Age and spatial distribution ofAbies firma saplings were analyzed in a mature mixed forest ofA. firma andFagus japonica. There were two major peaks in the age structure ofAbies saplings at around 10 and 75 years of age. Distribution of these age classes of saplings was contagious, while that of all saplings was approximately random. These facts showed that most of the establishment ofA. firma occurs just after a gap is formed, and the frequency at which such establishment occurs is about once every 60 years in a 1600 m2 area.  相似文献

A simple in vitro system is described for the synthesis ofAbies firma-Cenococcum geophilum ectomycorrhizas. SterilizedA. firma seedlings on both MMN and FH media were inoculated with hyphal discs from actively growing margins ofC. geophilum colonies. Typical ectomycorrhizas formed on seedlings on FH medium after 3 mo of incubation. By light microscopy, the synthesized mycorrhizas were seen to possess a thin mantle from which emanated extraradicle hyphae and highly branched, rarely septate intracortical Hartig net mycelium, characteristic ectomycorrhizal features. This is the first report of aseptic ectomycorrhization ofA. firma seedlings byC. geophilum. This model system will facilitate detailed studies on ectomycorrhizal development ofAbies species.  相似文献

Dematiaceous mitosporic fungi darkening decaying fir needles on the ground were studied. Fungal communities on decaying, blackish fir needles were investigated in nine sites of the Kanto district, Japan, using a washing method. A total 108 taxa was recorded from 540 sampled needles. Among abundantly occurring dematiaceous fungi,Anungitea continua, A. uniseptata andEndophragmiella uniseptata were recognized as the major colonizers, forming a hyphal network on the surface of decaying fallen needles and darkening them. The effects of climate on the distributions of seven dematiaceous fungi included the major colonizers were analyzed. The abundance (proportion of needles colonized by a fungal species) ofChaetopsina fulva showed a significantly positive correlation with annual mean air temperature at each sampling site. No other significant correlations between the selected climatic factors and the distributions of dematiaceous fungi were recognized. Molecular markers for alder,Alnus firma Sieb. et Zucc, have not been studied extensively. Here, we used amplified fragment length polymorphism (AFLP) to investigate genetic relationships among 15 natural populations. EcoRI-ACG + Msel-CTG combinations revealed the highest polymorphism (62.2%). A total of 171 DNA fragments were identified. On average, 58.1% of the AFLP markers that were generated using four primer pairs were polymorphic. Diversity was insignificant among the populations. The combination of a wind-pollinated, outcrossing breeding system along with large population sizes, and the ability to regenerate by stump sprouting may explain the high level of genetic diversity within this species. The majority (98%) of the genetic variance resided within populations. The average number of individuals that were exchanged between populations per generation was very high (N em = 12.3). Gene dispersal in alder is apparently by seed dispersalvia water and human activity as well as through pollen. Five individuals per population were claded in the same cluster.  相似文献

The objective of this study was to determine patterns of ectomycorrhizas (ECM) and arbuscular mycorrhizas (AM) colonization associated with Alnus acuminata (Andean alder), in relation to soil parameters (electrical conductivity, field H2O holding capacity, pH, available P, organic matter, and total N) at two different seasons (autumn and spring). The study was conducted in natural forests of A. acuminata situated in Calilegua National Park (Jujuy, Argentina). Nine ECM morphotypes were found on A. acuminata roots. The ECM colonization was affected by seasonality and associated positively with field H2O holding capacity, pH, and total N and negatively associated with organic matter. Two morphotypes (Russula alnijorullensis and Tomentella sp. 3) showed significant differences between seasons. Positive and negative correlations were found between five morphotypes (Alnirhiza silkacea, Lactarius omphaliformis, Tomentella sp. 1, Tomentella sp. 3, and Lactarius sp.) and soil parameters (total N, pH, and P). A significant negative correlation was found between field H2O holding capacity and organic matter with AM colonization. Results of this study provide evidence that ECM and AM colonization of A. acuminata can be affected by some soil chemical edaphic parameters and indicate that some ECM morphotypes are sensitive to changes in seasonality and soil parameters.  相似文献

