With the use of voltage clamp and current clamp techniques thesupposition was proved that during the hyperpolarizing response(HR) N. obtusa cells generate active electromotive force (emf)at the expense of metabolic energy. Threshold inward currentsent through the plasmalemma of the cell which was depolarizedwith 100 mol m–3 KG resulted in the HR with the transferof the membrane's excitable units from the high-conductive stateto the low-conductive state. During the HR the membrane potentialVm increased from –135±10 mV to –290±15mV, the membrane resistance increased from 3.3±1.5 kOhmcm2 to 5.8±1.2 kOhm cm2 and the membrane emf Em increasedfrom –20±4 mV to –93± 15 mV. Changesin the external concentration of K, Na+, Cl andH did not affect the patterns of HR. Cells which weredepolarized by light also generated HR (in normal medium) whichwas accompanied with the increase of Vm, Rm and Em. The highvalue of Em generated during the HR can be explained only withthe involvement of active electrogenic charge transfer acrossthe membrane. 0.05 mol m–3 DCCD added to the externalmedium inhibited the HR in both cases. To study the effect of chronically elevated CO2 on the excitability and function of neurons, we exposed mice to 7.5–8% CO2 for 2 wk (starting at 2 days of age) and examined the properties of freshly dissociated hippocampal neurons. Neurons from control mice (CON) and from mice exposed to chronically elevated CO2 had similar resting membrane potentials and input resistances. CO2-exposed neurons, however, had a lower rheobase and a higher Na+ current density (580 ± 73 pA/pF; n = 27 neurons studied) than did CON neurons (280 ± 51 pA/pF, n = 34; P < 0.01). In addition, the conductance-voltage curve was shifted in a more negative direction in CO2-exposed than in CON neurons (midpoint of the curve was –46 ± 3 mV for CO2 exposed and –34 ± 3 mV for CON, P < 0.01), while the steady-state inactivation curve was shifted in a more positive direction in CO2-exposed than in CON neurons (midpoint of the curve was –59 ± 2 mV for CO2 exposed and –68 ± 3 mV for CON, P < 0.01). The time constant for deactivation at –100 mV was much smaller in CO2-exposed than in CON neurons (0.8 ± 0.1 ms for CO2 exposed and 1.9 ± 0.3 ms for CON, P < 0.01). Immunoblotting for Na+ channel proteins (subtypes I, II, and III) was performed on the hippocampus. Our data indicate that Na+ channel subtype I, rather than subtype II or III, was significantly increased (43%, n = 4; P < 0.05) in the hippocampi of CO2-exposed mice. We conclude that in mice exposed to elevated CO2, 1) increased neuronal excitability is due to alterations in Na+ current and Na+ channel characteristics, and 2) the upregulation of Na+ channel subtype I contributes, at least in part, to the increase in Na+ current density. sodium ion channels; oxygen deprivation  相似文献

Membrane potential and resistance, each of which was the sumof those of the plasmalemma and tonoplast, measured in the coenocyticthallus of Boergesenia forbesii were 6.7 mv inside positiveand 2.8 k.cm2, respectively. Protoplasm squeezed from the thallus into artificial sea water(ASW) formed numerous spherical bodies, which are termed aplanospore-likecells (simply "spores"). The following electrical propertiesof the "spores" 20–40 hr after squeezing were obtained:potential difference (p.d.) across plasmalemma (Eco) was –66mv (– means inside negative), plasmalemma resistance 665cm2, p.d. across the tonoplast (Evc) +73 mv, and tonoplast resistance2.6 k.cm2. Tenfold increase in external [K+] caused +45 mv changein Eco and +17 mv in Evc. The plasmalemma was entirely depolarizedin Ca++-free ASW or ASW containing Triton X-100. When the "spore" was immersed in potassium-rich (277 mil) ASW,Eco was almost zero and the tonoplast showed two states (I andII, Eve about +70 mv and +20 mv, respectively). Evc went backand forth between the two states spontaneously or when a smallcurrent was applied. In most cases oscillatory changes in Evcoccurred after the lapse of a long time in the K+-rich sea water.Membrane resistances in states I and II were 5 and 9 k.cm2,respectively. Mice