Contingency analysis of r × c-tables is systematized as follows: Global contingency testing based on the total chisquare under H0 of independent row and column variables is least favorable to exhaustive interpretation. Local contingency testing based on chi-square components of the individual cells of an r × c-table and realized in Configural frequency analysis (KRAUTH and LIENERT, 1973) is more favourable to substantive interpretation. Compromising between global and local contingency testing leads to so-called generalized local and regional contingency testing. Every contingency testing (local, regional and naturally global) is including all N individuals of a sample supposed to have been drawn randomly from a defined population. Extension to 3-and t-dimensional contingency analysis is outlined, and biomedical applications are discussed.  相似文献

A medicinal mushroom, Phellinus linteus, was successfully cultivated using a cheese-processing waste, whey, and the optimal bioconversion conditions for the maximum mycelial growth rate was also estimated through solid-state cultivation experiments. Response surface analysis with a face-centered design (center point replication = 5) was applied to statistically approximate the simultaneous effects of the three variables, i.e., substrate concentration (10–30 g lactose l−1), temperature (20–30°C), and pH (4–6), on the mycelial growth rate of P. linteus. The following is a partial cubic model where η is the mycelial growth rate (K r ) and x k is the corresponding variable term (k = substrate concentration, temperature, and pH in order): η = −23.8 + 8.67 × 10−2 x 1 + 1.48x 2 + 1.77x 3 + 8.00 × 10−4 x 1 x 2 + 7.25 × 10−2 x 1 x 3 + 5.13 × 10−2 x 2 x 3 −1.28 × 10−2 x 12 –3.18 × 10−2 x 22. −2.64 × 10−1 x 32 −3.28 × 10−3 x 1 x 2 x 3 + 4.68 × 10−4 x 12 x 2. The produced response surface model proved to be significant (r 2 > 0.99, P-value <0.0001, coefficient of variation <5%) to describe the explored space. Temperature was found to be the most significant factor of dominant effects on the mycelial growth rate, and other variables such as temperature2, pH, pH2, and (substrate concentration2 × temperature) also showed significant effects on the model output. The maximum mycelial growth rate was predicted to be 2.80 mm d−1 at 29.7 g lactose l−1, 26.2°C, and pH 5. Our results proved a good potential of whey to serve as an alternative growth medium for cultivating P. linteus mycelia. This may provide another potential for managing this nutrient-rich waste in a cost-effective way.  相似文献

Atmospheric CO2 (ca) rise changes the physiology and possibly growth of tropical trees, but these effects are likely modified by climate. Such ca × climate interactions importantly drive CO2 fertilization effects of tropical forests predicted by global vegetation models, but have not been tested empirically. Here we use tree‐ring analyses to quantify how ca rise has shifted the sensitivity of tree stem growth to annual fluctuations in rainfall and temperature. We hypothesized that ca rise reduces drought sensitivity and increases temperature sensitivity of growth, by reducing transpiration and increasing leaf temperature. These responses were expected for cooler sites. At warmer sites, ca rise may cause leaf temperatures to frequently exceed the optimum for photosynthesis, and thus induce increased drought sensitivity and stronger negative effects of temperature. We tested these hypotheses using measurements of 5,318 annual rings from 129 trees of the widely distributed (sub‐)tropical tree species, Toona ciliata. We studied growth responses during 1950–2014, a period during which ca rose by 28%. Tree‐ring data were obtained from two cooler (mean annual temperature: 20.5–20.7°C) and two warmer (23.5–24.8°C) sites. We tested ca × climate interactions, using mixed‐effect models of ring‐width measurements. Our statistical models revealed several significant and robust ca × climate interactions. At cooler sites (and seasons), ca × climate interactions showed good agreement with hypothesized growth responses of reduced drought sensitivity and increased temperature sensitivity. At warmer sites, drought sensitivity increased with increasing ca, as predicted, and hot years caused stronger growth reduction at high ca. Overall, ca rise has significantly modified sensitivity of Toona stem growth to climatic variation, but these changes depended on mean climate. Our study suggests that effects of ca rise on tropical tree growth may be more complex and less stimulatory than commonly assumed and require a better representation in global vegetation models.  相似文献

