The bronchial sensitivity to acetylcholine (ACh) of guinea pigs of various strains was investigated to clarify strain differences. Inbred Strain 2, Strain 13 and JY-1 and non-inbred Hartley strain (two colonies) were used in this experiment. (1) Guinea pigs were exposed to 0.08% ACh aerosol and the time needed to produce falling down (TNPFD) was determined. Mean +/- standard error of TNPFD (n = 14 per group) of animals was 182 +/- 28 sec, 148 +/- 22 sec, 210 +/- 30 sec, 342 +/- 24 sec and 406 +/- 36 sec in Strain 2, Strain 13, JY-1, Hartley (Japan SLC) and Hartley (Hitachi), respectively. There was a significant difference in TNPFD between inbred strains and non-inbred strains (P less than 0.05 or P less than 0.01), indicating that inbred strains had higher sensitivity. (2) Guinea pigs were exposed to 20-5000 micrograms/ml ACh for 2 min. The mean dose threshold as determined by transcutaneous oxygen pressure was 524 micrograms/ml, 424 micrograms/ml, 614 micrograms/ml, 1317 micrograms/ml and 1651 micrograms/ml (n = 14 per group) in Strain 2, Strain 13, JY-1, Hartley (Japan SLC) and Hartley (Hitachi), respectively. Inbred strains showed lower dose thresholds than non-inbred strains. (3) Isolated trachea-lungs of 5 guinea pigs were perfused with 10(-9)-10(-5) g/ml ACh to determine strain differences. Dose response curves of animals of inbred strains shifted to the left (lower concentrations), unlike those of non-inbred strains, suggesting that inbred strains had higher sensitivity to ACh than non-inbred strains.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Frequencies and relative durations of eleven ethologically defined behavioral units were measured on paradise fish of two inbred strains and 9 recombinant inbred lines derived from their cross and then maintained under inbreeding. Significant genotype-dependent differences were found in all but one behavior units. Strain distribution patterns showed polygenic genetic determination of all units, and in several cases non-allelic genic interactions seem to contribute to the variation. Additive and interaction components of the progenitor means and minimal number of effective factors were estimated for each behavior unit.  相似文献

Random-bred Hartley and inbred Strain 2 and Strain 13 guinea pigs were inoculated for acute experimental autoimmune encephalomyelitis (EAE). Sixty-six percent (69/103) of the Hartleys developed signs of EAE while the remaining 34% (34/103) were resistant. No Strain 2 and all Strain 13 guinea pigs developed EAE. Histologic examination of nervous tissue revealed that susceptible Hartleys and Strain 13 and Strain 2 animals had lesions characteristic of EAE. Tissue from resistant Hartleys showed fewer and less severe changes. Lymphocyte-transformation assays with EAE-inducing and noninducing antigens and T-cell mitogens revealed three different sets of responses in vitro: (i) lymphocytes from all animals responded to mitogens; (ii) lymphocytes from susceptible animals responded to EAE-inducing antigens; and (iii) lymphocytes from resistant Hartleys were suppressed from responding to the mitogens solely by EAE-inducing antigens. Plasmas from all EAE-sensitized animals had equivalent anti-myelin basic proteins (MBP) antibody titers and skin tests of EAE-inoculated Hartleys were all positive for MBP sensitization. Therefore, resistance and reduced histologic changes characteristic of EAE correlated with a disease-specific antigen-induced suppression of lymphocyte responses to T-cell mitogens. This suggests that clinical resistance to EAE in Hartley guinea pigs is mediated by an immunologic suppressor mechanism.  相似文献

A new esterase polymorphism was identified in epididymal homogenates from inbred rat strains by polyacrylamide gel electrophoresis. The inbred rat strains showed either fast (A) or slow (B) bands. Strain distributions of the phenotypes differed from those of other esterase loci. Genetic analyses revealed that the polymorphism is controlled by codominant alleles (Es-19a and Es-19b) and is not linked to linkage groups, I, II, IV, V, VI, XIII of the rat.  相似文献

在我国,昆明小鼠作为一种实验动物广泛应用于药理和遗传学相关的研究领域。但由于昆明小鼠属于远交群,而且不同地区的种群间已经出现了严重分化,缺乏具有显著特征的近交系,这使得它在生物学上的应用受到了很大的限制。研究人员已经以昆明小鼠为背景培育出了几个可能的近交系,但由于缺乏可靠的遗传检测,至今未得到广泛的认可和应用。文章收集昆明小鼠的4个已经60代以上兄妹交配繁殖的可能近交系,并以两个标准近交系BALB/c和C57BL/6为参照,利用30个微卫星标记对每个品系的5只小鼠进行了微卫星基因分型,进而分析其遗传纯度。结果发现,品系A1和品系N4在本研究所用的30个位点均呈纯合状态;而T2和N2均在D15Mit16位点呈杂合状态。本研究第一次为我国昆明小鼠近交系的遗传学纯度提供了可靠的分子水平证据。今后应当加强昆明小鼠近交系的标准化,以扩大其在遗传学方面的应用。  相似文献

