We examined the effect of acclimation to moderate hyperthermic environment on the ACTH, TSH, T3, T4 and corticosterone level, as well as the relative weight of hypophysis, thyroid and adrenal glands in streptozotocin-diabetic rats. Increased activity of the hypothalamo-pituitary-adrenocortical (HPA) axis has been demonstrated in diabetic animals, whereas insulin treatment restores the changes. Heat acclimation reduces the level of ACTH and corticosterone in control animals and moderates the hormonal disturbances caused by diabetes. Simultaneously, our study revealed impairment in the activity of the hypothalamo-pituitary-thyroid (HPT) axis. Acclimation to 35±1 °C resulted in significantly lower T3 and T4 levels in control, diabetic and insulin-treated animals. Relative weight of the hypophysis, thyroid and adrenal glands is decreased in heat-acclimated rats. Our assumption is that there might be a cross tolerance between diabetes and heat acclimation on a hormonal level.  相似文献

Hypoxia events, or low dissolved oxygen (DO) conditions, occur frequently in North Carolina estuaries during the summer. These events may have harmful effects on important fish stocks, including spot (Leiostomus xanthurus) and Atlantic menhaden (Brevoortia tyrannus), but their consequences are not well understood. We investigated direct mortality due to hypoxia in juvenile spot and Atlantic menhaden to determine how the extent of mortality varies with the severity of hypoxia and the duration of exposure, and to explore how vulnerability to hypoxia changes across species, fish size, and temperature.Atlantic menhaden and spot were tested at two temperatures, 25 and 30 °C, and three dissolved oxygen concentrations, 0.6, 0.9, and 1.2 ppm. Survival analyses were performed on the data relating survival rate of each species to dissolved oxygen concentration, duration of exposure, fish size, and temperature. The data were analyzed using an LC50 approach for comparative purposes, and 12-h LC50 estimates ranged from 0.9 to 1.1 ppm O2. Spot and menhaden exposed to 1.2 ppm O2 showed no mortality in 24 h at 25 °C, and only 30-40% mortality at 30 °C. In contrast, both species experienced 100% mortality in 2-6 h at 0.6 ppm O2. There was an effect of size on hypoxia tolerance, with small spot being less tolerant than large spot, while the converse size effect was observed for menhaden. Spot were consistently less tolerant to hypoxia than menhaden and both species were less tolerant to hypoxia at 30 °C than at 25 °C. Preliminary experiments showed a 24-h acclimation to sublethal levels of hypoxia significantly reduced mortality upon subsequent exposure to lethal hypoxia concentrations.Our results indicate that direct mortality due to hypoxia will vary with species, size, and temperature, but will likely only be substantial when these species are exposed to oxygen concentrations less than about 1 ppm O2. Given the severity of hypoxia necessary to cause mortality and the ability of fish to behaviorally avoid hypoxia, direct mortality due to hypoxia may have limited impacts on fish population dynamics. Therefore, the greatest effects due to hypoxia may be caused by the stress imposed by sublethal hypoxic conditions alone or in concert with other stressors, or by indirect effects incurred by avoiding hypoxic areas.  相似文献

Remote measurements of body temperature (Tb) in animals require implantation of relatively large temperature-sensitive radio-transmitters or data loggers, whereas rectal temperature (Trec) measurements require handling and therefore may bias the results. We investigated whether ∼0.1 g temperature-sensitive subcutaneously implanted transponders can be reliably used to quantify thermal biology and torpor use in small mammals. We examined (i) the precision of transponder readings as a function of temperature and (ii) whether subcutaneous transponders can be used to remotely record subcutaneous temperature (Tsub). Five adult male dunnarts (Sminthopsis macroura, body mass 24 g) were implanted with subcutaneous transponders to determine Tsub as a function of time and ambient temperature (Ta), and in comparison to thermocouple readings of Trec. Transponder temperature was highly correlated with water bath temperature (r2=0.96–0.99) over a range of approximately 10.0–40.0 °C. Transponders provided reliable data (±0.6 °C) over the Tsub of 21.4–36.9 °C and could be read from a distance of up to 5 cm. Below 21.4 °C, accuracy was reduced to ±2.8 °C, but individual transponder accuracy varied. Consequently, small subcutaneous transponders are useful to remotely quantify thermal physiology and torpor patterns without having to disturb the animal and disrupt torpor. Even at Tsub<21.4 °C where the accuracy of the temperature readings was reduced, transponders do provide reliable data on whether and when torpor is used.  相似文献

