In starved newborn rats an increase in glucose turnover rate was observed 4 and 6h after birth, but a dramatic fall occurred at 16h. In suckling newborn rats, no decrease in glucose turnover rate was observed at 16h. The metabolic clearance of glucose did not change in fed or starved animals. The results are discussed in relation to metabolic adaptation to extra-uterine life.  相似文献

In analogy to “specific gravity” or “specific heat” the expression “weight specific metabolic rate” (Ultsch, 1973) would be correct if the metabolic rate were directly proportional to body weight. In that case the quotient metabolic rate divided by body weight would be a constant, independent of body weight like density or specific heat are constants. The metabolic rate, however, is not proportional to body weight but to its 34 power. I have stated that heat flow per unit body weight has no proper physical or physiological meaning (Kleiber, 1970), but since found such a physiological meaning: in work with tracers turnover rates are measured as quotients of transfer rates/pool content. For similometric animals pool contents are proportional to body weight. For such animals therefore the quotient metabolic rate/body weight may have a proper physiological meaning, namely the turnover rate of chemical energy in the animal body.The usefulness of the turnover rate is limited. For the calculation of the energy requirement of horizontal animal locomotion, for example, the calculation from the metabolic rate per animal is preferable to the calculation based on the metabolic rate per unit body weight.  相似文献

Estimation of the isotopic turnover rate in the tissues of organisms allows us to estimate the temporal relationship between the isotope ratio of an organism and its prey, and to analyze seasonal variations of food webs and migratory patterns of animals. We analyzed the isotopic turnover rate in the tissue of lacustrine sockeye salmon, Oncorhynchus nerka, of three age classes in a laboratory food alteration experiment to clarify the isotopic turnover rate. The contribution of growth to isotopic turnover was higher in young and fast-growing lacustrine sockeye salmon than in older and slow-growing fish. Maturation masks the relationship between the contributions of growth and metabolism to isotopic change. Our estimate of the isotopic time lag of lacustrine sockeye salmon can be used to research trophic relationships of anadromous sockeye salmon.  相似文献

Muscle ATP turnover rate during isometric contraction in humans   总被引:4,自引:0,他引:4  
ATP turnover and glycolytic rates during isometric contraction in humans have been investigated. Subjects contracted the knee extensor muscles at two-thirds maximal voluntary force to fatigue (mean +/- SE, 53 +/- 4 s). Biopsies were obtained before and after exercise and analyzed for high-energy phosphates and glycogenolytic-glycolytic intermediates. Total ATP turnover was 190 +/- 7 mmol/kg dry muscle, whereas the average turnover rate was 3.7 +/- 0.2 mmol . kg dry muscle-1 . S-1. The average ATP turnover rate was positively correlated with the percentage of fast-twitch fibers in the postexercise biopsy (r = 0.71; P less than 0.05) and negatively correlated with contraction duration to fatigue (r = -0.88; P less than 0.05). At fatigue, phosphocreatine ranged from 1 to 11 mmol/kg dry muscle (86-99% depletion of value at rest), whereas lactate ranged from 59 to 101. The mean glycolytic rate was 0.83 +/- 0.05 mmol . kg dry muscle-1 . S-1 and was positively correlated with the rate of glucose 6-phosphate accumulation (r = 0.83; P less than 0.05). It is concluded that a major determinant of the ATP turnover rate is the muscle fiber composition, which is probably explained by a higher turnover rate in fast-twitch fibers; fatigue is more closely related to a low phosphocreatine content than to a high lactate content; and the increase in prephosphofructokinase intermediates is important for stimulating glycolysis during contraction.  相似文献

Measurement of acetylcholine turnover rate in discrete areas of rat brain   总被引:6,自引:0,他引:6  
The turnover rate of ACh was estimated in brain stem, two cortical areas and striatum of rat brain. The turnover rate was highest in the striatum (1.3 μmoles/g/hr); lowest in brain stem (0.092 μmoles/g/hr); and intermediate values were observed in limbic and occipital cortex. The highest ACh concentrations were measured in striatum, those in brain stem were intermediate but in the two cortical areas the ACh concentrations were the lowest. The results of the turnover estimations with the finite difference method yielded values similar to those obtained with the procedure described in this paper. Moreover, once the baseline was established, this method could be reliably used to estimate turnover rate using a single infusion time. The latter simplication would be very useful to compare ACh turnover rate in drug studies.  相似文献

