通过15年的红壤稻田长期定位试验,研究了不同施肥模式下土壤微生物生物量磷(MB-P)对土壤有机碳和磷素变化的响应.结果表明红壤稻田有机碳源的长期投入和土壤有机碳的逐年升高使土壤微生物生物量碳(MB-C)维持在较高水平(>800 mg·kg-1),是稻田土壤MB-P(>16.0 mg·kg-1)提高的主要原因.长期不施磷肥条件下,土壤全磷含量与试验前相比显著降低(P<0.05),而土壤有机磷含量平均提高了29.3%;土壤亏损的磷形态主要是无机磷(Al-P、Fe-P、Ca-P和O-P),其中Al-P含量处于最低水平(平均0.5 mg·kg-1).另外,长期不施磷肥土壤的MB-P远高于Olsen法提取态磷(Olsen-P)(<7.0 mg·kg-1),而稻田土壤MB-P与Al-P呈显著相关(P<0.05),表明土壤微生物对稻田土壤Al-P、Fe-P、Ca-P和O-P的利用是促进其向有效磷方向转化的关键途径.磷肥配合有机养分循环利用不仅提高了土壤磷库的积累,而且通过土壤微生物的活化有效地提高了土壤磷的有效性.  相似文献

长期施磷对黄壤旱地磷库变化及地表径流中磷浓度的影响   总被引:20,自引:1,他引:19  
对贵州黄壤旱地进行采样以及盆栽试验,探索黄壤旱地磷库的变化及其对地表径流磷量的影响。结果表明,长期施磷后黄壤旱地无机磷占全磷的比例逐渐增高,Al-P、Fc-P和Ca-P积累的顺序为Fe-P>Al-P>Ca-P。长期施磷后黄壤旱地的有效磷和藻类可利用的总磷也不断积累,高磷(Olsen-P>25mg·kg^-1)旱地藻类可利用的总磷与Al-P、Fe-P和Ca-P的相关系数分别为0.859、0.903和0.650,Fc-P对藻类可利用磷量的影响起着最重要的作用。在模拟雨强为63.2mm·h^-1下,降雨30min后,低磷黄壤旱地(Olsen-P为4.62-15.9mg·kg^-1)径流中磷酸根磷含量仅为2.81-4.17μg·L^-1,生物有效性磷含量为0.723-0.876mg·L^-1;高磷黄壤旱地(Olsen-P)为29.4-59.2mg·kg^-1)径流中磷酸根磷含量达到0.026-0.714mg·L^-1,生物有效性磷含量增加到0.996-1.281mg·L^-1;高磷黄壤旱地地表径流磷量能加速水体富营养化的产生。  相似文献

黄壤稻田土壤微生物生物量碳磷对长期不同施肥的响应   总被引:3,自引:0,他引:3  
本研究以进行了22年的黄壤稻田长期定位试验为依托,分析了不同施肥模式下土壤碳(C)、磷(P)与微生物生物量C(MBC)、P(MBP)的变化及其耦合特征,旨在探讨黄壤地区合理培肥模式以提高土壤磷素有效性.试验包括10个处理,分别为不施肥(CK)、单施氮肥(N)、磷钾配合(PK)、氮钾配合(NK)、氮磷配合(NP)、氮磷钾配合(NPK)、单施有机肥(M)、3种有机无机肥配施(1/4M+3/4NP、0.5MNP、MNPK).结果表明:与不施肥相比,N和NK处理土壤有机碳(TOC)、全磷(TP)、MBC、MBP均有不同程度的降低,施磷处理(PK、NP、NPK)则有不同程度的提升;与不施肥和施用无机肥相比,施用有机肥各处理TOC、MBC、MBP及MBP/TP均显著增加,其中M和MNPK处理增幅最大.MBC/MBP、TOC/MBP、MBC/TP以施用有机肥处理最低,N处理最高.土壤MBC、MBP及其耦合关系与土壤TOC和有效磷均呈极显著相关,TOC是影响MBC、MBP、MBP/TP的直接因素,而有效磷则是影响MBC/MBP、TOC/MBP、MBC/TP的直接因素.土壤MBP及碳、磷耦合关系各指标可以有效区分单施化肥和施用有机肥的不同施肥方式,可作为评价黄壤稻田磷素肥力的生物学指标.配施有机肥是增加黄壤稻田磷有效性、提高土壤供磷潜力和保持土壤生物健康的有效途径.  相似文献

