Key message

Abstract

Melia azedarach is an important forest tree with pharmaceutical, insecticidal, pesticidal, and commercial significance. It is a good reforestation tree because of its fast growth and drought hardy nature. Genetic variation in a species allows itself to adapt, evolve and respond to environmental stress. It provides the basis for survival of a species and critically influences its evolutionary potential. Assessment of genetic diversity is necessary for improvement and conservation of a species. For this, microsatellite markers are of particular interest given the attributes like co-dominance, reproducibility, hyper variability and abundance throughout the genome. In the present study, we analyzed the genetic diversity and population structure of M. azedarach, an ecologically imperative species growing in the North-Western Plains of India. We developed 43 microsatellite markers, of which 20 were subsequently employed for analysis of diversity and population structure among 33 populations encompassing 318 genotypes representing North-Western Plains of India. A moderate level of diversity (Na = 5.1, Ho = 0.506, He = 0.712, I = 1.386) was assessed. The highest value of ΔK estimated using STRUCTURE indicated 2 subpopulations (K = 2). AMOVA exhibited 73 % variation within populations and 12 % variation was found among regions. Significant positive correlation between geographical and genetic distance was found (Rxy = 0.365, P = 0.010). The present study lays a foundation on a better understanding of genetic dynamics of the species and reveals its diversity and population structure in North-Western Plains of India.

Life-stage-based management of marine fishes requires information on juvenile habitat preferences to ensure sustainable population demographics. This is especially important in the Arctic region given very little is known about the life histories of many native species, yet exploitation by developing commercial and artisanal fisheries is increasing as the ice extent decreases. Through scientific surveys and bycatch data from gillnet fisheries, we document captures of rarely reported juvenile Greenland sharks (Somniosus microcephalus; ≤200 cm total length [TL]) during the ice-free period in the Canadian Arctic. A total of 22 juvenile animals (42 % of total catch; n = 54), including the smallest reliably measured individual of 117 cm TL, were caught on scientific longlines and bottom trawls in Scott Inlet and Sam Ford Trough over three consecutive years. Molecular genetic nuclear markers confirmed species identity for 44 of these sharks sampled; however, two sharks including a juvenile of 150 cm TL were identified as carrying a Pacific sleeper shark (Somniosus pacificus) mitochondrial cytochrome b (cyt b) haplotype. This represents the first record of a Pacific sleeper shark genetic signature in Greenland sharks in Eastern Arctic waters. Juvenile sharks caught as bycatch in gillnet fisheries were only observed offshore in Baffin Bay surrounding a fishery closure area, while larger subadult and mature Greenland sharks (>200 cm TL) were caught in all fishing locations, including areas where juveniles were observed. The repeatable occurrence of juvenile Greenland sharks in a fjord and their presence at two offshore sites indicates that these smaller animals either reside in nurseries or have defined home ranges in both coastal and offshore regions or undertake large-scale inshore–offshore movements.  相似文献

Two new species of Gentianella (Gentianaceae, Gentianeae, Swertiinae), G. grantii and G. wayqecha, are described from Departamento Cusco, Peru. These two species differ from other Peruvian species of Gentianella in a combination of stems 10–100 cm long; no rosette of basal leaves; cauline leaves 10–75 mm long; flowers in thyrses; and corollas 14–26 mm long, campanulate, and adaxially glabrous. The leaves of G. grantii consist of an appressed pseudopetiole and a spreading, narrowly elliptic-oblong to linear blade; the corollas are lavender; and the corolla lobes are 0.6–0.7× as long as the tube. Gentianella grantii is similar to G. lythroides, of Bolivia, but differs in having more closely spaced leaves and less deeply lobed corollas. The leaves of G. wayqecha are sessile, lanceolate to ovate; the corollas are rose-violet; and the corolla lobes are 0.75–1.35× as long as the tube. Gentianella wayqecha is similar to G. rapunculoides, of Colombia and Ecuador, and G. ruizii, of Peru, but differs from both in its adaxially glabrous corollas and from G. rapunculoides in its less deeply lobed corollas. Both G. grantii and G. wayqecha grow in moist habitats near tree line, and are known only from a area northeast of the city of Cusco.  相似文献

