We describe the sperm ultrastructure of five species of Colostethus from Brazil. The general structures of the spermatozoa (acrosomal complex and flagellar apparatus) were similar to species within the Bufonoidea. Colostethus brunneus, Colostethus sp. (aff. trilineatus), Colostethus nidicola and Colostethus sp. had biflagellated spermatozoa, whereas the spermatozoa of Colostethus stepheni had a single flagellum, a characteristic previously unknown for this genus. In addition, the spermatozoa of this species showed some mitochondria within the thick, undulating membrane. These characteristics are apparently common to dendrobatid species that have a single flagellum. Based on these findings, we tentatively consider biflagellarity to be an apomorphic condition in the Dendrobatidae. In addition, the results described here do not support the proposed regrouping of the ‘brunneus’ and ‘alagoanus’ groups in a monophyletic ‘trilineatus’ group.  相似文献

The sperm ultrastructure of Epipedobates trivittatus and E. hahneli is described. The spermatological characteristics are identical in both species, except for the extension of the acrosomal vesicle. The structure of the acrosomal complex is shared with species of the Bufonoidea neobatrachian lineage, which differs from the condition observed in species included in the ranoid lineage. The phylogenetic significance of the mitochondrial collar, also seen in Eubufonoidea groups, is still unclear. Also the significance of the alternative mitochondrial arrangement seen here is unclear. The perpendicular arrangement of the centrioles is also shared with the Bufonoidea and is considered a synapomorphic condition. An expanded undulating membrane and the absence of a juxtaxonemal fibre appear to be apomorphic characteristics of dendrobatid species, as also observed (homoplastically) for species of Litoria and Cyclorana. However, the notable expansion of the undulating membrane appears to be an autapomorphy of Dendrobatidae frogs. The ultrastructural data presented here do not support the proposed placement of E. trivittatus in a separate genus.  相似文献

We report on the international trade in South American poison arrow frogs (Dendrobatidae) in the period 2004–2008, and focus on the role of Asian countries. All species of dendrobatid frogs are included in Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES), regulating all commercial trade in these species. Based on data compiled in the WCMC CITES database, we establish that >63,000 dendrobatid frogs (of 32 species) were traded internationally. For 21 species the majority of individuals were reported as captive-bred. A quarter to a fifth of the commercial trade in dendrobatid frogs in terms of volume is destined for Asian markets (mainly Japan, Thailand and Taiwan, Province of China). Kazakhstan, the main supplier for the Thai market, is reported as a source country for 16 species, all captive-bred. We found large discrepancies between the reported export of dendrobatid frogs from Kazakhstan—none—and imports reported by Thailand as coming from Kazakhstan (>2,500 individuals). A significant part of the trade flow goes via Lebanon, a non-CITES Party. We urge the CITES Management Authorities of the countries involved to investigate the trade in dendrobatid frogs to ensure it does not violate the rules and intentions of CITES.  相似文献

Scanning and transmission electron microscopy and fluorochrome staining with DAPI were used to study the spermatozoa of Colostethus marchesianus from the type locality and Colostethus sp. (aff. marchesianus) from a different locality. The sperm cell of these species resembles those of most neobatrachian species, except for the presence of two complete flagella, a character previously described only for four unrelated anuran species belonging to the families Leptodactylidae, Rhacophoridae and Dendrobatidae. Although very similar, the spermatozoa described here showed slight differences in the length of their nucleus, in the extent of the subacrosomal cone over the nucleus, and in the curvature of the axial fiber seen in cross section. The absence of a juxtaxonemal fiber and the presence of a comma shaped axial fiber seem to be common characteristics of dendrobatid spermatozoa. The minor differences found in our study may indicate that the taxa studied here are distinct species. The evolutionary significance of spermatozoa biflagellarity is still unclear, although the presence of this trait in unrelated groups suggests independent origins.  相似文献

