The authors present the results of study of the protective function of cells of the local inflammatory focus caused by the administration of live brucella culture to guinea pigs immunized with a protective antigen in the optimal (0.6) and increased (2 mg) doses. Macrophages of guinea pigs vaccinated with the optimal immunized dose of the protective antigen phagocytized brucellae actively; an increased dose of the antigen suppressed the ingestive and the digestive functions of macrophages, this apparently being caused by the manifestation of the immunodepressive activity of the residual amount of lipopolysaccharide (endotoxin) in the protective antigen.  相似文献

Guinea pigs injected intradermally with antigen pulsed macrophages generate a population of immune T cells that proliferate in vitro on second exposure to antigen. T cells from F1 (2 X 13) guinea pigs immunized with DNP-OVA on one parental macrophage respond in vitro only to DNP-OVA on macrophages identical to those used for immunization and not to DNP-OVA associated with the other parental macrophages. These results demonstrate that the immunogenicity of antigen is dependent upon the macrophages used for priming in that, with this approach, strain 2 or 13 guinea pigs immunized with allogeneic macrophages pulsed with antigen do not respond to either allogeneic or syngeneic antigen-bearing macrophages. However, lysates of antigen-pulsed macrophages can still immunize either allogeneic or syngeneic recipient via their own macrophages. F1 (2 X 13) guinea pigs are immunized by insulin B chain pulsed strain 13 macrophages (responder) but not by strain 2 macrophages (nonresponder) suggesting that whether a F1 (nonresponder X responder) guinea pig recognizes antigen bound to a parental macrophage is genetically restricted before immunization to the same extent as the donor parental macrophages used for immunization.  相似文献

Spleen cells from C57BL/6 mice immunized with murine sarcoma virus (MSV) are capable of producing migration inhibition factor (MIF) in response to stimulation with a specific tumor-associated antigen prepared by solubilization with 3 M KCL. We have previously demonstrated that this response is T cell-dependent. Further investigations into the effector cells involved in the production of MIF have revealed that spleen cells from mice immunized with MSV cannot produce MIF when stimulated with tumor extract if the population has been previously depleted of macrophages. However, the response can be restored by adding nonimmune syngeneic macrophages but not by allogeneic macrophages. The inability of allogeneic macrophages to provide this function was not due to their increased suppressor activity since in mixing experiments they did not interfere with the ability of immune spleen cells to produce MIF. Furthermore, they were not defective since they could supply this "cooperative function" to appropriate F1 mice. The results indicate that macrophages are required for stimulation of MIF by soluble tumor antigens and that for efficient interaction the macrophages and lymphocytes must share some genetic similarities.  相似文献

In experiments on rabbits immunized with soluble protein antigen immune reactions were found to be accompanied by the production of lipofuscin in macrophages. This process was the morphological manifestation of the digestion of antigen by macrophages which thus acquired the ability to migrate in the organ and to form lymphoid follicules in the medullary zone of lymph nodes. The newly formed follicules seem to be the basis of pronounced specific immune response. In staphylococcal bacteriemia the phagocytic activity of macrophages was delayed, thus causing disturbances in lipofuscin production; as a result, the subsequent phases of immune response also lagged somewhat behind in time.  相似文献

Guinea pigs immunized with ABA-tyr in CFA respond well by skin test to ABA-tyr-pulsed macrophages but poorly to ABA-coupled macrophages and not at all to ABA-coupled red cells, thymocytes, or L2C cells. On the other hand, guinea pigs immunized with ABA-coupled macrophages do not respond to ABA-insulin or ABA-tyr-pulsed macrophages but do respond to ABA-coupled macrophages. Similarly guinea pigs immunized with Ars-NCS-coupled macrophages respond only to the homologous antigen. The specificity of these reactions is determined by how ABA is associated with the Ia-positive accessory cell. The presence of Ia molecule is not a sufficient condition since neither Ia-positive L2C cells nor spleen cells depleted of adherent macrophages are effective as immunogens or elicitors of response when coupled with ABA. These results suggest that the topography of the ABA and Ia complex formed on the accessory cell is the prime determinant of specificity for T-cells responses.  相似文献

