Intravenous inoculation of young rabbits with 1.4 × 105 spores ml-1 of Absidia corymbifera produced gross and microscopic changes principally in the kidneys. Grossly, there was congestion, haemorrhages and greyish-white necrotic foci giving a mottled appearance to the kidneys. Microscopically, besides congestion and haemorrhages, there was a granulomatous reaction characterized by a central necrotic mass containing mycelium surrounded by a reactive zone of neutrophils, lymphocytes, a few macrophages and giant cells. Fibrous tissue proliferation was also present. Characteristic dichotomously branched non-septate fungal hyphae could be demonstrated in the sections of kidneys up to 14 days of infection. In one rabbit, which had died on the 8th day, small granulomatous lesions with fungal hyphae were noticed in the liver's parenchyma. Zygomycosis was produced experimentally in 20 New Zealand white rabbits (Oryctolagus cuniculus) by intra-nasal administration of spores of Absidia corymbifera. Infected animals showed dullness, depression, coughing and mucopurulent nasal discharge, but no mortality. Haematology revealed no significant change in Hb and PCV, but leukocytosis due to neutrophilia in the initial stages of the experiment. There was a significant increase in serum total proteins, creatinine, AST, ALT, total Igs and CICs. A. corymbifera specific IgM and IgG antibodies were detected in the sera of the infected animals. Gross lesions consisted of pneumonic consolidations of the antero-ventral lobes of the lungs. Microscopically, histology showed formation of pyogranulommas in the lungs. Fungal elements typical of A. corymbifera were demonstrated in the tissues upto 15 days after infection by special stains and confirmed by indirect immunoperoxidase. Re-isolation of the fungus from lungs was also achieved consistently up to 15 days only. It was concluded that intra-nasal instillation of A. corymbifera in rabbits produced significant clinico-pathological alterations with the lesions confined mainly to the lungs. In the present study, neither systemic dissemination of the disease occurred nor were kidneys site of predilection as reported earlier. The present study, first of its kind, was conducted with the objectives to understand hitherto little known aspects of candidal mastitis, like its sequential pathology, pathogenesis and clinico-biochemical changes. For this purpose, unilateral intramammary inoculation of 10 goats with Candida albicans (1.2 × 107 yeast cells) resulted in the development of mastitis, with gross and microscopic lesions being restricted to the infected udder halves only and without dissemination of infection to the opposite uninfected udder halves as well as other organs of the body. The experiment was continued for 40 days and after infection, there was sharp fall in milk yield and Candida albicans was directly demonstrated in the milk and re-isolated from the milk and udder tissues up to 30th day after inoculation. An increase in total immunoglobulins in the milk and plasma along with increase in total plasma proteins were also observed. Haematology revealed leukocytosis and neutrophilia. Microscopically, there was acute purulent mastitis, which later became chronic, nonpurulent and interstitial with formation of granulomas. It was concluded that Candida albicans was highly pathogenic to the lactating goat mammary gland even without immunosuppression or antibiotic treatment, resulting in severe irreversible tissue damage and nearly complete agalactia. Zygomycosis was produced experimentally in Asian water buffalo calves (Bubalus bubalis) by intratracheal inoculation of sporangiospores of Absidia corymbifera. Infected animals exhibited dullness, depression, partial anorexia, initial pyrexia, mucopurulent nasal and ocular discharge and coughing during the first week. There was no mortality at any stage of the experiment, which continued for 30 days. The gross and microscopic lesions were restricted to the lungs and there was no dissemination of the fungus to other organs. Gross and microscopic changes in the lungs were observed up to the 20th day post-infection. Gross lesions consisted of pneumonic consolidation of the antero-ventral lobes of the lungs. Microscopic changes consisted of granulomatous reactions with well developed pulmonary granulomas. Distorted hyphae of A. corymbifera were demonstrated in tissue sections up to 15 days post inoculation. Re-isolation of the fungus was achieved consistently for up to 15 days. It is concluded that intratracheal inoculation of A. corymbifera in buffalo calves leads to significant pathological changes in the lungs, but the disease appears to be self limiting 20 days following inoculation.  相似文献