are useful animal models to study pathogenic mechanisms involved in pulmonary vascular disease. Altered expression and function of voltage-gated K+ (KV) channels in pulmonary artery smooth muscle cells (PASMCs) have been implicated in the development of pulmonary arterial hypertension. KV currents (IK(V)) in mouse PASMCs have not been comprehensively characterized. The main focus of this study was to determine the biophysical and pharmacological properties of IK(V) in freshly dissociated mouse PASMCs with the patch-clamp technique. Three distinct whole cell IK(V) were identified based on the kinetics of activation and inactivation: rapidly activating and noninactivating currents (in 58% of the cells tested), rapidly activating and slowly inactivating currents (23%), and slowly activating and noninactivating currents (17%). Of the cells that demonstrated the rapidly activating noninactivating current, 69% showed IK(V) inhibition with 4-aminopyridine (4-AP), while 31% were unaffected. Whole cell IK(V) were very sensitive to tetraethylammonium (TEA), as 1 mM TEA decreased the current amplitude by 32% while it took 10 mM 4-AP to decrease IK(V) by a similar amount (37%). Contribution of Ca2+-activated K+ (KCa) channels to whole cell IK(V) was minimal, as neither pharmacological inhibition with charybdotoxin or iberiotoxin nor perfusion with Ca2+-free solution had an effect on the whole cell IK(V). Steady-state activation and inactivation curves revealed a window K+ current between –40 and –10 mV with a peak at –31.5 mV. Single-channel recordings revealed large-, intermediate-, and small-amplitude currents, with an averaged slope conductance of 119.4 ± 2.7, 79.8 ± 2.8, 46.0 ± 2.2, and 23.6 ± 0.6 pS, respectively. Experiments were conducted to determine whether the Cl secretagogue, 1-ethyl-2-benzimidazolinone (EBIO), stimulates Cl transport in the rabbit conjunctival epithelium. For this study, epithelia were isolated in an Ussing-type chamber under short-circuit conditions. The effects of EBIO on the short-circuit current (Isc) and transepithelial resistance (Rt) were measured under physiological conditions, as well as in experiments with altered electrolyte concentrations. Addition of 0.5 mM EBIO to the apical bath stimulated the control Isc by 64% and reduced Rt by 21% (P < 0.05; paired data). Under Cl-free conditions, Isc stimulation using EBIO was markedly attenuated. In the presence of an apical-to-basolateral K+ gradient and permeabilization of the apical membrane, the majority of the Isc reflected the transcellular movement of K+ via basolateral K+ channels. Under these conditions, EBIO in combination with A23187 elicited nearly instantaneous 60–90% increases in Isc that were sensitive to the calmodulin antagonist calmidazolium and the K+ channel blocker tetraethyl ammonium. In the presence of an apical-to-basolateral Cl gradient and nystatin permeabilization of the basolateral aspect, EBIO increased the Cl-dependent Isc, an effect prevented by the channel blocker glibenclamide (0.3 mM). The latter compound also was used to determine the proportion of EBIO-evoked unidirectional 36Cl fluxes in the presence of the Cl gradient that traversed the epithelium transcellularly. Overall, EBIO activated apical Cl channels and basolateral K+ channels (presumably those that are Ca2+ dependent), thereby suggesting that this compound, or related derivatives, may be suitable as topical agents to stimulate fluid transport across the tissue in individuals with lacrimal gland deficiencies. We used the short-circuit current (Isc) technique to investigate the effects of the isoflavone genistein on the electrogenic Cl secretion of the mouse jejunum. Genistein stimulated a sustained increase in Isc that was dose dependent. Bumetanide inhibited 76 ± 5% of the genistein-stimulated Isc consistent with activation of Cl secretion. Genistein failed to stimulate Isc following maximal activation of the cAMP pathway by forskolin. In addition, forskolin had a reduced effect on Isc of the mouse jejunum in the presence of genistein. Glibenclamide, a blocker of CFTR, eliminated the