The rate of appearance of prototrophic revertants when Escherichia coli tyrA14 (ochre) or trpA23 bacteria were incubated on plates lacking the required amino acid was greatly elevated when the organisms also carried a mutT mutation. Previous studies with Arabidopsis accessions revealed that biomass correlates negatively to dusk starch content and total protein, and positively to the maximum activities of enzymes in photosynthesis. We hypothesized that large accessions have lower ribosome abundance and lower rates of protein synthesis, and that this is compensated by lower rates of protein degradation. This would increase growth efficiency and allow more investment in photosynthetic machinery. We analysed ribosome abundance and polysome loading in 19 accessions, modelled the rates of protein synthesis and compared them with the observed rate of growth. Large accessions contained less ribosomes than small accessions, due mainly to cytosolic ribosome abundance falling at night in large accessions. The modelled rates of protein synthesis resembled those required for growth in large accessions, but were up to 30% in excess in small accessions. We then employed 13CO2 pulse‐chase labelling to measure the rates of protein synthesis and degradation in 13 accessions. Small accessions had a slightly higher rate of protein synthesis and much higher rates of protein degradation than large accessions. Protein turnover was negligible in large accessions but equivalent to up to 30% of synthesised protein day?1 in small accessions. We discuss to what extent the decrease in growth in small accessions can be quantitatively explained by known costs of protein turnover and what factors may lead to the altered diurnal dynamics and increase of ribosome abundance in small accessions, and propose that there is a trade‐off between protein turnover and maximisation of growth rate.  相似文献

The validation of the urinary excretion of N-methylhistidine (N-MH) by quail as an index of the muscle protein turnover rate was tested using the criterion of the rate of recovery of radioactivity in urine following an intraperitoneal dose of l-[3-14C]methylhistidine. A genetic study on muscle protein turnover in quail was conducted using three genetically diverse lines (LL, large body size; SS, small body size; RR, random-bred control line) selected for body size. When l-[3-14C]methylhistidine was administered to 20-week-old male and female coturnix quail by direct intraperitoneal injection, approximately 90% of the l-[3-14C]methylhistidine was recovered by 96 hr postinjection. Recoveries were low in the egg and muscle. These results show that N-MH released from myofibrillar protein is not reutilized and the excretion of N-MH is a satisfactory index of muscle protein breakdown. In all lines, the amount of urinary N-MH excretion and fractional synthesis (Ks) and degradation (Kd) rates at the high growing period were higher than those at the low growing period. The Ks and Kd are significantly different among selected lines at both 3 and 6 weeks of age. At 3 weeks of age, the fractional rate of synthesis of the LL line (13.2%/day) was higher than that of the RR line (11.5%/day), whereas the SS (8.1%/day) was lower than that of the RR line (11.5%/day). The fractional rates of degradation of both the LL line (4.1%/day) and the SS line (5.6%/day) were lower than that of the RR line (7.0%/day) at 3 weeks of age. From these results, it was recognized that selection for body size gave rise to the changes in the muscle protein turnover rate.  相似文献

Geographic range, turnover rate and the scaling of species diversity   总被引:6,自引:0,他引:6  
The study of the relative roles of local and regional processes in determining the scaling of species diversity is a very active field in current ecology. The importance of species turnover and the species‐range‐size frequency distributions in determining how local and regional species diversity are linked has been recognised by recent approaches. Here we present a model, based on a system of fully nested sampling quadrats, to analyse species diversity at several scales. Using a recursive procedure that incorporates increasingly smaller scales and a multiplicative formula for relating local and regional diversity, the model allows the simultaneous depiction of alpha, beta and gamma diversity in a single “species‐scale plot”. Species diversity is defined as the number of ranges that are intersected by sampling quadrats of various sizes. The size, shape and location of individual species ranges determine diversity at any scale, but the average point diversity, measured at hypothetical zero‐area localities, is determined solely by the size of individual ranges, regardless of their shape and location. The model predicts that if the species‐area relationship is a power function, then beta diversity must be scale invariant if measured at constant scale increments. Applying the model to the mammal fauna of four Mexican regions with contrasting environmental conditions, we found that: 1) the species‐range‐size frequency distribution at the scale of the Mexican regions differs from the log‐normal pattern reported for the national and continental scales. 2) Beta diversity is not scale‐invariant within each region, implying that the species‐area relationship (SAR) does not follow a power function. 3) There is geographic variation in beta diversity. 4) The scaling of diversity is directly linked to patterns of species turnover rate, and ultimately determined by patterns in the geographic distribution of species. The model shows that regional species diversity and the average distribution range of species are the two basic data necessary to predict patterns in the scaling of species diversity.  相似文献