Dendrobatidae (dart‐poison frogs) exhibit some of the most complex spatial behaviors among amphibians, such as territoriality and tadpole transport from terrestrial clutches to widely distributed deposition sites. In species that exhibit long‐term territoriality, high homing performance after tadpole transport can be assumed, but experimental evidence is lacking, and the underlying orientation mechanisms are unknown. We conducted a field translocation experiment to test whether male Allobates femoralis, a dendrobatid frog with paternal extra‐territorial tadpole transport, are capable of homing after experimental removal, as well as to quantify homing success and speed. Translocated individuals showed a very high homing success for distances up to 200 m and successfully returned from up to 400 m. We discuss the potential orientation mechanisms involved and selective forces that could have shaped this strong homing ability.  相似文献

Few ultrastructural studies have been performed on members of the Dendrobatidae, although such investigations can be useful for the understanding of reproductive patterns, as a diagnostic method for males in breeding programs for endangered amphibians and for phylogenetic analysis. The sperm ultrastructure of the Green Poison Frog, Dendrobates auratus, from Panama is described following induced spermiation in living animals. To date only testicular spermatozoa in other dendrobatid frogs have been analysed. Moreover, an electron microscopic preparation method (transmission and scanning electron microscopy) for dendrobatid sperm cells in low concentration is presented. Sperm cells from stimulated frogs (100 IU human chorionic gonadotropin, hCG, twice at an interval of 1h) were recovered via cloaca lavage using 600 microl isotonic phosphate-free amphibian saline (IPS). Centrifuged flushings (5 min, 173 x g) were deposited on microscopic slides. Adherent spermatozoa were treated with Karnovsky fixative (overnight, 4 degrees C). After postfixation (2h, 1% osmium tetroxide), samples were dehydrated in series of ascending acetones (30-100%). For transmission electron microscopy sperm cells were encapsulated using Epon and 1.5% 2,4,6-tris(dimethylaminomethyl)phenol (DMP 30). Ultrathin sections (70 nm) were cut and stained with uranyl acetate (30 min) and lead citrate (5 min). Sperm cells are filiform with a 21.1+/-2.7 microm long and arcuated head and a single tail (35.0+/-4.2 microm length). Their acrosomal complex is located at the anterior portion of the head and consists of the acrosomal vesicle which has low electron density, and the subjacent electron-dense subacrosomal cone. In transverse section, the nucleus is circular (1.9+/-0.2 microm diameter) and conical in longitudinal section. It is surrounded by several groups of mitochondria. The chromatin is highly condensed and electron-dense but shows numerous electron-lucent inclusions. A short midpiece has a mitochondrial collar with a proximal and a distal centriole. The latter gives rise to the axoneme which alone forms the flagellum. The sperm ultrastructure of D. auratus differs from that of other Dendrobatidae because of the absence of a nuclear space and the absence of the undulating membrane associated with an axial fibre. This tail conformation is found in the Ranoidea but not in the Bufonoidea. These results show that the spermatozoa of D. auratus are the first within the Dendrobatidae without accessory tail structures. Methods of using sperm samples from hormonal treated frogs for ultrastructural studies is not only reasonable to examine e.g. amphibian phylogeny without killing frogs threatened with extinction but allows investigations in the field of assisted reproduction and male fertility for example in conservation programs for endangered amphibians.  相似文献

Next‐generation sequencing continues to revolutionize biodiversity studies by generating unprecedented amounts of DNA sequence data for comparative genomic analysis. However, these data are produced as millions or billions of short reads of variable quality that cannot be directly applied in comparative analyses, creating a demand for methods to facilitate assembly. We optimized an in silico strategy to efficiently reconstruct high‐quality mitochondrial genomes directly from genomic reads. We tested this strategy using sequences from five species of frogs: Hylodes meridionalis (Hylodidae), Hyloxalus yasuni (Dendrobatidae), Pristimantis fenestratus (Craugastoridae), and Melanophryniscus simplex and Rhinella sp. (Bufonidae). These are the first mitogenomes published for these species, the genera Hylodes, Hyloxalus, Pristimantis, Melanophryniscus and Rhinella, and the families Craugastoridae and Hylodidae. Sequences were generated using only half of one lane of a standard Illumina HiqSeq 2000 flow cell, resulting in fewer than eight million reads. We analysed the reads of Hylodes meridionalis using three different assembly strategies: (1) reference‐based (using bowtie2 ); (2) de novo (using abyss , soapdenovo2 and velvet ); and (3) baiting and iterative mapping (using mira and mitobim ). Mitogenomes were assembled exclusively with strategy 3, which we employed to assemble the remaining mitogenomes. Annotations were performed with mitos and confirmed by comparison with published amphibian mitochondria. In most cases, we recovered all 13 coding genes, 22 tRNAs, and two ribosomal subunit genes, with minor gene rearrangements. Our results show that few raw reads can be sufficient to generate high‐quality scaffolds, making any Illumina machine run using genomic multiplex libraries a potential source of data for organelle assemblies as by‐catch.  相似文献

