Our previous investigation of the naturally occurring indolecompounds in barley and tomato shoots suggested that the biosynthesisof indol-3yl-acetic acid (IAA) from tryptophan might proceedvia either the indol-3yl-pyruvic acid or tryptamine pathwaysin both species. The results further indicated that the indol-3yl-lacticacid pathway for IAA formation might also be operative in tomato.In the present study, tryptophan-3-14C and tryptamine-2-14Cwere fed to excised shoots of both barley and tomato, and indol-3yl-lacticacid-3-14C was also fed to shoots of tomato. All three compoundswere found to give rise to radioactive IAA with little dilutionin specific activity. Feeding tryptophan-3-14C also resultedin the labelling of indol-3yl-pyruvic acid, indol-3yl-acetaldehyde,and tryptamine, which were isolated and chemically identifiedfrom both species, and radioactive indol-3yl-lactic acid andtryptophol were also produced in tomato. Indol-3yl-acetaldehydewas found to be labelled in both species after administrationof tryptamine-2-14C, while the principal metabolite of indol-3yl-lacticacid-3-14C was radioactive tryptophan. These findings, alongwith the results from a quantitative study of the radioactivemetabolites, indicate that both the indol-3yl-pyruvic acid andtryptamine pathways can operate in both species, while the formationof IAA from indol-3yl-lactic acid in tomato probably occursindirectly, via tryptophan. These conclusions were supportedby the demonstration of the enzymes, L-tryptophan transaminase,L-trypto-phan decarboxylase, and indol-3yl-acetaldehyde dehydrogenasein cell-free extracts of both tissues, and of indol-3yl-pyruvicacid decarboxylase in the tomato extract. No indol-3yl-lacticacid decarboxylase activity was observed in the extracts fromeither tissue.  相似文献

The effects of methyl jasmonate and jasmonic acid on uptake of abscisic acid (ABA) by suspension-cultured runner-bean cells and subapical runner-bean root segments have been investigated. Increasing concentrations of methyl jasmonate inhibit ABA uptake by the cultured cells with a K i of 22±3 M. This is not due to cytoplasmic acidification or to effects on metabolism of ABA, and is not additive with inhibition of radioactive ABA uptake by nonradioactive ABA. Uptake of indol-3-yl acetic acid (IAA) is unaffected by methyl jasmonate. The maximum effect of nonradioactive ABA in inhibiting uptake of radioactive ABA, previously shown to reflect saturation of an ABA carrier, is generally greater than the effect of maximally inhibitory concentrations of methyl jasmonate. Similar results were obtained with root segments, but longer incubation times were necessary to observe inhibitory effects of methyl jasmonate. Demethylation of methyl jasmonate to jasmonic acid does not appear to be required since similar concentrations of jasmonic acid had no observable direct effect on ABA uptake other than that attributable to cytoplasmic acidification. Histidine reagents, a proton ionophore and acidic external pH all affect in parallel the inhibition by methyl jasmonate and nonradioactive ABA of uptake of radioactive ABA by the cultured cells. There is no effect of ABA or nonradioactive methyl jasmonate on uptake of radioactive methyl jasmonate by the cultured cells. It is proposed that methyl jasmonate interacts with the ABA carrier. The uptake of abscisic acid (ABA) by suspension-cultured Phaseoluscoccineus L. cv. ‘Prizewinner’ (runner bean) cellshas been studied. In addition to non-mediated diffusive uptakeof ABAH, a saturable component of uptake occurs which is demonstrableas inhibition of uptake of submicromolar concentrations of [2-14C]ABAby increasing concentrations of nonradioactive ABA to a constantplateau level. Saturation is not due to competition for extracellularbinding sites or cytoplasmic acidification and can only be partiallyaccounted for by saturation of metabolic enzymes. It is consideredlargely to represent carrier-mediated ABA uptake. The maximumvelocity of the carrier is greater at pH 4.0 than at pH 5.0,although the apparent Michaelis constants are similar (about2.0 mmol m–3). No carrier activity was detectable abovepH 6.0. The carrier is unaffected by indol-3-yl acetic acid(IAA), gibberellin A3, benzyladenine or 2, 3, 5-triiodobenzoicacid (TIBA). Use of protein-modifying reagents suggested a roleof histidine residues in carrier activity. Reagents expectedto modify the transmembrane pH ( pH) and electrical ( E) gradientswere used to study the driving forces for ABA uptake. Therewas no apparent effect of E, indirectly monitored by effectson an electrogenic TIBA-sensitive component of IAA transport,on ABA uptake. Both diffusive and saturable components