Fucus serratus and F. evanescens commonly occur on Northern European shores. Nine microsatellite loci were developed for F. serratus (8–22 alleles, observed heterozygosities = 0.367–0.850) and one for F. evanescens (seven alleles, observed heterozygosity = 0.804). Cross‐amplification was apparent, as five F. serratus loci were polymorphic in F. evanescens and 2–5 were polymorphic in F. vesiculosus, F. distichus, and F. spiralis.  相似文献

Eukaryotic mitochondria are mostly uniparentally (maternally) inherited, although mtDNA heteroplasmy has been reported in all major lineages. Heteroplasmy, the presence of more than one mitochondrial genome in an individual, can arise from recombination, point mutations, or by occasional transmission of the paternal mtDNA (=paternal leakage). Here, we report the first evidence of mtDNA paternal leakage in brown algae. In Denmark, where Fucus serratus L. and Fucus evanescens C. Agardh have hybridized for years, we found eight introgressed individuals that possessed the very distinct haplotypes of each parental species. The finding of heteroplasmy in individuals resulting from several generations of backcrosses suggests that paternal leakage occurred in earlier generations and has persisted through several meiotic bottlenecks.  相似文献

Patterns of nuclear and organelle inheritance among artificial hybrids of the seaweeds Fucus serratus and F. evanescens were detected using single-strand conformation polymorphism (SSCP). Three alleles were identified in the 231?bp rDNA-ITS1 gene (nuclear): two in F. serratus and one in F. evanescens. Alleles differed by 1–2?bp and all hybrids possessed one allele from each parent. Two haplotypes were present in the 288?bp Rubisco spacer (chloroplast), differentiated by a 33?bp indel. Two haplotypes differing by a single nucleotide were found in a 135?bp region of nad11 gene (mitochondrion). Both organelles are maternally inherited, as all hybrids contained the haplotypes of the parent contributing the egg. Although laboratory hybrids among Fucus spp. have been produced previously, this is the first time that both nuclear and cytoplasmic genetic markers have been used to document inheritance patterns. SSCPs analysed on an automated sequencer offer a rapid and powerful approach for identifying suspected hybrids from field samples, as well as a screen for intraspecific and intra-individual variation in DNA regions prior to confirmation of variations by sequencing.  相似文献

Invasion of a Habitat-Forming Seaweed: Effects on Associated Biota   总被引:1,自引:0,他引:1  
Fucus evanescens is a brown alga of arctic origin that has invaded European coasts. The epiphytic community of F. evanescens in southern Sweden was compared with that of the native Fucus vesiculosus, to examine to what extent an invading seaweed can modify local biodiversity. F. evanescens was much less fouled than F. vesiculosus, supporting both less biomass and fewer species of epiphytes. Multivariate analysis of the most common epiphyte taxa showed that the epiphytic community composition of F. evanescens was not entirely separated from that of F. vesiculosus, but host species contributed significantly to explain the variation in community composition. The biomass of free-living invertebrates was also lower on F. evanescens, although the pattern differed between taxonomic groups. While the biomass of amphipods was lower on F. evanescens, there was no significant difference in biomass of isopods or gastropods between the Fucus species. The good correlation between biomass of epiphytes and free-living animals suggests that the epiphytes play an important role in providing a suitable habitat for many species of free-living epifauna. The study shows that the invasion of F. evanescens affects the environmental conditions for many species associated with the Fucus community but that the direct effect on biodiversity is probably low.  相似文献

