BACKGROUND AND AIMS: Colleters are secretory structures consisting of a parenchymatic middle axis surrounded by a layer of palisade-like epidermal cells. Colleters occur in a large number of rubiaceous species. Their function is to protect the developing shoot apex. They are also taxonomically useful in the Rubiaceae. This study characterized the structure of the colleters of Simira glaziovii, S. pikia and S. rubra and the biochemistry of secretions in S. glaziovii. METHODS: Stipules of the shoot apices of the three species studied were collected at Barragem de Saracuruna, in Rio de Janeiro state, Brazil. The samples were fixed according to the usual methods for light and electron microscopy. Secretion stipules of S. glaziovii were washed with 0.1 m Tris-HCl plus 0.1 %Triton X-100 to extract proteins and carbohydrates. KEY RESULTS: Colleters in these species are located at the base of the stipule. Each species shows a different pattern of distribution. They form as emergentia from the stipules. Simira glaziovii was different from the other two species because it exhibited vascular traces. The epidermal cells of colleters have dense cytoplasm, nuclei, small vacuoles, endoplasmic reticulum, Golgi apparatus, mitochondria and extraplasmic spaces if they are secretory. The outer cell wall of the mature colleters differs from the outer cell wall of stipule cells and immature colleters. Both carbohydrates and proteins were found in secretions from the stipules of S. glaziovii. CONCLUSIONS: Few ultrastructural differences were noted among the three species. These secretory structures not only protect the shoot apex, but also have taxonomic importance below the genus level. 相似文献

The presence of extrafloral nectaries (EFNs) between leaflets is an usual feature in Inga edulis (Vell.) Mart. (Leguminosae). Extrafloral nectaries are secretory structures involved in production of nectar and which serve in the protection of plants against herbivores through association with ants. This study aimed to characterize the EFNs of I. edulis at different developmental stages and describe their morphology, histochemistry and ultrastructure. Leaf fragments, containing secretory structures, were processed according to standard methods for light, scanning and transmission electron microscopy. The EFNs were classified into three stages based on morphology: pre‐secretory, secretory and post‐secretory. The pre‐secretory stage occurs in young leaves, whereas secretory and post‐secretory stages occur in developed and senescent leaves, respectively. The EFNs possess a concave surface and a central cleft in which nectar is accumulated and which was not observed in pre‐secretory EFNs. Histochemical tests identified the presence of sugars, proteins, phenolic compounds, mucilage and lipids at all developmental stages of the EFNs. Calcium crystals were identified in all tissues and stages of the EFNs. The secretory cells of the EFNs exhibit a granular cytoplasm, small vacuoles, prominent nuclei, smooth endoplasmic reticulum and mitochondria. Post‐secretory stage EFNs exhibited intense cytoplasmic degradation and the presence of microorganisms. The performance of EFNs of I. edulis appear to follow the leaf development. 相似文献

Cavitated secretory trichomes are characterized by a short or absent stalk that is connected to a secretory hollow head. They are rare structures in angiosperms; in Fabaceae, they have been recorded in only seven genera, including Bauhinia s.s. Because B. curvula and B. rufa exhibit glands that are responsible for attracting pollinators to flowers, this study aimed to test whether the cavitated secretory trichomes present in the flowers of these species have an attraction function. As leaf trichomes are commonly related to plant defence, comparative analyses of the morphology, ontogeny, ultrastructure and chemical profile of the secretory trichomes present in flowers and leaves were conducted. It was found that cavitated secretory trichomes are similar in their external morphology and development, regardless of the organ or species analysed. However, interspecific differences were found in the secretion process and chemical profile of the exudate. The differences found in the cavitated secretory trichomes between species indicate that they secrete distinct compounds, whereas the similarities found in these structures between vegetative and reproductive organs indicate that the cavitated trichomes have equivalent ecological functions within a species, probably in plant defence during organ development. © 2015 The Linnean Society of London, Botanical Journal of the Linnean Society, 2016, 180 , 104–122. 相似文献