Polystoma sodwanensis n. sp. andMetapolystoma porosissimae n. sp. are described as new species of the Polystomatidae parasitic in the urinary bladder of adultPtychadena porosissima collected in northern Natal, South Africa. Of 26 frogs examined, 11 host individuals were infected with one of the two parasite species, while in five both parasite species occurred. This is the first record of two different genera of polystomatids occurring together in one host species and the first record ofMetapolystoma in South Africa. The exceptional nature of the reported double infection is discussed in terms of host specificity within the Polystomatidae. Spermatozoa ofPocillopora damicornis, Pocillopora elegans (Astrocoeniina, Pocilloporidae) andPavona gigantea (Fungiina, Agariciidae) from the eastern Pacific (Isla del Caño, Costa Rica) were examined using transmission electron microscopy. The hermaphroditic pocilloporidsP. damicornis andP. elegans are spermiomorphologically very similar to hermaphroditic acroporids, being characterized by bullet-shaped nuclei and elongated mitochondria. Such traits have not been found in other families. Thus, the suborder Astrocoeniina, including pocilloporids and acroporids, can clearly be distinguished from other scleractinian suborders. This separation underlines the isolated position of the Astrocoeniina within the order Scleractinia following the evolutionary scheme of Wells. A conical sperm type, known from gonochoric species (sexes separate) from all families except Acroporidae and Pocilloporidae, was found in the gonochoric agariciidPavona gigantea. This supports previous findings that gonochoric corals share a unique and common sperm structure regardless of which family they belong to. However, no gonochoric Astrocoeniina have ever been examined. Hence, the question whether the sperm type common to gonochorists is also represented in Astrocoeniina, which would undermine the ultrastructural distinction of Astrocoeniina and other suborders seen among hermaphrodites, as well as the systematic value of sperm structure within scleractinian corals, remains open.  相似文献

This paper investigates interspecies relationships within the genusAcomys(spiny mice) by analyzing entire mitochondrial cytochromebgene (1141 bp). This gene provides strong phylogenetic signal, as shown by high support of the topology obtained (bootstrap value and RNA support number). The phylogeny is congruent with inferences from allozymes for the species considered. Controversial taxonomy ofAcomys cahirinus, dimidiatus, airensis,andignitusis clarified, with their specific ranks confirmed on the basis of tree topology and nucleotide distances. The zoospores and isogametes ofUlvaria obscura var.blyttii, the isogametes ofMonostroma bullosum, and the anisogametes ofM. grevillei have a flagellar apparatus with counterclockwise absolute orientation and terminal caps, and therefore belong to theUlvophyceae. On the basis of the absence or presence of body scales and the morphologies of certain flagellar apparatus components,Ulvaria obscura var.blyttii is retained in theUlvales, whileM. bullosum, M. grevillei andM. oxyspermum are referred to theUlotrichales. Differences in scale morphology, certain flagellar apparatus components, and early thallus ontogeny support the transfer ofM. oxyspermum to the genusGayralia. Mating structures and their positional relationships within the cell are described from the gametes examined. Among vertebrates, comparable spatial learning abilities have been found in birds, mammals, turtles and fishes, but virtually nothing is known about such abilities in amphibians. Overall, amphibians are the most sedentary vertebrates, but poison frogs (Dendrobatidae) routinely shuttle tadpoles from terrestrial territories to dispersed aquatic deposition sites. We hypothesize that dendrobatid frogs rely on learning for flexible navigation. We tested the role of experience with the local cues for poison frog way-finding by (i) experimentally displacing territorial males of Allobates femoralis over several hundred metres, (ii) using a harmonic direction finder with miniature transponders to track these small frogs, and (iii) using a natural river barrier to separate the translocated frogs from any familiar landmarks. We found that homeward orientation was disrupted by the translocation to the unfamiliar area but frogs translocated over similar distances in their local area showed significant homeward orientation and returned to their territories via a direct path. We suggest that poison frogs rely on spatial learning for way-finding in their local area.  相似文献