The prevalence of antigrazing defense induction and the cues triggering induction in marine macroalgae are generally not well understood. We examined the capacity of defense and the mechanisms of regulation in five common perennial macroalgal species from the Baltic Sea, Furcellaria lumbricalis (Huds.) J. V. Lamour., Delesseria sanguinea (Huds.) J. V. Lamour., Phyllophora pseudoceranoides (S. G. Gmel.) Newroth et A. R. A. Taylor, Fucus serratus L., and Fucus evanescens C. Agardh. Specifically, we investigated whether direct feeding and/or waterborne cues from feeding on neighboring conspecifics decreased the palatability of the tested algae. Direct feeding by the local isopod Idotea baltica triggered the induction of chemical defense in Fur. lumbricalis, D. sanguinea, P. pseudoceranoides, F. serratus, and F. evanescens. Conversely, we did not find any evidence for waterborne cues associated with feeding to trigger defense induction in neighboring conspecifics.  相似文献

Reproductive allocation, the proportion of total dry weight allocated to receptacle tissue and reproductive effort, the proportion of reproductive carbon requirement contributed by receptacle photosynthesis, were measured in two fucoid algal species Fucus serratus and Himanthalia elongata at sites in NE Scotland. Reproductive development takes over ten months in H. elongata, a semelparous (single reproductive event) species, and reproductive allocation at receptacle maturity is over 98%. Following gamete release, the whole thallus dies. In contrast, reproductive development in F. serratus takes four months. Fucus serratus is iteroparous (capable of multiple reproductive events), reproductive allocation is 38.6% for the first reproductive event and 50.5% for the following year's event. In Fucus serratus, the receptacles appear to make a major contribution to their own carbon requirements after the first month of reproductive development. The receptacles of Himanthalia elongata contribute only a small proportion of the receptacle carbon requirements in the first four months of reproductive development, after which the contribution made through receptacle photosynthesis increases.  相似文献

Hybrid zones in fluvial fishes may be heterogeneous from drainage to drainage. The comparison of data from morphology, allozymes, and mitochondrial DNA (mtDNA) indicates variability in the causes and degree of restriction of gene flow between Notropis cornutus and Notropis chrysocephalus. Allozyme marker loci show frequency-dependent introgression; i.e., the rarer species, whichever it is at a particular locality, tends to exhibit a higher proportion of introgressed alleles. Unlike allozymes, introgression of mtDNA haplotypes varies geographically. In westward-flowing Michigan drainages, N. cornutus mtDNA haplotypes are more common in F1 hybrids and backcrosses, independent of parental frequencies. In eastward-flowing Michigan drainages, N. chrysocephalus mtDNA is more common in F1 hybrids and backcrosses; this pattern may be due to local ecological effects or frequency-dependent introgression. Morphological data alone are not sufficient to distinguish all classes of hybrids. The lack of concordance of morphological, allozymic, and mtDNA introgression patterns implies operation of one or two factors: 1) geographically variable patterns of selection against different hybrid and backcross combinations or 2) genetic differences between Michigan populations inhabiting eastward- and westward-flowing drainage systems accumulated during historical isolation.  相似文献

The macroalgal belt in the southern Baltic Sea may be partly structured by the interaction of physical and biological factors. A field study, spanning the 1990s, describes a rapid decline of the Fucus spp. stands along the wave-exposed Swedish southeast coast. During this period, a relative dominance of Fucus vesiculosus L. shifted to a relative dominance of Fucus serratus L. The decline of F. vesiculosus coincided with observations of large numbers of the grazing isopods Idotea baltica (Pallas) and Idotea granulosa Rathke, or with field observations of frequent grazing marks on Fucus fronds. I. baltica, but not I. granulosa, tended to aggregate in the declining Fucus spp. stands, indicating a strong preference for Fucus spp. In a mesocosm experiment I. baltica, when given a choice, grazed both Fucus species at weak water motion. At strong water motion grazing was concentrated on F. vesiculosus. It is hypothesized that one of the reasons I. baltica preferred F. vesiculosus to F. serratus at strong water motion may have been differences in habitat quality, like width of thallus, influencing the ability to cling to the plant. Smaller thallus, as in F. vesiculosus, thus is the preferred habitat for grazing of I. blatica. We postulate that the existence of F. serratus in the area may be favoured by strong wave action and moderate but not strong grazing by I. baltica, relaxing the interspecific competition from F. vesiculosus.  相似文献

