Application of juvenile hormone esterase inhibitor 3-octylthio-1,1,1- trifluropropan-2-one (OTFP) to 5th instar nymphs and virgin females of D. cingulatus revealed the profound role played by juvenile hormone esterase (JHE) in metamorphosis and reproduction. The ability of OTFP to cause delay and the formation of malformed nymphs, suggests that inhibition of JHE in vivo maintains a higher than normal hemolymph JH titer. It is obvious that OTFP does inhibit in vivo JHE activity in late instar nymphs. Further, the application of JHE inhibitor, OTFP to virgin females demonstrates that substituted trifluropropanones can indirectly stimulate egg development by inhibiting JHE activity in virgin females. 相似文献

Abstract: Mass-reared reduviid, Rhynocoris marginatus was released in large-sized cotton field cages against three cotton pests, Spodoptera litura, Mylabris pustulata and Dysdercus cingulatus . Pest species were introduced into separate cages in the absence of other pests, parasitoids and predators. Control experimental cages without R. marginatus were maintained for each prey set up during the evaluation period. R. marginatus greatly reduced the infestation of S. litura (57.5%), M. pustulata (52.3%) and D. cingulatus (45.8%). The leaves, flower and boll damages (32%, 35% and 28% by S. litura, M. pustulata and D. cingulatus , respectively) and seed-cotton yield loss (1.4, 1.6 and 1.25 times in S. litura, M. pustulata and D. cingulatus infested cages, respectively) were reduced by R. marginatus compared with such fields without R. marginatus . The field observations suggest this predator's pest suppression and damage reduction efficacy. 相似文献

Juvenile hormone esterase (JHE), a selective enzyme that hydrolyzes the methyl ester of insect juvenile hormone plays an important role in regulating metamorphosis in nymphs as well as reproduction in adults. Studies on JH degradation provide insight into the possibilities of physiological disruption in the insects. In the present study, the JH degrading enzyme, JHE from the cotton pest Dysdercus cingulatus (Heteroptera) is characterized. Electrophoretic analysis of haemolymph during various developmental stages showed the JHE bands prominent only on the final day of 5th instar nymph, and the esterase substrate specificity confirmed the presence of JHE isoforms. In an attempt to clone cDNA of JHE gene from the final instar nymphs, mRNA isolated from fat bodies was coupled with JHE gene-specific primers and the cDNA was synthesized using RT-PCR. The PCR amplified cDNA showed the presence of JHE isoforms in D. cingulatus. 相似文献

Investigations were carried out to determine the role of juvenile hormone (JH) and 20-hydroxy ecdysone in the synthesis and uptake of vitellogenins, which were earlier identified, purified and characterised, in Dysdercus koenigii. The concentration(s) of vitellogenin(s) in fat body, haemolymph and that of vitellin(s) in ovary were significantly lower after chemical allatectomy at eclosion. In addition, at 70 h after emergence, chemical allatectomy reduced ovarian vitellin concentration, but vitellogenin levels remained normal in the fat body and haemolymph. The haemolymph vitellogenins were not incorporated into oocytes in such insects. Administration of JH-III at 20 h after allatectomy restored vitellogenin levels in the fat body and haemolymph, but the ovary failed to incorporate the available vitellogenins from haemolymph in such insects. However, when JH-III was administered twice, one at 20 h and then at 70 h after allatectomy, vitellogenin concentrations in fat body and haemolymph and also vitellin concentrations in ovary approached control levels. It is suggested that JH has two separate roles, one in vitellogenin synthesis and the other in uptake. 20-hydroxy ecdysone had no apparent role in either vitellogenin synthesis or uptake in D. koenigii. 相似文献

ABSTRACT. Allatectomy did not affect either food ingestion or midgut protease and invertase activity in adult female Dysdercus cingulatus Fabr., but ingestion of methoprene (juvenile hormone analogue) stimulated food consumption in the late instar caterpillars of Hyblaea puera Cramer. Implantation of brain to raise the insect's own level of brain neurosecretion also stimulated food ingestion in Hyblaea. Caterpillars which either ingested methoprene or received brain-implants showed higher levels of midgut amylase activity. However, no increase in amylase occurred after ingestion of methoprene or implantation of brain followed by starvation. A probable mechanism of hormonal influence on food ingestion and enzyme activity is discussed in the light of these results. 相似文献

