Gemins 2-8 and Unr-interacting protein (UNRIP) are intimate partners of the survival motor neuron (SMN) protein, which is the determining factor for the neuromuscular disorder spinal muscular atrophy (SMA). The most documented role of SMN, Gemins and UNRIP occurs within the large macromolecular SMN complex and involves the cytoplasmic assembly of spliceosomal uridine-rich small nuclear ribonucleoproteins (UsnRNPs), a housekeeping process critical in all cells. Several reports detailing alternative functions for SMN in either motor neurons or skeletal muscles may, however, hold the answer to the extreme neuromuscular tissue specificity observed in SMA. Recent discoveries indicate that collaboration between SMN and Gemins also extends to these non-canonical functions, hence raising the possibility that mutations in Gemin genes may be the cause of unlinked neuromuscular hereditary syndromes. This review evaluates the functions of Gemins and UNRIP inside the SMN complex and discusses whether these less notorious SMN complex members are capable of acting independently of SMN.  相似文献

alpha2-Macroglobulin (alpha2M) has been identified as a carrier protein for beta-amyloid (Abeta) decreasing fibril formation and affecting the neurotoxicity of this peptide. The alpha2-macroglobulin receptor/low density lipoprotein receptor related protein (LRP) is involved in the internalization and degradation of the alpha2M/Abeta complexes and its impairment has been reported to occur in Alzheimer's disease. Previous studies have shown alpha2M to determine an enhancement or a reduction of Abeta toxicity in different culture systems. In order to clarify the role of alpha2M in Abeta neurotoxicity, we challenged human neuroblastoma cell lines with activated alpha2M in combination with Abeta. Our results show that in neuroblastoma cells expressing high levels of LRP, the administration of activated alpha2M protects the cells from Abeta neurotoxicity. Conversely, when this receptor is not present alpha2M determines an increase in Abeta toxicity as evaluated by MTT and TUNEL assays. In LRP-negative cells transfected with the full-length human LRP, the addition of activated alpha2M resulted to be protective against Abeta-induced neurotoxicity. By means of recombinant proteins we ascribed the neurotoxic activity of alpha2M to its FP3 fragment which has been previously shown to bind and neutralize transforming growth factor-beta. These studies provide evidence for both a neuroprotective and neurotoxic role of alpha2M regulated by the expression of its receptor LRP.  相似文献

已有的各个物种概念对物种的认识类似盲人摸象, 只包含了物种的某一个方面; 而一个分化后期的成熟物种应涵盖了所有的物种概念。但是, 尚未到达分化后期的物种往往又已开始新一轮的物种分化; 自然中存在的多数“物种”处于分化路上。这种循环往复连续分化产生的物种, 存在种间生殖隔离不彻底、基因流频繁发生、网状进化突出等现象。此外, 对于不同的物种对, 最早开始分化的基因以及不同物种概念所要求的条件的分化顺序不是统一的, 而是随机的。定义一个适合所有“分化路上的物种”概念存在较大困难。但是, 应采用尽可能多的物种概念来界定分化路上的物种、发表新种和进行分类处理; 也应承认种间可能广泛存在不完全的生殖隔离和有限的基因流, 即有不属于两个物种群体的杂交或回交个体的存在。这样划分的物种比只依据一个物种概念认定的物种具有更高的客观性和科学性。  相似文献

现代栽培马铃薯(Solanum tuberosum)是同源四倍体, 基因组高度杂合, 遗传组成复杂, 存在严重的自交衰退, 优异性状难以聚合, 使得育种周期漫长, 造成基于种子传代的马铃薯杂交育种难以突破。与此同时, 块茎无性繁殖导致马铃薯繁殖系数低、储运成本高和易携带病虫害等痼疾, 且种薯脱毒成本高, 限制其产业化发展。近期, 中国农业科学院深圳农业基因组研究所黄三文团队运用基因组设计理论和方法体系培育杂交马铃薯, 用二倍体育种替代四倍体育种, 实现了用杂交种子繁殖替代薯块繁殖, 创制了第1代高纯度的二倍体自交系和杂种优势明显的杂交马铃薯品系。该研究是马铃薯育种领域里程碑式的重大成果, 开启了基于基因组设计和种子迭代的马铃薯生物育种新纪元。  相似文献

Quercetin and other flavonoids have been reported to exhibit both antioxidant and pro‐oxidant properties. Most studies about the pro‐oxidative ability were conducted in the presence of metal ions, and the essential functional moiety of quercetin responsible for the pro‐oxidative effect is still unclear. In this study, we evaluated the pro‐oxidative abilities in the absence of metal ions of two quercetin derivatives, i.e., quercetin‐3′‐O‐β‐D ‐glucoside ( 1 ) and quercetin‐3‐Oβ‐D ‐glucoside ( 2 ), by assessing DNA cleavage and HO.‐radical production. The binding mode between these compounds and DNA was studied by fluorescence and viscometric titrations. The results showed that 1 can efficiently induce oxidative damage to plasmid DNA, while 2 shows poor activity. Both 1 and 2 bind to DNA via groove‐binding. These results proved that the α‐hydroxy‐β‐oxo‐α,β‐enone moiety contributes to the pro‐oxidative activity of quercetin.  相似文献

