Pressure drop across chromatography beds employing soft or semirigid media can be a significant problem in the operation of large-scale preparative chromatography columns. The shape or aspect ratio (length/diameter) of a packed bed has a significant effect on column pressure drop due to wall effects, which can result in unexpectedly high pressures in manufacturing. Two types of agarose-based media were packed in chromatography columns at various column aspect ratios, during which pressure drop, bed height, and flow rate were carefully monitored. Compression of the packed beds with increasing flow velocities was observed. An empirical model was developed to correlate pressure drop with the aspect ratio of the packed beds and the superficial velocity. Modeling employed the Blake-Kozeny equation in which empirical relationships were used to predict bed porosity as a function of aspect ratio and flow velocity. Model predictions were in good agreement with observed pressure drops of industrial scale chromatography columns. A protocol was developed to predict compression in industrial chromatography applications by a few laboratory experiments. The protocol is shown to be useful in the development of chromatographic methods and sizing of preparative columns.  相似文献

Monoliths are considered as a novel generation of stationary phases. They were applied for capillary electrochromatography and liquid chromatography exploiting every action principle such as ion-exchange, affinity recognition, reversed-phase, and hydrophobic interaction. The fast separation was explained by convective transport of the solutes through the bed. The contribution of this mode of transport is similarly explained as done for the beds packed with particles with gigapores. For monolithic beds, the concept of an ultrashort bed was frequently used. This mode of operation allows very short separation time. In many cases a gradient elution is necessary to achieve separation. Examples of applications for protein and polynucleotide separation performed on monoliths are given. Enzymatic conversion was described showing the examples of several immobilzed enzymes.  相似文献

The pioneering research work published by Hjertén et al. [J. Chromatogr. 473 (1989) 273] in 1989 dealing with development and application of the continuous bed (monolithic) technique as an attractive alternative for the classical packed columns in chromatography, stimulated further investigations in this direction. The research data published since that time on the development and application of the continuous beds formed using hydrophobic interaction-based phase separation mechanism are reviewed. Some innovative species of the beds, such as polyrotaxane beds or nonparticulate restricted-access materials for direct analysis of the biological fluids in the capillary format are also discussed. Characteristic features and practical details of the continuous bed technique are revealed. Due to many advantages, the continuous bed technique became a competitor with the traditional packings in capillary or chip-based microanalysis. The importance of the continuous bed morphology on the chromatographic characteristics is shown. The applicability of modern microscopic analysis to evaluate the morphology of the continuous beds is demonstrated.  相似文献

Prepacked chromatography columns and cassette filtration units offer many advantages in bioprocessing. These include reduced labor costs and processing times, ease of storage, and enhanced process flexibility. Rectangular formats are particularly attractive as they can be easily stacked and multiplexed together for continuous processing. Cylindrical chromatography beds have dominated bioprocessing even though their bed support and pressure-flow performance vary with bed dimensions. This work presents the performance of novel, rhombohedral chromatography devices with internally supported beds. They are compatible with existing chromatography workstations and can be packed with any standard commercial resin. The devices offer pressure-flow characteristics independent of container-volume, simple multiplexing, and separation performance comparable to cylindrical columns. Their bi-planar, internal bed support allows mechanically less-rigid resins to be used at up to four times higher maximal linear velocities, and productivities approaching 200 g/L/h for affinity resins, compared to the 20 g/L/h typical of many column-based devices. Three 5 L devices should allow processing of up to 3 kg of monoclonal antibody per hour.  相似文献

Mussel beds are known to affect fine sediment dynamics and morphology on mudflat scale, a clear example of ecosystem engineering. Current research into possible ecological engineering applications of mussel beds makes quantitative modeling desirable. In this study a process-based model of the interaction between a young mussel bed and fine sediment was set up for use in the hydrodynamic and morphological model Delft3D-FLOW. The model encompasses the hydraulic roughness of the mussel bed, active capture of suspended sediment by filter feeding and changed bed properties due to biodeposited matter. The mussel bed implementation in Delft3D-FLOW was applied in a test case: a Wadden Sea intertidal mudflat model. It was concluded that a combination of active deposition via filtration and slow down of the flow due to increased roughness leads to high net deposition in the mussel bed. In addition, the ability of young mussels to quickly climb on top of deposited material results in rapid trapping of large amounts of fine sediment. In the wake of the mussel bed, deposition is also high because of reduced flow velocities. The effects of different existing mussel bed patterns were also evaluated. Patchiness and specifically striped patterns cause mussel beds to experience less sedimentation than uniformly covered beds of the same size and may therefore be favorable to mussels.  相似文献

