The complete mitochondrial genomes of five tiger samples from three subspecies (P. t. sumatrae, P. t. altica, and P. t. tigris) were successfully obtained by using 26 specifically designed Panthera-specific primer sets. The genome organization and gene arrangement of the five tiger samples were similar to each other; however polymorphic tandem repeat sequences were observed in the control region (CR). This led to a difference in the genome lengths obtained from these five samples with an average size of 16,994 bp for the five tiger mitochondrial genomes. The nucleotide base composition was on average as follows: A, 31.8%; T, 27.0%; C, 26.6%; G, 14.6% and exhibited compositional asymmetry. Most of tiger mitochondrial genome characteristics are similar to those of other common vertebrate species; however, some distinctive features were observed in the CR. First, the repetitive sequence 2 (RS 2) contained two repeat units of 80 bp and the first 15 bp of what would be the third repeat motif. The repetitive sequence 3 (RS 3) contained 47–50 repeat motifs of a shorter 8 bp (ACGTAYAC)n. Second, length heteroplasmy polycystosine (poly-C) stretches was observed at the end of the HV I locus in all tiger samples.  相似文献

Long-term monitoring of tiger population is essential in any human-dominated landscape as globally their population is showing a declining trend due to unrelenting stress or stochastic events. In view of that, Bengal tiger population was monitored in Pench Tiger Reserve (Pench), Madhya Pradesh of Central India between May 2006 and April 2013. The population, age-sex ratios, survival rate, growth rate, and recruitment pattern of adult (≥36 month), subadult (≥18 to 36 month), and cubs (≤15 month) were studied using camera trap and radio-telemetry techniques. Overall, tiger density ± SE/100 km2 using Maximum Likelihood Spatial Explicit Capture Recapture method was 5.5 ± 1.6 in first trapping period or year and 3.7 ± 0.8 in last or seventh trapping year. The estimated survival rate of all tiger (n = 66) was 0.66 (±0.04), whereas mean annual growth rate (±SE) was 1.15 (±0.11) i.e. 15%. The sex ratio (male/female) found to be female biased and more than 80% of overall recruitment was contributed by female tiger. The dispersal of subadults in the tiger population contributed gradual fluctuation in survival rate and annual growth rate, as resident adult tiger population remained almost stable over the study period. As the overall tigers are surviving in a small population size and operating at carrying capacity, the important corridors connecting with neighboring population of Kanha Tiger Reserve and Satpura Tiger Reserve may ensure constant emigration and immigration to reduce the chances of genetic drift or inbreeding in the Pench tiger population.  相似文献

Haematological and biochemical studies were conducted on 12 clinically healthy tigers of Central India. The range and mean (with one standard deviation), respectively for the parameters examined were: red blood cells, 4.66 to 9.15, 7.9 ± 1.42, 106/μl; haemoglobin, 7.8 to 13.8, 12.8 ± 1.65 g/dl; packed cell volume, 36 to 45, 38 ± 2.54; icterus index, 2 to 5, 2 ± 1.51 U; erythrocyte sedimentation rate, 14 to 26, 21 ± 4.21 mm at 1 h; white blood cells, 6.2 to 11.05, 8.5 ± 1.49, 103/μl; neutrophils, 57 to 75, 60 ± 5.08%; lymphocytes, 18 to 35, 30 ± 4.56%; monocytes, 2 to 6, 5 ± 1.21%; eosinophils, 2 to 6, 4 ± 1.3; basophils, 0 to 4, 1 ± 1.21; plasma albumin, 2.1 to 4.6, 3.5 ± 0.99 mg/dl; total protein, 3.7 to 8.7, 6.4 ± 1.88 mg/dl; total bilirubin, 0.4 to 3.2, 1.9 ± 1.21 mg/dl; creatinine, 1.6 to 4.6, 2.90 ± 1.03 mg/dl; blood urea nitrogen, 6.5 to 48.2, 27.90 ± 13.77 mg/dl; glutamic pyruvic transaminase, 21.2 to 109.0, 67.88 ± 27.84 IU/L and glutamic oxaloacetate transaminase, 14.4 to 84, 57.96 ± 17.27 IU/L; index conspicuous erythrocyte sedimentation rate; absence of reticulocytes and predominance of neutrophils.  相似文献

