链霉菌属是绝大多数已知抗生素和一些重要活性物质的产生菌,它对高等植物是否具有化感作用尚缺乏研究. 从土壤中分离获得7个放线菌菌株,研究其对植物幼苗生长的抑制作用.结果表明: 链霉菌6803菌株在固体和液体发酵培养时均强烈抑制油菜根和稗草根的生长, 其液体发酵液的5倍稀释液对油菜和稗草苗生长的抑制分别达到60.7%和61.3%. 常规形态与生理生化试验表明,该菌株属于链霉菌属金色类群; 16S rRNA基因序列分析表明,该菌株为沙场链霉菌,基因序列相似性达到99.28%. 此菌株经过80~100 s紫外线照射筛选出的正突变菌株UV8024和UV100-2的10倍稀释发酵液对油菜幼苗生长的抑制作用比原始菌株分别提高了37.5%和38.1%, 经过1%硫酸二乙酯诱变50 min筛选出的正突变菌株D507的10倍稀释发酵液对油菜幼苗生长的抑制作用比原始菌株提高了29.8%. 链霉菌6803菌株对高等植物具有化感作用, 并可通过诱变育种提高其化感潜力.  相似文献

西双版纳不同演替状态热带次生林土壤节肢动物群落特征   总被引:7,自引:0,他引:7  
采用样地调查法,对西双版纳4种不同演替状况热带次生林:中平树(Macavanga denticulate)、崖豆藤(Millettia laptobotrya)、野芭蕉(Musa acuminata)与黄竹(Dendrocalamus membranaceae)林的土壤节肢动物群落结构与季节变化进行了研究.结果表明,4类不同演替状况次生林土壤节肢动物群落在数量优势类群组成上无较大差异,蜱螨目为所有4类林地的突出优势类群,膜翅目、弹尾目和鞘翅目在不同林地中分别为不同数量等级的次优势类群,而在常见和稀有类群的组成上,各林地表现出较大的差异.土壤节肢动物类群数、个体数和DG多样性指数以正向演替的崖豆藤林最高,偏途演替的黄竹林最低,但中平树、崖豆藤和野芭蕉林的差异不大.4类林地土壤节肢动物类群数和个体数的垂直分布分别以凋落物层和土壤表层(0~5cm)最高,其它各层分布因林地不同各异,并存在明显的季节差异.各林地土壤节肢动物个体数和类群数的季节消长总体表现出干季和雨季初期与末期高于雨量最大的雨季中期,由于不同林地植被结构、凋落物数量和质量以及土壤水热状况不同,其季节变化显现样地差异,除了受林地降水量和温度变化影响外,食物的丰欠和栖息场所的干扰状况也有重要的影响作用.  相似文献

抗生素LA5具有开发成抗真菌农用抗生素的广阔前景,但由于该菌株的野生型菌株发酵效价低,不能满足工业化开发的要求。通过以链霉菌LA5为出发菌株,采用紫外线(UV)、微波、亚硝基胍(NTG)、紫外线 亚硝基胍、亚硝基胍 紫外线等方法进行诱变筛选,结果表明,以紫外线照射80 s 亚硝基胍处理80 min的诱变的正突变率最高,为30%,其他依次为亚硝基胍处理80 min 紫外线照射40 s诱变和亚硝基胍处理60min 紫外线照射120 s诱变,正突变率均为26%。然后依次使用亚硝基胍处理80 min 紫外线照射80 s、亚硝基胍处理80 min 紫外线照射40 s、亚硝基胍处理60 min 紫外线照射120 s进行第2、第3、第4轮复合诱变筛选,最后选育出突变菌株U8-N8A-196,其产抗生素能力比链霉菌LA5出发菌株提高了34.5%,极显著高于LA5菌株。  相似文献

对菊科植物化感作用的研究进展进行了综述。菊科植物中至少有 39个属存在化感作用 ,特别是一枝黄花属、向日葵属、胜红蓟属、银胶菊属、蒿属植物等有较多的研究报道。鉴定出的化感物质多为萜类、聚乙炔类、酚类、有机酸类等 ,这些化感物质对多种受体植物表现出程度不同的抑制或促进的效应。其化感作用机理表现在破坏受体膜系统的稳定性及水分平衡关系、抑制氧化磷酸化、促进或阻滞叶绿素的合成、影响矿质元素的吸收利用等。并对菊科植物化感物质在植物生长调节剂、天然除草剂和生物杀虫剂 ,或人工合成除草剂和杀虫剂上应用的前景进行了探讨。本文显示菊科植物的化感作用将在控制外来恶性杂草及维护生态平衡上扮演重要的角色。在当前菊科植物化感作用研究的基础上 ,提出了进一步研究的 6个方向 :(1)化感物质的生物合成途径与关键酶的特性研究 ;(2 )具化感潜势物种资源的调查评价及利用研究 ;(3)化感作用在自然生态系统中的演变规律 ;(4 )菊科重要作物自毒的生化机制及克服途径 ;(5 )具应用前景的菊科植物化感关键酶的基因克隆和转基因 ,并对受体植物基因的表达与调控进行研究 ;(6 )化感作用在可持续发展农业应用上的研究与开发 ,特别是作为天然除草剂及杀虫剂  相似文献