genistein-stimulated increase of Isc and reduced the forskolin-activated Isc. Clotrimazole, a Ca2+-activated K+ channel blocker, failed to reduce the genistein-stimulated Isc. Vanadate, a blocker of tyrosine-dependent phosphatases, reduced the genistein-activated Isc. Tyrphostin A23, a tyrosine kinase inhibitor, reduced basal Isc, after which genistein failed to stimulate Isc. An inwardly rectifying swelling- and meiotic cell cycle-regulated anion current carried by the ClC channel splice variant CLH-3b dominates the whole cell conductance of the Caenorhabditis elegans oocyte. Oocytes also express a novel outwardly rectifying anion current termed ICl,OR. We recently identified a worm strain carrying a null allele of the clh-3 gene and utilized oocytes from these animals to characterize ICl,OR biophysical properties. The ICl,OR channel is strongly voltage dependent. Outward rectification is due to voltage-dependent current activation at depolarized voltages and rapid inactivation at voltages more hyperpolarized than approximately +20 mV. Apparent channel open probability is zero at voltages less than +20 mV. The channel has a 4:1 selectivity for Cl over Na+ and an anion selectivity sequence of SCN > I > Br > Cl > F. ICl,OR is relatively insensitive to most conventional anion channel inhibitors including DIDS, 4,4'-dinitrostilbene-2,2'-disulfonic acid, 9-anthracenecarboxylic acid, and 5-nitro-2-(3-phenylpropylamino)benzoic acid. However, the current is rapidly inhibited by niflumic acid, metal cations including Gd3+, Cd2+, and Zn2+, and bath acidification. The combined biophysical properties of ICl,OR are distinct from those of other anion currents that have been described. We investigated the effects of 5,6-dichloro-1-ethyl-1,3-dihydro-2H-benzimidazol-2-one(DCEBIO) on the Cl secretory response of the mouse jejunum using the Ussing short-circuit current (Isc) technique. DCEBIO stimulated a concentration-dependent, sustained increase in Isc (EC50 41 ± 1 µM). Pretreating tissues with 0.25 µM forskolin reduced the concentration-dependent increase in Isc by DCEBIO and increased the EC50 (53 ± 5 µM). Bumetanide blocked (82 ± 5%) the DCEBIO-stimulated Isc consistent with Cl secretion. DCEBIO was a more potent stimulator of Cl secretion than its parent molecule, 1-ethyl-2-benzimidazolinone. Glibenclamide or NPPB reduced the DCEBIO-stimulated Isc by >80% indicating the participation of CFTR in the DCEBIO-stimulated Isc response. Clotrimazole reduced DCEBIO-stimulated Isc by 67 ± 15%, suggesting the participation of the intermediate conductance Ca2+-activated K+ channel (IKCa) in the DCEBIO-activated Isc response. In the presence of maximum forskolin (10 µM), the DCEBIO response was reduced and biphasic, reaching a peak response of the change in Isc of 43 ± 5 µA/cm2 and then falling to a steady-state response of 17 ± 10 µA/cm2 compared with DCEBIO control tissues (61 ± 6 µA/cm2). The forskolin-stimulated Isc in the presence of DCEBIO was reduced compared with forskolin control tissues. Similar results were observed with DCEBIO and 8-BrcAMP where adenylate cyclase was bypassed. H89, a PKA inhibitor, reduced the DCEBIO-activated Isc, providing evidence that DCEBIO increased Cl secretion via a cAMP/PKA-dependent manner. The Na+-dependent nucleoside transporter 2 (CNT2) mediates active transport of purine nucleosides and uridine as well as therapeutic nucleoside analogs. We used the two-electrode voltage-clamp technique to investigate rat CNT2 (rCNT2) transport mechanism and study the interaction of nucleoside-derived drugs with the transporter expressed in Xenopus laevis oocytes. The kinetic parameters for sodium, natural nucleosides, and nucleoside derivatives were obtained as a function of membrane potential. For natural substrates, apparent affinity (K0.5) was in the low micromolar range (12–34) and was voltage independent for hyperpolarizing membrane potentials, whereas maximal current (Imax) was voltage dependent. Uridine and 2'-deoxyuridine analogs modified at the 5-position were substrates of rCNT2. Lack of the 2'-hydroxyl group decreased affinity but increased Imax. Increase