Summary A new control policy for the on-line optimization of the nutrient supply in bakers yeast process is proposed. A feed rate corresponding to minimal substrate uptake time was shown to be optimal for cell yield and specific growth rate.  In this paper we study the existence of one-dimensional travelling wave solutions u(x, t)=φ(xct) for the non-linear degenerate (at u=0) reaction-diffusion equation u t =[D(u)u x ] x +g(u) where g is a generalisation of the Nagumo equation arising in nerve conduction theory, as well as describing the Allee effect. We use a dynamical systems approach to prove: 1. the global bifurcation of a heteroclinic cycle (two monotone stationary front solutions), for c=0, 2. The existence of a unique value c *>0 of c for which φ(xc * t) is a travelling wave solution of sharp type and 3. A continuum of monotone and oscillatory fronts for cc *. We present some numerical simulations of the phase portrait in travelling wave coordinates and on the full partial differential equation. Aims: A microbiological bioassay using Geoacillus stearothermophilus was optimized to detect betalactams at concentrations near to the Maximum Residue Limits (MRLs), with low cross‐specificity for tetracycline. Methods and Results: A factorial design (3 × 4) was used to evaluate the effects of concentration of spores (2·0 × 106, 4·0 × 106 and 8·0 × 106 spores ml?1) and incubation time (3·0, 3·5, 4·0 and 4·5 h) on the response of the bioassay. Then, desirability function to raise the detection capabilities (CCβ) of tetracyclines and increase sensitivity to betalactams was implemented. Significant effects of Log[S] and incubation time [It] on the CCβ of betalactams and tetracyclines were observed. Finally, high value of global desirability (D = 0·853), adequate betalactams CCβ (3·8 μg l?1 of penicillin ‘G’, 27 μg l?1 of oxacillin, 8·1 μg l?1 of ampicillin, 48 μg l?1 of cloxacillin) and high tetracyclines CCβ (5260 μg l?1 chlortetracycline, 1550 μg l?1 of oxytetracycline, 1070 μg l?1 of tetracycline) were calculated. Conclusions: The application of chemometric tools allows the optimization of a bioassay that detects betalactam residues in milk. The more robust conditions have been achieved in Log[S] = 6·30 and [It] = 4·20 h. Significance and Impact of the Study: The logistic regression model and the desirability function are adequate chemometric techniques to improve the properties of the methods, because it is possible to increase sensitivity and decrease cross‐specificity simultaneously.  相似文献

There are conflicting opinions about the need to fertilize Miscanthus and, also, the question has been raised whether Miscanthus should be irrigated, especially if water resources are limited. Crop growth modeling can help answer such questions. In this article the FAO AquaCrop water‐driven model was selected to simulate Miscanthus biomass under different nutrient and water supply conditions. The article reports the outcomes of 6‐year experiments with Miscanthus on two locations in Serbia: Zemun, where three fertilizer treatments were applied (Nl – 100 kg ha?1, Nopt 50 kg ha?1 and Nf nonfertilized), and Ralja, where only Nl 100 kg ha?1 was applied. Model calibration focused on the measured data (root depth, crop phenology, and the above‐ground biomass by year of growth. Calibration results showed a very good match between measured and simulated values. The largest and only significant difference was noted in 2008, when the crop was establishing and exhibited uneven radication. The simulation results for the next 5 years showed a variance from ?4 to 5.7%, believed to be a very good match. A high coefficient of determination (R2 = 0.995) and high Willmott index of agreement (0.998) were also indicative of a good match between simulated and recorded biomass yields. The measured and simulated results for validated datasets at both locations were good. The average RMSE was 2.89 Mg ha?1; when compared to the deviations noted at the test site itself, it was apparent that they were smaller in all the years of research except the first year. The index of agreement was 0.97 and the coefficient of determination R2 0.947. The AquaCrop model can be used with a high degree of reliability in strategic planning of Miscanthus cultivation in new areas, under different nutrient and water supply and local weather and soil conditions.  相似文献

