1. Lineweaver-Burk plots revealed the presence of high (uptake 1) and low (uptake 2) affinity reuptake components for serotonin, dopamine and norepinephrine in the cerebral and in the visceral ganglia of M. edulis.2. The Km, affinity value for norepinephrine was 5 times greater in the visceral ganglia than in the cerebral ganglia, a finding which demonstrates ganglionic variations in norepinephrine uptake.3. The rank order of monoamine affinity for the uptake 1 system is serotonin > norepinephrine > dopamine in the cerebral ganglia, whereas in the visceral it is norepinephrine > serotonin > dopamine.4. A Michaelis-Menten analysis of the uptake data revealed that at low monoamine concentrations uptake 1 contributes more than uptake 2 to the total amount of amine accumulated, while at higher concentrations the reverse is true.  相似文献

The dopamine-stimulated adenylate cyclase activity was studied both in vivo and in vitro in the central nervous system of the bivalve mollusc Mytilus edulis. Dopamine, epinine, and apormorphine stimulated the enzyme system. Fluphenazine, haloperidol, chlorpromaxine, and to a lesser extent BOL inhibited the dopamine-stimulated adenylate cyclase. Etorphine, -endorphine, DALA, and methionine enkephalin depressed cyclic AMP levels. This phenomena was naloxone reversible. In addition, the opioids inhibited the stimulation of adenylate cyclase by dopamine. This phenomena was also naloxone reversible. The study demonstrates an interaction among dopamine, the opioids, and cyclic AMP.This work was partially supported by Grant 1-T32GM07641-01 from the M.A.R.C.   • 1.1. The glycogen content of the mantle tissue reached a maximum in the summer (May–July) with levels of 41.0–53.5% of the dry tissue weight.
  • 2.2. Seasonal changes in glycogen synthetase activity showed that the I-activity (independent of G6P) increased up to 10-fold in June as compared with December. The measured I-activity of glycogen synthetase was sufficient to account for the accumulation of mantle glycogen in the summer.
  • 3.3. The I-activity of glycogen synthetase declined rapidly in July of each year. A possible role for the inhibition of glycogen synthetase by high levels of tissue glycogen is suggested.
  • 4.4. The I-activity in the mantle tissue of mussels on the shore was higher than that for animals starved in the laboratory for 2–3 days. The differences were minimal in early May but increased markedly in late May–July. Starved mussels returned to the shore showed an increase in I-activity of glycogen synthetase.
  • 5.5. Injection of 30 μmol glucose into the adductor muscle increased the concentration of glucose in the mantle fluid to 2.0–2.5 mM. A similar injection of 60 μ mol glucose resulted in a time-dependent increase in the I-activity of glycogen synthetase.
  • 6.6. Injection of mussels with mammalian insulin or anti-insulin serum had no effect on the activity of glycogen synthetase. Our results are at variance with those of other workers who have used the mammalian hormone in molluscan studies (see Discussion).

Concentrations of zinc, cadmium, lead and iron were determined in whole soft parts of mussels, Mytilus edulis (L.), and in growing tips of the alga Fucus vesiculosus (L.) from a total of 20 locations in the area of the Sound between Sweden and Denmark. Pollution profiles produced for each metal were similar in any one of the species studied but the profiles for metals in M. edulis were distinct from those for metals in F. vesiculosus. Results for mussels agreed broadly with those reported previously for this region, indicating a northward decrease in available metal concentrations. By contrast, results for algae showed few differences in metal availability throughout the study area, although some local effects were evident.The differences in the pollution profiles exhibited by these two organisms depend on their response to different portions of the total trace metal load of ambient waters. Results from the alga agree well with available data on the concentrations of trace metals in solution in waters of the Sound. The alga thus appears to be responding only to metals in solution, as suggested by previous authors. By contrast, the major proportion of the total body load of metals in mussels is derived from ingested phytoplankton. The pollution profiles exhibited by mussels may be explained if phytoplankton from the Baltic Sea contain higher concentrations of trace metals than do those from Kattegat. Possible mechanisms for the production of such differences in the metal contents of the two phytoplankton populations, and the ecological implications of these differences, are briefly discussed.  相似文献