Abstract

A set of artiodactyl footprints preserved in the Pie de Vaca Formation from the Plio-Pleistocene of south-central Puebla in central Mexico is formally described. The sample consists of five footprints that represent a trackway produced by one individual. A comparative study and a taxonomic assessment were performed in order to identify the track-maker. The ichnites are characterized by having a rounded shape, length of about 5 cm, two hoof imprints that are longer than wide, and tips of the hooves directed forwards. Their size and morphology are comparable to those produced by extant peccaries. The spatial distribution of the footprints has been related to an individual in fast walking or even in a slow trot. Ichnotaxonomically, the record represents a new ichnospecies formally referred as to Tayassuichnum felixarangutii. A member of Platygonus, the most common peccary for the late Cenozoic of North America is proposed as the potential track-maker. The record contributes to the (indirect) presence of peccaries in central Mexico during the late Cenozoic, representing the first documented ichnites referable to that group of artiodactyls in the Mexican territory and the second mention in the North American subcontinent.

Lakes have an esthetic significance that is particularly important for attracting tourism. In this context, it is often preferable for lakes to have clear water, so many lake managers attempt to achieve clear lake water by various means. However, the lakes of Kashmir Himalaya are undergoing several complex ecological changes due to, for example, increasing tourism, overfishing, and intensive agriculture, which are making these lakes less clear. One such change is the vigorous growth and development of aquatic weeds in the shallow-water areas of Kashmir Himalayan lakes. We thus, investigated the response of Nymphoides peltatum, a rapidly multiplying clonal species, to water depth, in order to determine whether water depth can be used to control the spread of this proliferating macrophyte. Different traits of the given plant species, such as the mean number of ramets, were significantly higher (F = 55.412, p = 0.000) at depth zone D1 (0–100 cm) than at depth zones D2 (101–200 cm) and D3 (201–300 cm). In all of the lakes, mean spacer length—a tool for facilitating plant spread—was observed to be significantly higher (F = 45.890, p = 0.000) at lower water levels (0–100 cm). Also, the reproductive structures (flowers) of N. peltatum showed significant variation with depth (F = 51.909, p = 0.000) and with the lake examined (F = 9.909, p = 0.001). Thus, the results obtained during the present study indicate the importance of water depth in the management of N. peltatum in various Kashmir Himalayan lakes.  相似文献

Abstract

The southern conifer Agathis australis is amongst the largest and longest-lived trees in the world. We measured sap flux densities (F d) in kauri trees with a DBH range of 20–176 cm to explore differences in responses of trees of different sizes to seasonal conditions and summer drought. F d was consistently higher in larger trees than smaller trees. Peak F d was 20 and 8 g m?2 s?1 for trees of diameters of 176 and 20 cm, respectively, during the wet summer. Multiple regression analysis revealed photosynthetically active radiation (PAR) and vapour pressure deficit (D) were the main drivers of F d. During drought, larger trees were more responsive to D whilst smaller trees were more responsive to soil drying. Our largest tree had a sapwood area of 3,600 cm2. Preliminary analysis suggests stem water storage provides a buffer against drying soil in larger trees. Furthermore, F d of smaller trees had higher R 2 values for soil moisture at 30 and 60 cm depth than soil moisture at 10 cm depth (R 2 = 0.68–0.97 and 0.55–0.67, respectively) suggesting that deeper soil moisture is more important for these trees. Larger trees did not show a relationship between F d and soil moisture, suggesting they were accessing soil water deeper than 60 cm. These results suggest that larger trees may be better prepared for increasing frequency and intensity of summer droughts due to deeper roots and/or larger stem water storage capacity.