Colleters are secretory structures well distributed in many organs of Angiosperms. Ultrastructurally, the colleters secretory cell presents an enhanced endoplasmic reticulum, Golgi apparatus, and mitochondria. Secretion synthesis, transportation, and passage through outer cell wall is poorly characterized. This study characterized the anatomy and ultrastructure of BATHYSA NICHOLSONII (Rubiaceae) colleters and evaluated the presence of protein in the secretion and its antifungal property. Samples were collected and prepared according to usual techniques in light and electron microscopy, electrophoresis, and fungal growth inhibition assay. Colleters are of a standard type, cylindrical and elongated, formed by one secretory epidermal palisade layer, and a central axis formed by parenchymatic cells and a vascular trace. Epidermal cells have dense cytoplasm with abundant ribosome, a nucleus, enhanced endoplasmic reticulum and Golgi apparatus. The outer cell wall presented morphologically distinct layers. The presence of secretory cavities was noted in all outer cell wall extents. Secretion preparations analyzed by SDS-PAGE showed that B. NICHOLSONII secretion is a mixture of proteins with molecular masses covering a range of approximately 66 to 24 kDa. This preparation presented an inhibitory effect on the fungi spore growth. 相似文献

Colleters are secretory structure present on many families including Rubiaceae. Particular characteristics have been described about colleters secretory cells, however senescence process are still under debate. Tocoyena bullata (Vell.) Mart. (Rubiaceae) shoot apex were collected at Jardim Botânico do Rio de Janeiro, RJ/Brazil. Stipules were separated and fragments were fixed in 2.5% glutaraldehyde and 4.0% formaldehyde in 0.05 m sodium cacodylate buffer, pH 7.2, post fixed in 1.0% osmium tetroxide in the same buffer, dehydrated in acetone, critical‐point‐drying, sputtered coated and observed. For light microscopy fragments were fixed and dehydrated, infiltrated with historesin and stained with 1% toluidine blue. For transmission electron microscopy, the samples were infiltrated with Epoxi resin. Colleters are present on stipule adaxial surface. On the beginning of development, these structures are recognized as small projections. Later on, colleters differentiated and secrete by cuticle rupture. The colleters senescence occurs in a concomitant and indissoluble way of programmed cell death. Ultrastructural analyses during the process strongly suggest the senescence is based on a non‐autolitic programmed cell death. T. bullata colleters, present at stipule abaxial surface are cylindrical secretory structures. Colleters secretory cells originated as stipule projections; differentiate; secrete and senesce by programmed cell death. The secretion and the cell dead occurs in a concomitantly and indissoluble way. 相似文献

An ex-situ experiment was conducted to evaluate the growth performance of six medicinal species (Bergenia ciliata, Valeriana jatamansi, Dioscorea deltoidea, Paeonia emodi, Polygonum amplexicaule and Viola serpense) from upper Swat , Pakistan . Experiments were conducted at four different locations in the upper Swat valley at altitudes ranging from1200 to 1900 m. a. s. l . The objectives were : 1) to determine the suitability of ex-situ cultivation of different medicinal species, and; 2) to assess the economic geasibility of growing medicinal plants in the area . A highest mean survival of 80.7% across all locations was observed for Viola serpense, followed by 58.7% for Valeriana jatamansi. The remaining four species exhibited very poor survival rates , although Polygonum amplexicaule, did show encouraging signs of growth and flowered , before experiencing high mortality rates late in the trial . Altitude generally seemed to enhance the degree of sprouting for all species except Viola serpense. However, the productive yield of V. serpense was certainly not reduced, but rather slightly enhanced in the higher altitude sites. Overall, cultivation of only two of the investigated species, Valeriana jatamansi and Viola serpense, appearedsuccessful and potentially economically viable under farmland conditions at upper Swat . 相似文献

