The morphogenetic changes of the serosal membrane during embryonic development of Cardiochiles nigriceps Viereck (Hymenoptera : Braconidae) were investigated. Eggs observed soon after oviposition into the natural host Heliothis virescens (F.) (Lepidoptera, Noctuidae) showed a transparent chorion and a uniform texture. After 5 hr, the embryo exhibited a distinct granular appearance and by 12 hr attained the germ band stage. A serosal membrane originated from the anterior pole of the embryo between 14 and 15 hr after the egg was laid, eventually forming with the cells both in the anterior and posterior pole a continuous envelope around the developing embryo.Ultrastructural observations revealed that the serosal cells in contact with the abdominal region of the embryo, beginning 24–25 hr after oviposition, formed a syncytium. However, the syncytial tissue did not extend to the cells around the head and thorax. The serosal cells at both embryo poles increased in size without losing their structural organization, and developed into teratocytes when the larva hatched. In contrast, the serosal cells surrounding the body of the embryo persisted longer on the head and thorax region of the newly hatched larva, while the syncytial tissue degraded more rapidly after hatching.In vitro rearing experiments showed that C. nigriceps embryos removed from parasitized host larvae just before and just after serosa formation, hatched only when the medium used was formulated with the addition of fetal bovine serum. Embryos did not develop or hatch when placed in a serum-free medium. Once the syncytium deriving from the serosal membrane became evident, embryos readily developed and hatched in serumfree media. The results of this study seem to suggest that the serosal embryonic membrane could have a nutritional role for the developing parasitoid embryo. 相似文献

Insect endoparasitoids modulate the host physiology through the injection of maternal-derived substances into the host, inducing physiological and hormonal changes in the host's internal environment to benefit parasitoid development. These changes are direct to control host development and regulate nutrient availability to the developing parasitoid, and they are synchronized with parasitoid development. Eggs of some of these parasitoids have low yolk content and require nutrients from the host hemolymph to initiate and complete embryogenesis. We report changes in the amino acid composition and protein profile of the host hemolymph of the endoparasitoid Toxoneuron nigriceps, and improved the in vitro culture of pre-germ band stage eggs. The protein profile of parasitized larvae was similar to controls throughout the embryonic development, but total amino acid concentration decreased in the first 2 h after parasitization, significantly increasing in the following hours up to 8 h. Amino acid levels were higher in parasitized larvae from 16 to 28 h after parasitization. Comparison of single amino acids indicated amino acids involved in energy metabolism (Krebs cycle) followed a trend during parasitoid embryogenesis, and their changes were correlated with embryonic development. Improvement in the in vitro development of 6 h-old eggs of T. nigriceps was obtained by adding factors released by the host fat body to the artificial medium, while a cell lysate stimulated embryogenesis and allowed the full development of newly laid eggs in vitro. 相似文献

Heliothis virescens (F.) last instar larvae parasitized by the endophagous braconid Cardiochiles nigriceps Viereck fail to attain the pupal stage, due to a parasitoid-induced alteration of ecdysteroid biosynthesis and metabolism. Currently available information on host prothoracic gland inactivation in this host-parasitoid system is reported here. Prothoracic glands of H. virescens mature larvae show a depressed biosynthetic activity, without undergoing gross morphological disruption. The ultrastructure of gland cells is characterized by minor parasitoid-induced changes, with the rough endoplasmic reticulum appearing more developed and electrondense than in nonparasitized controls. Eventually, the cells of prothoracic glands of parasitized host last instar larvae die but maintain their structural integrity. The inactivation of pupally committed host prothoracic glands is achieved through the disruption of the PTTH signal transduction pathway. The second messenger cAMP appears to be normally produced in response to PTTH stimulation of glands explanted from parasitized host larvae, however the downstream activation of the cAMP-dependent protein kinase does not appear to occur. In fact, a marked underphosphorylation of regulatory target proteins is observed. This underphosphorylation is associated with a significant reduction in general protein synthesis, which appears to be blocked at the translational level, to a redirection of specific protein synthesis and to a drastic suppression of ecdysteroidogenesis. These parameters appeared to be correlated in a kinetic time-course study, confirming their functional link. C. nigriceps polydnavirus (CnPDV) plays a major role in the inactivation of pupally committed host prothoracic glands, while putative factors occurring in the host haemolymph do not seem to be of particular importance at that developmental stage. Southern blot hybridization indicates the occurrence of PKI(protein kinase inhibitor)-like genes in the C. nigriceps genome, which, in contrast, are undetectable in H. virescens. 相似文献

