A repeated batch fermentation system was used to produce ethanol using an osmotolerant Saccharomyces cerevisiae (VS3) immobilized in calcium alginate beads. For comparison free cells were also used to produce ethanol by repeated batch fermentation. Fermentation was carried for six cycles with 125, 250 or 500 beads using 150, 200 or 250 g glucose L−1 at 30°C. The maximum amount of ethanol produced by immobilized VS3 using 150 g L−1 glucose was only 44 g L−1 after 48 h, while the amount of ethanol produced by free cells in the first cycle was 72 g L−1. However in subsequent fed batch cultures more ethanol was produced by immobilized cells compared to free cells. The amount of ethanol produced by free cells decreased from 72 g L−1 to 25 g L−1 after the fourth cycle, while that of immobilized cells increased from 44 to 72 g L−1. The maximum amount of ethanol produced by immobilized VS3 cells using 150, 200 and 250 g glucose L−1 was 72.5, 93 and 87 g ethanol L−1 at 30°C. Clostridium beijerinckii DSM 6423 was studied using different continuous production methods to give maximum and stable production of isopropanol and n-butanol. In a single-stage continuous culture, when wood pulp was added as a cell holding material, we could increase the solvent productivity from 0.47 to 5.52 g L−1 h−1 with the yield of 54% from glucose. The overall solvent concentration of 7.51 g L−1 (39.4% isopropanol and 60.6% n-butanol) with the maximum solvent productivity of 0.84 g L−1 h−1 was obtained with two-stage continuous culture. We were able to run the process for more than 48 overall retention times without losing the ability to produce solvents.  相似文献

The fermentation characteristics of the novel, thermotolerant, isolate Kluyveromyces marxianus var marxianus were determined to evaluate its aptitude for use in an ethanol production process. Sustainable growth was not observed under anaerobic conditions, even in the presence of unsaturated fatty acid and sterol. A maximum ethanol concentration of 40 g L−1 was produced at 45°C, with an initial specific ethanol production rate of 1.7 g g−1 h−1. This was observed at ethanol concentrations below 8 g L−1 and under oxygen-limited conditions. The low ethanol tolerance and low growth under oxygen-limited conditions required for ethanol production implied that a simple continuous process was not feasible with this yeast strain. Improved productivity was achieved through recycling biomass into the fermenter, indicating that utilising an effective cell retention method such as cell recycle or immobilisation, could lead to the development of a viable industrial process using this novel yeast strain. Candida peltata NRRL Y-6888 to ferment xylose to xylitol was evaluated under different fermentation conditions such as pH, temperature, aeration, substrate concentration and in the presence of glucose, arabinose, ethanol, methanol and organic acids. Maximum xylitol yield of 0.56 g g−1 xylose was obtained when the yeast was cultivated at pH 6.0, 28°C and 200 rpm on 50 g L−1 xylose. The yeast produced ethanol (0.41 g g−1 in 40 h) from glucose (50 g L−1) and arabitol (0.55 g g−1 in 87 h) from arabinose (50 g L−1). It preferentially utilized glucose > xylose > arabinose from mixed substrates. Glucose (10 g L−1), ethanol (7.5 g L−1) and acetate (5 g L−1) inhibited xylitol production by 61, 84 and 68%, respectively. Arabinose (10 g L−1) had no inhibitory effect on xylitol production. Corn steep water (CSW) medium (1.6% solids plus 6% glucose) was evaluated for growth and butanol production by Clostridium beijerinckii NCIMB 8052 wild-type and hyper-amylolytic, hyper-butanol-producing mutant strain BA101. CSW alone was not a suitable substrate, whereas addition of glucose supported growth and butanol production by both strains. In a batch-scale fermentation using an optimized 6% glucose-1.6% solids CSW medium, C. beijerinckii NCIMB 8052 and strain BA101 produced 10.7 g L−1 and 14.5 g L−1 of butanol, respectively. The total solvents (acetone, butanol, and ethanol) produced by C. beijerinckii NCIMB 8052 and strain BA101 were 14 g L−1 and 20 g L−1, respectively. Initial fermentation in small-scale flasks containing 6% maltodextrin-1.6% solids concentration CSW medium resulted in 6 g L−1 and 12.6 g L−1 of butanol production by C. beijerinckii NCIMB 8052 and strain BA101, respectively. CSW can serve