in the size and decrease in the electronegativity of the residue at the 5-position affected the interaction with the transporter by decreasing both affinity and Imax. Fludarabine and formycin B were also transported with higher Imax than uridine and moderate affinity (102 ± 10 and 66 ± 6 µM, respectively). Analysis of the pre-steady-state currents revealed a half-maximal activation voltage of about –39 mV and a valence of about –0.8. K0.5 for Na+ was 2.3 mM at –50 mV and decreased at hyperpolarizing membrane potentials. The Hill coefficient was 1 at all voltages. Direct measurements of radiolabeled nucleoside fluxes with the charge associated showed a ratio of two positive inward charges per nucleoside, suggesting a stoichiometry of two Na+ per nucleoside. Pancreatic duct cells express a Ca2+-activated Cl- conductance (CaCC), upregulation of which may be beneficial to patients with cystic fibrosis. Here, we report that HPAF, a human pancreatic ductal adenocarcinoma cell line that expresses CaCC, develops into a high-resistance, anion-secreting epithelium. Mucosal ATP (50 µM) caused a fourfold increase in short-circuit current (Isc), a hyperpolarization of transepithelial potential difference (from -4.9 ± 0.73 to -8.5 ± 0.84 mV), and a fall in resistance to less than one-half of resting values. The effects of ATP were inhibited by mucosal niflumic acid (100 µM), implicating an apical CaCC in the response. RT-PCR indicated expression of hClC-2, hClC-3, and hClC-5, but surprisingly not hCLCA-1 or hCLCA-2. K+ channel activity was necessary to maintain the ATP-stimulated Isc. Using a pharmacological approach, we found evidence for two types of K+ channels in the mucosal and serosal membranes of HPAF cells, one activated by chlorzoxazone (500 µM) and sensitive to clotrimazole (30 µM), as well as one blocked by clofilium (100 µM) but not chromanol 293B (5 µM). RT-PCR indicated expression of the Ca2+-activated K+ channel KCNN4, as well as the acid-sensitive, four transmembrane domain, two pore K+ channel, KCNK5 (hTASK-2). Western blot analysis verified the expression of CLC channels, as well as KCNK5. We conclude that HPAF will be a useful model system for studying channels pertinent to anion secretion in human pancreatic duct cells. Mammary epithelial 31EG4 cells (MEC) were grown as monolayers onfilters to analyze the apical membrane mechanisms that help mediate ionand fluid transport across the epithelium. RT-PCR showed the presenceof cystic fibrosis transmembrane conductance regulator (CFTR) andepithelial Na+ channel (ENaC) message, and immunomicroscopyshowed apical membrane staining for both proteins. CFTR was alsolocalized to the apical membrane of native human mammary ductepithelium. In control conditions, mean values of transepithelialpotential (apical-side negative) and resistance(RT) are 5.9 mV and 829  · cm2, respectively. The apical membranepotential (VA) is 40.7 mV, and the mean ratioof apical to basolateral membrane resistance (RA/RB) is 2.8. Apicalamiloride hyperpolarized VA by 19.7 mV andtripled RA/RB. AcAMP-elevating cocktail depolarized VA by 17.6 mV, decreased RA/RB by60%, increased short-circuit current by 6 µA/cm2,decreased RT by 155  · cm2, and largely eliminated responses toamiloride. Whole cell patch-clamp measurements demonstratedamiloride-inhibited Na+ currents [linear current-voltage(I-V) relation] and forskolin-stimulated Clcurrents (linear I-V relation). A capacitance probe methodshowed that in the control state, MEC monolayers either absorbed orsecreted fluid (2-4µl · cm2 · h1). Fluidsecretion was stimulated either by activating CFTR (cAMP) or blockingENaC (amiloride). These data plus equivalent circuit analysis showedthat 1) fluid absorption across MEC is mediated byNa+ transport via apical membrane ENaC, and fluid secretionis mediated, in part, by Cl transport via apicalCFTR; 2) in both cases, appropriate counterions move throughtight junctions to maintain electroneutrality; and 3)interactions among CFTR, ENaC, and tight junctions allow MEC to eitherabsorb or secrete fluid and, in situ, may help control luminal[Na+] and [Cl].

