The effects of nitrogen form and phosphorus source on the growth, nutrient uptake and rhizosphere soil property of tea (Camellia sinensis L.) were investigated in a pot experiment. The experiment was performed with a compartmental cropping device, which enables the collection of rhizosphere soil at defined distances from the root of tea plant. Nitrogen was supplied as nitrate or ammonium in combination with soluble phosphorus as Ca(H2PO4)2 or insoluble P as rock phosphate. The leaf dry matter production of tea was significantly greater in the treatments with NH4 + than NO3 -, whereas dry matter production of root and stem was not significantly affected. Addition of phosphorus as either source did not influence the dry matter production. The concentrations of K in root, Mg and Ca in both the shoot and root supplied with NO3 - were significantly higher than in NH4 + and influence of P sources was minor. On the contrary, Al and Mn concentrations were significantly larger in NH4 --fed plants which could be attributed to remarkably increased availability of Al and Mn caused by acidification of the rhizosphere soil (the first 1-mm soil section from the root surface) with NH4–N nutrition. The concentration of N in shoot was also significantly higher in NH4- than in NO3-fed plants, indicating higher use efficiency of NH4–N. Whatever the phosphate source, rhizosphere pH declined in ammonium compared to in nitrate treatment. The pH decrease was much larger when no P or soluble P were applied and reached 0.85–1.30 units which extended to 3–5 mm away from the root surface. Exchangeable acidity, content of exchangeable Al and Mn were also considerably higher in the rhizosphere soils of NH4 + fed tea plants. Significant amounts of P dissolved from rock phosphate accumulated in rhizosphere of NH4 +, not NO3 -, suggesting that the dissolution of rock phosphate was induced by the proton excreted by tea root fed with ammonium. With soluble P addition, shoot and root P concentrations were greater in NH4 + than in NO3 - treatment and it appeared that this difference could not be sufficiently explained by the available P content in soil which was only slightly higher in NH4 + treatment. With rock phosphate addition, the shoot and root P concentrations were hardly affected by nitrogen form, although the available P content was much higher and accumulated in the rhizosphere soil supplied with ammonium. The experiment was set up to examine the influence of different nitrogen forms: (NH4)2SO4, Ca(NO3)2 or NH4NO3 on growth response, root induced pH changes in the rhizosphere, root-borne acid phosphatase activity in strawberry plants cv. Senga Sengana. The plants grown on sandy mineral soil were fertilized with 3 forms of nitrogen, in concentrations of 46 mg N·kg−1 soil. The plants were grown in rhizoboxes with removable plexiglass lids. To ensure the root growth along the plexiglass lids, the rhizoboxes were placed at an angle of about 50° with the lid on the lower side. In case of ammonium supply, the nitrification inhibitor DIDIN was added (10 mg·kg−1 of moist soil) to prevent conversion of ammonium into nitrate. The growth response (roots and shoots) of strawberry plants were determined after 11 weeks of treatment with different N forms. The best development of the root system and shoots (root and shoot dry weight and root length) was obtained, when ammonium nitrate was supplied. It is suggested therefore, that NH4NO3 stimulates vegetative growth of strawberry plants cv. Senga Sengana. However, there were no statistical differences in a leaf and flower number of the plants grown under different forms of N-fertilization. Determination of rhizosphere pH, and acid phosphatase activity were executed using non-destructive techniques, which enabled weekly measurement of chemical changes in the rhizosphere. The results revealed that the form of nitrogen supplied had a predominant effect on chemical changes in the rhizosphere of strawberry plants. The highest pH values (average pH 6.8) were measured in the rhizosphere of individual plants supplied with Ca(NO3)2. Whereas the lowest pH values (average pH 5.8) were detected in the presence of (NH4)2SO4. The curve of rhizosphere pH measured along individual roots of the plants treated with Ca(NO3)2 represents the highest pH values whereas the curve of rhizosphere pH under (NH4)2SO4 treatment had the lowest pH values. The highest activity of acid phosphatase were observed in the rhizosphere of strawberry plants grown in the presence of (NH4)2SO4, at pH 5.8.  相似文献