Miscanthus × giganteus is often regarded as one of the most promising crops to produce sustainable bioenergy. This perennial crop, renowned for its high productivity associated with low input requirements, in particular regarding fertilizers, is thought to have low environmental impacts, but few data are available to confirm this. Our study aimed at assessing nitrate leaching from Miscanthus × giganteus crops in farmers' fields, thus including a wide range of soil and cropping system conditions. We focused on the first years of growth after planting as experimental studies have suggested that Miscanthus × giganteus, once established, results in low nitrate leaching. We combined on‐farm measurements and modeling to estimate drainage, leached nitrogen, and nitrate concentration in drainage water in 38 fields located in Center‐East France during two winters (November 2010 to March 2011, November 2011 to March 2012). Nitrate leaching and nitrate concentration in drainage water were on average very low. Nitrate leaching averaged 6 kg N ha?1 whereas nitrate concentration averaged 12 mg l?1. These low values are attributable to the low estimates of drainage water (mean = 166 mm) but also to the low soil mineral nitrogen contents measured at the beginning of winter (mean = 37 kg N ha?1). Our results were, however, very variable, mainly due to the crop age: nitrate leaching and nitrate concentration were critically higher during the winter following the first growth year of Miscanthus × giganteus, reflecting the low development of the crop. This variability was also explained by the range of soil and cropping conditions explored in the on‐farm design: shallow and/or sandy soils as well as fields where establishment failed had a higher risk of nitrate leaching.  相似文献

Miscanthus × giganteus is an energy crop with many attributes that make it a potential biofuel feedstock. This study examined the chemical composition of M. × giganteus stems cut at different dates throughout the spring harvest window (January, February and March) and either left in a swath or left flat in a thin layer on the ground and compared the composition to that of the standing crop collected on the same date in April (control). The research then examined the effect of cutting date on the chemical composition of whole plant M. × giganteus biomass (leaf and stem). The parameters examined in both parts of this experiment were lower heating value on a wet basis (LHVWB), ash, chlorine, potassium, nitrogen, sulphur, carbon and hydrogen content. The range of values recorded for the parameters from both aspects of this trial were LHVWB 4.84–11.87 MJ kg?1; ash 1.44–1.97%; Cl 0.07–0.23%; K 0.15–0.32%; N 0.28–0.39%; S 0.13–0.19%; C 46.75–50.00%; H 5.76–6.09%. The length of time that the M. × giganteus remained in the field after cutting affected the LHVWB (increased with time) of the stem biomass material. Cutting the biomass and leaving it in the field lowered the ash, Cl and C content of the stem material compared to that of the control which was cut and collected on the same date. No differences were observed for the other parameters. Date of harvest affected the LHVWB, Cl and C content which all improved with later cutting dates. Thus, combustion quality can be improved by delaying the harvest date or by cutting the crop and leaving it in the field for a period prior to collection. Choosing the correct combination of time and harvest method can therefore improve biomass fuel quality.  相似文献

The estimator ?0(x) of the regression r(x) = E (Y | × = x) from measured points (xi, yi), i = 1(1) n, of a continuous two-dimensional random variable (X, Y) with unknown continuous density function f(x, y) and with moments up to the second order can be made with the help of a density estimation f?0(x, y) (see e.g. SCHMERLING and PEIL, 1980). Here f?0(x, y) still contains free parameters (so-called band-width-parameters), the values of which have to be optimally fixed in the concrete case. This fixing can be done by using a modification of the maximum-likelihood principle including jackknife techniques. The parameter values can be also found from the estimators for r(x). Here the cross-validation principle can be applied. Some numerical aspects of these possibilities for optimally fixing the bandwidth-parameter are discussed by means of examples. If ?0(x) is used as a smoothing operator for time series the optimal choice of the parameter values is dependent on the purpose of application of the smoothed time series. The fixing will then be done by considering the so-called filter-characteristic of ?C0(x).  相似文献