The theory and practice of applying the thermodynamics of irreversibleprocesses to mass-flow theories is presented. Onsager coefficientswere measured on cut and uncut phloem and cut xylem strandsof Heracleum muntegazzimum. In 0.3 N sucrose + 1 mN KC1 theyare as follows. In phloem, LEE = 5 ? 10–4 mho cm–1,LpE = 9 ? 10–6 cm3 s–1 cm–2 volt–1 cm,and LPP = 0.16 cm3 s–1 cm–2 (J cm–3)–1cm. In uncut phloem strands LEE is about 1 ? 10–3 mhocm–1. In xylem in 2 x 10–3 N KCI, Lpp = 50 to 225,LPE = 2 ? 10–4, and LEE = 4 ? 10–3. The measurementsare tentative since the blockage of the sieve plates is an interferingfactor, but if they are valid they lead to the conclusion thatneither a pressure-flow nor an electro-kinetic mechanism envisaginga ‘long distance’ current pathway can be the majormotive ‘force’ for transport in mature phloem. Measurementsof biopotentials along conducting but laterally detached phloembundles of Heracleum suggest, nevertheless, that there may bea small electro-osmotic component of at least 0.1 mV cm–1endogenous in the phloem.  相似文献

The cardiac Ca2+-independent transient outward K+ current (Ito), a major repolarizing ionic current, is markedly affected by Cl substitution and anion channel blockers. We reexplored the mechanism of the action of anions on Ito by using whole cell patch-clamp in single isolated rat cardiac ventricular myocytes. The transient outward current was sensitive to blockade by 4-aminopyridine (4-AP) and was abolished by Cs+ substitution for intracellular K+. Replacement of most of the extracellular Cl with less permeant anions, aspartate (Asp) and glutamate (Glu), markedly suppressed the current. Removal of external Na+ or stabilization of F-actin with phalloidin did not significantly affect the inhibitory action of less permeant anions on Ito. In contrast, the permeant Cl substitute Br did not markedly affect the current, whereas F substitution for Cl induced a slight inhibition. The Ito elicited during Br substitution for Cl was also sensitive to blockade by 4-AP. The ability of Cl substitutes to induce rightward shifts of the steady-state inactivation curve of Ito was in the following sequence: NO3 > Cl Br > gluconate > Glu > Asp. Depolymerization of actin filaments with cytochalasin D (CytD) induced an effect on the steady-state inactivation of Ito similar to that of less permeant anions. Fluorescent phalloidin staining experiments revealed that CytD-pretreatment significantly decreased the intensity of FITC-phalloidin staining of F-actin, whereas Asp substitution for Cl was without significant effect on the intensity. Measuring the Canopy Net Photosynthesis of Glasshouse Crops   总被引:3,自引:0,他引:3  
A null balance method is described for measuring net photosynthesisof mature canopies of cucumber and other protected crops overperiods of 10 min in a single-span glasshouse (c. 9m x 18m inarea). Accuracy of control of the CO2 concentration in the greenhouseatmosphere is within ±10 vpm of the normal ambient level(c. 350 vpm). The amounts of CO2 used in canopy net photosynthesisare measured with linear mass flowmeters accurate to within±0.80g. The total errors incurred in measuring canopynet photosynthesis at an ambient CO2 level are estimated tobe of the order of ± 1·2% in bright light (350W m–2, PAR)and ±3·6% in dull light (100W m–2, PAR). Measurements of the rates of net photosynthesis of a maturecanopy of a cucumber crop were made at