以链霉菌702-20为出发菌株,经HNO2诱变处理,获得高产突变株。实验结果表明:HNO2处理20 m in对菌株的致死率可达83.10%,突变率高达14.13%,经过摇瓶筛选获得高产突变株20-29-148,产链霉素能力达到1.404 mg/mL,比出发菌株提高了37.65%。经传代培养考察,该突变菌株具有良好的遗传稳定性。  相似文献

Root exudates have a key role in communication between plants and microbes in the rhizosphere. Fusarium wilt of watermelon, caused by Fusarium oxysporum f. sp. niveum (Fusarium oxysporum), drastically reduces watermelon yields in continuous cultivation systems, but it can be significantly alleviated using watermelon/aerobic rice intercropping system as shown by the research carried out in this laboratory. It is important to evaluate the interaction between root exudates from the two crops and the pathogen and thus to clarify the mechanism of disease suppressiveness in the intercropping system. The effects of phenolic acids, sugars and free amino acids in root exudates from watermelon (REW) and rice (RER) on the growth of Fusarium oxysporum were studied. The results obtained are listed as follows: (1) REW significantly increased spore germination and sporulation, whereas RER had inhibitory effects on those two parameters. (2) HPLC analysis showed that salicylic acid, p-hydroxybenzoic acid and phthalic acid were identified in exudates from both plants, but p-coumaric acid was only detected in rice and ferulic acid only in watermelon. Moreover, of the total rice exudates a high proportion (37.9 %) of p-coumaric acid was detected and the total amount of phenolic acids was 1.4-fold as high as that in watermelon. (3) Considerable differences in the components and contents of both sugars and amino acids were found between REW and RER exudates. (4) Exogenously applied alanine (Ala) increased spore germination and sporulation. In contrast, addition of exogenous p-coumaric acid reduced spore germination and sporulation, relative to controls. It was concluded that the rice root exudates had anti-fungal properties while that from watermelon promoted pathogen growth. This discovery provided a scientific basis for practicing watermelon/aerobic rice intercropping to control Fusarium wilt in watermelon.  相似文献

研究了五种高等水生植物(黑藻、金鱼藻、水花生、茭白、空心菜)对蛋白核小球藻、斜生栅藻生长的抑制作用。试验结果显示,各种受试植物均有不同程度的克藻效果,金鱼藻与黑藻的克藻作用最强,水花生的作用次之。克藻效果以受试植物与藻共培养最佳,其次为连续添加种植水,表明在植物的生长过程中其化感物质是连续释放且能迅速降解的,而黑藻干物质的浸提液所具有的克藻效应说明有些化感物质对一定温度和光照是较稳定的。  相似文献

采用人工气候箱培养试验,初步研究了入侵杂草加拿大一枝黄花对本地植物的化感作用.测定了根系和根状茎的不同浓度浸提液对白三叶发芽率和幼苗生长的影响;不同播种密度下根系1:60提取浓度对白三叶种子萌发和幼苗生长的影响;根状茎1:60提取浓度对白三叶、红三叶、天蓝苜蓿、黑麦草、北美车前、鸡眼草、苇状羊茅、胜红蓟、马齿苋、碱蓬和刺苋的种子萌发和幼苗生长的影响.结果表明,加拿大一枝黄花根系和根状茎的化感抑制作用均随浓度的升高而增强,且根状茎提取物的抑制效应高于根系提取物的效应;在1:60低浓度根系浸提液作用下,不同密度的白三叶幼苗萌发率和幼根、幼芽长度差异不显著;1:60低浓度根状茎提取物对11个物种的种子萌发均有显著抑制作用,但对不同物种的抑制程度有差异,对禾本科植物的抑制作用大于非禾本科和豆科;1:60低浓度根状茎提取物对10个物种的幼根影响与对萌发率的影响相似,但在对幼芽的影响上,仅对鸡眼草、刺苋和苇状羊茅起抑制作用,而对其他植物无显著抑制(黑麦草、北美车前、胜红蓟、马齿苋和刺苋)或起促进作用(白三叶、红三叶、天蓝苜蓿).  相似文献

Sucrose-phosphate synthase (SPS) from the cyanobacterium Synechocystis sp. PCC 6803 lacks all of the Ser residues known to be involved in the regulation of higher plant SPS by protein phosphorylation. The Synechocystis SPS is also not allosterically regulated by glucose 6-phosphate or orthophosphate. To investigate the effects of expressing a potentially unregulated SPS in plants, the Synechocystis sps gene was introduced into tobacco, rice and tomato under the control of constitutive promoters. The Synechocystis SPS protein was expressed at a high level in the plants, which should have been sufficient to increase overall SPS activity 2-8-fold in the leaves. However, SPS activities and carbon partitioning in leaves from transgenic and wild-type plants were not significantly different. The maximal light-saturated rates of photosynthesis in leaves from tomato plants expressing the Synechocystis SPS were the same as those from wild-type plants. Tomato plants expressing the maize SPS showed 2-3-fold increases in SPS activity, increased partitioning of photoassimilate to sucrose and up to 58% higher maximal rates of photosynthesis. To investigate the apparent inactivity of the Synechocystis SPS the enzyme was purified from transgenic tobacco and rice plants. Surprisingly, the purified enzyme was found to have full catalytic activity. It is proposed that some other protein in plant cells binds to the Synechocystis SPS resulting in inhibition of the enzyme.  相似文献