The continuous bed technique with its attractive features, such as fritless design, one-step in situ synthesis, low back pressure and no need for pressurising the electrode vessels to suppress bubble formation was applied to form polyrotaxane-based stationary phases for capillary electrochromatography (CEC). Rotaxanes are synthesized from two classes of substances, namely linear reactive monomers and inert cyclic compounds. Upon polymerisation, a gel forms with the cyclic molecules mechanically immobilized (see Fig. 1). We have employed this simple approach, using charged derivatives of cyclodextrins in order to introduce charged groups into continuous beds and thus render them appropriate for electrochromatography. The self-assembly of supramolecular structures to form rotaxanes during the synthesis of the continuous beds is treated. The electroosmotic and chromatographic properties of the various polyrotaxane-based stationary phases synthesized are discussed, as well as the synthesis of the continuous beds, including how to affect their porosity and its influence on the efficiency of the electrokinetic separation. The applicability of the rotaxane-based continuous bed is demonstrated by separation of model compounds by reversed- and normal-phase chromatography. A separation of enantiomers is also presented. This experiment is of particular interest because it indicates that the interaction with the cavity of beta-cyclodextrin (beta-CD) is not a fundamental requirement for enantioseparations.  相似文献

Rigid chromatography resins, such as controlled pore glass based adsorbents, offer the advantage of high permeability and a linear pressure‐flow relationship irrespective of column diameter which improves process time and maximizes productivity. However, the rigidity and irregularly shaped nature of these resins often present challenges in achieving consistent and uniform packed beds as formation of bridges between resin particles can hinder bed consolidation. The standard flow‐pack method when applied to irregularly shaped particles does not yield well‐consolidated packed beds, resulting in formation of a head space and increased band broadening during operation. Vibration packing methods requiring the use of pneumatically driven vibrators are recommended to achieve full packed bed consolidation but limitations in manufacturing facilities and equipment may prevent the implementation of such devices. The stop‐flow packing method was developed as an improvement over the flow‐pack method to overcome these limitations and to improve bed consolidation without the use of vibrating devices. Transition analysis of large‐scale columns packed using the stop‐flow method over multiple cycles has shown a two‐ to three‐fold reduction of change in bed integrity values as compared to a flow‐packed bed demonstrating an improvement in packed bed stability in terms of the height equivalent to a theoretical plate (HETP) and peak asymmetry (As). © 2014 American Institute of Chemical Engineers Biotechnol. The best hope for understanding global diversity patterns is to compare local assemblages, which are mostly preserved in taphonomically-complex shell beds. The present study investigates the variability in faunal composition and diversity at the scale of a single outcrop. A total of 152 species (3315 shells) occurred in 25 samples from 5 tempestitic shell beds. Although sampling intensity was high, total species richness was not captured by far at the hierarchical levels present (outcrop, shell beds, samples) because the majority of species is rare. In contrast, sampling intensity was sufficient to cover the most abundant species, as indicated by stable evenness values. Four taxa dominate the assemblage, but their rank order differs strongly between individual shell beds and individual samples; significant differences between some shell beds are evident for faunal composition, and one shell bed differs from all others with respect to species accumulation curves. Within shell beds, rarefaction curves are generally characterized by strongly overlapping confidence intervals, but outliers occur in three of five shell beds. Patchiness is additionally indicated by a wide scatter of diversity indices in some shell beds and by a wide scatter of samples of one shell bed in an ordination on faunal composition. Most of the outcrop-scale variability in faunal composition and diversity can be related to differences between shell beds. This suggests that sampling a single shell bed of the outcrop is insufficient to characterize the local fauna and its diversity, even when sampling intensity (i.e. the number of samples and shells) within the shell bed was high. Similarly, a single sample from such a shell bed may not be sufficient to characterize its diversity, even when the number of counted shells was high. It is therefore confirmed that sampling strategy and sampling intensity are crucial to confidently characterize the shelly assemblages at such a small spatial scale and that dispersed sampling effort with many small replicate samples will characterize a local assemblage and its diversity better than a few large samples. Diversity comparisons of individual samples between localities must account for the high variability present at the smaller spatial scale, as observed in our study.  相似文献