The influence of silicon-treatment on the levels of TSH and thyroid hormones was studied in rats. Concentrations of thyrotropin (TSH), triiodothyronine (T3), and thyroxine (T4) were estimated in sera of rats receiving per os a soluble silicon compound—sodium metasilicate nonahydrate (Na2SiO3·9H2O), dissolved in the animals' drinking water. An increase in the TSH level in the tested group was observed, without statistically significant differences in T3 and T4 concentrations between the two groups of animals. The results provide evidence for the influence of silicon on the endocrine balance. They could also prove that this chemical element is capable of modifying the rate of some hormones' synthesis.  相似文献

Alterations in circulatory levels of total T4 (TT4), total T3 (TT3), free T4 (FT4), free T3 (FT3), thyrotropin (TSH) and T3 uptake (T3U) were studied in male and female sea-level residents (SLR) at sea level, in Armed forces personnel staying at high altitude (3750 m) for prolonged duration (acclimatized lowlanders, ALL) and in high-altitude natives (HAN). Identical studies were also performed on male ALL who trekked to an extreme altitude of 5080 m and stayed at an altitude of more than 6300 m for about 6 months. The total as well as free thyroid hormones were found to be significantly higher in ALL and HAN as compared to SLR values. Both male as well as female HAN had higher levels of thyroid hormones. The rise in hormone levels in different ALL ethnic groups drawn from amongst the southern and northern parts of the country was more or less identical. In both HAN and ALL a decline in FT3 and FT4 occurred when these subjects trekked at subzero temperatures to extreme altitude of 5080 m but the levels were found to be higher in ALL who stayed at 6300 m for a prolonged duration. Plasma TSH did not show any appreciable change at lower altitudes but was found to be decreased at extreme altitude. The increase in thyroid hormones at high altitude was not due to an increase in hormone binding proteins, since T3U was found to be higher at high altitudes. A decline in TSH and hormone binding proteins and an increase in the free moiety of the hormones is indicative of a subtle degree of tissue hyperthyroidism which may be playing an important role in combating the extreme cold and hypoxic environment of high altitudes.  相似文献

Gamete production after exposure to hypoxia or sulphide was studied in the marine macroalga Ulva sp. collected in the Sacca di Goro, Italy. Experiments were carried out on discs (12 mm diameter) of thalli cultured in artificial sea water in laboratory at 20 ± 1 °C, 152 μmol m−2 s−1, 16 h photoperiod and 30‰ salinity. Dehydration of thallus was used as inducer of gametogenesis and growth and gamete release during recovery after 10, 20, 30 or 40 min dehydration (20 ± 1 °C, 25% humidity) were analysed. Unlike non-dehydrated thalli the dehydrated ones produced gametes. Thallus discs, non-dehydrated or subjected to 30 min dehydration, were exposed to hypoxia (1.78–4.02 μmol O2 L−1) or sulphide (1 mM) for 3, 5, or 7 days at 20 °C in the dark. Non-dehydrated and dehydrated thalli maintained in normoxic conditions in the dark were the controls. Gamete density was checked by counting at the end of the incubation period and during the subsequent 7 days of recovery under 16 h photoperiod in normoxic conditions. Non-dehydrated thalli maintained in normoxic conditions in the dark released gametes when returned to light suggesting that dark constitutes a stimulus to gamete production. The presence of gametes at the end of 3 days incubation of dehydrated thalli in normoxia demonstrated that gametogenesis can occur even in the dark. However, gametes were not present at the end of incubation in hypoxic and sulphidic conditions. Actually, during hypoxic incubation oxygen consumption in D-thalli was very low, only 0.117 × 10−3 μmol O2 mg−1 h−1 compared to 5.93 × 10−3 μmol O2 mg−1 h−1 in normoxia, denoting a reduction of the metabolic rate that could not sustain gametogenesis. During recovery after incubation in normoxic, hypoxic or sulphidic conditions densities of gametes from dehydrated thalli showed significant differences and resulted after hypoxia > after normoxia > after sulphide. Differences in non-dehydrated thalli were not significant. Dehydrated thalli, still green at the end of the incubation period, underwent blanching in the course of recovery in parallel to gamete production, while non-dehydrated thalli maintained their green colour even after exposure to sulphide. Our findings suggest that macroalga Ulva sp. can survive exposure to darkness, severe hypoxia and high sulphide levels and can maintain gamete production even when the exposure to these stress conditions is joined to dehydration.  相似文献

