In 34 patients with human immunodeficiency virus (HIV) infection at the asymptomatic stage and 29 patients with chronic viral hepatitis B at the period of exacerbation (of these 14 patients had chronic persistent hepatitis and 15 patients had chronic active hepatitis) the complex study of the functional activity of lymphocytes and neutrophils was carried out by cytochemical methods with the simultaneous determination of the content of immunoregulating lymphocyte subpopulations. In patients with chronic active hepatitis a decrease in the percentage and the absolute number of helper T-lymphocytes and the ratio of CD4/8 in comparison with those in patients with HIV infection were revealed. At the same time patients with HIV infection exhibited more pronounced decrease in the activity of all lymphocytic enzymes under study (neutrophil esterase, acidic phosphatase and succinate dehydrogenase in lymphocytes), as well as in the activity of myeloperoxidase and the content of cation proteins and glycogen in neutrophils in comparison with patients having chronic active hepatitis.  相似文献

Summary The histochemical distribution of various enzymes, such as alkaline phosphatase, acid phosphatase, esterase, -glycosidase, aminopeptidase, succinic dehydrogenese and TPN diaphorase, in human submaxillary glands has been determined.Acini and ducts of human submaxillary gland were devoid of alkaline phosphatase activity, but this enzyme was observed in capillaries and somewhat in myoepithelium.Activities of acid phosphatase, esterase, -glucuronidase and -galactosidase were generally observed in the entire cytoplasm of serous acini; but the cytoplasm of mucous acini was either negative or showed only trace amounts.Aminopeptidase reaction of both acini and ducts was generally negative.Succinic dehydrogenase and TPN diaphorase activities were strongly active in intralobuler ducts. Serous acini exhibited less activity with these enzymes; and mucous cells showed still less and were almost negative. Summary Active male gerbils were rendered unilaterally cryptorchid and the enzymatic changes were studied after 48, 60 and 100 days. The histochemical observations revealed inhibition of alkaline phosphatase, acid phosphatase, 5-nucleotidase, adenosine-tri-phosphatase and succinic dehydrogenase activities in the germinal elements of the seminiferous tubules. The non-specific esterase and succinic dehydrogenase activities increased in the Leydig cells to certain extent. The sudanophilic lipids increased in the Sertoli cells and the interstitial cells. An attempt has been made to approach the intricate problem of the spermatogenesis in order to understand the sequence of certain enzymatic activities in the gerbil testis in which spermatogenesis is suppressed when one of the testes is lodged inside the abdominal cavity.  相似文献

The proventriculus of White Leghorn chick embryos (stages 29–45) newly-hatched chicks, and adult chickens were frozen, sectioned in a cryostat and treated histochemically to identify localizations of alkaline and acid phosphatase, adenosine triphosphatase, 5-nucleotidase, nucleotide-diphosphatase, non-specific glycerophosphatase, glucose-6-phosphatase, non-specific esterase and succinic dehydrogenase. Ribonucleic acid, proteins and acid mucopolysaccharides were identified in tissues fixed in FAA. Acid phosphatase, nucleotide-diphosphatase, adenosine triphosphatase, succinic dehydrogenase, ribonucleic acid and proteins were present in the cells of the deep glands at all stages of development. Alkaline phosphatase and 5-nucleotidase were found only in mesenchymal derivatives of the proventriculus. After the chick begins swallowing and digesting albumen, enzymatic activity increased and non-specific esterase became very reactive. The surface epithelium is covered with a mucous coat. Ribonucleic acid, non-specific esterase, acid phosphatase and nucleotide-diphosphatase were localized in the basal portions of the epithelial cells. The functional significance of these different patterns is discussed.  相似文献

Cytochemical indices of leukocytes were determined in 16 patients with diabetes mellitus in the period of unbalancing and balancing. The following tests were made: content of glycogen and lipids, acid phosphatase (AP), alkaline phosphatase (AIP), myeloperoxidase (MPO) and nonspecific alpha-naphtol acetate esterase (NANAE) activity. In unbalanced diabetics an evident decrease in the activity of AP and MPO could be noted as well as a decrease of glycogen content and an increase of lipid content. An insignificant decrease could be observed in the activity of ALP and NANAE in granulocytes. A slight increase in the activity of NANAE in monocytes would be found. Balancing this disease induced the increase of all parameters in granulocytes except MPO activity. It is interesting to note that balancing diabetes mellitus deepened the observed changes in the decrease or increase of tested parameters. The presented findings clearly indicate the role of metabolic disorders in diabetes mellitus on the activity of some neutrophilic enzymes and the glycogen and the content of lipids in neutrophils.  相似文献