Cucumis × hytivus (2n = 4× = 38) is a synthetic allotetraploid obtained from interspecific hybridization between the cucumber (2n = 2× = 14) and its wild relative C. hystrix (2n = 2× = 24). The synthesis of this species built a bridge for cucumber improvement through gene introgression. Allotriploid and introgression lines (ILs) have previously been produced and characterized with respect to morphology, cytology, and molecular markers. However, no clear evidence of how the chromosomal segments of C. hystrix were introgressed and inherited was found owing to the small size of chromosomes. In the present study, cucumber-C. hystrix introgression lines were developed by backcrossing the allotriploid to North China cucumber breeding line “P01” followed by self-pollination. The introgressed segments of C. hystrix in the ILs were revealed by meiotic pachytene chromosome analysis. Fluorescence in situ hybridization (FISH) was performed on pachytene chromosomes using fosmid clones from cucumber, which confirmed that introgression occurred in the long arm of chromosome 7. Molecular analysis using a set of 53 simple sequence repeats (SSRs) indicated that the chromosomal segments of C. hystrix were introduced into 4 cucumber chromosomes, the short arms of chromosomes 2 and 6, and long arms of chromosomes 3 and 7. The inheritance of alien sequences in the long arm of chromosome 7 was investigated with 21 SSRs in self-pollinated progenies. C. hystrix-specific bands of several SSRs were still present in some individuals, indicating that the introgressed segment was partially preserved. The first unambiguous identification of alien chromosome segments in cucumber ILs using combined molecular cytogenetics could facilitate the determination of effects of wild alleles and promote cucumber improvement.  相似文献

Reproduction and maturation in the economically important, but data-deficient, Mustelus minicanis and M. norrisi were analysed using catches of populations exploited by a gillnet fishery during two years in the southern Caribbean Sea. In total, 691 female (mean ± SD total length–TL of 55.3 ± 5.8 cm) and 503 male (50.4 ± 4.9 cm TL) M. minicanis were assessed, with ~95% of all specimens deemed mature. Almost 25% of females were gravid (occurring between January and October) and with variable temporal development of up to six embryos (3.3 ± 1.2), implying protracted temporal parturition. Parity in the sex ratio of embryos, but not in landed catches, suggested sexual segregation across the fished area. The 50% sizes at maturity (M 50) (± SE) were similarly estimated at 45.11 (± 0.39) and 45.48 (± 0.42) cm TL for females and males, respectively. Relatively fewer (235) M. norrisi were landed, with samples comprising 150 females (82.6 ± 18.1 cm TL) and 85 males (75.5 ± 17.7 cm TL). More than 30% of both sexes were immature. Ten percent of females were gravid (up to 11 embryos) and present in catches between October and February, coinciding with the northern hemisphere autumn/winter. Female and male M 50s were 76.65 (± 1.16) and 69.63 (± 1.92) cm TL, respectively. The results imply variable inter-specific reproductive plasticity and the need for further life-history studies. Increasing gillnet selectivity might represent a simple precautionary management option for concurrently regulating catches of the smaller-bodied M. minicanis during peak abundances of gravid females and similar-sized juvenile M. norrisi.  相似文献

Two western Pacific triplefins, Enneapterygius fuscoventer Fricke 1997 and E. howensis Fricke 1997 (Perciformes: Tripterygiidae), are similar to each other in sharing 15–19 (usually 17) notched lateral-line scales and the mandibular pore formula 3–5 + 1 + 3–5 (usually 4 + 1 + 4), in addition to similar coloration, viz. body with four vertical bands, the first and second forked ventrally, dorsal-fin membrane semi-transparent, anal fin entirely blackish, and caudal fin blackish with a semi-transparent margin. These species have previously been known only from preserved specimens. Examination of additional specimens plus color photographs of males and females of both species when fresh, and comparisons with type specimens resulted in several features, including coloration and counts of second dorsal-fin spines, anal-fin soft rays, pored lateral-line scales and longitudinal scale rows, being regarded as new diagnostic characters. Enneapterygius fuscoventer and E. howensis have been newly recorded from southern Japan and coastal eastern Australia, respectively.  相似文献

