Summary Tests for enzymes of gluconeogenesis and of the synthesis and degradation of sucrose and polysaccharides have been carried out in the phloem exudate of Cucurbita pepo. All the enzymes which are necessary for the synthesis of sucrose and polysaccharides from metabolites of the citric acid cycle were found to be present in the exudate, except phosphoenolpyruvate carboxykinase. The polysaccharide synthetase was found to exhibit higher activity with glycogen (which is an unnatural polysaccharide in higher plants) than with starch. In addition, polysaccharide synthetase activity could be increased remarkably with 2 mM glucose-6-phosphate and glycogen as primer. Among the enzymes which catabolize sucrose and polysaccharides (phosphorylase, invertase, sucrose phosphorylase), only sucrose phosphorylase showed activity.  相似文献

Understanding the ecology of plant species in their whole range (native and introduced) can provide insights into those that become problematic weeds in the introduced range despite being benign components of the vegetative community in the native range. We studied the morphological traits of Impatiens glandulifera in the native (Indian Himalayas) and introduced (UK) range and evaluated what influences natural enemies and arbuscular mycorrhizal fungi (AMF) have on plant performance. We compared height, total leaf area, root: shoot ratio, natural enemy damage and the colonisation of AMF from individual plants within and between ranges twice in 2010 during the months of June and August. In addition, in August 2010, we estimated the number of reproductive units (expressed as the sum of flowers, seed capsule and seeds) at each site. We found that all morphological traits varied between populations and countries, though in general introduced populations, and the semi-natural population in India, showed higher performance compared to natural native populations. There was only an indication that natural enemy damage, which was significantly higher in the native range, negatively affected reproductive units. Within the introduced range, the percentage colonisation of AMF was negatively associated with plant performance indicating that I. glandulifera may associate with an incompatible AMF species incurring a cost to invasive populations. We conclude that species which are heavily regulated in the native range, though still show high levels of performance, should be considered undesirable introductions into similar ecoclimatic ranges due to the potential that these species will become highly invasive species.  相似文献

Zusammenfassung Licht- und elektronenmikroskopische Untersuchungen am Integument vonEchiniscus testudo (Heterotardigrada),Milnesium tardigradum, Hypsibius oberhaeuseri undMacrobiotus hufelandi (Eutardigrada) lassen acht, mindestens jedoch sechs Schichten erkennen, die in Anlehnung an Baccetti und Rosati (1971) benannt werden, wobei eine Homologie beiE. testudo nicht sicher ist.BeiE. testudo bildet die innere sklerotisierte Epicuticula ein Hohlraumsystem, dessen Wände von ca. 150 Å breiten Kanälen durchzogen werden. Die dorsalen Einsenkungen der Panzerplatten besitzen einen zentralen 500 Å breiten Kanal. Ventral ist die äußere Epicuticula mehrfach geschichtet. Ein darunterliegender Röhrchensaum und die weitgehend reduzierte innere Epicuticula lassen andere Permeabilitätseigenschaften vermuten. Die Intracuticula zeigt vertikale Filamente. Wachsschicht und innere Lage der Intracuticula sind morphologisch nicht eindeutig zu identifizieren. Die einschichtige Epidermis ist reich an Lipidtropfen, freien Ribosomen und granulärem endoplasmatischem Reticulum. Manche Zellen sind kontrastreicher.Besondere Cuticula-Bildungen der drei Eutardigraden sind die epicuticulären Einsenkungen vonM. hufelandi, in deren Bereich die innere Epicuticula fehlt, die dorsale und laterale Skulpturierung vonH. oberhaeuseri, die eine Bildung der Epicuticula ist, und die epicuticulären Kanäle vonM. tardigradum. Die Proticula aller untersuchten Tardigraden-Arten besitzt keine Porenkanäle.Die Epidermis vonM. hufelandi besteht aus einem einschichtigen Epithel. Seine Zellen enthalten zahlreiche membranbegrenzte osmiophile Einschlüsse, die die Färbung der Tiere bedingen. Einzelne Zellen fallen durch ihren hohen Kontrast und zahlreichere Organellen auf.Die Cuticula der Eutardigraden zeigt in allen Fällen eine morphologisch vergelichbare Schichtenfolge. Die Cuticula vonE. testudo, die Besonderheiten aufweist, kann jedoch aufgrund charakteristischer Merkmale (z. B. die äußere Lage der Intracuticula) vielleicht in den Vergleich einbezogen werden.
Comparative studies on the integument of Hetero- and Eutardigrada
Summary The structure of the integument ofEchiniscus testudo (Heterotardigrada),Milnesium tardigradum, Hypsibius oberhaeuseri andMacrobiotus hufelandi (Eutardigrada) was studied by light and electron microscopy. In all species the cuticle consists of 6–8 layers and can adequately be described following the terminology of Baccetti and Rosati (1971). This perhaps does not apply toE. testudo.InE. testudo the inner sclerotized epicuticle reveals a labyrinthine structure due to lacunae with numerous channels of about 150 Å in diameter. The central canals in the depressions of the dorsal plates have a diameter of 500 Å. The outer epicuticle of the ventral cuticle is formed by several layers. Basally there is a tubular layer of channels and an almost reduced epicuticle. Thus the permeability of the cuticle is supposed to be altered. In the intracuticle vertically oriented filaments occur. The inner layer of the intracuticle and the wax-layer cannot be properly distinguished. The one-layered epidermis contains many lipid droplets, ribosomes and rough ER. Some of the cells are rather electron-dense.Peculiar cuticular structures inM. hufelandi appear as cup-shaped depressions of the epicuticle, and here the inner epicuticle is absent. A similarly typical feature is the structured and sculptured dorsal and lateral epicuticle ofH. oberhaeuseri and the channels of the epicuticle inM. tardigradum. In all species investigated there are no pore canals in the procuticle.InM. hufelandi there is a single layer of epidermal cells, which contain many osmiophilic grana ensheathed by membranes. These grana cause the colour of the animal. Some of the cells contain more organelles and ribosomes and show a strong contrast.In spite of some cuticular peculiarities inE. testudo, perhaps this Heterotardigrade species fits into the general pattern of cuticle morphology of Tardigrada.

