Rabbit polyclonal antibodies prepared against faba bean phyllody MLO from the Sudan reacted with its homologous antigen and with extracts of Catharanthus roseus experimentally infected with the same or a related MLO from Crotalaria saltiana showing symptoms of phyllody disease, as well as with extracts of naturally MLO-infected C. saltiana growing in the field in the Sudan. The antibodies also reacted positively with extracts of C. roseus experimentally infected with Crotalaria juncea phyllody MLO and soybean phyllody MLO from Thailand. Polyclonal antibodies prepared against an MLO associated with witches' broom disease in C. juncea reacted positively in ELISA tests with homologous antigen extracts from naturally infected C. juncea as well as with extracts from experimentally infected C. roseus and with extracts prepared from Sesamum indicum plants with phyllody symptoms growing in Thailand. There was no reaction between these antibodies and extracts from C. roseus plants infected with the MLOs associated with C. juncea phyllody or with soybean phyllody. No cross reactions were observed among the antigens and antibodies of the two MLO groups by immunoflorescence, ELISA or western blotting. However, the molecular weight of the principal protein antigen, determined by SDS-polyacrylamide gel electrophoresis (SDS-PAGE) and western blotting was the same for both types of MLO. Serologically-similar MLOs thus occur in the Sudan and in Thailand, where they are associated with phyllody symptoms in C. saltiana and faba bean and with C. juncea and soybean, respectively. A second, serologically distinct MLO group was also found infecting C. juncea and S. indicum in Thailand but MLOs from this group have not yet been identified in crops from the Sudan.  相似文献

A lectin preparation obtained fromTetracarpidium conophorum (Nigerian walnut) by affinity chromatography of seed extracts on lactose-agarose has been shown to contain two components by gel filtration on Sephadex G150. The larger componentTetracarpidium conophorum agglutinin I (TCAI) is a disulphide-bonded 70 kDa homodimer whereas the second component TCAII is a 34 kDa monomeric protein. Amino terminal aminoacid sequencing shows identity in TCAI and TCAII for the first fifteen residues after which the sequences diverge. TheN-terminal sequences of TCAI and TCAII show identity with sequences in the B-chains of ricin andRicinus communis agglutinin I (RCAI) in eleven of the initial fifteen residues. Thereafter TCAI appears to be homologous to the ricin B chain whereas TCAII is more homologous with the B chain of RCAI. A limited screening of the carbohydrate-binding specificity of TCAII by affinity chromatography of defined oligosaccharides on TCAII Sepharose columns shows that the binding specificity reported earlier for affinity purifiedTetracarpidium conophorum isolectins (Sato S, Animashaun T, Hughes RC (1991)J Biol Chem 266:11485–94) reflects the binding properties of TCAII which is the major isolectin in unfractionated lectin preparations.  相似文献

A crop legume Vigna unguiculata L. (Walp.) and a wild legume Crotalaria juncea L. were evaluated for their relative responses to the oxidative stress injury induced by various doses of UV-B radiation (UV-B, 280–315 nm; 0, 1.0, 1.4, 4.7, and 6.0 kJ m−2 d−1). A dose-dependent damage in lipid peroxidation was determined as an index of membrane injury caused by UV-B. The impact was significantly higher in V. unguiculata than in C. juncea. The specific activities of superoxide dismutase, ascorbate peroxidase, monodehydroascorbate reductase, and dehydroascorbate reductase increased directly proportional to UV-B doses. However, the activities of these enzymes were significantly higher in V. unguiculata than in C. juncea indicating that V. unguiculata was inflicted with more severe oxidative stress injury under UV-B. In C. juncea the glutathione reductase and ascorbate oxidase activities were 35 and 40 % greater than in V. unguiculata, respectively. Further, the non-enzymatic antioxidants ascorbate and glutathione, and their reduced/oxidizes ratios in C. juncea were much greater than V. unguiculata indicating C. juncea has an inherently greater antioxidative potential than V. unguiculata. Thus C. juncea is better adapted to oxidative stress than V. unguiculata by means of efficient cellular antioxidant mechanisms helping to combat the photooxidative stress injury elicited by UV-B.  相似文献