An inexpensive, time-saving and reliable method, polymerase chain reaction with confronting two-pair primers (PCR-CTPP), was developed for sex identification in tiger (Panthera tigris) based on zinc finger alleles (ZFX/ZFY). A site of “C/G” transversion representing fixed differences that discriminated between ZFX and ZFY exons among felids was identified for primers designing. This primer set was successfully tested on samples including blood, shed hairs, dried skin, and stool which contained potential contamination caused by prey DNA. Cross species tests shown that this primer set was also useful for sex identification in four other endangered felids.  相似文献

Background

Bengal tiger Panthera tigris tigris the National Animal of India, is an endangered species. Estimating populations for such species is the main objective for designing conservation measures and for evaluating those that are already in place. Due to the tiger's cryptic and secretive behaviour, it is not possible to enumerate and monitor its populations through direct observations; instead indirect methods have always been used for studying tigers in the wild. DNA methods based on non-invasive sampling have not been attempted so far for tiger population studies in India. We describe here a pilot study using DNA extracted from faecal samples of tigers for the purpose of population estimation.

Results

In this study, PCR primers were developed based on tiger-specific variations in the mitochondrial cytochrome b for reliably identifying tiger faecal samples from those of sympatric carnivores. Microsatellite markers were developed for the identification of individual tigers with a sibling Probability of Identity of 0.005 that can distinguish even closely related individuals with 99.9% certainty. The effectiveness of using field-collected tiger faecal samples for DNA analysis was evaluated by sampling, identification and subsequently genotyping samples from two protected areas in southern India.

Conclusion

Our results demonstrate the feasibility of using tiger faecal matter as a potential source of DNA for population estimation of tigers in protected areas in India in addition to the methods currently in use.

Livestock depredation by carnivores cause substantial human carnivore conflict and subsequently decreased support for carnivore conservation. Thus, understanding carnivore diet with respect to wild prey availability has major implications to determine the reasons behind livestock depredation. A study was conducted to investigate food habits and prey use of tiger at four study sites (Dudhwa National Park, Kishanpur Wildlife Sanctuary, Pilibhit Forest Division and Katerniaghat Wildlife Sanctuary) in Dudhwa landscape, Terai Region, North India for further understanding of prey–predator relationship and partial impact of wild prey availability on livestock depredation by tiger through scat analysis. Scat analysis shows that the tigers depend mostly on medium sized prey throughout the study area (74.11, 73.58, 71.79, 47.62%). In Dudhwa National Park and Kishanpur Wildlife Sanctuary, predation was attempted subsequently on wider prey variety of eleven and nine different available prey species where livestock depredation were only 3.77 and 5.36% respectively. While, in absence of wider prey variety, large sized livestock (21.91, 16.55%) and nilgai (24.41, 5.57%) contributed much higher in tiger diet in Pilibhit Forest Division and Katerniaghat Wildlife Sanctuary respectively. Our study suggested that availability of prey variety has an important role in reduced livestock depredation. Medium sized preys were mostly contributing in tiger diet and seems to be a significant parameter for sustaining tiger population where abundance of large sized prey is rare. Conservation of medium sized preys is important but along with natural restoration of the population of large sized prey species like sambar and swamp deer is essential in order to reduce livestock depredation.  相似文献

The shallow (1.5 m) Pulicat Lagoon in India supports a variety of migratory birds (125 species), especially flamingos and storks. Flamingo populations in this area are dominated by Phoenicopterus roseuis, and they are densely distributed in shallow areas and fringes of the lake where the water level is below 40 cm. Eighteen flamingo groups are distributed around the lagoon with ~750 individuals per group. Flamingo populations are greatest in places where fish, algal, and benthic faunal biomass are highest. Physiological adaptations and morphology of flamingo are important criteria for selecting feeding locations.  相似文献