The study was envisaged for elucidating the effect of immunosuppression on the pathology and pathogenesis of experimental respiratory absidiomycosis in rabbits. Seventy rabbits used in the experiment were divided into different groups viz. non-immunosuppressed, immunosuppressed with cyclophosphamide and immunosuppressed with methylprednisolone respectively, and a control group. The experiment was continued for 50 days during which all the infected animals exhibited clinical signs of respiratory distress but no mortality. The infected rabbits showed gross lesions mainly in the form of pneumonic consolidations in the lungs in antero-ventral lobes initially followed by spread to the other lobes. Similarly, microscopic lesions were restricted to lungs and consisted of early pyogranulomas dominated by fungal invasion and spread, followed by chronic granulomatous reaction and a recovery during the later stages. Fungus was demonstrated and re-isolated only from lung lesions up to 15 days postinfection (DPI) in non-immuno-suppressed group and up to 30 DPI in both the immunosuppressed groups suggesting no systemic dissemination. The findings of the present study indicated that immunosuppression increased the extent, duration and severity of the pathological lesions associated with absidiomycosis in the lungs. Moreover, an assessment based on gross and histopathological lesions revealed cyclophosphamide to be a more potent immunosuppressant than methylprednisolone.  相似文献

Parenteral administration of bovine placental extract to mice in the period overlapping inoculation with spores of Absidia corymbifera did not increase their susceptibility to acute, lethal infection of the central nervous system. It did, however, exacerbate the chronic and sub-acute renal lesions which developed in those animals which survived inoculation without developing acute infection. This probably resulted from excretion by the kidneys of fungal growth stimulants derived from the placental extract.  相似文献

Five erythrocyte proteins (Adenosine deaminase, Ada; 6-Phosphogluconate dehydrogenase, Pgd; Esterase 1, Es-1; Esterase 3, Es-3; NADH-Diaphorase 2, Dia-2) and a serum beta-globulin protein (Esterase 7, Est-7) were studied in rabbits using starch gel electrophoresis. Samples were obtained from 317 Spanish Common individuals (38 families, 317 individuals). For the analyses of linkage, Morton's sequential probability ratio test was applied. Clear evidence for linkage between Es-1 and Est-7 (theta 0.2) was obtained, and no evidence of linkage was obtained for the remaining pairwise combinations of loci studied.  相似文献

Serum samples from 412 Spanish wild rabbits were analysed by starch and polyacrylamide gel electrophoresis. Three different transferrin (Tf) phenotypes (A, AB and B) were observed by both methods. The occurrence of two codominant alleles (TfA and TfB with frequencies of 0.89 and 0.11 respectively) at an autosomal locus (Tf) was supported by the population data on genetic equilibrium. Electrophoretic mobility differences between the Tf variants A and B could not be explained by differences in sialic acid or iron contents. Each of the two Tf variants were shown to have two sialic acid residues by neuraminidase treatment. These variants had similar affinities for iron, and iron binding did not lead to the conversion of one variant into the other.  相似文献

Rabbit haemorrhagic disease virus (RHDV) is a highly virulent lagovirus endemic in Europe and Australasian populations of the European rabbit, Oryctolagus cuniculus. It has also caused several unexplained disease outbreaks in domestic European rabbits in North America. Non-pathogenic spread of RHDV leading to persistent infection which later reactivated has recently been proposed as the cause of overt disease and death of a pet rabbit in Canada, the first confirmed case of Rabbit haemorrhagic disease in that country. We suggest that there is little evidence to support non-pathogenic spread of virulent RHDV, some evidence that is contradictory, and evidence to support a simpler alternative hypothesis. RHDV can be spread over long distances between sparse rabbit populations by fomites or flying insects. Although highly pathogenic, RHDV can be limited in its spread within rabbit populations, or its presence masked by closely related but non-pathogenic lagoviruses which can provide protection against acute disease. In the absence of any evidence from clinical studies to support reactivation of persistent RHDV infection, the simpler explanation seems more likely to be correct.  相似文献

The combination of artemether plus lumefantrine is a type of artemisinin‐based combination therapy (ACT) recommended by the World Health Organization for uncomplicated falciparum malaria except in the first trimester of pregnancy. The first trimester restriction was based on the marked embryotoxicity in animals (including embryo death and cardiac and skeletal malformations) of artemisinins such as artesunate, dihydroartemisinin, and artemether. Before recommending ACTs for use in the first trimester, the World Health Organization has requested that all information relevant to the assessment of risk of ACTs to the embryo be made available to the public. This report describes the results of embryo‐fetal development studies of artemether alone, lumefantrine alone, and the combination in rats and rabbits as well as toxicokinetic studies of lumefantrine in pregnant rabbits. The developmental no‐effect levels for lumefantrine were 300 mg/kg/day in rats (based on a 25% decrease in litter size at 1000 mg/kg/day) and 1000 mg/kg/day in rabbits. The calculated safety margins based on human equivalent dose and plasma Cmax and AUC values were in the range of 2.5‐ to 17‐fold. The developmental no‐effect levels for artemether were 3 mg/kg/day in rats and 25 mg/kg/day in rabbits. Lumefantrine caused no teratogenicity and was not a potent embryotoxin in rats and rabbits. Expected artemisinin‐like findings were seen with artemether alone and with artemether/lumefantrine combined except that no malformations were observed. There were no findings in pregnant rats and rabbits that would cause increased concern for the use of artemether–lumefantrine in the first trimester compared to other ACTs.  相似文献