We are presenting the first report of the occurrence of "gill disease" in Mytilus edulis trossulus from the southern Baltic Sea, Gulf of Gdańsk (Poland). The disease preliminary diagnosis was based on the presence of white and yellow "spots" causing deep indentations in the gills with degeneration, destruction and necrosis of gill filaments. Average prevalence of gill erosion in the blue mussels population was 15.8%. According to other authors, gill erosion may affect over 80% of the bivalve population resulting in mortality rates of up to 40%. The origin of the gill erosion remains unknown, but viruses are most likely involved in the etiology of these pathological conditions. The disease as such may indicate a decrease in the immunological resistance of organisms to infections and inflammations directly or indirectly caused by harmful factors in the ambient environment. Furthermore, the occurrence of numerous pathologies in bivalves is a particular problem in the Gulf of Gdańsk being a low biodiversity ecosystem. Pathologies reduce bilvalves' reproduction ability, worsen their physiological condition and increase their mortality rate. Those factors may also pose a significant ecological danger and lead to negative alterations of the ecosystem.  相似文献

Two giant neurons, d-RCDN (dorsal-right cerebral distinct neuron) and d-LCDN (dorsal left cerebral distinct neuron), with a diameter of about 100 microns, were found symmetrically on the dorsal surface of the cerebral ganglia of an African giant snail (Achatina fulica Férussac). They showed spontaneous spike discharges at a stable frequency. Two giant neurons, v-RCDN (ventral-right cerebral distinct neuron) and v-LCDN (ventral-left cerebral distinct neuron), (diameter, approx. 150 microns) were identified on the ventral surface of the same ganglia. No spontaneous spike discharges were evident. Both d-RCDN and d-LCDN were equally inhibited by dopamine, octopamine, 5-hydroxytryptamine and histamine. Acetylcholine sometimes showed inhibitory effects, but they were not so stable. No substance having excitatory effects on the neurons was found. Both v-RCDN and v-LCDN were equally excited by octopamine, 5-hydroxytryptamine, GABA and acetylcholine and inhibited by dopamine and beta-hydroxy-L-glutamic acid.  相似文献

1. Pharmacological characteristics of the two largest neurons, r-PLN (right-parietal large neuron) and d-VLN (dorsal-visceral large neuron), situated symmetrically on the anterior-dorsal surface in the suboesophageal ganglia of an African giant snail (Achatina fulica Férussac), were determined.2. Of the catecholamines examined, dopamine (DA) showed marked but complex effects on the two neurons. DA produced a transient excitation followed by either hyperpolarization or depolarization of the neuromembranes. L-Norepinephrine produced a slight depolarization of the neurons.3. 5-Hydroxytryptamine (5-HT) produced marked excitation of both neurons. Bufotenine showed the same effects, but they were weaker than those of 5-HT.4. Histamine showed inhibatory effects on the two neurons examined, whereas L-histidine had rather slight or no excitatory effects.5. Of the amino acids examined, L-homocysteic acid (L-HCA) and erythro-β-hydroxy-L-glutamic acid had marked excitatory effects on r-PLN and d-VLN. L-Homocysteine sulfinic acid also showed the excitatory effects, but they were weaker than those of L-HCA. Glycine and L-methionine had slight excitatory effects on both neurons, whereas L-aspartic acid, L-glutamic acid and GABA had none.6. Acetylcholine (ACh) exhibited marked and complex effects on r-PLN and d-VLN. In trials, ACh produced either excitation or inhibation of both neurons. Propionylcholine and butyrycholine had slightly less effect, producing a slight inhibation of r-PLN and either excitation or inhibation of d-VLN in trials.  相似文献

