  • 1.1. Polyethyleneglycol-modified bovine adenosine deaminase was administered (10–20 U/kg/week) intramuscularly to two opossums for 15 weeks and changes in red cell adenine ribo- and deoxyribonucleotides quantitated by HPLC.
  • 2.2. Only a moderate decline of erythrocyte dAXP was observed at the end of the study when compared to results of enzyme replacement seen in human adenosine deaminase deficient patients.

  • 1.1. The toxic effects of adenosine and deoxyadenosine on lymphocytes from horses were evaluated.
  • 2.2. Mitogen-stimulated peripheral blood lymphocytes (PBL) were found to be more sensitive to the inhibitory effects of adenosine than were lymphocytes from spleen, lymph node and thymus.
  • 3.3. Adenosine deaminase activity was approximately 10 times lower in horse lymphoid tissue in comparison to that found in human lymphoid tissue. In horse, the highest activity was in spleen while the lowest activity was in thymus.
  • 4.4. Adenosine inhibition of mitogenesis in PBL was prevented by uridine or cytidine, suggesting pyrimidine starvation as the major mechanism for adenosine toxicity.
  • 5.5. Deoxyadenosine inhibition of mitogenesis in PBL was not prevented by the addition of various ribo- or deoxyribonucleosides. This and the finding that treated cells show no increase in deoxyATP suggest that some other mechanism for deoxyadenosine toxicity other than deoxyATP inhibition of ribonucleotide reductase operates in horse PBL.

  • 1.1. The metabolism of purine bases and nucleosides in cotyledons and embryonic axes of black gram (Phaseolus mungo L.) was studied.
  • 2.2. A large portion of absorbed [8-14C]adenine, [8-14C]guanine and [8-14C]adenosine was salvaged in nucleotide and nucleic acid synthesis.
  • 3.3. Most of the radioactivity of [8-14C]hypoxanthine and [8-14C]inosine was incorporated into allantoin and allantoic acid.

  • 1.1. Exogenous and endogenous tyrosine protein phosphorylation activities were examined in soluble and partieulate fractions from various normal tissues by using poly-[Glu-80Na, Tyr20] and a monoclonal antibody specific for phosphotyrosine.
  • 2.2. Phosphorylation of the exogenous substrate by the partieulate forms of TPKs was 2- to 10-fold higher than by soluble forms. The activities of partieulate and soluble enzymes decreased in the following order: spleen > (thymus = kidney) > testes ⩾ (pancreas = liver = brain) > heart.
  • 3.3. The level of endogenous phosphorylation in the tissues decreased respectively in the following order: thymus > brain ⩾ (pancreas = liver) > spleen > testes > kidney > heart for the partieulate fractions, and spleen > thymus > brain > pancreas ⩾ liver > testes > kidney > heart for the soluble fractions.
  • 4.4. A large number of phosphotyrosine-containing proteins were detected. In addition, several phosphotyrosine-containing proteins of similar molecular weight were found in different tissues and fractions.

  • 1.1. Heparin stimulates the activity of nonactivated and activated skeletal muscle phosphorylase kinase in a Ca2+-dependent manner.
  • 2.2. The stimulatory effect of heparin on the activity of nonactivated phosphorylase kinase is also expressed in the presence of calmodulin and glycogen. Heparin acted in synergism with glycogen.
  • 3.3. Heparin increases the affinity of phosphorylase kinase to Ca2+ 5–12 fold depending upon the activation conditions.
  • 4.4. Ca2+ influences the stimulation of liver phosphorylase kinase by heparin in a similar way.

The capacity of avian erythrocytes for utilization of guanine and hypoxanthine is several fold less than that of adenine.
  • 3.4. The data are consistent with lower activity for hypoxanthine/guanine phosphoribosyltransferase than for adenine phosphoribosyltransferase in intact chicken erythrocytes.
  • 4.5. The results indicate that reutilization of adenine by chicken erythrocytes may be physiologically significant.

  • 1.1. Acid and alkaline phosphatase activities of eight different snake venoms were determined quantitatively by using synthetic substrates, o-carboxyphenylphosphate and p-nitrophenylphosphate respectively.
  • 2.2. It was found that most of Elapidae venoms investigated had both acid and alkaline phosphatase activities.
  • 3.3. Three Crotalidae venoms investigated did not show any alkaline phosphatase activity.