Significant variations in the number of white and red blood cells, haematocrit and haemoglobin were found throughout the year in sexually mature male and female tench Tinca tinca . In general, the lowest values were observed during autumn–winter and the highest during summer, with males exhibiting higher values than females. Plasma glucose, cholesterol and triglycerides were lower during the winter than during the summer–autumn seasons in both sexes. Gonado-somatic and hepato-somatic indices were inversely correlated in female tench throughout the year. Seasonal patterns in liver metabolic resources were very similar for both sexes. For males and females, liver glycogen and proteins increased during the autumn, whereas the liver stored lipid during spring. Dorsal muscle mainly deposited glycogen, whereas lipid was mainly stored in the ventral muscle. Relations between seasonal changes in environmental factors, such as feeding and temperature are discussed.  相似文献

通过光镜和电镜观察了中华硬蜱Ixodes sinensis叮咬初次和再次感染宿主新西兰兔后不同时间(叮咬后24 h、48 h、72 h以及第 5天、第8天)中肠上皮组织的形态学动态变化。结果显示: 中华硬蜱叮咬前中肠上皮主要由替代细胞和少量体积较大的消化细胞构成;替代细胞数量多、体积小、呈圆形、胞质染色浅 。中华硬蜱叮咬初次感染宿主后,消化细胞随叮咬时间延长而增多增大,微绒毛较密集,排列整齐,胞质内细胞器丰富,各单位膜结构清晰,并出现顶端小管、小泡、大量脂滴和高铁血红素颗粒;近基膜的细胞膜内褶形成发达的基底迷路系统。中华硬蜱叮咬再次感染宿主后,中肠可发生一系列明显的病理变化,中肠基膜出现变薄、松散和断裂现象,消化细胞破裂、空泡化,消化细胞数量减少;消化细胞微绒毛减少、变短、排列不整,线粒体肿大,体嵴减少、变短甚至髓样变,粗面内质网扩张,脂粒及高铁血红素颗粒减少,细胞膜吞饮、吞噬现象减弱,消化细胞内结构紊乱和破坏。该研究结果提示初次叮咬导致了宿主的免疫抗性,再次叮咬后蜱中肠是宿主免疫力的主要作用部位。  相似文献

Bacteriological and serological examinations were made on Pasteurella multocida infection in two rabbit-breeding colonies. The organism was localized in the paranasal sinuses of 53 of 54 infected rabbits, excreting to the external nares of 49 rabbits. It was also isolated from the trachea and middle and inner ear of half of the infected animals and occasionally from the conjunctiva, lung and heart. The infection rate of the organism was very low (4.3%) in sucklings younger than a month old, but increased with advancing age, reaching nearly 100% in adults more than five months of age. A rapid increase of the infection rate was observed at two to three months of age. Serum antibody against somatic antigen of P. multocida was demonstrated in infected rabbits by use of the tube agglutination test, showing a close correlation with isolation of the organism in adults. Sucklings younger than one month of age from infected dams were significantly resistant to experimental nasal infection of P. multocida.  相似文献

BACKGROUND: A frequent drawback in physiology of non-human primates is that normal values for a variety of indicators (haematological, biochemical and others) are scant. METHODS: We report here the blood values in a series of 92 healthy Cebus apella (divided by sex, age and pregnancy status). Health check-ups indicated that animals were healthy for the month prior to and after the sampling. Dietary intake was estimated on the basis of two semi-balance studies. RESULTS: Values of haematological indicators, serum LDH activity, micronutrient indicators (serum copper, iron and serum ceruloplasmin concentrations, Zn-Cu-superoxide dismutase activity in erythrocytes) agreed with previous results and provide some values that were not available. Activities of liver enzymes were lower than some previously reported. CONCLUSIONS: These results provide valuable information that help understanding the physiology of C. apella.  相似文献

Context: Nickel oxide (NiO) nanoparticles (NPs) with appropriate surface chemistry have been widely used for their potential new applications in biomedical industry. Increased usage of these NPs enhances the chance of exposure of personnel involved in the work place.

Objective: This study was designed to assess the ability of NiO NPs to cause biochemical alterations post-acute oral exposure in female Wistar rats.

Materials and methods: Rats were administered with 125, 250, and 500?mg/kg doses of NiO NPs for haematological, biochemical, and histopathological studies. Biodistribution patterns of NiO NPs in female Wistar rats were also monitored.