通过对活性污泥中硝化细菌的富集,使硝化细菌浓度达10^7个/ml。在硅酸胶平板上分离到硝化细菌的纯培养物。经鉴定主要有三种菌,简称N-1,N-2和N-3,N-1为亚硝化单胸菌,N-2为硝化杆菌,N-3为硝化刺菌,对三力的硝化强度进行了研究。16d时,N-1的亚硝化率为55.6%,N-2的硝化率为65.5%,N-3的硝化率为93.7%。三株菌在工业化污水处理中将有良好的应用前景。  相似文献

Strain differences among inbred mice is demonstrated for the relative amounts of peritoneal ascites fluid induced by the injection of Freund's adjuvant and antigen. Females tend to produce more fluid than males; the lower level in males is attributable to the action of male hormone. Both genetic and epigenetic factors are determinants in the response levels; genetic factors differentiate the inbred strains of mice, epigenetic factors underlie the variations within a strain and the difference between sexes.  相似文献

The Ah locus encodes a cytosolic receptor which controls the induction of enzymes that metabolize drugs, chemical carcinogens, and other environmental pollutants. B6NXC3N recombinant inbred lines have been developed from the progenitors C57BL/6N and C3H/HeN inbred mouse strains. Ah phenotyping at each generation has resulted in the establishment of some lines containing high levels of the high-affinity Ah receptor; other lines, very low levels. A genetic model involving two unlinked loci is offered to explain the distribution of Ah receptor levels among (C57BL/6N) (C3H/HeN)F2 individuals. Between generations 7 and 13, individual females and males from the B6NXC3N recombinant inbred lines were crossed with DBA/2N males and females. Presence of high levels of the high-affinity Ah receptor in both female and male B6NXC3N mice was found to be associated with greater fertility, fitness, and longer life span. The data suggest that these parameters are correlated with the Ah locus or a closely segregating gene.  相似文献

本文选用刺芹侧耳Pleurotus eryngii 24号(广杏)为自交材料,对其自交S1代群体的质量性状(颜色、畸形、生长特性)和数量性状(形状、产量、大小、数目)进行了综合分析.分析结果表明:自交后代的出菇率为70%,自交导致后代群体菌丝平均生长速度和平均产量都低于亲本,16%的后代菌株单产高于亲本,最高单产达204.0g,高出亲本45.7%.木屑培养基上菌丝日均生长速度与产量无显著相关性(R=-0.028).均菇数目与单产成显著正相关性(R=0.543).大部分菌株(52%)菌盖颜色为亲本颜色(灰色),自交后代出现20%非亲本形状(棒状)菌株,畸形菇占19%.后代群体盖径、柄径、柄长的变异程度相当,差异性不显著.可根据育种目标对自交子代的较优菌株进行定向选择利用.  相似文献

During late pregnancy, female mice of the DBA/2J inbred strain are more likely to exhibit aggressive behavior toward a standard stimulus intruder male than C57BL/6J females. This strain difference can not be accounted for by differences in circulating levels of progesterone (P) since pregnant DBA/2J and C57BL/6J females exhibit similar patterns of the steroid throughout pregnancy. Upon receiving subcutaneously implanted Silastic capsules containing P, virgin DBA/2J mice are more likely than virgin C57BL/6J to respond to the steroid by exhibiting aggression. Strain differences in the aggressive behavior exhibited by pregnant mice may be related to genotype-based variation in central neural tissue sensitivity to P.  相似文献

在筛选氮素利用高效茄子自交系的基础上,利用Griffing-Ⅱ设计,在两个氯素水平下(450kg/hm2争不施氮)分析了7个茄子自交系氮素利用效率相关性状的配合力.结果表明,两个氮素水平下,氮效率受加性效应和非加性效应的共同控制;氮素吸收总量和氮素利用效率则以加性遗传为主.氮高效育种中,自交系609和749在两个氮素水平下均有较高的氮效率一般配合力.与高氮处理相比,低氮条件下氮素利用效率相关性状的广义遗传力有所增大,而狭义遗传力多为递减趋势.  相似文献