Male New Zealand White rabbits were divided into three groups: (I) control, (II) high-fat-diet (HFD) fed, and (III) HFD fed+selenium supplemented. After 3 mo of treatment, there was a significant increase in serum cholesterol and triglycerides in the HFD-fed group as compared to the control. However in the selenium (Se)-supplemented group, the levels of serum cholesterol and triglycerides were significantly less as compared to group II. HFD feeding resulted in decreased serum Se levels, but supplementation of dietary Se along with HFD, as in group III, showed an apparent increase in its levels. The Se-dependent glutathione peroxidase (GSH-Px) activity in the liver and the aorta increased significantly in HFD-fed animals and also showed an additional significant increase on Se supplementation. Both serum T3 and T4 levels showed a significant decrease on HFD feeding. However, supplementation of Se led to a significant increase in the levels of these parameters viz-à-viz HFD-fed animals. HFD feeding significantly decreased the activity of type I iodothyronine 5′-deiodinase (5′-DI) in the liver from group II rats. On supplementation of Se along with HFD, the activity increased in the liver. However, there was no significant change in its activity in the aorta. The 5′-DI activity in the thyroid showed an opposite trend in comparison with peripheral tissues (i.e., liver). The important finding of this study is that in the hyperlipidemic state, deiodinase in the thyroid behaves in a different manner as compared to its activity in extrathyroidal tissues.  相似文献

The effects of equilibration under different air relative humidities (RH, 1 – 90 %) and temperatures (35 and 45 °C) on soybean (Glycine max) and wheat (Triticum aestivum) seeds were studied using different techniques. Seed moisture content, electrical conductivity (EC) of seed leachate and per cent seed germination were measured following standard procedures, and compared with nuclear magnetic resonance spin-spin relaxation time (T2) measurements. Moisture contents of soybean and wheat seeds, following the reverse sigmoidal trend, were greater at 35 than at 45 °C at any particular RH. Changes in T2 were related to the changes in germination percentage and leachate EC of both soybean and wheat seeds. Equilibrating soybean seeds at RH 11 % decreased germination percentage with corresponding decrease in T2. On the contrary, EC of seed leachate increased. In wheat seeds equilibrated at 45 °C, T2 was maximal at RH 5.5 %. T2 declined in seeds equilibrated at high RH (> 80 %) together with low germination percentage.  相似文献

Synopsis Gill ventilation, breathing frequency, breath volume, oxygen extraction from the ventilatory water current and oxygen uptake through the gills were measured in flounder, Platichthys flesus, and plaice, Pleuronectes platessa, at water O2 tensions ranging from 35 to 155 mm Hg at 10° C. Ventilation volumes were similar in the two species at high water O2 tension. Exposure to hypoxic water elicited a larger increase in ventilation in the flounder. The per cent extraction of O2 from water decreased slightly in both species as water O2 tension was lowered. At comparable levels of ventilation O2 extraction was higher in flounder. At the higher levels of water O2 tension, O2 uptake across the gills of flounder was stable, the critical O2 tension being between 60 and 100 mm Hg. The plaice behaved as an oxygen conformer over the entire range of O2 tensions investigated. The superior ability of the flounder in maintaining OZ uptake across the gills during a reduction in water O2 tension may in part explain why the species, unlike plaice, inhabits very shallow waters with large fluctuations in dissolved oxygen.  相似文献