Models and experimental studies of the rhizosphere of rice plants growing in anaerobic soil show that two major processes lead to considerable acidification (1–2 pH units) of the rhizosphere over a wide range of root and soil conditions. One is generation of H+ in the oxidation of ferrous iron by O2 released from the roots. The other is release of H+ from roots to balance excess intake of cations over anions, N being taken up chiefly as NH4 +. CO2 exchange between the roots and soil has a much smaller effect. The zone of root-influence extends a few mm from the root surface. There are substantial differences along the root length and with time. The acidification and oxidation cause increased sorption of NH4 + ions on soil solids, thereby impeding the movement of N to absorbing root surfaces. But they also cause solubilization and enhanced uptake of soil phosphate.  相似文献

The effect of potentially toxic concentrations of ammonium on root development of Scots pine seedlings raised on Perlite was investigated during growth periods of 3 or 10 weeks after sowing. It was shown that imbalanced ammonium nutrition led to conspicuous changes of root morphology provided the pH value in the medium was allowed to decrease to 3.9 due to the NH+4-dependent proton excretion into the rhizosphere. Ammonium toxicity could not be observed with seedlings treated either with ammonium nitrate or with ammonium chloride at pH 5.3 ? 6.8. While the supply of NH+4 considerably inhibited root development the biomass production of the shoot was increased. Determination of the endogenous level of ammonium in roots and the leaf whorl exclude a simple causal correlation between ammonium toxicity and accumulated ammonium as has been postulated for herbaceous plants.  相似文献

Black root rot of tobacco, caused by Thielaviopsis basicola, is generally severe at soil pH values >5.6 and suppressed under more acidic conditions (pH < 5.2). Soil acidifying fertilizers containing NH4–N are generally recommended for burley tobacco production in North Carolina, but the effects of N form and application rate on development of black root rot and on the population dynamics of T. basicola have not been determined. Greenhouse and laboratory studies were conducted to evaluate the effects of N form (NH4 + or NO3 ) and rate on pathogen and disease parameters at several initial soil pH levels. A moderately-conducive field soil, initial pH 4.7, was adjusted to a pH of 5.5 or 6.5 by the addition of CaOH2, then amended with the desired nitrogen form and rate. Pathogen populations were determined over time. In addition, spore production in extracts of roots from plants grown in the various nitrogen and pH treatments was determined. Finally, because tobacco responds to acidic soil conditions and exposure to NH4–N by accumulating high concentrations of the polyamine putrescine, the toxicity of putrescine on vegetative growth and reproduction of T. basicola was investigated. Low soil pH and high levels of NH4–N suppressed reproduction of T. basicola in soil and in root extract, while use of NO3–N and depletion of NH4–N resulted in rapid increases in populations of T. basicola. At 20 mM, putrescine inhibited hyphal growth by 60% and aleuriospore production by 98%. Fertilizers that reduced soil pH also reduced reproduction by T. basicola, and thus have potential for management of black root rot by suppressing populations of T. basicola over multiple years of crop production. The suppression of T. basicola and black root rot observed with NH4–N amendments may partially be due to development of an inhibitory environment in the root and not solely to changes in rhizosphere pH.  相似文献

The aim of the present review is to define the various origins of root-mediated changes of pH in the rhizosphere, i.e., the volume of soil around roots that is influenced by root activities. Root-mediated pH changes are of major relevance in an ecological perspective as soil pH is a critical parameter that influences the bioavailability of many nutrients and toxic elements and the physiology of the roots and rhizosphere microorganisms. A major process that contributes root-induced pH changes in the rhizosphere is the release of charges carried by H+ or OH to compensate for an unbalanced cation–anion uptake at the soil–root interface. In addition to the ions taken up by the plant, all the ions crossing the plasma membrane of root cells (e.g., organic anions exuded by plant roots) should be taken into account, since they all need to be balanced by an exchange of charges, i.e., by a release of either H+ or OH. Although poorly documented, root exudation and respiration can contribute some proportion of rhizosphere pH decrease as a result of a build-up of the CO2 concentration. This will form carbonic acid in the rhizosphere that may dissociate in neutral to alkaline soils, and result in some pH decrease. Ultimately, plant roots and associated microorganisms can also alter rhizosphere pH via redox-coupled reactions. These various processes involved in root-mediated pH changes in the rhizosphere also depend on environmental constraints, especially nutritional constraints to which plants can respond. This is briefly addressed, with a special emphasis on the response of plant roots to deficiencies of P and Fe and to Al toxicity. Finally, soil pH itself and pH buffering capacity also have a dramatic influence on root-mediated pH changes.  相似文献