A new variant of kidney esterase in the DK/Nac rat strain is reported. The new esterase was tentatively named ES-4C determined by a third allele of the Es-4 locus of Linkage Group V (LGV). Strain distribution was surveyed using 17 inbred strains, but no strain except for the DK/Nac strain possessed the ES-4C type. Although we surveyed outbred stocks (Jcl: Wistar and Jcl: SD) we could not find rats carrying the ES-4C type. Genetic analysis of the ES-4C type was carried out using mating experiments between DK/Nac and BUF/Nac (ES-4B). The results indicated that the new variant was controlled by the Es-4 locus and it was named the Es-4c allele.  相似文献

Summary Strain identification in Zea mays by restriction fragment length polymorphism should be feasible due to the high degree of polymorphism found at many loci. The polymorphism in maize is apparently higher than that currently known for any other organism. Five randomly selected maize inbred lines were examined by Southern filter hybridization with probes of cloned low copy sequences. Typically, several alleles could be distinguished among the inbred lines with any one probe and an appropriately selected restriction enzyme. Despite considerable polymorphism at the DNA level, 16 RFLP markers in three inbred lines of maize were examined for six to 11 generations and found be stable. Mapping of RFLP markers in maize can be accelerated by the use of B-A translocation stocks, which enable localization of a marker to chromosome arm in one generation. The use of recombinant inbred lines in further refinement of the map is discussed.  相似文献

Summary Inbred lines of corn (Zea mays L.) have been characterized, which exhibit differential sensitivity to the sulfonylurea herbicide primisulfuron (2-[3-(4,6-bis(di-fluoromethoxy) pyrimidin-2-yl)-ureidosulfonyl]-benzoic acid methylester). When treated postemergence with 160 g a.i. per hectare, inbred 4CO exhibited complete tolerance while inbred 4N5 was killed. The F1 hybrid 4C0 x 4N5 was uniformly tolerant indicating dominance of the tolerance trait. The field observations correlated with laboratory tests in which seedling root growth was measured. Based on IC50, inbred 4CO was more than ten times more tolerant than inbred 4N5. In the F2 and F3 generations, a 31 segregation of tolerant and sensitive individuals was observed, consistent with tolerance being inherited as a single dominant trait. Backcrosses of heterozygous F1 plants with the sensitive parent (4N5) yielded progeny that segreated at the expected 11 ratio. Backcrosses with 4C0 yielded tolerant offspring only. Inhibition characteristics of acetohydroxyacid synthase (AHAS; E.C. 4.1.3.18) were determined. The enzymes from both inbreds and their F1 hybrid were equally sensitive and strongly inhibited by primisulfuron (IC50: 7 nM). The fate of 14C-labeled primisulfuron in seedling tissues of inbred 4C0 and the hybrid, 4C0 x 4N5, indicated rapid metabolism with a half-life (t 1/2) of approximately 3 h. On the other hand, the herbicide-sensitive inbred 4N5 was considerably slower to metabolize primisulfuron (t 1/2 >24 h). These data indicate that differential metabolism is the mechanism of tolerance to the sulfonylurea herbicide primisulfuron in tolerant corn.Deceased  相似文献

Inheritance of dermatoglyphic configurations was studied on the palmar III interdigital pad of the rat (Rattus norvegicus). Four rats from each of the WKS and ACI/N inbred strains were mated with each other, and 54 F1 and 88 F2 hybrids were obtained. In addition, 52 offspring were produced by backcross mating between F1 hybrids and WKS and ACI/N parents. Whorls were the predominant pattern in the WKS strain and triradial patterns characterized the ACI/N strains. The F1 hybrids showed a wide range of pattern types, consisting of whorls, loops, cusps, arches, and triradial patterns. These patterns were intermediate in size between the two inbred strains. The F2 hybrids presented a distribution of patterns with a similar range as the F1, but the frequencies of some types were different. Patterns in the offspring of each backcross demonstrated a slight shift towards the characteristic pattern of the parental inbred strain. No sex difference was observed. Generally, the bilateral differences were striking, with a radial direction predominating on the left palm, and an ulnar one on the right palm, respectively. This study suggests that the dermal patterns are genetically determined, but also are influenced by environmental factors, especially in the hybrids.  相似文献