Efficient plant regeneration is essential for successful transformation and in vitro polyploidy induction in mulberry. A high frequency (80%) of plant regeneration from hypocotyls occurred under in vitro conditions in mulberry (Morus multicaulis Poir.). We identified three key factors for enhancing successful regeneration based on earlier work: (1) hypocotyl position, (2) the combination and concentration of growth regulators, and (3) the addition of AgNO3. The highest frequency of shoot regeneration was achieved using hypocotyl segments, which are proximal to apical meristems, and the optimal culture conditions were Murashige and Skoog’s (MS) (Murashige and Skoog, 1962) basal medium supplemented with 3.0 mg l−1 6-benzylamino purine, 0.3 mg l−1 indole-3-acetic acid, 0.1% polyvinypyrrolidone, and 1.0 mg/l silver nitrate (AgNO3) under subdued light at 25 ± 2°C. Treating the shoots with 0.2% colchicine (dipping for 72 h) resulted in a 14% tetraploid frequency, whereas a 20% tetraploid frequency resulted from using a 0.25% colchicine (dripping for 5 d) treatment, as determined by chromosome number counts. The diploid plant chromosome number was 28 (2n = 2x = 28) and that of tetraploid plants was 56 (2n = 4x = 56). Regenerated shoots rooted easily in 8–10 d using half-strength basal MS medium with 0.5 mg l−1 indole-3-butyric acid and were successfully established in the soil.  相似文献

A novel flow injection chemiluminescence (CL) method for the determination of rutin was reported. The proposed method was based on the enhanced effect of rutin on the chemiluminescence intensity of luminol and potassium hexacyanoferrate(III) reaction in NaOH medium. The variables of reaction system, such as luminol concentration, potassium hexacyanoferrate(III) concentration and NaOH concentration, were optimized with the aid of response surface methodology. For the responses prediction, a second‐order polynomial model (SOPM) was applied. The optimal conditions for determination of rutin estimated by the model equation were as follows: NaOH concentration of 0.13 mol/L luminol concentration of 0.94 × 10?6 mol/L, and K3Fe(CN)6 concentration of 1.09 × 10?4 mol/L. The theoretical increased ratio of CL intensity (IRI) predicted and actual IRI for 0.05 mg/L rutin under the above conditions were 99.40 and 99.74%, respectively. The SOPM model proved to be powerful for navigating the design space. Under the above optimum conditions, the increased IRI was linearly related to the concentration of rutin in the range from 0.008 to 0.100 mg/L with the regression equation IRI = 1948.20c + 5.24 (r = 0.9994) and in the range from 0.100 to 1.000 mg/L with the regression equation IRI = 1362.50 c + 61.94 (r = 0.9996). The detection limit (3σ) was of 1.95 × 10?3 mg/L. The sampling frequency of this method was 72/h. The method was used directly to determine rutin in tablets. By combining gel permeation chromatography (GPC) and light-scattering spectroscopy, including photon correlation and angular distribution of absolute scattered intensity, we were able to characterize immunologically active Haemophilus influenzae type b polysaccharide (HIB Ps) bovine serum albumin (BSA) conjugates in terms of equivalent hydrodynamic radius rh ~ (6.2 ± 0.6) × 102 Å, apparent radius of gyration rg ~ (5.4 ± 0.3) × 102 Å, apparent molecular weight Mw ~ (3.5 ± 0.4) × 106 g/mol, and a second virial coefficient A2 ~ (1.9 ± 0.3) × 10?4 cm3 mol/g2. We could study the effects of each of the processes in the conjugate formation according to the following procedure: BSA (dialysis, modification, fractionation) + HIB Ps → HIB Ps/BSA conjugate (conjugate formation, fractionation). Narrow distributions of HIB Ps BSA conjugate formation can be achieved using fractionated BSA.  相似文献