Abstract: This study has shown that in Fucus serratus and Fucus distichus, young zygotes and embryos are highly susceptible to elevated levels of both UVA (UVAR) and UVB radiation (UVBR). Zygotes treated with UVAR are able to polarise and germinate, but are very slow to divide; if they do, they often have skewed division planes or deformed rhizoids. Those treated with UVAR and UVBR remain spherical, they do not polarise, germinate to form rhizoids or undergo cell division. We suggest that the UVR may be affecting the cytoskeleton. Conversely, zygotes and embryos of Fucus spiralis are able to withstand these same UVR levels and, at the light microscope level, appear to develop normally. When the brown algal phenolic compound phloroglucinol was placed in a filter covering the developing embryos, normal development was seen under all treatments. Phenolic compounds protect the developing fucoids from UVR. In comparison with the other two species, Fucus spiralis grows high up on the shore and is exposed for much longer periods of time and, presumably, to higher levels of natural UVR. The failure of the juvenile stages of F. serratus and F. distichus to withstand UVR stress may have implications for the continued survival of these species in the intertidal, and may prove detrimental to the population as a whole if UVR levels increase.  相似文献

Rhinanthus minor andRhinanthus angustifolius are known to hybridize in mixed populations in nature. These hybridization events can have important evolutionary consequences. The development and use of species-specific RAPD and ISSR markers allowed the detection of hybrid individuals not always distinguishable with morphological characters. Two mixed populations of different ages were studied. In a young mixed 2-year-old population, both individuals of the two parental species and F1 hybrids were found using genetic analysis, showing that hybridization occurred rapidly. Flower morphology of F1 hybrids was too variable to distinguish all these hybrids from the parental species. This morphological variability of F1 hybrids was also confirmed in artificial crosses in the greenhouse. In an old and no longer mixed 30-year-old population, onlyR. angustifolius plants and a few genetically introgressed individuals close toR. angustifolius were present. Genetic markers showed traces of past hybridization and introgression. Unidirectional introgression ofR. minor intoR. angustifolius with the complete disappearance ofR. minor from this population was observed.  相似文献

We examined six allozyme markers and shell morphology to study gene flow between the naturally sympatric freshwater snail species Viviparus ater and V. contectus in Lake Garda, Italy, using offspring from experimental crosses and snails collected from natural populations. Hybrid offspring obtained from experimental crosses and hybrids collected from natural populations were heterozygous at four or five out of five diagnostic loci. Shell morphology was a poor predictor of the hybrid status of individual snails. Backcrossed offspring from experimentally bred hybrids and either of the parental species morphologically resembled the parental species. There was no evidence of segregation distortion at the six loci. Linkage analysis revealed one pair of linked loci (GPI and PNP). These cross experiments indicated a Mendelian type hybrid system in which gene introgression may occur. F1 hybrids were found at four out of six sampling sites of Lake Garda, Italy. Local frequencies of F1 hybrids ranged from 0% to 1.6% (estimated average = 0.74%) of the total population including both species. Alleles typical of V. ater were found at low frequencies in V. contectus at all six sampling sites. Alleles typical of V. contectus were found at low frequencies in V. ater at three out of six sampling sites. This is consistent with the hypothesis of introgression in both directions.  相似文献

Background and Aims

Quercus suber and Q. ilex are distantly related and their distributions partially overlap. They hybridize occasionally, but the complete replacement of Q. suber chloroplast DNA (cpDNA) by that of Q. ilex was identified in two specific geographical areas. The objective of this study was to determine whether the contrasting situation reflected current or recent geographical interspecies gene flow variation or was the result of ancient introgression.