The ultrastructure of resting state colonies of Botryococcus braunii Kützing from two localities is compared to active state colonies maintained in the laboratory. Qualitative hydrocarbon analyses confirmed the physiological status of each sample according to the precedent in the literature; resting state colonies contained botryococcenes, while active state colonies contained a predominance of straight-chain olefins. The ultrastructure of resting and active state colonies is fundamentally similar. The chloroplast of resting state cells contains fewer thylakoids and larger relative numbers of plastoglobuli than the chloroplast of active state cells maintained under favorable growth conditions. Previously undescribed phenomena include the cytochemical demonstration of polyphosphate bodies microbody-like organelles and ER-ribosomal-mitochondrial complexes. Attempts to elicit the transition of active to resting state colonies utilizing different light intensities and/or nitrogen deficient media were unsuccessful. Instead, these experiments induced the formation of yellow or whitish-brown senescent colonies which always contained a predominance of olefins. Senescent cells contain reduced, peripherally displaced organelles. Most of the cytoplasm is occupied by vacuoles and lipid (probably hydrocarbon) inclusions. Morphologically identifiable polyphosphate bodies are usually small or absent. Observations pertinent to the mechanism of hydrocarbon secretion were in basic agreement with previously published information. The outer cell wall, or trilamellar structure (TLS), was recently as the primary site of hydrocarbon accumulation and production. The role of the TLS in hydrocarbon biosynthesis, however, should be cautiously regarded without additional evidence. 相似文献

This paper describes the development of colleters of Bathysa cuspidata, Rubiaceae, considering anatomical, histochemical and ultrastructural aspects and going from first differentiation stages until senescence. Further, the chemical composition of the secretion is investigated. The samples were prepared according to the usual techniques for light microscopy and scanning and transmission electron microscopy. Electrophoresis and thin layer chromatography (TLC) were used to confirm the results obtained in the histochemical tests. The colleters occur at the ventral surface of the stipules which protect the leaf primordia as well as the shoot meristem. The origin of the colleters is mixed, involving protoderm and ground meristem. The Bathysa colleters are of the standard type or are bifurcated; this latter type is documented here for the first time for Rubiaceae. Colleter secretion is a mucilage rich in protein, as determined by histochemical tests and confirmed by chemical analysis. Phenolic compounds and terpenes were detected only in the colleters themselves, but not in the secretion. The epithelial cells present conspicuous nuclei and nucleoli and the cytoplasm is rich in dictyosomes, endoplasmic reticulum, mitochondria, vesicles and small vacuoles with a fibrillar content. The accumulation of phenolic compounds and terpenes, the formation of a large central vacuole, the increase of the intercellular and subcuticular spaces occupied by the secretion and, finally, the darkening and the wilting of the colleters characterize the senescence of these structures. The secretion process of the colleters of B. cuspidata suggests a process of programmed cell death. 相似文献

