within the generations and between those groups among generations. This was due to the highly variable larger group of the younger generation, in which an additional gene pool was identified by Bayesian analysis of population structure. Based on these results we discuss scenarios of local and regional dynamics within and among G. aspera populations.  相似文献

Meals have long been considered relevant units of feeding behavior. Large data sets of feeding behavior of cattle, pigs, chickens, ducks, turkeys, dolphins, and rats were analyzed with the aims of 1) describing the temporal structure of feeding behavior and 2) developing appropriate methods for estimating meal criteria. Longer (between-meal) intervals were never distributed as the negative exponential assumed by traditional methods, such as log-survivorship analysis, but as a skewed Gaussian, which can be (almost) normalized by log-transformation of interval lengths. Log-transformation can also normalize frequency distributions of within-meal intervals. Meal criteria, i.e., the longest interval considered to occur within meals, can be estimated after fitting models consisting of Gaussian functions alone or of one Weibull and one or more Gaussian functions to the distribution of log-transformed interval lengths. Nonuniform data sets may require disaggregation before this can be achieved. Observations from all species were in conflict with assumptions of random behavior that underlie traditional methods for criteria estimation. Instead, the observed structure of feeding behavior is consistent with 1) a decrease in satiety associated with an increase in the probability of animals starting a meal with time since the last meal and 2) an increase in satiation associated with an increase in the probability of animals ending a meal with the amount of food already consumed. The novel methodology proposed here will avoid biased conclusions from analyses of feeding behavior associated with previous methods and, as demonstrated, can be applied across a range of species to address questions relevant to the control of food intake.  相似文献

Summary The paper formulates a one locus two allele diploid model influenced by temporal random selection intensities. The concept of stochastic local stability for equilibria states is formulated. Necessary and sufficient conditions are derived for the stochastic local stability of the fixation states for non-dominant and dominant traits. A general model, where a fixed polymorphic equilibrium is maintained is investigated. Background  

Structural similarities among proteins can provide valuable insight into their functional mechanisms and relationships. As the number of available three-dimensional (3D) protein structures increases, a greater variety of studies can be conducted with increasing efficiency, among which is the design of protein structural alphabets. Structural alphabets allow us to characterize local structures of proteins and describe the global folding structure of a protein using a one-dimensional (1D) sequence. Thus, 1D sequences can be used to identify structural similarities among proteins using standard sequence alignment tools such as BLAST or FASTA.  相似文献

Neuropsychological evidence indicates that the global aspect of complex visual scenes is preferentially processed by the right hemisphere, and local aspects are preferentially processed by the left hemisphere. Using letter-based hierarchical stimuli (Navon figures), we recently demonstrated, in a directed-attention task, lateralized neural activity (assessed by positron emission tomography) in the left prestriate cortex during local processing, and in the right prestriate cortex during global processing. Furthermore, temporal-parietal cortex was critically activated bilaterally in a divided-attention task that involved varying the number of target switches between local and global levels of letter-based hierarchical stimuli. Little is known about whether such stimulus categories influence such hemispheric lateralization. We now present data on brain activity, derived from positron emission tomography, in normal subjects scanned during either local or global processing of object-based hierarchical stimuli. We again observe attentional modulation of neural activity in prestriate cortex. There is now greater right-sided activation for local processing and greater left-sided activation for global processing, which is the opposite of that seen with letter-based stimuli. The results suggest that the relative differential hemispheric activations in the prestriate areas during global and local processing are modified by stimulus category.  相似文献

DNA synthesis in the S phase of V79 and CHO Chinese hamster cells was examined in detail using an automated system for selection and subculturing of mitotic cells and four different assays for DNA synthesis. Flow microfluorometric (FMF) analysis showed that the selected populations were highly synchronous with few noncycling cells. In CHO cells changes in mean and modal fluorescence in the FMF suggested that DNA content increased in a saltatory fashion with 10-20% of the DNA replicated in early S, 40% in mid S, and 40-50% in late S. Pulse labeling and acid precipitation revealed a repeatable pattern of fluctuations in the rate of isotope incorporation with the maximum rate occurring late in S both V79 and CHO. Autoradiography proved to be the best means of accurately determining the beginning of S phase. Cumulative labeling from mitosis to points in S exaggerated the differences in rate between early and late S, so that significant DNA synthesis in early S might easily be overlooked using this method. In CHO cells DNA-specific fluorescence by the Kissane and Robins assay supported the isotopic incorporation data and the FMF analyses by exhibiting a stepwise increase. In V79 cells, S phase lasts only 5 or 5.5 hr, and consequently the mid S and late S steps in fluorescence are compressed. In V79, greater than 80% of the increase in DNA-specific fluorescence occurred between 4.5 and 7 hr after mitotic selection. In both cell lines, fluorescence in early S phase frequently increased transiently to maximum and then decreased.  相似文献

We studied the local response to wounding in Arabidopsis thaliana leaves using a two-step microarray analysis. A microarray containing 3500 cDNA clones was first screened to enrich for genes affected by wounding in the immediate vicinity of the wound (4 h post wounding). 359 non-redundant putative wound responsive genes were then spotted on a smaller wound-response array for detailed analysis of spatial expression (local, adjacent and systemic), timing of expression (0.5, 4, 8, 17 h), and effect of hormone treatments (methyl jasmonate, ethylene and abscisic acid). Our results show that genes that respond early at the site of the wound also respond throughout the plant, with similar kinetics. Early-induced genes which respond systemically encode predominantly signal transduction and regulatory factors (36%), and the expression of many of them is also controlled by methyl jasmonate (about 35% of the 36%). Genes specific to the wound site and the wounded leaf have a slower response to wounding and are mainly metabolic genes. At the wound, many genes of the lignin biosynthesis pathway were induced. In silico analysis of the 5′ promoter regions of genes affected by wounding revealed G-box-related motifs in a significant proportion of the promoters. These results show that the establishment of a systemic response to wounding is a priority for the plant, and that the local response at the wound site is established later. Ethylene and abscisic acid are involved in the local response, regulating repression of photosynthetic genes and expression of drought responsive genes respectively.  相似文献
