Vertebrate species display consistent left-right asymmetry in the arrangement of their internal organs. This asymmetry reflects the establishment of the left-right axis and the alignment of the organs along this axis during development. Members of the TGF-β family of molecules have been implicated in both the establishment and signaling of left-right axis information. Asymmetric expression of one member, nodal (called Xnr-1 in the frog, Xenopus laevis), is highly conserved among species. The nodal-related genes are normally expressed in the left lateral plate mesoderm prior to the development of morphologic asymmetry. Expression patterns of nodal have been correlated with heart situs in mouse, chick, and frog and our previous work has implicated the dorsal midline structures in the regulation of nodal expression and cardiac laterality. In this study, three approaches were used to address the embryologic and molecular basis of asymmetric Xnr-1 expression. First, notochord and lateral plate recombinants were performed and showed that notochord can repress Xnr-1 expression in lateral plate mesoderm explants derived from either the left or the right side. Second, lateral plate mesoderm grafts indicated that Xnr-1 expression is specified but not determined at neurula stages and can subsequently be repatterned. These experiments suggest that a repressive signal from the notochord is required for maintenance of asymmetric Xnr-1 expression and that Xnr-1 expression is regulated by signals outside of the lateral plate mesoderm. Third, candidate molecules were injected to test for their ability to alter Xnr-1 expression pattern in the lateral plate. Late injection of activin protein on the right side of the embryo induced ectopic Xnr-1 expression and randomized cardiac orientation. Restriction-endonuclease analyses of mitochondrial DNAs from all six color-pattern classes (A–F) of the parthenogenetic lizard Cnemidophorus tesselatus yield estimates of nucleotide divergence that are extremely low (π = 0.06%). In digests of 75 C. tesselatus mtDNAs with 20 different restriction enzymes, only four cleavage-site differences were noted, three of which were found only in pattern class F. The near-identity of these mitochondrial DNAs with those from C. tigris marmoratus shows unequivocally that C. t. marmoratus was the species to which the maternal parent(s) of all C. tesselatus belonged. Mitochondrial-DNA analyses of another unisexual species, C. neomexicanus, led to the same conclusion. Mitochondrial DNAs from 96 individuals of these three species were extensively analyzed for cleavage-site differences; only 13 were found. The low interspecific sequence diversity found within C. neomexicanus and the C. tesselatus complex suggests a recent origin for both. Based on diversity data for mitochondrial DNA and allozymes, we estimate that a minimum of two hybridizations were required to produce all diploid C. tesselatus (C–F), followed by at least two more to generate the triploids (A and B). These data and those presented in the two accompanying papers indicate that events leading to parthenogenesis in Cnemidophorus are rare and strengthen the hypothesis that interspecific hybridization is a necessary, causal event in its establishment.  相似文献