  • 1.1. The effect of adenosine separately or in combination with alpha-1 adrenergic antagonist prazosin and alpha-2 adrenergic antagonist yohimbine as well as adenosine antagonists 8-phenyltheophylline and xanthine amine conjugate on glucose-induced insulin secretion from isolated rat pancreatic islets was studied.
  • 2.2. Their in vivo effects on serum glucose and insulin levels were also investigated. Adenosine at 10 and 100 μM inhibited significantly, insulin secretion from the isolated islets whereas at 10 mM slightly increased the secretion of insulin.
  • 3.3. Prazosin used at 100 μM inhibited insulin secretion. When it combined with adenosine (10 μM) it augmented the inhibitory effect of adenosine.
  • 4.4. In vivo prazosin (21 mg/kg bodywt) caused a hyperglycaemia which was accompanied by hypoinsulinaemia.
  • 5.5. Concurrent administration of this drug with adenosine neither affect the hyperglycaemic nor the hypoinsulinaemic effects of adenosine.
  • 6.6. On the other hand, yohimbine (100 μM) has no effect neither separately nor in combination with adenosine (10 μM) in modulating the inhibitory effect of adenosine on insulin secretion.
  • 7.7. When Yohimbine administered at 19.5 mg/kg body wt it did not alter serum glucose but it markedly increased the serum insulin level. Its combined administration with adenosine reduced the hyperglycaemic effect of adenosine with a remarkable increase in serum insulin.
  • 8.8. Both adenosine-antagonists were ineffective in alteration of insulin secretion.
  • 9.9. However, combination of 8-phenyltheophylline with adenosine (10 μM) totally blocked the inhibitory effect of adenosine on insulin secretion while xanthine amine conjugate failed to prevent this effect of adenosine.
  • 10.10. These results indicate that the inhibitory effect of adenosine on insulin secretion is neither mediated via alpha-1 nor alpha-2 adrenoceptors. It might be via activation of specific adenosine receptors on rat islets which are sensitive to blockade by 8-phenyltheophylline.

  • 1.1. Glucose formation from lactate by the perfused liver of 48 hr starved chickens was strongly inhibited by adenosine (Ado); the half-maximal inhibition was attained at 40 μM. This effect was paralleled by a four- to five-fold increase of ATP content as determined in freeze-clamped liver.
  • 2.2. In chicken liver homogenate gluconeogenesis from precursors such as alanine, glutamate, glutamine and aspartate, which are not converted into glucose by the perfused chicken liver, proceeded at rates equal to or higher than that with lactate, being markedly inhibited by Ado.
  • 3.3. In the perfused guinea-pig liver glucose synthesis with lactate, propionate, glycerol and fructose was also inhibited by Ado; however, when precursors such as pyruvate, glutamine and a mixture of lactate + pyruvate were supplied to the liver Ado did not inhibit gluconeogenesis.
  • 4.4. Assay of adenine nucleotides in the perfused guinea-pig liver, stopped by freeze-clamping technique in a number of experimental variants, revealed no correlation between the rate of gluconeogenesis and the changes induced by Ado in the adenine nucleotide pool.
  • 5.5. In the perfused liver of both chicken and guinea-pig Ado produced an increase of the lactate to pyruvate ratio and, in general, a diminution of the content of malate-aspartate shuttle intermediates.
  • 6.6. The results are interpreted as suggesting that the inhibitory effect of Ado on hepatic gluconeogenesis is not necessarily mediated by the changes in the adenine nucleotide pool.

In rat heart perfused with adenosine (10−6M), dilazep (10−4M) inhibited incorporation of adenosine into nucleotides (an index of nucleoside transport and phosphorylation) to a greater extent (70%) than metabolism to inosine and uric acid (40%) and actually increased the recovery of inosine to 30% of the adenosine infused.
  • 2.2. Extrapolating for complete inhibition of transport suggested that 60% of adenosine metabolism was intracellular and 40% extracellular.
  • 3.3. Static incubations of atria also gave an estimate for extracellular metabolism of 40%.
  • 4.4. Adenosine deaminase was localised by immunocytochemistry to the extracellular surface of endothelial cells of small coronary arteries.
  • 5.5. Extracellular deamination may explain the lack of effect of nucleoside transport inhibitors on responses to adenosine in rat heart.

