Pro-inflammatory cytokines secreted by tissue macrophages recruit polymorphonuclear leukocytes and evoke fever, cachexia and production of acute phase proteins. This study investigates whether Gram-positive and Gram-negative bacteria equally and efficiently trigger production of the pro-inflammatory cytokines IL-1 beta, IL-6, IL-8 and TNF-alpha in human monocytes. A range of aerobic and anaerobic Gram-positive and Gram-negative bacteria were killed by UV-light and added in different concentrations to human monocytes. Cytokines were measured in 24 h supernatants by ELISA. Gram-positive and Gram-negative bacteria were equally efficient inducers of IL-1 beta, but Gram-positive bacteria generated twice as much TNF-alpha as did Gram-negative bacteria (p<0.001 for 25 and 250 bacteria/cell). In contrast, Gram-negative bacteria induced at least twice as much IL-6 and IL-8 as did Gram-positive bacteria (p<0.001 for 2.5, 25 and 250 bacteria/cell). While the cytokine responses to LPS were similar to those induced by the corresponding amount of Gram-negative bacteria, the strong IL-1 beta and TNF-alpha responses to Gram-positive bacteria could not be induced by soluble peptidoglycan or lipotheicoic acid. The particular nature of the bacteria, thus seem to modify the response to Gram-positive bacterial components. The different cytokine profiles evoked by Gram-positive and Gram-negative bacteria might optimize clearance of bacteria that differ in cell wall structure.  相似文献

Extensive colonization of the rhizosphere by beneficial bacteria is desirable but it is not yet clear whether the feeding patterns of protozoa exert any ecological impact on the competence of bacteria in the rhizosphere. To determine whether the successful colonization of the rhizosphere by bacteria can be explained by inherent abilities of the bacteria to resist predation, six bacteria categorized as poor, mediocre or good rhizosphere colonists were tested. The populations of protozoa in the rhizosphere consistently increased to a maximum density in two days but then declined to a stable level. Grazing by protozoa increased the doubling time of all of the test bacteria but did not significantly affect the final densities of bacteria in the rhizosphere. The differences in the colonization ability of the six bacteria tested were not attributable to inherent differences in their susceptibility to predation by protozoa.  相似文献

棉花根际亲和性高效促生细菌的分离筛选   总被引:3,自引:0,他引:3  
为了从棉花根际土壤筛选能与棉花凝集素具有亲和作用的高效促生细菌,以选择性培养基从棉花根部初步筛选具有固氮能力、解磷能力及解钾能力的促生细菌,再以异硫氰酸磺(FITC)标记的棉花凝集素为复筛工具,从棉花根际促生细菌中筛选能与棉花凝集素结合的亲和性菌株,分别挑选2株固氮菌、2株解磷细菌和2株解钾细菌作为微生物肥料接种到棉花根部进行盆栽试验.观察其在根部定殖情况.结果是在选择性平板上有20%~30%的菌株具有凝集素染色阳性.盆栽试验显示,接种的6株亲和性菌株能在棉花根部成功定殖,根际细菌数量约是灭活对照的`0倍.通过初步鉴定,固氮菌株N1111为固氮菌属(Azotobacter),N2121属于德克斯氏菌属(Derxia);解磷菌株P2126属于黄单胞菌属(Xanthomonas),P1108菌株为假单胞菌属(Pseudomonas);解钾菌株K2204和K2116属于芽孢杆菌属(Bacillus).  相似文献

A study has been made of the promoting effect of starch on cellulose digestion by mixed rumen bacteria in a cellulose-urea medium. Starch supplementation of the medium promoted the growth of bacteria that required neither amino acids (AA) nor branched-chain fatty acids (BrFA). The growth of these bacteria was followed by the growth of AA-dependent bacteria, AA- or BrFA-dependent bacteria, BrFA-producing bacteria, and finally, BrFA-dependent cellulolytic bacteria. Population changes of these bacterial groups corresponded with a cross-feeding of AA and BrFA and the overall disappearance of cellulose. The data suggest that the nutritional interdependence among rumen bacteria affects the rate of cellulose digestion.  相似文献

