A microwave acid digestion method for the preparation of biological samples for PIXE analysis is presented. The precision and accuracy of the entire PIXE analytical procedure, including the micro-wave digestion step, were evaluated by analyzing eight certified reference materials. For elements heavier than K, and for concentration levels from 2 μg/g upward, the total random error of a single analysis is in the range of 2–5%. The accuracy is better than 5%. The detection limits are down to 0.3 μg/g.

Epithermal instrumental neutron activation analysis (EINAA) has been used to determine the iodine content of many individual food materials that constitute the typical Libyan diet. The selected samples include different varieties of local and imported foods such as wheat and barley products, rice, bread, legumes such as chick peas and lentil, table salt, and commonly used spices, including thyme and fenugreek. Both conventional and anticoincidence γ-ray spectrometry techniques have been employed. Epithermal INAA in conjunction with anticoincidence counting has been found to provide the most reliable results. For quality control purposes, a number of NIST biological reference materials were analyzed. The range of daily dietary intake has been calculated as 100–180 μg of iodine per day, which is within the recommended range. Bread was identified as a significant source of iodine in the Libyan diet, as it contributed 99 μg/d.  相似文献

The National Institute of Standards and Technology has developed a new Standard Reference Material 1941, “Organics in Marine Sediment.” In addition to the organic constituents, over 30 elements have been determined by instrumental neutron activation analysis and prompt-gamma activation analysis. The homogeneity of the material was investigated and relative standard deviations of single-element concentrations in 250-mg samples were found to be 1% or less with regard to major inorganic constituents and rare earth elements. A slightly higher relative SD was found for elements that may stem from biological or anthropogenic input. The element concentrations determined in this work are discussed in comparison to concentrations in other similar reference materials. Concentrations for 31 elements will be included for information on the certificate.

ABSTRACT

Students often have misconceptions about terrestrial carbon flows and there is a lack of coherence in students’ explanations regarding the different levels of biological organisation at which these processes occur. In this study, problem-based teaching materials on the topic of terrestrial carbon flows were developed and tested with 15 students 18–19 years old (grade 12) using a pre/post-test design. Students focused on specific carbon flows in an ecosystem at the macro level (e.g. CO2 fixation and heterotrophic respiration) including specific related concepts at the micro level (e.g. photosynthesis and cellular respiration). The findings indicate that the teaching materials improved the students’ understanding of terrestrial carbon flows and their ability to reason across the different levels of biological organisation. On the basis of these findings, implications for teaching terrestrial carbon flows in biological education are discussed.  相似文献

Abstract

We have developed a rapid method for producing homogeneous mono-[12]A14 iodoinsulin with high specific activity and yield. After iodination by the lactoperoxidase method, the labeled peptides were applied to a C18 PorasilR prs-column, washed with aqueous buffer to eliminate the free [12]-iodide and placed “in-line” with a C-18 HPLC column; mono-[12]A14 iodoinsulin was then eluted isocratically with 29% aceto-nitrile in 16 minutes. The labeled hormone was extremely stable, and proved suitable for various biological studies.  相似文献

Synthesis and biological evaluation of a novel library of fused 1,2,3-triazole derivatives are described. The in-house developed multicomponent reaction based on commercially available starting materials was applied and broad biological screening against various viruses was performed, showing promising antiviral properties for compounds 14d, 14n, 14q, 18f and 18i against human coronavirus 229E. Further in silico studies identified the key molecular interactions between those compounds and the 3-chymotrypsin-like protease, which is essential to the intracellular replication of the virus, supporting the hypothesis that the protease is the target molecule of the potential antiviral derivatives.  相似文献

A microcomputer-controlled irradiation and measurement system and a microprocessor-controlled sample changer have been installed at the SLOWPOKE-2 Facility at the Royal Military College of Canada (RMC). These systems can provide the gamut of instrumental neutron activation analysis (INAA) techniques for the analyst. Custom software has been created for system control, data acquisition, and off-line spectral analysis using programs that incorporate Gaussian peak-fitting methods of analysis. The design and use of the equipment is discussed, and the performance is illustrated with results obtained from the analysis of marine sediment and biological reference materials.  相似文献