We studied the relationship between changes in intracellular pH (pH i ), intracellular Ca2+([Ca2+] i ) and charybdotoxin sensitive (CTX) maxi-K+ channels occurring after modest `physiological' swelling in guinea pig jejunal villus enterocytes. Villus cell volume was assessed by electronic cell sizing, and pH i and [Ca2+] i by fluorescence spectroscopy with 2,7, biscarboxyethyl-5-6-carboxyfluorescein and Indo-1, respectively. In a slightly (0.93 × isotonic) hypotonic medium, villus cells swelled to the same size they would reach during d-glucose or l-alanine absorption; the subsequent Regulatory Volume Decrease (RVD) was prevented by CTX. After the large volume increase in a more hypotonic (0.80 × isotonic) medium, RVD was unaffected by CTX. After modest swelling associated with 0.93 × isotonic dilution, the pH i alkalinized but N-5-methyl-isobutyl amiloride (MIA) prevented this ΔpH i and the subsequent RVD. Even in the presence of MIA, alkalinization with added NH4Cl permitted complete RVD which could be inhibited by CTX. The rate of 86Rb efflux which also increased after this 0.93 × isotonic dilution was inhibited an equivalent amount by CTX, MIA or Na+-free medium. Modest swelling transiently increased [Ca2+] i and Ca2+-free medium or blocking alkalinization by MIA or Na+-free medium diminished this transient increase an equivalent amount. RVD after modest swelling was prevented in Ca2+-free medium but alkalinization still occurred. After large volume increases, alkalinization of cells increased [Ca2+] i and volume changes became sensitive to CTX. We conclude that both alkalinization of pH i and increased [Ca2+] i observed with `physiological' volume increase are essential for the activation of CTX-sensitive maxi-K+ channels required for RVD. Abstract: In the postmortem human brain (27 specimens of frontal cortex, Brodmann area 9), the specific binding of the antagonists [3H]RX 821002 (2-methoxyidazoxan) to α2A-adrenoceptors and that of [3H]idazoxan to l2-imidazo-line sites (a nonadrenoceptor mitochondrial site) were determined in parallel to study the effect of aging (range, 4–89 years) on both brain proteins. The density of α2A-adrenoceptors and age were negatively correlated (r=-0.71; p < 0.001). In contrast, the density of l2-imidazo-line sites was positively correlated with aging (r= 0.59; p < 0.005). The ratio of receptor densities (α2A/l2) also showed a marked negative correlation with age (r=-0.76; p < 0.001). In an age-selected group (range, 10–89 years), the density of monoamine oxidase (MAO)-B sites labeled by [3H]Ro 19–6327 (lazabemide) also showed a positive correlation with age (r= 0.80; p < 0.005). In these subjects, the density of l2-imidazoline sites correlated well with the density of MAO-B sites (r= 0.70; p < 0.005). The ratio of the density of these sites (MAO-B/l2) did not correlate with the age of the subject at death (r=-0.15). In the human frontal cortex, idazoxan displayed very low affinity (Ki= 89 μM) against the binding of [3H]Ro 19–6327 to MAO-B, which discounted a direct interaction of [3H]idazoxan with the active center of the enzyme and indicated that the l2-imidazoline site cannot be identified with MAO-B. However, l2-imidazoline sites and MAO-B show a clear coexpression not only in the human frontal cortex during the process of aging, but also in various brain regions of the human and rat brains. It is suggested that the l2-imidazoline site has a specific location on glial (astrocyte) cells.  相似文献

A step‐by‐step strategy is reported for improving capacitance of supercapacitor electrodes by synthesizing nitrogen‐doped 2D Ti2CTx induced by polymeric carbon nitride (p‐C3N4), which simultaneously acts as a nitrogen source and intercalant. The NH2CN (cyanamide) can form p‐C3N4 on the surface of Ti2CTx nanosheets by a condensation reaction at 500–700 °C. The p‐C3N4 and Ti2CTx complexes are then heat‐treated to obtain nitrogen‐doped Ti2CTx nanosheets. The triazine‐based p‐C3N4 decomposes above 700 °C; thus, the nitrogen species can be surely doped into the internal carbon layer and/or defect site of Ti2CTx nanosheets at 900 °C. The extended interlayer distance and c‐lattice parameters (c‐LPs of 28.66 Å) of Ti2CTx prove that the p‐C3N4 grown between layers delaminate the nanosheets of Ti2CTx during the doping process. Moreover, 15.48% nitrogen doping in Ti2CTx improves the electrochemical performance and energy storage ability. Due to the synergetic effect of delaminated structures and heteroatom compositions, N‐doped Ti2CTx shows excellent characteristics as an electrochemical capacitor electrode, such as perfectly rectangular cyclic voltammetry results (CVs, R2 = 0.9999), high capacitance (327 F g?1 at 1 A g?1, increased by ≈140% over pristine‐Ti2CTx), and stable long cyclic performance (96.2% capacitance retention after 5000 cycles) at high current density (5 A g?1).  相似文献