Summary Detailed histochemical studies on the distribution of various oxidative and dephosphorylating groups of enzymes have been made in the olfactory glomeruli of the squirrel monkey. The olfactory glomeruli showed strongly positive activity for succinic dehydrogenase, lactic dehydrogenase, monoamine oxidase, alkaline phosphatase, adenosinetriphosphatase and simple esterase. They showed moderately positive activity for cytochrome oxidase, specific cholinesterase, 5'nucleotidase; mildly positive activity for acid phosphatase; and negligible activity for nonspecific cholinesterase and glucose-6-phosphatase. The glomeruli did not show the presence of any thiamine pyrophosphatase-positive Golgi apparatus. The blood vessels surrounding the glomeruli were strongly positive for the nonspecific cholinesterase test. The significance of these results are discussed briefly.  相似文献

Summary Histochemical localization of various groups of oxidative and dephosphorylating enzymes and esterases has been made in the peritoneal mesothelial cells. These cells were strongly positive for lactic dehydrogenase and succinic dehydrogenase, moderately positive for cytochrome oxidase, simple esterase, acid phosphatase, 5nucleotidase and nonspecific cholinesterase, and negative for monoamine oxidase and specific cholinesterase. They showed moderate positive activity for alkaline phosphatase, localized in the microvilli of these cells. They contain scanty amount of thiamine pyrophosphatase-positive Golgi complex and showed diffuse thiamine pyrophosphatase-positive activity in the cell cytoplasm. Adenosine triphosphatase-positive spaces found in between the adjacent cells and at the bases of the individual cells have been demonstrated. These results are discussed in view of the great absorptive power of the peritoneal mesothelium.  相似文献

The effect of malathion (50 mg/kg) and radiation (900R) separately and jointly upon phosphatases and succinic dehydrogenase has been studied in female albino rats. Malathion and radiation when given separately, at the dose levels used in this study, appear to have no effect on acid and alkaline phosphatase of kidney, heart and brain. In preirradiated rats given malathion a significant decrease in phosphatases was observed in kidney and brain tissue after as little as 2 h and the values persisted for up to 24 h. No significant change was observed in heart tissue. At the dose level of 50 mg/kg of malathion, no significant change in succinic dehydrogenase was observed; however, radiation caused a significant increase in succinic dehydrogenase of brain and kidney tissue. In preirradiated rats given malathion the effect appeared to be enhanced in time to a certain degree. One could therefore speculate that simple non-specific stress caused by malathion adds to a similar effort of radiation by reducing phosphatases and enhancing the succinic dehydrogenase enzyme.  相似文献

Summary The histochemistry and DNA turnover of postcapillary high-endothelial venules in lymph nodes and Peyer's patches have been investigated. Autoradiographic studies showed that the cells are long lived. They exhibit a distinct metachromatic reaction that is indicative of a moderate amount of RNA. They are PAS positive due to the presence of glycogen. Considerable amounts of mucopolysaccharides are not found inside the cells. The reactions for lactate dehydrogenase, NADH-tetrazolium reductase and unspecific esterase are strong. The intensities of the reactions are comparable to those of the epithelium of the intestinal villus and of reticular cells and definitely stronger than that of other endothelial cells. Tests for acid phosphatase, alkaline phosphatase, succinic dehydrogenase, glucose-6-phosphate dehydrogenase and NADPH-tetrazolium reductase showed that these enzymes are either absent or present in small amounts.  相似文献

It was shown that the vagotomy leads to the elevation of the activity of alkaline phosphatase (AP) 7 days after the operation and to the decrease of its activity 30 days after the operation, and to the decrease of the activity of succinic dehydrogenase (SDG) 7 and 30 days after the operation. Quantitative cytochemistry of components of blood neutrophil azurophilic granules (myeloperoxidase, chloroacetate esterase, beta-glucuronidase, and acid phosphatase) and specific granules (lactoferrin) has been performed by scanning and integrating microdensitometry in 13 patients with a myelodysplastic syndrome and 11 patients with chronic granulocytic leukaemia. Both patient groups showed a reduction of enzyme activity in azurophilic granules, and also of lactoferrin, consistent with abnormal development of neutrophil granules. These cytochemical changes in blood neutrophils are similar to those found in acute myeloid leukaemia, are consistent with a leukaemic maturation defect, and may be of diagnostic value.  相似文献