Scanning and transmission electron microscopy studies were conducted on the antennal sensory sensilla of the hymenopteran parasitoid, Cardiochiles nigriceps Viereck, of the family Braconidae. Distinct morphological differences were found between the chemoreceptors of the male and female. Curved, non-fluted, thin-walled sensilla were found to be very abundant on the male and restricted in location and number on the female. Trichoid, placoid and fluted basiconic sensilla were numerous on the antennal flagella of both sexes. Smooth basiconic sensilla were restricted in number to one per flagellar segment in both sexes. Behavioral data suggest that bent-tipped, thick-walled sensilla unique to the female are involved in detecting a chemical(s) emitted from the host, Heliothis virescens (Fab.). 相似文献

To compare host-searching and ovipositional behavior of Cardiochiles nigriceps Viereck (CN) in tobacco and cotton, behavior of CN females was observed on Hellothis virescens (Febricius)-infested plants and recorded using a Psion Organizer II with Observer software. Hover, search, oviposit, preen, rest, walk, agony-search, drop-search, and miss-search were the nine behaviors observed for each plant species. Agony-search was a searching behavior exhibited when CN females were prevented from ovipositing in a host because they were unable to reach the host. Drop-search and miss-search were the searching behaviors that occurred by CN females after a host dropped from the plant and when CN females missed ovipositing in a host, respectively. Hovering, searching, oviposition, miss-search, resting, and walking were equally common for CN on tobacco and cotton. Agony-search was exhibited more on tobacco than cotton, while drop-search and preening occurred more on cotton than tobacco. Female CN spent more time hovering in tobacco than in cotton, indicating that these females preferred tobacco to cotton. Female parasitoids also spent more time in the main ovipositional sequence, hover–search–oviposit, in tobacco than in cotton, mainly because females hovered more in tobacco than in cotton. Frequency and time of behavioral sequences associated with experiencing frustration in ovipositing due to difficulty in reaching or finding a host and subsequent level of success in ovipositing in a new host were determined. Overall, the rate of successful oviposition by CN females was significantly higher in tobacco (87.7%) than in cotton (79.5%). For cotton and tobacco, the rate of successful oviposition by CN females was significantly reduced (ca. 40.0%) when they experienced frustration in ovipositing compared to the rate of ovipositional success (100%) for CN females when hosts were present and easily accessible. Encountering ovipositional frustration also increased the duration of time for successful oviposition in comparison to the time spent for problem-free oviposition in cotton and tobacco. Interestingly, with successful oviposition, the amount of time spent in a behavioral sequence was not significantly different for tobacco and cotton regardless of whether females experienced ovipositional frustration or not. However, female CN invested more time in tobacco than in cotton when they were unsuccessful in ovipositing due to the inaccessibility or disappearance of hosts. CN females' preference for tobacco over cotton probably gave them more motivation to continue searching for hosts, especially those not easily attained, on tobacco than on cotton. Nevertheless, CN females readily searched in cotton for HV even in the presence of host-infested tabacco. CN females possibly could be successful in maintaining levels of HV below an economic threshold when using a tobacco trap crop to protect cotton as the main crop. 相似文献