The selenium determination in biological materials by the classical fluorometric method (FM) is time-consuming and also hazardous, as it requires the destruction of the organic matrix samples with hot HNO3/HClO4 mixtures prior to analysis. Accordingly, commercial analytical laboratories are increasingly using faster instrumental methods; for sample digestion, avoid using HClO4. Because of these procedural changes, the results obtained by commercial laboratories may be unreliable, especially for samples containing Se in organic forms. One such “difficult” substrate is Se yeast, which contains most of its Se as selenomethionine. To establish which methods for Se analysis and sample digestion are applicable, samples of Se yeast and of selenomethionine standards were sent to laboratories employing either flame atomic absorption spectrometry (FAAS), inductively coupled plasma-mass spectrometry (ICP-MS), or hydride generation atomic absorption spectrometry (HGAAS). The result were compared with those obtained by FM and non-destructive instrumental neutron activation analysis (INAA). ICP-MS, after microwave digestion of sample with HNO3/H2O2, produced results within 5% of the expected values, as did those obtained by FM and INAA. With FAAS, acceptable results were obtained after digestion with HNO3/HCl. With HGAAS, sample digestion with HNO3/H2O2 produced values that were systematically elevated by about 10% and exhibited standard deviations of ≥10%. Thus, current methods of sample digestion are applicable for Se yeast analysis by ICP-MS and FAAS, but not by HGAAS.  相似文献

Purpose

The main goal of this work is to evaluate the environmental impact of a 63-m blade for wind generators. The embodied energy and the carbon footprint are used as supporting tools for material selection in the initial project stages.

Methods

Real industrial data regarding the most used materials for wind turbine blade construction are used. Two eco-parameters, embodied energy and carbon footprint, were calculated from each selected material together with values of manufacture, transport, use, and final disposal. The blades must be built to have a mechanical strength high enough to withstand vibrations caused by manufacturing flaws, turbulence, or irregular loading. In this sense, Young’s modulus, yield strength, and density were compared to the environmental footprint data to support the final material choice. This evaluation process of the possible materials to be used in the blade manufacture was carried out in the initial stages of the project.

Results

Composite materials such as glass fiber-reinforced polymer (GFRP) and carbon fiber-reinforced polymer (CFRP), bonded together with an adhesive material, are used to build modern wind turbine blades. Those composites comprise a considerable number of different materials that can be mixed to reach adequate performance. Comparisons were made with 46 pre-selected materials, considering the mechanical behavior and environmental impacts. The final selected materials have better properties than the reference material. Finally, two materials with the desired mechanical properties and with a potential lower negative environmental impact than the reference material were selected.

Conclusions

Replacing the reference resin—epoxy/E-glass fiber—with the epoxy resin with the lowest environmental impact—epoxy/S-glass fiber—will reduce the total value of the environmental load to 102 GJ of energy and 3.4 t of CO2. As important as the material selection in the early stages of product development is the end of life (EoL) choice. In this case, the glass fiber has an EoL potential of 370 GJ of energy and 460 t of CO2 in the remanufacturing option, against zero for the landfill. This work shows that carefully selected raw materials and EoL alternatives for WTB can significantly reduce the environmental impact of this component.

Background: Chemoradiotherapy (CRT) is currently performed for patients with advanced esophageal carcinoma. Sensitivity of tumours to CRT differs from one case to another and may be influenced by the expression of biological molecules. The aim of this study was to identify biological markers which could predict sensitivities of esophageal squamous cell carcinoma (ESCC) to CRT.

Methods: A total of 84 patients with stage I–IV ESCC were evaluated. The cytokeratin 19 fragment antigen 21-1 (CYFRA21-1) and carcinoembryonic antigen (CEA) levels were measured before CRT by enzyme-linked immunosorbent assays in patients with primary ESCCs using 3.4?ng ml?1 and 3.3?ng ml?1, respectively, as cut-off values. The relationships between pretreatment expression of CYFRA 21-1 and CEA and the effectiveness of CRT were analysed.

