Toxin- (T)from the Brazilian scorpion Tityusserrulatus venom caused a concentration- andtime-dependent increase in the release of norepinephrine andepinephrine from bovine adrenal medullary chromaffin cells. T was~200-fold more potent than veratridine judged fromEC50 values, although the maximalsecretory efficacy of veratridine was 10-fold greater than that of T(1.2 vs. 12 µg/ml of catecholamine release). The combination of both toxins produced a synergistic effect that was particularly drastic at 5 mM extracellular Ca2+concentration([Ca2+]o),when 30 µM veratridine plus 0.45 µM T were used. T (0.45 µM) doubled the basal uptake of45Ca2+,whereas veratridine (100 µM) tripled it. Again, a drastic synergism in enhancing Ca2+ entry was seenwhen T and veratridine were combined; this was particularlypronounced at 5 mM[Ca2+]o.Veratridine induced oscillations of cytosolicCa2+ concentration([Ca2+]i)in single fura 2-loaded cells without elevation of basal levels. Incontrast, T elevated basal[Ca2+]ilevels, causing only small oscillations. When added together, T andveratridine elevated the basal levels of[Ca2+]iwithout causing large oscillations. T shifted the current-voltage (I-V) curve forNa+ channel current to the left.The combination of T with veratridine increased the shift of theI-V curve to the left, resulting in agreater recruitment of Na+channels at more hyperpolarizing potentials. This led to enhanced andmore rapid accumulation of Na+ inthe cell, causing cell depolarization, the opening of voltage-dependent Ca2+ channels, andCa2+ entry and secretion.

The role of the Na+ pump2-subunit in Ca2+ signaling was examined inprimary cultured astrocytes from wild-type(2+/+ = WT) mouse fetuses and thosewith a null mutation in one [2+/ = heterozygote (Het)] or both [2/ = knockout (KO)] 2 genes. Na+ pump catalytic() subunit expression was measured by immunoblot; cytosol[Na+] ([Na+]cyt) and[Ca2+] ([Ca2+]cyt) weremeasured with sodium-binding benzofuran isophthalate and fura 2 byusing digital imaging. Astrocytes express Na+ pumpswith both 1- (80% of total ) and2- (20% of total ) subunits. Het astrocytesexpress 50% of normal 2; those from KO express none.Expression of 1 is normal in both Het and KO cells.Resting [Na+]cyt = 6.5 mM in WT, 6.8 mMin Het (P > 0.05 vs. WT), and 8.0 mM in KO cells(P < 0.001); 500 nM ouabain (inhibits only2) equalized [Na+]cyt at 8 mMin all three cell types. Resting[Ca2+]cyt = 132 nM in WT, 162 nM in Het,and 196 nM in KO cells (both P < 0.001 vs. WT).Cyclopiazonic acid (CPA), which inhibits endoplasmic reticulum (ER)Ca2+ pumps and unloads the ER, induces transient (inCa2+-free media) or sustained (in Ca2+-repletemedia) elevation of [Ca2+]cyt. TheseCa2+ responses to 10 µM CPA were augmented in Het as wellas KO cells. When CPA was applied in Ca2+-free media, thereintroduction of Ca2+ induced significantly largertransient rises in [Ca2+]cyt (due toCa2+ entry through store-operated channels) in Het and KOcells than in WT cells. These results correlate with published evidencethat 2 Na+ pumps andNa+/Ca2+ exchangers are confined to plasmamembrane microdomains that overlie the ER. The data suggest thatselective reduction of 2 Na+ pump activitycan elevate local [Na+] and, viaNa+/Ca2+ exchange, [Ca2+] in thetiny volume of cytosol between the plasma membrane and ER. This, inturn, augments adjacent ER Ca2+ stores and therebyamplifies Ca2+ signaling without elevating bulk[Na+]cyt.