The effects of intercycle cover crops on Rotylenchulus reniformis population densities in pineapple were evaluated in one greenhouse and two field experiments. In the greenhouse, Crotalaria juncea, Brassica napus, and Tagetes erecta were planted for 3 months and then incorporated. These treatments were compared to weedy fallow with or without 1,3-dichloropropene (1,3-D) in three soils (Makawao fallow, Wahiawa fallow, and Wahiawa pineapple) naturally infested with R. reniformis. All cover crop incorporation suppressed R. reniformis numbers in cowpea more than did the weedy treatment in the Makawao (P < 0.05) but not in the Wahiawa soils. Crotalaria juncea treatment increased bacterivorous nematodes and nematode-trapping fungal population densities more than the other treatments in Makawao fallow and Wahiawa pineapple-planted soils. The field trials included the same plants as well as Sinapis alba. Treatments with Crotalaria juncea and 1,3-D maintained lower R. reniformis population densities on pineapple longer than other cover crops or weedy fallow treatments. Crotalaria juncea could have suppressed R. reniformis because it is a poor host and because it enhances nematode-trapping fungi when incorporated into soil. Treatment with 1,3-D reduced microbial activities but produced the greatest pineapple yield.  相似文献

The influence of Chloris gayana, Crotalaria juncea, Digitaria decumbens, Tagetes patula, and a chitin-based soil amendment on Hawaiian populations of Rotylenchulus reniformis was examined. Chloris gayana was a nonhost for R. reniformis. The nematode did not penetrate the roots, and in greenhouse and field experiments, C. gayana reduced reniform nematode numbers at least as well as fallow. Tagetes patula was a poor host for reniform nematode and reduced reniform nematode numbers in soil better than did fallow. Crotalaria juncea was a poor host for R. reniformis, and only a small fraction of the nematode population penetrated the roots. Crotalaria juncea and D. decumbens reduced reniform nematode populations at least as well as fallow. A chitin-based soil amendment, applied at 2.24 t/ha to fallow soil, did not affect the population decline of reniform nematode.  相似文献

Abstract

This study evaluated remediation of the herbicide sulfentrazone in soils with three different mineralogies (kaolinite, hematite, and gibbsite) and three remediation sulfentrazone treatments (Canavalia ensiformis L., Crotalaria juncea L., and natural attenuation). This study was conducted in a factorial scheme, in triplicate with randomized block design. Sulfentrazone was applied at 0 and 400?g ha?1. We analyzed sulfentrazone residue in the soils by high-performance liquid chromatography and confirmed the results with bioassays of Pennisetum glaucum. Herbicide movement was greater in the kaolinitic soil without plant species. The retention of herbicide in the kaolinitic soil occurred in larger quantities in the 0–12?cm layer, with higher levels found in the treatments with plants. In the hematitic soil with C. juncea, all applied herbicides were concentrated in the 0–12?cm layer. In the other hematitic soil treatments, sulfentrazone was not detected by chemical analysis at any soil depth, although in many treatments, it was detected in the bioassay. Phytoremediation was more efficient with C. ensiformis grown in gibbsitic soil, reducing the sulfentrazone load by approximately 27%. Natural attenuation was more efficient than phytoremediation in oxidic soils due to soil pH and texture soils favored microbial degradation of the compound.

A lectin was purified from Crotalaria paulina seeds by ion-exchange and FPLC molecular exclusion chromatography. CrpL had an apparent molecular mass of 30 kDa, as determined by SDS-PAGE under non-reducing and reducing conditions. CrpL effectively agglutinated human and cow erythrocytes, and this activity was not affected by 20 mM EDTA, showing no dependence of divalent cations. Hemagglutination was inhibited by N-acetyl- D-galactosamine, D-galactose and was also inhibited by glycoproteins, fetuin and asialofetuin. The N-terminal amino acid sequence of CrpL was identical to those of other lectins from the genus Crotalaria, and amino acid composition showed high amounts of Asx and Glx, and was rich in Gly, Ala and Ser, as also reported for lectins from other Crotalaria species. CrpL inhibited the growth of Xanthomonas axonopodis pv. phaseoli and Xanthomonas axonopodis pv. passiflorae, suggesting a role of this lectin in the defense of seeds against bacterial infections.  相似文献