Purified, recombinant-derived murine granulocyte-monocyte colony-stimulating factor was found to enhance the primary in vitro immune response to SRBC by murine spleen cells. In determining the mechanism of this augmentation, it was found that only splenic adherent cells and neither resting nor activated T cells nor B cells expressed specific receptors for GM-CSF. When splenic adherent cells were pulsed briefly with GM-CSF before addition to macrophage-depleted cultures, they reconstituted the PFC response to a significantly greater degree than did control macrophages. Splenic adherent cells incubated overnight with SRBC plus GM-CSF were also more efficient antigen-presenting cells than splenic adherent cells incubated with antigen alone. The mechanism of this enhanced antigen presentation was found to be due to a GM-CSF-dependent increase in the level of IL 1 secretion and Ia antigen expression. Consistent with these data was the finding that GM-CSF augmented IL 2 production by splenic T cells in response to suboptimal concentrations of Con A. Finally, the day 5 in vivo antibody response (as measured by serum titers) of mice immunized with a low dose of SRBC was enhanced by two daily inoculations of GM-CSF. Thus, the role that GM-CSF plays in augmenting immune responses may not be solely accounted for by its ability to cause the proliferation or differentiation of macrophages, but more than likely includes its ability to enhance the function of antigen-presenting macrophages.  相似文献

Mice protected against Schistosoma mansoni infection by intradermal (i.d.) immunization with nonliving larval or adult worm antigens plus bacterial adjuvant developed 24-hr skin test responsiveness to schistosome antigens with the histologic features of delayed hypersensitivity. Intraperitoneal antigen injection elicited a mononuclear cell-enriched exudative population containing macrophages activated for direct cytotoxicity against schistosomula and tumor cell targets. This was likely to be due to in vivo exposure to macrophage-activating lymphokines, since these cells were unresponsive to further lymphokine stimulation in vitro and splenocytes from immunized mice reacted to specific in vitro antigen challenge by production of lymphokines capable of conferring larvacidal activity upon control macrophages. In contrast, mice treated with schistosome antigens by i.v. injection, which were not protected against challenge infection, failed to develop delayed hypersensitivity or activated macrophages in response to specific antigen challenge in vivo, and the titers of macrophage-activating lymphokine produced by in vitro antigen-stimulated splenocytes from these mice were threefold to fourfold lower than those produced by cells from animals immunized by the i.d. route. Thus, sensitization for cell-mediated immune responses including lymphokine production and macrophage activation correlated with induction of resistance to S. mansoni in this model of vaccination.  相似文献

The data in this report describe a T cell proliferation assay with nylon wool column-purified murine lymph node lymphocyte from animals immunized by footpad injection of antigen in CFA. It was found that the in vitro immune response of sensitized T cells to soluble protein antigens was functionally dependent on the presence of adherent cells, more specifically macrophages, at all concentrations of in vitro antigen challenge. The response was due to T cells in that cytotoxic treatment of the immune lymphocyte cells with anti-Thy 1.2 serum and complement effectively eliminated the antigen-specific DNA synthetic responses. The antigen-specific proliferation of murine lymphocytes depleted of adhereent cells could not be reconstituted with either guinea pig macrophages nor murine fibroblasts, indicating the existence of species and cell type specificity. In contrast to previous observations in the guinea pig, soluble products of cultured adherent cells could at least partially replace the function of intact macrophages in the response to antigen.  相似文献

It is known that excretory/secretory antigens of Fasciola hepatica (ESFh) trigger a Th2-like immune response. Anapsos (A) is an aqueous hydrosoluble extract obtained from the rhizomes of the fern Polypodium leucotomos that has shown immunomodulator effects in some parasitic infections and immunological disorders. In this work we assess the effect of Anapsos and ESFh and Quillaja saponaria extract (Qs) on BALB/c mice and rat alveolar macrophages. Anapsos modulates the response of mice immunized with ESFh, decreasing IgG antibodies in A+ESFh- and A+Qs+ESFh-treated mice and triggering high levels of gammaIFN in spleen cell culture in comparison with ESFh- and Qs + ESFh--treated groups. Moreover, Anapsos showed statistically significant inhibitory effects on the nitrite production by rat alveolar macrophages prestimulated with lipopolysaccharide (LPS) as well as ESFh antigen in comparison with macrophages stimulated only with LPS. The application of ESFh and Anapsos combined avoids this inhibitory effect. Thus, Anapsos modulates the immune response against ESFh in naive mice and on the nitrite production in prestimulated rat aveolar macrophages.  相似文献