Na+-K+-Cl cotransporter isoform 1 (NKCC1) and reverse mode operation of the Na+/Ca2+ exchanger (NCX) contribute to intracellular Na+ and Ca2+ overload in astrocytes following oxygen-glucose deprivation (OGD) and reoxygenation (REOX). Here, we further investigated whether NKCC1 and NCX play a role in mitochondrial Ca2+ (Cam2+) overload and dysfunction. OGD/REOX caused a doubling of mitochondrial-releasable Ca2+ (P < 0.05). When NKCC1 was inhibited with bumetanide, the mitochondrial-releasable Ca2+ was reduced by 42% (P < 0.05). Genetic ablation of NKCC1 also reduced Cam2+ accumulation. Moreover, OGD/REOX in NKCC1+/+ astrocytes caused dissipation of the mitochondrial membrane potential (m) to 42 ± 3% of controls. In contrast, when NKCC1 was inhibited with bumetanide, depolarization of m was attenuated significantly (66 ± 10% of controls, P < 0.05). Cells were also subjected to severe in vitro hypoxia by superfusion with a hypoxic, acidic, ion-shifted Ringer buffer (HAIR). HAIR/REOX triggered a secondary, sustained rise in intracellular Ca2+ that was attenuated by reversal NCX inhibitor KB-R7943. The hypoxia-mediated increase in Cam2+ was accompanied by loss of m and cytochrome c release in NKCC1+/+ astrocytes. Bumetanide or genetic ablation of NKCC1 attenuated mitochondrial dysfunction and astrocyte death following ischemia. To study the effects of -opioid receptor stimulation onintracellular Ca2+ concentration([Ca2+]i)homeostasis during extracellular acidosis, we determined the effects of-opioid receptor stimulation on[Ca2+]iresponses during extracellular acidosis in isolated single ratventricular myocytes, by a spectrofluorometric method. U-50488H (10-30 µM), a selective -opioid receptor agonist, dosedependently decreased the electrically induced[Ca2+]itransient, which results from the influx ofCa2+ and the subsequentmobilization of Ca2+ from thesarcoplasmic reticulum (SR). U-50488H (30 µM) also increased theresting[Ca2+]iand inhibited the[Ca2+]itransient induced by caffeine, which mobilizesCa2+ from the SR, indicating thatthe effects of the -opioid receptor agonist involved mobilization ofCa2+ from its intracellular poolinto the cytoplasm. The Ca2+responses to 30 µM U-50488H were abolished by 5 µMnor-binaltorphimine, a selective -opioid receptorantagonist, indicating that the event was mediated by the -opioidreceptor. The effects of the agonist on[Ca2+]iand the electrically induced[Ca2+]itransient were significantly attenuated when the extracellular pH(pHe) was loweredto 6.8, which itself reduced intracellular pH(pHi) and increased[Ca2+]i.The inhibitory effects of U-50488H were restored during extracellular acidosis in the presence of 10 µM ethylisopropyl amiloride, a potentNa+/H+exchange blocker, or 0.2 mM Ni2+,a putativeNa+/Ca2+exchange blocker. The observations indicate that acidosismay antagonize the effects of -opioid receptor stimulation viaNa+/H+andNa+/Ca2+exchanges. When glucose at 50 mM, known to activate theNa+/H+exchange, was added, both the resting[Ca2+]iand pHi increased. Interestingly,the effects of U-50488H on [Ca2+]iand the electrically induced[Ca2+]itransient during superfusion with glucose were significantly attenuated; this mimicked the responses during extracellular acidosis. When a high-Ca2+ (3 mM) solutionwas superfused, the resting[Ca2+]iincreased; the increase was abolished by 0.2 mMNi2+, but thepHi remained unchanged. Like theresponses to superfusion with high-concentration glucose andextracellular acidosis, the responses of the[Ca2+]iand electrically induced[Ca2+]itransients to 30 µM U-50488H were also significantly attenuated. Results from the present study demonstrated for the first time thatextracellular acidosis antagonizes the effects of -opioid receptorstimulation on the mobilization ofCa2+ from SR. Activation of bothNa+/H+andNa+/Ca2+exchanges, leading to an elevation of[Ca2+]i,may be responsible for the antagonistic action of extracellular acidosis against -opioid receptor stimulation.