全球气候变暖对高纬度、高海拔地区的植物形态产生强烈影响。川西林线交错带是青藏高原东部高寒生态系统的重要组成部分,对全球变化极度敏感。以川西林线交错带岷江冷杉(Abies faxoniana)幼苗异龄叶为对象,采用原位开顶式生长室(Open-top chamber,OTCs)模拟增温,研究了长期模拟增温下岷江冷杉幼苗异龄叶片叶长、叶厚等叶形态的响应,采用表型可塑性指数和变异系数对叶形态的可塑性进行分析。结果表明:增温限制岷江冷杉幼苗叶片的增长、增宽和叶面积、体积的扩大,使叶长、叶宽、叶面积、叶体积分别较对照减小12.77%、11.86%、17.76%、11.49%;增温促进叶片厚度的增加,较对照增加7.27%;除叶长宽比外,增温对其余叶形态均产生显著影响(P0.05)。叶形态对模拟增温的响应具有显著的年龄差异(P0.05)。温度、叶龄的交互效应对叶长和叶面积影响显著(P0.05),对叶宽和叶厚影响不显著(P0.05)。两种表型可塑性分析结果表明,除1 a叶叶长外,增温不同程度增大各叶形态可塑性。长期增温使冷杉幼苗叶片有旱生倾向且形态值更发散。研究提供了岷江冷杉幼苗叶片对长期增温的差异性响应证据,为评估青藏高原东缘优势植物对响应气候变暖提供了基础数据和理论参考。  相似文献

Suppression of hepatic stellate cell (HSC) growth and activation have been proposed as therapeutic strategies for the treatment and prevention of liver fibrosis. In the course of screening antifibrotic activity of natural products, the methanolic extract of Alnus firma barks (Betulaceae) showed inhibitory activity of cell proliferation on HSC-T6 cells. A new triterpenoid characterized as lup-20(29) en-2,28-diol-3-yl caffeate (13) was isolated with 12 known diarylheptanoids (1-12) from the barks of A. firma using bioactivity-guided fractionation. Among these compounds, 2 and 13 significantly inhibited the proliferation of HSCs in dose- and time-dependent manners at concentrations from 10 to 100 μM. Taken together, antifibrotic activities of A. firma and its active constituents might suggest the therapeutic potentials against liver fibrosis.  相似文献

Mycorrhizal fungi were isolated from the nonphotosynthetic orchid Chamaegastrodia sikokiana and identified as members of Ceratobasidiaceae by phylogenetic analysis of the internal transcribed spacer (ITS) region of ribosomal deoxyribonucleic acid. The ITS sequences were similar among geographically separated samples obtained from Mt. Kiyosumi in Chiba Prefecture and Mt. Yokokura in Kochi Prefecture. One of the isolated fungi, KI1-2, formed ectomycorrhiza on seedlings of Abies firma in pot culture, suggesting that tripartite symbiosis exists among C. sikokiana, mycorrhizal fungi, and A. firma in nature, and carbon compounds are supplied from A. firma to C. sikokiana through the hyphae of the mycorrhizal fungi. To our knowledge, this is the second study to suggest the involvement of Ceratobasidiaceae fungi in tripartite symbiosis with achlorophyllous orchids and photosynthetic host plants.  相似文献

Apparent photosynthetic rates (APS) of two Zostera noltii Hornem. morphotypes were measured in air and in water at different temperatures with a closed infra-red gas analysis system (IRGA).Hyperbolic functions accurately described the photosynthesis-CO2 relationships when the leaves were exposed to air. The photosynthetic behaviour in water, on the contrary, could not be described by Michaelis type kinetics, due to the existence of a rapid transition from the initial slope to the saturation phase. Both morphotypes (narrow-leaved, NLM and large-leaved, LLM) showed higher APS rates in water than in air, although the highest APS rates, in air as well in water, were recorded for the NLM.Temperature had a significant influence on the photosynthetic parameters: APSmax (maximum photosynthetic rate) decreased (in air and in water) with increased temperature in both morphytypes; compensation points (CP) in air increased at high temperature, especially in the LLM. NLM specimens showed enhanced affinity (lower Km) with increasing temperature in air. On the contrary, Km values in water were not significantly affected by temperature.The results suggest that NLM specimens are better adapted than the LLM to occur exposed to air. The distributional pattern of the two morphotypes in the Palmones Estuary is discussed on the basis of their photosynthetic behaviour.  相似文献