The Fused toes (Ft) mouse mutation was created by insertional mutagenesis, resulting in the deletion of several hundred kb of genomic sequences of mouse Chromosome (Chr) 8. Mice heterozygous for the Ft mutation are characterized by partial syndactyly of forelimbs and massive thymic hyperplasia indicating that programmed cell death is affected. Homozygous Ft/Ft embryos die at midgestation and show severe malformations of craniofacial structures. Furthermore, establishment of left-right asymmetry is random. Here we report on the positional cloning of a novel gene by exon trap analysis of a genomic clone encoding wild-type sequences corresponding to parts of the deletion in Ft mutants. RT-PCR experiments demonstrated that the newly identified gene, Fatso (Fto), is expressed throughout embryonic development. Wide expression was also found in tissues of adult mice. We show that expression of Fto is completely absent in mouse embryonic fibroblasts homozygous for the Ft mutation. In addition, we isolated the full-length cDNA which encodes a putative 58-kDa protein showing no similarities to known proteins or protein motifs. The expression data of Fto define it as a candidate gene involved in processes such as programmed cell death, craniofacial development, and establishment of left-right asymmetry. The underlying Boltzmann characteristics of motility-related gating currents of the outer hair cell (OHC) are predicted to generate distortion components in response to sinusoidal transmembrane voltages. We studied this distortion since it reflects the mechanical activity of the cell that may contribute to peripheral auditory system distortion. Distortion components in the OHC electrical response were analyzed using the whole-cell voltage clamp technique, under conditions where ionic conductances were blocked. Single or double-sinusoidal transmembrane voltage stimulation was delivered at various holding voltages, and distortion components of the current responses were detected by Fourier analysis. Current response magnitude and phase of each distortion component as a function of membrane potential were compared with characteristics of the voltage-dependent capacitance, obtained by voltage stair-step transient analysis or dual-frequency admittance analysis. The sum distortion was most prominent among the distortion components at all holding voltages. Notches in the sum (f1+f2), difference (f2−f1) and second harmonic (2f) components occur at the voltage where peak voltage-dependent capacitance resides (V pkCm ). Rapid phase reversals also occurred at V pkCm , but phase remained fairly stable at more depolarized and hyperpolarized potentials. Thus, it is possible to extract Boltzmann parameters of the motility-related charge movement from these distortion components. In fact, we have developed a technique to follow changes in the voltage dependence of OHC motility and charge movement by tracking the voltage at phase reversal of the f2−f1 product. When intracellular turgor pressure was changed, V pkCm and distortion notch voltages shifted in the same direction. These data have important implications for understanding cochlear nonlinearity, and more generally, indicate the usefulness of distortion analysis to study displacement currents. Experiments have been performed to investigate the mechanisms maintaining enzyme polymorphisms in natural populations. We have measured effects on fitness of genotypic variants at three loci, Est-5, Odh, and Mdh-2, in D. pseudoobscura. Significant differences exist among the genotypes in the rate of development from egg-to-adult; there is also indication of differences in larval survival. In a population segregating for allelic variants at all three loci, there is indication that segregation distortion at meiosis or some form of gametic selection might be involved. The relative fitnesses of alternative genotypes are reversed when either different fitness components are considered, or the genotypic frequencies are changed, or the larval density is increased. Segregation distortion has been reported repeatedly in soybean (Glycine max [L.] Merr.) inbred line populations segregating for the soybean cyst nematode (SCN) (Heterodera glycines Ichinohe) resistance gene rhg1. In each reported case, the frequency of the SCN resistance allele at the rhg1 locus was lower than expected. Segregation distortion was studied in 51 F4 populations by counting the number of plants predicted to be homozygous resistant, susceptible, and heterozygous for rhg1 based on the genetic markers Satt309, CTA, or TMA5. Significant (P<0.05) segregation distortion was observed in 44 out of the 51 F4 populations. When the heterozygotes were ignored, there were significantly fewer homozygous-resistant plants than expected in 33 populations. To study whether differential field emergence was a cause of the segregation distortion, three near isogenic line (NIL) populations segregating at the rhg1 locus for SCN resistance from plant introduction 88788 were tested. Population sizes ranged from 32 to 44 NILs and emergence was determined in field experiments in three environments. In each population, SCN-resistant NILs had significantly (P<0.05) less field emergence than susceptible NILs. In the population with the greatest effect, field emergence of resistant NILs was 6% less than susceptible NILs, with the entire population having an average emergence rate of 46%. Equations were derived to describe the effect of selection on segregation ratios over generations of population development and the observed emergence rates were transformed into fitness factors. Depending on assumptions of gene action, it was predicted from these fitness factors that segregation distortions were in the range of those reported previously for the rhg1 locus and were similar to what was observed on average across the 51 F4 populations. While other factors might also be involved, the results suggest that reduced field emergence associated with the SCN resistance allele contributes to previously reported segregation distortion at the rhg1 locus.  相似文献

The measurement of distortion-product otoacoustic emissions is a noninvasive method that can be used for assessing the sensitivity and the frequency tuning of nonlinear cochlear mechanics. During stimulation with two pure tones f1 and f2, the acoustic 2f1-f2 distortion was recorded in the ear canal of Cryptomys spec. to study specializations in cochlear mechanics that could be associated with the presence of a frequency expanded cochlear region between 0.8–1 kHz. In addition, a distortion threshold curve was obtained which describes relative threshold of nonlinear cochlear mechanics. Sensitive distortion thresholds could be measured for stimulus frequencies between 0.4 to 18 kHz with a broad minimum between 0.75 to 2.5 kHz. The distortion threshold curve extends to higher frequencies than previous neuronal data indicated.As a measure of mechanical tuning sharpness in the cochlea, suppression tuning curves of 2f1-f2 were recorded. The tuning curves reflected the typical mammalian pattern with shallow low frequency and steep high frequency slopes. Their tuning sharpness was poor with Q10dB values between 0.3 and 1.88. In the range of the frequency expanded region, the Q10dB values were below 0.5. This finding emphasizes that the presence of frequency expansion does not necessarily lead to enhanced mechanical tuning in the cochlea and one has to consider if in certain bat species with cochlear frequency expansion and particularly sharp cochlear tuning, the two phenomena may not be interlinked.Abbreviations CF constant frequency component of echolocation call - STC suppression tuning curve  相似文献