Summary Enzyme activity is not uniformly distributed through the cotyledon of Pisum arvense. Initially the peripheral region, certain scattered cells of the storage tissue and the procambium show a high level of activity of succinic dehydrogenase, cytochrome oxidase, acid phosphatase and esterase. Activity of acid phosphatase declines sharply after the first day of germination; activity of the other enzymes declines after about three days. In the storage tissue, where activity is lower initially, it declines after about five days and is correlated with the disappearance of the reserves. The pattern of alkaline phosphatase activity is similar except that activity is lower in the procambium but increases in the sieve-elements during differentiation of the phloem. 5-nucleotidase and glucose-6-phosphatase activity is low throughout the cotyledon but it also increases to a significant level in the sieve-elements. Activity of starch synthesizing enzymes is high in the parenchymatous bundle sheath, where they may be involved in the pathway from lipids to soluble carbohydrates.  相似文献

Mild structural and functional changes were observed in liver and testes of rats after a single intraperitoneal administration of diazinon (21.6 mg/kg). Kidney, however, showed no pathological lesion. Attempts are made to correlate the pathological changes in these organs with the activity of succinic dehydrogenase, adenosine triphosphatase and alkaline phosphatase.  相似文献

Summary Changes in liver enzymes of Weanling CDF1 mice inoculated with mouse hepatitis virus (MHV3) were studied by histochemical techniques for the demonstration of alkaline phosphatase, acid phosphatase, and esterase. Marked changes were observed in the distribution of these enzymes 22 to 70 hours after infection. These included a generalized increase in peribiliary alkaline phosphatase together with a localized increase in acid phosphatase and a decrease in esterase associated with parenchymal damage and subsequent necrosis. Summary The morphology and histochemical enzyme pattern of the small intestine were investigated in chicks undergoing feed restriction. Corresponding intestinal sites were compared in both restricted birds and in control birds under normal feeding. Intestines from the restricted birds showed some atrophy, the villi being slightly shorter and thinner than normal after eight days restriction, and there was an increase in the activity of alkaline phosphatase, leucine naphthylamidase, acid phosphatase, -glucuronidase, non-specific esterase and succinic dehydrogenase in the absorptive cells.The significance of these findings has been discussed in relation to the enhanced absorptive capacity of the intestine during feed restriction and its similarity to other dietary stress factors that produce enhanced absorption. Possible mechanisms for the production of such mucosal changes have been considered. It was concluded that the enhanced absorption of nutrients in semi-starved animals is correlated with increased mucosal enzyme activities.  相似文献

Acute renal failure was induced in male rats by the subcutaneous injectioon of 4 mg HgC12 per kg body weight. Enzyme activities of the proximal tubule were studied histochemically at six time intervals from 15 min to 24 h. The enzyme studied were alkaline phosphatase, 5'-nucleotidase, acid phosphatase, alpha-glycerophosphate dehydrogenase (NAD-independent), malic dehydrogenase, succinic dehydrogenase, latic dehydrogenase, glucose-6-phosphate dehydrogenase and glucose-6-phosphatase. Decreases in activity were observed for alkaline phosphatase and 5'-nucleotidase after 15 min. Acid phosphatase was decreased after 30 min. These three enzymes returned to control levels after 3 h, but malic dehydrogenase and alpha-glycerophosphate dehydrogenase were decreased at this time interval. Succinic dehydrogenase was first decreased after 6 h. The earliest morphological changes detectable by light microscopy were observed in pars recta tubules in the medullary rays after 6 h, a time when all enzymes studied showed widespread decreased activity throughout the proximal tubule. After 24 h, the pars convoluta appeared morphologically normal but the pars recta was necrotic and exhibited calcification, whereas enzyme activity was decreased (absent in some cases) in both pars convoluta and pars recta. These results support the hypothesis that Hg++, when given in a sublethal dose, is associated with early histochemical changes in the brush border of the proximal tubule, which may be related to early changes in sodium reabsorption and to the subsequent development of acute renal failure. The observation that changes in plasma membrane-associated enzymes occur early and prior to alterations in enzymes of mitochondria and the endoplasmic reticulum suggests that Hg++ interacts initially with the plasma membrane.  相似文献

