Aim We investigate the population genetic structure of the Maghrebian bat, Myotis punicus, between the mainland and islands to assess the island colonization pattern and current gene flow between nearby islands and within the mainland. Location North Africa and the Mediterranean islands of Corsica and Sardinia. Methods We sequenced part of the control region (HVII) of 79 bats across 11 colonies. The phylogeographical pattern was assessed by analysing molecular diversity indices, examining differentiation among populations and estimating divergence time. In addition, we genotyped 182 bats across 10 colonies at seven microsatellite loci. We used analysis of molecular variance and a Bayesian approach to infer nuclear population structure. Finally, we estimated sex‐specific dispersal between Corsica and Sardinia. Results Mitochondrial analyses indicated that colonies between Corsica, Sardinia and North Africa are highly differentiated. Within islands there was no difference between colonies, while at the continental level Moroccan and Tunisian populations were highly differentiated. Analyses with seven microsatellite loci showed a similar pattern. The sole difference was the lack of nuclear differentiation between populations in North Africa, suggesting a male‐biased dispersal over the continental area. The divergence time of Sardinian and Corsican populations was estimated to date back to the early and mid‐Pleistocene. Main conclusions Island colonization by the Maghrebian bats seems to have occurred in a stepping‐stone manner and certainly pre‐dated human colonization. Currently, open water seems to prevent exchange of bats between the two islands, despite their ability to fly and the narrowness of the strait of Bonifacio. Corsican and Sardinian populations are thus currently isolated from any continental gene pool and must therefore be considered as different evolutionarily significant units (ESU).  相似文献

Understanding how the skull transmits and dissipates forces during feeding provides insights into the selective pressures that may have driven the evolution of primate skull morphology. Traditionally, researchers have interpreted masticatory biomechanics in terms of simple global loading regimes applied to simple shapes (i.e., bending in sagittal and frontal planes, dorsoventral shear, and torsion of beams and cylinders). This study uses finite element analysis to examine the extent to which these geometric models provide accurate strain predictions in the face and evaluate whether simple global loading regimes predict strains that approximate the craniofacial deformation pattern observed during mastication. Loading regimes, including those simulating peak loads during molar chewing and those approximating the global loading regimes, were applied to a previously validated finite element model (FEM) of a macaque (Macaca fascicularis) skull, and the resulting strain patterns were compared. When simple global loading regimes are applied to the FEM, the resulting strains do not match those predicted by simple geometric models, suggesting that these models fail to generate accurate predictions of facial strain. Of the four loading regimes tested, bending in the frontal plane most closely approximates strain patterns in the circumorbital region and lateral face, apparently due to masseter muscle forces acting on the zygomatic arches. However, these results indicate that no single simple global loading regime satisfactorily accounts for the strain pattern found in the validated FEM. Instead, we propose that FE models replace simple cranial models when interpreting bone strain data and formulating hypotheses about craniofacial biomechanics.  相似文献

The plains topminnow Fundulus sciadicus is a freshwater killifish endemic to the Great Plains of North America. Rising concerns for future viability of this species have prompted recent changes in its conservation status. In this study, the results of a range-wide population genetic survey based on the sequence of entire mitochondrial control region (CR) are presented. A total of 181 fish were collected from 11 sites in Nebraska and 10 sites in Missouri spanning the distribution range of the species. Seven polymorphic sites were found in the 966 base pairs of the CR, and only nine unique haplotypes were detected among all fish. Phylogenetic analysis and statistical parsimony networks identified two distinct clades. The first included fish in the Osage, Gasconade and Spring River drainages in Missouri, while the second included individuals from Nebraska and the Lamine River in Missouri, although the Lamine River is much closer to the other Missouri sites than to the Nebraska sites. Pair-wise F ST and average population distances indicated that populations from most drainages are genetically distinct, as 93% of the total molecular variance was attributed to among-drainage effects. Four sites within the main distributions of this species and a highly disjunct site from the south-western portion of the range are suggested as potential targets for conservation.  相似文献