Results: The complete response (CR) rates of the primary tumours estimated by computed tomography in patients with high levels of CYFRA21-1 and CEA were 10% (3/30) and 4.2% (1/24), while in cases with low CYFRA21-1 and CEA the CR rates were 50% (27/54) and 48.3% (29/60), respectively (p?=?0.002 and 0.003). The effective rates (CR+PR) in CYFRA21-1 high and low groups were 60% (18/30) and 96.3% (52/54), while in CEA high and low groups they were 58.3% (14/24) and 93.3% (56/60), respectively (p?=?0.013 and 0.013).

Conclusion: CYFRA21-1 and CEA may be helpful in predicting the responsiveness in ESCC of primary lesions to CRT, although the results should be confirmed in larger, more homogeneous studies.  相似文献

Abstract

Arsenic (As) is found naturally in the geological strata within the Ba Men Region of Inner Mongolia, China. A study was conducted to compare the total As measurements from two analytical techniques: instrumental neutron activation analysis (INAA) and atomic fluorescence spectrometry (AFS), and to verify nails as an exposure biomarker in this population. In 1999, nail and water samples were collected in a pilot study. Fingernails and toenails were pooled from 32 participants and analysed for total As by both INAA and AFS. Mean nail As values were 14.8±2.4 and 19.4±2.8 µg?g?1 (±SEM) for INAA and AFS, respectively. Results from these two methods were significantly correlated (r=0.93, p<0.0001). In 2000, a second study was conducted and INAA was used to measure total As in toenails from 314 Ba Men residents. Well water samples were collected from 121 households and analysed by AFS. A significant correlation was observed between toenail and well water As (r=0.84, p<0.0001). Based on the results, INAA was significantly correlated with AFS and proved to be a reliable measure of nail As levels. In this population, toenail samples are a useful internal As exposure biomarker from drinking water sources.  相似文献

The metabolism of 14C-labeled fatty acids and triacylglycerols was followed in intact germinating oil palm seedlings as well as in tissue slices. In the germinating seedling, the shoot contained a normal pattern of membrane fatty acids (mainly C16, C18:1, C18:2) but the kernel contained about 68% C12 and C14 fatty acids. Haustorium fatty acids were intermediate between the two. [14C]Acetate was actively metabolized by shoot and haustorium slices but not so actively by the kernel. Approximately 9% to 17% was converted to water-soluble substances, 4% to 6% to CO2, and 0.5% to 5.9% to lipids. The fatty acids synthesized in the shoot and haustorium were mainly C16, C18, and C18:1 fatty acids but in the kernel about 18% to 32% of the 14C-fatty acids were C12 fatty acids.

[14C]Lauric acid was absorbed and metabolized by haustorium slices and by the haustorium in intact seedlings; it was partly esterified to triacylglycerols and also converted to water-soluble substances and insoluble tissue material. In contrast, tri-[14C]laurin was absorbed but not metabolized. The haustorium also absorbed other fatty acids but the longer chain (C16 and C18) fatty acids were not esterified or metabolized further. Preincubation of the haustorium with plant hormones or in the presence of kernel tissue did not alter its inactivity towards tri-[14C]laurin.

When tri-[14C]laurin or [14C]lauric acid were injected into the seed or the shoot, there was no movement or radioactivity to other parts of the seedling. When injected into the shoot, but not into the seed, tri-[14C] laurin was hydrolyzed and partly metabolized to water-soluble substances.

Selenium is an important and essential trace element to living systems. In the article, two methods of instrumental neutron activation analysis and hydride generation-atomic fluorescence spectrometry were applied to determine Se in biological samples and the accuracy was evaluated by several reference materials. The subcellular distribution of selenium in human liver samples, which were obtained from normal subjects who had an accidental death, was investigated by differential centrifugation combined with INAA. Selenium was mainly enriched in mitochondria, nuclei, and cytosol. Almost half of the total Se content existed in nuclei as a result of the large amount in liver and the high Se concentration. Generally, the highest Se concentration in the mitochondrial fractions of each liver sample suggested that Se had important functions in this liver component.  相似文献

Purpose

Construction and demolition waste (C&DW) is the largest waste stream in the European Union (EU) and all over the world. Proper management of C&DW and recycled materials—including the correct handling of hazardous waste—can have major benefits in terms of sustainability and the quality of life. The Waste Framework Directive 2008/98/EC aims to have 70% of C&DW recycled by 2020. However, except for a few EU countries, only about 50% of C&DW is currently being recycled. In the present research, the environmental impact of concrete with recycled aggregates and with geopolymer mixtures is analysed. The aim of the present research is to propose a comparative LCA of concrete with recycled aggregates in the context of European politics.