T cells that recognize the cross-reactive idiotype expressed on the heavy (H) chain of M104E (IgM, lambda 1) were induced in BALB/c mice by immunization with Dextran B-1355. T cells derived from mice immunized with 1 mg of Dextran B-1355 showed a marked proliferative response against M104E, whereas T cells from mice immunized with Ficoll or lesser amounts of Dextran B-1355 did not. BCL1Id, which had an immunoglobulin isotype identical to M104E, did not induce proliferation of the T cells. These T cells also proliferated against J558 (IgA, lambda 1) which shared the cross-reactive idiotype of the anti-alpha (1----3) glucosidic linkage antibody with M104E on the H chain. The T cells proliferated more efficiently against F(ab')2-104E, Fab-104E and H104E, the H chain of M104E, than against intact M104E. The T cell proliferative response against the idiotype on M104E or even H104E required macrophages as antigen-presenting cells (APC) and the response was inhibited when APC were treated with NH4Cl or chloroquine, inhibitors of antigen processing. Moreover, anti-CD4 antibody or anti-Ia antibody inhibited the proliferative response. These results indicated that anti-idiotypic T cells of the helper type, which recognized a cross-reactive idiotype associated with Ia molecules in processed form, could be induced physiologically through a network mechanism.  相似文献

Rabbits were immunized with human or bovine albumin at different intervals after birth and antibody formation was studied by haemagglutination of red cells sensitized with the relevant antigen. The intraperitoneal injection of antigen in amounts of 5 mg. induced antibody formation in some litters 16–20 days after immunization, if the animals were over three days old when immunized. In younger rabbits the same dose induced tolerance. Even when different methods of enhancing the effect of the antigen (Freund’s adjuvant, Al (OH)3, antigen-conjugated red cells, immune precipitates) or very small doses of antigen were used, antibody formation was still not detected before the 20th day of life. The use of131I-BSA did not demonstrate the immune phase of elimination of the antigen during 17 days after administration of the antigen, even in rabbits immunized 14 days after birth. The relationship of antibody formation to the induction of tolerance and the difference in the response of newborn rabbits to immunization with the different types of antigen is discussed.  相似文献

ATP-dependent Lon protease-deficient Salmonella enterica serovar Typhimurium (strain CS2022) appeared to invade successfully the mesenteric lymph nodes (MLN) and Peyer's patches (PP) of BALB/c mice and appeared to be easily eradicated by the host after oral immunization. As detected by flow cytometry, the population of major histocompatibility complex class I (MHC-I)-expressing macrophages and dendritic cells (DCs) was increased in the PP of mice immunized with CS2022 on day 6 after immunization. Thereafter, the population of splenic surface CD69(+) T lymphocytes prepared from mice immunized with CS2022 6 weeks prior to measurement increased as a result of the administration of the extracellular vesicles of RAW264.7 macrophage-like cells derived by Salmonella challenge. In addition, the proliferation of CD8(+) and even of CD4(+)T cells isolated from mouse spleens immunized with CS2022 was enhanced after cocultivation with naive DCs in the presence of the extracellular vesicles. These findings indicate that the extracellular vesicles prepared from the Salmonella-challenged macrophages carried salmonellae antigens to bystander DCs, thereby stimulating T-cell responses. Therefore, as antigen presentation after phagocytosis should be a central process in the T-cell activation that occurs in response to Salmonella infection, an oral immunization with CS2022 sufficiently induces T cell-mediated immunity in mice.  相似文献