The Hawaiian strain (CB4856) of Caenorhabditis elegans is one of the most divergent from the canonical laboratory strain N2 and has been widely used in developmental, population, and evolutionary studies. To enhance the utility of the strain, we have generated a draft sequence of the CB4856 genome, exploiting a variety of resources and strategies. When compared against the N2 reference, the CB4856 genome has 327,050 single nucleotide variants (SNVs) and 79,529 insertion–deletion events that result in a total of 3.3 Mb of N2 sequence missing from CB4856 and 1.4 Mb of sequence present in CB4856 but not present in N2. As previously reported, the density of SNVs varies along the chromosomes, with the arms of chromosomes showing greater average variation than the centers. In addition, we find 61 regions totaling 2.8 Mb, distributed across all six chromosomes, which have a greatly elevated SNV density, ranging from 2 to 16% SNVs. A survey of other wild isolates show that the two alternative haplotypes for each region are widely distributed, suggesting they have been maintained by balancing selection over long evolutionary times. These divergent regions contain an abundance of genes from large rapidly evolving families encoding F-box, MATH, BATH, seven-transmembrane G-coupled receptors, and nuclear hormone receptors, suggesting that they provide selective advantages in natural environments. The draft sequence makes available a comprehensive catalog of sequence differences between the CB4856 and N2 strains that will facilitate the molecular dissection of their phenotypic differences. Our work also emphasizes the importance of going beyond simple alignment of reads to a reference genome when assessing differences between genomes.  相似文献

The human polymorphism in the hepatic enzyme N-acetyltransferase (NAT) affects the rate at which individuals acetylate, and in many cases detoxify, aromatic amine and hydrazine drugs and xenobiotics. Differences in NAT activity are known to affect individual susceptibility to drug toxicities and are thought to play a part in some spontaneous disorders. A mouse model for the human acetylation polymorphism has been previously characterized and involves the A/J (slow acetylator) and C57BL/6J (rapid acetylator) inbred strains. Strain distribution analysis of 40 A x B and B x A recombinant inbred (RI) strains indicated linkage between the N-acetyltransferase gene (Nat) and the esterase 1 (Es-1) gene, located on mouse chromosome 8. A double backcross involving 107 animals confirmed the recombination frequency between Nat and Es-1 to be 12 +/- 3% (mean +/- SE). The information obtained in the backcross and RI studies was combined, yielding a 13 +/- 2.8% (mean +/- SD) recombination frequency. The Es-1 genotype was determined in our newly developed congenic strains A.B6-Natr and B6.A-Nats. The B6.A-Nats strain has the Es-1 genotype of its inbred partner, the B6 strain, and the A.B6-Natr strain has the Es-1 genotype of the donor strain. These congenic strains will be important in determining the role of the NAT genotype in susceptibility to arylamine-induced cancer and other disorders.  相似文献

During the course of studies in inbred Strain 2 Guinea pigs on in vitro cell-mediated cytotoxicity to viral induced fibrosarcomas, it was observed that fetal calf serum in the medium in which the target cells were grown was responsible in some instances for much of the cytotoxicity observed. This fetal calf serum-specific cytotoxicity seemed mainly mediated by T cells and appeared to be independent on histocompatibility between target and killer cells.  相似文献

Seven genes and two anonymous markers were mapped to a single linkage group on rat chromosome 10 using progeny of an F2 intercross of Fischer (F344/N) and Lewis (LEW/N) inbred rats. Two genes, the neu oncogene or cellular homologue of the viral oncogene erbb2 (ERBB2) and growth hormone (GH) were mapped by Southern blot analysis of restriction fragment length polymorphisms. Five genes, embryonic skeletal myosin heavy chain (MYH3), androgen binding protein/sex hormone binding globulin (SHBG), asialoglycoprotein receptor (hepatic lectin)-1 (ASGR1), ATP citrate lysase (CLATP), and pancreatic polypeptide (PPY), and two anonymous markers, F16F2 and F10F1, were mapped using PCR amplification techniques. The PCR-typable polymorphic markers for the five genes were also highly polymorphic in 10 other inbred rat strains (SHR/N, WKY/N, MNR/N, MR/N, LOU/MN, BN/SsN, BUF/N, WBB1/N, WBB2/N, and ACI/N). These markers should be useful in genetic analysis of traits described in inbred rat strains, as well as in genetic monitoring of such strains. The loci in this linkage group covered 50 cM of rat chromosome 10 with the following order: MYH3, SHBG/ASGR1 (no recombinants detected), F16F2, ERBB2, CLATP, PPY, GH, and F10F1. Comparative gene mapping analysis indicated that this region of rat chromosome 10 exhibits linkage conservation with regions of human chromosome 17 and mouse chromosome 11.  相似文献

Two inbred strains of mice, C57BL/6J and DBA/2J, ranging in age from 2 to 38 months, were tested in an open field using the free exploration method. Scores were obtained for locomotor activity, exploratory behavior and emotionality. Strain differences were observed for all three variables. Beginning at late maturity (12 months), locomotor activity decreased with increasing age. Exploratory behavior was at a low level for DBA/2J mice at all ages. For C57BL/6J mice, exploratory behavior decreased significantly between 2 and 6 months and remained stable thereafter. Emotionality remained unchanged with advancing age for both strains of mice.  相似文献