Fertilizers labelled with 32P were used to measure amounts of phosphorus, Ps and PF, taken up by Lolium perenne from available soil P and from P fertilizer respectively, when applied at a rate of 66 mg P·(kg soil–1) in greenhouse experiments. The quantity Ps of phosphorus taken up from soil in the presence of P fertilizer was compared to the quantity Po taken up from soil without P fertilizer. The quantity (Ps–Po) is positive for low Po values, i.e. in soils poor in available phosphorus, but is negative for high Po values indicating that an input of P fertilizer can induce a decrease in the utilization of available soil phosphorus. Moreover, for a given soil, the quantity (Ps–Po) depends on the chemical form of the fertilizer. The standard method of evaluation of P fertilizer efficiency is based on the assumption that Ps=Po, but Ps can differ from Po. This result can explain the contradictory data published from field experiments about the efficiency of the various P fertilizers.  相似文献

采用土壤培养方法研究了 3种纤毛虫对土壤微生物量及氮磷转化的影响 ,结果表明 ,向土壤接种肾形虫 ( Colopodia sp.)、尖毛虫 ( Oxytricna sp.)和澳毛虫 ( Australothrix sp.) ,特别是澳毛虫 ,显著地降低了土壤微生物碳。说明供试的原生动物与微生物之间存在消长关系。接种澳毛虫显著地降低了土壤有效磷含量 ,而肾形虫和尖毛虫对土壤有效磷含量影响很小 ,仅在培养后期显著地降低了土壤铵态氮含量 ,3种原生动物特别是澳毛虫 ,显著地降低了土壤氮矿化量和硝态氮含量 ,但提高了土壤铵态氮含量 ,说明 3种原生动物抑制了硝化作用 ,而增强了氨化作用。  相似文献

Synthetic sheep urine additions (59 g N m−2) were made to pastures on two soils, at Fasset, a semi-natural grassland, and Strathfinella, an improved pasture. Urine was applied to microplots in May and the soil, grass and soil solution analyzed 1, 2, 4, 5, 12 and 23 weeks after the addition. At Fasset, the grass was scorched by urine and the standing biomass decreased compared to the control, increasing only after 5 weeks. The absence of scorching at the Strathfinella site was attributed to a greater biomass of root mat to buffer the roots from exposure to NH3and a greater rainfall immediately following treatment. Scorching reduced the uptake of N and it was not clear if the greater contents of NH4 + and the increases in soil pH at Fasset compared with Strathfinella were the causes or symptoms of the scorch effect. Amounts of extractable organic N (DON) were similar in both soils and increased during the first 4 weeks and then decreased. Urine addition both increased and decreased DON at different times, but the overall mean values were unchanged. Urine application changed the distribution of P in the two soils, increasing the soil solution P at Fasset by 80 mg P m−2 and raising the P content of herbage at Strathfinella by 600 mg P m−2. In the soil solution, dissolved forms of molybdate reactive P, organic P and condensed P fractions were all increased by the urine addition. After 23 weeks, condensed P made the greatest contribution to soil solution P in both soils indicating that this fraction was the least available for plant uptake. 以7个磷效率不同的大豆基因型为材料,通过土壤盆栽试验进行石灰和磷肥处理,研究酸性土壤上缺磷和铝毒对大豆生长的交互影响及其基因型差异.结果表明:缺磷和铝毒是酸性土壤上同时存在的影响大豆生长的主要障碍因子,其中铝毒对大豆生长的限制更为严重;缺磷和铝毒对酸性土壤上大豆生长的影响具有显著的交互作用.同时施用石灰(降低铝毒)和磷肥(提高磷有效性)比单施石灰或单施磷肥处理对大豆生长的促进效果更显著;缺磷和铝毒对大豆磷吸收的影响远大于对氮、钾吸收的影响.合理种植大豆对酸性土壤具有较好的改良作用.同时施用石灰[1.103 g Ca(OH)2·kg-1土]和磷肥(2.018 g KH2PO4·kg-1土)可使酸性土壤pH值平均提高38.4%,交换性铝含量降低96.3%,有效磷含量提高3223.8%.种植磷高效大豆基因型比磷低效大豆基因型对酸性土壤的改良效果更好.  相似文献