near-ambient CO2 concentrationsover a range (0–350 W m–2) of natural light fluxdensities. A model of light absorption and photosynthesis applicableto row crops was used to obtain a net photosynthesis versuslight response curve for the cucumber crop. At a light fluxdensity of 350 W m–2 the fitted value of canopy net photosynthesiswas 2.65 mg CO2 m–2s–1 (equivalent to over 95 kgCO2 ha–1h–1). The results are discussed in relationto the need for CO2 supplements to avoid depletion in both ventilatedand unventilated glasshouses during late spring and summer. Yield stress threshold (Y) and volumetric extensibility () arethe rheological properties that appear to control root growth.In this study they were measured in wheat roots by means ofparallel measurement of the growth rate (r) of intact wheatroots and of the turgor pressures (P) of individual cells withinthe expansion zone. Growth and turgor pressure were manipulatedby immersion in graded osmoticum (mannitol) solutions. Turgorwas measured with a pressure probe and growth rate by visualobservation. The influence of various growth conditions on Yand was investigated; (a) At 27 °C.In 0.5 mol m–3 CaCl2 r, P, Y and were20.7±4.6 µm min–1, 0.77±0.05 MPa,0.07±0.03 MPa and 26±1.9 µm min–1MPa–1 (expressed as increase in length), respectively.Following 24 h growth in 10 mol m–3 KC1 these parametersbecame 12.3±3.5 µm min–1, 0.72±0.04MPa, 0.13±0.01 MPa and 21±0.7 µm min–1MPa–1. After 24 h osmotic adjustment in 150 mol m–3mannitol/0.5 mol m–3 CaCl2 r= 19.6±4.2 µmmin–1, P = 0.68±0.05 MPa and Y and were 0.07±0.04MPa and 30±0.2 µm min–1 MPa–01, respectively.After 24 h growth in 350 mol m–3 mannitol/0.5 mol m–3CaCl2 r= 13.3±4.1 µm min–1, P= 0.58±0.07MPa, Y=0.12±0.01 MPa and ø 32±0.2 tim min–1MPa–1. During osmotic adjustment in 200 mol m–3mannitol/0.5 mol m–3 CaCl2, with or without KCl, the recoveryof growth rate corresponded to turgor pressure recovery (t1/2approximately 3 h). (b) At 15 °C. Lowered temperature dramatically influencedthe growth parameters which became r= 8.3±2.8 um min–1,P=0.78 MPa, r=<0.2 MPa and =15±0.1 µm min–1MPa–1. Therefore, Y and are influenced by 10 mol m–3 K+ ionsand low temperature. In each case the effective pressure forgrowth (P-Y) was large indicating that small fluctuations ofsoil water potential will not stop root elongation. Effects of cytoplasmic Ca2+ on the electrical properties ofthe plasma membrane were investigated in tonoplast-free cellsof Chara australis that had been internally perfused with media,containing either 1 mM ATP to fuel the electrogenic pump orhexokinase and glucose to deplete the ATP and stop the pump. In the presence of ATP, cytoplasmic Ca2+ up to 2.5?10–5M did not affect the membrane potential (about -190 mV), butmembrane resistance decreased uniformly with increasing [Ca2+]i.In the absence of ATP, the membrane potential, which was onlyabout -110 mV, was depolarized further by raising [Ca2+]i from1.4?10–6 to 2.5?10–5 M. Membrane resistance, whichwas nearly the twofold that of ATP-provided cells, decreasedmarkedly with an increase in [Ca2+]i from zero to 1.38?10–6M, but showed no change for further increases. Internodal cellsof Nitellopsis obtusa were more sensitive to intracellular Ca2+with respect to membrane potential than were those of Charaaustralis, reconfirming the results obtained by Mimura and Tazawa(1983). The effect of cytoplasmic Ca2+ on the ATP-dependent H+ effluxwas