Metabolism of the plant sulfolipid—sulfoquinovosyldiacylglycerol (SQDG)—was studied in animal tissues. In vivo experiments with [35S]SQDG in guinea pigs showed that this lipid is not absorbed intact in the gastrointestinal tract. In these experiments, 3 h after administration of [35S]SQDG, the intestinal mucosa contained 1 to 5% of the radioactivity as SQDG, while the remainder was in a water-soluble form. Analysis of the water-soluble components showed that about 60% of the radioactivity was present as sulfoquinovosylglycerol (SQG) and the remainder was present as free SO2−4. In the blood, 99% of the radioactivity was present as SO2−4, SQG was not observed. In liver, only very little radioactivity was observed and appeared to be mainly in the form of SO2−4. Experiments with everted intestinal sacs of guinea pigs confirmed the formation of SQG, SO2−4, and, in addition, sulfoquinovosylmonoacylglycerol (SQMG) in this tissue. In vitro experiments with saline extracts of acetone powders of pancreas and intestinal mucosa of guinea pig, sheep, and rat showed that [35S]SQDG was deacylated to SQMG (sulfolipase A activity) and SQG (sulfolipase B activity). It is concluded that animal tissues deacylate SQDG in a stepwise manner to SQG. It is further metabolized to yield free SO2−4 by cleavage of the C-S bond which appears to be brought about by the intestinal microflora. Sheep pancreatic sulfolipases were characterized. Bile salts, sodium dodecyl sulfate, and Triton X-100 inhibited the pancreatic sulfolipases, while CaCl2 activated them. Substrate competition experiments and investigations on substrate specificity with a partially purified preparation indicated that relatively specific sulfolipase(s) may exist in pancreas. Among the species tested, guinea pig tissues showed the highest sulfolipase A and B activities followed sheep and rat tissues. Pancreatic enzymes were 18 to 60 times more active than intestinal enzymes.  相似文献

The sublethal toxic potency of malathion in inhibiting acetylcholinesterase activity of brain, muscle, gill and liver tissues of the fish,Tilapia mossambica was studied at 12 h intervals. Maximum in hibition at 36 and 48 h, and complete revival of acetylcholinesterase activity after 72 h was noticed, suggestive of the loss of inhibition of the enzyme activity was probably by suitable (acetylcholine) accumulation.  相似文献

Bleomycin (blm) solutions containing the nickel(II) ion have been investigated through 1H nmr, ligand field and circular dichroism spectroscopies. It has been found the blm binds the metal ion in a pH dependent fashion. The spectral data are consistent with the presence of at least two species. It is suggested that in the low pH region blm binds to nickel(II) through the β-aminoalanino residue, whereas in the high pH region, the 4-amino-pyrimidine, imidazole, and amido group of β-hydroxy-histidine are also involved in coordination.  相似文献