Fate and turnover rate of ovarian follicular cysts in dairy cattle   总被引:2,自引:0,他引:2  
Non-lactating, multiparous dairy cows diagnosed as having cysts by palpation per rectum were used. Cysts were induced with oestradiol-17 beta (15 mg) and progesterone (37.5 mg) dissolved in ethanol and injected s.c. twice daily for 7 days. Following initial diagnosis of cysts, ovaries were exposed by midventral laparotomy, and the perimeter of the base of each cyst was marked with subepithelial injections of charcoal. Ovaries were removed from cows by transvaginal incision at 10 days (Group 1; N = 8), 20 days (Group 2; N = 8), or 40 days (Group 3; N = 7) after marking of cysts. Ovaries were examined for structures present and their relationship to the marked site. Corpora lutea with ovulation papilla were present in 7/23 cows (1/8, 4/8 and 2/7, for Groups 1, 2 and 3, respectively). In these 7 cows, corpora lutea were at a site different from the original structure that was marked. Marked structures persisted for the duration of the experimental period in 1 and 2 cows, in Groups 1 and 3, respectively. In the remaining 13 cows, new large follicular structures (cysts) were present at a site other than that marked with charcoal. These structures developed on the ovary contralateral to the one originally marked in 9 of 13 cows. Cysts are therefore dynamic in nature and may persist or may be replaced by others.  相似文献

Valproate is an important anticonvulsant currently in clinical use for the treatment of seizures. We used electrophysiological and tracer uptake methods to examine the effect of valproate on a gamma-aminobutyric acid (GABA) transporter (mouse GAT3) expressed in Xenopus laevis oocytes. In the absence of GABA, valproate (up to 50 mm) had no noticeable effect on the steady-state electrogenic properties of mGAT3. In the presence of GABA, however, valproate enhanced the GABA-evoked steady-state inward current in a dose-dependent manner with a half-maximal concentration of 4.6 +/- 0.5 mm. Maximal enhancement of the GABA-evoked current was 275 +/- 10%. Qualitatively similar observations were obtained for human GAT1 and mouse GAT4. The valproate enhancement did not alter the Na(+) or Cl(-) dependence of the steady-state GABA-evoked currents. Uptake experiments under voltage clamp suggested that the valproate enhancement of the GABA-evoked current was matched by an enhancement in GABA uptake. Thus, despite the increase in GABA-evoked current, ion/GABA co-transport remained tightly coupled. Uptake experiments indicated that valproate is not transported by mouse GAT3 in the absence or presence of GABA. Valproate also enhanced the rate of the partial steps involved in transporter presteady-state charge movements. We propose that valproate increases the turnover rate of GABA transporters by an allosteric mechanism. The data suggest that at its therapeutic concentration, valproate may enhance the activity of neuronal and glial GABA transporters by up to 10%.  相似文献

Phosphoinositides (PtdInss) play key roles in cell polarization and motility. With a series of biosensors based on Förster resonance energy transfer, we examined the distribution and metabolism of PtdInss and diacylglycerol (DAG) in stochastically migrating Madin-Darby canine kidney (MDCK) cells. The concentrations of phosphatidylinositol (4,5)-bisphosphate, phosphatidylinositol (3,4,5)-trisphosphate (PIP3), phosphatidylinositol (3,4)-bisphosphate, and DAG were higher at the plasma membrane in the front of the cell than at the plasma membrane of the rear of the cell. The difference in the concentrations of PtdInss was estimated to be less than twofold between the front and rear of the migrating MDCK cells. To decode the spatial activities of PtdIns metabolic enzymes from the obtained concentration maps of PtdInss, we developed a one-dimensional reaction diffusion model of PtdIns metabolism. In this model, the activities of phosphatidylinositol monophosphate 5-kinase, phosphatidylinositol 3-kinase, phospholipase C, and PIP3 5-phosphatases were higher at the plasma membrane of the front than at the plasma membrane of the rear of the cell. This result suggests that, although the difference in the steady-state level of PtdInss is less than twofold, PtdInss were more rapidly turned over at the front than the rear of the migrating MDCK cells.  相似文献