measured. No marked difference in H+ effluxes was detectedbetween zero and 2.5?10–5 M [Ca2+]i; but, at 10–4M the ATP-dependent H+ efflux was almost zero. Ca2+ efflux experimentswere done to investigate dependencies on [Ca2+]i and [ATP]i.The efflux was about 1 pmol cm–2 s–1 at all [Ca2+]iconcentrations tested (1.38?10–6, 2.5?10–5, 10–4M).This value is much higher than the influx reported by Hayamaet al. (1979), and this efflux was independent of [ATP]i. The cellular mechanism for Cl and K+ secretion in the colonic epithelium requires K+ channels in the basolateral and apical membranes. Colonic mucosa from guinea pig and rat were fixed, sectioned, and then probed with antibodies to the K+ channel proteins KVLQT1 (Kcnq1) and minK-related peptide 2 (MiRP2, Kcne3). Immunofluorescence labeling for Kcnq1 was most prominent in the lateral membrane of crypt cells in rat colon. The guinea pig distal colon had distinct lateral membrane immunoreactivity for Kcnq1 in crypt and surface cells. In addition, Kcne3, an auxiliary subunit for Kcnq1, was detected in the lateral membrane of crypt and surface cells in guinea pig distal colon. Transepithelial short-circuit current (Isc) and transepithelial conductance (Gt) were measured for colonic mucosa during secretory activation by epinephrine (EPI), prostaglandin E2 (PGE2), and carbachol (CCh). HMR1556 (10 µM), an inhibitor of Kcnq1 channels (Gerlach U, Brendel J, Lang HJ, Paulus EF, Weidmann K, Brüggemann A, Busch A, Suessbrich H, Bleich M, and Greger R. J Med Chem 44: 3831–3837, 2001), partially (50%) inhibited Cl secretory Isc and Gt activated by PGE2 and CCh in rat colon with an IC50 of 55 nM, but in guinea pig distal colon Cl secretory Isc and Gt were unaltered. EPI-activated K+-secretory Isc and Gt also were essentially unaltered by HMR1556 in both rat and guinea pig colon. Although immunofluorescence labeling with a Kcnq1 antibody supported the basolateral membrane presence in colonic epithelium of the guinea pig as well as the rat, the Kcnq1 K+ channel is not an essential component for producing Cl secretion. Other K+ channels present in the basolateral membrane presumably must also contribute directly to the K+ conductance necessary for K+ exit during activation of Cl secretion in the colonic mucosa. Human lung epithelial (Calu-3) cells were used to investigate the effects of protease-activated receptor (PAR) stimulation on Cl secretion. Quantitative RT-PCR (QRT-PCR) showed that Calu-3 cells express PAR-1, -2, and -3 receptor mRNAs, with PAR-2 mRNA in greatest abundance. Addition of either thrombin or the PAR-2 agonist peptide SLIGRL to the basolateral solution of monolayers mounted in Ussing chambers produced a rapid increase in short-circuit current (Isc: thrombin, 21 ± 2 µA; SLIGRL, 83 ± 22 µA), which returned to baseline within 5 min after stimulation. Pretreatment of monolayers with the cell-permeant Ca2+-chelating agent BAPTA-AM (50 µM) abolished the increase in Isc produced by SLIGRL. When monolayers were treated with the cyclooxygenase inhibitor indomethacin (10 µM), nearly complete inhibition of both the thrombin- and SLIGRL-stimulated Isc was observed. In addition, basolateral treatment with the PGE2 receptor antagonist AH-6809 (25 µM) significantly inhibited the effects of SLIGRL on Isc. QRT-PCR revealed that Calu-3 cells express mRNAs for CFTR, the Ca2+-activated KCNN4 K+ channel, and the KCNQ1 K+ channel subunit, which, in