This study was conducted to elucidate the effect of P fertilisation on the function of field communities of arbuscular mycorrhizal fungi (AMF) measured as P transport to flax. Two methods were applied to soil from a long-term field experiment with NaHCO3-extractable soil P levels of 24 and 50 mg kg-1in an experiment under controlled conditions: i) Measurement of plant growth and P uptake in the presence or absence of the fungicide benomyl and ii) measurement of hyphal P transport from a root-free compartment labelled with 32P. Benomyl successfully prevented mycorrhizal function. The absolute contribution of AMF to plant P uptake was of the same magnitude with or without P fertilisation at 27 days after sowing. Therefore, even though plants grown at the higher soil P level had greater P uptake, the relative contribution of AMF to P uptake was greater at the lower P level than at the higher P level (77 and 49% of total P uptake, respectively). The AMF in P-fertilized soil transported less P32 from the root-free compartment to the plant after 23 days than the AMF in unfertilized soil, but this difference disappeared in plants harvested after 27 and 32 days. The production of hyphae was largely similar in both fertilization treatments, indicating that the capacity for P uptake and transport by hyphae of the two AMF communities was similar. In spite of its central roles in cell cycle progression, senescence, and aging, knowledge about the posttranslational regulation of P16 (also known as INK4A and MTS1) remains limited. While it has been reported that P16 could be phosphorylated at Ser7, Ser8, Ser140, and Ser152, the corresponding kinases have not been identified yet. Here we report that IKKβ, a primary kinase for IκBα phosphorylation, is involved in P16 phosphorylation. Immunoprecipitation and kinase assays showed that IKKβ specifically binds to P16 and phosphorylates P16 at Ser8 in WI38 cells. Biochemical characterization of phosphomimetic Ser → Glu P16 mutants demonstrated that phosphorylation at Ser8 of P16 brings about a significant loss of its cyclin-dependent kinase (CDK) 4-inhibitory activity while P16 retains structurally and functionally intact upon phosphorylation at Ser7, Ser140, and Ser152. Our results reveal the novel role of IKKβ in P16 phosphorylation and broaden our understanding of the regulation of P16.  相似文献

为了明确低铁胁迫下磷素用量对大豆光合和磷/铁性状的影响及基因型差异,为磷、铁肥的合理施用提供理论依据,以前期筛选的6个磷高效基因型和6个磷低效基因型大豆为供试材料,设4个P∶Fe配比处理,分别为0∶30、30∶30、150∶30和300∶30(μmol·L-1),对大豆叶绿素荧光特性和磷、铁利用率进行了测定,利用单株粒重建立逐步回归方程并进行通径分析,通过因子得分综合评价磷高效和磷低效基因型对不同P∶Fe处理的响应。结果表明: 基因型效应、P∶Fe处理效应和两者互作效应对始花期(R1)光系统Ⅱ的相对电子传递速率(ETR)、光系统Ⅱ吸收的能量用于耗散为热量的比例(NPQ)、光系统Ⅱ吸收的能量用于进行光化学反应的比例(qL)的影响均达显著水平。典型相关分析表明,磷高效大豆基因型完熟期(R8)籽粒磷利用率与R1期光合速率呈负相关;磷低效大豆基因型R8期籽粒铁利用率与R1期NPQ呈正相关关系,而与R1期qL呈负相关关系;R1期PSⅡ实际光化学效率(ΦPSⅡ)与磷高效基因型呈负相关,而与磷低效基因型呈正相关,这表明R1期ΦPSⅡ可以作为鉴定低铁条件下不同磷效率大豆基因型的一个重要指标。利用因子得分综合评价发现,磷高效基因型表现为随着磷水平的上升而先降后升,而磷低效基因型则为先升后降,但两者拐点均出现在P∶Fe为30∶30处理下,这表明在低铁条件下P∶Fe为30∶30可以作为鉴定不同磷效率基因型的一个临界值。因而,在低铁地区种植磷高效大豆基因型时,磷肥的施用量至少要大于1∶1 (P∶Fe);而种植磷低效基因型时,磷肥的施用量不宜超过1∶1 (P∶Fe)。  相似文献