Effect of thyroidectomy (Tx) and subsequent treatment with 3,5,3′-triiodo-l-thyronine (T3) or replacement therapy (TR) with T3 + l-thyroxine (T4) on the temperature kinetics properties of FoF1 adenosine triphosphatase (ATPase, ATP synthase, H+-translocating ATP synthase EC 3.6.3.14) and succinate oxidase (SO) and on the lipid/phospholipid makeup of rat kidney mitochondria were examined. Tx lowered ATPase activity, which T3 treatment restored. SO activity was unchanged in Tx but decreased further by T3 treatment. TR restored both activities. The energies of ATPase activation in the high and low temperature ranges (E H and E L) increased in the Tx and T3 animals with decrease in phase transition temperature (Tt). TR restored E H and E L but not Tt to euthyroid levels. E H and E L of SO decreased in Tx animals. T3 and TR restored E H whereas E L was restored only in the TR group; Tt increased in both groups. Total phospholipid and cholesterol contents decreased significantly in Tx and T3-treated animals. In Tx animals, sphingomyelin (SPM) and phosphatidylcholine (PC) components decreased, while phosphatidylserine (PS) and diphosphatidylglycerol components increased. T3 and TR treatments caused decreases in SPM, phosphatidylinositol and PS. PC and phosphatidylethanolamine (PE) increased in the T3 group. TR resulted in increased lysophospolipids and PE. Changes in kinetic parameters of the two enzymes were differently correlated with specific phospholipid components. Both T3 and TR regimens were unable to restore normal membrane structure-function relationships.  相似文献

T4, T3 and TSH serum levels were measured in 25 patients with paracoccidioidomycosis. Thyroid T3 reserves were measured on the basis of the increase in T3 (T3) 2 h after intravenous injection of 200 g TRH, and pituitary TSH reserves were measured on the basis of TSH increase (TSH) 20 min after the same injection. Twenty healthy volunteers with no history of thyroid disease were used as controls. When the two groups were compared, the following results were obtained: (a) there was no significant difference in mean T4, T3, TSH between groups; (b) reduced T3 levels were detected more frequently in patients with paracoccidioidomycosis, especially among those with the acute form of the disease or with the severely disseminated chronic form. The results suggest the occurrence of a reduction in peripheral conversion of T4 to T3, but do not indicate the occurrence of hypothyroidism in any of its forms (thyroid, pituitary or hypothalamic).  相似文献

Transferrin (Tf) is a kind of non-heme β-globulin with two iron ions (Fe3+)-binding sites. To prove Tf’s physiological functions, Fe3+-proteins, serum iron contents, and total iron-binding capabilities were tested for Tfs of crucian carps (Carassius auratus) and sliver carps (Hypophthalmichthys molitrix). The above results demonstrated that sliver carps shared 1/3 Tf alleles with crucian carps; Tf of crucian carps had stronger Fe3+-binding ability and transportation ability in plasma than that of sliver carps. In addition, the results of oxygen consumption experiments indicated that crucian carps had the higher oxygen utility rate than sliver carps. For acute hypoxia exposure assay, normoxic gas mixture, hypoxic gas mixture A, and hypoxic gas mixture B were used to induce oxygen-regulated gene expression of crucian carps in acute hypoxia. The results of quantitative real-time PCR revealed that mRNA levels of Tf gene, Tfr gene and ATPase gene were down-regulated in acute hypoxia but mRNA level of LDHa gene was up-regulated in acute hypoxia. The results of crucian carp Tf-cDNA sequence analysis showed that cDNA regions of two Fe3+-binding sites were T747–T1026 and T1737–A1884 based on the principle of bioinformatics. The sequence conservation of two Fe3+-binding sites was higher than that of the other five regions, which were confirmed according to the subregion model of Tf-cDNA sequence.  相似文献