Availability of molecular methods, gene sequencing, and phylogenetic species recognition have led to rare fungi being recognized as opportunistic pathogens. Fungal keratitis and onychomycosis are fairly common mycoses in the tropics, especially among outdoor workers and enthusiasts. The frequently isolated etiological agents belong to genera Candida, Aspergillus, and Fusarium. Within the genus Fusarium, known to be recalcitrant to prolonged antifungal treatment and associated with poor outcome, members of the Fusarium solani species complex are reported to be most common, followed by members of the Fusarium oxysporum SC and the Fusarium fujikuroi SC (FFSC). Morphological differentiation among the various members is ineffective most times. In the present study, we describe different species of the FFSC isolated from clinical specimen in south India. All twelve isolates were characterized up to species level by nucleic acid sequencing and phylogenetic analysis. The molecular targets chosen were partial regions of the internal transcribed spacer rDNA region, the panfungal marker and translation elongation factor-1α gene, the marker of choice for Fusarium speciation. Phylogenetic analysis was executed using the Molecular Evolutionary Genetics Analysis software (MEGA7). In vitro susceptibility testing against amphotericin B, voriconazole, posaconazole, natamycin, and caspofungin diacetate was performed following the CLSI M38-A2 guidelines for broth microdilution method. The twelve isolates of the FFSC were F. verticillioides (n = 4), F. sacchari (n = 3), F. proliferatum (n = 2), F. thapsinum (n = 1), F. andiyazi (n = 1), and F. pseudocircinatum (n = 1). To the best of our knowledge, this is the first report of F. andiyazi from India and of F. pseudocircinatum as a human pathogen worldwide. Natamycin and voriconazole were found to be most active agents followed by amphotericin B. Elderly outdoor workers figured more among the patients and must be recommended protective eye wear.  相似文献

The conditions of storage, cultivation and maintenance of microbial cultures should preserve the microbiological homogeneity, phenotypic and genotypic characteristics to ensure better reproducibility of metabolic production. To evaluate the influence of the storage condition on the composition of cell fatty acids, genetic profile and biochemical characteristics of Xanthomonas campestris pv. mangiferaeindicae IBSBF 2103, as well as, to identify its relationship with the yielding and viscosity of the xanthan gum produced, this study monitored the strain preserved in two simple and widely used conditions, ultra-freezer (?80 °C) and refrigeration (3–8 °C) during 5 months. Were identified and quantified 13 fatty acids. The cells preserved at ?80 °C showed more stable concentration of all fatty acids, producing more xanthan gum and with higher viscosity. The chromosomal analysis obtained with the enzyme XbaI revealed 17 distinct fragments with maximum size of 485 kilobases, without variations among the subcultures maintained in both storage conditions. The X. campestris pv. mangiferaeindicae subcultures preserved at ?80 °C showed less pronounced phenotypic variations, which had positive influence in the qualitative and quantitative characteristics of the xanthan gum produced.  相似文献