The effects of metabolic and protein synthesis inhibitors on NH4 + uptake by Pisum arvense plants at low (0.05 mM) and high (1 mM) external ammonium concentration were studied. In short-time experiments cycloheximide decreased the ammonium uptake rate at low level of NH4 + and increased the absorption of NH4 + from uptake medium containing high ammonium concentration. Arsenate and azide supplied into uptake solutions at low ammonium concentration strongly decreased or completely suppressed the NH4 + uptake rate, respectively. When the experiments were carried out at high level of ammonium only azide decreased the uptake rate of NH4 + and arsenate stimulated this process. Dinitrophenol very strongly repressed the uptake rate of NH4 + at both ammonium concentrations. After removing dinitrophenol from both solutions, neither at low nor high external ammonium level the recovery of NH4 + uptake rate was achieved within 150 min or 3 h, respectively. The recovery of NH4 + uptake rate after removing azide was observed within 90 min and 3 h at low and high ammonium concentrations, respectively. The regulation of NH4 + uptake by some inhibitors at low external ammonium level was investigated using plasma membrane vesicles isolated from roots by two-phase partitioning. Orthovanadate completely suppressed the uptake of NH4 + by vesicles and quinacrine decreased the NH4 + uptake which 55 suggests that ammonium uptake depends on activities of plasma membrane-bound enzymes. On the other hand, it was found that dinitrophenol completely reduced the NH4 + uptake by vesicles. The various effects of inhibitors on ammonium uptake dependent on external ammonium concentration suggest the action of different ammonium transport systems in Pisum arvense roots. Pisum arvense plants were subjected to 5 days of nitrogen deprivation. Then, in the conditions that increased or decreased the root glutamine and asparagine pools, the uptake rates of 0.5 mM NH4 + and 0.5 mM K+ were examined. The plants supplied with 1 mM glutamine or asparagine took up ammonium and potassium at rates lower than those for the control plants. The uptake rates of NH4 + and K+ were not affected by 1 mM glutamate. When the plants were pre-treated with 100 μM methionine sulphoximine, an inhibitor of glutamine synthesis, the efflux of NH4 + from roots to ambient solution was enhanced. On the other hand, exposure of plants to methionine sulphoximine led to an increase in potassium uptake rate. The addition of asparagine, glutamine or glutamate into the incubation medium caused a decline in the rate of NH4 + uptake by plasma membrane vesicles isolated from roots of Pisum arvense, whereas on addition of methionine sulphoximine increased ammonium uptake. The results indicate that both NH4 + and K+ uptake appear to be similarly affected by glutamine and asparagine status in root cells. Biochar may affect the root morphology and nitrogen (N) use efficiency (NUE) of rice at seedling stage, which has not been clearly verified until now. To clarify it, we conducted a pot experiment regarding to two soil types (Hydragric Anthrosol and Haplic Acrisol), two biochar application rates (0.5 wt% and 1.5 wt %) and two rice varieties (common rice var. Xiushui134 and hybrid super rice var. Zhongkejiayou12-6) meanwhile. Seedling NUE of common rice Xiuhui134 was significantly increased (p < 0.05) by 78.2% in Hydragric Anthrosol and by 91.4% in Haplic Acrisol following biochar addition with 1.5 wt%. However, biochar addition exerted no influence on seedling NUE of super rice Zhongkejiayou12-6 in both soils. Overall, 0.09–0.10 units higher soil pH and 105– 116% higher soil NH4+ -N were observed in Xiushui134 growing two soils with 1.5 wt% biochar. In addition, improved root morphology (including longer root length, larger root surface area, bigger root volume, and more root tips) contributed to the higher seedling NUE of Xiushui134 in two soils. The soil pH and NH4+ -N content, also the root morphology were influenced by biochar, which though could not thoroughly explained the NUE of Zhongkejiayou12-6. In conclusion, biochar application to paddy soil changed soil pH and NH4+ -N content, root growth, and the consequent seedling NUE of rice, which effects are relative with rice cultivar, biochar addition rate, and soil type.  相似文献