The taxonomic positions of soil isolates known as Streptomyces groups A, B and C were clarified. Comparative 16S rDNA sequence studies indicated that representatives of all three taxa formed distinct phyletic lines within the Streptomyces tree though the group A strains were shown to be related to Streptomyces griseus and associated validly described species. The taxonomic integrity of all three groups was highlighted by DNA:DNA relatedness and ribotype data though the group A strains encompassed a higher degree of genetic variation than the group B and C strains. In light of these and earlier phenotypic data it is proposed that Streptomyces groups A, B and C be given species status as Streptomyces sanglieri sp. nov., Streptomyces aureus sp. nov. and Streptomyces laceyi sp. nov., respectively. 采用模拟的方式,利用生物测定和室内培养的方法,研究了不同浓度的己二酸二异丁酯对辣椒种子萌发、幼苗生长及辣椒枯萎菌的化感效应.结果表明:己二酸二异丁酯对辣椒种子萌发和幼苗生长具有低浓度促进、高浓度抑制的化感效应;己二酸二异丁酯对辣椒和茄子种子的化感效应存在较大差异,在低浓度时对辣椒种子萌发和幼苗生长的促进作用强于茄子种子,而高浓度时对辣椒种子萌发和幼苗生长的抑制强度弱于茄子种子;各浓度己二酸二异丁酯对辣椒枯萎菌菌丝生长有极显著的抑制作用,且作用强度随着浓度增加而增强,当浓度为1 mnol·L~(-1)时抑制作用最强,抑制率为10.75%;在田间抗病性调查期间内各浓度处理均提高了辣椒幼苗的抗病性,其中以1 mmol·L~(-1)处理抗病性最好,病情指数比对照降低了49.88%.
Abstract:
By the methods of laboratory incubation and bioassay, a simulation test was conducted to study the allelopathic effects of different concentration diisobutyl adipate on pepper seed germi-nation and its seedling growth and Fusarium oxysporum f. sp. vasinfectum. For the pepper seed germination and seedling growth, diisobutyl adipate exhibited allelopathic promotion at low con-centration, but allelopathic inhibition at high concentration. There existed greater differences in the allelopathic effects of diisobutyl adipate on the seed germination and seedling growth of pepper and eggplant. The promotion effects of low concentration diisobutyl adipate on pepper were stron-ger than those on eggplant, while the inhibition effects of high concentration diisobutyl adipate on pepper were weaker than those on eggplant. All test concentration diisobutyl adipate had signifi-cant inhibition effects on the mycelium growth of Fusarium oxysporum f. sp. vasinfectum, and the effects increased with increasing diisobutyl adipate concentration, being the strongest (an inhibi-tory rate of 10. 75%) at 1 mnol·L~(-1). Field text indicated that diisobutyl adipate at all test con-centrations enhanced the disease resistance of pepper seedlings. The best effect was observed at 1 mmol·L~(-1) of diisobutyl adipate, with the disease index decreased by 49.88%, compared to the control.  相似文献

During the past years ragweed has been coming to the forefront of interest in Hungary and in other European countries as well because its serious health risk. Results of the 5th National Weed Survey has proven that ragweed is the most important weed species on Hungarian field lands, its coverage shows a rising tendency in cereals moreover it not only occurs in cultivated plants. Allelopathic effects of aqueous extracts derived from different parts of ragweed plants (air dried leafy stems, seeds) on the germination and growth of other cultivated plants [maize (Zea mays L.), winter wheat (Triticum aestivum L.), rye (Secale cereale L.), oat (Avena sativa L.)] were studied. The extracts made for the trials were prepared with distilled water. Petri dishes were used for the germination experiments and distilled water was used as a control treatment. The seven days long experiment was carried out within a Binder-type thermostat under dark conditions. The germination percentage was checked in every two days and the growth of sprouts was evaluated after a week counting the germinated seeds and measuring the length of the radicle and plumule. The measured data were statistically analysed and the effect of extracts on germinating and length of sprouts were assessed.  相似文献

Three cryptic plasmids have been isolated from cyanobacterium Synechocystis sp. 6803::pSS2 (1.4 Md), pSS3 (36 Md), pSS4 (60 Md). Plasmid DNA was isolated in Cs-Cl-EB density gradient and analyzed by gel electrophoresis and electron microscopy by gel electrophoresis and electron microscopy techniques. The restriction map is constructed for plasmid pSS2 having the cleavage sites for Sau3a, HincII, HindIII, MspI restriction endonucleases. The plasmid may be used to construct the recombinant vector DNAs capable of autonomous replication in cyanobacterium Synechocystis sp. 6803. cells.  相似文献