In situ edaphic factors affecting seed germination and seedling emergence of three framework species of Acacia were investigated with the intent of developing fundamental and scalable restoration capacity for Arabian dryland restoration. Direct seeding represents the most efficient means to restore vegetation at the landscape scale and this study provides insight into edaphic and ecological limitations, as well as effective protocols governing the use of native seeds for restoration in hyper-arid environments. The study was conducted in extant Acacia woodland habitat on conserved land (Thumamah Nature Park) in close proximity to Riyadh, Saudi Arabia. Broad-scale direct seeding using un- and pretreated Acacia gerrardii, A. tortilis, and A. ehrenbergiana seed, and two seed burial depths were implemented across three sites with distinct soil surface characteristics. Eight weeks post-sowing, random samples for each species × seed treatment × burial depth combination were excavated, sieved, and categorized as follows: failed to germinate, germinated but died prior to emerging, or successfully emerged. We show that germination and emergence of Acacia gerrardii, A. tortilis, and A. ehrenbergiana were driven by a three-way interaction among species, site, and seed burial depth. Treating seed with the signaling compound Moddus did not have a definitive effect, positive or negative, on any of the species investigated. Acacia gerrardii was the only species that exhibited widespread emergence, though emergence was not consistent across sites or burial depths. Germination was highest in disturbed soil (up to 69% for A. gerrardii), but very few (<2%) successfully emerged; a greater proportion of germinants in sandy soil emerged (up to 44% for A. gerrardii) even though the overall germination was less. Though species-dependent, a 2-cm sowing depth was most effective in sand; while in disturbed soil, sowing depths of 1 and 2 cm were comparable; and no germination was observed in gravelly clay soil. Sandy soil exhibited rapid water infiltration (107.6 mm min?1), and post-sowing surface crusting was a non-factor (0.44 kg cm?2). Disturbed soil exhibited moderate water infiltration (1.46 mm min?1) and post-sowing surface crusting was double that of sand (0.88 kg cm?2) and restrictive on seedling emergence. Gravelly clay exhibited extremely poor water infiltration (0.12 mm min?1), and surface crusting was severe (4.49 kg cm?2) and an order of magnitude greater than sand. The medium-coarse sand fraction, a key driver of the observed soil surface processes, was greatest in sand (55%) and significantly less and uniform in the disturbed (22%) and gravelly clay (22%) soils. Our findings demonstrate that soil surface characteristics and associated processes can dictate ecological processes at depths as shallow as 1–2 cm, and that soil crusts that slow water infiltration and impede seedling emergence rapidly reconstitute after disturbance; both are important considerations for restoring dryland vegetation.  相似文献

Abstract

Sauropod tracks from the Early Jurassic Maanshan Member of the Ziliujing Formation in the Dazhuanwan area of Guizhou represent the first Jurassic dinosaur track record for this province. The best preserved and longest trackway is narrow gauge (Brontopodus type), and indicates a relatively small trackmaker (footprint length ~35.0 cm). In conjunction with sauropodomorph skeletal remains these tracks suggest that basal sauropodomorphs and primitive sauropods coexisted in this region during the Early Jurassic. This pattern is same in the Lufeng and Sichuan basins. Thus, sauropod tracks from southwest China are diverse in the Early Jurassic, and include narrow gauge Parabrontopodus, wide gauge Brontopodus-type, and basal sauropodomorph tracks.  相似文献

Background

Male European seabass, already predominant (~?70%) in cultured stocks, show a high incidence (20–30%) of precocious sexual maturation under current aquaculture practices, leading to important economic losses for the industry. In view of the known modulation of reproductive development by swimming exercise in other teleost species, we aimed at investigating the effects of sustained swimming on reproductive development in seabass males during the first year of life in order to determine if swimming could potentially reduce precocious sexual maturation.

Methods

Pre-pubertal seabass (3.91?±?0.22 g of body weight (BW)) were subjected to a 10 week swimming regime at their optimal swimming speed (Uopt) in an oval-shaped Brett-type flume or kept at rest during this period. Using Blazka-type swim tunnels, Uopt was determined three times during the course of the experiment: 0.66 m s??1 at 19?±?1 g BW, 10.2?±?0.2 cm of standard length (SL) (week 1); 0.69 m s??1 at 38?±?3 g BW, 12.7?±?0.3 cm SL (week 5), and also 0.69 m s??1 at 77?±?7 g BW, 15.7?±?0.5 cm SL (week 9). Every 2 weeks, size and gonadal weight were monitored in the exercised (N?=?15) and non-exercised fish (N?=?15). After 10 weeks, exercised and non-exercised males were sampled to determine plasma 11-ketotestosterone levels, testicular mRNA expression levels of genes involved in steroidogenesis and gametogenesis by qPCR, as well as the relative abundance of germ cells representing the different spermatogenic stages by histological examination.