Abies firma is a dominant coniferous tree species endemic to Japan. We isolated eight microsatellite loci from needles of this tree species and tested their polymorphism among 26 A. firma individuals. Six of them showed polymorphism, with two to 16 alleles per locus. Their expected heterozygosities ranged from 0.075 to 0.922. Moreover, interspecific amplification among Abies sachalinensis, Abies mariesii and Abies veitchii was successful in majority of the isolated loci, suggesting that these loci may be useful for characterization of other fir species.  相似文献

Cultures of the cyanobacterium Microcystis firma show rhythmic uptake and release of ammonia under conditions of carbon limitation. The massive removal of ammonia from the medium during the first light phase has little impact on the intracellular pH: a pH shift of less than 0.2 U towards the alkaline can be measured by in vivo 31P NMR. Furthermore, the energy status of the cells remains regulated. In vivo 15N NMR of M. firma, cultivated either with labelled nitrate or ammonia as the sole nitrogen source, reveals only gradual differences in the pool of free amino acids. Additionally both cultivation types show -aminobutyric acid, acid amides and yet unassigned secondary metabolites as nitrogen storing compounds. Investigating the incorporation of nitrogen under carbon limitation, however, only the amide nitrogen of glutamine is found permanently labelled in situ. While transamination reactions are blocked, nitrate reduction to ammonia can still proceed. Ectomycorrhizal synthesis on seedlings of Afzelia quanzensis was initiated in the greenhouse and in the field using basidiospores or soil inoculum originating from fungi associated with adult trees of A. quanzensis, Brachystegia microphylla, B. spiciformis, and Julbernardia globiflora. Of the spore inocula used, only a Pisolithus sp. associated with adult A. quanzensis formed mycorrhizae. Seedlings raised in contact with all soil inocula formed mycorrhizae; however, the mycorrhizal types formed differed between soil inoculum used in the greenhouse and soil inoculum directly used in the field. A. quanzensis has a low specificity for mycorrhizal association. The concepts of ectomycorrhizal succession are also applicable to African savanna ecosystems.  相似文献

The extreme phenotypic variability recognized among the species of Gracilaria has highlighted the need for the application of refined methods to help solve taxa identifications. In Chile, there still exists uncertainty about the exact number of Gracilaria species. Our investigations are centered on DNA analyses of morphotypes collected from different geographical locations, namely Lenga and Isla Santa María, Region VIII (36°00 S to 38°00 S), and Maullín, Region X (39°30 S to 43°40 S). These two regions of Chile are considered as areas of confluence of G. chilensis, G. verrucosa, and a species of Gracilariopsis. In this study four morphotypes, from a natural bed located in Maullín, were analyzed for RFLP of plastid DNA and the results compared with data of four morphotypes from a bed in Lenga. The DNA banding patterns from each enzyme digest were identical irrespective of morphotypes and/or locations. In an attempt to unravel the nature of the morphological differences found among Lenga and Maullín morphotypes, RAPD analyses of nuclear DNA were also performed; however, no polymorphism has been found yet. Therefore, the data of this study, as well as concurrent data from preliminary interfertility tests, suggest that all morphotypes belong to a single taxon, Gracilaria chilensis.Departamento de Botánica, Facultad de Ciencias Naturales y Oceanográficas  相似文献

Megagametophytes of Abies alba containing the immature embryos were dissected from the seed coats and divided by longitudinal and transverse sections. They were placed with the cut surface down on modified Schenk & Hildebrandt medium containing 50 mgl-1 myo-inositol and 2% sucrose, supplemented with 1 mgl-1 N6-benzyladenine (BAP). An embryogenic type of callus proliferated after one month of culture. Closer examination revealed the presence of structures resembling early stages of embryogenesis as well as of single elongated, vacuolated cells and clusters of cells with dense cytoplasm. Under appropriate conditions, some of the somatic embryos elongated and formed cotyledons.  相似文献