Genetic structure and diversity can reveal the demographic and selective forces to which populations have been exposed, elucidate genetic connections among populations, and inform conservation strategies. Beds of the clonal marine angiosperm Zostera marinaL. (eelgrass) in Chesapeake Bay (Virginia, USA) display significant morphological and genetic variation; abundance has fluctuated widely in recent decades, and eelgrass conservation is a major concern, raising questions about how genetic diversity is distributed and structured within this metapopulation. This study examined the influence of bed age (<65years versus<6years) and size (>100ha versus<10ha) on morphological and genetic (allozyme) structure and diversity within Chesapeake Bay eelgrass beds. Although both morphology and genetic diversity varied significantly among individual beds (F ST=0.198), neither varied consistently with bed age or size. The Chesapeake eelgrass beds studied were significantly inbred (mean F IS=0.680 over all beds), with inbreeding in old, small beds significantly lower than in other bed types. Genetic and geographic distances within and among beds were uncorrelated, providing no clear evidence of isolation by distance at the scale of 10's of km. These results suggest that local environmental conditions have a greater influence on plant morphology than do bed age or size. They support the hypotheses that eelgrass beds are established by multiple founder genotypes but experience little gene flow thereafter, and that beds are maintained with little loss of genetic diversity for up to 65 years. Since phenotypic and genotypic variation is partitioned among beds of multiple ages and sizes, eelgrass conservation efforts should maximize preservation of diversity by minimizing losses of all beds.  相似文献

Electrospun polymeric nanofiber adsorbents offer an alternative ligand support surface for bioseparations. Their non‐woven fiber structure with diameters in the sub‐micron range creates a remarkably high surface area. To improve the purification productivity of biological molecules by chromatography, cellulose nanofiber adsorbents were fabricated and assembled into a cartridge and filter holder format with a volume of 0.15 mL, a bed height of 0.3 mm and diameter of 25 mm. The present study investigated the performance of diethylaminoethyl (DEAE) derivatized regenerated cellulose nanofiber adsorbents based on criteria including mass transfer and flow properties, binding capacity, and fouling effects. Our results show that nanofibers offer higher flow and mass transfer properties. The non‐optimized DEAE‐nanofiber adsorbents indicate a binding capacity of 10% that of packed bed systems with BSA as a single component system. However, they operate reproducibly at flowrates of a hundred times that of packed beds, resulting in a potential productivity increase of 10‐fold. Lifetime studies showed that this novel adsorbent material operated reproducibly with complex feed material (centrifuged and 0.45 µm filtered yeast homogenate) and harsh cleaning‐in‐place conditions over multiple cycles. DEAE nanofibers showed superior operating performance in permeability and fouling over conventional adsorbents indicating their potential for bioseparation applications. Biotechnol. Seagrass habitat structure influences epifaunal density, diversity, community composition and survival, but covariation of structural elements at multiple scales (e.g., shoot density or biomass per unit area, patch size, and patch configuration) can confound studies attempting to correlate habitat structure with ecological patterns and processes. In this study, we standardized simulated seagrass shoot density and bed area among artificial seagrass beds in San Diego Bay, California, USA to evaluate the singular effect of seagrass bed configuration (“patchiness”) on the density and diversity of seagrass epifauna. Artificial seagrass beds all were 1 m2, but were composed of a single large patch (“continuous” treatment), four smaller patches (“patchy” treatment), or 16 very small patches (“very patchy” treatment). We allowed epifauna to colonize beds for 1 month, and then sampled beds monthly over the next 3 months. Effects of seagrass bed patchiness on total epifaunal density and species-specific densities were highly variable among sampling dates, and there was no general trend for the effects of fragmentation on epifaunal densities to be positive or negative. Epifaunal diversity (measured as Simpson's index of diversity) was highest in very patchy or patchy beds on two out of the three sampling dates. Very patchy beds exhibited the highest dissimilarity in community composition in the first two sampling periods (August and September), but patchy beds exhibited the highest dissimilarity in the third sampling period (October). Our results indicate that seagrass patch configuration affects patterns of epifaunal density, diversity, and community composition in the absence of covarying bed area or structural complexity, and that patchy seagrass beds may be no less valuable as a habitat than are continuous seagrass beds. The spatial pattern employed when harvesting or planting seagrass may influence epifaunal habitat use and should be a key consideration in restoration plans.  相似文献