Epithelioid cells from BCG-induced granulomas and macrophages from Mycobacterium leprae-induced granulomas were examined for their ability to act as accessory cells for T-cell proliferation to mitogen (Con A) and antigen (PPD). The granuloma cells were separated on a FACS using monoclonal antibody specific to guinea pig macrophages. Epithelioid cells (which are Ia negative) were able to support proliferation to Con A but not to antigen. Cultures containing Ia positive granuloma macrophages from M. leprae sensitized animals did not show responsiveness to Con A or to PPD. Oil-induced peritoneal exudate macrophages from BCG or M. leprae immunized animals were able to act as accessory cells for both mitogen and antigen proliferation. The nonresponsiveness of cultures containing epithelioid cells stimulated with PPD or M. leprae granuloma macrophages stimulated with Con A was not due to suboptimal or supraoptimal accessory cell:lymphocyte ratios.  相似文献

We reported previously that thyrotropin (TSH) enhanced the in vitro antibody response to a T cell-dependent antigen, sheep red blood cells (SRBC), as determined by direct plaque-forming cell (PFC) assay. The present studies were designed to determine the possible immunoregulatory function of TSH on lymphocytes immunized with the T-independent antigen Brucella abortus-TNP (BA-TNP) and the cellular components involved in such function. We report here that TSH enhanced the in vitro antibody response to BA-TNP as determined by direct PFC assays. Cell depletion studies showed that the TSH effect, although independent of macrophages, required the presence of T cells. Thus, pituitary--and possibly leukocyte--TSH appears to function as a lymphokine which may act via T cells to augment antibody production.  相似文献

The aim of the study was to determine the influence of twelve antibacterial antibiotics (various concentrations) on the activation of rabbit peritoneal macrophages. Macrophages were stimulated by filtrates of culture of lymphocytes T obtained from OVA immunized rabbits. Phagocytic activity and intracellular killing against Listeria monocytogenes were tested by fluorescence method. Penicillin G (0.4-50 mg/l), erythromycin and lincomycin (2.5-40 mg/l) used at all concentrations, were not exerting significant effects on activation of peritoneal macrophages. Cephalosporins, aminoglycosides, and rifampicin at low concentrations (0.4-5.0 mg/l) had no influence on phagocytosis and intracellular killing, also. Cephalosporins at concentration 10 mg/l (cephradine and cefamandole) and 50 mg/l (cefotaxime) inhibited intracellular killing and phagocytic activity. The same results were observed with ampicillin and ticarcillin (50 mg/l). The highest suppression effect was demonstrated using rifampicin at concentration 10 mg/l or more. Gentamicin, streptomycin and amicacin at concentrations 40 mg/l or more significantly inhibited macrophage activation in response to filtrates lymphocytes of culture. These inhibitions were more marked with gentamicin (10 mg/l) than amicacin (20 mg/l) or streptomycin (40 mg/l). All antibiotics did not stimulated the activity of peritoneal macrophages. The suppression activity of peritoneal macrophages by some antibiotics probably acts at the level of specific immune system by interfering with cytokine production.  相似文献

In a bid to characterize the antigens and immunization mechanisms which may be used to produce a protective response against L. donovani, role of lipid associated polysaccharide (LPS) antigen and whole antigen was evaluated. BALB/C mice were immunized with whole or LPS antigen in combination with one of three putative adjuvents (anti CD-2 antibody/FIA/0.85% Saline). LPS antigen emulsified in anti CD-2 antibody was found to induce significant antibodies in mice on day 28 against challenge with lethal dose of L. donovani. Immunoprophylactic properties of LPS and whole antigen was investigated on day 40 through cytokine elicitation (IL-2), MIF) in culture supernatants of spleen cells, but before that MHC-II expressed on macrophage was studied. The LPS antigen in combination with anti CD-2 antibody was found to be most immuno-reactive inducing higher MHC-II expression on macrophages which was associated with substantial rise in the level of MIF and IL-2. It coincided with decline in antibody titre in 100% mice immunized with LPS antigen while Leishmania injected as whole antigen failed to induce specific macrophage and T-cell response with all the above formulations. We surmise from our data that lipid associated polysaccharide antigen linked to anti CD-2 antibody has potential for eliciting protective immunity against Leishmania.  相似文献