Methods

cpDNA PCR-RFLPs (restriction fragment length polymorphisms) and variation at ten nuclear microsatellite loci were analysed in populations of each species, in 16 morphologically intermediate individuals and the progeny of several of them. Interspecies nuclear introgression was based on individual admixture rates using a Bayesian approach with no a priori species assignment, and on a maximum-likelihood (ML) method, using allele frequencies in the allopatric populations of each species as controls. Gene flow was compared specifically between populations located within and outside the specific areas.

Key Results

High interspecies nuclear genetic differentiation was observed, with twice the number of alleles in Q. ilex than in Q. suber. According to Bayesian assignment, approx. 1 % of individuals had a high probability of being F1 hybrids, and bidirectional nuclear introgression affected approx. 4 % of individuals in each species. Hybrid and introgressed individuals were identified predominantly in mixed stands and may have a recent origin. Higher proportions including allospecific genes recovered from past hybridization were obtained using the ML method. Similar rates of hybridization and of nuclear introgression, partially independent of cpDNA interspecies transfer suggestive of gene filtering, were obtained in the populations located within and outside the areas of complete cpDNA replacement.

Conclusions

The results did not provide evidence for geographical variation in interspecies gene flow. Transgenic plants have increased interest in the study of crop gene introgression in wild populations. Genes (or transgenes) conferring adaptive advantages persist in introgressed populations, enhancing competitiveness of wild or weedy plants. This represents an ecological risk that could increase problems of weed control. Introgression of cultivar alleles into wild plant populations via crop–wild hybridisations is primarily governed by their fitness effect. To evaluate this, we studied the second generation of seven wild–crop interspecific hybrids between weedy Helianthus petiolaris and cultivated sunflower, Hannuus var. macrocarpus. The second generation comprised open‐pollinated progeny and backcrosses to the wild parent, mimicking crosses that occur in natural situations. We compared a number of morphological, life history and fitness traits. Multivariate analysis showed that the parental species Hannuus and Hpetiolaris differed in a number of morphological traits, while the second hybrid generation between them was intermediate. Sunflower crop introgression lowered fitness of interspecific hybrids, but fitness parameters tended to recover in the following generation. Relative frequency of wild/weedy and introgressed plants was estimated through four generations, based on male and female parent fitness. In spite of several negative selection coefficients observed in the second generation, introgressed plants could be detected in stands of <100 weedy Hpetiolaris populations. The rapid recovery of fecundity parameters leads to prediction that any trait conferring an ecological advantage will diffuse into the wild or weedy population, even if F1 hybrids have low fitness.  相似文献

Protoplasts were isolated enzymatically from meristematic tissues of the brown algae, Fucus serratus, using a combination of 2% cellulase R-10 Onozuka, 0.5% macerozyme and 1% crude extract of gland gut of Aplysia vaccaria. The main factors affecting protoplast yield were identified. Protoplasts were produced in large quantities from apical region of thallus and from plantlets compared to mature explants. Yields were greatly improved by the addition of sodium citrate and bovine serum albumin in the enzymatic solution and could reach 5.8 × 106 protoplasts per gram of fresh wt. The applicability of these optimal parameters to other species Fucus vesiculosus was shown.  相似文献

Marginal populations are often geographically isolated, smaller, and more fragmented than central populations and may frequently have to face suboptimal local environmental conditions. Persistence of these populations frequently involves the development of adaptive traits at phenotypic and genetic levels. We compared population structure and demographic variables in two fucoid macroalgal species contrasting in patterns of genetic diversity and phenotypic plasticity at their southern distribution limit with a more central location. Models were Ascophyllum nodosum (L.) Le Jol. (whose extreme longevity and generation overlap may buffer genetic loss by drift) and Fucus serratus L. (with low genetic diversity at southern margins). At edge locations, both species exhibited trends in life‐history traits compatible with population persistence but by using different mechanisms. Marginal populations of A. nodosum had higher reproductive output in spite of similar mortality rates at all life stages, making edge populations denser and with smaller individuals. In F. serratus, rather than demographic changes, marginal populations differed in habitat, occurring restricted to a narrower vertical habitat range. We conclude that persistence of both A. nodosum and F. serratus at the southern‐edge locations depends on different strategies. Marginal population persistence in A. nodosum relies on a differentiation in life‐history traits, whereas F. serratus, putatively poorer in evolvability potential, is restricted to a narrower vertical range at border locations. These results contribute to the general understanding of mechanisms that lead to population persistence at distributional limits and to predict population resilience under a scenario of environmental change.  相似文献