Summary In glasshouse experiments, invasion of wheat and grass (Dactylis glomerata) seedling roots byMicrodochium bolleyi was strongly correlated with the pattern and rate of natural senescence of the root cortex. The fungus did not enhance cortical senescence and did not damage roots except in a few instances when it invaded and killed their tips.M. bolleyi behaved as a weak parasite, largely restricted to invasion of naturally senescing cortices of cereal and grass roots.  相似文献

Spatial dispersal patterns of fine litter were analyzed in relation to forest structure within a temperate mixed stand, in northeastern Japan. The canopy layer was a mosaic of two types of patches, those of shade-tolerant Abies firma (estimated age being 81–146 yrs.) and early successional deciduous broad-leaved trees (about 45 yrs. of age). Litter input to the forest floor was observed for a period of three years using 38 traps set at a height of 1.2 m. The average annual total of fine litter was 6.233 (dw.) metric tons hectare-1, 85.7% of the total litterfall. Fine litter was mostly leaves from the overstory (about 64%). Litter distribution maps were drawn, using the Davis's grid method, of the total fine litter and its two main elements, namely A. firma (a 36.1% share) and deciduous broad-leaved species (36.7%). The mosaic distribution of heterogeneous patches of litter was seem to corresponded with distribution of the canopy structure.  相似文献

We investigated the species richness and composition of ectomycorrhizal (EM) fungi colonizing Pinus sylvestris L. seedlings naturally regenerating in boreal forest, in three different microhabitats: on forest ground, on decaying stumps, and within moss layer on erratic boulders. We tested the hypothesis that habitat differences would affect the composition of the EM community of regenerating pine seedlings. In total, 16 EM species were detected, from which none occurred on seedlings growing in all three microhabitats. Piloderma croceum and Cenococcum geophilum were common for seedlings growing in forest ground and on boulders, while Tricholoma aestuans and Suillus luteus were shared between seedlings growing on forest ground and decaying stumps. EM species richness and composition were strikingly different between seedlings regenerating in different microhabitats. Results are discussed as a function of dispersal and niche differentiation of EM fungi.  相似文献

In the present study ectomycorrhizal development of Laccaria bicolor, Rhizopogon luteolus and Suillus bovinus associated with Scots pine (Pinus sylvestris) seedings was studied as affected by primary stand humus, secondary stand humus, podsolic sandy soil or peat in perspex growth chambers. After 9 weeks, ectomycorrhizal development with S. bovinus was significantly greater in peat and primary stand humus than in secondary stand humus or podsolic sandy soil. Ectomycorrhizal development with R. luteolus in secondary stand humus was higher than in primary stand humus. Degree of ectomycorrhizal development of L. bicolor, R. lutuelus and S. bovinus on Scots pine was related to potassium concentration, organic matter content and pH of the soils suggesting that chemical composition of the soils affects ectomycorrhizal development.  相似文献

 Although Pinus pinea L. is an important forest species in the Mediterranean region, few reports exist on its ectomycorrhizal associates. Sixty isolates, obtained from fungal sporocarps collected in mixed forests of P. pinea in Catalonia (northeastern Spain), were tested for ectomycorrhiza formation on containerized P. pinea seedlings when applied as mycelial inoculum produced in peat-vermiculite. A total of 17 isolates, in 8 genera (Amanita, Hebeloma, Laccaria, Lactarius, Pisolithus, Rhizopogon, Scleroderma and Suillus), formed ectomycorrhizas and the percentages of mycorrhizal short roots varied among isolates and species from 13% to 89%. Some of these fungi are cited for the first time in association with P. pinea. The results indicate further fungal candidates for controlled inoculation of P. pinea seedlings in the nursery. Seedlings of O. violaceus were cultured on MS media and treated at low temperature. Cold treatment at 5–7 °C for more than 7 days was needed for flower induction of seedlings in vitro originated from germinated seeds. When cultured on MS media supplemented with 2.5 mg l−1 zeatin and 2 mg l−1 gibberellin (GA3), seedlings in vitro did initiate flowers without cold treatment. When MS media was used with a reduced amount of NH4NO3, flower induction of seedlings in vitro could be accelerated.