To clarify differences in community structures and habitat utilization patterns of fishes in Enhalus acoroides- and Thalassia hemprichii-dominated seagrass beds on fringing coral reefs, visual censuses were conducted at Iriomote and Ishigaki islands, southern Japan. The numbers of fish species and individuals were significantly higher in the E. acoroides bed than in the T. hemprichii bed, although the 15 most dominant fishes in each seagrass bed were similar. Cluster and ordination analyses based on the number of individuals of each fish species also demonstrated that fish community structures were similar in the two seagrass beds. Species and individual numbers of coral reef fishes which utilized the seagrass beds numbered less than about 15% of whole coral reef fish numbers, although they comprised about half of the seagrass bed fishes. Of the 15 most dominant species, 5 occurred only in the two seagrass beds, including seagrass feeders. Ten other species were reef species, their habitat utilization patterns not differing greatly between the two seagrass beds. Some reef species, such as Lethrinus atkinsoni and L. obsoletus, showed ontogenetic habitat shifts with growth, from the seagrass beds to the coral areas. These results indicate that community structures and habitat utilization patterns of fishes were similar between E. acoroides- and T. hemprichii-dominated seagrass beds, whereas many coral reef fishes hardly utilized the seagrass beds.  相似文献

This work demonstrates an experimental method for studying breakthrough behaviour in expanded beds. The behaviour of beds made with differently sized particles were studied at varying flowrates. The use of a dimensionless residence time measurement allowed a more valid comparison of breakthrough characteristics in expanded bed operation by compensating for the changes in bed volume that occur during expansion. We demonstrate that bed breakthrough behaviour can be compared directly even when the beds contain different-sized particles and hence have different expanded volumes. By utilising this concept we demonstrate that, in the case of the Alcohol Dehydrogenase (ADH) / STREAMLINE Phenyl system used here, there was little or no variation in ADH breakthrough behaviour between beds of differently sized particles operating at flowrates above 100 cm/h. This suggests that the higher specific surface area and hence binding capacity of smaller particles is negated in this case due to mass transfer limitations and the increase in system void volume even at normal operating flowrates of 200-300 cm/h.  相似文献

The potential of reed beds to act as biofilters of pathogenic and environmental mycobacteria was investigated through examination of the fate of mycobacteria in a constructed reed bed filtering effluent from a large captive wildfowl collection. Particular emphasis was placed on the presence and location of Mycobacterium avium – the causal agent of avian tuberculosis (ATB) – in an effort to clarify the potential role of reed beds in the control of this disease. Water, sediment, and stems and roots of common reed ( Phragmites australis ) and greater reedmace ( Typha latifolia ) were taken from 15 locations within the reed bed plus sites upstream and downstream. Samples were analysed for mycobacteria using PCR and specifically for M. avium using nested PCR. Environmental mycobacteria were found throughout the entire reed bed but M. avium was not found downstream of the first vegetation growth. The reed bed was found to effectively remove M. avium from the water through a combination of sedimentation and adsorption onto vegetation stems. The results of this study show that constructed reed beds composed of a settlement lagoon and one or more vegetation beds can act as valuable and ecologically friendly tools in the environmental control of ATB.  相似文献

