利用电子探针微区分析(EPMA)技术, 分析了2018年8月采自湖南省岳阳市洞庭湖畔水产市场2尾刀鲚(Coilia nasus)干制个体的矢耳石中锶和钙微化学特征。定量线分析结果显示, 2尾样本的耳石锶钙比值(Sr/Ca×1000)波动显著, 从耳石核心到边缘可分别出现对应淡水生境的锶钙比值<3的低值区, 对应于半咸水生境的3—7高值区和再次为对应于淡水生境<3的低值区。面分布分析结果更直观显示两尾刀鲚个体的耳石从核心到边缘对应于不同盐度生境的锶含量变化图谱, 即从耳石核心到边缘可分别出现对应于淡水生境的蓝色区域, 对应长江口外半咸水生境的黄绿色区域和再次对应于淡水生境的蓝色区域。这种淡水-半咸水-淡水的生境转换显示出了上述个体的典型溯河洄游生活史“履历”。研究显示目前距长江河口约1400 km的洞庭湖中仍可能存在溯河洄游型刀鲚个体。  相似文献

The ontogenetic patterns of habitat use and the migratory history of the tapertail anchovy, Coilia mystus, collected in the Changjiang estuary around Chongming Island, China, were studied by examining the environmental signature in the otolith strontium (Sr) and calcium (Ca) fingerprints using electron probe microanalysis (EPMA). Our results suggest that the migration strategy of C. mystus is much more flexible than supposed in the literature to date. The spring spawning population of C. mystus from the studied area was found to consist of individuals with different migration histories. Although the tapertail anchovy seems to be an anadromous fish that spawns and hatches in a freshwater habitat, it can also use a freshwater environment in non‐spawning seasons. The otolith EPMA of the elemental fingerprint (Sr x‐ray maps and Sr : Ca ratios) is an environmental indicator that can be applied to the migratory ecology of other important diadromous species in China.  相似文献

The chemical composition of common carp Cyprinus carpio asteriscus (vaterite) and lapillus (aragonite) otoliths from the same individual and reflecting the same growth period was measured to (1) determine whether there are differences in the uptake of trace metals (Mg:Ca, Mn:Ca, Sr:Ca and Ba:Ca ) and Sr isotope ratios (87Sr:86Sr) in co‐precipitating lapilli and asterisci and (2) compare the ability of multi‐element and isotopic signatures from lapilli, asterisci and both otolith types combined to discriminate C. carpio populations over a large spatial scale within a river basin. Depth profile analyses at the otolith edge using laser‐ablation inductively coupled plasma mass spectrometry showed that asterisci were enriched in Mg and Mn and depleted in Sr and Ba relative to lapilli, whilst 87Sr:86Sr values were nearly identical in both otolith types. Significant spatial differences among capture locations were found when all trace element and Sr isotope ratio data were aggregated into a multi‐element and isotopic signature, regardless of which otolith type was used or if they were used in combination. Discriminatory power was enhanced, however, when data for both otolith types were combined, suggesting that analysis of multiple otolith types may be useful for studies attempting to delineate C. carpio populations at finer spatial or temporal scales.  相似文献

The migratory history of tapertail anchovy Coilia nasus in the Yangtze River Estuary, China was investigated using otolith Sr:Ca ratios and two-dimensional images of the Sr level from an X-ray electron probe microanalyzer (EPMA). The results showed that 17 of the 22 young-of-the-year (YOY) specimens had low Sr:Ca ratios (1.2–2.4?×?10?3;1.5?±?0.3?×?10?3) at the central otolith area, indicating their riverine origin and initial freshwater residence. In addition, 11 of the 14 adult specimens had low Sr:Ca ratios (1.3–2.2?×?10?3; 1.7?±?0.4?×?10?3) at the central otolith area but showed alternating changes between high (>4.0?×?10?3) and low (<2.5?×?10?3) values outside of this region, reflecting their riverine origin and the migration between freshwater and estuarine habitats. These 28 specimens represented the anadromous population in this region. The other 5 YOY specimens had high Sr:Ca ratios (3.6–5.9?×?10?3; 4.8?±?0.8?×?10?3) throughout the life history. Similarly, the other 3 adult specimens had high Sr:Ca ratios (4.0–5.7?×?10?3; 4.8?±?0.7?×?10?3) at the central otolith area but showed alternating changes between low and high values outside this region, suggesting that estuarine-origin non-anadromous individuals occurred in this region. The average of the otolith Sr:Ca ratios and Sr level mapping along the life-history transects could be used as a scalar for charting the migratory history of the tapertail anchovy in the Yangtze River Estuary: <2.0?×?10?3 for freshwater residence and 3.5–6.0?×?10?3 for estuarine residence.  相似文献