Two lectins were purified by affinity chromatography from mature peanut (Arachis hypogaea L.) nodules, and compared with the previously characterised seed lectin of this plant. One of the nodule lectins was similar to the seed lectin in its molecular weight and amino-acid composition and ability to bind derivatives of galactose. However, unlike the seed lectin, this nodule lectin appeared to be a glycoprotein and the two lectins were only partially identical in their reaction with antibodies prepared against the seed lectin. A lectin was isolated from Ulex europaeus seeds by affinity chromatography on affinity adsorbent prepared by copolymerization of acrylamide, N,N'-methylene bisacrylamide and maleylated hog stomach peptone. The lectin is homogeneous as judged by ultracentrifugation (s20,w = 6.4 S), electrophoretic and gel chromatography criteria; it contains 4.2% neutral sugar and 1.4% glucosamine. Its molecular weight is approx. 110,000 and the molecule consists of two noncovalently linked protomers which are formed by two covalently bound basic subunits (Mr = 30,000). The preparation contains three isolectins differing in the strength of interaction with specific sugars (cellobiose, N-acetyl-D-glucosamine) under the conditions of affinity electrophoresis. The lectin is non-specific with human ABO blood group system, the agglutination is inhibited by partial chitin hydrolysate, hog stomach peptone and high concentration of cellobiose.  相似文献

The carbohydrate specificity of three novel lectins, Boletopsis leucomelas lectin (BLL), Aralia cordate lectin (ACL), and Wasabia japonica lectin (WJL), was examined by frontal affinity chromatography using a panel of fluorescently labeled 47 oligosaccharides. The results indicate that BLL recognizes an agalacto structure of the biantennary chain and its bisecting structure. ACL showed strong affinity for triantennary oligosaccharides, but no affinity for tetraantennary structure. WJL showed no appreciable affinity for any of the 47 glycans examined. These lectins with a unique affinity specificity might be useful for examining alterations in the glycan structures of the glycoconjugates in association with development and various diseases.  相似文献

The development and implementation of sustainable and environmentally friendly agricultural practices are indispensable as alternatives to pesticide use and to keep populations of soil-borne plant pathogens at levels that do not affect crop productivity. The present research evaluates the incidence of soil-borne phytopathogens on the pineapple variety MD-2, which was subjected to different treatments: Incorporation of Crotalaria juncea into the soil (organic amendment), application of dolomitic lime to soil (inorganic amendment), and the use of plastic mulch covering the soil. During the crop cycle (15 months), the following variables were evaluated: plant height (cm), fruit weight (kg·plant−1 ), crop yield (ton·ha−1 ), the bud root disease incidence caused by Phytophthora nicotianae, number of soil phytoparasitic nematodes and colony-forming-units (CFUs) of soil fungi and oomycetes. The results indicate that Crotalaria juncea treatment reduced the pathogen population (nematode and oomycetes) at levels that did not affect crop development, so that yield increased (18–20%). The incorporation of C. juncea into the soil as an organic amendment favors the populations of fungi disease suppressors (Trichoderma-Aspergillus). The phytoparasitic nematodes (Meloidogyne sp., Pratylenchus sp., and Mesocriconema sp.) and oomycetes (Phytophthora spp., and Pythium spp.) showed a reduction of their population levels by effects of organic amendment (C. juncea). The plastic mulch was also effective, probably due to the maintenance of optimal condition to crop growth and weed control. However, the dolomitic lime application had the poorest effect under the conditions of the study area on the variables analyzed. The described observations are characteristics of a system-based approach for the potential management of soil-borne pathogens of pineapple MD-2 in Veracruz, México.  相似文献

Several aspects of the interaction of various lectins with the surface of Ehrlich ascites carcinoma cells are described. The order of agglutinating activity for various various lectins is Ricinuscommunis > wheat germ concanavalin A soybean >Limuluspolyphemus. No agglutination was noted for Ulex europaeus. Using 125I-labeled lectins it was determined that there are 1.6 and 7 times as many Ricinus communis lectin binding sites as sites for concanavalin A and soybean lectins. Sodium deoxy-cholate-solubilized plasma membrane material was subjected to lectin affinity chromatography and sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The lectin receptors of the plasma membrane appeared to be heterogeneous and some qualitative differences could be discerned among the electrophoretically analyzed material, which bound to and was specifically eluted from the various lectin affinity colums. The characteristics of elution of bound material from individual lectin columns indicated secondary hydrophobic interactions between concanavalin A or wheat germ agglutinin and their respective lectin receptor molecules.  相似文献