The effect of l-arginine on transepithelial ion transport was examined in cultured M-1 mouse renal cortical collecting duct (CCD) cells using continuous short circuit current (I SC ) measurements in HCO3 /CO2 buffered solution. Steady state I SC averaged 73.8 ± 3.2 μA/cm2 (n= 126) and was reduced by 94 ± 0.6% (n= 16) by the apical addition of 100 μm amiloride. This confirms that the predominant electrogenic ion transport in M-1 cells is Na+ absorption via the epithelial sodium channel (ENaC). Experiments using the cationic amino acid l-lysine (radiolabeled) as a stable arginine analogue show that the combined activity of an apical system y+ and a basal amino acid transport system y+L are responsible for most cationic amino acid transport across M-1 cells. Together they generate net absorptive cationic amino acid flux. Application of l-arginine (10 mm) either apically or basolaterally induced a transient peak increase in I SC averaging 36.6 ± 5.4 μA/cm2 (n= 19) and 32.0 ± 7.2 μA/cm2 (n= 8), respectively. The response was preserved in the absence of bath Cl (n= 4), but was abolished either in the absence of apical Na+ (n= 4) or by apical addition of 100 μm amiloride (n= 6). l-lysine, which cannot serve as a precursor of NO, caused a response similar to that of l-arginine (n= 4); neither L-NMMA (100 μm; n= 3) nor L-NAME (1 mm; n= 4) (both NO-synthase inhibitors) affected the I SC response to l-arginine. The effects of arginine or lysine were replicated by alkalinization that mimicked the transient alkalinization of the bath solution upon addition of these amino acids. We conclude that in M-1 cells l-arginine stimulates Na+ absorption via a pH-dependent, but NO-independent mechanism. The observed net cationic amino acid absorption will counteract passive cationic amino acid leak into the CCD in the presence of electrogenic Na+ transport, consistent with reports of stimulated expression of Na+ and cationic amino acid transporters by aldosterone. S-Adenosyl-l-methionine (AdoMet) was produced by a mutant strain Kluyveromyces lactis AM-65 grown on whey. A full factorial design method of three factors – (NH4)2SO4 (factor x 1), corn steep liquor (factor x 2) and l-methionine (factor x 3) on three levels – was used to determine the optimal medium conditions for the production of AdoMet. A time course shake-flask experiment in optimal whey medium (x 1=3.1 g l–1, x 2=12.7 g l–1, x 3=4.6 g l–1) was also carried out and the results confirmed the results of the factorial design and subsequent quadratic modelling and optimization of AdoMet production which reached 90 mg g–1 cell dry wt.  相似文献

L-lysine Transport in Chicken Jejunal Brush Border Membrane Vesicles   总被引:2,自引:0,他引:2  
The properties of l-lysine transport in chicken jejunum have been studied in brush border membrane vesicles isolated from 6-wk-old birds. l-lysine uptake was found to occur within an osmotically active space with significant binding to the membrane. The vesicles can accumulate l-lysine against a concentration gradient, by a membrane potential-sensitive mechanism. The kinetics of l-lysine transport were described by two saturable processes: first, a high affinity-transport system (K mA= 2.4 ± 0.7 μmol/L) which recognizes cationic and also neutral amino acids with similar affinity in the presence or absence of Na+ (l-methionine inhibition constant KiA, NaSCN = 21.0 ± 8.7 μmol/L and KSCN = 55.0 ± 8.4 μmol/L); second, a low-affinity transport mechanism (KmB= 164.0 ± 13.0 μmol/L) which also recognizes neutral amino acids. This latter system shows a higher affinity in the presence of Na+ (KiB for l-methionine, NaSCN = 1.7 ± 0.3 and KSCN = 3.4 ± 0.9 mmol/L). l-lysine influx was significantly reduced with N-ethylmaleimide (0.5 mmol/L) treatment. Accelerative exchange of extravesicular labeled l-lysine was demonstrated in vesicles preloaded with 1 mmol/L l-lysine, l-arginine or l-methionine. Results support the view that l-lysine is transported in the chicken jejunum by two transport systems, A and B, with properties similar to those described for systems b 0,+ and y+, respectively.