Reintroduction is a viable conservation strategy for large carnivores however such reintroductions require robust feasibility assessments, considering ecological, management, and social factors, prior to implementation. Plans are being developed in a number of tiger range countries, including Cambodia, for tiger Panthera tigris reintroductions in response to local and national extinctions. We provide a framework for undertaking feasibility assessments for tiger, and other large carnivore, reintroductions and present a number of methodological tools, and appropriate indicators, for conducting such assessments. We apply the framework to plans by the Royal Government of Cambodia for tiger reintroduction into Srepok Wildlife Sanctuary (formally Mondulkiri Protected Forest), eastern Cambodia. Srepok Wildlife Sanctuary forms part of a large (>13,000-km2) trans-boundary conservation landscape with potential to support a tiger source population. Current ungulate prey densities, assessed through robust line-transect sampling, at approximately 5.0 individuals per km2 may be sufficient to support a breeding tiger population. However levels of protected area management and law enforcement fall below global standards for tiger recovery. Local communities, though supportive of conservation efforts, also identified a number of concerns regarding reintroduction. Therefore current ecological, social, and management conditions within SWS are not currently suitable for tiger reintroduction. However with improved and more effective law enforcement, combined with robust monitoring of the indicators within our framework, such conditions could be met. We recommend that our framework for assessing landscape suitability for reintroductions offers an effective road map for reintroduction-based recovery of tiger populations across tiger range countries.  相似文献

Many grouper (Epinephelidae) species aggregate to spawn at specific times of year, at consistent locations, and at specific times of day and phases of the moon. The tiger grouper (Mycteroperca tigris) is widely distributed in the Caribbean Sea and aggregates to spawn just after the full moon. In January and February 2003–2005, we conducted visual surveys of a tiger grouper spawning aggregation at Glover’s Reef, Belize. The primary objectives of this study were to describe grouper spawning activities with respect to the timing of the lunar cycle, identify and describe male and female spawning behaviors, and estimate the number of grouper spawning events each day. The peak number of both male and female tiger groupers counted each year occurred from 6 to 10 d after the full moon. The peak count of males was 1.6–2.2 times greater than the peak count of females. Six categories of male–male and six categories of male–female interactions were identified. All male tiger groupers displayed an ability to rapidly change color; this was often triggered by a close encounter with other male or female tiger groupers. The daily spawning activity was remarkably predictable. The first spawning occurred on average 8.1 min before sunset, and the last occurred 5.1 min after sunset. On average, less than half of the females at the spawning site spawned each day, but some females spawned more than once in a given day. Females appeared to choose their mates.  相似文献

New combinations are proposed in anticipation of the Polygonaceae treatment in the forthcoming volume of Intermountain Flora: Polygonum kelloggii var. esotericum, P. kelloggii var. watsonii , Rumex densiflorus var. pycnanthus , R. salicifolius var. utahensis, and R. occidentalis var. tomentellus. Typifications are proposed to facilitate ongoing studies in Polygonaceae and to maintain current usage.  相似文献

Two repeated DNA sequences isolated from a partial genomic DNA library of Helianthus annuus, p HaS13 and p HaS211, were shown to represent portions of the int gene of a Ty3 /gypsy retroelement and of the RNase-Hgene of a Ty1 /copia retroelement, respectively. Southern blotting patterns obtained by hybridizing the two probes to BglII- or DraI-digested genomic DNA from different Helianthus species showed p HaS13 and p HaS211 were parts of dispersed repeats at least 8 and 7 kb in length, respectively, that were conserved in all species studied. Comparable hybridization patterns were obtained in all species with p HaS13. By contrast, the patterns obtained by hybridizing p HaS211 clearly differentiated annual species from perennials. The frequencies of p HaS13- and p HaS211-related sequences in different species were 4.3x10(4)-1.3x10(5) copies and 9.9x10(2)-8.1x10(3) copies per picogram of DNA, respectively. The frequency of p HaS13-related sequences varied widely within annual species, while no significant difference was observed among perennial species. Conversely, the frequency variation of p HaS211-related sequences was as large within annual species as within perennials. Sequences of both families were found to be dispersed along the length of all chromosomes in all species studied. However, Ty3 /gypsy-like sequences were localized preferentially at the centromeric regions, whereas Ty1/ copia-like sequences were less represented or absent around the centromeres and plentiful at the chromosome ends. These findings suggest that the two sequence families played a role in Helianthusgenome evolution and species divergence, evolved independently in the same genomic backgrounds and in annual or perennial species, and acquired different possible functions in the host genomes.  相似文献