association with KCNE3, is known to be regulated by cAMP. Stimulation with SLIGRL produced an increase in apical Cl conductance that was blocked in cells expressing short hairpin RNAs designed to target CFTR. These results support the conclusion that PAR stimulation of Cl secretion occurs by an indirect mechanism involving the synthesis and release of prostaglandins. It has been reported thatsecretory mammary epithelial cells (MEC) release ATP, UTP, and UDP uponmechanical stimulation. Here we examined the physiological changescaused by ATP/UTP in nontransformed, clonal mouse mammary epithelia(31EG4 cells). In control conditions, transepithelial potential (apicalside negative) and resistance were 4.4 ± 1.3 mV (mean ± SD, n = 12) and 517.7 ± 39.4  · cm2, respectively. The apicalmembrane potential was 43.9 ± 1.7 mV, and the ratio of apicalto basolateral membrane resistance (RA/RB) was 3.5 ± 0.2. Addition of ATP or UTP to the apical or basolateral membranescaused large voltage and resistance changes with an EC50 of~24 µM (apical) and ~30 µM (basal). Apical ATP/UTP (100 µM)depolarized apical membrane potential by 17.6 ± 0.8 mV (n = 7) and decreasedRA/RB by a factor of3. The addition of adenosine to either side (100 µM) hadno effect on any of these parameters. The ATP/UTP responses werepartially inhibited by DIDS and suramin and mediated by a transientincrease in free intracellular Ca2+ concentration (427 ± 206 nM; 15-25 µM ATP, apical; n = 6). This Ca2+ increase was blocked by cyclopiazonic acid, by BAPTA,or by xestospongin C. 31EG4 MEC monolayers also secreted or absorbedfluid in the resting state, and ATP or UTP increased fluid secretion by5.6 ± 3 µl · cm2 · h1(n = 10). Pharmacology experiments indicate that 31EG4epithelia contain P2Y2 purinoceptors on the apical andbasolateral membranes, which upon activation stimulate apicalCa2+-dependent Cl channels and cause fluid secretion acrossthe monolayer. This suggests that extracellular nucleotides could playa fundamental role in mammary gland paracrine signaling and theregulation of milk composition in vivo.

Three marine phytoplankton species (Skeletonema costatum, Olisthodiscusluteus andGonyaulax tamarensis) were grown in batch culturesat 15°C and a 14:10 L:D cycle at irradiance levels rangingfrom 5 to 450 µEinst m–2 s–1. At each irradiance,during exponential growth, concurrent measurements were madeof cell division, carbon-specific growth rate, photosyntheticperformance (both O2 and POC production), dark respiration,and cellular composition in terms of C, N and chlorophyll a.The results indicate that the three species were similar withrespect to chemical composition, C:N (atomic) = 6.9 ±0.4, photo-synthetic quotient, 1.43 ± 0.09, and photosyntheticefficiency, 2.3 ±0.1 x 10–3 µmol O2 (µgChl a)–1 h–1 (µEinst m–2 s–1)–1.Differences in maximum growth rate varied as the –0.24power of cell carbon. Differences in growth efficiency, werebest explained by a power function of Chl a:C at µ = 0.Compensation intensities, ranged from 1.1 µEinst m–2s–1 for S. costatum to 35 forG. tamarensis and were foundto be a linear function of the maintenance respiration rate.The results indicate that interspecific differences in the µ–Irelationship can be adequately explained in terms of just threeparameters: cell carbon at maximum growth rate, the C:Chl aratio (at the limit as growth approaches zero) and the respirationrate at zero growth rate.