weremodified in proportion by changes in pH, suggesting a commonsensitivity to this driving force. The carrier is suggestedto act as an ABA/H+ symport. Abscisic acid (ABA) applied to the decapitated second internodeof runner bean plants enhanced outgrowth of lateral buds onlywhen internode stumps were no longer elongating. Applied toelongating internodes of slightly younger plants, ABA causesinhibition of bud outgrowth. Together with 10–4 M indol-3-ylacetic acid (IAA), ABA stimulated internode elongation and interactedadditively in the inhibition of bud outgrowth. A mixture of10–5 M ABA and 10–6 M gibberellic acid (GA3 ) causedsimilar effects on internode growth as IAA + ABA, but was mutuallyantagonistic in effect on growth of the lateral buds. Indole-3-acetic acid (IAA) and 2-(indol-3-ylmethyl)indol-3-yl acetic acid were identified in lipid extracts of Sulfolobus acidocaldarius; they occurred at concentrations of 0.57 and 0.59 mumol/g (dry weight), respectively. The amount of IAA found in these cells is more than a thousand times greater than that found in a typical extract of a plant in which IAA serves as a plant growth hormone. Neither of these compounds was detected in the other archaebacteria that were analyzed; these included Sulfolobus sulfataricus, Halobacterium salinarium, and several strains of methanogenic bacteria. This is the first report of the natural occurrence of 2-(indol-3-ylmethyl)indol-3-yl acetic acid.  相似文献

Two properties of phytotropins, their ability to bind to 1-N-naphthylphthalamic acid (NPA) receptors located on microsomal vesicles isolated from Cucurbita pepo L. hypocotyls, and to stimulate auxin (indol-3-yl acetic acid, IAA) accumulation into such vesicles by blocking its efflux from them, were assessed in double labelling experiments using [2,3,4,5-3H]1-N-naphthylphthalamic acid and 3-indolyl-[2-14C]acetic acid. Two sites of differing affinities and activities on IAA accumulation were found. 1-N-Naphthylphthalamic acid was found to have high affinity (KD at 10-8mol·l-1) for one site and low affinity (KD at 10-6 mol·l-1) for the other, whereas 2-(1-pyrenoyl)benzoic acid displaced NPA with high efficiency (KD below 10-8 mol·l-1) from both sites. Other phytotropins had intermediate affinities for either site. The substrate for the abscisic acid (ABA) carrier in Phaseoluscoccineus L. suspension-cultured cells is shown to be the (S)ABAenantiomer, Km = 1?0 mmol m–3. The methyl (MeABA) andphenyl (PheABA) esters of ABA inhibit carrier-mediated uptakeof ABA with half-maximal inhibition achieved at about 7?0 mmolm–3 and 10 mmol m–3 respectively: with (S)MeABAthis value is decreased to about 2?0 mmol m–3. There isno demethylation of radioactive MeABA by the cells during 5min incubations. Although MeABA reversibly inhibits the ABAcarrier, it is not a transport substrate: association of radioactiveMeABA with living cells is unaffected by non-radioactive MeABAor ABA and, by comparison with frozen-and-thawed cells, it isshown that the radioactivity remains extracellular. It is proposedthat MeABA binds to the carrier to form an abortive complexthat is not translocated. The terpenoid ABA analogue LAB 144143also inhibits carrier-mediated ABA uptake. At concentrationsup to about 20 mmol m–3 - and ß-ionone specificallyinhibit the ABA carrier with the half-maximal effect at about0?6 mmol m–3 ß-ionone. However, at higher iononeconcentrations, the uptake of ABA, indol-3-yl acetic acid andof 5,5-dimethyloxazolidine-2,4-dione (DMO) are all stimulated:this may reflect general permeabilization of the membrane toweak acids by ionone. Effects of three phytohormones (IAA1, GA3 and kinetin) on grain-fillingand the pattern of 32P translocation from individual leavesto grains were studied at intervals of 7 days during the progressof reproductive development of rice (Oryza saliva L. cv. Jaya).The plants were sprayed with 100 µg ml–1 aqueoussolutions of the hormones at 100 days, when the plants wereentering the reproductive stage. Kinetin produced a pronouncedeffect on grain-filling as well as on 32P mobilization fromindividual leaf to grains and increased yield, possibly by increasingleaf longevity. GA3 and IAA also increased the grain-fillingand 32P mobilization significantly over control but the effectswere less marked than those of kinetin. The effect of continuous exposure to indol-3-yl acetic acid(IAA) on primordium initiation and their subsequent emergenceas lateral roots was determined in excised and attached rootsof Pisum sativum. IAA was found to stimulate the number of primordiainitiated per centimetre of attached or excised primary. Similarly,lateral emergence in terms of the number produced per centimetreof primary