Wheat (Triticum aestivum L.) was grown in nutrient solution with low or high N supply (NH4NO3 as N source). To further evaluate the influence of N form and its interaction with the nutrient solution pH, wheat plants were grown with NH 4 + or NO 3 - either in an conventional nutrient solution or in a nutrient solution in which the pH was maintained at pH 6.5 using a pH-stat system. The nutrient solution was inoculated with Pseudomonas fluorescens 2-79RLI, a genetically modified bacterium that contains lux genes activated by a ribosomal promoter. Cell numbers and physiological status of P. fluorescens 2-79RLI (length of the lag phase of bioluminescence) in the rhizosphere were determined at the root tip and in the lateral root zone. Nitrogen deficiency decreased both plant growth and root colonization by P. fluorescens 2-79RLI at the root tip while it had no effect on root colonization in the lateral root zone. The physiological status of P. fluorescens 2-79RLI was not affected by nitrogen deficiency. Ammonium nutrition increased root colonization by P. fluorescens 2-79RLI at the root tip and in the lateral root zone when the pH of the nutrient solution was allowed to change according to the N form provided. Under these conditions, the physiological status of P. fluorescens 2-79RLI was higher in the lateral root zone than at the root tip. In contrast, N source had no effect on root colonization or physiological status of P. fluorescens 2-79RLI in the nutrient solution maintained at pH 6.5. It is concluded that the stimulation of root colonization by NH 4 + in the nutrient solution, not maintained at a constant pH, may be due to increased leakage of solutes into the rhizosphere as a result of impaired exudate retention by high H+ concentration in the rhizosphere or the apoplast. To test the hypothesis that rhizosphere acidification would enhance the hydrolyzation of organic phosphates by increasing phosphatase activity. A Petri dish experiment with sterile agar and a pot experiment with a low P soil were used. In the Petri dish experiment, roots of each plant were cultured in two compartments, each of which contained agar with one of three nitrogen combinations: NH 4 + /N0 (N0 = nitrogen free), NH 4 + /NO 3 - , and NO 3 - /N0. Phytin was supplied as the sole phosphorus (P) source to all compartments. In the pot experiment, the soil in each pot was treated with N0, KNO3, or (NH4)2SO4) together with 0 or 75 mg kg?1 phytin-P. Dry weight, P concentration, and P content of roots were highest in the NH 4 + compartments in the Petri dish experiment. In the pot experiment, dry weight, P concentration, and P content of both shoots and roots were higher with NH 4 + than with NO 3 - . NH 4 + treatments reduced rhizosphere pH, promoted the hydrolization of phytin, enhanced acid phosphatase activity in the rhizosphere, and increased phytin-P utilization relative to N0 and NO 3 - treatments. Phosphatase activity was negatively correlated with rhizosphere pH but was positively correlated with plant P content in both experiments. Rhizosphere acidification optimized the activity of acid phosphatase excreted by maize roots and promoted phytin mineralization. NH 4 + -induced acidification in the maize rhizosphere improved the growth of maize roots by improving P uptake from phytin; the improved growth, in turn, increased NH 4 + uptake and acidification.  相似文献

Two experiments were carried out in a growth chamber to investigate the phosphorus (P)-uptake efficiency of Fagopyrum esculentum Moench (buckwheat) and Triticum aestivum (spring wheat) from a Ca-bound form. The first experiment was based on a sand-culture system with either rock phosphate (RP) or CaHPO4 (CaHP) as the P source and nitrate or ammonium nitrate as nitrogen source. A highly calcareous soil was used in the second experiment. Buckwheat was shown to be highly efficient in taking up Ca-bound P compared to spring wheat. When plants were supplied with nitrate, the total P uptake by buckwheat from RP was nearly 10-fold higher than that of spring wheat (20.1 compared with 2.1 mg P pot–1). Changing nitrogen source from nitrate only to ammonium nitrate increased P uptake by spring wheat substantially, but not buckwheat. High P-uptake efficiency of buckwheat was also demonstrated using the field soil, but to a lesser extent, which may be related to the difference in Zn supply between sand culture and field soil. It is suggested that buckwheat may be included in intercropping or crop rotation systems to activate P sources in calcareous soils. The principal mechanism of P uptake efficiency of buckwheat may be its ability to acidify the rhizosphere; however, further study is needed to unravel the regulation of root excretion of H+ and its molecular basis in order to exploit buckwheat's genetic capability to utilise sparingly soluble P from soil.  相似文献

