A previously presented model for an airlift fermentor is extended by considering the variations of pressure along the tubes. The fluid dynamics of the system is represented with the acid of correlations obtained from experiments. Only oxygen concentrations are considerations are considered. The influences of the various parameters affecting the system are analyzed. The model allows prediction of oxygen concentrations in the different points of an airlift fermentor and calculation of the best values of the gas flow rate.  相似文献

Summary The hydrodynamics and mass transfer behaviour of an airlift fermentor with an external loop (height 10m) has been investigated by measuring gas and liquid velocities, gas hold-up, liquid mixing and oxygen transfer coefficients. Liquid phase properties, i.e., ionic strength, viscosity and surface tension have been varied by altering the fermentation media. Results are compared with those from bubble column experiments performed in the same unit. Oxygen transfer to mycelial fermentation broths in an airlift fermentor   总被引:2,自引:0,他引:2  
Oxygen transfer rates and gas holdups were measured in mycelial fermentation broths of Chaetomium cellulolyticum and Neurospora sitophila, each cultured in a 1300-L pilot-plant-scale airlift fermentor. These cultures exhibited highly non-Newtonian flow behavior coupled with a substantial decrease in oxygen transfer rates. The volumetric mass transfer coefficients in these cultures were found to be 65-70% lower than those in water. The data were compared with the available correlations obtained for simulated fermentation broths. In general, the data for C. cellulolyticum are in satisfactory agreement with the correlations for the model media but the data for N. sitophila are higher than that predicted by the correlations. Model media based correlations are found to be applicable to the fermentation processes if the culture medium does not possess a high yield stress.  相似文献

The effects of aeration on the flow characteristics of water in a glass pilot-scale airlift fermentor have been examined. The 55-L capacity fermentor consisted of a 15.2-cm-i.d. riser column with a 5.1-cm-i.d. downcomer tube. It was found that the average bubble size diminished with increased aeration. Typically, average bubble sizes ranged from 4.32 mm at a superficial gas velocity of 0.64 cm/s to 1.92 mm at 10.3 cm/s. A gas holdup of 0.19 was attained with superficial gas velocities (vs) on the order of 10 cm/s, indicating the highly gassed nature of the fluid in the riser section of the fermentor. Circulation velocities of markers placed in the fermentor decreased with increasing aeration rates due to increased turbulence and axial liquid back mixing within the riser section. Actual volumetric liquid circulation rates remained relatively constant (0.36–0.49 L/s) for values of (vs) up to 10 cm/s. Based on theoretical calculations, the ascending velocity of bubbles in a swarm reached 54 cm/s in the range of (vs) values studied.  相似文献

This article deals with the modeling of the oxygen transfer in an industrial airlift fermentor used for lactic yeast production on whey substrates. The purpose of this study was to improve the understanding of the interactions among the various parameters that govern the oxygen transfer phenomena in this type of fermentor. The reliability of the proposed model is demonstrated. The results of the investigations have been put into practice on the industrial scale and have contributed to monitor better the fermentation process. The model was also used to develop new ways of industrial fermentor design.  相似文献

Results of pilot plant studies using a glass airlift fermentation device (55 liter fermentation volume) have proven the relative merits of such a system in the fermentation of a filamentous mold, Monascus purpureus, on 4% (w/w) starch media. The resultant overall yield of cell mass (Yx/s) of 0.38 was an appreciable increase over the 0.32 obtained with a pilot scale stirred tank fermentor previously studied. Power requirements of the airlift fermentor were approximately 50% of those for the mechanically agitated system. The lack of mechanical shear in the airlift system provides a more gentle environment or the cultivation of organisms than does the high degree of shear prevalent in the mechanically agitated vessels. Mass transfer of oxygen to the aqueous phase of the fermentation volume is improved significantly through use of the airlift device. Mass transfer coefficients in the range of 200 reciprocal hr were obtained to approximately 80 reciprocal hr in the stirred tank fermentor.  相似文献