Methods

Life cycle assessment (LCA) methodology is applied using Simapro© software. A cradle to grave analysis is carried out. The results are analysed based on the database Ecoinvent 3.3 and Impact 2002+.

Results

Results show that the concrete with 25% recycled aggregates is the best solution from an environmental point of view. Furthermore, geopolymer mixtures could be a valid alternative to reduce the phenomenon of “global warming”; however, the production of sodium silicate and sodium hydroxide has a great environmental impact.

Conclusions

A possible future implementation of the present study is certainly to carry out an overall assessment and to determine the most cost-effective option among the different competing alternatives through the life cycle cost analysis.

Uredospores of Puccinia graminis (Pers.) tritici (Eriks. and Henn.) were uniformly labeled with 14C by permitting the host (Triticum aestivum L.) to carry out photosynthesis in 14CO2 during the process of spore production by the obligate parasite. The use of 14C labeled spores provided advantages in a study of the utilization of endogenous substrates at frequent intervals with small amounts of spores under conditions conducive to germination.

Because of previous uncertainties about the nature of the substrates of importance to germination, a detailed study of carbohydrate and lipid components, both in the spores and in the germination medium, was made during the first 7 hours after placing the spores on aqueous media. Diethyl ether and 80% ethanol soluble metabolites each constituted approximately 20% of the total spore carbon. During the first hour nearly 60% of the 80% alcohol solubles disappeared from the spores while the total ether soluble material did not change appreciably. A significant part of the 80% ethanol soluble materials appeared in the germination medium.

During germination and germ tube extension, there was rapid utilization of trehalose, arabitol and mannitol even though appreciable amounts of these materials were present as exogenous pools in the germination medium. Although the total amounts of ether soluble components did not change as drastically as the carbohydrate fraction, there was extensive utilization of palmitic, oleic, linolenic and 9,10-epoxyoctadecanoic acids.

The results indicate that the germination process in spores of obligate parasites is not based solely on the utilization of lipids and some possible roles of the changes in internal and external pools of soluble carbohydrates are discussed.

Purpose

The purpose of this article is to find a suitable life cycle assessment (LCA) method to quantify the most important environmental burdens caused by construction processes of torrent control structures. To find these environmental burdens, 17 construction projects of the “Austrian Service for Torrent and Avalanche Control” (WLV) were analyzed using the “cradle to gate with options” LCA methodology (CEN, 2013).

Methods

This article explains an LCA methodology for the product stage and the construction process of torrent control structures following existing standards. The iterative approach of LCA methodology (ISO, 2006a) was used to record all important processes of the system and to supplement missing information. The LCA methodology has been developed from existing standards of the construction and product sector. Since the production of some construction materials takes place locally, the generic data, for Austria, was adapted. Wood inherent biogenic carbon and primary energy, used as raw material, are treated as materials inherent properties (CEN, 2014). The contribution of the various processes was reproduced by hotspot.

Results and discussion

Hotspots of the different stages are related to the construction materials used. The emissions and primary energy inputs in the product stage are clearly dominated by concrete and steel. If these two materials are used sparingly, the focus is on machine application and transportation. Depending on the selected scenarios, the smallest share of emissions, in relation to the total result of product and construction stage emitted by transport, is 3% and the maximum share is 69%. The greatest environmental impacts in the construction stage are caused by excavation work and transportation on-site. With an average of 4% in the construction stage, the transport of workers to the construction site cannot be neglected as is done in the building sector.

Conclusions

The conclusion of this study is that existing LCA models can be adapted to protective structures. In contrast to conventional buildings, the construction process and transportation are much more important and cannot be neglected. Shifting the hotspots to these processes requires specific calculation rules for that particular field. There is still a need for research to find a suitable functional unit and to develop a methodology for the use and end of life stage of these structures.