The directional hearing of male and female cicadas of the species Tympanistalna gastrica was investigated by means of laser vibrometry. The results show that the tympanic organs act as pressure difference receivers. This mechanism can produce left-right differences of more than 10 dB. The main acoustic inputs to the inner surfaces of the ears are the tympana, in males supplemented by the timbals, and by the third spiracles in females. In addition the hollow abdomen of males seems to play a minor role. Tympanic membrane input is the source of left-right differences in the tympanic vibration velocity at frequencies below 9 kHz in males and below 15–18 kHz in females. The input via the (contralateral) timbal in males is responsible for a null in vibration velocity appearing between 12 and 14 kHz when the sound is coming from the contralateral direction. The highest energy components of the calling song are found in this frequency range. The mechanical sensitivity of the ears depends upon the sex. At low frequencies males are about 10 dB more sensitive than females.  相似文献

1. Allozyme electrophoresis was used to describe the genetic structure of Helicopsyche borealis caddisflies collected from three sites in each of three streams in northern California, U.S.A.: the Rice Fork of the Eel River; Big Sulphur Creek; and Alameda Creek. Between 7 and 11km separated adjacent sites within these three streams. Helicopsyche borealis from three additional streams in eastern North America (Christiana Creek, Indiana; Byrd's Mill Creek, Oklahoma; Susquehanna River, Pennsylvania) were also analysed electrophoretically to address taxonomic questions that arose during the study. 2. Four genetically distinct groups of individuals were identified (i.e. Helicopsyche types A, B, C, and D). Lack of interbreeding between sympatric groups (as evidenced by fixed allelic differences) and large genetic differences (mean Nei's genetic distances = 0.396–0.693) indicate that these four groups of Helicopsyche were actually reproductively isolated species rather than genetic variants of a single species. 3. Occurrence of Helicopsyche type A at multiple sites permitted an analysis of spatial variation in genetic structure. Within a drainage basin, small differences in allele frequencies were observed among sites in the Rice Fork and Big Sulphur Creek, but not in Alameda Creek. Larger genetic differences were found among sites in separate drainage basins. Genetic distances (Nei's) between Helicopsyche type A from California and from eastern North America sites (mean = 0.236) were greater than interpopulation differences commonly observed for insects, which suggests that Helicopsyche type A from California may represent a different species than Helicopsyche type A from eastern North America. 4. Geographical and taxonomic differences observed in this study underscore the importance of understanding both population structure and genetic relationships among populations in the design and interpretation of stream faunal studies.  相似文献

Abstract

Biometrical observations on Cistus salvifolius L. (Cistaceae), a species with a very variable morphology, are presented: data, collected in Sicily, are relative to three environments in which populations of Cistus salvifolius live: garigue, maquis, pinewood. Measurements are compared with other ones, found in the literature, and in some cases they disagree or supply useful indications. On the basis of the different light regimes in the three environments, and of the parameters in which the difference among the three populations is most evident, some observations about the reaction to shade in Cistus salvifolius were made. On the basis of the few data collected, the intraspecific differences were given a possible explanation with the light factor: these differences are relative to morphological parameters such as leaf area, petiole length, stem length, and also to “derived” parameters such as specific leaf area and petiole length/leaf length ratio.  相似文献