To grow yeast rapidly, it is necessary to supply sufficient oxygen to the yeast and to effectively remove the heat of the fermentation. We succeeded in developing a commercial-scale fermentor for growing a food yeast (Candida utilis) to produce RNA rapidly. This fermentor is an internal-loop airlift type with vertical heat transfer tubes between inner and the outer columns. The volume of the fermentor is 145 m3 (working volume 75 m3). The oxygen transfer rate (OTR) was 9.9 kg-O2/m3/h using a superficial gas velocity of 30 cm/s based on the outer column. Much of the heat of fermentation and the energy resulting from aeration could be removed effectively by the heat transfer tubes. This unique airlift fermentor was driven at a dilution rate of 0.43 h−1 for about 70 d, with the yeast concentration being maintained at 22.8 kg-dry cell/m3. The yeast production rate was 9.79 kg-dry cell/m3/h. Compared with a traditional stirred-type fermentor, two Vogelbush-type fermentors and another airlift fermentor, our fermentor was far superior with respect to OTR and yeast productivity.  相似文献

Despite the increasing importance of airlift fermentors, very little published information is available on how the geometric configurations of the draft tubes and the air-sparging system affect the mixing and oxygen transfer characteristics of the fermentor. A 14-L air-lift fermentor was designed and build with a fixed liquid height to diameter ratio of 1.5 utilizing four equally spaced air jets at the bottom. Two jet orifice sizes were used, 1.27 and 3.81 mm i.d., and for each jet size the following four geometric configurations were used: Single inner concentric draft tube, single outer concentric draft tube, two concentric draft tubes, and no draft tubes where the fermentor was operated as a shallow bubble column. It was found that the presence of draft tubes stabilized liquid circulation patterns and gave systemically higher mixing times than those obtained in the absence of draft tubes. In addition, the double draft tube geometry resulted in higher mixing times than the single draft tubes. For the power unit volume range 20 to about 250 W/m3 the larger 3.81-mm orifices gave systemically higher kL a values than the smaller 1.27-mm i.d. orifices. At 200 W/m3 the use of a single outer draft tube with the 3.81-mm orifices resulted in 94% increase in kL a values over that obtained with no draft tubes. However, the effect of draft tube geometry on kL a values when the 1.27-mm orifices were used was not significant. The air bubble formation characteristics at the jet orifices were found to be different, which reflected the differences observed in mass transfer and mixing characteristics. The power economy for oxygen transfer was found to be depend strongly on the orifice size and less on the geometric configuration of draft tubes.  相似文献

A laboratory model constant-depth film fermentor was developed. Film grew on the surface of polytetrafluoroethylene (PTFE) plugs and was limited to a predetermined depth by mechanically removing excess film. Six film-forming organisms were isolated from river water and used to assess the operating characteristics of the fermentor. Film accumulation was logarithmic, and a steady state was maintained. Electron micrographs show early film development. The fermentor enables film to be grown on any substratum and allows discrete, reproducible, and representative samples to be taken.  相似文献

A mathematical model is presented for a microporous hollow-fiber membrane extractive fermentor (HFEF). The model is based on the continuous flow of the aqueous nutrient phase and cells through the shell space of the fermentor where the fermentation reaction occurs. The product diffuses from the shell space through the hollow-fiber membrane where it is continuously removed by solvent flowing concurrently through the fiber lumen. Results for ethanol production show that the HFEF has a volumetric productivity significantly higher than that possible using conventional methods. The model predicts the existence of an optimum volume fraction of hollow fibers in the fermentor that maximizes the total volumetric productivity. This optimum is the result of a classic trade-off between the volume fraction of the fermentor required for fermentation and that required for efficient removal of the ethanol product to minimize product inhibition.  相似文献