Previous investigations have demonstrated that photosystem II (PSII) thermostability acclimates to prior exposure to heat and drought, but contrasting results have been reported for cotton (Gossypium hirsutum). We hypothesized that PSII thermotolerance in G. hirsutum would acclimate to environmental conditions during the growing season and that there would be differences in PSII thermotolerance between commercially-available U.S. cultivars. To this end, three cotton cultivars were grown under dryland conditions in Tifton Georgia, and two under irrigated conditions in Marianna Arkansas. At Tifton, measurements included PSII thermotolerance (T15, the temperature causing a 15% decline in maximum quantum yield), leaf temperatures, air temperatures, midday (1200 to 1400 h) leaf water potentials (ΨMD), leaf-air vapor pressure deficit (VPD), actual quantum yield (ΦPSII) and electron transport rate through PSII (ETR) on three sample dates. At Marianna, T15 was measured on two sample dates. Optimal air and leaf temperatures were observed on all sample dates in Tifton, but PSII thermotolerance increased with water deficit conditions (ΨMD = −3.1 MPa), and ETR was either unaffected or increased under water-stress. Additionally, T15 for PHY 499 was ∼5 °C higher than for the other cultivars examined (DP 0912 and DP 1050). The Marianna site experienced more extreme high temperature conditions (20–30 days Tmax ≥ 35 °C), and showed an increase in T15 with higher average Tmax. When average T15 values for each location and sample date were plotted versus average daily Tmax, strong, positive relationships (r2 from .954 to .714) were observed between Tmax and T15. For all locations T15 was substantially higher than actual field temperature conditions. We conclude that PSII thermostability in G. hirsutum acclimates to pre-existing environmental conditions; PSII is extremely tolerant to high temperature and water-deficit stress; and differences in PSII thermotolerance exist between commercially-available cultivars.  相似文献

The aim of present study was to investigate the effects of 3,3′,5-triiodothyronine (T3) on rat testis both morphometrically and immunohistochemically with determining of insulin-like growth factor I (IGF-I) expression. Adult male Wistar-albino rats used in the study were divided into two groups; control and T3-treated groups. After T3 treatment there was observed to be a decrease in testicular weights, diameters of seminiferous tubules and the number of sertoli cells, and an increase in the number of leydig cells (P<0.05). Some of the seminiferous tubule lumens of T3 administrated rats had cellular debris. IGF-I was localized in sertoli cells, late spermatids and leydig cells of all groups. IGF-I immunoreactivity in T3 treated rats was higher than in controls in all stages of the cycle of rat seminiferous epithelium, but the staining intensity of leydig cells were similar in both groups. In conclusion, the present results suggest that T3 may modulate the testicular function by affecting IGF-I activity at the gonadal level.  相似文献

The effects of varying leaf temperature (T 1) on some ecophysiological characteristics of photosynthesis for Quercus liaotungensis Koiz. under ambient radiation stress around midday on clear summer days were investigated using an IRGA equipped with a temperature-controlled cuvette. Net photosynthetic rate (P N) decreased as T 1 increased from 30 to 35 °C as a result of stomatal closure, whereas non-stomatal limitation led to decreased P N in the T 1 range of 35–45 °C. Decreased transpiration rate (E) and stomatal conductance (g s) at leaf temperatures above 30 °C were interpreted as a combined feedward effect as a result of enhanced leaf-air vapour pressure deficit (VPD) and stomatal closure. Changes in E from T 1 30 to 20 °C depended on VPD when g s was maintained constant. Water use efficiency (WUE) varied inversely with T 1 by following a hyperbola. A decrease in intercellular CO2 concentration (C i) occurred as a result of stomatal closure and a relatively high carboxylation capacity, whereas inactivation of mesophyll carboxylation in combination with photorespiration might be associated with the observed increase in C i in the T 1 range of 40 to 45 °C.  相似文献