To determine how the cost of reproduction varies with brood size, a population of blue tits (Parus caeruleus) breeding in Wytham Wood, England, was manipulated over a three year period. Two hundred sixteen pairs were randomly assigned 3, 6, 9, 12, or 15 nestlings; nestlings were exchanged soon after hatching. Survival of adult females (as measured by the proportion recaptured in the following winter and/or spring) declined significantly with increasing brood size in two out of three years; there was significant year-to-year variation in the relationship of recapture rate to brood size. Mean female recapture rates (averaged over the three years) declined in a linear fashion (P < 0.01). There was no significant linear or curvilinear relationship between male-recapture rate and brood size in any of the three years nor was there a significant linear or curvilinear relationship for the data averaged over the three years. Nevertheless, recapture proportions for males differed significantly with respect to brood size (χ2 test, P < 0.05). The possibility that experimental brood size influences subsequent dispersal (and therefore biases measures of survival based on recapture rates to differing degrees) was examined by comparing distances moved by breeding adults from one year to the next. There was no relationship between brood size and dispersal distance within the study area for either sex, except that females given broods of three were significantly more likely to move more than 300 m than were those given broods of 6–15 young. Both males and females showed evidence of a cost with respect to future fecundity: as brood size increased, the number of surviving offspring produced in the following year decreased from 1.5–1.6 (for adults that had reared 3–6 young) to 0.4 (for those that had reared 15 young). The relationship of future reproductive success to experimental brood size did not differ among years or between the sexes. The number of eggs laid and number of young hatched in year n + 1 did not differ significantly with respect to brood size in year n; rather, differences in future fecundity reflected differences in the survival prospects of young reared in year n + 1.  相似文献

Chromosomal regions associated with marker segregation distortion in rice were compared based on six molecular linkage maps. Mapping populations were derived from one interspecific backcross and five intersubspecific (indica / japonica) crosses, including two F2 populations, two doubled haploid (DH) populations, and one recombinant inbred (RI) population. Mapping data for each population consisted of 129–629 markers. Segregation distortion was determined based on chi-square analysis (P < 0.01) and was observed at 6.8–31.8% of the mapped marker loci. Marker loci associated with skewed allele frequencies were distributed on all 12 chromosomes. Distortion in eight chromosomal regions bracketed previously identified gametophyte (ga) or sterility genes (S). Distortion in three other chromosomal regions was found only in DH populations, where japonica alleles were over-represented, suggesting that loci in these regions may be associated with preferential regeneration of japonica genotypes during anther culture. Three additional clusters of skewed markers were observed in more than one population in regions where no gametophytic or sterility loci have previously been reported. Some of the species within the genus Cyclops O. F. Müller, 1785 can only be determined exactly by combining different methods and techniques. Based on the comprehensive analysis of morphometrical data, some populations in periodical ponds near Lake Constance were examined for their pattern of chromatin diminution and were compared by enzyme electrophoresis. These studies provided evidence of two new species. Together with C. furcifer Claus, 1857 and C. vicinus Uljanin, 1875 one of the ponds (‘Litzelsee’) was inhabited by four Cyclops species. In addition to morphological differences and distinct electrophoretic characteristics, C. furcifer and the new species differ in the timing of their chromatin diminution: C. singularis in the fourth, C. heberti in the fifth, and C. furcifer in the sixth cleavage. The qualitative course of chromatin diminution is rather similar in all three species. The same combination of three Cyclops species (without C. vicinus) was also found in an older sample (1990) from another ephemeral pond near Lake Constance. A close genetical relationship between the three species seems probable.  相似文献

The body plan of Drosophila, and presumably that of other insects, develops under the control of anterio-posterior and dorsal ventral axes, but no evidence for a left-right axis has yet been found. We used geometric morphometrics to study the wings in three species of flies: Drosophila melanogaster, Musca domestica and Glossina palpalis gambiensis. In all three species, we found that both size and shape showed subtle, but statistically significant directional asymmetry. For size, these asymmetries were somewhat inconsistent within and between species, but for shape, highly significant directional asymmetry was found in all samples examined. These systematic left-right differences imply the existence of a left-right axis that conveys distinct positional identities to the wing imaginal discs on either body side. Hence, the wing discs of Drosophila may be a new model to study the developmental genetics of left-right asymmetry. The asymmetries of shape were similar among species, suggesting that directional asymmetry has been evolutionarily conserved since the three lineages diverged. We discuss the implications of this evolutionary conservatism in conjunction with results from earlier studies that showed a lack of genetic variation for directional asymmetry in Drosophila.  相似文献