The Na+/K+-ATPase (NKA) is the main route for Na+ extrusion from cardiac myocytes. Different NKA -subunit isoforms are present in the heart. NKA-1 is predominant, although there is a variable amount of NKA-2 in adult ventricular myocytes of most species. It has been proposed that NKA-2 is localized mainly in T-tubules (TT), where it could regulate local Na+/Ca2+ exchange and thus cardiac myocyte Ca2+. However, there is controversy as to where NKA-1 vs. NKA-2 are localized in ventricular myocytes. Here, we assess the TT vs. external sarcolemma (ESL) distribution functionally using formamide-induced detubulation of rat ventricular myocytes, NKA current (IPump) measurements and the different ouabain sensitivity of NKA-1 (low) and NKA-2 (high) in rat heart. Ouabain-dependent IPump inhibition in control myocytes indicates a high-affinity NKA isoform (NKA-2, K1/2 = 0.38 ± 0.16 µM) that accounts for 29.5 ± 1.3% of IPump and a low-affinity isoform (NKA-1, K1/2 = 141 ± 17 µM) that accounts for 70.5% of IPump. Detubulation decreased cell capacitance from 164 ± 6 to 120 ± 8 pF and reduced IPump density from 1.24 ± 0.05 to 1.02 ± 0.05 pA/pF, indicating that the functional density of NKA is significantly higher in TT vs. ESL. In detubulated myocytes, NKA-2 accounted for only 18.2 ± 1.1% of IPump. Thus, 63% of IPump generated by NKA-2 is from the TT (although TT are only 27% of the total sarcolemma), and the NKA-2/NKA-1 ratio in TT is significantly higher than in the ESL. The functional density of NKA-2 is 4.5 times higher in the T-tubules vs. ESL, whereas NKA-1 is almost uniformly distributed between the TT and ESL. We employed a glycogen-depleting session of exercise followed by a low-carbohydrate (CHO) diet to investigate modifications that occur in muscle sarcoplasmic reticulum (SR) Ca2+-cycling properties compared with low-CHO diet alone. SR properties were assessed in nine untrained males [peak aerobic power (O2 peak) = 43.6 ± 2.6 (SE) ml·kg–1·min–1] during prolonged cycle exercise to fatigue performed at 58% O2 peak after 4 days of low-CHO diet (Lo CHO) and after glycogen-depleting exercise plus 4 days of low-CHO (Ex+Lo CHO). Compared with Lo CHO, Ex+Lo CHO resulted in 12% lower (P < 0.05) resting maximal Ca2+-ATPase activity (Vmax = 174 ± 12 vs. 153 ± 10 µmol·g protein–1·min–1) and smaller reduction in Vmax induced during exercise. A similar effect was observed for Ca2+ uptake. The Hill coefficient, defined as slope of the relationship between cytosolic free Ca2+ concentration and Ca2+-ATPase activity, was higher (P < 0.05) at rest (2.07 ± 0.15 vs. 1.90 ± 0.10) with Ex+Lo CHO, an effect that persisted throughout the exercise. The coupling ratio, defined as the ratio of Ca2+ uptake to Vmax, was 23–30% elevated (P < 0.05) at rest and during the first 60 min of exercise with Ex+Lo CHO. The 27 and 34% reductions (P < 0.05) in phase 1 and phase 2 Ca2+ release, respectively, observed during exercise with Lo CHO were not altered by Ex+Lo CHO. The role of Na+/Ca2+ exchange inregulating intracellular Ca2+ concentration([Ca2+]i) in isolated smooth muscle cellsfrom the guinea pig urinary bladder was investigated. Incrementalreduction of extracellular Na+ concentration resulted in agraded rise of [Ca2+]i; 50-100 µMstrophanthidin also increased [Ca2+]i. Asmall outward current accompanied the rise of[Ca2+]i in low-Na+ solutions(17.1 ± 1.8 pA in 29.4 mM Na+). The quantity ofCa2+ influx through the exchanger was estimated from thecharge carried by the outward current and was ~30 times that which isnecessary to account for the rise of [Ca2+]i,after correction was made for intracellular Ca2+ buffering.Ca2+ influx through the exchanger was able to loadintracellular Ca2+ stores. It is concluded that the levelof resting [Ca2+]i is not determined by theexchanger, and under resting conditions (membrane potential 50 to60 mV), there is little net flux through the exchanger. However, asmall rise of intracellular Na+ concentration would besufficient to generate significant net Ca2+ influx.