as an economic source of nitrogen, vitamins, amino acids, minerals, and other nutrients. Continuous production of acetone, n-butanol, and ethanol (ABE) was carried out using immobilized cells of Clostridium acetobutylicum DSM 792 using glucose and sugar mixture as a substrate. Among various lignocellulosic materials screened as a support matrix, coconut fibers and wood pulp fibers were found to be promising in batch experiments. With a motive of promoting wood-based bio-refinery concept, wood pulp was used as a cell holding material. Glucose and sugar mixture (glucose, mannose, galactose, arabinose, and xylose) comparable to lignocellulose hydrolysate was used as a substrate for continuous production of ABE. We report the best solvent productivity among wild-type strains using column reactor. The maximum total solvent concentration of 14.32 g L−1 was obtained at a dilution rate of 0.22 h−1 with glucose as a substrate compared to 12.64 g L−1 at 0.5 h−1 dilution rate with sugar mixture. The maximum solvent productivity (13.66 g L−1 h−1) was obtained at a dilution rate of 1.9 h−1 with glucose as a substrate whereas solvent productivity (12.14 g L−1 h−1) was obtained at a dilution rate of 1.5 h−1 with sugar mixture. The immobilized column reactor with wood pulp can become an efficient technology to be integrated with existing pulp mills to convert them into wood-based bio-refineries.  相似文献

Production of the bacteriocin enterocin 1146 (E1146) by Enterococcus faecium DPC1146 was studied in batch and continuous fermentation. Growth was strongly inhibited by lactic acid. In batch fermentations maximum E1146 activity (2.8 MBU L−1) was obtained in 9 h with 20 g L−1 glucose. Increase in initial glucose concentration did not lead to a proportional increase in E1146 activity. A simple linear model was found to be adequate to explain the relationship between specific bacteriocin production rate and specific growth rate in batch fermentations with initial glucose concentration higher than 20 g L−1. Maximum bacteriocin activity (2.9–3.2 MBU L−1) was obtained in continuous fermentations at dilution rates between 0.12 and 0.17 h−1 and specific bacteriocin production rate increased linearly with dilution rate. The bioconversion of xylose to xylitol by Candida guilliermondii FTI 20037 cultivated in sugar cane bagasse hemicellulosic hydrolyzate was influenced by cell inoculum level, age of inoculum and hydrolyzate concentration. The maximum xylitol productivity (0.75 g L−1 h−1) occurred in tests carried out with hydrolyzate containing 54.5 g L−1 of xylose, using 3.0 g L−1 of a 24-h-old inoculum. In a study screening anaerobic microbes utilizing d-galactitol as a fermentable carbon source, four bacterial strains were isolated from an enrichment culture producing H2, ethanol, butanol, acetic acid, butyric acid, and hexanoic acid. Among these isolates, strain BS-1 produced hexanoic acid as a major metabolic product of anaerobic fermentation with d-galactitol. Strain BS-1 belonged to the genus Clostridium based on phylogenetic analysis using 16S rRNA gene sequences, and the most closely related strain was Clostridium sporosphaeroides DSM 1294T, with 94.4% 16S rRNA gene similarity. In batch cultures, Clostridium sp. BS-1 produced 550 ± 31 mL L−1 of H2, 0.36 ± 0.01 g L−1 of acetic acid, 0.44 ± 0.01 g L−1 of butyric acid, and 0.98 ± 0.03 g L−1 of hexanoic acid in a 4-day cultivation. The production of hexanoic acid increased to 1.22 and 1.73 g L−1 with the addition of 1.5 g L−1 of sodium acetate and 100 mM 2-(N-morpholino)ethanesulfonic acid (MES), respectively. Especially when 1.5 g L−1 of sodium acetate and 100 mM MES were added simultaneously, the production of hexanoic acid increased up to 2.99 g L−1. Without adding sodium acetate, 2.75 g L−1 of hexanoic acid production from d-galactitol was achieved using a coculture of Clostridium sp. BS-1 and one of the isolates, Clostridium sp. BS-7, in the presence of 100 mM MES. In addition, volatile fatty acid (VFA) production by Clostridium sp. BS-1 from d-galactitol and d-glucose was enhanced when a more reduced culture redox potential (CRP) was applied via addition of Na2S·9H2O.  相似文献