Results: NiO NPs caused significant (p?Conclusions: This study revealed that exposure to nanosized NiO particles at acute doses may cause adverse changes in animal biochemical profiles. Further, the in vivo studies on toxicity evaluation help in biomonitoring of the potential contaminants.  相似文献

Female ICR mice were infected with HSV-1 and HSV-2 by inserting a cotton pellet soaked in viral solution (10(7-8) PFU/m1) into the vagina. The appearance of giant cells and formation of intranuclear inclusions were detected in the epithelial layer of the uterus 24 h after intravaginal inoculation. These histopathological changes were pronounced 3 to 4 days after virus inoculation and then gradually disappeared in the next few days. Results of fluorescent antibody studies on the appearance of viral antigens in infected uterine tissues and results of viral infectivity titrations of emulsified samples of infected uteri coincided well with the histopathological observations on the general course of virus infection. The degree of histopathological involvement caused by HSV-1 was somewhat less than that caused by HSV-2, and the laboratory strains of HSV-1 so far examined (HF and Miyama) were found to be especially weakly pathogenic.  相似文献

The analysis of haematological parameters is an important element of the assessment of the physiological condition of animals. Haematological parameters may change both under the influence of various external factors, and in the course of normal pregnancy, which has been found in various species of mammals, including rabbits. Our study showed statistically significant (p<0.05) changes in basic haematological parameters: RBC (decrease; 5.87±0.48 at day 15 vs. 5.42±0.32 T/L at day 26), MCH (increase; 1.35±0.04 before matching vs. 1.41±0.03 fmol at day 26), RDW (decrease; 15.77±1.80 at day 15 vs. 14.27±1.57% at day 26) MPV (increase; 5.17±0.31 at day 15 vs. 5.92±0.70 fL at day 26), WBC (decrease; 8.60±2.57 at day 15 vs. 4.94±0.88 G/L at day 26) and PLT (decrease; 398.17±91.67 before matching vs. 271.67±61.72 G/L at day 26) in Termond White rabbits and RBC (decrease; 6.18±0.68 before matching vs. 5.68±0.54 T/L at day 26), Hb (decrease; 8.00±0.90 before matching vs. 7.32±0.71 mmol/L at day 26), MCH (decrease; 1.32±0.05 at day 15 vs. 1.29±0.04 fmol at day 26) and WBC (decrease; 9.62±1.81 before matching vs. 5.85±2.23 G/L at day 26 as well as 9.58±2.35 at day 15 vs. 5.85±2.23 G/L at day 26) in Popielno White rabbits. Moreover, in the Popielno White rabbits we recorded a significant (p<0.05) decrease in the percentage of irregular erythrocytes at the end of pregnancy (11.00±10.02 at day 15 vs. 3.00±4.94 at day 26). The changes appear to be physiological but should be considered in studies using rabbits as model organisms.  相似文献

Diniconazole [(E)-(RS)-1-(2,4,-dichlorophenyl)-4,4-dimethyl-2-(1H-1,2,4-triazole-1-yl)pent-1-en-3-ol)] is a potent triazole fungicide. The enantioselective pharmacokinetics of diniconazole enantiomers in rabbits was studied via intravenous (i.v.) injection. The pharmacokinetics and the enantiomer fraction (EF) were determined using normal high-performance liquid chromatography with diode array detection and a cellulose-tris-(3,5-dimethylphenylcarbamate)-based chiral stationary phase (CDMPC-CSP). The time-concentration curves in plasma were fitted by a two-compartment open mode. The results showed that the concentration of S-diniconazole in plasma decreased faster than that of R-diniconazole, and EFs increased with time after administration of racemic diniconazole (rac-diniconazole). The R-/S-enantiomer ratio of the area under the time-plasma concentration curve (AUC(0-infinity)) after administration was 1.52. The total plasma clearance value of S-enantiomer was 1.57-fold higher than that of the R-diniconazole. These results indicate substantial stereoselectivity in the kinetics of diniconazole enantiomers in rabbit.  相似文献

Intraocularly circulating hot water was used to produce cataracts in nine eyes of seven rabbits by maintaining their retrolental temperatures between 43 degrees C and 45 degrees C. A rapid rate of heating (1.3 degrees C/min) plus a sharp temperature gradient across the eye may have been contributing factors in the consistent production of cataracts at these temperatures. Biomicroscopy and light microscopy showed lens changes similar to those associated with acute exposure to microwave radiation. These findings support the assumption that microwave cataractogenesis is due to the local production of elevated temperatures.  相似文献