  • 1.1. The axonal pathways of thirteen giant neurons identified in the right parietal and the visceral ganglia, found in the suboesophageal ganglia of an African giant snail (Achatina fulica Férussac), were investigated by intracellular injections of Lucifer Yellow, with regard to their axonal projections into the following six peripheral nerves: lap n (left anterior palliai nerve), lpp n (left posterior palliai nerve), int n (intestinal nerve), anal n (anal nerve), rpp n (right posterior palliai nerve) and rap n (right anterior palliai nerve).
  • 2.2. These projections were confirmed by the recording of the axonal responses from the nerves.
  • 3.3. On the dorsal surface of the right parietal ganglion, the following four giant neurons were identified: PON (periodically oscillating neuron), TAN (tonically autoactive neuron), RAPN (right anterior palliai neuron), and d-RPLN (dorsal-right parietal large neuron).
  • 4.4. The PON axonal pathways projected into int n; those of TAN into all of the nerves examined; those of RAPN into lap n, lpp n, int n, anal n and rap n.; and those of d-RPLN into pd nn (pedal nerves) through the pedal ganglia, lpp n, anal n, rap n and sometimes lap n.
  • 5.5. On the dorsal surface of the visceral ganglion, the following four giant neurons were also identified: VIN (visceral intermittently firing neuron), FAN (frequently autoactive neuron), INN (intestinal nerve neuron) and d-VLN (dorsal-visceral large neuron).
  • 6.6. The VIN axonal pathways, which had no branch into the six nerves examined, went to both the right and the left pedal ganglia, sending a branch into the cerebro-pleural connective; those of FAN projected into lap n, anal n and rap n, and sometimes into lpp n and rpp n; those of INN into int n; and those of d-VLN into pd nn, lap n, lpp n, anal n and rap n.
  • 7.7. On the ventral surface of the right parietal ganglion, v-RPLN (ventral-right parietal large neuron) was identified. The axonal pathways went to pd nn, lap n, lpp n, anal n and rap n.
  • 8.8. On the ventral surface of the visceral ganglion, the four giant neurons, v-VNAN (ventral-visceral noisy autoactive neuron), v-VLN (ventral-visceral large neuron), r-VMN (right-visceral multiple spike neuron) and 1-VMN (left-visceral multiple spike neuron) were identified.
  • 9.9. The axonal pathway of v-VNAN projected into rpp n and rap n; those of v-VLN into pd nn, lap n, anal n, rap n and sometimes to lpp n; those of r-VMN into int n and rpp n; and those of 1-VMN also into int n and rpp n.
  • 10.10. The present morphologial investigations of the giant neurons confirmed well the identifications of the neurons previously studied. The axon of the neurons examined here, except for VIN, projected into some of the peripheral nerves, while the VIN axon extended into the cerebro-pleural connective.
  • 11.11. The five neurons, PON, TAN, v-VNAN, r-VMN and 1-VMN, formed fine axonal arborizations terminating at the neuropile, while the arborizations of the other neurons were not clearly observed.
  • 12.12. Although the anatomical structures of the portion examined of the suboesophageal ganglia are asymmetrical, three pairs of symmetrically-situated neurons, d-RPLN and d-VLN, v-RPLN and v-VLN, and r-VMN and 1-VMN, were found, indicating the existence of symmetrical components in the ganglia.

To understand vago-vagal reflexes, one must have an appreciation of the events surrounding the encoding, integration, and central transfer of peripheral sensations by vagal afferent neurons. A large body of work has shown that vagal afferent neurons have nonuniform properties and that distinct subpopulations of neurons exist within the nodose ganglia. These sensory neurons display a considerable degree of plasticity; electrophysiological, pharmacological, and neurochemical properties have all been shown to alter after peripheral tissue injury. The validity of claims of selective recordings from populations of neurons activated by peripheral stimuli may be diminished, however, by the recent demonstration that stimulation of a subpopulation of nodose neurons can enhance the activity of unstimulated neuronal neighbors. To better understand the neurophysiological processes occurring after vagal afferent stimulation, it is essential that the electrophysiological, pharmacological, and neurochemical properties of nodose neurons are correlated with their sensory function or, at the very least, with their specific innervation target.  相似文献

Low molecular weight zinc(II) and copper(II) binding ligands were detected in ultrafiltered human, bovine, and goat milk by the application of the method of modified gel chromatography. Human milk contains at least three detectable low molecular weight copper binders, whereas bovine and goat milk contain at least two. All three milks show two copper binding peaks with the same elution volumes. Zinc chromatograms were less specific than copper. Zinc showed only a single detectable low molecular weight binding ligand common to all three milks. Elution volumes for both zinc(II) and copper(II) citrate and picolinate systems were measured. Elution volumes of both copper(II) and zinc(II) citrate complexes are identical to elution volumes of an intense peak observed with all three milks; it is reasonable to assume that at least part of this peak corresponds to citrate. Human milk alone has a relatively intense binding peak for copper(II) at the same elution volume as the glutamate complex. Human and goat milk have another low intensity copper(II) binding ligand peak at the same elution volume; a number of amino acid complexes have binding peaks at this position. No peak characteristic of the zinc(II) or copper(II) picolinate systems could be found with any of the milks.  相似文献