Field surveys over 2 years in contiguous beds of the seagrassesZostera capricorni andPosidonia australis showed that the green algaCodium duthieae was consistently more abundant inZ. capricorni than inP. australis. In 1 year, mature plants were also more abundant at the boundary between the seagrass beds than in either bed. Field experiments and programmes of sampling were used to investigate three potential explanations for the unusual distribution of this alga: (1) that the availability of substrata suitable for attachment of the alga differed between the two seagrass beds; (2) that mortality of matureC. duthieae differed between the seagrass beds; and/or (3) that the intensity of recruitment was different in the two seagrass beds.C. duthieae plants were exclusively epibionts of the bivalveAnadara trapezia. Detailed sampling showed that the abundance ofA. trapezia was similar in both seagrass beds and that the distribution of bivalves suitable as substrata forC. duthieae plants was not obviously related to proximity to the boundary between the beds. Two experiments investigated the survival ofC. duthieae plants in each bed. In the first, matureC. duthieae plants transplanted into theP. australis bed suffered similar rates of mortality to plants which were disturbed and moved within theZ. capricorni bed or which were left undisturbed in theZ. capricorni bed. Fewer of the host bivalves were recovered from theZ. capricorni bed, however, indicating that the mechanism of mortality differed between the beds, hosts being more frequently dislodged in theZ. capricorni bed. Removal of the leaves of the seagrasses had consistently greater effects on near-bottom current velocities in theZ. capricorni bed than in theP. australis bed and significantly increased mortality ofC. duthieae in theZ. capricorni bed. Survival of plants was greater in plots of artificial leaves ofP. australis placed in theZ. capricorni bed than in plots of the naturalZ. capricorni leaves or plots where the natural leaves were removed. Most mortality in theZ. capricorni bed was due to dislodgement of the alga and its bivalve substratum. Corresponding manipulations of leaves in theP. australis bed had consistently smaller effects on survivorship of both the alga and its host. Patterns in the recruitment of the alga most clearly reflected the distribution of adults.C. duthieae recruits were 5 times more abundant in theZ. capricorni bed and at the boundary between the two beds than in theP. australis bed. The results demonstrate how habitat structure, provided by the canopy of leaves of the two species of seagrass, can have contrasting effects on the recruitment and mortality of a macroalga. In the case ofC. duthieae, it appears that the differential pattern of recruitment is the primary determinant of the distribution of adult plants.  相似文献

In a wide variety of biotechnological and medical applications it is necessary to separate different cell populations from one another. A promising approach to cell separations is demonstrated to be the adoption of chromatographic techniques conducted in expanded beds. The high voidage between the adsorbent beads in an expanded bed allows for the efficient capture of particulate entities such as cells together with washing and subsequent elution without entrapment and loss. In addition, the combination of a gentle hydrodynamic environment, a high surface area and low mixing within the expanded bed make this technique highly favourable. A model system for the separation of two types of microbial cells using STREAMLINE DEAE adsorbent in expanded bed procedures has been investigated. The use of a less selective ligand such as an ion exchange group, which is often characterised by gentle elution procedures, has been investigated as an alternative to affinity ligands whose strong binding characteristics can result in harsh elution procedures with consequent loss of yield and cell viability. Expanded bed experiments have demonstrated selective and high capacity capture of cells from feedstocks containing either a single type of cell or as a mixture of cells of Saccharomyces cerevisiae and Eschericia coli. The capture, washing and elution phases of the separation have been studied with respect to capacity, selectivity and yield of released cells. In these procedures, separation of cell types is achieved by the presence of multiple equilibrium stages within the expanded bed. The results show the potential for carrying out cell separations in expanded beds as an alternative to immunomagnetic cell separations. The combination of these recently developed technologies promises to be a powerful, but economic technique for cell separations involving simple equipment that can readily be scaled up.  相似文献

Vegetated areas of rivers and estuaries are capable of affecting the concentration of dissolved and particulate matter in water masses traversing those plant beds. We examined whether different sizes of water chestnut (Trapa natans) beds in the Hudson River, USA, alter dissolved oxygen, nutrients and turbidity of water masses. Ebb–tide water was sampled from four water chestnut beds in the tidal freshwater portion of the Hudson River estuary and each site was sampled multiple times during the growing season and once following plant senescence. Water quality variables included dissolved oxygen, turbidity, dissolved organic carbon, and inorganic nutrients. Samples from the small beds (575 m2 and 624 m2) were compared with large beds (16 600 m2 and 24 820 m2). Dissolved oxygen of water flooding vegetated beds in the hour before high tide was 7.18±1.03 mg/l (mean±standard deviation) with a range of 5.5–9.8 mg/l throughout the growing season. Water samples collected as water ebbed from the plant beds showed that only the large beds had an effect on dissolved oxygen with the largest declines in oxygen exhibited by the largest bed. Decline of dissolved oxygen in the water ebbing from the largest bed averaged 1.5±0.4 mg/l/h with a minimum of 4.5 mg/l, equivalent to 54% of saturation, a level at which sensitive fauna are negatively affected. There were no significant relationships between bed size or plant presence and inorganic nutrients, turbidity or DOC. Ebb–tide nitrate was never lower than 87% of flood tide means. Effective management of invasive plants must consider both the variability in effects among plant beds and the areal coverage of plant bed sizes.  相似文献