Larval development of the parasitoid Cardiochiles nigriceps Viereck occurs in the last instar larva of its host, Heliothis virescens (F.). This allows the parasitoid to exploit the nutritional increase in the biosynthetic activity occurring in the host in preparation for metamorphosis. To understand the biochemical basis of this host parasitoid developmental synchrony, we undertook host ligation studies and analyzed host hemolymph for proteins and glycerol esters. Parasitization affected the biochemical profile of the host. The hemolymph protein concentration of parasitized last instar H. virescens larvae increased through time, whereas unparasitized (control) larvae were characterized by a decrease in the protein titer when they reached the prepupal stage. The effect of parasitism on glyceride titers of host hemolymph was not as pronounced as the effect on proteins. Ligation conducted on 5th instar hosts, which were parasitized as 4th instars, affected parasitoid development in a time-dependent way. The percentage of successfully developing C. nigriceps larvae increased with the increase of the time interval between parasitization and ligation. Ligation performed before day 2 of the 5th larval instar of H. virescens completely inhibited parasitoid development. Ligations that disrupted parasitoid developmentwere associated with a low host hernolymph protein concentration. Parasitoid development was successful when hernolymph protein titer was high, as occurred when ligations were performed after day 3 of the 5th host instar in both control and parasitized larvae. Ligations in both situations resulted in a slight increase in glyceride titers. The results suggest that host proteins and/or some factor(s) associated with them may play a role in parasitoid growth and development. © 1993 Wiley-Liss, Inc. 相似文献

Teratocytes deriving from the serosal membrane of Cardiochiles nigriceps Viereck, obtained “in vitro” from embryos hatched on a semidefined medium, were injected at different numbers and in different developmental stages of nonparasitized Heliothis virescens (F.) last instar larvae. Host development was affected by teratocyte injections and the responses registered ranged from normal to complete inhibition of pupation, according to the number of teratocytes injected and the developmental stage of the larva at time of injection. Complete pupation failure was observed when teratocytes derived from 4C nigriceps embryos were injected into 1st day 5th instar (new-slender stage) host larvae. Complete pupation occurred when teratocytes from 2 embryos were injected into 3rd or 4th day 5th instars (burrow-digging or day 1 cell formation stage). Intermediate responses, such as the formation of pupal cuticle without ecdysis or with only partial ecdysis, were obtained with intermediate teratocyte numbers, or host developmental stages. All pupae derived from teratocyte injected larvae failed to develop into adults normally obtained from control injected larvae. The larval weight just before pupation was negatively affected only when teratocyte injections were performed on 1st day 5th instar H. virescens larvae. Teratocyte injections altered the hemolymph protein titer to a level similar to that occurring in parasitized larvae. At the same time the ecdysteroid titer was characterized by a late significant increase, which reached values almost 3 times greater than found in normally parasitized larvae, and also surpassed the highest values registered for nonparasitized larvae. Ligation of parasitized larvae between the meso- and metathorax demonstrated that when the prothoracic glands were excluded, there was almost no ecdysteroid production posterior to the ligation. Ligations performed on parasitized larvae to isolate parasitoid eggs before hatching in the last abdominal segments, demonstrated that only virus and venom determined a reduction of the ecdysteroid titer. On the basis of these results the possible role of teratocytes in affecting the biological activity of ecdysteroids is postulated and discussed in a wider context of host-parasitoid physiological interactions. 相似文献

We evaluated the toxicity of three insecticides (lambda cyhalothrin, spinosad, and S-1812) to the natural enemies Bracon mellitor Say, Cardiochiles nigriceps Viereck, Coleomegilla maculata De Geer, Cotesia marginiventris (Cresson), Geocoris punctipes (Say), and Hippodamia convergens Guérin-Méneville, in topical, residual, and field assays. Lambda cyhalothrin exhibited the greatest toxicity to the natural enemies. In topical toxicity tests, lambda cyhalothrin adversely affected each natural enemy species studied. Residues of lambda cyhalothrin on cotton leaves were toxic to B. mellitor, C. nigriceps, C. maculata, and C. punctipes. Interestingly, residues of this insecticide were not very toxic to C. marginiventris and H. convergens. Geocoris punctipes and C. maculata numbers in the field generally were significantly lower for lambda cyhalothrin treatments than for the other four treatments, substantiating the previous tests. Although cotton aphids began to increase over all treatments around the middle of the test period, the number of cotton aphids in the lambda cyhalothrin plots was significantly higher than the number in any of the other treatments. As cotton aphids increased in lambda cyhalothrin field plots, the predator H. convergens also increased in number, indicating that lambda cyhalothrin did not adversely affect it in accordance with the residual tests. Spinosad exhibited marginal to excellent selectivity, but was highly toxic to each parasitoid species and G. punctipes in topical toxicity tests and to B. mellitor in residual tests. Spinosad generally did not affect the number of G. punctipes, H. convergens, and C. maculata in the field except for one day after the second application for G. punctipes. S-1812 exhibited good to excellent selectivity to the natural enemies. Some reduction of G. punctipes occurred for only a short period after the first and second application of this insecticide in the field. H. convergens and C. maculata were affected very little by S-1812. 相似文献