2015年年初,美国总统奥巴马在国情咨文中提出了一个预算2.15亿美元的“精准医学计划”,希望以此“引领一个医学时代”。新闻一经发布,“精准医学”立刻成为了媒体和百姓嘴边的热词,受此影响国内亦有不少人士纷纷为美国总统的这一计划点“赞”。有人用“医学革命”来形容它,有人用“开创性”来抬高它,还有一个传闻,受奥巴马“精准医学计划”的影响,中国将在15年内投入600亿元人民币启动并发展中国版的“精准医学计划”。对此,有人提出了质疑,美国版精准医学计划是否符合中国国情?是否存在“水土不服”的可能?直接套用美国总统的智慧能否解决具有中国特色的实际问题?争论由此引发一个让人思考的问题,究竟什么才是现代医学的核心?在盲目堆钱的行动前,我们确实有必要从科学和临床应用的角度来探讨和思考一下现代医学的发展方向。 为了能“精准”地看到问题的实质,我将从当下时髦并且相关的词汇谈起,通过梳理,期待找出解决人类健康问题的真正钥匙。  相似文献

Entomophilous flowers form the food resources for insect pollinators. Many pollinator species forage at the landscape scale and depend on floral resources that are highly variable in space and time. We present a general model approach in which the floral resources of plant communities are estimated by the floral phenology and the cover of entomophilous plant species. We applied this landscape model in a case study for three landscape sections (1.5–2.2 km2) with strongly differing land-use patterns. The comparison between a conservation area and two agricultural landscapes shows extreme differences in the quantities and in the course of floral resources.

In a stepwise simplification of the landscape model we tested the effects of input data with lower spatio-temporal resolution. Even if input data for floral phenology and vegetation have a low resolution, the landscape model allows a ranking of landscape-specific floral resource potentials. The results of the case study encourage the use of landscape models to estimate floral resource potentials. The assessment of floral resource potentials may help to define this essential landscape quality for evaluation in practical nature conservation.  相似文献

香菇‘申香1644’是以传统优质栽培品种‘申香215’为亲本,采用多孢自交育种技术选育的新品种。其菌盖纵切面呈凸形,菌盖直径(6.15±0.38) cm,菌盖厚度(2.27±0.42) cm,菇型圆整,菇质紧实,产量高,生物学转化率95%以上。与亲本相比,‘申香1644’在分子标记和栽培性状上均具有明显差异性,其菌盖为浅黄褐色,颜色较亲本浅;菌龄100-105 d,较亲本缩短5-10 d。‘申香1644’菌丝生长适宜温度为22-26 ℃,原基发育适宜温度为16-22 ℃,可在全国范围内进行代料栽培。  相似文献

SUMMARY In the early decades of the nineteenth century, the most important disagreement among comparative anatomists was not evolution versus "special creation" but between advocates of "transcendental morphology" and those of teleological anatomy—form versus function. In France this dichotomy was represented by the 1830–1832 public debate between Geoffroy St.-Hilaire (form) and Cuvier (function). Geoffroy's aim was to establish links of homology (known to him as "analogies") between the four "embranchements" into which Cuvier had divided the animal kingdom. Despite the fanciful nature of some of his homologies, Geoffroy, who was guided by his " principe de connections ," set in motion a school of morphology, some of whose conclusions, notably the homology of the dorsal surface of segmented invertebrates with the ventral surface of vertebrates, has been corroborated by recent studies in developmental genetics.  相似文献

The question raised in the title was answered. (3R, 3'S)-meso-Zeaxanthin was submitted to iodine catalyzed photochemical stereoisomerisation. The enantiomeric (9Z) and (9'Z) geometrical isomers were isolated by semipreparative HPLC and separated as diastereomeric dicarbamates on a chiral column only. Cleavage of the carbamate could not be effected. CD-Spectra of (1"S, 1"S)- and (1"R, 1"R)-dicarbamates of geometrical isomers of (3R, 3'R)- and (3R, 3'S)-meso-zeaxanthin were systematically studied and the contribution from the carbamate moieties revealed. It was concluded that (9Z, 3R, 3'S)-"meso"-zeaxanthin, in spite of having no symmetry elements, is optically inactive. The result has been rationalised in line with the current hypothesis on the origin of carotenoid CD spectra.  相似文献