Roots, stems, rhizomes and leaves of Rhaponticum carthamoides (Willd.) Iljin (a Siberian adaptogenic plant, originating from the Altai and Saian Mountains) of different ages were investigated by means of light and electron microscopy. Schizogenous secretory reservoirs occurred in every organ, and were located within the secondary xylem (adventitious roots and rhizome of young plants), at the interface of endodermis/cortical parenchyma (roots and hypocotyl), along phloem and primary xylem (older rhizome), around the vascular bundles (inflorescence stem, petiole and leaf midrib veins) and along phloem (cotyledonary and leaf veins). At the interface of endodermis/inner parenchyma, secretion accumulated in the intercellular spaces prior to the formation of proper epithelial cells. The secretion as observed by transmission electron microscopy comprised three components: soluble (i.e. absent from sections; probably phenolic), insoluble and strongly osmiophilic (probably phenolic) and insoluble, moderately osmiophilic (probably lipidic). Numerous osmiophilic oil droplets, similar to the lipidic secretion inside the reservoirs, local proliferation of rough endoplasmic reticulum and numerous multivesicular bodies characterized epithelial cells in all organs. Leucoplasts (in subterranean organs) with osmiophilic inclusions and peroxisomes with crystalloid inclusions were specific for parenchyma cells. BACKGROUND AND AIMS: Digestive structures of carnivorous plants produce external digestive enzymes, and play the main role in absorption. In Lentibulariaceae, the ultrastructure of digestive hairs has been examined in some detail in Pinguicula and Utricularia, but the sessile digestive hairs of Genlisea have received very little attention so far. The aim of this study was to fill this gap by expanding their morphological, anatomical and histochemical characterization. METHODS: Several imaging techniques were used, including light, confocal and electron microscopy, to reveal the structure and function of the secretory hairs of Genlisea traps. This report demonstrates the application of cryo-SEM for fast imaging of whole, physically fixed plant secretory structures. KEY RESULTS AND CONCLUSION: The concentration of digestive hairs along vascular bundles in subgenus Genlisea is a primitive feature, indicating its basal position within the genus. Digestive hairs of Genlisea consist of three compartments with different ultrastructure and function. In subgenus Tayloria the terminal hair cells are transfer cells, but not in species of subgenus Genlisea. A digestive pool of viscous fluid occurs in Genlisea traps. In spite of their similar architecture, the digestive-absorptive hairs of Lentibulariaceae feature differences in morphology and ultrastructure. 相似文献

用石蜡切片法制片,光学显微镜观察,研究白木香叶的解剖结构。结果表明,白木香叶为典型的异面叶,但同时还表现出许多旱生形态。表皮由一层排列紧密形状不规则的表皮细胞组成,细胞外壁角质膜较厚。上表皮角质膜一般较下表皮角质膜厚1μm,下表皮上零星分布着单细胞表皮毛。气孔仅分布在下表皮上,微下陷,气孔类型为无规则型。叶肉组织发达,其间分布着较多的长方晶体,其细胞长轴与表皮垂直。栅栏组织由1—2层排列整齐的圆柱形细胞组成。栅栏组织的外层细胞转化为异细胞,海绵组织内具有一层由异细胞组成的下皮层;横切面上栅栏组织厚度与海绵组织厚度之比为1:3。主脉发达,有异细胞组成的维管束鞘,具双生韧皮部。叶内具有发达的木质部外纤维。这些特征与旱生环境相适应。 相似文献

- In Metrodorea nigra, a Rutaceae species with rhythmic growth, the shoot apex in the dormant stage is enclosed by modified stipules. The young organs are fully covered with peltate secretory trichomes, and these structures remain immersed in a hyaline exudate within a hood-shaped structure. Our study focused on the morpho-functional characterization of the peltate trichomes and cytological events associated with secretion.
- Shoot apices were collected during both dormant and active stages and processed for anatomical, cytochemical and ultrastructural studies.
- Trichomes initiate secretion early on, remain active throughout leaf development, but collapse as the leaves expand; at which time secretory cavities start differentiation in the mesophyll and secretion increases as the leaf reaches full expansion. The subcellular apparatus of the trichome head cells is consistent with hydrophilic and lipophilic secretion. Secretion involves two vesicle types: the smaller vesicles are PATAg-positive (periodic acid/thiocarbohydrazide/silver proteinate) for carbohydrates and the larger ones are PATAg-negative. In the first phase of secretory activity, the vesicles containing polysaccharides discharge their contents through exocytosis with the secretion accumulating beneath the cuticle, which detaches from the cell wall. Later, a massive discharge of lipophilic substances (lipids and terpenes/phenols) results in their accumulation between the wall and cuticle. Release of the secretions occurs throughout the cuticular microchannels.
- Continued protection of the leaves throughout shoot development is ensured by replacement of the collapsed secretory trichomes by oil-secreting cavities. Our findings provide new perspectives for understanding secretion regulation in shoot apices of woody species with rhythmic growth.