Decapitated blood-fed Aedes aegypti mosquitoes do not undergo normal oöcyte maturation. Topical application of 1.25 ng JH analogue (ZR 515) or 250 ng JH-I restored ovarian development in 70–80% of the treated females. The rate of vitellogenin synthesis in these animals was 80% of normal blood-fed controls.When ligated abdomens were treated, 125 pg ZR 515 or 12.5 ng JH-I were sufficient to restore ovarian development in 80% of the animals. The rate of vitellogenin synthesis in these animals was 70% of normal blood-fed controls. On the other hand, injection of 1.25 μg 20-hydroxyecdysone was needed to restore ovarian development and vitellogenin synthesis in decapitated and abdominally ligated females.These experiments indicate that JH concentrations closer to the physiological norm than 20-hydroxyecdysone, can restore ovarian development and vitellogenin synthesis in vivo. 相似文献

The chemosterilant activity of two naturally occurring napthaquinones, plumbagin from Plumbago sp. and juglone from Juglans regia has been evaluated using the red cotton bug Dysdercus koenigii . Their activity were compared with synthetic napthaquinones, menadione, two benzoquinones, 2,6-dimethyl and 2,3,6-trimethylbenzoquinone and a hydroquinone 2,6-dimethylhydroquinone. As far as the authors are aware, the present investigation is the first systematic attempt to investigate the effects of quinones on different aspects of reproduction, namely mating behaviour, fecundity and fertility. All the above types of quinones have revealed adverse effects on the reproduction of D. koenigii , which were topically treated as 1-day-old adults with different doses of the compounds depending on their 50% lethality (LD50 ) values. Analysis of the data revealed a highly statistically significant reduction in the number of eggs, their hatching and further development up to the final instar stage. The sterility index increased with the increase in the dose of any of the above compounds and was highest when both sexes were treated. Among the natural products, juglone induced more sterilizing effects than plumbagin at 5–10 μ g/insect. Menadione caused sterility at a very low dose such as 0.5 μ g. Of the two benzoquinones, the dimethylbenzoquinone acted at a much lower dose. 相似文献

Juvenile hormone esterase (JHE) is the primary juvenile hormone (JH) metabolic enzyme in insects and plays important roles in the regulation of molt and metamorphosis. We investigated its mRNA expression profiles and hormonal control in Bombyx mori larvae. JHE mRNA was expressed at the end of the 4th and 5th (last) larval instars in the midgut and in all the three (anterior, middle, posterior) parts of the silk gland. In the fat body, JHE expression peaked twice in the 5th instar, at wandering and before pupation, while it gradually decreased through the 4th instar. When 20-hydroxyecdysone (20E) was injected into mid-5th instar larvae, JHE mRNA expression was induced in the anterior silk gland but suppressed in the fat body. Topical application of a juvenile hormone analog fenoxycarb to early-5th instar larvae induced JHE expression in both tissues. In the anterior silk gland, JHE expression was accelerated and strengthened by 20E plus fenoxycarb treatments compared with 20E or fenoxycarb single treatment, indicating positive interaction of 20E and JH. JHE mRNA is thus expressed in tissue-specific manners under the control of ecdysteroids and JH. 相似文献

The effect of an experimentally increased octopamine content (feeding flies with OA) on the levels of juvenile hormone (JH) degradation, dopamine (DA), and 20-hydroxyecdysone (20E) contents, oogenesis, and fecundity of wild type Drosophila flies has been studied. OA feeding of the flies was found to (1) cause a considerable decrease in JH degradation in females, but not males, of D. melanogaster and D. virilis; (2) have no effect on DA content in D. melanogaster and D. virilis; (3) increase 20E contents in D. virilis females; (4) decrease to a large extent the number of vitellogenic (stages 8-10) and mature (stage 14) oocytes in D. virilis; and (5) decrease the fecundity of D. melanogaster and D. virilis. A possible mechanism of action of OA as a neurohormone on the reproductive function of Drosophila is discussed. 相似文献