Localization and activity of five hydrolases (alkaline phosphatase, adenosine triphosphatase, acid phosphatase, nonspecific esterase and leucylamino-peptidase) were evaluated histochemically in the epididymides of mature dogs. In the ductuli efferentes, cilia and apical parts of the epithelial cells displayed high activity of alkaline phosphatase and adenosine triphosphatase. Strong activity of acid phosphatase, nonspecific esterase and leucylamino-peptidase was present in the basal and supranuclear zones of the epithelium of the ductuli efferentes. Stereocilia of all three segments of the ductus epididymidis showed a high activity of alkaline phosphatase. Positive adenosine triphosphatase reaction was confined to the stereocilia of the initial segment. A complex pattern of acid phosphatase activity was observed in the middle segment. The subdivision of the middle segment in four subsegments was therefore suggested. In the epithelium of the initial segment only a few nonspecific esterase-positive cells were seen. The infranuclear and basal areas of the epithelium in the middle segment and the supranuclear zone of the terminal segment displayed distinct nonspecific esterase activity. The possible contribution of the hydrolases to the function of the epididymis is discussed.  相似文献

The cytochemistry and staining of granula in peripheral blood granulocytes in healthy carp ( Cyprinus carpio L.) are described. Blood smears were stained for periodic acid-Schiff (PAS), peroxidase, oxidase, alkaline and acid phosphatase, α-naphthyl-acetate esterase, α-naphthyl-butyrate esterase, naphthol-AS-chloroacetate esterase (AS-D), naphthol-AS-acetate esterase and β-glucuronidase. Different granula types were shown by triazid-staining (eosinophil and neutrophil granula) and methylenblue-staining for basophil granulation. Toluidinblue-staining was used for basophil granulocytes. Lipids were shown by the Sudan-black-reaction. Four granulocyte subpopulations are described: neutrophil, heterophil, basophil and eosinophil granulocytes. Neutrophils possess all tested granula types, whereas heterophil and basophil granulocytes show only basophil granula. Neutrophils and heterophils show no activity of the tested esterases with the exception of AS-D. Only neutrophils were peroxidase-positive. Alkaline phosphatase and β-glucuronidase were not detected in granulocytes. Basophils and especially eosinophils were rarely found in peripheral blood.  相似文献

The magnetic field of 0.008 T and 0.15 T inductions influence lasting 7 weeks (7 days a week), 1 h daily determines the increase of the activity of cytoplasmatic enzymes (glutamic pyruvic transaminase, glutamic oxalacetic transaminase, lactic dehydrogenase), the decrease of cholinesterase activity and the growth of alkaline phosphatase activity in the plasma of the examined animals. The observed changes were reversible. 2 months after the exposure had been stopped, the tested parameters were back to normal.  相似文献

Microwave-stimulated enzyme incubations for acetylcholinesterase, 5'-nucleotidase, alkaline phosphatase, lactate dehydrogenase, malate dehydrogenase, succinic dehydrogenase and isocitric dehydrogenase were studied, and compared with incubations in a waterbath. Temperature settings of 37 degrees C and 50 degrees C were used, and the incubation times were varied from 30 seconds to 30 minutes. The desired temperature of the incubation solution was reached in the microwave oven within 1 minute, whilst in the waterbath it took 10 to 25 minutes. The microscopic results for alkaline phosphatase and succinic dehydrogenase at a temperature setting of 50 degrees C were superior in the microwave method for incubation times less than 15 minutes. It is postulated that the increased reaction product of alkaline phosphatase and succinic dehydrogenase is due to a temperature effect, which has to be large enough to be of practical value. For the other enzymes studied, microwave-stimulated incubations were no better than the conventional incubations at corresponding temperatures. For 5'-nucleotidase there were aspecific lead deposits in the microwave method. All enzymes performed at the elevated, unphysiological temperature of 50 degrees C proved to have advantages, except for 5'-nucleotidase, whilst for malate dehydrogenase there was an aspecific reduction of the colour developer at this temperature.  相似文献