The effects of serotonin[5-hydroxytryptamine (5-HT)] on the transepithelial electricalproperties of the short-circuited rabbit conjunctiva were examined.With this epithelium, the short-circuit current(Isc) measures Cl secretion plusan amiloride-resistant Na+ absorptive process. Apicaladdition of 5-HT (10 µM) elicited a prompt Iscreduction from 14.2 ± 1.2 to 10.9 ± 1.2 µA/cm2 and increased transepithelial resistance from0.89 ± 0.05 to 1.03 ± 0.06 k · cm2(means ± SE, n = 21, P < 0.05).Similar changes were obtained with conjunctivae bathed withoutNa+ in the apical bath, as well as with conjunctivaepreexposed to bumetanide with the Cl-dependentIsc sustained by the parallel activities ofbasolateral Na+/H+ andCl/HCO exchangers. In contrast, the5-HT-evoked effects were attenuated by the absence of Cl(Isc = 0.5 ± 0.2, n = 5), suggesting that reduced Clconductance(s) is an effect of 5-HT exposure. In amphotericin B-treatedconjunctiva and in the presence of a transepithelial K+gradient, 5-HT addition reduced K+ diffusion across thepreparation by 13% and increased transepithelial resistance by 4%(n = 6, P < 0.05), indicating that aninhibition in K+ conductance(s) was also detectable.Significant electrical responses also occurred under physiologicalconditions when 5-HT was introduced to epithelia pretreated withadrenergic agonists or protein kinase C, phospholipase C,phosphodiesterase, or adenylyl cyclase inhibitors or after perturbationof Ca2+ homeostasis. Briefly, the conjunctiva harbors theonly known Cl-secreting epithelium in which 5-HT evokesCl transport inhibition; receptor subtype and signaltransduction mechanism were not determined.

Role of caveolae in signal-transducing function of cardiac Na+/K+-ATPase   总被引:2,自引:0,他引:2  
Ouabain binding toNa+/K+-ATPase activates Src/epidermal growthfactor receptor (EGFR) to initiate multiple signal pathways thatregulate growth. In cardiac myocytes and the intact heart, the earlyouabain-induced pathways that cause rapid activations of ERK1/2 alsoregulate intracellular Ca2+ concentration([Ca2+]i) and contractility. The goal of thisstudy was to explore the role of caveolae in these early signalingevents. Subunits of Na+/K+-ATPase were detectedby immunoblot analysis in caveolae isolated from cardiac myocytes,cardiac ventricles, kidney cell lines, and kidney outer medulla byestablished detergent-free procedures. Isolated rat cardiac caveolaecontained Src, EGFR, ERK1/2, and 20-30% of cellular contents of1- and 2-isoforms ofNa+/K+-ATPase, along with nearly all ofcellular caveolin-3. Immunofluorescence microscopy of adult cardiacmyocytes showed the presence of caveolin-3 and -isoforms inperipheral sarcolemma and T tubules and suggested their partialcolocalization. Exposure of contracting isolated rat hearts to apositive inotropic dose of ouabain and analysis of isolated cardiaccaveolae showed that ouabain caused 1) no change in totalcaveolar ERK1/2, but a two- to threefold increase in caveolarphosphorylated/activated ERK1/2; 2) no change in caveolar 1-isoform and caveolin-3; and 3) 50-60%increases in caveolar Src and 2-isoform. These findings,in conjunction with previous observations, show that components of thepathways that link Na+/K+-ATPase to ERK1/2 and[Ca2+]i are organized within cardiac caveolaemicrodomains. They also suggest that ouabain-induced recruitments ofSrc and 2-isoform to caveolae are involved in themanifestation of the positive inotropic effect of ouabain.