The naked carp (Gymnocypris przewalskii) is a native teleost of Lake Qinghai (altitude, 3.2 km) on the Qinghai-Tibet Plateau in China. Hypoxia-inducible factor (HIF)-1α of Gymnocypris przewalskii was cloned and a phylogenetic tree for vertebrate HIF-1α was constructed. By analysis of maximum likelihood models of codon substitutions for HIF-1α, three positive sites in the branch lineages of crucian carp, eelpout, and flounder, and a higher proportion of neutral sites in naked carp, antarctic eelpout, rainbow trout, and grayling, were detected among all teleosts. It seems that low habitat temperatures relax the purifying selection of HIF-1α in these stenothermal coldwater fish, and both cold and hypoxic lake water contributed to the evolution of the HIF-1α gene in the naked carp. Furthermore, Glut 1 mRNA, a gene downstream from HIF-1α, has a time-course- and tissue-specific dependent response to hypoxic challenge.  相似文献

The migratory histories of Japanese freshwater sculpins, one Trachidermus and four Cottus species, were studied by examining strontium (Sr) and calcium (Ca) concentrations in their otoliths using wavelength dispersive X‐ray spectrometry on an electron microprobe. The Sr : Ca ratios in the otoliths changed with salinity of the habitat. The otoliths of Cottus nozawae showed consistently low Sr : Ca ratios, with an average of 3·37 × 10?3 from the core to the edge, suggesting a freshwater resident life cycle. In contrast, the otolith Sr : Ca ratios for Trachidermus fasciatus and Cottus kazika changed along the life history transects possibly in accordance with their migration patterns from sea to fresh water. The ratios of T. fasciatus and C. kazika averaged 5·4 × 10?3 and 5·3 × 10?3 respectively, in the otolith region from the core to the points 450–890 μm, and changed to the lower levels, averaging 2·0 × 10?3 and 2·7 × 10?3, in the outer otolith region. These data suggest that both the species have a catadromous life cycle. The otoliths of Cottus hangiongensis had low Sr : Ca ratios in the two regions from the core to the points 15–30 μm and the points 415–582 μm to the edge, averaging 2·0 × 10?3 and 1·9 × 10?3, with significantly higher ratios in the narrow area between these regions, averaging 4·6 × 10?3. Similar ontogenetic changes in otolith Sr : Ca ratios were found in the otoliths of Cottus amblystomopsis, suggesting their amphidromous life cycle. These findings suggest that otolith Sr : Ca ratios reflect individual life histories and that Japanese Trachidermus and Cottus species have diverse migratory histories.  相似文献

Synopsis We studied the life histories of the ninespine sticklebacks, Pungitius pungitius and Pungitius tymensis, collected from Japanese freshwater and brackish (sea) water habitats by examining the strontium (Sr) and calcium (Ca) concentrations in their otoliths. The Sr:Ca ratios in the otoliths changed with the salinity of the habitat regardless of identification as freshwater or brackish water type based on morphological characteristics. The ninespine sticklebacks living in a freshwater environment showed consistently low Sr:Ca ratios throughout the otolith. These samples were identified as a standard freshwater type. In contrast, all freshwater-type fishes collected from the intertidal zone showed higher otolith Sr:Ca ratios than those in the standard freshwater type, and the ratios fluctuated with the growth phase. All brackish water-type fishes collected in the intertidal zone showed the highest otolith Sr:Ca ratio throughout the otolith. In the present study, besides the two representative life history types of P. pungitius, i.e., freshwater and brackish water life history types, other sticklebacks had an anadromous life history type. These findings clearly indicate that the ninespine stickleback has a flexible migration strategy with a high degree of behavioral plasticity and an ability to utilize the full range of salinity in its life history.  相似文献