Synopsis Oxygen uptake (Vo 2) was measured in carp of approximately 40 cm length swimming at controlled variable oxygen tensions (Po 2). At Po 2> 120 mm Hg Vo 2 increased with an increase in swimming speed from 5.6 to 11.3 cm · sec–1. Extrapolation of Vo 2 to zero activity at Po 2 = 140 mm Hg revealed a standard O2 uptake of 36.7 ml O2 · kg–1 · h–1 at 20° C. At the lowest swimming speed (5.6 cm · s–1) the oxygen uptake increased when the water Po 2 was reduced. A near doubling in Vo 2 was seen at Po 2 = 70 mm Hg compared to 140 mm Hg. At higher swimming speeds in hypoxic water Vo 2 decreased relative to the values at low swimming speeds. As a result the slope of the lines expressing log Vo 2 as a function of swimming speed decreased from positive to negative values with decreasing Po 2 of the water. pH of blood from the caudal vein drawn before and at termination of swimming at Po 2 = 70 mm Hg and 100 mm Hg did not show any decrease in relation to rest values at Po 2 = 140 mm Hg. Blood lactate concentration did not increase during swimming at these tensions.  相似文献

Studies were conducted on male adult rabbits to find out the changes in blood glucocorticoid levels along with the changes in aspartate aminotransferase activity in blood and the role of pyridoxine on the glucose tolerance pattern under hypoxic stress. Hypoxic stress was produced by exposing the animals to a simulated altitude of 7,000 m for 6 h. In the first set of experiments 10 rabbits were used. Blood haemoglobin level, plasma and erythrocyte glucocorticoid levels and erythrocyte GOT activity were measured just before and after the exposure to hypoxia. Erythrocyte GOT activity was measured both without and with 50 mg of pyridoxal phosphate addition to the incubation mixture. Glucocorticoid levels in plasma increased by 11% whereas in erythrocytes the increase was 55% after hypoxia. Percent stimulation of erythrocyte GOT activity with pyridoxal phosphate before exposure to hypoxia was 180% but increased to 321% after exposure. In the second set of experiments another 10 rabbits were used. First they were exposed to hypoxia without pyridoxine hydrochloride feeding and then after 7 days with 3 mg of pyridoxine hydrochloride feeding. For glucose tolerance tests the animals were fed with 1 g of glucose immediately after the hypoxic exposures. Plasma reduced glutathione (GSH), LDH and ICDH activities increased and GOT activity was depressed after hypoxic stress, but when the animals were fed pyridoxine hydrochloride prior to the exposure the enzyme activities remained unaltered after hypoxic stress. Pyridoxine hydrochloride did not alter the pattern of glucose tolerance after hypoxic stress.  相似文献

The respiratory responses and tolerance of hypoxia were studied in two marine teleosts, the red grouper (Epinephelus akaara, a sluggish species) and the black sea bream (Mylio macrocephalus, an active species). Neither species showed abnormal behaviour or mortality when exposed to 2 mg O2 l–1 for 7 h. The black sea bream was, however, comparatively more tolerant when exposed to 1 mg O2 l–1, but tolerance of both species became similar under extremely hypoxic conditions (i.e. 0.5 mg O2 l–1). In contrast to most other teleosts, both species showed a reduction in opercular beating rate during hypoxia, and oxygen conformity was found in the range of 0.5 to 7.0 mg O2l –1. O2 dissociation curves were constructed, and the P50 value of the black sea breams (27 ± 5.6 mm Hg) was found to be much lower than that of the red groupers (50 ± 2.5 mm Hg). For both species, the general levels of venous PO2 showed a direct relationship to ambient PO2, and were markedly reduced after 1 h exposure to various levels of hypoxia. Compared with the red groupers, the black sea breams appeared to be more able to maintain its venous PO2 levels during prolonged hypoxic exposure.  相似文献

Summary The effects of ambient temperatures of 10°C and 30°C and of E. coli endotoxin on brain temperature and plasma iron level were investigated in unrestrained wild house mice, Mus musculus. In control animals (i.p. saline-injected) exposed to cold environmenta the brain temperature decreased and plasma iron levels were lower than those observed under thermoneutral conditions (30°C). Animals injected i.p. with endotoxin (0.5 g·kg-1) and placed at 30°C showed a drop in plasma iron level during the fever episode. The results provide strong evidence for a relationship between brain temperature and plasma iron level in control mice under thermoneutral conditions, and show that during cold exposure or after injection of endotoxin, there is no linear correlation between brain temperature and plasma iron.