As it is previously shown (Tsyrlova et al., 1986), the level of humoral immune response is not only determined by the reaction of peripheral lymphoid system on antigenic effect, but also is bound up with the observed stem blood cell (SBC) proliferation in bone marrow (Frindel et al., 1976, Kozlov et al., 1982). Dynamics of label accumulation in bone marrow was examined when injecting antigen--sheep red blood cells labeled by radioisotope 51Cr, 125I. The peak of label accumulation in bone marrow, accompanied by the increase in proliferative SBC, was observed on the 3rd day after antigen injection. Furthermore in the course of immunization 51Cr labeled macrophage assortment was changed in time in such a way that a greater number of macrophages was accumulated in bone marrow on the 3rd and 4th days in the immunized animals in comparison with the intact ones. The macrophages in bone marrow are likely to take part in antigen uptake and to mediate its effect on SBC proliferation.  相似文献

Mice were immunized with antigen (Rabbit Fab' fragments) attached to syngeneic, or f1 (semi-syngeneic) irradiated spleen cells. Specific anti-rabbit Fab' plaque forming cell numbers showed that the response towards antigen on syngeneic or F1 cells, was significantly lower than that towards the same antigen on allogeneic cells. By subsequent in vitro incubation of immune spleen cells with antigen followed by plaque assay, it was found that those spleen cells exhibiting lowered plaque forming cell numbers initially, (i.e., those mice immunized with antigen on syngeneic or F1 cell surfaces) showed, after incubation, a response equal to or greater than those cells which initially (before in vitro incubation) demonstrated a larger response (i.e. those mice immunized with antigen on allogeneic cells).  相似文献

Summary Lithium 3,5 diiodosalicylate (LIS), a chemical utilized for the noncytolytic extraction of cell surface antigens, was used in this study to extract glycoproteins from the cell membranes of L2C-EN leukemic blast cells. The crude soluble antigen (LIS-L2C) preparation was found to confer immunoprotection in syngeneic guinea pigs against a lethal challenge of L2C-EN. Titration of the crude LIS-L2C soluble antigen extract revealed that 1 mg antigen gave 100% protection against a 2×105 viable tumor cell challenge 2 weeks after immunization and that immunizing doses of 0.1 mg, 0.25 mg, and 0.5 mg soluble antigen afforded 17%, 66%, and 83% protection, respectively. The specificity of this immune response was demonstrated by the failure of guinea pigs immunized with 1 mg LIS extract prepared from another guinea pig tumor (line 10 hepatoma) to be refractory to a similar L2C tumor cell challenge. A cell-mediated immune response to the LIS-L2C soluble antigen was observed in animals, based on a positive delayed hypersensitivity response to the soluble antigen 5 weeks after immunization. Similarly, in vitro testing revealed a specific blastogenic recognition of the soluble antigen by immune leukocytes.  相似文献

In contrast to many other strains, inbred P strain mice fail to develop significant levels of resistance to challenge Schistosoma mansoni infection as a result of prior vaccination with radiation-attenuated cercariae. In this study, the relationship between defects in resistance and development of cell-mediated immune reactivity was examined. Although splenocytes from immunized P mice demonstrated deficiencies in production of macrophage-activating lymphokine(s) in response to either antigenic or mitogenic stimulation, other aspects of T lymphocyte responsiveness including blastogenesis, production of interleukin 2, interleukin 3 and macrophage chemotactic factor, as well as helper cell function for secondary plaque-forming cell response to a T-dependent antigen and allospecific cytolytic T cell reactivity, appeared to be comparable with those of C57BL/6 mice, a strain that is protected by vaccination against S. mansoni. FACS comparison revealed no significant deficits in percentages of Thy-1+, Lyt-1+, or L3T4+ splenocytes in vaccinated P mice. The P-associated defect in production of macrophage-activating factor appeared to be at the level of the T cell rather than the antigen-presenting cell, because macrophages from P mice could reconstitute the lymphokine-producing capacity of T-enriched splenocytes from immunized, resistant (C57BL/6 X P) F1 or B10.P mice, whereas the converse was not true. These results indicate that vaccinated P mice have a selective defect in T cell function for production of macrophage-activating lymphokine, which is manifested as a failure to produce activated larvicidal macrophages at the site of specific antigen challenge in vivo and may be associated with the failure of this strain to become resistant to S. mansoni.  相似文献