Although otolith Strontium (Sr)/calcium (Ca) ratios have been widely used to reconstruct the past salinity environmental history of anguillid eels, factors affecting the Sr/Ca ratios in otoliths are incompletely understood. Japanese Eel (Anguilla japonica) elvers (mean length 54.7 ± 2.1 mm) were collected in the estuary during their upstream migration and reared at 5 different salinities (0, 5, 15, 25, and 35 psu) and 3 types of feeding conditions (formulated feed, tubifex, and starvation) for 30 days to evaluate the effects of salinity and diets on otolith Sr/Ca ratios. Ca and Sr concentrations in the ambient water significantly increased with salinity (SAL) as [Ca] water = 15.50SAL − 5.56, and [Sr] water = 0.21SAL + 0.03, respectively. Sr/Ca ratios in otoliths increased with salinity (SAL) of the rearing water as [(Sr/Ca) × 1000] otolith = 0.091SAL + 3.790. In diets, Sr/Ca ratios were 4 times higher in tubifex than in formulated feed. However, in otoliths, ANOVA indicated that Sr/Ca ratios did not differ significantly between groups fed on tubifex or formulated feed (p = 0.118). Otolith Sr/Ca ratios were negatively correlated with fish growth rates while the growth rates differed significantly among rearing conditions with different salinities and diets. Partition coefficients of the Sr/Ca ratios from ambient water to fish tissues and otoliths significantly increased with salinity. The Sr/Ca ratios of Japanese Eel otoliths thus were positively correlated with the ambient salinity and decreased with increasing fish growth rate, but was not affected by fish diet.  相似文献

The variation of migration patterns in Dolly Varden Salvelinus malma in a stream with an artificial dam (erosion-control-dam) in the Shiretoko Peninsula, Hokkaido Island, Japan, at the southernmost part of its distribution, was examined by otolith Sr:Ca ratio analysis. All specimens from the above-dam area showed consistently low Sr:Ca ratios (<5.0 × 10−3) throughout the otolith. In contrast, many specimens from the below-dam area had higher ratios (>5.0 × 10−3) throughout the otolith or only at the outer part than do those from the above-dam area. This higher ratio is probably due to salinity effects. Specimen from the below-dam area could be divided into three types: (1) Consistently low Sr:Ca ratios throughout the otolith, being freshwater residents. (2) Low Sr:Ca ratios around the inner part of the otolith, and thereafter higher ratios toward the outer part, being typically anadromous. (3) Relatively higher Sr:Ca ratios throughout many or in some parts of the otolith, indicating that these have migrated from the freshwater to brackish water or seawater from the early life stage. These findings suggest that Dolly Varden has a high degree of plasticity in its migratory behavior.  相似文献

Much has been revealed about fish migration, including diadromous behaviour, through the use of otolith chemistry. Manipulative experiments assist with unravelling information on otolith chemical composition and incorporation thereby answering specific questions on diadromous movements. In this study, a laboratory-based experiment was used to determine the relative and interactive effects of salinity and water temperature on the composition of three key elements (Sr, Ba and Mg) within the otolith of a catadromous fish, Percalates novemaculeata, endemic to south-eastern Australia. Otolith incorporation of Sr and Ba was positively related to ambient water concentration, whereas Mg incorporation was not. Sr and Ba increased and decreased significantly across salinity gradients, respectively, with minor positive effects of temperature also being detected. Salinity and temperature interacted to significantly affect the elemental concentration ratios for Ba: Ca in otoliths. Discrimination between fresh water and marine environments shows promise for interpreting P. novemaculeata residency based on these elements alone. However, deciphering finer scale movements within estuarine environments may be difficult. Our data highlights the importance of multifactorial validation experiments and suggests complementary use of multiple approaches for unravelling species-specific patterns of fish movement and habitat use.  相似文献

对鲤稚鱼开展了不同浸泡浓度和不同标记时间梯度的SrCl2·6H2O暴露标记试验,以确认其耳石Sr标记的可行性及基本条件.首先,基于4个浓度(0、4、8、12 mg·L-1)水平的SrCl2·6H2O溶液,浸泡标记2 d来筛选基本浸泡标记浓度.然后,在SrCl2·6H2O为8 mg·L-1浓度下,基于5个浸泡时间(1、2、3、4、5 d)来筛选基本浸泡标记时间.电子探针分布分析结果显示:对照组(0 mg·L-1)耳石Sr/Ca比值低且稳定,标记组均出现了高Sr/Ca比值区.对照组耳石剖面为均一的低Sr蓝色图谱,而标记组耳石上均有高Sr红色标记环带,且标记成功率为100%.试验期间,标记组和对照组的死亡率、平均全长和平均体质量无显著差异,表明Sr标记对供试鱼无不良影响.由于耳石上清晰、完整的高Sr红色标记环带出现在标记浓度为8 mg·L-1及以上,标记时间为2 d及以上,故建议分别选择8 mg·L-1和2 d为基本浸泡标记浓度和基本浸泡标记时间.本研究证实耳石Sr标记技术对鲤稚鱼具有很好的可行性.  相似文献