was promoted in the presence of IAA. This stimulationof lateral emergence even took place in excised roots whichwere 1 cm in length at the onset of culture and which neverproduced secondary roots over a 6-d culture period when grownin the absence of auxin. These effects of IAA on lateral rootdevelopment have been considered in relation to the concurrentchanges which take place in proliferative activity in the apicalmeristem of the primary root during exposure to auxin. The hypothesis that indol-3yl-acetic acid (IAA) stimulates solutemobilization in stems by increasing metabolite utilization (i.e.sink strength) and/or sieve tube unloading mechanisms was investigatedusing seedlings of Phaseolus vulgaris L. Sink strength was assessedby obtaining estimates of rates of growth, 14C incorporationinto protein, sucrose metabolism, and sucrose uptake. The dataobtained suggest that, whilst enhancing assimilate mobilization,IAA had no short term effect on the sink strength of the treatedstumps. In several experiments involving long distance transportof 14C-assimilates, the magnitude of auxin-promoted transportwas found to be insensitive to potential changes in sink strength.Furthermore, 14C transport profiles demonstrated that the siteof hormone action was not confined to its point of application.  相似文献

The radish varieties Cherry Belle and Long White Icicle wereused to investigate the role of the shoot and the effects ofsynthetic growth promoters in controlling cambial activity inthe seedling axis. Development was compared in excised roots, roots with hypocotylsattached and intact seedlings cultured aseptically on a nutrientmedium. No cambial divisions were seen in isolated radicleswhich had been cultured for ten days following excision butretention of hypocotyl tissue or the entire shoot resulted incambial activity and the production of secondary vascular tissues.Enriching the culture medium by raising the sucrose conantrationto 8% and including 10–5 M indol-3yl acetic acid (IAA)5 x 10–6 M 6-benzylaminopurine (BA) and 5 x 10–4Minositol enhanced root thickening, increasing stele and xylemdiameters in roots cultured both with and without attached shoottissues. The effects of shoot tissues and enrichment of themedium were additive. The effects of auxin, cytokinin and gibberellin (gibberellicacid, GA2) were also studied on daxpitated seedlings. BA wasmuch more effective in inducing cell divisions in the hypocotylthan either IAA or GA supplied separately but a mixture of IAA+GAalso produced clearly defined arcs of cambial tissue. Littlesecondary tissue had been produced after seven days' treatment,and stelar enlargement was due to the development of a cambialzone and cell expansion in the primary tissues. Only minor differencesin response were observed between the two varieties. No stimulation of storage organ development occurred when auxin,cytokinin or inositol was inwrporated into the inorganic culturesolution in which plants of Cherry Belle were grown. To study the influence of morphogenetic gradients on vasculardifferentiation patterns, a new technique was developed whichallows different substances to be applied at opposite ends ofa tissue block. It yielded information on the mobility of particularmorphogens and on the dependence of callus formation and trachearyelement differentiation on their presence. Application of indol-3ylacetic acid (1AA) (10 mg l–1), zeatin (0.1 mg l–1)and sucrose (3 per cent, w/v) in various combinations to theends of cylindrical explants of lettuce pith (Lactuca sativaL.) showed that (a) callus formation was stimulated by IAA,whereas induction of tracheary elements required both IAA andzeatin; (b) callus was confined to a few millimetres at theends of the explants, and tracheary elements occurred mainlywithin the callus; (c) sucrose or its metabolic products diffusedthe 10 mm length of the explants, while IAA and zeatin wereeffective only close to the application site; and (d) some callusand tracheary elements formed when no sucrose was applied, butboth increased with sucrose application, though inhibition oftracheary elements formation occurred with high sucrose concentrations. differentiation, pith explant, tissue culture, xylogenesis, indol-3yl acetic acid, sucrose, zeatin, lettuce, Lactuca sativa  相似文献