Previous studies have shown that inhibition of L-type Ca2+ current (ICa) by cytosolic free Mg2+ concentration ([Mg2+]i) is profoundly affected by activation of cAMP-dependent protein kinase pathways. To investigate the mechanism underlying this counterregulation of ICa, rat cardiac myocytes and tsA201 cells expressing L-type Ca2+ channels were whole cell voltage-clamped with patch pipettes in which [Mg2+] ([Mg2+]p) was buffered by citrate and ATP. In tsA201 cells expressing wild-type Ca2+ channels (1C/2A/2), increasing [Mg2+]p from 0.2 mM to 1.8 mM decreased peak ICa by 76 ± 4.5% (n = 7). Mg2+-dependent modulation of ICa was also observed in cells loaded with ATP--S. With 0.2 mM [Mg2+]p, manipulating phosphorylation conditions by pipette application of protein kinase A (PKA) or phosphatase 2A (PP2A) produced large changes in ICa amplitude; however, with 1.8 mM [Mg2+]p, these same manipulations had no significant effect on ICa. With mutant channels lacking principal PKA phosphorylation sites (1C/S1928A/2A/S478A/S479A/2), increasing [Mg2+]p had only small effects on ICa. However, when channel open probability was increased by 1C-subunit truncation (1C1905/2A/S478A/S479A/2), increasing [Mg2+]p greatly reduced peak ICa. Correspondingly, in myocytes voltage-clamped with pipette PP2A to minimize channel phosphorylation, increasing [Mg2+]p produced a much larger reduction in ICa when channel opening was promoted with BAY K8644. These data suggest that, around its physiological concentration range, cytosolic Mg2+ modulates the extent to which channel phosphorylation regulates ICa. We reported previously that inhibition ofNa+-K+-Cl cotransporter isoform 1 (NKCC1) by bumetanide abolishes high extracellular K+concentration ([K+]o)-induced swelling andintracellular Cl accumulation in rat cortical astrocytes.In this report, we extended our study by using cortical astrocytes fromNKCC1-deficient (NKCC1/) mice. NKCC1 protein andactivity were absent in NKCC1/ astrocytes.[K+]o of 75 mM increased NKCC1 activityapproximately fourfold in NKCC1+/+ cells (P < 0.05) but had no effect in NKCC1/ astrocytes.Intracellular Cl was increased by 70% inNKCC1+/+ astrocytes under 75 mM[K+]o (P < 0.05) butremained unchanged in NKCC1/ astrocytes. Baselineintracellular Na+ concentration([Na+]i) in NKCC1+/+ astrocyteswas 19.0 ± 0.5 mM, compared with 16.9 ± 0.3 mM[Na+]i in NKCC1/ astrocytes(P < 0.05). Relative cell volume ofNKCC1+/+ astrocytes increased by 13 ± 2% in 75 mM[K+]o, compared with a value of 1.0 ± 0.5% in NKCC1/ astrocytes (P < 0.05).Regulatory volume increase after hypertonic shrinkage was completelyimpaired in NKCC1/ astrocytes.High-[K+]o-induced 14C-labeledD-aspartate release was reduced by ~30% inNKCC1/ astrocytes. Our study suggests that stimulationof NKCC1 is required for high-[K+]o-inducedswelling, which contributes to glutamate release from astrocytes underhigh [K+]o.

These experiments were performed to determine the effects ofreducing Ca2+ influx(Cain) onK+ currents(IK) inmyocytes from rat small mesenteric arteries by1) adding externalCd2+ or2) lowering externalCa2+ to 0.2 mM. When measured froma holding potential (HP) of 20 mV(IK20),decreasing Cain decreasedIK at voltageswhere it was active (>0 mV). When measured from a HP of 60 mV(IK60),decreasing Cain increasedIK at voltagesbetween 30 and +20 mV but decreased IK at voltagesabove +40 mV. Difference currents(IK) weredetermined by digital subtraction of currents recorded under controlconditions from those obtained whenCain was decreased. At testvoltages up to 0 mV,IK60 exhibitedkinetics similar to controlIK60, with rapidactivation to a peak followed by slow inactivation. At 0 mV, peakIK60 averaged75 ± 13 pA (n = 8) withCd2+ and 120 ± 20 pA(n = 9) with lowCa2+ concentration. At testvoltages from 0 to +60 mV,IK60 always had an early positive peak phase, but its apparent "inactivation" increased with voltage and its steady value became negative above +20mV. At +60 mV, the initial peakIK60 averaged115 ± 18 pA with Cd2+ and 187 ± 34 pA with low Ca2+. With 10 mM pipette BAPTA, Cd2+ produced asmall inhibition ofIK20 but stillincreased IK60 between 30 and +10 mV. InCa2+-free external solution,Cd2+ only decreased bothIK20 andIK60. In thepresence of iberiotoxin (100 nM) to inhibitCa2+-activatedK+ channels(KCa),Cd2+ increasedIK60 at allvoltages positive to 30 mV while BAY K 8644 (1 µM) decreasedIK60. Theseresults suggest that Cain, through L-type Ca2+ channels and perhapsother pathways, increases KCa(i.e., IK20) and decreases voltage-dependent K+currents in this tissue. This effect could contribute to membrane depolarization and force maintenance.