  • 1.1. An affinity chromatography technique for the determination of turnover parameters has been applied to the study of carbonic anhydrase in a variety of mouse tissues.
  • 2.2. The methodology has allowed the establishment of the relative turnover characteristics in these tissues, and the evaluation of the half lives and degradation rate constants.
  • 3.3. Considerable variation was evident between the individual tissues, with synthesis being indicated as most rapid in intestine while degradation was highest in liver.

  • 1.1. Five major forms of nuclear DNA-dependent RNA polymerases from seven transplantable murine tumors and from five tissues from normal, adult, male Swiss mice (Mus musculus) were separated chromatographically on DEAE-Sephadex A-25. Forms Ia and Ib were insensitive to α-amanitin, whereas forms IIa and IIb were highly sensitive and form III was slightly sensitive.
  • 2.2. The polymerases from all tumors or ascites tumors only were compared statistically with those from the normal tissues in regard to elution patterns, specific activities, activities per mg of nuclear DNA. degrees of purification and yields. Similarly, individual tumors were compared with homologous normal tissues. All parameters were characterized by relatively large variance.
  • 3.3. Activities of all RNA polymerases per mg DNA were higher in all tumors compared with normal tissues. The order of statistical significance of these differences was lib > III > IIa > Ib > Ia.
  • 4.4. Activities per mg DNA of all RNA polymerases from 6C3HED and L1210 tumors, with the exception of L1210 tumor enzyme III, were significantly greater than those from spleen, but only the activities of Lewis lung tumor enzyme IIb and of Taper hepatoma enzymes Ia and III were higher than those from the homologous tissues, lung and liver, respectively.

  • 1.1. Adenine nucleotide concentrations and metabolism in red blood cells (RBC)2 and RBC ghosts from psoriatic patients and healthy subjects were compared.
  • 2.2. The ATP and total adenine nucleotide levels and the adenylate energy charge (EC) were elevated in the blood from psoriatic patients.
  • 3.3. The rate of glycolytic production of ATP by intact RBC was unchanged, but the Na+, K+-ATPase activity of RBC ghosts was decreased significantly in psoriasis.
  • 4.4. Results suggest that the defect in adenine nucleotide metabolism is a systemic manifestation of psoriasis, and that the quantification of adenine nucleotides in RBC and in whole blood samples may be of pathophysiological value in psoriatic lesion.

  • 1.1. An artificial diet, consisting of a dry aggregate of 59 chemical substances, was used to assess the requirements of the sea slater Ligia pallasii for vitamins, carbohydrates, fatty acids, cholesterol and minerals.
  • 2.2. Good growth and survival of L. pallasii was obtained on the diet, comparable to that on seaweeds and to that shown by a field population.
  • 3.3. No dietary requirements for vitamins, fatty acids or cholesterol were shown for periods of 40 weeks or more for L. pallasii.
  • 4.4. Carbohydrates were shown to be required by L. pallasii in its diet, in the order: starch, lactose > maltose, glucose > sucrose, cellulose.
  • 5.5. Dietary requirements for minerals were, in order: calcium, magnesium, phosphorus > copper, nickel, zinc > iron, manganese, sulphur > iodine, silicon.
  • 6.6. The results are discussed in relation to the role of gut bacteria in supplying required nutrients to their isopod hosts and the enhancement of this process through coprophagic behaviour.

  • 1.1. Amino acid sequences of bovine and porcine band 3, an erythrocyte anion transporter, were determined.
  • 2.2. The sequence of bovine band 3 was positioned to residues 519–599 (the numbering is based on human band 3), in which probably 6 residues were unidentified.
  • 3.3. Binding site of DIDS (4,4'-diisothiocyanostilbene-2,2'-disulfonate), a potent anion transport inhibitor, was identified as Lys-539 in the bovine case.

  • 1.1. Substantial differences were found in the acyi donor and lyso-acceptor specificities among subcellular membranes and with respect to different regions of the adrenal gland.

In the presence of Mg2+-ATP and CoASH, adrenal microsomes were actively transferring arachidonate to lysophospholipids with acyl acceptor specificity in the order: 1-acyl-GPI > 1-acyl-GPC > 1-acyl-GP. However, when oleoyl-CoA was used, acyi acceptor specificity for the microsomal transferases was in the order: 1-acyl-GPC > 1-acyl-GP > 1-acyl-GPI.
  • 3.3. Mitochondrial membranes had very low acyi transfer activity and they preferred 1-acyl-GPC over other lyso-acceptors.
  • 4.4. The chromamn granules were apparently lacking this type of activity.