Genetic variation and population structure of hair crab (Erimacrus isenbeckii) were examined using nucleotide sequence analysis of 580 base pairs (bp) in the 3′ portion of the mitochondrial cytochrome c oxidase subunit I gene (COI) of 20 samples collected from 16 locales in Japan (the Hokkaido and Honshu Islands) and one in Korea. A total of 27 haplotypes was defined by 23 variable nucleotide sites in the examined COI region. Pairwise population F ST estimates and neighbor-joining tree inferred distinct genetic differentiation between the representative samples from the Pacific Ocean off the Eastern Hokkaido Island and the Sea of Japan, while others were intermediate between these two groups. AMOVA also showed a weak but significant differentiation among these three groups. The present results suggest a moderate population structure of hair crab, probably influenced by high gene flow between regional populations due to sea current dependent larval dispersal of this species.  相似文献

The peculiar bioclimatic and geographic features of Corso–Sardinian islands may provide an ideal scenario for investigating microevolutionary processes, given their large heterogeneity of environments, which could affect dispersal and gene flow among populations, as well as processes of local adaptation. The genetic variation and differentiation among populations of the endemic lizard Archaeolacerta bedriagae were studied by allozyme electrophoresis at 20 presumptive loci. The genetic structure of this species is characterized by relatively high levels of polymorphism and low differentiation among populations. The pattern of genetic differentiation cannot be explained by genetic drift as a function of geographic distance. Genetic distance data show that genetic variation is distributed into three geographically coherent population groups and suggest a recent (Late Pleistocene) origin for the observed geographic fragmentation. The analysis of environmental correlates of allozymic variation indicates a strong correlation of the Idh-1 locus with climatic variables. The frequency of the Idh-1106 allele is negatively correlated with annual temperature, and positively correlated with annual precipitation. In addition, the observed heterozygosity at this locus decreases towards more arid climatic regimes. The results obtained support the assumption of differential selection acting on Idh-1 allozymes under diverse climates. An association between Idh-1 allozymes and local bioclimatic regimes was also observed for the sympatric lizard Podarcis tiliguerta , suggesting a key role for such selective agents on Idh-1 polymorphism in these two Corso–Sardinian lacertids.  © 2009 The Linnean Society of London, Biological Journal of the Linnean Society , 2009, 98 , 661–676.  相似文献

North American freshwater mussel species have experienced substantial range fragmentation and population reductions. These impacts have the potential to reduce genetic connectivity among populations and increase the risk of losing genetic diversity. Thirteen microsatellite loci and an 883 bp fragment of the mitochondrial ND1 gene were used to assess genetic diversity, population structure, contemporary migration rates, and population size changes across the range of the Sheepnose mussel (Plethobasus cyphyus). Population structure analyses reveal five populations, three in the Upper Mississippi River Basin and two in the Ohio River Basin. Sampling locations exhibit a high degree of genetic diversity and contemporary migration estimates indicate that migration within river basins is occurring, although at low rates, but there is no migration is occurring between the Ohio and Mississippi river basins. No evidence of bottlenecks was detected, and almost all locations exhibited the signature of population expansion. Our results indicate that although anthropogenic activity has altered the landscape across the range of the Sheepnose, these activities have yet to be reflected in losses of genetic diversity. Efforts to conserve Sheepnose populations should focus on maintaining existing habitats and fostering genetic connectivity between extant demes to conserve remaining genetic diversity for future viable populations.  相似文献