Summary Relationships between root zone temperature, concentrations and uptake rates of NH 4 + and NO 3 were studied in non-mycorrhizal roots of 4-year-old Norway spruce under controlled environmental conditions. Additionally, in a forest stand NH 4 + and NO 3 uptake rates along the root axis and changes in the rhizosphere pH were measured. In the concentration (Cmin) range of 100–150 M uptake rates of NH 4 + were 3–4 times higher than those of NO 3 The preference for NH 4 + uptake was also reflected in the minimum concentration (Cmin) values. Supplying NH4NO3, the rate of NO 3 uptake was very low until the NH 4 + concentrations had fallen below about 100 M. The shift from NH 4 + to NO 3 uptake was correlated with a corresponding shift from net H+ production to net H+ consumption in the external solution. The uptake rates of NH 4 + were correlated with equimolar net production of H+. With NO 3 nutrition net consumption of H+ was approximately twice as high as uptake rates of NO 3 In the forest stand the NO 3 concentration in the soil solution was more than 10 times higher than the NH 4 + concentration (<100 M), and the rhizosphere pH of non-mycorrhizal roots considerably higher than the bulk soil pH. The rhizosphere pH increase was particularly evident in apical root zones where the rates of water and NO 3 uptake and nitrate reductase activity were also higher. The results are summarized in a model of water and nutrient transport to, and uptake by, non-mycorrhizal roots of Norway spruce in a forest stand. Model calculations indicate that delivery to the roots by mass flow may meet most of the plant demand of nitrogen and calcium, and that non-mycorrhizal root tips have the potential to take up most of the delivered nitrate and calcium.  相似文献

Ammonia-oxidizing bacteria (AOB) populations were studied on the root surface of different rice cultivars by PCR coupled with denaturing gradient gel electrophoresis (DGGE) and fluorescence in situ hybridization (FISH). PCR-DGGE of the ammonium monooxygenase gene (amoA) showed a generally greater diversity on root samples compared to rhizosphere and unplanted soil. Sequences affiliated with Nitrosomonas spp. tended to be associated with modern rice hybrid lines. Root-associated AOB observed by FISH were found within a discrete biofilm coating the root surface. Although the total abundance of AOB on root biofilms of different rice cultivars did not differ significantly, there were marked contrasts in their population structure, indicating selection of Nitrosomonas spp. on roots of a hybrid cultivar. Observations by FISH on the total bacterial community also suggested that different rice cultivars support different bacterial populations even under identical environmental conditions. The presence of active AOB in the root environment predicts that a significant proportion of the N taken up by certain rice cultivars is in the form of NO3 -N produced by the AOB. Measurement of plant growth of hydroponically grown plants showed a stronger response of hybrid cultivars to the co-provision of NH4 + and NO3 . In soil-grown plants, N use efficiency in the hybrid was improved during ammonium fertilization compared to nitrate fertilization. Since ammonium-fertilized plants actually receive a mixture of NH4 + and NO3 with ratios depending on root-associated nitrification activity, these results support the advantage of co-provision of ammonium and nitrate for the hybrid cultivar.  相似文献

Summary Plants that absorb nitrogen as NO3 tend to raise the pH in the rhizosphere. Those absorbing nitrogen as NH4 + or N2 lower the pH. The change in pH near the root surface may be calculated approximately from the H+ or HCO3 efflux and radius of the root; and the pH buffering capacity, moisture content, initial pH and pCO 2 of the soil. An accurate equation, solved numerically, also takes account of root hairs, mass flow and slow acid-base reaction in the soil. The pH at the root surface will often differ from the pH a few mm away by 1–2 units.  相似文献