Delta-endotoxin production by a strain of Bacillus thuringiensis subsp kurstakion complex media based on crude gruel and fish meal was investigated. High proteolytic activities were concomitantly produced with the bioinsecticide. In such complex media, the repressive regulation due to readily consumed carbon sources was partially overcome. In order to improve substrate assimilation, 0.5 g L−1 sodium chloride and 0.1% Tween-80 were supplemented to the production medium, increasing delta-endotoxin yields when using gruel concentrations below 59 g L−1. At and beyond 75 g L−1 gruel, delta-endotoxin yields were not affected in the presence of 0.5 g L−1 NaCl and 0.1% Tween-80, but proteolytic activity yields were remarkably reduced. Thus, the use of sodium chloride and Tween-80 allowed reduction of the initial gruel concentration to 42 g L−1 for the production of 3350 mg L−1 delta-endotoxin, while it was only 3800 mg L−1 with 92 g L−1 gruel. Moreover, similar to 0.5 g L−1 NaCl and 0.1% Tween-80, the use of 10 g L−1 sodium acetate significantly improved delta-endotoxin production and also reduced the proteolytic activity to 250 U ml−1. 1,3-Propanediol inhibition during glycerol fermentation to 1,3-propanediol by Clostridium butyricum CNCM 1211 has been studied. The initial concentration of the 1,3-propanediol affected the growth of the bacterium more than the glycerol fermentation. μ max was inversely proportional to the initial concentration of 1,3-propanediol (0–65 g l−1). For glycerol at 20 g l−1, the growth and fermentation were completely stopped at an initial 1,3-propanediol concentration of 65 g l−1. However, for an initial 1,3-propanediol concentration of 50 g l−1 and glycerol at 70 g l−1, the final concentration (initial and produced) of 1,3-propanediol reached 83.7 g l−1(1.1 M), with complete consumption of the glycerol. Therefore, during the fermentation, the strain tolerated a 1,3-propanediol concentration higher than the initial inhibitory concentration (65 g l−1). The addition of 1,2-propanediol or 2,3-butanediol (50 g l−1) in the presence of glycerol (50–100 g l−1), showed that 2-diols reduced the μ max in a similar way to 1,3-propanediol. The measurement of the osmotic pressure of glycerol solutions, diols and diol/glycerol mixtures did not indicate any differences between these compounds. The hypothesis of diol inhibition was discussed. Taking into account the strain tolerance of highly concentrated 1,3-propanediol during fermentation, the fermentation processes for optimising production were considered. Environmental copper contamination is a serious human health problem. Copper reductase is produced by microorganisms to facilitate copper uptake by ATPases into the cells increasing copper biosorption. This study assessed the reduction of Cu(II) by cell-free extracts of a highly copper-resistant bacterium, Pseudomonas sp. strain NA, isolated from vineyard soil contaminated with copper. Both intact cells and cell-free extract of Pseudomonas sp. strain NA displayed substantial reduction of Cu(II). Intact cells reduced more then 80 mg L−1 of Cu(II) from medium amended with 200 mg L−1 of copper after 24 h of incubation. Cell-free extract of the isolate reduced more than 65% of the Cu(II) at initial copper concentration of 200 mg L−1 after 24 h. Soluble protein production was high at 72 h of incubation at 100 mg L−1 of copper, with more then 60 μg L−1 of total soluble protein in cell-free extract recorded. Cu(II) reduction by isolate NA was increased when copper concentration increased for both intact cells and cell-free extract. Results indicate that Pseudomonas sp. strain NA produces copper reductase enzyme as the key mechanism of copper biotransformation.  相似文献

A yeast strain Kluyveromyces sp. IIPE453 (MTCC 5314), isolated from soil samples collected from dumping sites of crushed sugarcane bagasse in Sugar Mill, showed growth and fermentation efficiency at high temperatures ranging from 45°C to 50°C. The yeast strain was able to use a wide range of substrates, such as glucose, xylose, mannose, galactose, arabinose, sucrose, and cellobiose, either for growth or fermentation to ethanol. The strain also showed xylitol production from xylose. In batch fermentation, the strain showed maximum ethanol concentration of 82 ± 0.5 g l−1 (10.4% v/v) on initial glucose concentration of 200 g l−1, and ethanol concentration of 1.75 ± 0.05 g l−1 as well as xylitol concentration of 11.5 ± 0.4 g l−1 on initial xylose concentration of 20 g l−1 at 50°C. The strain was capable of simultaneously using glucose and xylose in a mixture of glucose concentration of 75 g l−1 and xylose concentration of 25 g l−1, achieving maximum ethanol concentration of 38 ± 0.5 g l−1 and xylitol concentration of 14.5 ± 0.2 g l−1 in batch fermentation. High stability of the strain was observed in a continuous fermentation by feeding the mixture of glucose concentration of 75 g l−1 and xylose concentration of 25 g l−1 by recycling the cells, achieving maximum ethanol concentration of 30.8 ± 6.2 g l−1 and xylitol concentration of 7.35 ± 3.3 g l−1 with ethanol productivity of 3.1 ± 0.6 g l−1 h−1 and xylitol productivity of 0.75 ± 0.35 g l−1 h−1, respectively.  相似文献