采用氯仿熏蒸提取法和磷酸苯二钠比色法分别测定了长期土壤培肥定位试验地所设置的不施肥、单施化肥、玉米秸秆 (低、中、高量 ) 化肥、厩肥 化肥以及休闲 (低量秸秆 化肥 )处理土壤微生物磷和磷酸酶活性。结果表明 ,各施肥处理均不同程度提高了土壤微生物磷 ;除休闲处理外 ,各施肥处理也不同程度提高了磷酸酶活性 ,这有利于为作物生长创造一种良好的土壤条件。石灰性土壤微生物磷与土壤全磷、速效磷之间存在明显的正相关 ,表明要改善石灰性土壤P利用率低的状况 ,提高土壤微生物磷不失为一条行之有效的生物学途径  相似文献

应用免疫组化SP法观察34例骨肉瘤中P53及P21WAF1的表达情况,探讨P53及P21WAF1表达与骨肉瘸临床病理学特征之间的关系,分析P53和P21WAF1在骨肉瘤中的作用及其相关性。结果可见,在34例骨肉瘤中,18例表达P53,阳性表达率52.94%;12例表达P21WAF1,阳性表达率35.29%。P53阳性表达与性别、肿瘤分化及是否转移复发有关(P<0.05),P21WAF1与肿瘤分化有关(P<0.01)。P53在中、低分化骨肉瘤中阳性表达率较高,而在高分化骨肉瘤中阳性表达率较低(70%、28.57%);P21WAF1在高分化骨肉瘤中阳性表达率较高,而在中、低分化骨肉瘤阳性表达率较低(85.71%、0%)。P53及P21WAF1表达呈负相关性(r=-0.537,P=0.001)。在骨肉瘤组织中P53表达上调及P21WAF1表达下调与骨肉瘤的恶性进展有关。  相似文献

Cholesterol plays an important role in cellular function and membrane compartmentalization and is involved in the interaction with more than a dozen of different proteins. Using three cholesterol-metabolizing cytochrome P450s (P450s 7A1, 46A1, and 11A1), we have developed a rapid and simple assay for measurements of nanomolar to micromolar cholesterol affinities. In this assay, the P450 is incubated with a fixed amount of radiolabeled cholesterol and varying concentrations of cold cholesterol followed by separation of free and protein-bound cholesterol via filtration through a membrane. Free cholesterol is found in the flow-through fraction, whereas P450 binds to the membrane. The radioactivity of the membranes is then measured, and a saturation curve is generated after correction for nonspecific binding of cholesterol to the filter. The validity of the filter assay was confirmed by spectral assay, a traditional method to evaluate the interaction of the P450 enzymes with their substrates. Two types of membranes, one binding positively charged proteins and another binding negatively charged proteins, were identified. These membranes were also found to hold proteins through hydrophobic interactions. Thus, the cholesterol binding properties of a wide variety of proteins could be characterized using this filter assay.  相似文献

洞庭湖区不同利用方式对土壤微生物生物量碳氮磷的影响   总被引:20,自引:3,他引:17  
以湖南省沅江市典型湖垸为代表,通过密集取样分析,研究了洞庭湖区不同利用方式条件下农田土壤微生物生物量碳、氮、磷的变化及其和土壤碳、氮、磷的关系,发现水田土壤碳、氮和微生物生物量碳、氮明显高于旱地,水田土壤中双季稻高于一季稻;土壤磷的含量旱地稍高于水田,但土壤微生物生物量磷水田稍高于旱地.尽管在水田土壤中微生物生物量碳、氮有明显的不同,但水田土壤微生物生物量磷维持在相对稳定的水平.典型样区土壤微生物生物量碳占有机碳的比例为0.65%~7.24%,平均3.00%;土壤微生物生物量氮占全氮的比例为0.98%~7.41%,平均3.81%;土壤微生物生物量磷占全磷的比例为0.16%~7.54%,平均2.80%.土壤C/N为3.87~17.31,平均9.15;BC/BN为4.06~9.29,平均7.26.土壤微生物生物量碳、氮与土壤碳、氮之间存在极其显著的线性相关关系,但土壤微生物生物量磷占全磷之间相关关系不显著.土壤微生物生物量碳、氮、磷之间的相关关系达到了极显著水平.不同的利用方式和耕作制度导致了土壤碳、氮和微生物生物量碳、氮的差异,土壤微生物生物量碳、氮能够很好地反映洞庭湖区农田土壤碳、氮水平.  相似文献