Coenagrionid damselflies are in general decline in the British Isles. Numerous factors have been implicated in the loss of these species including recent speculation that hybridisation between congeners may result in species decline. Here we use a panel of 12 microsatellite loci to examine levels of genetic divergence and the likely occurrence of hybridisation in five populations of Coenagrion puella and C. pulchellum using samples from four sites in south-east England. Coenagrion puella and C. pulchellum were highly genetically divergent, and there was no evidence of hybridisation between any of the populations examined, even where C. puella and C. pulchellum were sympatric. There was some suggestion that C. pulchellum was less genetically diverse than C. puella, though this may have been a result of ascertainment bias associated with cross-species application of microsatellite markers. We conclude that there is no evidence that hybridisation between C. puella/pulchellum could be responsible for the on-going demographic decline in C. pulchellum. Nevertheless, further genetic studies such as this one are likely to provide estimates of diversity, population structure and dispersal capacity that will be invaluable in future conservation management strategies for coenagrionid damselflies. Background

Gonadotropin releasing hormone (GnRH) is responsible for stimulation of gonadotropic hormone (GtH) in the hypothalamus-pituitary-gonadal axis (HPG). The regulatory mechanisms responsible for brain specificity make the promoter attractive for in silico analysis and reporter gene studies in zebrafish (Danio rerio).

Results

We have characterized a zebrafish [Trp7, Leu8] or salmon (s) GnRH variant, gnrh 3. The gene includes a 1.6 Kb upstream regulatory region and displays the conserved structure of 4 exons and 3 introns, as seen in other species. An in silico defined enhancer at -976 in the zebrafish promoter, containing adjacent binding sites for Oct-1, CREB and Sp1, was predicted in 2 mammalian and 5 teleost GnRH promoters. Reporter gene studies confirmed the importance of this enhancer for cell specific expression in zebrafish. Interestingly the promoter of human GnRH-I, known as mammalian GnRH (mGnRH), was shown capable of driving cell specific reporter gene expression in transgenic zebrafish.

Conclusions

The characterized zebrafish Gnrh3 decapeptide exhibits complete homology to the Atlantic salmon (Salmo salar) GnRH-III variant. In silico analysis of mammalian and teleost GnRH promoters revealed a conserved enhancer possessing binding sites for Oct-1, CREB and Sp1. Transgenic and transient reporter gene expression in zebrafish larvae, confirmed the importance of the in silico defined zebrafish enhancer at -976. The capability of the human GnRH-I promoter of directing cell specific reporter gene expression in zebrafish supports orthology between GnRH-I and GnRH-III.