We conducted a laboratory experiment to validate the relationship between the otolith strontium/calcium (Sr/Ca) ratio of Japanese eels (Anguilla japonica) and water Sr/Ca ratio when the ratio in water was changed. A linear and additive mixed modeling approach was used to assess otolith Sr/Ca ratio for elver-juvenile Japanese eels when ambient water was changed from seawater to freshwater. There was a significant difference between otolith Sr/Ca ratios of eels reared in freshwater and in seawater (freshwater: 1.3–2.3; seawater: 7.0–7.8 mmol/mol). The response of otolith Sr/Ca ratios of eels was not detected until after 10 d and models suggested that it might not be completed until at least 30–60 d. This study indicated the detailed ability of otolith Sr/Ca ratio to be used as a proxy for reconstructing the individual environmental history of Japanese eels. These findings can provide some assurances for future otolith Sr/Ca studies of eels in this system or in other areas that have similar environmental conditions.  相似文献

Synopsis In order to determine whether the trace element composition in otolith of the Japanese eel Anguilla japonica could be used to determine its habitat use, we used laser ablation inductivity coupled plasma mass spectrometry (LA-ICPMS) to assay sectioned otoliths of both marine-resident (sea eels) and freshwater-resident (river eels) eels. A close linear relationship in the Sr:Ca ratios between EPMA (X-ray analysis with an electron microprobe) and LA-ICPMS analyses was found, suggesting that the latter technique could be used to separate the marine and freshwater life phases. Elemental signatures in the otolith outside the elver mark showed significant differences in Cr:Ca, Mn:Ca, and Ba:Ca ratios as well as Sr:Ca ratios between sea and river eels. These results indicate that the elemental compositions may reflect environmental variability between marine and fresh water masses. Thus, those elemental ratios determined by LA-ICPMS analysis seem to have the potential to help distinguish the habitat of the eel.  相似文献

Strontium (Sr) and calcium (Ca) concentrations in the otoliths of the shirauo, Salangichthys microdon, collected from Japanese coastal waters were examined by wavelength‐dispersive X‐ray spectrometry on an electron microprobe. The otolith Sr : Ca ratios gradually increased from 15.7?19.7 × 10?3 around the core to 32.8?39.4 × 10?3 around the edge of otoliths, with no transition point from the low Sr : Ca ratio phase to the high phase. The fluctuation pattern of otolith Sr : Ca ratios was different from the anadromous form reported in a previous study. The evidence indicates that the fish do not necessarily migrate into freshwater during the growth phase.  相似文献

This study used otolith microchemistry to evaluate whether the moray eel Gymnothorax chilospilus uses different habitats throughout its life (mainly juvenile and adult phases). Of the most informative trace elements within otoliths (the twelve isotopes 23Na, 25Mg, 43Ca, 55Mn, 59Co, 60Ni, 63Cu, 66Zn, 86Sr, 111Cd, 138Ba and 208Pb) only three ratios of Ca (Na:Ca, Sr:Ca and Ba:Ca) were informative and therefore used in a multivariate regression-tree analysis. Using a multivariate partitioning, three main phases were described from profiles, including the larval life phase (leptocephali), the intermediate phase (longest section between the larval life phase and the terminal phase) and the terminal phase (final section i.e., the most recent months preceding the death of fish). According to concentrations of the three ratios to Ca, G. chilospilus can be separated into three groups during their larval life stage (very different in Sr and Na), four groups during the intermediate phase (few differences in Sr and Na) and three groups during the terminal phase (differences in Sr), illustrating that G. chilospilus inhabit different habitats during these three phases. Our results showed that the leptocephali encountered different oceanic water masses with fluctuating Sr:Ca ratios during the early larval phase. During the intermediate phase (main part of their life-span), they lived in lagoonal waters such as fringing reefs or reef flats of lagoonal islets, characterized by a lower Sr:Ca ratio. During the latter part of their life, approximately one third of G. chilospilus encountered more oceanic waters close to or at barrier reefs, suggesting possible movements of these fish along a coast-to-ocean gradient.  相似文献