This study used allozyme and mtDNA variation to examine genetic structure in Rhadinocentrus ornatus and Hypseleotris compressa in southern Queensland. The aim of this study was to test the hypothesis that dispersal between coastal streams was greater in R. ornatus than in H. compressa and to test whether R. ornatus would reflect a similar pattern seen in another obligate freshwater fish in the same region, that is, R. ornatus will show genetic homogeneity among streams of the Sunshine Coast but significant differentiation among all other streams. Evidence from the current study suggests that, in R. ornatus, there was significant genetic differentiation among populations within regions, which explains why this species has very restricted dispersal abilities between streams, even between those very close to one another. On the other hand, H. compressa showed very low levels of genetic differentiation suggesting some gene flow among regions by virtue of dispersal. High levels of genetic differentiation were identified between all coastal streams in the area, including those hypothesized to have been recently connected in the Sunshine Coast area in the case of R. ornatus.  相似文献

The wreckfish Polyprion americanus , a large [>1 m total length ( L T)] demersal teleost, is distributed globally in temperate waters, including both sides of the North and South Atlantic Oceans, the Mediterranean, the western South Pacific, and the southern Indian Ocean. Wreckfish spawn off the south-eastern U.S. on an area of the Blake Plateau (the Charleston Bump) characterized by an extensive ridge having approximately 100 m relief, in 450–600 m depths. Juvenile wreckfish (<60 cm L T) are pelagic and, in the North Atlantic, are not reported from the Blake Plateau fishing area, but occur in by-catch and fishery landings in the eastern Atlantic. Analysis of nine restriction fragment length profiles from a PCR-amplified fragment (∼1.5 kb) of the ND1 mitochondrial gene indicated no stock separation between eastern North Atlantic (Azores, Majorca, Madeira), and western North Atlantic (Blake Plateau) wreckfish. Restriction site differences separate western South Atlantic wreckfish from the North Atlantic; however, South Atlantic wreckfish share restriction-site similarities with western Pacific wreckfish that are not shared with North Atlantic wreckfish. North Atlantic circulation provides a mechanism for a long-lived pelagic stage to be dispersed from Blake Plateau spawning grounds to the eastern North Atlantic. Global circulation patterns may explain both the dispersal of mtDNA haplotypes and the disjunct distribution of wreckfish body lengths in a temperate, deep-water vagile species with an extended pelagic juvenile stage such as wreckfish.  相似文献

Hares (Lepus capensis Linnaeus 1758) were probably introduced into Sardinia in historical times. Previous studies indicated North Africa as the most likely source area but did not exclude the occurrence of hybridization events with continental brown hares (L. europaeus Pallas 1778) perhaps introduced for hunting purposes. We implemented both morphometric and genetic approaches to verify the genetic isolation of the Sardinian population. Specifically, we conducted a multivariate analysis of craniometric data and analysed 461 bp of the mitochondrial control region and 12 autosomal microsatellites in Sardinian hares, using North African cape hares and European brown hares as reference populations. Sardinian hares displayed a peculiar skull shape. In agreement, both nuclear and mitochondrial markers remarked the distinctiveness of this population. Observed and expected heterozygosity were 0.52 and 0.61, while haplotype and nucleotide diversity were 0.822 and 0.0129. Self‐assignment based on Bayesian cluster analysis was high (average membership 0.98), and no evident signs of introgression from continental brown hares were found. Our results support the hypothesis that the Sardinian hares have been introduced from North Africa, remained genetically isolated since the founding event and evolved independently from the source population. This long‐lasting isolation and the consequent genetic drift resulted in a differentiation, perhaps accompanied by an adaptation to local environmental conditions.  相似文献

In avian brood parasitism, egg phenotype plays a key role for both host and parasite reproduction. Several parrotbill species of the genus Paradoxornis are parasitized by the common cuckoo Cuculus canorus, and clear polymorphism in egg phenotype is observed. In this article, we develop a population genetics model in order to identify the key parameters that control the maintenance of egg polymorphism. The model analyses show that egg polymorphism can be maintained either statically as an equilibrium or dynamically with frequency oscillations depending on the sensitivity of the host against unlike eggs and how the parasite targets host nests with specific egg phenotypes. On the basis of the model, we discuss egg polymorphism observed in parrotbills and other host species parasitized by the cuckoo. We suggest the possibility that frequencies of egg phenotypes oscillate and we appeal for monitoring of cuckoo-host interactions over a large spatiotemporal scale.  相似文献