红颈常室茧蜂Peristenus spretus Chen et van Achterberg是一种绿盲蝽若虫的优势内寄生蜂,本研究利用超景深三维显微系统、显微镜观察了红颈常室茧蜂老熟幼虫发育特征、蛹形态特征及蛹发育后期卵巢的超微结构。结果表明:红颈常室茧蜂蛹期为12~15 d,其中雌蛹期比雄蛹期长2~3 d。根据其形态特征,可将蛹划分为预蛹期、第一蛹阶段、第二蛹阶段、第三蛹阶段。蛹期发育前9 d,蛹期组织结构开始分化形成,成蜂的外部形态基本显现,但性别难以区分;蛹期发育至10~14 d,可通过幼蜂体表颜色的变化或腹部末端的形状区分性别,在相同的适宜温度条件下,雌蜂比雄蜂颜色深,雌蜂腹部末端比较尖,有突出的产卵瓣,雄峰腹部末端钝圆,无凸起;雌蜂卵巢在化蛹后的第11天开始形成,卵巢初期的形状如细丝,此时卵巢管已开始初步分化,但卵室尚未分化;蛹发育至第12天时,卵巢管逐渐加粗,卵室开始分化;第13天时,卵巢管、卵室数量逐渐增多;第14天卵巢基部开始有少量成熟卵子产生,呈乳白色。10~12 d,雄蜂腹部末端绒毛逐渐增多,体壁变硬,逐渐角质化,虫体可活动,雄蜂胸部略带黑色,翅逐渐伸展开,整个虫体呈浅红棕色。本研究结果明确了红颈常室茧蜂蛹发育的形态变化过程,提出了蛹期雌雄区分的方法,叙述了蛹后期卵巢发育特征,为研究该蜂繁殖机理奠定了形态学基础。 相似文献

Characters previously used to delimit both Dinotrema and species groups within Aspilota s.l. are discussed, with emphasis on difficulties in interpreting transformation series. Phylogenetic relationships within the Aphidiinae, and between this and other subfamilies of Braconidae (Hymenoptera), were investigated using sequence data from three genes: elongation factor-1α, cytochrome b, and the second expansion segment of the 28S ribosomal subunit. Variation in both protein-coding genes was characterized by a high level of homoplasy, but analysis of the expansion segment—robust over a range of alignment methods and parameters—resolved some of the older divergences. Parsimony analysis of the combined data suggests the following tribal relationships: (Ephedrini + (Praini + (Aphidiini + Trioxini))). In addition, the cyclostome subfamilies were found to form a clade separate from the Aphidiinae, but relationships between the Aphidiinae and the noncyclostome braconids could not be resolved. The inferred phylogeny also supported a secondary loss of internal pupation within the Praini and a polyphyletic origin of endoparasitism within the Braconidae. 相似文献

Abstract.
- 1 We report on the mating system of a field population of the parasitic wasp, Bracon hebetor, on a corn pile infested by the Indian meal moth, Plodia interpunctella. We demonstrate that the mating system is based upon male scramble competition polygyny with male aggregations on high places on the corn.
- 2 The sex ratio among adults was greater than 80% males on the surface of the corn, whereas below the surface the sex ratio was less than 45%. Males actively courted females on the surface, but there were no aggressive interactions among males during courtship or mating.
- 3 Approximately 20% of the females found on the surface of the corn had no sperm in their spermathecae, regardless of age, but the numbers of unmated females decreased later during the day.
- 4 In laboratory studies we showed that females from this population oviposit a female biassed sex ratio, and that only 14% of females were mated before dispersing from their place of emergence.
- 5 Thus sib-mating is unlikely in this gregarious parasitoid. This outcrossing mating system probably arose because of severe inbreeding depression that B.hebetor suffers via a sex locus: diploids that are heterozygous at the sex locus develop into females, but homozygous diploids are male and are generally inviable. The female biassed sex ratio may have evolved in B. hebetor in response to males being the more expensive sex, females dispersing more frequently from the population than males, or a fraction of females remaining unmated in the population.