The spotted flagtail, Kuhlia marginata (Kuhliidae) is commonly found in streams throughout the Ryukyu Archipelago, including Okinawa Island, Japan. Although it has been suggested that they spawn at sea, little is known about their migratory history. The aim of the study was to clarify their migration history based on otolith microchemistry analysis, size composition and the gonadal development of fish collected from the Genka River on Okinawa Island. All strontium/calcium (Sr/Ca) ratios of otoliths remained high around their cores, and then dropped rapidly at some distance from the core. The estimated standard length corresponding to decreases correlated with the size of new recruits collected in the river (ca. 20 mm in standard length: SL). Multiple increases, the sign for migrations into the high saline area, were recognized intermittently around the otolith margin. As the estimated SL at these increases in the Sr/Ca ratio occurred above the minimum maturation size (female: 95.5 mm SL; male: 83.5 mm SL), such multiple increases in Sr/Ca ratios were caused by spawning migration. Therefore, we conclude that the catadromous pattern of K. marginata is as follows; this species grows in the sea during the early life stage until ca. 20 mm SL and then grow in the freshwater area until maturation, before they migrate again to the sea for spawning. Sr/Ca ratio profiles also suggested that a large number of males did not return to the freshwater area after spawning, whereas females might spawn several times during their lives around the inshore area.  相似文献

We tested the hypothesis that otolith trace elemental signatures (microchemistries) of mottled sculpin Cottus bairdi, slimy sculpin C. cognatus, and juvenile coho salmon Oncorhynchus kisutch were predictive of those of juvenile steelhead O. mykiss across many sites within the Lake Michigan basin. Laser ablation inductively coupled plasma mass spectrometry was used to generate otolith microchemistry signatures for each individual fish. For each species pair, statistical correlations of mean otolith concentrations of Mg, Mn, Cu, Zn, Sr, Ba, and Pb for each site were estimated. Linear equations describing these relationships were used to transform juvenile steelhead otolith microchemistry data to those of each of the other species. Transformed otolith microchemistry data were subjected to random forest classifications developed for mottled sculpin, slimy sculpin, and juvenile coho salmon to assess interspecific natal source assignment accuracies. Steelhead otolith concentrations of Sr were significantly correlated with those of each of the other species, whereas otolith concentrations of Ba and Mn were significantly correlated among some species pairs, but not others. Natal source assignment accuracies of juvenile steelhead to site and watershed generally decreased when otolith microchemistry data were transformed to those of mottled sculpin, slimy sculpin, and coho salmon. Miss-assigned fish often classified into nearby watersheds within larger hydrologic units, leading to higher assignment accuracies at coarser geographical resolutions (75–97% correct assignment to hydrologic unit for each species). These findings suggest that applications of otolith microchemistry data may extend beyond the species from which they are collected.  相似文献

Otolith elemental (Sr:Ca, Ba:Ca, Mn:Ca, Mg:Ca and Rb:Ca) and isotopic (87Sr:86Sr) profiles from several annual cohorts of juvenile Atlantic salmon Salmo salar were related to the physico‐chemical characteristics (chemical signatures, flow rate, temperature and conductivity) of their natal rivers over an annual hydrological cycle. Only Sr:Ca, Ba:Ca and 87Sr:86Sr in otoliths were determined by their respective ratios in the ambient water. Sr:Ca ratios in stream waters fluctuated strongly on a seasonal basis, but these fluctuations, mainly driven by water flow regimes, were not recorded in the otoliths. Otolith Sr:Ca ratios remained constant during freshwater residency at a given site and were exclusively related to water Sr:Ca ratios during low flow periods. While interannual differences in otolith elemental composition among rivers were observed, this variability was minor compared to geographic variability and did not limit classification of juveniles to their natal stream. Success in discriminating fish from different sites was greatest using Sr isotopes as it remained relatively constant across years at a given location.  相似文献

The elemental signature in otolith nuclei was determined using laser ablation inductively coupled plasma mass spectrometry (LA-ICPMS) for stock discrimination of adult anadromous tapertail anchovy, Coilia nasus, in five Chinese estuaries. Five elements (Na, Mg, K, Sr, and Ba) were well detected in the otolith nuclei of the adult fish. Results showed that the elemental composition in the otolith nuclei varied substantially among the estuaries. Age and fish length data showed no significant influences on the elemental concentration ratios across the sample sites. The Sr:Ca and Ba:Ca ratios were inter-site distinct and could be used as natal tags for discriminating among stocks. Discriminant function analyses (DFA) showed that these ratios can be used in discriminating the Liaohe River estuary (LD, 92.3?%), the Yangtze River estuary (CJ, 86.7?%), and the Yellow River estuary (HH, 76.9?%) samples with high classification accuracy, followed by the Haihe River estuary (BH, 58.3?%) and the Daguhe River estuary (JZ, 46.2?%) samples. An overall classification accuracy rate of 72.7?% from the discriminant functions indicated that elemental fingerprinting appeared to have the potential to discriminate between tapertail anchovy stocks in these estuaries.  相似文献