为验证以下3个假设: 1) NO3 -和NH4 +及其不同供给方式显著影响根系生长; 2) NO3 -和NH4 +以及不同供给方式对根内激素含量影响显著; 3)根构型(1级根长、单位2级根上1级侧根密度(分枝强度)和1级根在2级根上的根间距)与根内激素(生长素(IAA)、脱落酸(ABA)和细胞分裂素(玉米素核苷+玉米素) (CK (ZR + Z))含量显著相关, 采用营养液培养方法, 使实验植物翠菊(Callistephus chinensis)在两种氮肥(NO3 -和NH4 +)、不同施氮浓度(NO3 -: 0.2、1.0和18.0 mmol·L -1; NH4 +: 0.2、4.0和20.0 mmol·L -1), 以及脉冲和稳定两种施用方式处理下生长。在处理35天后收获植物, 测定根系生物量、根系构型指标(根系1级根长、单位2级根上1级侧根数和1级根在2级根上的根间距)和根系中激素含量(IAA、ABA和CK (ZR + Z))。结果显示: 1)实验处理对根生物量和根系中IAA、ABA和CK (ZR + Z)含量均有不同程度的显著影响: 施用NH4 +使根生物量和根内IAA含量显著低于施用NO3 -; 高浓度NO3 -和NH4 +处理亦使根生物量和IAA降低; 相对于稳定处理, 脉冲施氮显著降低根生物量和根内IAA含量; NO3 -使根内CK (ZR + Z)含量显著高于施用NH4 +, 且与施氮浓度及施氮方式无关; NO3 -处理下, 高浓度使根内ABA含量提高, 且脉冲处理使ABA含量升高。NH4 +处理下, 高浓度使根内ABA含量降低, 而施氮方式对其没有显著影响。2)根构型因素与根内激素关系各异: 各激素与1级根间距无显著关系; IAA和CK (ZR + Z)与1级根长和侧根密度有显著回归关系。3)根构型因素与根生物量的关系是根生物量与1级根长和侧根密度有显著正回归关系, 与1级根间距无显著回归关系。实验结果表明翠菊根生长的 “反常”可能是由于其对脉冲高浓度NH4 +耐受阈值低所致。该研究通过实验建立了氮养分种类/供应方式通过改变激素、影响根构型而影响根生长的联系, 进一步探究了植物根养分捕获塑性机制。  相似文献

采用水培法,研究了旱柳苗在外源添加不同氮水平(贫氮、中氮、富氮、过氮)的铵态氮(NH+4-N)和硝态氮(NO-3-N)的生长、氮吸收、分配和生理响应。结果表明:一定范围氮浓度的增加能够促进旱柳苗的生长,但过量氮会抑制其生长,且NH+4-N的抑制作用大于NO-3-N;两种氮处理下,旱柳表现出对NH+4-N的吸收偏好,在同一氮水平时,旱柳各部位氮原子百分含量Atom%15N(AT%)、15N吸收量和来自氮源的N%(Ndff%)均为NH+4-N处理大于NO-3-N处理,且随着氮浓度的增加,差异增大,且在旱柳各部位的分布为根﹥茎﹥叶;2种氮素过量和不足均会对旱柳根和叶生理指标产生不同的影响,其中在过氮水平时,NH+4-N和NO-3-N处理下根系活力比对照减少了50.61%和增加了19.53%;在过氮水平时,NH+4-N处理柳树苗根总长、根表面积、根平均直径、根体积和侧根数分别对照下降了30.92%、29.48%、19.44%、27.01%和36.41%,NO-3-N处理柳树苗相应的根系形态指标分别对对照下降了1.66%、5.65%、1.49%、5.06%和25.72%。可见,高浓度NH+4-N对旱柳苗的胁迫影响大于NO-3-N,在应用于水体氮污染修复时可通过改变水体无机氮的比例,削弱其对旱柳的影响,从而提高旱柳对水体氮污染的修复效果。  相似文献