A genetic transformation system has been developed for callus cells of Crataegus aronia using Agrobacterium tumefaciens. Callus culture was established from internodal stem segments incubated on Murashige and Skoog (MS) medium supplemented with 5 mg l−1 Indole-3-butyric acid (IBA) and 0.5 mg l−1 6-benzyladenine (BA). In order to optimize the callus culture system with respect to callus growth and coloration, different types and concentrations of plant growth regulators were tested. Results indicated that the best average fresh weight of red colored callus was obtained on MS medium supplemented with 2 mg l−1 2,4-dichlorophenoxyacetic acid (2,4-D) and 1.5 mg l−1 kinetin (Kin) (callus maintenance medium). Callus cells were co-cultivated with Agrobacterium harboring the binary plasmid pCAMBIA1302 carrying the mgfp5 and hygromycin phosphotransferase (hptII) genes conferring green fluorescent protein (GFP) activity and hygromycin resistance, respectively. Putative transgenic calli were obtained 4 weeks after incubation of the co-cultivated explants onto maintenance medium supplemented with 50 mg l−1 hygromycin. Molecular analysis confirmed the integration of the transgenes in transformed callus. To our knowledge, this is the first time to report an Agrobacterium-mediated transformation system in Crataegus aronia.  相似文献

The wolf spider Pardosa cribata Simon is the most abundant ground-dwelling spider inhabiting citrus orchards in eastern Spain. However, little is known about its activity-density and its predatory role in the citrus agrosystem. Here we report on the activity-density of P. cribata monitored by pitfall traps, and on its capacity to prey on two citrus pests that appear both in the citrus canopy and the ground cover, Ceratitis capitata (Wiedemman) and Myzus persicae (Sulzer), respectively. Pardosa cribata was present in citrus orchards throughout the year, with a peak in spring and a higher peak in summer. Pardosa cribata preyed on adults and third-instar larvae but not on pupae of C. capitata. A type II functional response was obtained for teneral-like adults, with an estimated attack rate (a′) of 0.771 ± 0.213 days−1 and a handling time (T h) of 0.051 ± 0.013 days. Pardosa cribata also preyed efficiently on M. persicae, giving a type II functional response with an estimated attack rate and handling time of 2.833 ± 0.578 days−1 and 0.031 ± 0.001 days, respectively. The data reported here indicate that this wolf spider could play an important role in regulating both these pests, and therefore might contribute to developing conservation biological control strategies for citrus pests. We have investigated the floral ontogeny of Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis (of the eucalypt group, Myrtaceae) using scanning electron microscopy and light microscopy. Several critical characters for establishing relationships between these genera and to the eucalypts have been determined. The absence of compound petaline primordia in Arillastrum, Allosyncarpia, Stockwellia and Eucalyptopsis excludes these taxa from the eucalypt clade. Post-anthesis circumscissile abscission of the hypanthium above the ovary in Stockwellia, Eucalyptopsis and Allosyncarpia is evidence that these three taxa form a monophyletic group; undifferentiated perianth parts and elongated fusiform buds are characters that unite Stockwellia and Eucalyptopsis as sister taxa. No floral characters clearly associate Arillastrum with either the eucalypt clade or the clade of Stockwellia, Eucalyptopsis and Allosyncarpia.We gratefully acknowledge Clyde Dunlop and Bob Harwood (Northern Territory Herbarium) for collecting specimens of Allosyncarpia, and Bruce Gray (Atherton) for collecting specimens of Stockwellia. Single (unpaired) vegetative cells of freshwater pennate diatom Neidium cf. ampliatum differentiated into gametangia and produced a single zygote (auxospore) via a pedogamic process. Hippophae rhamnoides ssp. sinensis occurs mainly in the arid regions of northwest China. The wood stands of this subspecies play an important role in maintaining the local ecosystems in these regions. In addition, the genetic characteristics are essential to understand the historical range changes of this subspecies and its morphological differentiation with other subspecies. In this study, we developed nine microsatellite loci for this subspecies for the first time. We used the combining biotin capture method to enrich AG/CT/AC/GT/CG/GTG/CCA microsatellites. Twenty-six microsatellites were isolated from the enriching library and nine of them were found to be polymorphic through screening 12 distantly distributed individuals. The number of alleles per locus ranged from three to twelve and expected heterozygosity from 0.2659 to 0.4767, respectively. We further performed cross-priming tests in another subspecies and two congeneric species. These firstly isolated loci will provide a useful tool to investigate the genetic structure of this subspecies and its morphological differentiation from the other subspecies.  相似文献