Pretreatment of 2?0 mm segments of etiolated zucchini (Cucurbitapepo L.) hypocotyl with cycloheximide (CH) or 2-(4-methyl-2,6-dinitroanilino)-N-methylpropionamide(MDMP) eliminated the stimulation by N-1-naphthylphthalamicacid (NPA) of net uptake of [1-14C]indol-3yl-acetic acid ([1-14C]IAA),but had relatively little effect on the net uptake of IAA inthe absence of NPA. The efflux of [1-14C]IAA from preloadedsegments was not substantially affected by inhibitor pretreatmentin the absence of NPA, but CH pretreatment significantly inhibitedthe reduction of efflux caused by NPA. Pretreatment with CHor MDMP did not affect net uptake by segments of the pH probe[2-14C]5,5-dimethyl-oxazolidine-2,4-dione ([2-14C]DMO), or thenet uptake of [14C]-labelled 3-O-methylglucose ([14C]3-0-MeGlu),suggesting that neither inhibitor affected intracellular pHor the general function of proton symporters in the plasma membrane.Both compounds reduced the incorporation of label from [35S]methionineinto trichloroacetic acid (TCA)-insoluble fractions of zucchinitissue, confirming their inhibitory effect on protein synthesis. The steady-state association of [3H]IAA with microsomal vesiclesprepared from zucchini hypocotyl tissue was enhanced by theinclusion of NPA in the uptake medium. The stimulation by NPAof [3H]IAA association with microsomes was substantially reducedwhen the tissue was pretreated with CH. However, CH pretreatmentdid not affect the level of high affinity NPA binding to themembranes indicating that treatments did not result in lossof NPA receptors. It is suggested that the auxin transport site on the effluxcarrier system and the receptor site for NPA may reside on separateproteins linked by a third, rapidly turned-over, transducingprotein. 14C from indol-3-yl-(acetic acid-2-14C) (IAA-14C) was transportedin a weak but definitely polar manner through segments of youngand matured regions of pea roots. Greater quantities of 14C-labelledmaterial moved acropetally than basipetally. Up to 70 per centof radioactivity originally present in donor agar blocks wastaken up by the root segments, but only approximately 2 to 3per cent of this emerged into the receiver agar blocks. Anydifferences in uptake, transport, or binding of auxin were veryslight in the three regions of root studied. The IAA-14C wasmetabolized during passage through the root segments, yieldingtwo principal radioactive products. The identities of thesewere not determined, but they appeared to have auxin activityand may be formed spontaneously, but more slowly, in solutionsof IAA-14C. The permeability (P) of a lipophilic cation, triphenylmethylphosphonium(TPMP+) which is frequently used as a membrane potential probe,has been measured in Chara australis (Charophyceae). PTPMP+across biological membranes is usually thought to be very highbut this is not the case across the plasmalemma of Chara. Thepermeability of TPMP+ across the plasmalemma was found to betypical of inorganic cations, about 1.0 nm s–1. Estimateswere made of the permeability of lipophilic cations across someother cell membranes, based on previously published work. Thepermeability of TPMP+ across the plasma membranes of the redalga, Griffithsia monilis and the blue-green alga, Anabaenavariabilis was about 2–5 nm s–1. The permeabilityof TPMP+ across the plasma membranes of eukaryotes and prokaryotesappears to be similar. The permeability of lipophilic cationsacross the cristae of isolated mitochondria are exceptionallyhigh, about 170 nm s–1. TPMP+ did not behave as a thiamineanalogue in Chara, unlike in the case of yeast. The means ofentry of TPMP+ into the Chara cell, driven by the electrochemicalgradient across the plasmalemma, has not been identified. Thepresence of a second lipophilic cation probe, DDA+ (dibenzyldimethylammonium),caused a decrease in the uptake flux of TPMP+; this suggeststhat the two lipophilic cations compete for the same site atthe surface of the plasmalemma. The effect of morphactin (methyl-2-chloro-9-hydroxyfluorene-9-carboxylate) on the content of several plant growth substances in bean roots was determined. Beans (Phaseolus vulgaris L. cv. Spartan) were soaked in aqueous solutions of morphactin, 1 x 10-4, 1 x 10-5, and 1 x 10-6M and grown in moist vermiculite. As controls were used beans grown in water-moistened vermiculite either intact or having the root tips removed (decapped). The roots, morphactin-treated, controls, and the decapped ones were analyzed for indol-3-yl acetic acid (IAA), indol-3-yl acrylic acid (IAcA), indol-3-yl pyruvic acid (IPyA), indol-3-yl lactic acid (1LA), abscisic acid (ABA), gibberellins GA1, GA3, GA4, and GA9 using gas-liquid chromatographic methods. Morphactin, while affecting the geotropical responses, changed also the growth substance content of roots. IAA, ABA, GA1, and GA9 contents decreased, IPyA, IAeA, GA3, and GA4 contents were not affected and ILA content increased slightly with increasing dosages of morphactin. Growth substance pattern of decapped roots resembled that of the roots treated with the highest dose, 1 x 10-4M, of morphactin.  相似文献