In the annelid Enchytraeus albidus the ovary is composed of packets containing eight synchronously developing oocytes. Each oocyte in the packet is connected, via a bridge, to a common cytoplasmic mass. Developmental synchrony of oocytes within individual packets is probably related to the ooplasmic continuity. The young previtellogenic oocyte contains many polysomes, a few cisternae of smooth and rough endoplasmic reticulum, small Golgi complexes, and mitochondria. Many of the mitochondria are dumbbell-shaped and may thus represent division stages. Vitellogenesis is marked by the appearance of peripherally located lipid yolk and small, densely staining granules scattered throughout the ooplasm. There is an increase of smooth endoplasmic reticulum, mitochondria, and enlarged Golgi elements. Small multivesicular-like bodies, the early stages of developing yolk, are derived from the Golgi complex. The mature yolk sphere is bipartite and consists of (a) a variable number of dense spheres, the core bodies, which are produced in the ooplasm by the Golgi complex and which become embedded in (b) a dense matrix. The electron opaque tracer, horseradish peroxidase is incorporated into the oocyte and deposited in the matrix suggesting that this component of the yolk sphere is obtained by micropinocytosis. Enzyme digestions and various cytochemical techniques suggest that the core bodies are rich in carbohydrate, probably as glyco- or mucoproteins, and that the matrix is rich in lipid. 相似文献

Twenty-three species are included of which the following eighteen are new to science: achterbergi, anicus, arrogator, depressus, gloriosus, josieas, laevigatus, lindae, obscurus, ottus, palmeri, pinguicornis, sedlaceki, sritus, strabismus, tarius, townesi, varicosus . Primary type-material of five previously-described taxa has been examined: antipodalis Ashmead, bicolor Szépligeti, luteus (Cameron), trichogrammae Wilkinson, dumbletoni Muesebeck. The study of spermatogenesis in Hymenoptera is limited to few taxa due to limitations to access the material in the right stage for analysis. The information available up to now states that the gametogenesis in males involves a modified meiosis, similar to mitosis. This is due to the haplodiploid sex determination system present in the order, where females are diploid and males are haploid. In the present work, we examined the spermatogenesis process in the parasitoid Diachasmimorpha longicaudata (Ashmead) (Hymenoptera: Braconidae). We performed squash preparations from testes of individuals from the third larval instar until the adult stage. Some cytological traits, such as the presence of a monopolar spindle, were analysed in detail by means of histological semi‐thin sections. The spermatogenesis in this species comprises an abortive first division with the formation of an anucleated cytoplasmic bud and a nucleated cell, and an equal second division of the nucleated cell, leading to the generation of two mature spermatozoa. The results presented herein provide more information about the process of spermatogenesis in Hymenoptera, which has not been studied in depth in parasitic species. 相似文献

The early second instar larvae of Toxoneuron nigriceps, a larval endoparastioid of Heliothis virescens, were incubated in artificial rearing media, supplemented with hemolymph of the unparasitized and parasitized fifth instar larvae of the host, H. virescens. The parasitoid larvae were incubated in both a semisolid and liquid form of the artificial rearing medium, and their growth and development were evaluated. The growth in size (increase in length and width), development (molting), and survival of the incubated larvae were observed for 10 days. The incubated larvae exhibited some level of growth in all nine types of media tested, including the control (without host hemolymph). However, ingesting the semisolid rearing media supplemented with the hemolymph from the late fifth instar (day 5, 7 and 9) parasitized host resulted in 100% of the larvae molting to third instars. Some of the in vitro reared third instar larvae demonstrated behavioral changes that could be interpreted as the preparation for cocoon formation or pupation i.e. oral secretion of a whitish material and lots of twisting and turning; however, none produced a cocoon nor pupa. 相似文献