Salvia miltiorrhiza Bunge (Lamiaceae) hairy root cultures were inoculated (at 0.02 and 0.2% v/v) and co-cultured with Bacillus cereus bacteria. The root biomass growth was inhibited significantly by the bacteria inoculated to the root culture on the first day (day 0) but not by the bacteria inoculated on days 14 or 21 (in a 28-day overall period). On the other hand, the growth of the bacteria in the hairy root culture was also strongly inhibited by the hairy roots, partially because of the antibacterial activity of the secondary compounds produced by the roots. Most interestingly, the tanshinone production was promoted by the inoculation of bacteria at any of these days but more significantly by an earlier bacteria inoculation. With 0.2% bacteria inoculated on day 0, for example, the total tanshinone content of roots was increased by more than 12-fold (from 0.20 to 2.67 mg g−1 dry weight), and the volumetric tanshinone yield increased by more than sixfold (from 1.40 to 10.4 mg l−1). The tanshinone production was also stimulated by bacterial water extract and bacterial culture supernatant but less significantly than by the inoculation of live bacteria. The results suggest that the stimulation of tanshinone production by live bacteria in the root cultures may be attributed to the elicitor compounds originating from the bacteria, and the hairy root–bacteria coculture may be an effective strategy for improving secondary metabolite production in plant tissue cultures.  相似文献

PURPOSE: To identify the metabolites produced from an isoflavonoid, daidzein, by colonic bacteria of rhesus monkeys. METHODS: The metabolism of daidzein by the fecal bacteria of nine monkeys was investigated. Daidzein was incubated anaerobically with fecal bacteria, and the metabolites were analyzed by use of liquid chromatography and mass spectrometry. RESULTS: The fecal bacteria of all of the monkeys metabolized daidzein to various extents. Dihydrodaidzein was found in cultures of fecal bacteria from two monkeys; dihydrodaidzein and equol were found in cultures from four monkeys; dihydrodaidzein, equol, and an unknown metabolite (MW = 244) were found in cultures from one monkey; and dihydrodaidzein and the unknown metabolite were found in cultures from two monkeys. CONCLUSIONS: Similar to that in humans, variation was evident in the metabolism of isoflavonoids by fecal bacteria from rhesus monkeys. Some metabolites produced by fecal bacteria from monkeys were the same as those produced by fecal bacteria from humans.  相似文献

The character of interaction between saprophytic soil bacteria via gaseous metabolites was studied. It was established that, at the metabolic level, a diverse character of interspecies interrelationships between bacteria exist, directly influencing their reproduction and preservation in soil. Volatile compounds produced by bacteria are able to act as both intra-and interspecies regulators of microbial communities. The soil microbiocenosis composition may be therefore regulated by volatile products of metabolism of saprophytic soil bacteria. Methanol released by bacteria into the environment plays an important role in this process.  相似文献

目的初步筛选牛瘤胃中纤维素降解菌。方法分别采用基本培养基(牛肉膏蛋白胨培养基、马丁培养基),利用好氧、兼性和厌氧3种不同的培养方法进行初选,初步分离牛瘤胃中的细菌与真菌,再通过复选培养基(加入微晶纤维素),筛选降解纤维素的菌种。结果筛选分离出降解纤维素的1株厌氧细菌和1株厌氧真菌。结论此实验方法简单易行,能够有效地从牛瘤胃中筛选出生长良好的纤维素降解菌。  相似文献

The behavior of collections of oceanic bacteria is controlled by metabolic (chemotaxis) and physical (fluid motion) processes. Some sulfur-oxidizing bacteria, such as Thiovulum majus, unite these two processes via a material interface produced by the bacteria and upon which the bacteria are transiently attached. This interface, termed a bacterial veil, is formed by exo-polymeric substances (EPS) produced by the bacteria. By adhering to the veil while continuing to rotate their flagella, the bacteria are able to exert force on the fluid surroundings. This behavior induces a fluid flow that, in turn, causes the bacteria to aggregate leading to the formation of a physical pattern in the veil. These striking patterns are very similar in flavor to the classic convection instability observed when a shallow fluid is heated from below. However, the physics are very different since the flow around the veil is mediated by the bacteria and affects the bacterial densities.  相似文献

Many different bacteria secrete proteases into the culture medium. Extracellular proteases produced by Gram-positive bacteria are secreted by a signal-peptide-dependent pathway and have a propeptide located between the signal peptide and the mature protein. Many extracellular proteases synthesized by Gram-negative bacteria are also produced as precursors with a signal peptide. However, at least two species of Gram-negative bacteria secrete one or more proteases via a novel signal-peptide-independent route. Most proteases secreted by Gram-negative bacteria also have a propeptide whose length and location vary according to the protease. Specific features of protease secretion pathways and the mechanisms of protease activation are discussed with particular reference to some of the best-characterized extracellular proteases produced by Gram-positive and Gram-negative bacteria.  相似文献