The permeability of the plasmalemma of Chlamydomonas reinhardtiicells was increased by treatment with poly-L-lysine or dimethylsulphoxideas indicated by 3-phosphoglyceric acid dependent O2 evolution.These treatments decreased the ability of the cells to accumulateinorganic carbon internally and hence their photosynthetic affinityfor inorganic carbon in the medium. With saturating light andinorganic carbon, the photosynthetic rate was less affectedby the poly-L-lysine and dimethylsulphoxide treatments. Thusthe poly-L-lysine and dimethylsulphoxide did not alter the activityof the chloroplasts but rather made the intracellular inorganiccarbon pool more freely exchangeable with the medium. It isconcluded that the transporting system for inorganic carbonis located at the plasmalemma. Treatment with Diamox, an inhibitor of carbonic anhydrase, didnot affect photosynthetic rate and accumulation of inorganiccarbon when CO2 was supplied but strongly inhibited both parameterswhen HCO3 was supplied. In a mutant of Chlamydomonasreinhardtii lacking a cell wall, carbonic anhydrase leaks tothe medium and uptake of inorganic carbon is much faster whenCO2 is supplied than when HCO3 is supplied. The initiation of lateral root primordia and their subsequentemergence as secondary roots have been examined in attachedand excised roots of Zea mays grown in the presence or absenceof indol-3-yl acetic acid (IAA). Exposure to IAA enhanced anlageinception in both batches of roots. In the attached roots, theIAA-induced stimulation of primordium initiation was followedby a similar increase in lateral emergence. IAA treatment, however,had no effect on the number of laterals produced, per centimetreof root, in the excised primaries. Thus, exposure to IAA didnot directly enhance lateral emergence in the attached rootsnor did it stimulate such emergence in the excised ones. Nocorrelation was found between proliferative activity in themeristem at the apex of the primary or the rate of root elongationon the one hand, and either the number of primordia initiated,or the number of laterals produced, per centimetre of primary,on the other. The effects of calmodulin antagonists on indol-3-yl acetic acid (IAA) uptake by Cucurbita pepo L. hypocotyl segments have been studied. Chlorpromazine (CP) and trifluoperazine (TFP) stimulate uptake of IAA, but not in the presence of 1-naphthylphthalamic acid (NPA), which itself stimulates net IAA uptake by inhibiting the IAA efflux carrier. Maximum stimulation of IAA uptake is observed at 300 μM CP, at both pH 4.0 and 5.0, with a decrease below this maximum at 1 mM CP. CP also stimulates uptakes of abscisic acid (ABA) and 5,5-dimethyl-oxazolidine-2,4-dione (DMO) with the same concentration-dependence. Indirect evidence suggests that enhanced permeability to lipophilic molecules rather than cytoplasmic alkalinification is involved. At submaximal stimulatory concentrations, CP and NPA have apparently additive effects. However, the carrier antagonism by CP is less specific than that of NPA since it also renders the IAA uptake carrier undetectable. The local anaesthetics, dibucaine and tetracaine, and the calcium entry blocker, flunarizine, had similar effects to CP on uptake. However, only CP increased the binding of IAA (but not of ABA or DMO) to frozen-and-thawed segments. It is suggested that CP may increase membrane permeability to apolar molecules, probably non-specifically, and also interfere with auxin carrier action by hydrophobic interaction with phospholipids or membrane proteins.  相似文献

Carbohydrate accumulation in young, fully expanded leaves ofCitrus sinensis L. Osbeck is affected by the presence of thefruitlet on the shoot. Previous work gave evidence that gibberellinsmay be involved in this 'fruit effect'. In the present workwe have studied the effect of gibberellic acid (GA3) on 14C-sucroseuptake by leaf discs and whether its action could be due toa modulation of the plasma membrane ATPase, which maintainsthe H+ gradient that drives H+/sucrose co-transport. The effect of GA3 on 14C-sucrose uptake depended on the osmolarityof the assay medium. At 300 mOsm a reduction in the uptake ratewas observed. The inhibitory effect of the hormone disappearedafter preincubating the leaf discs with para-chloromercuri-phenylsulphonicacid (PCMPS), a sulphydril binding inhibitor. ATPase activityof isolated plasma membrane vesicles was inhibited by IAA treatments,while GA3 or ABA did not affect this enzyme, even after a 3h preincubation period. However, in the absence of a surfactantin the assay medium, GA3, together with turgor pressure, modulatedplasma membrane ATPase activity, possibly through modificationsof membrane permeability.