G protein-coupled receptors (GPCRs) control neuronal functions via ion channel modulation. For voltage-gated ion channels, gating charge movement precedes and underlies channel opening. Therefore, we sought to investigate the effects of G protein activation on gating charge movement. Nonlinear capacitive currents were recorded using the whole cell patch-clamp technique in cultured rat sympathetic neurons. Our results show that gating charge movement depends on voltage with average Boltzmann parameters: maximum charge per unit of linear capacitance (Qmax) = 6.1 ± 0.6 nC/µF, midpoint (Vh) = –29.2 ± 0.5 mV, and measure of steepness (k) = 8.4 ± 0.4 mV. Intracellular dialysis with GTPS produces a nonreversible 34% decrease in Qmax, a 10 mV shift in Vh, and a 63% increase in k with respect to the control. Norepinephrine induces a 7 mV shift in Vh and 40% increase in k. Overexpression of G protein 14 subunits produces a 13% decrease in Qmax, a 9 mV shift in Vh, and a 28% increase in k. We correlate charge movement modulation with the modulated behavior of voltage-gated channels. Concurrently, G protein activation by transmitters and GTPS also inhibit both Na+ and N-type Ca2+ channels. These results reveal an inhibition of gating charge movement by G protein activation that parallels the inhibition of both Na+ and N-type Ca2+ currents. We propose that gating charge movement decrement may precede or accompany some forms of GPCR-mediated channel current inhibition or downregulation. The lipid bilayertechnique was used to characterize theCa2+ dependence of a fastK+ channel formed by a synthetic17-amino acid segment [OaCNP-39-(1-17)] ofa 39-amino acid C-type natriuretic peptide (OaCNP-39) found in platypus (Ornithorhynchusanatinus) venom (OaV). TheOaCNP-39-(1-17)-formed K+ channel was reversiblydependent on1,2-bis(2-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid-buffered cis (cytoplasmic)Ca2+ concentration([Ca2+]cis).The channel was fully active when[Ca2+]ciswas >104 M andtrans (luminal)Ca2+ concentration was 1.0 mM, butnot at low[Ca2+]cis.The open probability of single channels increased from zero at1 × 106 McisCa2+ to 0.73 ± 0.17 (n = 22) at103 McisCa2+. Channel openings to themaximum conductance of 38 pS were rapidly and reversibly activated when[Ca2+]cis,but not transCa2+ concentration(n = 5), was increased to >5 × 104 M(n = 14). Channel openings to thesubmaximal conductance of 10.5 pS were dominant at5 × 104 MCa2+.K+ channels did not open whencisMg2+ orSr2+ concentrations were increasedfrom zero to 103 M or when[Ca2+]ciswas maintained at 106 M(n = 3 and 2). The Hill coefficientand the inhibition constant were 1 and 0.8 × 104 McisCa2+, respectively. Thisdependence of the channel on high[Ca2+]cissuggests that it may become active under1) physiological conditions whereCa2+ levels are high, e.g., duringcardiac and skeletal muscle contractions, and2) pathological conditions that leadto a Ca2+ overload, e.g., ischemicheart and muscle fatigue. The channel could modify a cascade ofphysiological functions that are dependent on theCa2+-activatedK+ channels, e.g., vasodilationand salt secretion.

To investigate theeffects of reactive oxygen species (ROS) on NHpermeation in Xenopus laevis oocytes, we used intracellulardouble-barreled microelectrodes to monitor the changes in membranepotential (Vm) and intracellular pH(pHi) induced by a 20 mM NH4Cl-containingsolution. Under control conditions, NH4Cl exposure induceda large membrane depolarization (to Vm = 4.0 ± 1.5 mV; n = 21) and intracellularacidification [reaching a change in pHi(pHi) of 0.59 ± 0.06 pH units in 12 min]; theinitial rate of cell acidification (dpHi/dt) was0.06 ± 0.01 pH units/min. Incubation of the oocytes in thepresence of H2O2 or -amyloid protein had nomarked effect on the NH4Cl-induced pHi. Bycontrast, in the presence of photoactivated rose bengal (RB),tert-butyl-hydroxyperoxide (t-BHP), orxanthine/xanthine oxidase (X/XO), the same experimental maneuverinduced significantly greater pHi anddpHi/dt. These increases in pHiand dpHi/dt were prevented by the ROS scavengershistidine and desferrioxamine, suggesting involvement of the reactivespecies 1gO2 and ·OH. Using thevoltage-clamp technique to identify the mechanism underlying theROS-measured effects, we found that RB induced a large increase in theoocyte membrane conductance (Gm). ThisRB-induced Gm increase was prevented by 1 mMdiphenylamine-2-carboxylate (DPC) and by a low Na+concentration in the bath. We conclude that RB, t-BHP, andX/XO enhance NH influx into the oocyte via activationof a DPC-sensitive nonselective cation conductance pathway.