A working analytical procedure for selenium determinations by INAA, using γ spectrometry with a germanium well-type detector, is presented. The spectral line at 400.7 keV is employed, enhanced with the effect of energy summing in true coincidence of cascade photons. The main advantage of the method is high efficiency and reliable, interference-free results; potential drawbacks are increases in input count rate and pile-up losses, decrease in resolution, and sensitivity to errors in sample-to-detector geometry. The procedure is applied to certification analysis of reference materials, large-area biomonitoring by oak leave samples, and determination of separated proteins.  相似文献

Visualizing regions of conserved synteny between two genomes is supported by numerous software applications. However, none of the current applications allow researchers to select genome features to display or highlight in blocks of synteny based on the annotated biological properties of the features (e.g., type, function, and/or phenotype association). To address this usability gap, we developed an interactive web-based conserved synteny browser, The Jackson Laboratory (JAX) Synteny Browser. The browser allows researchers to highlight or selectively display genome features in the reference and/or the comparison genome according to the biological attributes of the features. Although the current implementation for the browser is limited to the reference genomes for the laboratory mouse and human, the software platform is intentionally genome agnostic. The JAX Synteny Browser software can be deployed for any two genomes where genome coordinates for syntenic blocks are defined and for which biological attributes of the features in one or both genomes are available in widely used standard bioinformatics file formats. The JAX Synteny Browser is available at: http://syntenybrowser.jax.org/. Photosynthetically active chloroplasts retaining high rates of fatty acid synthesis from [1-14C]acetate were purified from leaves of both 16:3 (Solanum nodiflorum, Chenopodium album) and 18:3 plants (Amaranthus lividus, Pisum sativum). A comparison of lipids into which newly synthesized fatty acids were incorporated revealed that, in 18:3 chloroplasts, enzymic activities catalyzing the conversion of phosphatidate to diacylglycerol and of diacylglycerol to monogalactosyl diacylglycerol (MGD) were significantly less active than in 16:3 chloroplasts. In contrast, labeling rates of MGD from UDP-[14C]gal were similar for both types of chloroplasts.

The composition and positional distribution of labeled fatty acids within the glycerides synthesized by isolated 16:3 and 18:3 chloroplasts were similar and in each case only a C18/C16 diacylglycerol backbone was synthesized. In nodiflorum chloroplasts, C18:1/C16:0 MGD assembled de novo was completely desaturated to the C18:3/C16:3 stage.

Whereas newly synthesized C18/C18 MGD could not be detected in any of these chloroplasts if incubated with [14C]acetate after isolation, chloroplasts isolated from acetate-labeled leaves contained MGD with labeled C18 fatty acids at both sn-1 and sn-2 positions. Taken together, these results provide further evidence on an organellar level for the operation of pro- and eucaryotic pathways in the biosynthesis of MGD in different groups of plants.

Abstract

Endogenous Rhythms in Mimosa pudica L. Leaf Movements.

The rhythmic movements performed by the leaves of the “Sensitive plant”, Mimosa pudica L., observed by time lapse photography, result of periodical turgor variations taking place in the parenchymatous cells of specialized motor organs. These turgor variations are associated with membrane permeability changes and ionic movements. These leaf movements allow to specify the temporal organization of this plant. Statistical analysis of observed periodicities in leaf movement shows that, in alternating conditions of light and dark (L/D:14/10) three distinct rhythms exist: a circadian rhythm synchronized by the photoperiodic cycle (τ = 24 hrs), and two ultradian rhythms with mean period values 3.8 hrs and 0.5 hrs respectively. In constant conditions from germination (L/L), the leaf behavior is strongly modified, but the three period values are found again (mean period values of 25.1 hrs, 3.5 hrs and 0.6 hrs respectively). The occurence of many rhythms with various periods taking place in the same organ is discussed in reference to observations effected on other biological subjects. Then, it appears that the period value within 2 and 4 hrs may be considered as a characteristic one in plants.  相似文献