Summary Seeds ofVicia faba L. var.minor (2n=12) were submerged in tap water for 12 to 96 hours. After submersion and recovery times of 48 to 144 hours root tips were fixed in intervals of 24 hours. The presoaking of seeds before germination led to an increase of the spontaneous chromosomal aberration rate. Up to 65% of all cells in the root tips showed fragments, chromatid-translocations and triradials after 72 hours of presoaking and up to 12% of all chromosomes showed breaks or participated in chromatidal reunions. Submersion of seeds for 12 hours already caused a statistically significant increase of chromosomal aberrations in comparison to the controls. The maximum of aberrations was found after a recovery period of 120 hours, afterwards the aberration rate dropped. Parallel experiments with the N2-atmosphere showed that the increase in aberration rate is caused by metabolic changes produced by anaerobiosis. Probably the changed metabolic conditions led to the formation of automutagenic substances. The distribution of breaks between the mand SAT-chromosomes was not at random (theoretical expectation 5∶2); more breaks appeared in the mthan in the SAT chromosomes (actual relation 5∶1,1). The breaks in the mand SAT chromosomes were localized in a few special regions.   相似文献

Zusammenfassung Durch Vergleich fluoreszenz- und elektronenmikroskopischer Untersuchungen am zentralen Nervensystem von Planorbarius corneus L. wird nachgewiesen, daß in den Schlundringganglien Neurosekretzellen vorkommen (Nachweis mit Pseudoisocyaninchlorid), die mit Nervenzellen nicht identisch sind, die durch ihren hohen Gehalt an biogenen Aminen auffallen (Nachweis durch die Methode von Falck und Owman, 1965). Es können daher im Schlundring von Planorbarius corneus peptiderge und aminerge Neurosekretzellen unterschieden werden. Die PSC-positiven Neurosekretzellen enthalten elektronendichte Elementargrana und die aminergen Neurone dense-core Vesikel. Der Nachweis biogener Amine in einigen Nervenzellen von Planorbarius corneus spicht für deren chemische Identität mit Transmittersubstanzen, ihre hohe Konzentration aber für eine Abgabe in die Körperflüssigkeit.
Comparative fluorescence- and electron microscopic studies of the central nervous system of Planorbarius corneus L. (Basommatophora)
Summary The neurosecretory system of the snail Planorbarius corneus has been investigated by fluorescence and electron microscopy. With the fluorochrome Pseudoisocyanin the established neurosecretory system in the cerebral ganglia and single neurosecretory cells in the other ganglia show an intensive yellow fluorescence. Electron micrographs reveal the presence of electron dense granules (elementary granules) in the pericarya and the axons of neurones which have the same localisation in the ganglia as the pseudoisocyanin-positive cells. The fluorescence technique for biogenic amines produces yellow and green fluorescence within neurons and in the neuropil and nerves. The fluorescence obtained in determinable areas and neurones correlates well with the electron microscopic location of dense-core vesicles within the pericarya and axons of cells in even the same areas. It is discussed, that in this animal both types of cells are so-called neurosecretory cells, because the high content of elementary granules in the peptidergic neurosecretory cells and of dense-core vesicles in the aminergic neurosecretory cells is an indication for secretion of these products in neurohaemal areas (circulatory channels).