The anatomy of the neurosecretory cells in the brain-subesophageal ganglion complex of female European corn borer moth Ostrinia nubilalis (Lepidoptera: Pyralidae) was studied using histological and cobalt backfilling techniques. Histological staining revealed the presence of 2 median and one lateral neurosecretory cell groups in the brain. These brain neurosecretory cells are made up of mainly type A cells with a few type B cells in the median group. Three type C neurosecretory cell clusters occupy the apparent mandibular, maxillary, and labial neuromeres at the ventral median aspect of the subesophageal ganglion. Axonal pathways of the neurosecretory cell groups were delineated by retrograde cobalt filling from the corpora cardiaca. Fibers of the 3 brain neurosecretory cell groups merged to form a distinct axonal tract that exits the brain via the fused nervi corporis cardiaci-1 + 2. Cobalt backfilling from the corpora cardiaca filled 4 groups of cell bodies in the subesophageal ganglion. The presence in the subesophageal ganglion of extensive dendritic arborizations derived from the brain suggests interactions between neurosecretory cell groups in the 2 head ganglia.  相似文献

Interaction of cobalt(II) at micromolar concentrations with live cells of the plant-growth-promoting rhizobacterium Azospirillum brasilense (strain Sp245) and further transformations of the metal cation were monitored using 57Co emission Mössbauer spectroscopy (EMS). Cell suspensions of the bacterial culture (2.4×108 cells ml?1) were doped with radioactive 57CoCl2 (1 mCi; final concentration 2×10?6 M 57Co2+), kept under physiological conditions for various periods of time (from 2 min up to 1 hour) and then rapidly frozen in liquid nitrogen. Analysis of emission Mössbauer spectra of the frozen aqueous suspensions of the bacterial cell samples shows that the primary absorption of cobalt(II) at micromolar concentrations by the bacterial cells is rapid and virtually complete, giving at least two major forms of cobalt(II) species bound to the cells. Within an hour, the metal is involved in further metabolic transformations reflected by changes occurring in the spectra. The Mössbauer parameters calculated from the EMS data by statistical treatment were different for suspensions of live and dead (thermally killed) bacterial cells that had been in contact with 57Co2+ for 1 h, as well as for the cell-free culture medium containing the same concentration of 57Co2+. Chemical after-effects of the nuclear transition (57Co →57Fe), which provide additional information on the chemical environment of metal ions, are also considered. The data presented demonstrate that EMS is a valuable tool for monitoring the chemical state of cobalt species in biological matter providing information at the atomic level in the course of its uptake and/or metabolic transformations.  相似文献

In a previous paper (Bellman, Jacquez, and Kalaba,Bull. Math. Biophysics,22: 181–198, 1960) a model of the processes occurring in the exchange of a drug between capillary plasma, extracellular space and intracellular space was developed. This included the possibility of a reaction between the drug and a component of the intracellular space. The equations developed thus describe the events within a capillary bed. In the present paper, a simplified model of the body is set up. Each organ is treated as a single capillary bed and is linked to other organs via the circulation, in the parallel and/or series arrangements found in the body. Mixing in the circulation is included at the simplest possible level. The concentration of drug entering any one capillary bed is determined by the concentrations leaving all other capillary beds, the time lags, and mixing involved in the circulation. The equations describing these processes in conjunction with the equations of the processes occurring within each capillary bed lead to a large set of differential-difference equations.  相似文献

The anticonvulsant activity of bis(acetato)tetrakis(imidazole) copper(II), Cu(OAc)2(Im)4, was studied in normal mice using chemical convulsions induced by strychnine, thiosemicarbazide, picrotoxin, and pentelenetetrazol. Intraperitoneal administration of Cu(OAc) 2(Im)4, 50 mg/kg body mass, has delayed the onset of strychnine (3 mg/kg)-induced convulsion by 204% (p≤0.005) and thiosemicarbazide (20 mg/kg)-induced convulsant by 61% (p≤0.005). The changes in the onset of picrotoxin-(6 mg/kg) and pentelenetetrazol (50 mg/kg)-induced convulsions were not significant. The same dosage of the copper compound was effective in delaying the lethal time and reducing the mortality rate of treated animals. The anticonvulsant activity of Cu(OAc) 2(Im)4 complex against strychnine was not related to its constituents because the inorganic form of copper such as copper chloride, copper acetate, and the parent imidazole has no anticonvulsant activity. Other copper(II) complexes like copper(II)aspirinate and bis(acetato)bis(2-methyl imidazole) copper(II) were less effective.  相似文献