Results

Our results indicate that sustained swimming exercise at Uopt delays testicular development in male European seabass as evidenced by decreased gonado-somatic index, slower progression of testicular development and by reduced mRNA expression levels of follicle stimulating hormone receptor (fshR), 3-beta-hydroxysteroid dehydrogenase (3βhsd), 11-beta hydroxysteroid dehydrogenase (11βhsd), estrogen receptor-beta (erβ2), anti-mullerian hormone (amh), structural maintenance of chromosomes protein 1B (smc1β), inhibin beta A (inhba) and gonado-somal derived factor 1 (gsdf1) in exercised males as compared with the non-exercised males.

Conclusions

Swimming exercise may represent a natural and non-invasive tool to reduce the incidence of sexually precocious males in seabass aquaculture.

In this study, we assessed geographic patterns of genetic variations in nuclear and chloroplast genomes of two related native oaks in Japan, Quercus aliena and Q. serrata, in order to facilitate development of genetic guidelines for transfer of planting stocks for each species. A total of 12 populations of Q. aliena and 44 populations of Q. serrata were analyzed in this study. Genotyping of nuclear microsatellites in Q. aliena was done with only nine populations (n = 212) due to limited numbers of individuals in two populations, while all 12 populations (n = 89) were used in sequencing chloroplast DNA (cpDNA). In Q. serrata, 43 populations (n = 1032) were genotyped by nuclear microsatellite markers, while cpDNA of 44 populations (n = 350) was sequenced. As anticipated, geographic patterns detected in the variations of Q. aliena’s nuclear genome and its chloroplast haplotype distribution clearly distinguished northern and southern groups of populations. However, those of Q. serrata were inconsistent. The geographic distribution of its chloroplast haplotypes tends to show the predicted differentiation between northern and southern lineages, but geographic signals in the genetic structure of its nuclear microsatellites are weak. Therefore, treating northern and southern regions of Japan as genetically distinct transferrable zones for planting stocks is highly warranted for Q. aliena. For Q. serrata, the strong NE-SW geographic structure of cpDNA should be considered.  相似文献

To identify potential linkages between specific bacterial populations and process performance in anaerobic digestion, the dynamics of bacterial community structure was monitored with high-throughput sequencing in triplicate anaerobic digesters treating animal waste. Firmicutes and Bacteroidetes were found as the two most abundant populations, however, with contrasting population dynamics in response to organic overloading. Firmicutes dominated the bacterial community during stable process performance at low organic loading rate, representing over 50 % of the bacterial abundance. In contrast, the onset of organic overloading raised the relative abundance of Bacteroidetes from 20 ± 2.6 to 44 ± 3.1 %. In addition to the significant negative correlation between the relative abundance of Firmicutes and Bacteroidetes, populations of Firmicutes and Bacteroidetes were found to be linked to process parameters including organic loading rate, volatile fatty acids concentration, and methane production. Therefore, the population abundance ratio of Firmicutes to Bacteroidetes (F/B ratio) was suggested as a potential indicator for process performance. The interactions between Firmicutes and Bacteroidetes populations could be exploited to develop strategies for the prevention of performance perturbation in anaerobic digestion processes.  相似文献