We examined the effects of human cytomegalovirus (HCMV)infection on theNa+-K+-Clcotransporter (NKCC) in a human fibroblast cell line. Using the Cl-sensitive dye MQAE, weshowed that the mock-infected MRC-5 cells express a functional NKCC.1) IntracellularCl concentration([Cl]i)was significantly reduced from 53.4 ± 3.4 mM to 35.1 ± 3.6 mMfollowing bumetanide treatment. 2)Net Cl efflux caused byreplacement of external Clwith gluconate was bumetanide sensitive.3) InCl-depleted mock-infectedcells, the Cl reuptake rate(in HCO3-free media) was reduced inthe absence of external Na+ and bytreatment with bumetanide. After HCMV infection, we found that although[Cl]iincreased progressively [24 h postexposure (PE), 65.2 ± 4.5 mM; 72 h PE, 80.4 ± 5.0 mM], the bumetanide andNa+ sensitivities of[Cl]iand net Cl uptake and losswere reduced by 24 h PE and abolished by 72 h PE. Western blots usingthe NKCC-specific monoclonal antibody T4 showed an approximatelyninefold decrease in the amount of NKCC protein after 72 h ofinfection. Thus HCMV infection resulted in the abolition of NKCCfunction coincident with the severe reduction in the amount of NKCCprotein expressed.

To investigate the biology of the malegenital duct epithelium, we have established cell cultures from theovine vas deferens and epididymis epithelium. These cells develop tightjunctions, high transepithelial electrical resistance, and alumen-negative transepithelial potential difference as a sign of activetransepithelial ion transport. In epididymis cultures the equivalentshort-circuit current (Isc) averaged 20.8 ± 0.7 µA/cm2 (n = 150) and was partially inhibited byapical application of amiloride with an inhibitor concentration of 0.64 µM. In vas deferens cultures, Isc averaged 14.4 ± 1.1 µA/cm2 (n = 18) and was also inhibited byapical application of amiloride with a half-maximal inhibitorconcentration (Ki) of 0.68 µM. The remainingamiloride-insensitive Isc component in epididymisand vas deferens cells was partially inhibited by apical application ofthe Cl channel blocker diphenylamine-2-carboxylicacid (1 mM). It was largely dependent on extracellularCl and, to a lesser extent, on extracellularHCO3. It was further stimulated bybasolateral application of forskolin (105 M), which increasedIsc by 3.1 ± 0.3 µA/cm2 (n=65) in epididymis and 0.9 ± 0.1 µA/cm2 (n =11) in vas deferens. These findings suggest that cultured ovine vasdeferens and epididymis cells absorb Na+ viaamiloride-sensitive epithelial Na+ channels (ENaC) andsecrete Cl and HCO3via apical cystic fibrosis transmembrane conductance regulator (CFTR)Cl channels. This interpretation is supported byRT-PCR data showing that vas deferens and epididymis cells express CFTRand ENaC mRNA.

Patients treated with glucocorticoids have elevated skeletal muscle ouabain binding sites. The major Na+-K+-ATPase (NKA) isoform proteins found in muscle, 2 and 1, are increased by 50% in rats treated for 14 days with the synthetic glucocorticoid dexamethasone (DEX). This study addressed whether the DEX-induced increase in the muscle NKA pool leads to increased insulin-stimulated cellular K+ uptake that could precipitate hypokalemia. Rats were treated with DEX or vehicle via osmotic minipumps at one of two doses: 0.02 mg·kg–1·day–1 for 14 days (low DEX; n = 5 pairs) or 0.1 mg·kg–1·day–1 for 7 days (high DEX; n = 6 pairs). Insulin was infused at a rate of 5 mU·kg–1·min–1 over 2.5 h in conscious rats. Insulin-stimulated cellular K+ and glucose uptake rates were assessed in vivo by measuring the exogenous K+ infusion () and glucose infusion (Ginf) rates needed to maintain constant plasma K+ and glucose concentrations during insulin infusion. DEX at both doses decreased insulin-stimulated glucose uptake as previously reported. Ginf (in mmol·kg–1·h–1) was 10.2 ± 0.6 in vehicle-treated rats, 5.8 ± 0.8 in low-DEX-treated rats, and 5.2 ± 0.6 in high-DEX-treated rats. High DEX treatment also reduced