The ultrastructure of the type species of the genusDunaliella, D. salina, has been reinvestigated in an attempt to clarify the relationships betweenDunaliella andSpermatozopsis. Dunaliella salina differs in the following ultrastructural characters fromSpermatozopsis (as exemplified byS. similis Preisig etMelkonian): presence of a distinctive surface coat covering the plasmalemma; presence of a prominent pyrenoid (with pairs of thylakoids partially entering the pyrenoid matrix); dictyosomes parabasal; endoplasmic reticulum closely underlying the plasmalemma around most of the cell; contractile vacuoles absent; cell form ovoid to elongated and not spirally twisted; mitochondrial profiles near the flagellar apparatus. Differences in the ultrastructure of the flagellar apparatus: basal body angle more or less fixed; distal connecting fibre cross-striated; system II fibre (rhizoplast) present, associated with mitochondrial profile; system I fibre underlying two-stranded microtubular root; mating structure present. These ultrastructural differences justify distinction between the two taxa at generic level. The problematical status of freshwater species ofDunaliella is briefly discussed.  相似文献

Zoospore ultrastructure (incl. flagellar apparatus) has been investigated in three species ofTrebouxia (T. glomerata, T. erici, T. pyriformis) and one species ofPseudotrebouxia (P. impressa) using an absolute configuration analysis. Zoospores in all taxa studied are nearly identical in ultrastructure and exhibit a very distinctive disposition of cell organelles: cells are naked, biflagellate and considerably flattened along the plane of flagellar beat, the single contractile vacuole is located anteriorly in the ventral region of the cell, the nucleus is anteriorly to centrally located in the dorsal region of the cell. A single dictyosome is located close to the anterior, ventral edge of the nucleus. The chloroplast occupies a posterior position in the cell and usually has an anterior profile in the left region of the cell. There are two branched mitochondria per cell or a single mitochondrial reticulum with profiles anterior to the nucleus (in the dorsal region of the cell), and posterior to the nucleus. In zoospores ofTrebouxia spp. the posterior mitochondrial profile is associated with a microbody, inP. impressa zoospores the anterior mitochondrial profiles are associated with a microbody. The zoospores contain a distinctive system of three ER-cisternae: one system links to both basal bodies and extends to the nucleus, the other two systems subtend the plasmamembrane on the left and right broad cell surfaces and extend to the posterior region of the cell. The flagellar apparatus is structurally identical to that previously described for zoospores ofFriedmannia israelensis and exhibits basal body displacement by one basal body diameter into the 11/5 o'clock direction, a non-striated distal connecting fiber, a cruciate microtubular root system lacking system I fibers and presence of a single system II fiber which connects the basal bodies with the nucleus and runs parallel to one of the ER-strands. The left flagellar roots (X-roots) are subtended by a complex set of amorphous and striated material that connects each left root with both basal bodies.—This study demonstrates the close systematic relationship between the phycobiontsTrebouxia andPseudotrebouxia and the generaFriedmannia, Pleurastrum, andMicrothamnion and supports recent classification schemes which place all these taxa into a single order separate from otherChlorophyta. Patterns of gonad structure in hermaphroditic gobies (Teleostei Gobiidae)   总被引:5,自引:0,他引:5  
Synopsis Hermaphroditism has been reported for a small number of gobiid fishes, but the extent of this sexual pattern within the family is not known. Gonad structure was examined in one or more species from twenty-one gobiid genera. No evidence of hermaphroditism was found in the species selected from 14 genera. Laboratory studies supported the conclusion of gonochorism for the examined species in four of them:Asterropteryx, Bathygobius, Gnatholepis, andPsilogobius. Currently, the absence of precursive testicular tissues associated with the ovary in females, in conjunction with no retained ovarian features in the testes of males, appear to be reliable indicators of a gonochoristic sexual pattern in gobiid fishes. Evidence for hermaphroditism was observed in seven genera:Eviota, Trimma, Fusigobius, Lophogobius, Priolepis, Gobiodon, andParagobiodon. Protogyny was experimentally confirmed inE. epiphanes, and the gonad structure in another nine of ten species ofEviota suggested either protogyny or protogynous tendencies. With the exception ofGobiodon andParagobiodon, which exhibited similar gonadal structure, ovarian and testicular structure varied considerably among the hermaphroditic genera examined, both with regard to the configuration and to the degree of development of ovarian and testicular tissues, or testicular tissue precursors. Findings of this study indicate that hermaphroditic gonad structure will prove to be a useful trait in determining evolutionary relationships within the Gobiidae.  相似文献