Bacterial seed and boll rot is a newly emerging cotton disease in Pakistan. Twenty-one cotton varieties were screened to find resistance source against the disease. None of these was found to be resistant. Five cotton varieties (CIM-595, MK2, BT-986, BT-986 & SG-1) having 700–1400 Area under disease progress curve (AUDPC) units were found to be moderately resistant to the disease. SLH-317, FH-942, BT-222, BT-666, MNH-457 ranging from 1401–1700 AUDPC units were moderately susceptible while MNH-456, SLH-336, 9811, FH-942, MNH-886 susceptible to boll rot. Seven varieties (FH-114, FH-113, BT-7, BT-212, SLH-BT-4, BT-212 and FH-941) were highly susceptible to bacterial seed and boll rot indicated by 2001–2300 AUDPC units. Biochemical tests identified bacterial isolates as Pantoea agglomerans. Different inoculation techniques were assessed for bacterial pathogenicity and symptoms of boll rot were only observed in needle punctured bolls. One, two and three weeks old bolls were mechanically inoculated by injecting bacterial suspension to evaluate the boll’s age impact on disease severity. Maximum severity was observed in two weeks old bolls. Red cotton bugs (Dysdercus cingulatus) were fed on artificially inoculated diseased bolls and then transferred on healthy bolls. Diseased symptoms were noticed on healthy cotton bolls. Bacterial colonies were recovered and red cotton bug was confirmed as the disease-transmitting vector. 相似文献

Summary By use of osmium ferricyanide (OsFeCN) staining the fate of cytoplasmic membranes was followed during spermiogenesis in the cotton seed bug (Dysdercus intermedius). During early spermiogenesis interzonal lamellae of endoplasmic reticulum become aggregated as a stack of membranes traversing the entire cell body from the nucleus to the cytoplasmic bridge connecting neighbouring spermatids. Cisternae of endoplasmic reticulum ensheath the acroblast from which vesicles of different sizes are pinched off into the cytoplasm. The oxalate method was used to show that acroblast and associated vesicles are calcium-sequestering sites in spermatids. Membrane profiles with dense calcium oxalate precipitate derived from the acroblast form an uninterrupted membranous sheath at the apical side of the nucleus where the proacrosome will be attached. With further development of the spermatids, the vesicles derived from the acroblast also participate in forming a calciumsequestering sheath enveloping the axoneme and the mitochondrial nebenkern derivatives. 相似文献

Bacterial seed and boll rot disease is a newly emerging threat to the cotton growers. Disease prediction model was devised to predict the disease progression impacted by the vector (Dysdercus cingulatus) and environmental variables (maximum air temperature, minimum air temperature, relative humidity and rainfall) on four varieties to minimise its losses and disease management cost. Impact of a-biotic environmental variables (maximum and minimum air temperature, relative humidity and rainfall) was assessed on bacterial seed and rot of cotton disease and its vector (D. cingulatus) on FH-941, FH-942, MNH-886 and FH-114 cotton varieties. Maximum red cotton bug population was assessed at 29–31 °C maximum temperature and at 15–17 °C minimum temperature. Disease severity was noticed maximum when maximum and minimum temperature was measured at 28–29 °C and 13–14.5 °C, respectively. Vector population was maximum when relative humidity and rainfall were 63–66% and 1.50–2.5 mm, respectively. Relative humidity at 66–68% and 0.5–1.5 mm rainfall favoured disease development. With increase in number of bugs, increase in disease severity was noted, maximum disease severity 45–48% noticed when 7–8 bugs were recorded. Red cotton bug (Dysdercus cingulatus) population prediction model was devised based on a-biotic factors (maximum and minimum air temperature, relative humidity and rainfall) on four cotton varieties. Disease forecasting model was developed based on biotic (D. cingulatus) and a-biotic factors. A close resemblance between observed and the predicted red cotton bugs and disease severity was seen. 相似文献

Sensitivity to juvenile hormone and to 20-hydroxyecdysone has been investigated during the last-larval stages of Tenebrio molitor. Topical applications of a juvenile hormone analogue (K-421d) showed that the sensitive period, occurring before apolysis, is relatively short (less than 4 days in a 3-week instar) and divided into two phases. Treatment during the first and longest phase induced a delay in development and then an increase in larval moult percentage. Treatment during the second phase induced several abnormal moults (prothetelic larvae and larval-pupal intermediates).Injections of massive doses of 20-hydroxyecdysone (10 μg per animal) also evidenced a period of disturbance of the morphogenetic programme, beginning before pupal apolysis but continuing several days after.Comparison of the sensitive periods to both hormones suggests that a very important and rapid step of the larval-pupal programme change is controlled hormonally just before pupal apolysis. 相似文献