insulin-stimulated K+ uptake. (in mmol·kg–1·h–1) was 0.53 ± 0.08 in vehicle-treated rats, 0.49 ± 0.14 in low-DEX-treated rats, and 0.27 ± 0.08 in high-DEX-treated rats. DEX treatment did not alter urinary K+ excretion. NKA 2-isoform levels in the low-DEX-treated group, measured by immunoblotting, were unchanged, but they increased by 38 ± 15% (soleus) and by 67 ± 3% (gastrocnemius) in the high-DEX treatment group. The NKA 1-isoform level was unchanged. These results provide novel evidence for the insulin resistance of K+ clearance during chronic DEX treatment. To examine theeffect of hyperosmolality on Na+/H+ exchanger(NHE) activity in mesangial cells (MCs), we used apH-sensitive dye,2',7'-bis(2-carboxyethyl)-5(6)-carboxyfluorescein-AM, to measure intracellular pH (pHi) in a single MC from ratglomeruli. All the experiments were performed inCO2/HCO3-free HEPESsolutions. Exposure of MCs to hyperosmotic HEPES solutions (500 mosmol/kgH2O) treated with mannitol caused cellalkalinization. The hyperosmolality-induced cell alkalinization wasinhibited by 100 µM ethylisopropylamiloride, a specific NHEinhibitor, and was dependent on extracellular Na+. Thehyperosmolality shifted the Na+-dependent acid extrusionrate vs. pHi by 0.15-0.3 pH units in thealkaline direction. Removal of extracellular Cl byreplacement with gluconate completely abolished the rate of cellalkalinization induced by hyperosmolality and inhibited the Na+-dependent acid extrusion rate, whereas, under isosmoticconditions, it caused no effect on Na+-dependentpHi recovery rate or Na+-dependent acidextrusion rate. The Cl-dependent cell alkalinizationrate under hyperosmotic conditions was partially inhibited bypretreatment with 5-nitro-2-(3-phenylpropylamino)benzoic acid, DIDS,and colchicine. We conclude: 1) in MCs, hyperosmolality activates NHE to cause cell alkalinization, 2) the acidextrusion rate via NHE is greater under hyperosmotic conditions thanunder isosmotic conditions at a wide range of pHi,3) the NHE activation under hyperosmotic conditions, but notunder isosmotic conditions, requires extracellularCl, and 4) theCl-dependent NHE activation under hyperosmoticconditions partly occurs via Cl channel andmicrotubule-dependent processes.

In the presentstudy, we examined the ability of adenosine 3',5'-cyclicmonophosphate (cAMP) to reduce elevated levels of cytosolicCa2+ concentration([Ca2+]i)in pancreatic -cells.[Ca2+]iand reduced pyridine nucleotide, NAD(P)H, were measured in rat single-cells by fura 2 and autofluorescence microfluorometry. Sustained[Ca2+]ielevation, induced by high KCl (25 mM) at a basal glucose concentration (2.8 mM), was substantially reduced by cAMP-increasing agents, dibutyryl cAMP (DBcAMP, 5 mM), an adenylyl cyclase activatorforskolin (10 µM), and an incretin glucagon-likepeptide-1-(7-36) amide (109 M), as well as byglucose (16.7 mM). The[Ca2+]i-reducingeffects of cAMP were greater at elevated glucose (8.3-16.7 mM)than at basal glucose (2.8 mM). An inhibitor of protein kinase A (PKA),H-89, counteracted[Ca2+]i-reducingeffects of cAMP but not those of glucose. Okadaic acid, a phosphataseinhibitor, at 10-100 nM also reduced sustained [Ca2+]ielevation in a concentration-dependent manner. Glucose, but not DBcAMP,increased NAD(P)H in -cells.[Ca2+]i-reducingeffects of cAMP were inhibited by 0.3 µM thapsigargin, an inhibitorof the endoplasmic reticulum (ER)Ca2+ pump. In contrast,[Ca2+]i-reducingeffects of cAMP were not altered by ryanodine, an ERCa2+-release inhibitor,Na+-free conditions, or diazoxide,an ATP-sensitive K+ channelopener. In conclusion, the cAMP-PKA pathway reduces[Ca2+]ielevation by sequestering Ca2+ inthapsigargin-sensitive stores. This process does not involve, but ispotentiated by, activation of -cell metabolism. Together with theknown[Ca2+]i-increasingaction of cAMP, our results reveal dual regulation of -cell[Ca2+]iby the cAMP-signaling pathway and by a physiological incretin.
