铁皮石斛茎段快繁技术的研究   总被引:1,自引:0,他引:1  
通过组织培养建立铁皮石斛快繁体系,为更好利用开发铁皮石斛提供原料。以铁皮石斛茎段为外植体,研究不同种类和浓度配比的激素和天然添加物对组培过程中不定芽的诱导和丛生芽的增殖的影响,筛选出各阶段最适培养基。结果表明,不定芽诱导最适激素配比为NAA 1.0 mg/L+BA 2.0 mg/L,丛生芽增殖最适培养基为NAA 0.5 mg/L+BA 1.0 mg/L+香蕉100 g/L+活性炭100 g/L。以茎段为途径建立铁皮石斛快繁体系高效稳定的获得组培苗是可行的。  相似文献

以玉竹[Polygonatum odoratum (Mill.) Druce]根状茎、叶片和茎段为外植体,于附加不同激素配比的MS培养基中诱导愈伤组织、不定芽和不定根,探讨增殖培养和植株再生的条件.结果表明,叶片和茎段外植体诱导愈伤组织和芽的分化率很低;而根状茎外植体易于培养,有较高的诱导率和增殖倍数,其愈伤组织、不定芽和不定根的诱导率分别可达87%、90%和99%以上.适宜根状茎外植体愈伤组织诱导的培养基为MS+1.0 mg/L 6-BA+0.5 mg/L NAA,有利于增殖和丛生芽分化的培养基为MS+2.0 mg/L 6-BA+0.5 mg/L IBA和MS+3.0 mg/L 6-BA+0.1 mg/L NAA,而1/2MS+3.0~5.0 mg/L NAA适宜诱导试管苗生根培养.试管苗的移栽成活率可达85%以上.  相似文献

本试验以芦笋新品种"津宁"为材料,探讨植物生长调节剂对不同外植体产生丛生芽的效应。试验结果表明,NAA、6-BA和KT三者配合使用有利于丛生芽的增殖:以茎尖为增殖外植体时,培养基用MS+NAA0.1 mg/L+6-BA0.2~0.3 mg/L+KT 0.1 mg/L,30 d时平均每瓶可以产生20.5个丛生芽;以茎中段为外植体时,培养基用MS+NAA 0.1 mg/L+6-BA 0.3~0.4 mg/L+KT 0.1 mg/L,30 d时平均每瓶可产生33.5个丛生芽;以茎基为外植体时,培养基用MS+NAA 0.1 mg/L+6-BA 0.3~0.5 mg/L+KT 0.1 mg/L,30 d时平均每瓶可以产生43.1个丛生芽。进行试管苗工厂化生产时,增殖外植体以茎基为最佳。  相似文献

本研究以滇黄芩不带腋芽茎段为外植体,采用不同配比激素的MS培养基建立了滇黄芩器官发生,植株再生体系.结果表明,茎段在MS+6-BA 2 mg/L+NAA 0.5 mg/L培养基上培养时,愈伤组织诱导率达到100%:以相同培养基进行分化,并在MS+6-BA 2 mg/L+NAA 0.2 mg/L培养基上扩繁丛生芽.在1/2MS+IBA 0.2 mg/L培养基诱导生根,扦插继代.利用RAPD分析不同继代次数再生植株,所用的39条随机引物中只有1条引物带型发生变化.说明经组织培养获得的再生植株遗传稳定,可多次继代培养,为滇黄芩进一步遗传操作和扩大药材资源奠定基础.  相似文献

本试验采用正交设计,探讨春石斛组培以丛生芽途径进行快速繁殖的方法。研究主要集中在不同基本培养基类型、不同生长调节剂配比及不同添加物等因素对丛生芽增殖的影响,从中筛选最优技术参数组合,提高丛生芽增殖系数,建立春石斛最优再生体系。结果表明:影响丛生芽增殖的显著因子分别是基本培养基、6-BA、KT;最适宜的培养基配方是1/2 MS + 6-BA 1.0 mg/L + NAA 1.0 mg/L + KT 0.5 mg/L +蔗糖30.0 g/L +琼脂7.0 g/L +椰汁150.0 ml/L,pH 5.4。此外,春石斛丛生芽增殖的外植体以1.5 cm带节茎段、接入密度每瓶3株较适宜。  相似文献

以金线莲茎段、叶片、茎片、不定芽为试材,分别在添加6-BA、ZT、NAA、KT 5个不同处理的MS及1/2MS培养基上培养,诱导愈伤组织。结果表明,以茎段、茎片、不定芽为外植体,在MS + 6-BA 2.0 mg/L + NAA0.5 mg/L、MS + NAA 2.0 mg/L + KT 0.1 mg/L和MS + 6-BA 2.0 mg/L + NAA 0.5 mg/L + ZT 0.2 mg/L培养基中培养,均能成功诱导愈伤组织。不定芽为诱导愈伤组织最佳外植体,最佳培养基为MS + 6-BA 2.0 mg/L + NAA 0.5 mg/L + KT 0.2 mg/L。  相似文献

香蕉间接器官发生再生体系是一种较好的突变育种和基因转化受体再生系统。本研究以巴西蕉低代试管苗茎段薄层切片、中代试管苗茎段薄层切片、吸芽薄层切片、雄花薄层切片为外植体,对巴西蕉间接器官发生再生体系进行了系统研究。研究结果表明,以MS为基本培养基,巴西蕉低代试管苗茎段薄层切片在0.5~1.0mg/L2,4-D+4~6mg/L6-BA激素条件下;雄花薄层切片在0.5~1.0mg/L2,4-D+0.2~0.5mg/LZT激素条件下能通过花菜体间接器官发生途径实现高效再生;同时,花菜体薄层切片在1.0mg/L2,4-D+5mg/L6-BA激素条件下,再次通过花菜体途径实现循环高效再生。本研究为建立香蕉高效间接器官发生再生体系和相关育种技术奠定了基础。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