Summary To determine the degree of systematic affinity between branchiobdellids and lumbriculids, the ultrastructures of muscle and sperm cells ofBranchiobdella pentodonta andBythonomus lemani were studied.The structure ofBranchiobdella sperms is similar to that of the oligochaete model in terms of nucleus and the middle piece and to that of the hirudinean model in the peculiarity of the flagellum and acrosomal complex. The karyotype, nucleolar organizer region (NOR) location and C-banding pattern of two species of Hylodes (H. phyllodes and H. asper) and two of Crossodactylus (Crossodactylus sp. n. and Crossodactylus cf. caramaschi) were studied. All species had a diploid number of 2n = 26, with differences in the chromosomal morphology of the Hylodes species while the two Crossodactylus species were cytogenetically indistinguishable. The NOR was located on pair 1 in both species of Hylodes, and on pair 8 in the Crossodactylus species. In the latter, the NOR was heteromorphic between the homologues. The NOR was coincident with a secondary constriction in the four species. Except to H. phyllodes, such secondary constrictions were clearly seen strongly stained after C-banding treatment. The C-banding pattern varied between the two species of Hylodes, but was identical in the Crossodactylus species. The results from conventionally stained karyotypes confirmed the uniformity within the genus Crossodactylus, and the relatively conserved karyotypes within Hylodes, in agreement with other literature reports. We conclude that the cytogenetic data do not provide further evidence which could be useful to corroborate the supposed relationships between the hylodines and dendrobatids since there are no unambiguous homeologies between the karyotypes of these groups.  相似文献

The ultrastructure of the mature spermatozoa of four polychaetes is described:Eulalia sp. (Phyllodocidae),Lepidonotus sp. (Polynoidae),Lumbrineris sp. (Lumbrineridae) andOwenia fusiformis (Oweniidae). All the sperm show features typical of externally fertilizing sperm in having a rounded nucleus, a short unmodified midpiece, and a simple flagellum with a 9+2 axoneme.Owenia fusiformis andLepidonotus sp. have a nuclear cone extending into the subacrosomal space that may act to present the inner acrosomal membrane to the egg during fertilization. The acrosome ofLumbrineris sp. is flattened and crenulated. The sperm ofEulalia sp. is unusual in having the four mitochondria of the midpiece ensheathed by a membrane. Comparisons are made with other polychaete sperm, and the use of sperm ultrastructure as a taxonomic tool within the Polychaeta is discussed.  相似文献

Two new gynandrosporous species of Oedocladium from the United States were isolated into unialgal culture and comparative studies were made. The new taxa described, O. carolinianum sp. nov. and O. cir-ratum sp. nov., are morphologically very similar. Both species have an opercufate oogonium with inferior division, oospore walls with the middle layer ungulate, and hyaline suffultory cells containing a small amount of vacuolated cytoplasm. Conditions for sexuality in culture zuere determined for both species. Zoospores and akinetes serve as means for asexual reproduction.  相似文献

Two new homothallic species ofPichia, associated with exudates ofPopulus trichocarpa andSalix sp. have been described.Pichia trehalophila sp.n. was isolated from slime exudates in two widely separatedPopulus trichocarpa trees andPichia salictaria sp.n. was isolated from two exudates ofSalix sp. The latter species was previously present as an unidentified strain ofPichia in the culture collection of the Centraalbureau voor Schimmelcultures and had been isolated from a patient in Germany. The natural habitat ofP. salictaria is considered to be in association with willow trees.  相似文献