Zusammenfassung Das Recessus praeopticus-Organ (RPO) des 3. Ventrikels von Urodelen und Anuren wurde licht- und elektronenmikroskopisch untersucht. Es besteht aus einem speziellen Ependym und einer kleinzelligen Neuronengruppe, in der sich intraependymale, hypendymale und distale Nervenzellen unterscheiden lassen. Die Neurone senden Fortsätze in den Recessus praeopticus, wo sie freie, knöpfchenförmige Endigungen bilden. In den Nervenzellen, ventrikulären Dendriten und Liquorendigungen sowie in den peripheren Axonen ist fluoreszenzhistochemisch ein primäres Katecholamin, wahrscheinlich Noradrenalin, nachweisbar.Mit dem Elektronenmikroskop sind in den ventrikulären Nervenendigungen Mitochondrien, wenige Ergastoplasmacisternen, Mikrotubuli und dense-core vesicles in wechselnder Menge und Größe (Durchmesser 500–850 Å) nachweisbar. Es ist charakteristisch für die Nervenendigung, daß sie meist über ein Zilium verfügt, von dessen Basalkörper und akzessorischem Zentrosom lange, quergestreifte Basalwurzeln ausgehen. Querschnitte atypischer Zilien (Typ 8+1, 9+0, 10+1, 20+0) werden beschrieben. Die ventrikulären Nervenfortsätze sind durch Zonulae adhaerentes mit den benachbarten Ependymzellen verbunden. Die Perikaryen der intraependymalen und hypendymalen Neurone enthalten dense-core vesicles in wechselnder Menge. Die distalen Nervenzellen und ihre Fortsätze weisen relativ wenige Katecholamingranula auf. Im Neuropil wurden verschiedene Synapsen beobachtet, die wahrscheinlich unterschiedliche Funktionen ausüben. — An Hand der morphologischen Daten wird die Frage einer etwaigen Rezeptorfunktion des RPO diskutiert.
Light and electron microscopic studies of the preoptic recess organ in amphibians
Summary The preoptic recess organ (PRO) of the 3rd brain ventricle of urodela and anura has been studied by light and electron microscopy. — The PRO consists of a special ependyma and a parvocellular group of neurons. The latter is built up of intraependymal, hypendymal and distal nerve cells. The neurons send processes into the preoptic recess where they form free, club-like nerve terminals. In the perikarya, their ventricular dendrites and liquor endings as well as in their peripheral axons a primary catecholamine, probably noradrenaline, can be demonstrated by fluorescence histochemistry.As demonstrated electron microscopically, the ventricular nerve endings contain mitochondria, a few ergastoplasmic cisternae, microtubuli and dense-core vesicles of various amounts and size (500–850 Å). It characterizes the neurons that they mostly possess a cilium; its basal body and accessory centriol give rise to long rootlet fibers. Cross sections of atypical cilia (type 8+1, 9+0, 10+1, 20+0) are described. The ventricular nerve processes are connected with the surrounding ependymal cells by zonulae adhaerentes. The perikarya of the intraependymal and hypendymal neurons contain various amounts of dense-core vesicles. The distal nerve cells and their processes contain relatively few catecholamine granules. In the neuropil, different types of synapses are to be observed probably acting in different manner. — On the basis of the morphological data, the question of a receptor function of the PRO is discussed.

Zusammenfassung Fluoreszierende aminerge Nervenfasern bzw. Endigungen und elektronendichte Granula mit einem Durchmesser von 500–1000 Å wurden bei Passer domesticus in den Perikaryen und im Neuropil des Nucleus infundibularis, im Neuropil des Nucleus supraopticus, im rostralen Abschnitt der neurosekretorischen Bahn (Region der Sehnervenkreuzung) und in der retikulären und Palisadenschicht der Eminentia mediana beobachtet. 2000–2500 Å große kontrastreiche Granula finden sich in den Perikaryen des Nucleus supraopticus, im Tractus supraoptico-paraventriculo-hypophyseus und im Hypophysenhinterlappen. Nach Reserpinbehandlung schwanden sowohl die gelbgrüne Fluoreszenz als auch das elektronendichte Material der 1000 Å und z. T. auch der 500–800 Å großen Bläschen. Dieses Ergebnis spricht dafür, daß beide Granulatypen Monoamine enthalten. Der Nucleus infundibularis ist nach unseren Befunden die wichtigste Quelle der etwa 1000 Å großen Granula in der Palisadenschicht der Eminentia mediana.
Fluorescence- and electronmicroscopic studies on the hypothalamo-hypophysial system of Passer domesticus
Summary In Passer domesticus, fluorescent aminergic fibers and axon terminals and dense-core vesicles of 500 to 1000 Å diameter were found in the perikarya and the neuropile of the infundibular nucleus, in the neuropile of the supraoptic nucleus, in the rostral common neurosecretory pathway (region of the optic chiasma), in the tubero-infundibular tract, and in the reticular and palisade zones of the median eminence. 2000–2500 Å electron-dense vesicles were found in the perikarya of the supraoptic nucleus, in the proximal division of the common neurosecretory pathway and in pars nervosa of the hypophysis. After reserpine treatment, the yellow-green fluorescence as well as the dense material contained in the 1000 Å vesicles and partly also in the 500–800 Å vesicles disappeared. This suggested that the 1000 Å and the 500 Å dense-core vesicles in the hypothalamo-hypophysial system of Passer domesticus contain monoamines and that the infundibular nucleus is an important source of the 1000 Å dense-core vesicles in the palisade layer of the median eminence.