Endangered species with small and isolated populations has been a key topic of conservation biology studies in the last decade. Lamyropsis microcephala is among the most significant narrow endemic plants in the Mediterranean region, lying on the Gennargentu massif of the Sardinia island (Italy). Due to heavy threat factors, this species has rapidly become threatened with extinction. The inter-simple sequence repeat technique was used to assess the genetic variation and structure of the individuals growing in the four remnant localities known to date, with the aim to implement further conservation strategies. Results indicated a degree of differentiation among the four subpopulations, in particular for the Fonni one. The estimates of Nei's genetic diversity (H) ranged from 0.0563 (Fonni) and 0.1104 (Bau ‘e Laccos). Analysis of molecular variance values showed that 53% of the total variation may be attributed to the individuals within subpopulations, while 47% is due to differences among subpopulations (P < 0.001). Results also highlighted a scarce gene flow (Nm = 0.503).  相似文献

Hermaphroditism has evolved several times independently in nematodes. The model organism Caenorhabditis elegans and Pristionchus pacificus are self-fertile hermaphrodites with rare facultative males. Both species are members of different families: C. elegans belongs to the Rhabditidae and P. pacificus to the Diplogastridae. Also, both species differ in their ecology: C. elegans is a soil-dwelling nematode that is often found in compost heaps. In contrast, field studies in Europe and North America indicate that Pristionchus nematodes are closely associated with scarab beetles. In C. elegans, several recent studies have found low genetic diversity and rare out-crossing events. Little is known about diversity levels and population structure in free-living hermaphroditic nematodes outside the genus Caenorhabditis. Taking a comparative approach, we analyse patterns of molecular diversity and linkage disequilibrium in 18 strains of P. pacificus from eight countries and four continents. Mitochondrial sequence data of P. pacificus isolates reveal a substantially higher genetic diversity on a global scale when compared to C. elegans. A mitochondrial-derived hermaphrodite phylogeny shows little geographic structuring, indicating several worldwide dispersal events. Amplified fragment length polymorphism and single strand conformation polymorphism analyses demonstrate a high degree of genome-wide linkage disequilibrium, which also extends to the mitochondrial genome. Together, these findings indicate distinct patterns of genetic variation of the two species. The low level of genetic diversity observed in C. elegans might reflect a recent human-associated dispersal, whereas the P. pacificus diversity might reflect a long-lasting and ongoing insect association. Thus, despite similar lifestyle characteristics in the laboratory, the reproductive mode of hermaphroditism with rare facultative males can result in distinct genetic variability patterns in different ecological settings.  相似文献

The population genetic structure of the Anopheles gambiae in western Kenya was studied using length variation at five microsatellite loci and sequence variation in a 648-nt mtDNA fragment. Mosquitoes were collected from houses in villages spanning up to 50 km distance, The following questions were answered, (i) Are mosquitoes in a house more related genetically to each other than mosquitoes between houses? (ii) What degree of genetic differentiation occurs on these geographical scales? (iii) How consistent are the results obtained with both types of genetic markers? At the house level, no differentiation was detected by FST and RST, and the band sharing index test revealed no significant associations of alleles across loci. Likewise, indices of kinship based on mtDNA haplotypes in houses were even lower than in the pooled sample. Therefore, the hypothesis that mosquitoes in a house are more related genetically was rejected. At increasing geographical scales, microsatellite allele distributions were similar among all population samples and no subdivision of the gene pool was detected by FST or RST. Likewise, estimates of haplotype divergence of mtDNA between populations were not higher than the within population estimates, and mtDNA-based FST values were not significantly different from zero. That sequence variation in mtDNA provided matching results with microsatellite loci (while high genetic variation was observed in all loci), suggested that this pattern represents the whole genome. The minimum area associated with a deme of A. gambiae in western Kenya is therefore larger than 50 km in diameter.  相似文献