V. cholerae, V. parahaemolyticus, and V. vulnificus are recognized human pathogens. Although several studies are available worldwide, both on environmental and clinical contexts, little is known about the ecology of these vibrios in African coastal waters. In this study, their co-occurrence and relationships to key environmental constraints in the coastal waters of Guinea-Bissau were examined using the most probable number-polymerase chain reaction (MPN-PCR) approach. All Vibrio species were universally detected showing higher concentrations by the end of the wet season. The abundance of V. cholerae (ISR 16S-23S rRNA) ranged 0–1.2 × 104 MPN/L, whereas V. parahaemolyticus (toxR) varied from 47.9 to 1.2 × 105 MPN/L. Although the presence of genotypes associated with virulence was found in environmental V. cholerae isolates, ctxA+ V. cholerae was detected, by MPN-PCR, only on two occasions. Enteropathogenic (tdh+ and trh+) V. parahaemolyticus were detected at concentrations up to 1.2 × 103 MPN/L. V. vulnificus (vvhA) was detected simultaneously in all surveyed sites only at the end of the wet season, with maximum concentrations of 1.2 × 105 MPN/L. Our results suggest that sea surface water temperature and salinity were the major environmental controls to all Vibrio species. This study represents the first detection and quantification of co-occurring Vibrio species in West African coastal waters, highlighting the potential health risk associated with the persistence of human pathogenic Vibrio species.  相似文献

Inulin is a readily available feedstock for cost-effective production of biochemicals. To date, several studies have explored the production of bioethanol, high-fructose syrup and fructooligosaccharide, but there are no studies regarding the production of d-lactic acid using inulin as a carbon source. In the present study, chicory-derived inulin was used for d-lactic acid biosynthesis by Lactobacillus bulgaricus CGMCC 1.6970. Compared with separate hydrolysis and fermentation processes, simultaneous saccharification and fermentation (SSF) has demonstrated the best performance of d-lactic acid production. Because it prevents fructose inhibition and promotes the complete hydrolysis of inulin, the highest d-lactic acid concentration (123.6 ± 0.9 g/L) with a yield of 97.9 % was obtained from 120 g/L inulin by SSF. Moreover, SSF by L. bulgaricus CGMCC 1.6970 offered another distinct advantage with respect to the higher optical purity of d-lactic acid (>99.9 %) and reduced number of residual sugars. The excellent performance of d-lactic acid production from inulin by SSF represents a high-yield method for d-lactic acid production from non-food grains.  相似文献

Background

All animals have mechanisms for healing damage to the epithelial sheets that cover the body and line internal cavities. Epithelial wounds heal either by cells crawling over the wound gap, by contraction of a super-cellular actin cable (“purse string”) that surrounds the wound, or some combination of the two mechanisms. Both cell crawling and purse string closure of epithelial wounds are widely observed across vertebrates and invertebrates, suggesting early evolution of these mechanisms. Cnidarians evolved ~600 million years ago and are considered a sister group to the Bilateria. They have been much studied for their tremendous regenerative potential, but epithelial wound healing has not been characterized in detail. Conserved elements of wound healing in bilaterians and cnidarians would suggest an evolutionary origin in a common ancestor. Here we test this idea by characterizing epithelial wound healing in live medusae of Clytia hemisphaerica.

Results

We identified cell crawling and purse string-mediated mechanisms of healing in Clytia epithelium that appear highly analogous of those seen in higher animals, suggesting that these mechanisms may have emerged in a common ancestor. Interestingly, we found that epithelial wound healing in Clytia is 75 to >600 times faster than in cultured cells or embryos of other animals previously studied, suggesting that Clytia may provide valuable clues about optimized healing efficiency. Finally, in Clytia, we show that damage to the basement membrane in a wound gap causes a rapid shift between the cell crawling and purse string mechanisms for wound closure. This is consistent with work in other systems showing that cells marginal to a wound choose between a super-cellular actin cable or lamellipodia formation to close wounds, and suggests a mechanism underlying this decision.

Conclusions

1. Cell crawling and purse string mechanisms of epithelial wound healing likely evolved before the divergence of Cnidaria from the bilaterian lineage ~ 600mya 2. In Clytia, the choice between cell crawling and purse string mechanisms of wound healing depends on interactions between the epithelial cells and the basement membrane.