Drosophila simulans strains originating from Madagascar and nearby islands in the Indian Ocean often differ from those elsewhere in the number of sex comb teeth and the degree of morphological anomaly in hybrids with D. melanogaster. Here, we report a strong segregation distortion in the F1 intercross between two D. simulans strains originating from Madagascar and the US, possibly at both the gametic and zygotic levels. Strong bias against alleles of the Madagascar strain was observed for all ten marker loci distributed over the entire second chromosome in the F1 intercross, but only a few showed a weak distortion in the isogenic backgrounds of either strains. Significant deviations of genotype frequencies from Hardy–Weinberg proportions were consistently observed for the second chromosome. By contrast, the X and third chromosomes did not show any strong segregation distortion. Crossover frequency on the second chromosome was uniformly reduced in isogenic backgrounds whereas the map lengths in the F1 intercross were comparable to or larger than that of the standard D. melanogaster map. We discuss these findings in relation to previous studies on other traits and interspecific differences between D. mauritiana, which is endemic to Mauritius Island, and D. simulans.  相似文献

Summary Female gametophytes of knobcone pine were used to study genetic variation at 58 loci in 26 enzyme systems. Mendelian segregation and linkage were tested at 21 loci. Got1, Pgi2, Mnr3, Adh2, and Lap2 were linearly arrayed in a single linkage group. Est and Acp3, and Flest and Lap1, formed two independent linkage groups. Although Mendelian segregation was the rule, several cases of segregation distortion were observed. Pooled over trees, Lap1 and Aap1 showed significant distortion. Of 11 cases of distortion observed for individual trees, 10 showed an excess of common alleles. Pooled over both loci and trees, giving a total sample of 17,183 gametes, the common alleles were significantly overrepresented by 1.1%, and heterogeneity was highly significant. Our results, and others in the literature, suggest that segregation distortion may affect the genetic structure of conifer populations.  相似文献

Amphidinium operculatum Claparède et Lachmann, the type species of the dinoflagellate genus Amphidinium, has long had an uncertain identity. It has been considered to be either difficult to distinguish from other similar species or a morphologically variable species itself. This has led to the hypothesis that A. operculatum represents a “species complex.” Recently, the problem of distinguishing A. operculatum from similar species has become particularly acute, because several morphologically similar species have been found to produce bioactive compounds of potential interest to the pharmaceutical industry. In this study, we cultured and examined existing cultures of several species of Amphidinium, most of which have been previously identified as A. operculatum or as species considered by some to be synonyms or varieties of A. operculatum. Thirty strains were examined using comparative LM, SEM, and partial large subunit (LSU) rDNA sequence data. Through morphological and molecular phylogenetic analyses, six distinct species were identified, including Amphidinium trulla sp. nov. and Amphidinium gibbosum comb. nov. Amphidinium operculatum was redescribed based on four cultures. Genetic variability within the examined Amphidinium species varied greatly. There was little difference among strains in partial LSU rDNA for most species, but strains of A. carterae and A. massartii Biencheler differed by as much as 4%. In both A. carterae and A. massartii, three distinct genotypes based on partial LSU rDNA were found, but no morphological differences among strains could be observed using LM or SEM. In the case of A. carterae, no biogeographically related molecular differences were found.  相似文献

The taxonomic positions of soil isolates known as Streptomyces groups A, B and C were clarified. Comparative 16S rDNA sequence studies indicated that representatives of all three taxa formed distinct phyletic lines within the Streptomyces tree though the group A strains were shown to be related to Streptomyces griseus and associated validly described species. The taxonomic integrity of all three groups was highlighted by DNA:DNA relatedness and ribotype data though the group A strains encompassed a higher degree of genetic variation than the group B and C strains. In light of these and earlier phenotypic data it is proposed that Streptomyces groups A, B and C be given species status as Streptomyces sanglieri sp. nov., Streptomyces aureus sp. nov. and Streptomyces laceyi sp. nov., respectively. Genetic variability of AFLP markers was studied in 20 populations of Siberian fir (Abies sibirica, Pinaceae) and in two populations of Far-Eastern species Manchurian fir A. nephrolepis and Sakhalin fir A. sachalinensis each. Four pairs of selective primers were used. In total, 168 samples from three fir species were genotyped for 117 polymorphic loci. According to the AMOVA results, the variability proportion characterizing the differences among three Abies species was several times higher (F CT = 0.53) than that acounting for population differences within the species (F SC = 0.125). Differentiation of the A. sibirica populations based on AFLP markers exceeded 14% (F ST = 0.141). Significant correlation between the genetic distances calculated from the AFLP data and the geographic distances between populations was found. The results of AFLP variability analysis supported and supplemented the conclusions inferred previously from allozyme and cpSSR data: several genetically similar geographic groups of Siberian fir were identified. These groups differ both in allele frequencies and in the levels of genetic variation.  相似文献