鱼类是脊椎动物中最原始而在种属数量上又最占优势的类群,其起源复杂,分布广泛,拥有丰富的遗传多样性。鱼类线粒体DNA(mitochondrial DNA,mtDNA)同其他脊椎动物的mtRNA一样,呈共价闭合环状,是细胞核外具自主复制、转录和翻译能力的遗传因子。与核DNA相比,鱼类mtDNA具有分子较小、结构简单、进化速度快、遗传相对独立性和母系遗传等特点,是一个相对独立的复制单位。由于鱼类线粒体DNA具有上述特点,以mtDNA作为分子标记,探讨鱼类的群体遗传结构与系统演化,已成为鱼类分子群体遗传学和系统学研究中的热点。综述了鱼类mtDNA的结构特征、进化和多态性检测方法及其在鱼类分子群体遗传学和鱼类系统学研究中的应用。  相似文献

Schreiber’s bent-winged bat Miniopterus schreibersii and the greater horseshoe bat Rhinolophus ferrumequinum are widespread and common cavernicolous species across southern Europe that host numerous specialized ectoparasite species. The objective of this study was to characterize the species assemblage, genetic diversity, and host specificity of bat flies (Nycteribiidae, Diptera) and wing mites (Spinturnicidae, Acari) found on these bat hosts in Serbia and Bosnia and Herzegovina. Notably, while bat flies lay puparia on the cave walls and can thus be transmitted indirectly, wing mites require direct body contact for transmission. Morphological identification and sequencing of a 710-bp fragment of cytochrome oxidase I gene of 207 bat flies yielded 4 species, 3 on M. schreibersii and 1 on R. ferrumequinum. Sequencing of a 460-bp small subunit ribosomal RNA fragment, in all 190 collected wing mites revealed 2 species, 1 per host. In no case was a parasite associated with 1 host found on the other host. Species and genetic diversity of flies were higher in M. schreibersii, likely reflecting their host’s larger colony sizes and migratory potential. Mite species of both hosts showed similarly low diversity, likely due to their faster life history and lower winter survival. Our findings highlight a remarkably high host-specificity and segregation of ectoparasite species despite direct contact among their hosts in the roost, suggesting a defined host preference in the investigated ectoparasite species. Furthermore, the differences in ectoparasite genetic diversity exemplify the interplay between host and parasite life histories in shaping parasite population genetic structure.  相似文献

Two strains of Drosophila melanogaster, rover and sitter, differing in locomotion while foraging were simultaneously exposed to females of either Leptopilina boulardi or Ganaspis xanthopoda (parasitic Hymenoptera). These two parasitoids show different modes of host-searching behavior, ovipositor searching, or vibrotaxis, respectively. L. boulardi parasitized the sitter host strain significantly more than the rover. In contrast, G. xanthopoda parasitized the rover strain more than the sitter. In one case, L. boulardi selected far more sitters than rovers in population cage experiments. We also describe the frequencies of rovers and sitters in three natural populations where the local parasitoid community may have partially contributed to the differences in rover and sitter frequencies.  相似文献

Genetic diversity is a major component of the biological diversity of an ecosystem. The survival of a population may be seriously threatened if its genetic diversity values are low. In this work, we measured the genetic diversity of the stingless bee Plebeia remota based on molecular data obtained by analyzing 15 microsatellite loci and sequencing two mitochondrial genes. Population structure and genetic diversity differed depending on the molecular marker analyzed: microsatellites showed low population structure and moderate to high genetic diversity, while mitochondrial DNA (mtDNA) showed high population structure and low diversity in three populations. Queen philopatry and male dispersal behavior are discussed as the main reasons for these findings.  相似文献