Butanol, a four-carbon primary alcohol (C4H10O), is an important industrial chemical and has a good potential to be used as a superior biofuel. Bio-based production of butanol from renewable feedstock is a promising and sustainable alternative to substitute petroleum-based fuels. Here, we report the development of a process for butanol production from glycerol, which is abundantly available as a byproduct of biodiesel production. First, a hyper butanol producing strain of Clostridium pasteurianum was isolated by chemical mutagenesis. The best mutant strain, C. pasteurianum MBEL_GLY2, was able to produce 10.8 g l−1 butanol from 80 g l−1 glycerol as compared to 7.6 g l−1 butanol produced by the parent strain. Next, the process parameters were optimized to maximize butanol production from glycerol. Under the optimized batch condition, the butanol concentration, yield, and productivity of 17.8 g l−1, 0.30 g g−1, and 0.43 g l−1 h−1 could be achieved. Finally, continuous fermentation of C. pasteurianum MBEL_GLY2 with cell recycling was carried out using glycerol as a major carbon source at several different dilution rates. The continuous fermentation was run for 710 h without strain degeneration. The acetone–butanol–ethanol productivity and the butanol productivity of 8.3 and 7.8 g l−1 h−1, respectively, could be achieved at the dilution rate of 0.9 h−1. This study reports continuous production of butanol with reduced byproducts formation from glycerol using C. pasteurianum, and thus could help design a bioprocess for the improved production of butanol.  相似文献

Maltose and sucrose were efficient carbon sources for the production of curdlan by a strain of Agrobacterium sp. A two-step, fed-batch operation was designed in which biomass was first produced, followed by curdlan production which was stimulated by nitrogen limitation. There exists an optimal timing for nitrogen limitation for curdlan production in the two-step, fed-batch operation. Maximum curdlan production (60 g L−1) was obtained from sucrose with a productivity of 0.2 g L−1 h−1 when nitrogen was limited at a cell concentration of 16.0 g L−1. It was also noted that the curdlan yield from sucrose was as high as 0.45 g curdlan g−1 sucrose, and the highest specific production rate was 1.0 g curdlan g−1 cells h−1 right after nitrogen limitation. Of particular importance was the use of molasses as a cheap carbon source to produce curdlan in the two-step, fed-batch cultivation. As high as 42 g L−1 of curdlan with a yield of 0.35 g curdlan g−1 total sugar was obtained after 120 h of fed-batch cultivation. Phycocyanin production by high cell density cultivation of Spirulina platensis in batch and fed-batch modes in 3.7-L bioreactors with a programmed stepwise increase in light intensity program was investigated. The results showed that the cell density in fed-batch culture (10.2 g L−1) was 4.29-fold that in batch culture (2.38 g L−1), and the total phycocyanin production in the fed-batch culture (0.795 g L−1) was 3.05-fold that in the batch culture (0.261 g L−1). An unstructured kinetic model to describe the microalga culture system including cell growth, phycocyanin formation, as well as glucose consumption was proposed. The data fitted the models well (r 2 > 0.99). Furthermore, based on the kinetic models, the potential effects of light limitation and photoinhibition on cell growth and phycocyanin formation can be examined in depth. The models demonstrated that the optimal light intensity for mixotrophic growth of Spirulina platensis in batch or fed-batch cultures using a 3.7-L bioreactor was 80160 μE m−2 s−1, and the stepwise increase in light intensity can be replaced by a constant light intensity