French bean seedlings grown on choline, ammoniacal and nitrate forms of nitrogen together with equivalent basal application of P as KH2PO4 were tested for nutrient uptake from the rhizosphere. Statistical tests on soil (rhizosphere and non-rhizosphere) and plant (root and shoot) revealed that with the exception of P, levels of all other estimated macro-(Na+, K+, Ca2+, Mg2+) and micro-nutrients (Fe2+, Mn2+, Zn2+) were significantly changed after 42 days growth as compared to 21 days growth period. The higher uptake into shoots of Na+, K+, Fe2+, Mn2+, Zn2+ and H2PO4 and higher biomass accumulation in the rhizosphere were associated with lower rhizosphere pH. The uptake of Ca2+ and Mg2+ increased with higher rhizosphere pH. While ammoniacal and choline forms decreased rhizosphere pH and increased the P uptake, nitrate form reversed the trend showing significant inverse relationship between shoot phosphate and rhizosphere pH. Calcium and iron were associated with an inhibition of the translocation of P from root to shoot. However, no causal relationships could be established. Both shoot weight and shoot P content were closely associated with a number of rhizosphere soil parameters. The paper forms a part of the Ph. Rising atmospheric carbon dioxide (CO2) is expected to increase forest productivity, resulting in greater carbon (C) storage in forest ecosystems. Because elevated atmospheric CO2 does not increase nitrogen (N) use efficiency in many forest tree species, additional N inputs will be required to sustain increased net primary productivity (NPP) under elevated atmospheric CO2. We investigated the importance of free amino acids (AAs) as a source for forest N uptake at the Duke Forest Free Air CO2 Enrichment (FACE) site, comparing its importance with that of better‐studied inorganic N sources. Potential proteolytic enzyme activity was monitored seasonally, and individual AA concentrations were measured in organic horizon extracts. Potential free AA production in soils ranged from 190 to 690 nmol N g−1 h−1 and was greater than potential rates of soil NH4+ production. Because of this high potential rate of organic N production, we determined (1) whether intact AA uptake occurs by Pinus taeda L., the dominant tree species at the FACE site, (2) if the rate of cycling of AAs is comparable with that of ammonium (NH4+), and (3) if atmospheric CO2 concentration alters the aforementioned N cycling processes. A field experiment using universally labeled ammonium (15NH4+) and alanine (13C3H715NO2) demonstrated that 15N is more readily taken up by plants and heterotrophic microorganisms as NH4+. Pine roots and microbes take up on average 2.4 and two times as much NH4+ 15N compared with alanine 15N 1 week after tracer application. N cycling through soil pools was similar for alanine and NH4+, with the greatest 15N tracer recovery in soil organic matter, followed by microbial biomass, dissolved organic N, extractable NH4+, and fine roots. Stoichiometric analyses of 13C and 15N uptake demonstrated that both plants and soil microorganisms take up alanine directly, with a 13C : 15N ratio of 3.3 : 1 in fine roots and 1.5 : 1 in microbial biomass. Our results suggest that intact AA (alanine) uptake contributes substantially to plant N uptake in loblolly pine forests. However, we found no evidence supporting increased recovery of free AAs in fine roots under elevated CO2, suggesting plants will need to acquire additional N via other mechanisms, such as increased root exploration or increased N use efficiency.  相似文献

Abstract. An accepted view, that unless nitrate concentrations in the soil solution are very low (e.g. below 0.1–0.2 mol m?3) the growth of high-yielding crops is not limited by the availability of nitrogen, is challenged. Conventional analyses of nutrient supply and demand, based on calculations of apparent inflow rates (uptake rates per unit total root length) are invalid. Apparent inflow rates are inversely proportional to root length. The convention of using total root length grossly overestimates the fraction of the root system active in nutrient uptake. Consequently, inflow rates based on total root lengths underestimate the true values, indicating unrealistically low nutrient concentration differentials between bulk soil and root surfaces required to drive uptake. An alternative method of analysis is suggested. This is based on total nutrient uptake rather than on inflow rate. Measurements of the former do not depend on estimates of active root length and can be made directly and reliably. The method was applied to data obtained from a pot experiment using spring wheat (Triticum aestivum L., cv. Wembley) grown in soil without nitrogen fertilizer (N0) or with nitrogen fertilizer equivalent to 200kg N ha?1 (N+). Soil nitrate concentrations calculated using the conventional method based on total root length, suggested that any increases in concentration above those measured in the N0 treatment should not have resulted in increased uptake and growth. However, the N+ plants were always bigger than those in the No treatment, refuting this suggestion. Theoretical uptakes of nitrogen (calculated initially on the basis of a fully active root system) were adjusted, by reducing the effective root length incrementally, until the theoretical uptake matched the measured net uptake of nitrogen. The mean fractions of the root systems likely to have been involved in nitrate uptake were 11% and 3.5% of the total lengths of root in the N0 and N+ treatments, respectively.  相似文献