Approaches to increasing the productivity of benzophenanthridine alkaloids in suspension cultures in Escherichia californica were made in an airlift fermentor under different culture conditions. Elicitation with yeast extract elicitor reduced the time required to obtain a certain amount of alkaloid production. In a two-phase airlift fermentor with compounded silicone fluid, total alkaloid concentration in silicone fluid was 153.1 mg/L and that in the aqueous cellular phase was 8.2 mg/L at day 21 from inoculation. The large accumulation capacity of silicone fluid made it possible to store correspondingly large amounts of total alkaloid and increased the alkaloid production. Act day 21 from inoculation, the volumetric alkaloid productivity and the netproduction in a two-phase airlift fermentor were 1.4 and 1.5 times higher than those of normal airlift fermentor operation. This performance was furthermore enhanced by elicitation. Elicitation in two-phase airlift fermentor operation increased the volumetric productivity and the new production 3.3- and 3.5-fold compared to those of normal airlift fermentor operation. This paper uses a multikinetic approach to predict gluconic acid (GA) production performance in a 4.5 L airlift bioreactor (ALBR). The mathematical model consists of a set of simultaneous firstorder ordinary differential equations obtained from material balances of cell biomass, GA, glucose, and dissolved oxygen. Multikinetic models, namely, logistic and contois equations constitute kinetic part of the main model. The main model also takes into account the hydrodynamic and mass transfer parameters. These equations were solved using ODE solver of MATLAB v6.5 software. The mathematical model was validated with the experimental data available in the literature and is used to predict the effect of change in initial biomass and air sparging rate on the GA production. It is concluded that the mathematical model incorporated with multikinetic approach would be more efficient to predict the change in operating parameters on overall bioprocess of GA production in an ALBR.  相似文献

An electronic liquid level control system was developed to maintain constant working volume in a continuously fed and aerated fermentor from which spent medium was continously withdrawn by microfiltration in an external recycle loop. The level control system was comprised of an admittance sensor in the fermentor and an external transmitter and controller, which together regulated the speed of a recycle pump and thus the rate of liquid withdrawal from the fermentor. During test bacterial culture the liquid level was maintained usually within +/- 1% and always within +/- 5%. The control system could be applied to various other types and scales of continuous-flow fermentors.  相似文献

As a function of the gas throughput the following parameters were measured in an external loop reactor with a riser diameter of 0.6 m and a gassed liquid height of 8.6 m: integral and local values of gas hold-up; liquid velocities; mixing times and axial dispersion coefficients of the liquid phase. The height of the reactor could be altered by reconstruction. Measurements were also carried out with lower heights than 8.6 m. Besides pure water, aqueous solutions of coalescing, non-coalescing and viscosity-increasing substances were used as model systems. With the results a general relationship between superficial gas velocity, gas hold-up and liquid velocity was established. This hydrodynamic model uses the relative velocity between gas and liquid phase as the fundamental parameter. The generally valid model consists of one term for the homogeneous and of two additional terms for the heterogeneous flow regime.  相似文献

We will be concerned with optimal intervention policies for a continuous-time stochastic SIR (susceptible-->infective-->removed) model for the spread of infection through a closed population. In previous work on such optimal policies, it is common to assume that model parameter values are known; in reality, uncertainty over parameter values exists. We shall consider the effect upon the optimal policy of changes in parameter estimates, and of explicitly taking into account parameter uncertainty via a Bayesian decision-theoretic framework. We consider policies allowing for (i) the isolation of any number of infectives, or (ii) the immunisation of all susceptibles (total immunisation). Numerical examples are given to illustrate our results.  相似文献

In this work we address the problem of the robust identification of unknown parameters of a cell population dynamics model from experimental data on the kinetics of cells labelled with a fluorescence marker defining the division age of the cell. The model is formulated by a first order hyperbolic PDE for the distribution of cells with respect to the structure variable x (or z) being the intensity level (or the log10-transformed intensity level) of the marker. The parameters of the model are the rate functions of cell division, death, label decay and the label dilution factor. We develop a computational approach to the identification of the model parameters with a particular focus on the cell birth rate α(z) as a function of the marker intensity, assuming the other model parameters are scalars to be estimated. To solve the inverse problem numerically, we parameterize α(z) and apply a maximum likelihood approach. The parametrization is based on cubic Hermite splines defined on a coarse mesh with either equally spaced a priori fixed nodes or nodes to be determined in the parameter estimation procedure. Ill-posedness of the inverse problem is indicated by multiple minima. To treat the ill-posed problem, we apply Tikhonov regularization with the regularization parameter determined by the discrepancy principle. We show that the solution of the regularized parameter estimation problem is consistent with the data set with an accuracy within the noise level in the measurements.   相似文献