mode. Simultaneous isomerisation and fermentation (SIF) of xylose and simultaneous isomerisation and cofermentation (SICF) of a glucose/xylose mixture was carried out by Saccharomyces cerevisiae in the presence of xylose isomerase. The SIF of 50 g l−1 xylose gave an ethanol concentration and metabolic yield of 7.5 g l−1 and 0.36 g (g xylose consumed)−1. These parameters improved to 13.4 g l−1 and 0.40 respectively, when borate was added to the medium. The SICF of a mixture of 50 g l−1 glucose and 50 g l−1 xylose gave an ethanol concentration and metabolic yield of 29.8 g l−1 and 0.42 respectively, in the presence of borate. Temperature modulation from 30 °C to 35 °C during fermentation further enhanced the above parameters to 39 g l−1 and 0.45 respectively. The approach was extended to the bioconversion of sugars present in a real lignocellulose hydrolysate (peanut-shell hydrolysate) to ethanol, with a fairly good yield. Molasses “B” is a rich co-product of the sugarcane process. It is obtained from the second step of crystallization and is richer in fermentable sugars (50–65%) than the final molasses, with a lower non-sugar solid content (18–33%); this co-product also contains good vitamin and mineral levels. The use of molasses “B” for ethanol production could be a good option for the sugarcane industry when cane sugar prices diminish in the market. In a complex medium like molasses, osmotolerance is a desirable characteristic for ethanol producing strains. The aim of this work was to evaluate the use of molasses “B” for ethanol production using Saccharomyces cerevisiae ITV-01 (a wild-type yeast isolated from sugarcane molasses) using different initial sugar concentrations (70–291 g L−1), two inoculum sizes and the addition of nutrients such as yeast extract, urea, and ammonium sulphate to the culture medium. The results obtained showed that the strain was able to grow at 291 g L−1 total sugars in molasses “B” medium; the addition of nutrients to the culture medium did not produce a statistically significant difference. This yeast exhibits high osmotolerance in this medium, producing high ethanol yields (0.41 g g−1). The best conditions for ethanol production were 220 g L−1 initial total sugars in molasses “B” medium, pH 5.5, using an inoculum size of 6 × 106 cell mL−1; ethanol production was 85 g L−1, productivity 3.8 g L−1 h−1 with 90% preserved cell viability.  相似文献

A bench-scale biofilter with a 0.5-m high filter bed, inoculated with a toluene-degrading strain of Acinetobacter sp. NCIMB 9689, was used to study toluene removal from a synthetic waste air stream. Different sets of continuous tests were conducted at influent toluene concentrations ranging over 0.1–4.0 g m−3 and at superficial gas velocities ranging over 17.8–255 m h−1. The maximum volumetric toluene removal rate for the biofilter (242 g m−3 h−1) was obtained at a superficial gas velocity of 127.5 m h−1 (corresponding to a residence time of 28 s) and a toluene inlet concentration of 4.0 g m−3. Under these operating conditions, toluene removal efficiency was only 0.238, which suggested that effective operation required higher residence times. Removal efficiencies higher than 0.9 were achieved at organic loads less than 113.7 g m−3 h−1. A macro-kinetic study, performed using concentration profiles along the bioreactor, revealed this process was limited by diffusion at organic loads less than 100 g m−3 h−1 and by biological reaction beyond this threshold. Batch and continuous cultivation of Anaerobiospirillum succiniciproducens were systematically studied for the production of succinic acid from whey. Addition of 2.5 g l−1 yeast extract and 2.5 g l−1 polypeptone per 10 g l−1 whey was most effective for succinic acid production from both treated and nontreated whey. When 20 g l−1 nontreated whey and 7 g l−1 glucose were used as cosubstrates, the yield and productivity of succinic acid reached at the end of fermentation were 95% and 0.46 g (l h)−1, respectively. These values were higher than those obtained using nontreated whey alone [93% and 0.24 g (l h)−1 for 20 g l−1 whey]. Continuous fermentation of A. succiniciproducens at an optimal dilution rate resulted in the production of succinic acid with high productivity [1.35 g (l h)−1], high conversion yield (93%), and higher ratio of succinic acid to acetic acid (5.1:1) from nontreated whey.