A lectin was isolated fromCanavalia brasiliensis Mart. seeds by combining solubility fractionation with affinity chromatography on Sephadex G-50. The lectin showed a carbohydrate specificity for D-mannose (D-glucose) binding and a requirement for Ca2+ and Mn2+. All the hemagglutinating activity was found in the cotyledons and the presence of the lectin was followed during the first 15 days of plant germination, through the activity against rabbit erythrocytes, the presence of the “lectin peak” in Sephadex G-50 affinity chromatography, presence of the “lectin bands” in SDS-polyacrylamide gel electrophoresis and the “lectin arcs and rockets” in immunoelectrophoresis in agarose gel. On application of all these methods the lectin showed a differentiated metabolism, disappearing more slowly than the other high molecular weight proteins of the seed.  相似文献

Tomato lectin is specific for oligomers of poly-N-acetyllactosamine containing 3 repeating Gal(beta 1-4)GlcNAc (beta 1-3)-disaccharides. As such it is highly useful for purifying oligosaccharides or glycopeptides with poly-N-acetyllactosamine character. We have found the lectin very useful as an affinity reagent for isolating glycoproteins or glycoprotein domains having poly-N-acetyllactosamine glycosylation. Conventional preparation of tomato lectin by ovomucoid-Sepharose affinity chromatography was found to be unsatisfactory due to instability of column and bleeding of ovomucoid into eluents requiring the necessity for additional purification steps following affinity chromatography. We prepared a column of human erythrocyte band 3 carbohydrate glycopeptide (erythroglycan) attached to Sepharose as an affinity matrix. The purification of tomato lectin to homogeneity in one step on this column matrix is described in this report.  相似文献

Abstract This paper describes an assessment of the effect of exposure to fire‐related cues (heat shock, smoke and nitrate) and the interactions between the cues on seed dormancy release of tropical savanna legumes in north‐eastern Australia. Ten legume species were tested, comprising both native and exotic species. The ten species responded variously to the treatments. Brief exposure to temperatures between 80 and 100°C was found to break the seed dormancy of the native ephemeral herbs Chamaecrista mimosoides, Crotalaria calycina, Crotalaria montana, Indigofera hirsuta and Tephrosia juncea, as well as the exotic ephemeral herb Crotalaria lanceolata. Exposure to 80°C combined with treatment with a nitrate solution produced an additive effect on the germination of Chamaecrista mimosoides and Crotalaria lanceolata. However, the four species with the heaviest seeds, two exotic ephemeral herbs (Chamaecrista absus and Crotalaria pallida) and two native perennials (Galactia tenuiflora and Glycine tomentella) displayed no significant increase in germination with exposure to fire‐related cues. Exposure to 120°C for 5 min produced seed mortality in all species tested. Two of the largest seeded species, Crotalaria pallida and Galactia tenuiflora, displayed the lowest tolerance to heat shock, with seed mortality after exposure to 100°C for 5 min. These data indicate that fire can promote the germination of some tropical savanna legumes. As a proportion of seeds of each species displayed no innate dormancy, some germination may occur in the absence of fire, especially of exotic species.  相似文献

In this work, we characterized chemically the N-acetyl-D-galactosamine specific lectin from Amaranthus leucocarpus syn hypocondriacus lectin (ALL). It is a dimeric glycoprotein composed by three isoforms with pl at 4.8, 4.9, and 5.2. Circular dichroism analysis indicated that the secondary structure of ALL contains 45% of -sheet and 5% of -helix. Amino acid sequence of the purified lectin and its isoforms was determined from peptides obtained after trypsin digestion by MALDI-TOF (Matrix assisted laser desorption ionization-time of flight). The tryptic peptides prepared from the purified lectin and the three isoforms showed different degrees (80 to 83%) of identity with the amino acid sequence belonging to a previously described high nutritional value protein from A. hypocondriacus not shown at the time to be a lectin. Furthermore, analysis of tryptic peptides obtained from ALL previously treated with peptide N-glycosidase, revealed a 93% identity with the aforementioned protein. Presence of N-glycosidically linked glycans of the oligomannosidic type and, in minor proportion, of the N-acetyllactosaminic type glycans was determined by affinity chromatography on immobilized Con A.  相似文献

A lectin was isolated from the roots of Sesbania aculeata. This is a glucose specific lectin having 39 kDa subunit molecular weight. The expression of this lectin was found to be developmentally regulated and observed to be the highest in the second week. The lectin was purified by affinity chromatography using Sephadex G-50 and found to have 28% homology with Arabidopsis thaliana lectin-like protein (accession No. CAA62665). The lectin binds with lipopolysaccharide isolated from different rhizobial strains indicating the plants interaction with multiple rhizobial species.