Material exchange, biodiversity and trophic transfer within the food web were investigated in two different types of intertidal seagrass beds: a sheltered, dense Zostera marina bed and a more exposed, sparse Z. noltii bed, in the Northern Wadden Sea. Both types of Zostera beds show a seasonal development of above-ground biomass, and therefore measurements were carried out during the vegetation period in summer. The exchange of particles and nutrients between seagrass beds and the overlying water was measured directly using an in situ flume. Particle sedimentation [carbon (C), nitrogen (N) and phosphorus (P) constituents] from the water column prevailed in dense seagrass beds. In the sheltered, dense seagrass bed, a net particle uptake was found even on windy days (7–8 Beaufort). Dissolved inorganic N and orthophosphate were mainly taken up by the dense seagrass bed. At times of strong winds, nutrients were released from the benthic community to tidal waters. In a budget calculation of total N and total P, the dense seagrass beds were characterised as a material sink. The seagrass beds with sparse Z. noltii were a source of particles even during calm weather. The uptake of dissolved inorganic N in the sparse seagrass bed was low but significant, while the uptake of inorganic phosphate and silicate by seagrasses and their epiphytes was exceeded by release processes from the sediment into the overlying water. Estimates at the ecosystem level showed that material fluxes of seagrass beds in the Sylt-Rømø Bight are dominated by the dense type of Zostera beds. Therefore, seagrass beds act as a sink for particles and for dissolved inorganic nutrients. During storms, seagrass beds are distinct sources for inorganic nutrients. The total intertidal area of the Sylt-Rømø Bight could be described as a sink for particles and a source for dissolved nutrients. This balance of the material budget was estimated by either including or excluding seagrass beds. Including the subtidal part, the function of the ecosystem as a source for particles increased, supposing that all seagrass beds were lost from the area. During the vegetation period, seagrass beds act as a storage compartment for material accumulated in the living biomass of the community. There was great biodiversity among the plant and animal groups found in intertidal seagrass beds of the Sylt-Rømø Bay, representing 50–86% of the total number of species investigated, depending on the particular group. Since most species are not exclusively seagrass residents, the loss of intertidal seagrass beds would be of minor importance for biodiversity at the ecosystem level. Food web structure in seagrass beds is different from other intertidal communities. Primary production and detritus input is high, but secondary production is similar to that of unvegetated areas, although the relative importance of the trophic guilds is different. The loss of seagrass beds leads to profound alterations in the food web of the total ecosystem. Historical as well as recent changes in material fluxes and energy flow due to man-made alterations to the ecosystem are discussed.  相似文献

In this paper we describe bed (nest) and bed-site reuse by western lowland gorillas (Gorilla g. gorilla) in Moukalaba-Doudou National Park, south-eastern Gabon. During an eight-month study 44 bed sites and 506 beds were found. Among these, 38.6% of bed sites and 4.1% of beds were reused. We analyzed the monthly frequency of bed-site reuse in relation to rainfall, fruit abundance, and fruit consumption by the gorillas. The different frequency of bed-site reuse in the rainy and dry seasons was not significant. More bed-site reuse was observed during the fruiting season than during the non-fruiting season. Results from fecal analysis suggested that gorillas ate more fruit in the fruiting season than in the non-fruiting season. The frugivorous diet of western gorillas may possibly cause gorillas to stay in some areas and, consequently, reuse their bed sites. Reuse of bed sites by gorillas suggests their frequent return to an area where preferred fruit is readily available. A higher percentage of arboreal beds may also affect bed-site reuse, because of the shortage of bed material.  相似文献