Natural F1 hybrids between the outcrossingPhlox drummondii and the predominantly selfingP. cuspidata were examined to ascertain the proportion of hybrid individuals mothered by each species. Species-specific restriction fragment patterns (both nrDNA and cpDNA) were established as markers, and synthetic hybrids of known parentage were utilized to determine that the chloroplast genome is maternally inherited. Of 89 mature natural hybrids examined, approximately two thirds were mothered byP. drummondii, the outcrosser. That the outcrosser should mother most hybrids is expected since it is dependent upon incoming pollen for fertilization, and hybrids may result when heterospecific pollen is received. The fact that the highly selfingP. cuspidata mothered nearly one third of the hybrids is surprising, and may be related to both pre-zygotic and post-zygotic factors. Which species mothers hybrids has important implications for the potential for introgression as well as its direction.  相似文献

Natural hybrids between Ficus septica and two closely related dioecious species, F. fistulosa and F. hispida, were confirmed using amplified fragment length polymorphisms (AFLP) and chloroplast DNA markers. Ficus species have a highly species‐specific pollination mutualism with agaonid wasps. Therefore, the identification of cases in which breakdown in this sophisticated system occurs and the circumstances under which it happens is of interest. Various studies have confirmed that Ficus species are able to hybridize and that pollinator‐specificity breakdown can occur under certain conditions. This study is the first example in which hybrid identity and the presence of hybrids in the natural distribution of parental species for Ficus have been confirmed with molecular markers. Hybrid individuals were identified on three island locations in the Sunda Strait region of Indonesia. These findings support Janzen's (1979) hypothesis that breakdown in pollinator specificity is more likely to occur on islands. We hypothesized that hybrid events could occur when the population size of pollinator wasps was small or had been small in one of the parental species. Later generation hybrids were identified, indicating that backcrossing and introgression did occur to some extent and that therefore, hybrids could be fertile. The small number of hybrids found indicated that there was little effect of hybridization on parental species integrity over the study area. Although hybrid individuals were not common, their presence at multiple sites indicated that the hybridization events reported here were not isolated incidences. Chloroplast DNA haplotypes of hybrids were not derived solely from one species, suggesting that the seed donor was not of the same parental species in all hybridization events.  相似文献

The high degree of morphological plasticity displayed by species of the brown algal genus Fucus L. is well documented. Such variation is especially pronounced for those estuarine taxa lacking holdfasts (termed ecads) that often bear little resemblance to the attached species from which they are derived. To better understand the systematics of salt marsh fucoids, we developed a suite of four microsatellite‐containing loci capable of distinguishing between F. vesiculosus L. and F. spiralis L. The genetic markers were used to determine the relationships of the fucoid ecads F. vesiculosus ecad volubilis (Hudson) Turner and a muscoides‐like Fucus in the Brave Boat Harbor (ME, USA) estuary. Ecad populations had 2‐ to 3‐fold higher levels of heterozygosity than attached populations of F. vesiculosus and F. spiralis. Further, ecads were “intermediate” between F. vesiculosus and F. spiralis in their allele frequencies and genotype composition. Our data indicate that populations of muscoides‐like Fucus in Brave Boat Harbor mainly consist of F1 hybrids between F. vesiculosus and F. spiralis, whereas F. vesiculosus ecad volubilis may arise through backcrosses between F. vesiculosus and other fertile hybrids. Finally, our data support the hypothesis that introgression has occurred between attached populations of F. vesiculosus and F. spiralis.  相似文献