The marine bacteria Beneckea harveyi and Photobacterium leiognathi were shown to bear mannose-containing binding sites for the mannosephilic lectins of Pseudomonas aeruginosa and concanavalin A (Con A). The interaction between the lectins and the marine bacteria was demonstrated by the bacteriagglutination test, by adsorption of the lectins onto the bacteria and by mannose-specific peroxidase-binding to the lectin-coated bacteria. Treatment of the bacteria with formaldehyde, phenol, ethanol or boiling them for 15 min, did not alter their ability to adsorb the lectins. The growth rate of the marine bacteria was unaffected when either the Pseudomonas lectins or Con A was added to the culture medium.  相似文献

目的肠道细菌对家蚕具有重要的生理作用,探明家蚕肠道产蛋白酶细菌菌群对家蚕肠道细菌的高效开发与利用具有十分重要的意义。方法从菁松×皓月品种4龄家蚕肠液中分离与纯化肠道细菌,获得一株产高活力蛋白酶细菌,对该菌菌落进行形态学鉴定及菌体的显微镜检并结合16S rDNA方法进行了鉴定。结果该菌为革兰阴性杆菌,属于嗜麦芽寡养单胞菌(Stenotrophom onas maltophilia)。结论该产蛋白酶细菌产酶能力较高,可进一步开发研究并用作微生态制剂。  相似文献

Aims:  To expose bacteria to anolyte and subsequently investigate the effect of anolyte on the protein profiles of treated bacteria.
Methods and Results:  Proteins were extracted from bacteria treated with different concentrations of anolyte and analysed using SDS-PAGE. Fewer and more faint protein bands were observed for concentrated halide anolyte treated bacteria when compared to untreated bacteria while extra protein bands were observed for bacteria exposed to dilute concentrations.
Conclusions:  The undiluted and the 10−1 dilution of halide derived anolyte was effective in killing the test bacteria. Anolyte caused bacterial death by complete destruction of proteins or by causing oxidative stress which resulted in protein fragmentation.
Significance and Impact of the Study:  The results of this study provide information on the antimicrobial mechanism of anolyte on other bacteria for which the information is currently unavailable.  相似文献

In this study we analyzed the numbers and types of cultivable tylosin-resistant bacteria from six agricultural soils that differed with respect to their association with subtherapeutic antibiotic use. The proportion of tylosin-resistant bacteria to the total number of bacteria cultivated was significantly higher (7.2-16.5%) at three sites affected by subtherapeutic antibiotic use compared to three sites unaffected by subtherapeutic antibiotic use (0.7-2.5%). We also detected differences in the types of cultivable tylosin-resistant bacteria. At a site affected by subtherapeutic antibiotic use, we detected tylosin-resistant bacteria from the alpha- and beta-subdivisions of Proteobacteria. In contrast, at a site unaffected by subtherapeutic use, we detected only Streptomyces-like (high-G+C Gram-positive) tylosin-resistant bacteria. Our results suggest a link between subtherapeutic use of antibiotics and the numbers and types of antibiotic-resistant bacteria in nearby soil. However, other factors, such as soil type and temporal variation, may have also contributed to the differences observed.  相似文献

为了考察生物脱氮系统中硝化菌群(氨氧化菌和亚硝酸氧化菌)的种群多样性及硝化菌群随溶解氧降低的种群变化规律,并建立一套行之有效的用于自养生物脱氮系统中功能微生物菌群的快速分子检测技术,采用DGGE(变性梯度凝胶电泳)分子检测技术对硝化菌群的16SrDNA的特异性PCR扩增产物进行了分析,结果表明:OLAND生物脱氮系统中氨氧化菌和亚硝酸氧化菌随溶解氧的降低表现出了不同的种群变化规律,氨氧化菌种群多样性受溶解氧的影响非常大,而非亚硝酸氧化菌的种群多样性比较单一,且不受溶解氧的影响。结合FISH(全细胞荧光原位杂交)分析结果表明,在OLAND限氧稳定运行后期,亚硝化单胞菌属(Nitrosomonas)是主要的氨氧化菌,占OLAND限氧亚硝化阶段反应器中总细菌数的72.5%左右。  相似文献