Temporal variations in plant production, plant P and some soil P (and N) pools were followed over 21 months in two New Zealand pasture soils of widely different P fertility status. Plant growth rates, and herbage composition at the high-fertility site, were closely linked to soil water use, with growth rates falling when soil water deficits exceeded 60 mm. Herbage P concentrations reflected P fertility, and varied with season, being generally higher in winter and lower in summer. A similar temporal pattern was also observed for labile organic P (NaHCO3-extractable P0) in both soils. In the low-fertility soil in spring, net mineralization was especially strong, but from early winter net immobilization occurred. Surprisingly, Olsen P also changed temporally in the high-fertility soil. The microbial biomass remained fairly constant throughout the year, whereas the P content of the biomass varied seasonally. Although microbial biomass was not a useful index of soil fertility, highest microbial P0 contents coincided with periods of maximum labile P0 mineralization, when herbage production was also at a peak. Net N-mineralization in the low-fertility soil, in contrast to the high-fertility soil, was low but varied seasonally, under standardised incubation conditions. Soil P and N dynamics were apparently synchronised in the low-fertility soil through soil microbial processes, with mineral N being negatively correlated with microbial P0 in samples collected two months later. The results of this investigation suggest that the demands of rapid and sustained pasture growth in spring and early summer can best be met by maximising the build-up of organic matter during the preceding autumn and winter. This practice could help to alleviate the common problem of feed shortage in North Island hill country pastures in late winter-early spring.  相似文献

A procedure has been described for the large-scale micropropagation of three Phragmites species, P. karka, P. communis and P. australis, from axillary buds excised from the main and side branches. Position of the buds on the branches had an effect on the bud break and establishment of the cultures under in vitro conditions. Lower buds of P. australis and middle buds of P. karka and P. communis were the most suitable. The presence of yeast extract as one of the ingredients of the sprouting medium helped in the early detection of systemic contamination. Multiple shoot formation and root initiation were obtained on Murashige and Skoog's basal medium supplemented with different concentrations of BA – 0.5 mg/l for P. karka, 0.25 mg/l for P. communis and 0.1 mg/l for P. australis– 0.5 mg/l Kn and 2% sucrose (w/v). Shoots and roots elongated on half-strength MS basal medium with 2% sucrose but without any plant growth regulators. A zone of root hair was observed in the case of P. australis. Hardening occurred on 95% of the plantlets within 30 days of transfer to the polyhouse. Over 10,000 plants were produced from three buds of each species within 9 months. 目的 了解宫颈癌中hMLH1、P27、P53表达情况及其与宫颈癌发生和发展的关系.方法 采用免疫组织化学技术SP法检测临床资料较完整的72例子宫颈鳞癌和31例慢性宫颈炎组织hMLH1、P27、P53蛋白表达.结果 (1)hMLH1在宫颈炎及宫颈癌组的表达率分别为61.3%和58.3%,差异无显著性(P>0.05);hM-LH1在宫颈癌各期中表达率分别为Ⅰ期60.0%,Ⅱ期52.6%,Ⅲ期33.3%,差异无显著性(P>0.05);hMLH1表达率与宫颈癌分级无关(P>0.05);(2)P27在宫颈炎及宫颈癌组的表达率分别为74.2%%和76.4%,差异无显著性(P>0.05);P27的表达率与宫颈癌分级和分期无关(P>0.05);(3)P53在宫颈炎及宫颈癌组的表达率分别为32.3%和65.3%,差异有显著性(P<0.05);P53在宫颈癌各级中的表达率分别为Ⅰ级36.4%,Ⅱ级73.0%,Ⅲ级67.9%,Ⅰ级与Ⅱ、Ⅲ级的表达率差异有显著性(P<0.05);P53表达率与宫颈癌分期无关(P<0.05);(4)hMLH1表达阳性与阴性的宫颈癌中P27的表达率为94.6%和60.0%,差异有显著性(P<0.05);P53的表达率为62.8%和62.1%,差异无显著性(P>0.05).结论 P53基因突变与宫颈癌的发生和组织分化程度有关;hMLH1与P27表达缺失和宫颈癌发生的关系尚不确切.hMLH1可能使宫颈癌中P27表达上调,而对p53基因突变累积无明显抑制作用.  相似文献
